Direct and non-destructive measurements of plant-water relations of plants exhibiting the Crassulacean acid metabolism (CAM) photosynthetic pathway are seldom addressed, with most findings inferred from gas exchange measurements. The main focus of this paper was to study how the water exchange between the chlorenchyma and the hydrenchyma depends on and follows the CAM photosynthetic diel pattern using non-invasive and continuous methods. Gas exchange and leaf patch clamp pressure probe (LPCP) measurements were performed on Aloe vera (L.) Burm f., a CAM species, and compared to measurements on banana (Musa acuminata Colla), a C3 species. The LPCP output pressure, Pp, of Aloe vera plants follows its diel CAM photosynthetic cycle, reversed to that observed in banana and other C3 species. The four phases of CAM photosynthesis can also be identified in the diel LPCP output pressure, Pp, cycle. The Pp values in Aloe vera are determined by the hydrenchyma turgor pressure, with both parameters being reversely related. A non-invasive and continuous assessment of the water exchange between the chlorenchyma and the hydrenchyma in CAM plants, namely, by following the changes in the hydrenchyma turgor pressure, is presented. However, showing once more how the LPCP output pressure, Pp, depends on the leaf structure, such an approach can be used to study plant-water relations in other CAM species with a leaf structure similar to Aloe vera, with the hydrenchyma composing most of the leaf volume. Allopolyploidy is a common speciation mechanism in plants; however, its physiological and ecological consequences in niche partitioning have been scarcely studied. In this sense, leaf traits are good proxies to study the adaptive capacity of allopolyploids and diploid parents to their respective environmental conditions. In the present work, leaf water relations (assessed through pressure–volume curves) and structural and anatomical traits of the allotetraploid fern Oeosporangium tinaei and its diploid parents, Oeosporangium hispanicum and Oeosporangium pteridioides, were studied under controlled conditions in response to a water stress (WS) cycle. O. hispanicum showed the lowest osmotic potential at turgor loss point (πtlp) and leaf capacitance, together with higher leaf mass per area (LMA), leaf thickness (LT), leaf density (LD), and leaf dry matter content (LDMC), whereas O. pteridioides presented the opposite set of traits (high πtlp and capacitance, and low LMA, LT, LD, and LDMC). O. tinaei showed an intermediate position for most of the studied traits. The responsiveness (osmotic and elastic adjustments) to WS was low, although most of the traits explained the segregation of the three species across a range of drought tolerance according to the rank: O. hispanicum > O. tinaei > O. pteridioides. Previous studies have reported correlation of leaf hydraulic vulnerability with pressure–volume parameters related to cell turgor. This link has been explained on the basis of the effects of turgor on connectivity among cells and tissue structural integrity, which affect leaf water transport. In this study, we tested the hypothesis that osmotic adjustment to water stress would shift the leaf vulnerability curve toward more negative water potential (Ψleaf) by increasing turgor at low Ψleaf. We measured leaf hydraulic conductance (Kleaf), Kleaf vulnerability [50 and 80% loss of Kleaf (P50 and P80); |Ψleaf| at 50 and 80% loss of Kleaf, respectively), bulk leaf water relations, leaf gas exchange and sap flow in two Vitis vinifera cultivars (Tempranillo and Grenache), under two water treatments. We found that P50, P80 and maximum Kleaf decreased seasonally by more than 20% in both cultivars and watering treatments. However, Kleaf at −2 MPa increased threefold, while osmotic potential at full turgor and turgor loss point decreased. Our results indicate that leaf resistance to hydraulic dysfunction is seasonally plastic, and this plasticity may be mediated by osmotic adjustment. [ABSTRACT FROM AUTHOR]

This paper concerns tolerance to 50–200 mM NaCl of submerged rice (Oryza sativa cv. Amaroo) during germination and the first 138–186 h of development in aerated solution. Rice was able to germinate and the seedlings even tolerated exposure to 200 mM NaCl, albeit with severe growth restrictions. After return to 0.3 mM NaCl, growth increased, indicating that even at 200 mM NaCl there was no irreparable injury. Osmotic adjustment was achieved by using Na+ and Cl– as the major osmotica. At 200 mM NaCl commenced at sowing, the shoot Na+ and Cl– concentrations between 50–110 h were about 210 and 260 mM, respectively, i.e. above the external concentration. Thus, there was a high tissue tolerance to NaCl. The internal concentrations declined subsequently, concurrent with a decline in growth. At 50–200 mM NaCl, the contributions from ions to πsap were 81–92% in roots and 62–74% in shoots. The assessed turgor pressures at 200 mM NaCl were 0.33 MPa in shoots and 0.15 MPa in roots, compared to 0.62 and 0.43 MPa at 0.3 mM NaCl. In the General Discussion section, we compare the different responses of submerged seedlings to the responses of transpiring rice plants, reported in the literature, and suggest that the submerged system is useful to evaluate effects of NaCl on turgor pressure and particularly to establish whether there are specific effects of Na+ and Cl– in tissues. Relative changes in cell turgor of leaves of well-watered tomato plants were evaluated using the leaf patch clamp pressure probe (LPCP) under dynamic greenhouse climate conditions. LPCP changes, a measure for relative changes in cell turgor, were monitored at three different heights of transpiring and non-transpiring leaves of tomato plants on sunny and cloudy days simultaneously with whole plant water uptake. Clear diel patterns were observed for relative changes of cell turgor of both transpiring and non-transpiring leaves, which were stronger on sunny days than on cloudy days. A clear effect of canopy height was also observed. Non-transpiring leaves showed relative changes in cell turgor that closely followed plant water uptake throughout the day. However, in the afternoon the relative changes of cell turgor of the transpiring leaves displayed a delayed response in comparison to plant water uptake. Subsequent recovery of cell turgor loss of transpiring leaves during the following night appeared insufficient, as the pre-dawn turgescent state similar to the previous night was not attained. In this article, we measured the relative growth rate (RGR) of leaves of Robinia pseudoacacia seedlings under well-watered and water-stressed conditions (mid-day Ψw = leaf water potential estimated with a pressure bomb of −0.48 and −0.98 MPa, respectively). Pressure-volume (PV) curves were done on growing leaves at 25, 50 and 95% of the mature size (growth stage) in order to compute solute potential (Ψ) and turgor pressure (ΨP) as a function of Ψw. The PV curves and diurnal measurements of Ψw and RGR allowed us to evaluate the parameters (cell wall extensibility m and growth turgor threshold Y) of the Lockhart equation, RGR = m(ΨP− Y), at each growth stage. Our data showed that m and Y did change with leaf age, but the changes were slow enough to evaluate m and Y on any given day. We believe this is the first study to provide evidence that the Lockhart equation adequately quantifies leaf growth of trees over a range of time domains. The value of m linearly declined and Y linearly increased with growth stage. Also, mild drought stress caused a decline in m and increase in Y relative to controls. Although water stress caused an osmotic adjustment which, in turn, increased ΨP in stressed plants relative to controls, the RGR and final leaf sizes were reduced in water-stressed plants because of the impact of water stress on decreased m and increased Y. Type-II, singlet oxygen-mediated photosensitized damage has already been shown to occur in epicotyls of dark-germinated pea ( Pisum sativum L.) seedlings upon illumination, resulting in fast turgor loss and wilting. In this study we show evidence that the palette of reactive oxygen species (ROS) is more complex. Hydrogen peroxide, superoxide