Ample studies have shown that housing can affect the health, welfare and behavior of mice and therefore, the outcomes of certain experiments. The aim of this study was to investigate if three widely used housing systems, Open Top Cages (OTC), Motor Free Ventilated Cages (MFVC) and Individually Ventilated Cages (IVC) may affect exploratory and anxiety-related behaviors in mice. Subjects were 8 week-old male C57Bl/6 J mice (n = 36) divided into three groups, OTC, IVC and MFVC groups, respectively. The experimental procedure consisted of two behavioral tests: the open field and the elevated plus maze test. Although there were no differences in the open field test, the results from the elevated plus maze showed that animals housed in the MFVCs exhibited increased exploratory and less anxiety-like behavior. It is concluded that the different caging systems may have an impact on the outcome of behavioral tests used to assess exploratory and anxiety like behavior in mice. Therefore, it is essential to take into consideration housing conditions when reporting, analyzing, and/or systematically reviewing the results of behavioral testing in mice. [ABSTRACT FROM AUTHOR]

Abstract: Environmental enrichment (EE) has been largely used to investigate behavioral modifications and neuroplasticity in the adult brain both in normal and pathological conditions. The interaction between individual behavioral traits with EE responsiveness has not been investigated within the same strain. By using two extremes of CF1 mice that differ by their exploratory behavior in the Open Field (OF) task (Kazlauckas V, 2005), denominated as Low (LE) and High (HE) Exploratory Mice, the present study evaluated if EE during adulthood could modify the putative differences between LE and HE mice on exploratory behavior, memory performance and hippocampal BDNF levels. To this end, we investigated the effect of adult LE and HE mice after 2months of enriched or standard housing conditions on the open field, on novel object recognition, on the inhibitory avoidance task and on hippocampal BDNF immunocontent. LE showed low exploratory behavior, less retention in the inhibitory avoidance and lower hippocampal BDNF levels. EE enhanced exploratory behavior, memory performance and hippocampal BDNF levels both in LE and HE mice. Importantly, the general profile of LE mice submitted to EE was similar to HE mice housed in standard conditions. These results show that internalized behavior of LE mice can be significantly modified by exposure to an enriched environment even during adulthood. These observations may contribute to investigate biological mechanisms and therapeutical interventions for individuals with internalized psychiatric disorders. [Copyright &y& Elsevier]

Abstract: The motor activity of mice in tests of “exploration” is organized. Mice establish home bases, operationally defined as places where they spend long periods of time, near physical objects and nesting material from which they make excursions. This organization raises the question of the extent to which mouse motoric activity is modulated by innate predispositions versus environmental influences. Here the influence of contextual cues (visual and tactile) on the motor activity of C57BL/6 mice was examined: (1) on an open field that had no walls, a partial wall, or a complete wall, (2) in the presence of distinct visual cues, room cues, or in the absence of visual cues (infrared light), and (3) in the presence of configurations of visual and tactile cues. Mice were generally less active in the presence of salient cues and formed home bases near those cues. In addition, movement speed, path distribution, and the number and length of stops were modulated by contextual cues. With repeated tests, mice favored tactile cues over visual cues as their home base locations. Although responses to cues were robust over test days, conditioning to context was generally weak. That the exploratory behavior of mice is affected by experience and context provides insights into performance variability and may prove useful in investigating the genetic and neural influences on mouse behavior. Although rats are known to emit ultrasonic vocalizations (USVs), it remains unclear whether these calls serve an auditory communication purpose. For USVs to be part of communication, the vocal signals will need to be a transfer of information between two or more conspecifics, and with the possibility to induce changes in the behavior of the recipient. Therefore, the aim of our study was to investigate the role of USVs in adult rats' social and non-social investigation strategies when introduced into a large novel environment with unfamiliar conspecifics. We quantified a wide range of social and non-social behaviors in the seminatural environment, which could be affected by subtle