The present study aimed to assess the preclinical toxicity of two plants commonly used to treat "stomach ailments" in Brazil: Schinus terebinthifolius Raddi (S) and Myracrodruon urundeuva Allemao (M). In male rats, chronic treatment (83 days) with both pepper trees (17.6 and 13.8 mg/kg, S and M, respectively) has been shown to decrease hematocrit. However, a reduction in the number of red blood cells and hemoglobin was only seen following administration of S. terebinthifolius. None of the plants caused anatomopathological alterations following chronic treatment, and mating ability and fertility were not affected. Both pepper trees showed moderate toxicity following acute and chronic treatment by gavage, particularly S. terebinthifolius. Moreover, bone malformations were induced in fetuses, and a slight delay in recovery time of the postural reflex was observed in pups from female animals treated (18 days) with S. terebinthifolius. Given these results, a better assessment of the risks and benefits of the internal use of these plants is necessary, especially when used by women of childbearing age.

The present work showed the effects of Myracrodruon urundeuva Fr. All., popularly known as ‘aroeira’ (AE), in the form of enemas prepared from the stem bark, on several morphologic and morphometric parameters after acetic acid-induced colitis in rats. Enemas from 5-ASA were used as standard while the vehicle, carboxymethylcellulose, was used as a control. The results of the morphological evaluation showed that on day 1 acetic acid produced significantly more necrosis in the groups treated with AE (10% and 20%) or 5-ASA than the controls. However, on day 60, there were more caliciform and absorptive cells in the treated groups compared with the controls. A significantly higher number of eosinophil and mononuclear cells and also collagen deposition in the controls compared with the treated groups were observed on day 60. However, a higher number of polymorphonuclear cells was detected on day 60 only in the AE treated group but not in the 5-ASA group. These data indicate that animals treated with AE or 5-ASA showed complete epithelial tissue regeneration, while in the controls chronic inflammatory exudate persisted and tissue regeneration occurred through fibrosis. The bark of M. urundeuva (Anacardiaceae) used in popular medicine as an antidiarrhoeal agent has been investigated for its effects on castor oil-induced diarrhoea and on intestinal motility. In rats, the ethanol extract of stem bark significantly inhibited the castor oil-induced diarrhoea, small intestinal transit and accumulation of fluid volume at an oral dose of 400mg/kg. Furthermore, the plant extract caused marked inhibition of the contractile responses evoked by acetylcholine and histamine on guinea-pig ileum in vitro. These results point to a possible antidiarrhoeic effect of M. urundeuva bark extract since inhibition of intestinal motility and secretion can greatly control clinical diarrhoea.

The present work showed a significant antinociceptive activity in the tannin fraction (TF) extracted from the bark of Myracrodruon urundeuva Fr. All. This inhibitory effect was demonstrated not only against abdominal contractions but also in the formalin test in mice. In the first case, at doses of 0.1 and 1 mg/kg, i.p. the TF caused inhibitions of the order of 39.6% and 80.8%, respectively, and in the second one, inhibitions of 47.8% and 77.2% (phase I) and 59.2% and 100% (phase II), after the administration of 5 and 10 mg/kg, i.p. The antinociceptive effect was partially reverted by naloxone. The TF presented also an antioedematogenic effect in rat paw oedema induced by carrageenan as well as dextran. In the carrageenan model, significant inhibitions were seen at 2 h (29.7% and 41.7%) and 3 h (40.5% and 44.2%), after administration of 5 and 10 mg/kg, i.p. In the dextran induced oedema, the TF (10 mg/kg, p.o.) caused inhibitions of 29.2%, 42.4% and 54.5% at 2 h, 3 h and 4 h, respectively. The TF (10 and 25 mg/kg, i.p.) significantly inhibited the inflammatory events (vesical oedema and increased vascular permeability) which occur at the onset of the haemorrhagic cystitis induced by cyclophosphamide. After subcutaneous or oral administration, the TF (5–50 mg/kg) also blocked neutrophil migration induced by direct (fMLP) as well as indirect (carrageenan) stimuli. © 1997 by John Wiley & Sons, Ltd.