and hydroxyl radicals are also formed, suggesting the occurrence of type-I reactions as well. Moreover, hydrogen peroxide injection into the epicotyls in the dark was able to provoke wilting directly. Formation of hydroxyl radicals could also be triggered by the addition of hydrogen peroxide in the dark, preferentially in the mid-sections where wilting occurs, showing that potential mediators of a Fenton reaction are present in the epicotyls, but unevenly distributed. Localization of light-inducible ROS formation fully (hydrogen peroxide) or partially (superoxide radicals) overlaps with the distribution of monomer protochlorophyllide complexes, showing that these pigment forms are capable of provoking both type-I and type-II reactions. Low concentrations of some trace metals markedly reduce root elongation rate and cause ruptures to root rhizodermal and outer cortical cells in the elongation zone. The interactions between the trace metals and plant components responsible for these effects are not well understood but may be linked to changes in water uptake, cell turgor and cell wall extensibility. An experiment was conducted to investigate the effects of Al, La, Cu, Gd, Sc and Ru on the saturated hydraulic conductivity of bacterial cellulose (BC)–pectin composites, used as plant cell wall analogs. Hydraulic conductivity was reduced to ≈30% of the initial flow rate by 39 µ M Al and 0.6 µ M Cu, ≈40% by 4.6 µ M La, 3 µ M Sc and 4.4 µ M Ru and ≈55% by 3.4 µ M Gd. Scanning electron microscopy (SEM) revealed changes in the ultrastructure of the composites. The results suggest that trace metal binding decreases the hydraulic conductivity through changes in pectin porosity. The experiment illustrates the importance of metal interactions with pectin, and the implications of such an interaction in plant metal toxicity and in normal cell wall processes. Potassium (K+) is the most abundant ion in the plant cell and is required for a wide array of functions, ranging from the maintenance of electrical potential gradients across cell membranes, to the generation of turgor, to the activation of numerous enzymes. The majority of these functions depend more or less directly upon the activities and regulation of membrane-bound K+ transport proteins, operating over a wide range of K+ concentrations. Here, we review the physiological aspects of potassium transport systems in the plasma membrane, re-examining fundamental problems in the field such as the distinctions between high- and low-affinity transport systems, the interactions between K+ and other ions such as NH4+ and Na+, the regulation of cellular K+ pools, the generation of electrical potentials and the problems involved in measurement of unidirectional K+ fluxes. We place these discussions in the context of recent discoveries in the molecular biology of K+ acquisition and produce an overview of gene families encoding K+ transporters. [ABSTRACT FROM AUTHOR]

Water relation parameters including elastic modulus (ε), half-times of water exchange (Tw1/2), hydraulic conductivity and turgor pressure (P) were measured in individual root cortical and cotyledon midrib cells in intact figleaf gourd (Cucurbita ficifolia) seedlings, using a cell pressure probe. Transpiration rates (E) of cotyledons were also measured using a steady-state porometer. The seedlings were exposed to low ambient (approximately 10 μmol m−2 s−1) or high supplemental irradiance (approximately 300 μmol m−2 s−1 PPF density) at low (8°C) or warm (22°C) root temperatures. When exposed to low irradiance, all the water relation parameters of cortical cells remained similar at both root temperatures. The exposure of cotyledons to supplemental light at warm root temperatures, however, resulted in a two- to three-fold increase in Tw1/2 values accompanied with the reduced hydraulic conductivity in both root cortical (Lp) and cotyledon midrib cells (Lpc). Low root temperature (LRT) further reduced Lpc and E, whether it was measured under low or high irradiance levels. The reductions of Lp as the result of respective light and LRT treatments were prevented by the application of 1 μM ABA. Midrib cells required higher concentrations of ABA (2 μM) in order to prevent the reduction in Lpc. When the exposure of cotyledons to light was accompanied by LRT, however, ABA proved ineffective in reversing the inhibition of Lp. LRT combined with high irradiance triggered a drastic 10-fold reduction in water permeability of cortical and midrib cells and increased ε and Tw1/2 values. Measurement of E indicated that the increased water demand by the transpiring plants was fulfilled by an increase in the apoplastic pathway as principal water flow route. Wheat (Triticum aestivum L.) seedlings of a drought-resistant cv. C306 were subjected to severe water deficit directly or through stress cycles of increasing intensity with intermittent recovery periods (drought acclimation). The antioxidant defense in terms of redox metabolites and enzymes in leaf cells, chloroplasts, and mitochondria was examined in relation to ROS-induced membrane damage. Drought-acclimated seedlings modulated growth by maintaining favorable turgor potential and RWC and were able to limit H2O2 accumulation and membrane damage as compared with non-acclimated plants during severe water stress conditions. This was due to systematic upregulation of H2O2-metabolizing enzymes especially ascorbate peroxidase (APX, EC 1.11.1.11) and by maintaining ascorbate–glutathione redox pool in acclimated plants. By contrast, failure in the induction of APX and ascorbate–glutathione cycle enzymes makes the chloroplast susceptible to oxidative stress in non-acclimated plants. Non-acclimated plants protected the leaf mitochondria from oxidative stress by upregulating superoxide dismutase (SOD, EC 1.15.1.1), APX, and glutathione reductase (GR, EC 1.6.4.2) activities. Rewatering led to rapid enhancement in all the antioxidant defense components in non-acclimated plants, which suggested that the excess levels of H2O2 during severe water stress conditions might have inhibited or downregulated the antioxidant enzymes. Hence, drought acclimation conferred enhanced oxidative stress tolerance by well-co-ordinated induction of antioxidant defense both at the chloroplast and at the mitochondrial level. Pressure–volume (P–V) curves for leaves or terminal shoots summarize leaf-level responses to increasing water deficit. P–V curve traits and field-measured shoot xylem pressures were characterized across 62 species from four sites differing in rainfall and soil phosphorus. Within-species variation in the measured traits was small relative to differences among species and between environments. P–V curve traits tended to differ with site rainfall but not with soil phosphorus. Turgor loss points (TLPs) varied widely and averaged more negative in species from lower-rainfall sites. Differences between species in TLP were driven mainly by differences in solute potential, rather than by differences in cell wall elasticity. Among species at individual sites, species seemed to vary in leaf-response strategy reflected in TLP independently from water-uptake strategy reflected in predawn xylem pressures and in xylem pressure drop from predawn to midday. The interdependence between changes in growth and water relations after waterlog-ging was investigated by recording simultaneously growth, transpiration, water potential, turgor, leaf diffusion resistance and abscisic acid content in Phaseolus vulgaris L. cv. bruine Noord-Hollandse. Growth was inhibited immediately after flooding, whereas transpiration decreased gradually to a low level in about three days. The first two days after flooding a small increase in abscisic acid content in the leaves was observed which was accompanied by an increase in diffusion resistance. The increase in abscisic acid content could result from an inhibited export from the leaves. After the first two days a decrease in water potential and turgor was accompanied by a drastic increase in both abscisic acid content and diffusion resistance. This large increase in abscisic acid content occurred before the turgor had reached its minimum value. The change in diffusion resistance