signals, including USVs. We found that during the first hour in the seminatural environment the ability to vocalize did not affect how quickly adult rats met each other, their overall social investigation behavior, their passive social behavior nor their aggressive behavior. Furthermore, the non-social exploratory behaviors and behaviors reflecting anxiety/stress-like states were also unaffected. These results demonstrated that a disability to vocalize did not result in significant disadvantages (or changes) compared to intact conspecifics regarding social and non-social behaviors. • A modified open-field test can assess affective vocal communication in rats. • Male aversive ultrasonic vocalizations evoke defensive-like behaviors in receivers. • Female appetitive ultrasonic vocalizations evoke approach-like behaviors in males. Juvenile and adult rats emit two affectively different types of ultrasonic vocalizations (USVs), namely aversive 22-kHz and appetitive 50-kHz USVs. Aversive 22-kHz USVs are considered to be alarm calls that communicate negative affective states to conspecific receivers. Although the alarming effects of playback of 22-kHz USVs were reported recently, behavioral data showing those effects are still not abundant. Appetitive 50-kHz USVs are considered to communicate positive affective states to conspecific receivers, to pace and coordinate social behavior. In line with this, playback of 50-kHz USVs has been found to initiate behavioral activation and induce approach behavior in receiver rats. However, most of these playback studies have used male 50-kHz USVs; thus, it seems to remain unclear whether female 50-kHz USVs exert a similar social attractant effect on male rats. To investigate these issues, we performed modified open-field tests, during which USVs were continuously presented for 15 min to male receivers. In these tests, if negative affective changes are evoked in subject rats, the time spent in the open arena decreases, while the time spent on defensive behaviors increases. In contrast, when positive affective changes are evoked, the opposite phenomenon is observed. Playback of male aversive 22-kHz USVs induced anxiety-related defensive responses in receivers. However, playback of female appetitive frequency-modulated (FM) 50-kHz USVs increased opposite, appetitive pattern of exploratory behavior with increased exploration. The results indicate that playback of male aversive 22-kHz and female appetitive 50-kHz USVs might induce behavioral responses probably associated with negative and positive affective states in male rats, respectively, suggesting the validity of rat USVs as an animal model of vocal communication of emotion. Small-animal models are the most widely used preclinical model for studying the etiology, pathology and treatment of diabetes, prediabetes and diabetic comorbidities. Diabetic patients are burdened with higher rates of depression, anxiety and cognitive decline due to inadequate control of blood glucose levels, vascular damage and aberrant CNS insulin signaling. The C57BL/6J model is amongst the most widely used mouse model due to its susceptibility to diet-induced obesity (DIO). This strain has also been well-characterized in behavioral research studies. However the C57BL/6J model has a number of limitations: [1] overt fasting hyperglycemia can only be induced by dietary manipulation and/or chemical ablation of the pancreatic beta cells. [2] There is heterogeneity in the obesogenic response to hypercaloric feeding as well as sex-dependent differences, with males being more responsive. The KK inbred strain has been used to study aspects of the metabolic syndrome and prediabetes due to inherent glucose intolerance, hyperinsulinemia and insulin resistance. However KK/HlJ mice are less well-characterized and there have been fewer behavioral studies reported. The aim of this study was to examine differences in male and female glucocentric parameters between KK/HlJ and C57BL/6J mice, and to compare their performance in a variety of standard behavioral tests relating to general, anxiogenic and cognitive paradigms. Strain differences in male and female KK/HlJ and C57BL/6J mouse adiposity, glucose and insulin parameters were studied together with group differences in standard Open Field, Object Recognition, Elevated Plus Maze, Light-Dark Transition, Porsolt test, Marble Burying, Social Recognition and Morris Water Maze tests. Correlations between behavioral variables were analyzed. In addition to being uniformly larger, hyperinsulinemic and more insulin intolerant than C57BL/6J mice, we observed marked strain and sex-differences in KK/HlJ behavior. KK/HlJ mice exhibited less locomotor and vertical exploratory behavior in comparison to C57BL/6J, whereas object exploration and novel object discrimination were superior in KK/HlJ mice.
