Aims: Phytohormones are a group of signal compounds that regulate plant growth, development, health and plant-soil interactions. We aimed to investigate the distribution and controlling factors of a diverse set of phytohormones found in the plant-soil continuum. Methods: Twenty phytohormones belonging to seven categories were studied in maize roots, rhizosphere and bulk soil with ultra-high-performance liquid chromatography–tandem mass spectrometry. In addition, the microbiomes shaped by gamma-sterilization and reinoculation were analyzed using high throughput sequencing. Results: We found three major distribution patterns of the 17 phytohormones detected along the root-soil continuum. In the first of these, the phytohormones were predominant in roots, followed by rhizosphere and then by bulk soil. In the second pattern, concentrations of the phytohormones were smallest in the rhizosphere. While, in the third pattern, concentrations of the phytohormones were greatest in the rhizosphere. The concentrations of the phytohormones were closely related to those of root phytohormone or roots biomass as well as relative abundances of wide range of microbial taxa. Conclusions: The results indicated that the distribution patterns of six categories of phytohormones in the root-soil continuum are driven by a balance between the production and uptake by the roots and the production, utilization and degradation by the soil microbiome in a phytohormone-specific manner. Background and aims: Silicon (Si) deficiency is a major constraint on rice production. The objective of this study was to evaluate the long-term influence of phytolith-rich straw return and groundwater table management on labile Si fractions in paddy soil and subsequent plant Si uptake. Methods: A field experiment was conducted over 36 years in subtropical China with different application doses of phytolith-rich straw and a groundwater table of either 20 or 80 cm. An optimized sequential chemical extraction procedure allowed us to determine labile Si fractions, represented by CaCl2-Si, Acetic-Si, H2O2-Si, Oxalate-Si, and Na2CO3-Si. Additional analyses included the determination of amorphous silica particles in soil, phytoliths in supplied straw, Si in planted rice straw, and the dissolution rate of phytoliths extracted from supplied straw. Results: Long-term application of phytolith-rich straw significantly increased the H2O2-Si and Na2CO3-Si contents. The CaCl2-Si (5.21–7.91 mg kg− 1), H2O2-Si (50.0–72.4 mg kg− 1) and Na2CO3-Si (3.33–4.60 g kg− 1) contents were positively correlated with soil organic carbon. The Si content (13.6–28.9 g kg−‍1) in planted rice straw significantly (p < 0.05) increased with the application dose of phytolith-rich straw under both groundwater tables. This effect was significantly (p < 0.05) greater under 80 cm groundwater table than under 20 cm groundwater table for matching straw amendments. Conclusions: This study indicates that long-term application of phytolith-rich straw and groundwater management significantly increase soil Si bioavailability by promoting accumulation of organic matter and phytoliths, and enhancing the soil-plant Si cycle. Background and aims: We investigated the effects of silicon (Si) on chlorophyll concentration, photosynthesis, leaf chloroplast ultrastructure, and expression of genes involved in photosynthesis to elucidate the mechanisms through which Si mediated alleviation of manganese (Mn) toxicity in rice ( Oryza sativa L.). Methods: Rice seedlings were grown hydroponically with normal Mn (6.7 μM) or high Mn (2 mM) concentrations, both with (1.5 mM) and without Si supplementation. Leaf chloroplast ultrastructure was observed by scanning and transmission electron microscopy. Differentially expressed genes relating to photosynthesis were identified by high-throughput sequencing, and their relative expression levels were evaluated by real-time quantitative PCR. Results: Chlorophyll and carotenoid concentrations and net photosynthesis decreased with chloroplast degradation under high Mn stress. High Mn concentrations may have inhibited photosynthesis through several mechanisms, including suppressing chlorophyll and ATP synthesis, decreasing light-harvesting processes, impairing photosystem I (PSI) stability and structure, and slowing activity of phosphoribulokinase. Si enhanced Mn tolerance efficiently by increasing chlorophyll concentration, light-use efficiency, and ATP concentration as well as by stabilizing the structure of PSI and promoting CO assimilation. Background and aims: This study aimed to investigate the roles of silicon (Si) in ameliorating manganese (Mn) toxicity in two rice ( Oryza sativa L.) cultivars: i.e. cv. Xinxiangyou 640 (XXY), a Mn-sensitive cultivar and cv. Zhuliangyou 99 (ZLY), a Mn-tolerant cultivar. Methods: Plants were cultured in nutrient solution containing normal Mn (6.7 μM) or high Mn (2.0 mM), both with or without Si supply at 1.5 mM Si. Results: Plant growth was severely inhibited by high Mn in cv. XXY, but was enhanced by Si supply. In cv. XXY, Si-enhanced tolerance resulted from a restriction of Mn transport, whereas in cv. ZLY Mn uptake was depressed. In cv. XXY, high Mn significantly increased superoxide dismutase (SOD), catalase and ascorbate peroxidase activities but decreased non-protein thiols and glutathione concentrations, leading to accumulation of HO and malondialdehyde. The addition of Si significantly counteracted high Mn-elevated malondialdehyde and HO concentrations and enhanced plant growth. In cv. ZLY, high Mn considerably raised SOD activities and glutathione concentrations, thus leading to relatively low oxidative damage. Conclusions: Si-enhanced Mn tolerance was attributed mainly to restricted Mn transport in cv. XXY but to depressed Mn uptake in cv. ZLY. The objective of this study is to elucidate the roles of silicon (Si) in enhancing tolerance to excess zinc (Zn) in two contrasting rice ( Oryza sativa L.) cultivars: i.e. cv. TY-167 (Zn-resistant) and cv. FYY-326 (Zn-sensitive). Root morphology, antioxidant defense reactions and lipid peroxidation, and histochemical staining were examined in rice plants grown in the nutrient solutions with normal (0.15 μM) and high (2 mM) Zn supply, without or with 1.5 mM Si. Significant inhibitory effects of high Zn treatment on plant growth were observed. Total root length (TRL), total root surface area (TRSA) and total root tip amount (TRTA) of both cultivars were decreased significantly in plants treated with high Zn, whereas these root parameters were significantly increased when Zn-stressed plants were supplied with 1.5 mM Si. Supply of Si also significantly decreased Zn concentration in shoots of both cultivars, indicating lower root-to-shoot translocation of Zn. Moreover, superoxide dismutase (SOD), catalase (CAT), and asorbate peroxidase (APX) activities were increased, whereas malondialdehyde (MDA) and hydrogen peroxide (HO) concentrations were decreased in Si-supplied plants of both Zn-sensitive and Zn-resistant rice cultivars exposed to Zn stress. These alleviative effects of Si, further confirmed by the histochemical staining methods, were more prominent in the Zn-resistant cultivar than in the Zn-sensitive one. Taken together, all these results suggest that Si-mediated alleviation of Zn toxicity is mainly attributed to Si-mediated antioxidant defense capacity and membrane integrity.