kept showing a lag of about one day with the change in abscisic acid content. The possibility is discussed that besides abscisic acid also its metabolite phaseic acid is involved in stomatal closure. After the formation of adventitious roots on the hypocotyl, abscisic acid level, diffusion resistance, water potential and turgor returned to the control values. Transpiration showed a slow recovery from the sixth day after flooding, whereas growth was inhibited for at least nine days. The relationship between the inhibiting effect of Ca2+ and of low turgor pressure on K+ release from fresh‐cut discs of carrot (Daucus carota var. Nantes) storage tissue was studied. A range of Ca2+ concentrations in the tissue was obtained by adding 0.5 mM EDTA or CaSO4 at different concentrations to the medium. Calcium inhibited K+ release in fully turgid cells (2.5 μmol K+ g-1 h-1 in 0.5 mM EDTA vs 0.4 μmol K+ g-1 h-1 in 10 mM CaSO4). Less turgid cells, obtained by equilibration with 0.2 M mannitol, released K+ at only 30% of the rate of the turgid cells, yet the pattern of K+ release as a function of Ca2+ level was similar in both turgid and non‐turgid cells. Removal of calcium by EDTA occasionally injured cell membranes in the fully turgid discs but never in the less turgid ones. In view of the additive effect of Ca2+ and low turgor on K+ release regardless of the treatment order, it is suggested that the two factors exert their effect on membrane permeability independently of each other. The article focuses on the regulation and unloading in long distance transport systems. Long distance translocation of assimilates along sieve elements is believed by many investigators to be driven by a turgor pressure gradient existing between source and sink tissues. In soybean, for instance, the sucrose concentration in the sieve element-companion cell complex of the source leaf is double that in the roots. Thus the calculated maximum turgor pressure difference between source and sink is 410 kPa, which is sufficient to drive a pressure flow at an observed velocity.

The response to salt spray and soil salinity of two sand dune strandline species (Cakile maritima Scop. and Salsola kali L.) and two salt marsh strand‐line species (Atriplex hastata L. and A. littoralis L.) was compared in sand‐compost cultures. The growth of the salt‐marsh species remained unaffected, while the growth of the sand dune species Cakile maritima was strongly reduced by NaCl (150 and 300 mM) absorbed via the root system. All four species were resistant to airborne salinity, and under conditions of low soil fertility, salt spray increased the dry matter production, especially of the sand dune species. Mineral analysis revealed foliar uptake of Na, K, Cl, Ca and Mg. Na and Cl ions absorbed from seawater droplets induced succulence. Both salt spray and soil salt increased the methylated quaternary ammonium compound content in the shoot tissue. Under non‐saline conditions a considerable amount of these osmotic solutes was still present, while turgor pressure potential in these plants was rather low. The relation between salt, compatible osmotic solutes, turgor pressure potential and growth is discussed. Next to the major constituents of seawater, Na and Cl, especially magnesium and to a lesser extent, calcium, accumulated in the shoot tissue. Based on the positive growth response of the sand dune species to airborne salt, they should be termed ‘aerohalophytes’, whereas ‘soil halophytes’ should be used when referring to the Atriplex species, which are more specifically adapted to the increased salinity of salt marsh soils. The impairment of the turgor operated modulation of stomatal movement and changes in photosynthetic capacity were studied using two maize (Zea mays L.) varieties of different chilling sensitivity. In the tolerant variety stomatal opening exhibited characteristic transients, while in the more susceptible strain chilling induced a monotonous increase of stomatal apertures. Photosynthetic capacity of the leaves (expressed in terms of 14CO2 incorporation at 25°C) declined as a result of chilling regardless of stomatal opening. Lettuce seeds cv. Noran germinate at 23°C in light as well as in darkness. However dormancy can be induced either by a long exposure (24 h) to far-red radiation or by an exposure of 48-72 h to a temperature of 37°C. The difference in response of these two types of dormant seeds to conditions inducing germination indicate that in both types Pfr, is inactivated, but that a dark process required for immediate action of Pfr does not proceed at 37°C as it does during far-red radiation. The effect of increasing external osmotic pressure on. potassium fluxes in aged and fresh-cut discs of Daucus carota L. storage tissue was investigated. An increase of the external osmotic pressure by 5 bars of mannitol solution increased the rate of K+ net uptake of aged discs to 180% of their control rate. At 3°C and in 0.1 mM azide, in which a net efflux of potassium was observed, the mannitol treatment caused a reduction in the net efflux, In fresh-cut discs, in which the capability of net influx was rather low and a substantial net release of potassium was noted, the increase in the external osmotic pressure by mannitol caused a 70% inhibition in the net efflux. This effect was also observed at 3°C. Measurements of separate fluxes confirmed the assumption that the mannitol treatment brought about two distinct effects on K+ fluxes: (a) raised the metabolically-dependent influx and (b) lowered the membrane permeability-dependent efflux. When a permeating solute (ethylene glycol) was used instead of mannitol, no effect on K+ flux was detectable. The water content‐water potential relation in stressed and unstressed cassava (Man‐ihot species) was examined to ascertain (i) the magnitude of osmotic adjustment in response to water stress and (ii) the mechanisms of such adjustments. Water stress resulted in a displacement of the water content‐potential relation such that at any leaf water potential the water content was higher in the stressed plants. The osmotic potentials of turgid leaves (100% relative water content) were ‐0.97 and ‐1.00 MPa in the unstressed cultivars CMC 9 and MCOL 113 respectively. In the stressed plants, the values were‐1.13 MPa (CMC 9) and‐1.14 MPa (MCOL 113). The 0.14 to 0.16 MPa osmotic potential difference between the stressed and unstressed plants suggests that a stress‐induced osmotic adjustment occurred in both cultivars. The biiSk volumetric elastic moduli at turgor pressures above 0.10 MPa were 9.84 MPa (CMC 9) and 13.58 MPa (MCOL 113) in the unstressed plants. Tbe higher values found in the stressed plants, 14.56 MPa in CMC 9 and 16.91 MPa in MCOL 113, suggest a stress‐induced decrease in cell wall elasticity. Hence, the observed shift in the wafer content‐potential relations in the cassava involved both an osmotic adjustment and a decrease in cell wall elasticity. Measures of leaf tissue elasticity were determined by analyzing the turgor pressure-water content relation developed from pressure-volume experiments in four naturally occurring forest tree species. The relation between the bulk tissue elastic modulus and tissue turgor pressure varied by spades and varied with state of leaf maturation through the growing season. Although the values of the elastic modulus reported here agree in magnitude with values reported in general for higher plant tissue, the functional dependency of the apparent elasticity on tissue water status was more complex in this study than can be accounted for by current models. Plant growth is reduced under saline conditions even when turgor in mature leaves is maintained by osmotic adjustment. The objective of this study was to determine if young leaves from salt-affected plants were also osmotically adjusted. Pepper plants (Capsicum annuum L. cv. California Wonder) were grown in several levels of solution osmotic potential and various components of the plants' water relations were measured to determine if young, rapidly growing leaves could accumulate solutes rapidly enough to maintain turgor for normal cell enlargement. Psychrometric measurements indicated that osmotic adjustment is similar for both young and mature leaves although osmotic potential is slightly lower for young leaves. Total water potential is also lower for young leaves, particularly at dawn for the saline treatments. The result is reduced turgor under saline conditions at dawn for young but not mature leaves. This reduced turgor at dawn, and presumably low night value, is possibly a cause of reduced growth under saline conditions. No differences in leaf turgor occur at midday. Porometer measurements indicated that young leaves at a given salinity level have a higher stomatal conductance than mature leaves, regardless of the time of day. Leaf water (ψ) and solute (ψs) potential were measured in field sorghum and maize under well irrigated (I) and dryland (NI) conditions throughout a season. Despite decreases in ψ due to slow soil water depletion and to apparent increases in liquid phase plant resistance, midday leaf turgor (ψp) in the NI sorghum was maintained at similar levels as in the I treatment throughout the season due to concomitant decreases in ψs. Osmotic adjustment was also observed in maize, although ψp was significantly lower in the NI treatment as compared to I during the final stages of grain filling. A seasonal shift in the ψ vs. relative water content relation of NI sorghum leaves was observed, more water being retained by the older leaf at any particular ψ. The major factor for turgor maintenance was a net increase in solutes per unit of tissue. The role played by increases in the proportion of tissue volume occupied by cell wall was also evaluated. No stomatal closure due to water stress was found in NI sorghum even though leaf ψ reached -20 bars late in the season. Experiments were designed to test the hypothesis that the internal water relations of leaves are altered when cotton plants (Gossypium hirsutum L.‘Acala SJ‐2′) are conditioned by several cycles of water stress. Preliminary experiments suggested that plants so conditioned are less sensitive to water deficits and that the change might be partly explained by an accumulation of solutes or by structural alterations attendant on development under conditions of water stress. Leaves of preconditioned plants maintained turgor to lower values of water potential than did leaves of well‐watered plants. Accompanying this change was a lower osmotic potential at any given leaf water content in preconditioned plants. Tissue analysis of several osmotically active solutes indicated that soluble sugars and malate accumulate to about the same levels (dry‐weight basis) in both conditioned and unconditioned plants exposed to stress. These accumulations could not account for the turgor change. Analysis of the data on relative water content indicated that the leaves of conditioned plants had less water per unit dry weight than did leaves of controls. This change accounts for a substantial fraction of the difference between the osmotic potential of conditioned and control plants. The results of a simple model suggest that structural changes may play a significant role in explaining differences in the responses of conditioned and control plants to water stress. The rate of leaf elongation in radiata pine (Pinus radiata) and wheat seedlings was closely related to the osmotic potential of the rooting solution. Sudden stress application and removal treatments caused immediate changes in the leaf elongation rate and a new steady-state rate independent of the old was quickly established. The osmotic potential in the active elongation zones of a leaf differed from that of the remainder of the leaf and was considered more appropriate for deriving turgor pressure to be related to leaf elongation. Shifts in elongation rate were associated with corresponding shifts in total leaf water potential in both species and with corresponding shifts in turgor pressure in wheat. There was a linear correlation between the pitch of the helical thickenings in the protoxylem of radiata pine needles and their elongation rate. Over the past twenty years many studies have been undertaken to elucidate the regulation of seed germination. Abscisic acid (ABA) and the gibberellins (GAs) are the hormones proposed to control this process, the first by inhibiting and the second by inducing germination. It has been proposed that a high water potential increases the growth potential of the embryo, presumably permitting the production or activation by GA of the cell wall hydrolases and thus decreasing the yield threshold of the endosperm close to the radicle tip. A low water potential, e.g., imbibition in an osmoticum, imposes a stress on cell metabolism, by reducing the turgor of the radicle cells, and there is a decrease in growth potential. Exogenous ABA also causes a decline in growth potential of the radicle; however, the actions of low water potential in preventing germination are not mediated through an increase in ABA in the seeds. In the present paper an attempt is made to asses the role of ABA and polyethylene glycol (PEG) in the germination of chick-pea (Cicer arietinum L.) seeds. The endogenous ABA of chick-pea seeds was purified by reversed-phase HPLC and quantified by GC-ECD. The variations in the ABA levels in the embryonic axes and the cotyledons were studied during 120 h of imbibition. The highest ABA level in the embryonic axes was found at 18 h, coinciding with an increase in fresh weight and a high germination percentage. ABA was not detected in the cotyledons during incubation which probably indicates that the hormone is more involved in the active growth of the embryonic axes itself than in the mobilization process of the reserves. When seeds were treated with different PEG-cycles, PEG delayed germination, reduced the fresh weight of embryonic axes, and retarded the onset of ABA synthesis. It is concluded that endogenous ABA is related to the onset of germination and the growth of the embryonic axis. In addition, there is no correlation among the different PEG-cycles and the levels of ABA and germination. We highlight the novel observation that the responses of maize leaf growth to abscisic acid (ABA) signals can be amplified both by mild water deficits and by nutrient stress. Under our experimental conditions these stresses alone had no effect on leaf growth rate. In most cases leaf growth responses were not attributable to changes in the turgors of growing cells, focusing attention on a regulatory role for cell wall biochemistry. Roles for xyloglucan endotransglycosylase (XET), expansins and peroxidases are discussed. An effect of drought on the activity of expansins seems particularly attractive if xylem sap pH is considered as a chemical signal. In early studies, negative turgor pressure was obtained by indirect methods and was mainly related to inaccurate measurements of osmotic and/or water potential. The uncertainties in the measurements made any discussion of the underlying reasons and phenomena premature and risky, perhaps even useless. Yet, the idea of relating negative turgor to the strength of small-sized cells with thick walls, 10 resist collapse when subjected to water deficit, seemed attractive. Negative turgor pressures are difficult to detect in higher plants, but they are theoretically possible. Regarding their origin, a hypothesis is discussed here, that the negative turgor detected in leaves might indicate a reversal of the water flow that has occurred in roots, under conditions of severe and prolonged drought. Movements of ions are considered to be governed by the electroneutrality rule. Therefore, a cation moving across the cell membrane into the cell either passively or actively should move together with its counterion, an anion, in equal amounts of charge or in exchange for another cation inside the cell. This means that the net influx of the cation in question should be affected by the permeability of its counterion and/or another cation inside the cell. To examine osmotic and ionic regulation in Chara cells, cell fragments of Chara having a lower osmotic pressure than normal (L‐cell fragments) were prepared. The L‐cell fragments were individually put into various dilute electrolyte solutions and their osmotic potentials were measured with a turgor balance. Concentrations of K+, Na+, Ca2+, Mg2+, Cl−, NO−3. and SO2−4. in the external electrolyte solutions in which L‐cells had been incubated were also analysed by ion chromatography. The results showed that in 0.5 mM KCL + 0.1 mM CaCl2 solution, Chara L‐cell fragments absorbed K+ and Cl− to maintain electroneutrality and then regained their osmotic potential very rapidly. When the anion was Cl, the cation absorbed at the highest rate was K+ On the other hand, when the cation was K, the anion absorbed at the highest rate was Cl, Other ions Ca2+, SO2−4 and NO−3 showed much less permeability than K+ and Cl −for the Chara plasma membrane. The conclusion from these findings was that due to the constraint of electroneutral transport, the uptake rate of a salt into L‐cells is limited by the permeability of the least permeable ion. A reduction in cell turgor has been shown to stimulate sugar uptake in several plant sink tissues and it may regulate the import of assimilate into the sink apoplast, as well as maintain cell turgor. To determine whether cell turgor influences sugar uptake by strawberry (Fragaria × ananassa Duch. cv. Brighton) fruit cortex tissue, disks were cut from greenhouse-grown primary fruit at the green-white stage of development and placed in buffered incubation solutions containing either mannitol or ethylene glycol as an osmoticum. Cell turgor of fruit disks was calculated from the difference between the water potential of bathing solution and tissue solute potential after incubation at various osmolarities. Cell turgor increased when tissue disks were placed into mannitol incubation solutions more dilute than the water potential of fresh tissue (about 415 mOsmol kg-1). The rate of uptake of [14C]-sucrose or [14C]-glucose decreased as osmolarity of the incubation solution increased, i.e. as cell turgor declined. Cell turgor and the rate of [14C]-sucrose uptake were unaffected when rapidly permeating ethylene glycol was used as an osmoticum. A decrease in cell turgor reduced both the Vmax of the saturable (carrier mediated) kinetic component of sucrose uptake, and the slope of the linear (diffusional) component. The sulfhydryl binding reagent p-chloromercuibenzenesulfonic acid, an inhibitor of the plasma membrane sucrose carrier, strongly inhibited only the saturable component of sucrose uptake. Increased uptake of the nonmetabolizable sugar, O-methyl-glucose, at high turgor was similar to that of glucose, indicating that carrier activity was influenced by cell turgor, not cell metabolism. Turgor did not influence efflux of [14C]-sucrose from disks and had no effect on cell viability. The effects of auxin and osmotic stress on elongation growth of maize (Zea mays L.) coleoptile segments are accompanied by characteristic changes in the extensibility of the growth-limiting cell walls. At full turgor auxin causes growth by an increase in wall extensibility (wall loosening). Growth can be stopped by an osmotically produced step-down in turgor of 0.45 MPa. Under these conditions auxin causes the accumulation of a potential for future wall extension which is released after restoration of full turgor. Turgor reduction causes a reversible decrease in wall extensibility (wall stiffening) both in the presence and absence of auxin. These changes in vivo are correlated with corresponding changes in the theological properties of the cell walls in vitro which can be traced back to specific modifications in the shape of the hysteretic stress-strain relationship. The longitudinally load-bearing walls of the coleoptile demonstrate almost perfect viscoelasticity as documented by a nearly closed hysteresis loop. Auxin-mediated wall loosening causes an increase of loop width and thus affects primarily the amount of hysteresis in the isolated wall. In contrast, turgor reduction by osmotic stress reduces loop length and thus affects primarily the amount of viscoelastic wall extensibility. Pretreatment of segments with anoxia and H2O2 modify the hysteresis loop in agreement with the conclusion that the wall-stiffening reaction visualized under osmotic stress in vivo is an O2-dependent process in which O2 can be substituted by H2O2. Cycloheximide specifically inhibits auxin-mediated wall loosening without affecting wall stiffening, and this is mirrored in specific changes of the hysteresis loop. Corroborating a previous in vivo study (Hohl et al. 1995, Physiol. Plant. 94: 491–498) these results show that cell wall stiffening in vivo can also be demonstrated by theological measurements with the isolated cell wall and that this process can be separated from cell wall loosening by specific changes in the shape of the hysteresis loop. The ripening of discs cut from the pericarp of green tomato (Lycopersicon esculentum Mill.) fruits is inhibited by treatments with GA3 and several divalent cations, including calcium. Normal ripening is marked by an increase in the solubility of wall pectins. Calcium and GA3 alter the pattern of pectin solubility changes. In part this may be because polygalacturonase synthesis and/or secretion to the apoplast is reduced. The impact of divalent cations on ripening-related tissue softening appears to have a nonmetabolic component. Auxin-induced changes of wall-rheological properties during different growth rates of rye coleoptile segments (Secale cereale L.) were investigated. In addition, changes of osmotic concentration and turgor pressure were measured. Decrease of turgor and of osmotic concentration followed a synchronous time course. Auxin-incubated segments exhibited a faster decrease and eventually lower values of both parameters. Creep test extensibility measurements demonstrate that apparent plastic as well as elastic extensibility of distilled-water-incubated segments strongly decreased during 24 h. In auxin-incubated segments apparent plastic as well as elastic extensibilities were strongly increased, even in the absence of growth due to insufficient turgor pressure. The increasing effect of auxin on elastic wall properties is also reflected by an increase in relative reversible length (part of segment length by which segments shrink after freezing/thawing as referred to total length) and a complementary decrease of relative irreversible length (remaining length after turgor elimination as referred to turgid length); again the effects were independent of growth rate and turgor pressure. Cellulose synthesis inhibition of approx. 80% by dichlorobenzonitrile (DCB) had no significant effect either on growth or on wall-rheological properties. Independent of whether the changed rheological wall behaviour of auxin-incubated segments is causally related to the mechanism of auxin-induced wall loosening, it indicates changes of wall polymer properties and/or interactions which are conserved when no actual length increase occurs due to insufficient turgor pressure. The results suggest that IAA-induced wall loosening may be primarily mediated by cell wall changes other than cleavage of covalent, load-bearing bonds as hypothesized in various wall loosening models. Given the close relationship between a plant's growth rate and its pattern of biomass allocation and the effects of abscisic acid (ABA) on biomass allocation, we studied the influence of ABA on biomass allocation and growth rate of wildtype tomato (Lycopersicon esculentum Mill. cv. Moneymaker) plants and their strongly ABA-deficient mutant sitiens. The relative growth rate of sitiens was 22% lower than that of the wildtype, as the result of a decreased specific leaf area. The net assimilation rate and the leaf weight ratio were not affected. The mutant showed a much higher transpiration rate and lower hydraulic conductance of the roots. These two factors resulted in sitiens having a significantly lower leaf water potential and turgor, resulting in reduced leaf expansion and, consequently, a lower specific leaf area relative to the wildtype. Addition of ABA to the sitiens roots resulted in phenotypic reversion to the wildtype. We conclude that the influence of ABA-deficiency on biomass allocation and relative growth rate is the result of altered water relations in the plants, rather than of a direct effect on sink strength of different plant organs. Redland Pioneer) and Vicia faba (cv. Coles Prolific). The osmotic potentials of the seed apoplast were low. As a result, the osmotic gradients to the seed coat symplast were relatively small (i.e. 0.1 to 0.3 MPa). Sap concentrations of sucrose and potassium in the seed apoplast and coat symplast accounted for some 45 to 60% of the osmotic potentials of these compartments. Estimated turnover times of potassium and sucrose in the seed apoplast of < 1 h were some 5 to 13 times faster than the respective turnover times in the coat symplast pools. The small osmotic gradient between the seed apoplast and coat symplast combined with the relatively rapid turnover of solutes in the apoplast pool, confers the potential for a small change in assimilate uptake by the cotyledons to be rapidly translated into an amplified shift in the cell turgor of the seed coat. Observed adjustments in the osmotic potentials of solutions infused between the coat and cotyledons of intact seed were consistent with the in vivo operation of turgor‐dependent unloading of solutes from the coat. Homeostatic regulation of turgor‐dependent unloading was indicated by the maintenance of apoplast osmotic potentials of intact seeds when assimilate balance was manipulated by partial defoliation or elevating pod temperature. In contrast, osmotic potentials of the coat symplast adjusted upward to new steady values over a 2 to 4 h period. The resultant downward shift in coat cell turgor could serve to integrate phloem import into the seed coat with the new rates of efflux to the seed apoplast. To determine how tissue water relations vary and contribute to turgor maintenance in species from contrasting ecological zones, seedlings of jack pine (Pinus banksiana Lamb.), black spruce (Picea mariana [Mill] B.S.P) and flooded gum (Eucalyptus grandis W. Hill ex Maiden) were subjected to an 8 day drought stress by water withholding with and without prior mild water stress conditioning. Jack pine, a deep-rooted species from dry, sandy boreal sites, lost turgor at the lowest relative water content (75–65%) and water potential, and had lowest maximum bulk elastic modulus (εmax of 5.2–5.8 MPa). Although this suggests a high inherent dehydration tolerance, jack pine did not further adjust its elasticity when repeatedly stressed. Black spruce, a shallow-rooted species from predominantly moist sites in the boreal region, lost turgor at intermediate relative water content (86–76%) and water potential, but could adjust its elasticity to maintain turgor in repeatedly stressed tissues. Flooded gum, a deeprooted species from moist, warm temperate-subtropical regions, had a low inherent drought tolerance since it lost turgor at higher relative water content (88–84%) and water potential, but was capable of some adjustment when the stress was repeated. Elastic adjustment (< 3.7 MPa) was more important for turgor maintenance than osmotic adjustment (< 0.13 MPa), which was statistically nonsignificant. Maximum bulk modulus of elasticity, but not osmotic potentials at full turgor, was significantly correlated with the relative water content and water potential at zero turgor in droughted seedlings. These results highlight the importance of tissue shrinkage for dehydration tolerance. Both the inherent capacity for turgor maintenance of a species under drought and its ability to adjust to repeated drought should be considered in genetic selections for drought tolerance. We examined the roles of turgor potential and osmotic adjustment in plant growth by comparing the growth of spring wheat (Triticum aestivum cv. Siete cerrors) and sudangrass (Sorghum vulgare var. Piper) seedlings in response to soil water and temperature stresses. The rates of leaf area expansion, leaf water potential and osmotic potential were measured at combinations of 5 soil water potentials ranging from -0.03 to -0.25 MPa and 6 soil temperatures ranging from 14 to 36°C. Spring wheat exhibited little osmotic adjustment while sudangrass exhibited a high degree of osmotic adjustment. However, the rate of leaf area growth for sudangrass was more sensitive to water stress than that of spring wheat. These results were used to evaluate the relationship between growth and turgor potential. The modified Arrhenius equation based on thermodynamic considerations of the growth process was evaluated. This equation obtains growth rate as a function of activation energy, enthalpy difference between active and inactive states of enzymes, base growth rate and optimum temperature. Analyses indicate that the modified Arrhenius equation is consistent with the Lockhart equation with a metabolically controlled cell wall extensibility. [ABSTRACT FROM AUTHOR]

A relatively drought tolerant cultivar of maize (Zea mays L. cv. Pioneer 3950) and a drought tolerant line of sorghum (Sorghum bicolor [L.] Moench cv. ICSV 112) were grown hydroponically for 11 days. Treatments for non-ionic osmotic and salt stresses were started at the 8th day by addition of polyethylene glycol 6000 and NaCl, respectively, at 200 mOsm equivalent concentrations in the presence or absence of 0.1 µM abscisic acid. Relative growth rate was depressed by both stress factors, more severely for maize than sorghum. Abscisic acid increased the growth rate and reverted the negative effect of NaCl in maize, while sorghum was only slightly affected. In general, sorghum had higher levels of K+ and lower levels of Na+ and the K+/Na+ ratio was further increased by abscisic acid treatment. From the pressure-volume curves, osmotic potential, the water potential at turgor loss point, bulk elastic modulus and the water saturation deficit at initial turgor loss were estimated. Most significantly, sorghum had a higher elastic modulus than maize and it decreased under osmotic treatment, while in maize it increased under NaCl stress. The results suggest that bulk tissue turgor was not limiting growth under these conditions and underscores the possible implications of changes in the elastic condition of the cell walls in stress responses. Osmotic regulation of assimilate efflux from excised coats of developing Vicia faba (cv. Coles Prolific) seed was examined by exposing these to bathing solutions (adjusted to –0. 02 to –0. 75 MPa with sorbitol) introduced into the cavity vacated by the embryo. 14C photosynthate efflux was found to be independent of solution osmotic potentials below –0. 63 MPa. At higher osmotic potentials, efflux was stimulated and exhibited a biphasic response to osmotic potential with apparent saturation being reached at –0. 37 MPa. Efflux could be repeatedly stimulated and slowed by exposing seed coats to solutions of high and low osmotic potentials, respectively. Manipulation of components of tissue water potential, with slowly‐ and rapidly‐permeating osmotica, demonstrated that turgor functioned as the signal regulating 14C photosynthate efflux. Com‐partmental analysis of 14C photosynthate preloaded seed coats was consistent with exchange from 4 kinetically‐distinct compartments. The kinetics of turgor‐dependent efflux exhibited characteristics consistent with the transport mechanism residing in the plasma membranes of the unloading cells. These characteristics included the rapidity (<2 min) of the efflux response to turgor increases, similar rate constants for efflux from the putative turgor‐sensitive and cytoplasmic compartments and the apparent small pool size from which turgor‐dependent efflux could repeatedly occur. In contrast, influx of [14C] sucrose across the plasma and tonoplast membranes was found to be insensitive to turgor. The plasma membrane [14C] sucrose influx was unaffected by p‐chloromercuribenzenesulfonic acid and erythrosin B and exhibited a linear dependence on the external sucrose concentration. This behaviour suggested that influx across the plasma membrane occurs by passive diffusion. Preloading excised seed coats with a range of solutes demonstrated that turgor‐dependent efflux exhibited partial solute selectivity. Based on these findings, it is proposed that turgor controls assimilate exchange from the seed coat by regulating an efflux mechanism located in the plasma membranes of the unloading cells. The dependence of elongation on the osmotic potential of the medium was investigated, using coleoptile segments (CS) of Triticim aestivum L. (cv. Hartri) and an optoelectronic device. The study aimed at separating the osmoelastic response from the irreversible growth response when an osmoticum (mannitol) was added, and to compare both processes in order to consider the possibility of growth‐induced reduction in turgor pressure. The prompt inhibition of elongation registered just after addition of 50 mM mannitol as well as the subsequent resumption of the original elongation rate could be quantitatively explained by the extent and the kinetics of the osmoelastic relaxation. An initial reduction in the irreversible elongation component by mild osmotic stress could not be demonstrated. Above a critical value, the irreversible growth was insensitive to a further increase in water potential. The minimum turgor pressure required to drive steady growth was not far from zero in both the presence and absence of auxin. The rate (r) of osmotically caused shortening per unit change of water potential r = 1/l x dl/dt x 1/Δψ was determined from the kinetics of CS shortening induced by addition of mannitol at nearly isotonic concentration (300 mM). This parameter relates a fractional change in length to the difference in water potential between inside and outside, and was assumed to depend largely on the hydraulic resistance of the tissue and cuticle. It was found to be independent of IAA. The relatively low value of Γ suggests significant reduction of turgor at high growth rates. In accordance with this conclusion, the extent of osmoelastic shortening after a transfer to 300 mM mannitol (dependent on wall strain) was significantly decreased in the presence of IAA. Addition of 100 μM IAA to CS growing at a constant rate induced pronounced oscillations in the rate of elongation, which may be connected with the change in elastic cell wall strain. Whereas the steady state growth rate before the addition of IAA was the same in the presence and in the absence of 50 mM mannitol, the maximum growth rate found after addition of IAA was substantially reduced in the mannitol variant. The significance of the osmotic potential of the seed apoplast sap as a regulator of assimilate transfer to and within coats of developing seed of Vicia faba (cv. Coles Prolific) was assessed using attached empty seed coats and intact developing seed. Following surgical removal of the embryos, through windows cut in the pod walls and underlying seed coats, the resulting attached "empty" seed coats were filled with solutions of known osmotic potentials (-0.02 venus -0.75 MPa). Sucrose efflux from the coats was elevated at the higher osmotic potential (low osmotic concentration) for the first 190 mill of exchange. Thereafter, this efflux was depressed relative to efflux from coats exposed to the low osmotic potential (high osmotic concentration) solution. This subsequent reversal in efflux was attributable to an enhanced diminution of the coat sucrose pools at the high external osmotic potential. Indeed, when expressed as a proportion of the current sucrose pool size, relative efflux remained elevated for coats exposed to the high osmotic potential solution. Measurement of potassium and sucrose fluxes to and from their respective pools in the coat tissues demonstrated that the principal fluxes, sensitive to variation in the external osmotic potential, were phloem import into and efflux from the "empty" coats. Phloem import, consistent with a pressure-driven phloem transport mechanism, responded inversely with changes in the external osmotic potential. In contrast, sucrose and potassium efflux from the coats exhibited a positive dependence on the osmotic potential. Growth rates of whole seed were approximately doubled by enclosing selected pods in water jackets held at temperatures of 25°C compared to 15°C. The osmotic potential of sap collected from the seed apoplast remained constant and independent of the temperature-induced changes in seed growth rates and hence phloem import. Based on these findings, it is proposed that control of phloem import by changes in the external osmotic potential observed with "empty" seed coats has no significance as a regulator of assimilate import by intact seed. Rather, maintenance of the seed apoplast osmotic potential, independent of seed growth rate, suggests that the observed osmotic regulation of efflux from the coats may play a key role in integrating assimilate demand by the embryo with phloem import. While solute transport and ethylene production by plant tissue are sensitive to the osmotic concentration of the solution bathing the tissue, the influence of tissue water relations and specifically tissue turgor potential on the kinetics of 1-aminocyclopropane-1-carboxylic acid (ACC) uptake into the vacuolar compartment and ethylene production have not been examined. 1-Aminocyclopropane-1-carboxylic acid transport and ethylene production were examined in tomato (Lycopersicon esculentum Mill. cv. Liberty) pericarp slices incubated in solutions having a range of mannitol, polyethylene glycol 3350 and ethylene glycol concentrations known to affect tissue water relations. Tissue osmotic and turgor potentials were derived from osmolality measurements of cell saps recovered by freeze-thawing and corrected for the contribution of the free-space solution. When relatively nonpermeable (mannitol or polyethylene glycol 3350) osmotica were used, both ACC uptake and ethylene production were greatest at a solution osmolality of 230 milliosmolal where tissue turgor potential ranged between 120 and 140 kPa. At higher and lower turgor potentials, the high-affinity saturating component of ACC uptake and ethylene production were inhibited, and ACC efflux from the vacuolar compartment was increased. The inhibition of ACC uptake was evident as a decrease in Vmax with no effect on Km. Turgor potential changes caused by adjusting solution osmolality with mannitol or polyethylene glycol 3350 were accompanied by changes in the osmotic potential and water potential of the tissue. The effects of turgor potential vs the osmotic and water potentials of tomato pericarp slices were differentiated by comparing responses to nonpermeable osmotica and mixtures of nonpermeable and permeable osmotica. Ethylene glycol-mannitol mixtures had effects on the osmotic potential and water potential of the tissue similar to those of nonpermeable osmotica but had less effect on tissue turgor, ACC transport and ethylene production. Incubating tissue in solutions without nonpermeable osmotica osmotically shocked the tissue. Increasing solution osmolality with ethylene glycol in the absence of nonpermeable osmotica increased tissue turgor and ethylene production. The present study indicates that tissue turgor is an important factor affecting the kinetics of ACC uptake into the vacuolar compartment and ethylene production in tomato pericarp slices. Seasonal and diurnal variation and rehydration effects of pressure-volume param- eters in Pseudotsuga menziesii (Mirb.) Franco from a plantation in central Pennsylvania, USA, were evaluated during May-September. 1989. Predawn elastic modulus was lowest in overwintering and newly expanded shoots in May and June, respectively, whereas predawn osmotic potentials at full and zero turgor were lowest in May and in early September, following an August drought. Seasonal variation in predawn relative water content at zero turgor was highly correlated with increases and decreases in elastic modulus and osmotic potential. Diurnal osmotic adjustment resulted in nearly constant turgor pressure, despite decreases in bulk shoot water potential. Elastic modulus decreased diurnally on 1 August and increased `on 3 September. Decreases in osmotic potential and/or elastic modulus on 24 June and I August lowered the relative water content at zero turgor. Correcting for plauteaus via linear regression had no significant effect on osmotic potential at full turgor: however, uncorrected elastic modulus and relative water content at zero turgor were often significantly lower than the plateau-corrected values, particularly in artificially rehydrated shoots. Plateau-corrected osmotic potential at full turgor and osmotic potential at zero turgor were significantly higher in most artificially re-hydrated shoots than in those naturally rehydrated as the result of loss of symplastic solutes, `Corrected elastic modulus decreased following 12 and 24 h of rehydration and corrected relative water content at zero turgor increased in as little as 3 h of rehydration. These results indicate that seasonal and diurnal patterns of tissue-water parameters in Pseudotsuga menziesii vary with plant phenology and drought conditions, and that the length of rehydration period is an important consideration for pressure-volume studies. Rose (Rosa hybrida cv. Madame G. Delbard) was micropropagated in vitro on agar at varying concentrations: 0, 4, 5.5, 6, 7.5, 8, 9 and 15 g l-1. Water status of the culture medium, leaf water potential, plant growth and propagation rate were investigated. Water potential of the medium was a linear function of agar concentration, and gel strength increased linearly from zero as agar concentration increased from 5 to 15 g l-1. Except for liquid medium, where shoot proliferation (total shoot number) was imparied by vitrification, fresh and dry weights and number of total shoots decreased linearly with increasing agar concentration. At low agar concentration, where water potential of the gelled medium was high, only few among numerous shoots that had attained a length of > 0.3 cm were usable. Number of usable (elongated) shoots was significantly higher at a moderate level of agar, namely 7.5 g l-1. This can be explained in terms of the antagonistic actions of cytokinin and water on shoot elongation. At low agar concentration, leaf water potential and absolute water content were high. At high agar concentration, under gel water deficit, leaf water potential and absolute water content were low but an osmotic adjustment maintained turgor pressure. Seasonal changes in the relationship between stomatal responses and water potential components in detached leaves of sweet cherry (Prunus avium L. cv. Bigarreau Morreau) and plum (Prunus domestica L. cv. Queen Victoria) were examined by the simultaneous measurement of stomatal conductance and water relation parameters derived from pressure-volume curves. The water potential threshold for stomatal closure in sweet cherry leaves throughout the season and in plum leaves until mid-July varied nearly in parallel with changes in the osmotic potential at the turgor loss point. From August onward, closing reactions of plum stomata had no relationship to the turgor loss point. Stomata of sweet cherry leaves opened more widely and showed slower reactions to dehydration early in the season than later. Endocytosis in animal cells has been heavily documented. Both fluid-phase and receptor-mediated modes of uptake have been frequently studied, and the endocytic pathway is well defined. This contrasts markedly with the situation in plants where our knowledge of this process is still rudimentary. Partly responsible for this situation has been the view, widely held among plant physiologists, that because of turgor, endocytosis cannot occur in plant cells. As discussed below, the case against endocytosis is no longer water-tight. Endocytosis is a fact in protoplasts. It can be demonstrated with electron-dense tracers as well as with membrane impermeant dye Lucifer Yellow CH. The latter has also been used with success on both suspension-cultured and tissue cells of higher plants, suggesting that fluid-phase endocytosis is also a feature of cells with walls. Through the application of fluorescently labelled elicitor molecules, which specifically bind to the cell surface of suspension-cultured cells, it has also been possible to provide convincing evidence for the operation of receptor-mediated endocytosis in plants. A number of studies on protoplasts and cells clearly indicate that endocytosis in plants can be mediated by coated pits in the plasma membrane. At least one of the organdIes that lie on the endocytic pathway in plants has a structurally similar counterpart in animal cells: the multivesicular body. The first recipient of internalized molecules is the partially coated reticulum, although its relationship to the Golgi apparatus and Golgi function remain to be clarified. Understanding the origin and role of growth-induced water potentials helps to explain how cell enlargement is inhibited without a decrease in turgor when water is depleted in the soil or more rapidly lost by transpiration. In many cases, changes in growth-induced water potentials probably cause the inhibition initially, but eventually the cell walls lose extensibility and specific proteins accumulate so that metabolic changes in the walls probably limit later growth. Tissue elasticity can affect plant response to drought, in terms of turgor maintenance and water uptake from drying soils. The purpose of this study was to determine the effect of mycorrhizal colonization and drought acclimation on rose (Rosa hybrida L. cv. Samantha) leaf elasticity. Bulk elasticity was characterized by the pressure-volume method using plots of the elastic modulus as a function of leaf turgor pressure, total water potential and relative water content. The treatments, arranged in a 2 × 3 factorial design, included acclimated and unacclimated plants, and either Glomus intraradices Schenck and Smith, Glomus deserticola Trappe, Bloss and Menge, or a non-mycorrhizal control. Plants with root mycorrhizal colonization showed reduced leaf elasticity (i.e. higher elastic moduli) over a broad range of leaf water potential and water content. Both mycorrhizal colonization and acclimation facilitated the maintenance of positive values of turgor and elasticity at lower leaf water potential and water content than in controls. The diurnal and seasonal changes in plant water relations of two Japonica rice (Oryza sativa L.) cultivars, Nipponbare and Tachiminori, were studied under flooded conditions at Kyoto University. The dryland cv. Tachiminori maintained higher predawn and midday leaf osmotic potentials relative to the wetland cv. Nipponbare during the vegetative stage, but the ranking was reversed after flowering. The relationship between leaf water potential and leaf osmotic potential showed that prior to panicle emergence Nipponbare was able to adjust osmotically to maintain turgor, whereas after heading there was little turgor maintenance. Tachiminori showed little difference in osmotic adjustment before and after panicle emergence. Fertilizer treatment during panicle development also helped to maintain the degree of osmotic adjustment in both cultivars.

Changes in water status, membrane permeability, ethylene production and levels of abscisic acid (ABA) were measured during senescence of cut carnation flowers (Dianthus caryophyllus L. cv. White Sim) in order to clarify the temporal sequence of physiological events during this post‐harvest period. Ethylene production and ABA content of the petal tissue rose essentially in parallel during natural senescence and after treatment of young flowers with exogenous ethylene, indicating that their syntheses are not widely separated in time. However, solute leakage, reflecting membrane deterioration, was apparent well before the natural rise in ethylene and ABA had begun. In addition, there were marked changes in water status of the tissue, including losses in water potential (ψw), and turgor (ψp), that preceded the rise in ABA and ethylene. As senescence progressed, ψw continued to decline, but ψp returned to normal levels. These temporal relationships were less well resolved when senescence of young flowers was induced by treatment with ethylene, presumably because the time‐scale had been shortened. Thus changes in membrane permeability and an associated water stress in petal tissue appear to be earlier symptoms of flower senescence than the rises in ABA or ethylene. These observations support the contention that the climacteric‐like rise in ethylene production is not the initial or primary event of senescence and that the rise in ABA titre may simply be a response to changes in water status.
