The aim of the present work was to develop and evaluate sustained release colon targeted micropellets of lornoxicam in order to achieve release of the drug at colon which could result in enhanced local absorption and thereby improved bio-availability. The present worker prepared micropellets of lornoxicam in nine batches using pure drug, DCP, PVPK-30 and different ratios of HPMCK-4M taking into account the direct pelletization technique. Resulted micropellets were fiilled in capsules and coated with eudragit S-100 to achieve colon targeted release. Compatibility studies were by using FTIR and TLC methods. Particle size determination of micronized lornoxicam was performed by SEM which revealed that the mean particle diameter was in the range of 104-263 μm while percentage yield was in the range of 88.4-98.4%.Various pre-formulation physiochemical parameters of formulation blends were evaluated such as bulk density, tapped density, carr's index, hausner's ratio and angle of repose and coating durability test for coated capsules (passed). The optimized batch was compared for its release profile in pH 7.4 phosphate buffers and simulated colonic fluid, which were found to be 94.64% and 90.58% respectively. The actual responses were in accordance with the predicted values which showed validity of the model. A simple, rapid, precise and accurate derivative spectrophotometric method has been developed for the determination of Lornoxicam and Paracetamol in combined dosage forms (Tablets). The zero crossing point of Paracetamol (247.42 nm) has been selected for the quantification of Lornoxicam whereas the zero crossing point of Lornoxicam (233.28 nm) has been selected for the quantification of Paracetamol from the first order derivative spectrum observed in borate buffer. The method obeys Beer-Lambert's law over the concentration range 5-50 μg/ml for Lornoxicam and 5-60 μg/ml for Paracetamol. Introduction: Lornoxicam is a selective cyclooxygenase-1 and 2 inhibitor that exhibit anti inflammatory, analgesic and anti pyretic activities. It is used in the management of osteoarthritis, rheumatoid arthritis, postoperative pain and primary dysmenorrhoea. Aim: The present work describes a simple, precise and accurate HPTLC method for its estimation as bulk and in tablet dosage form. Methods: The chromatographic separation was carried out on precoated silica gel 60 GF254 aluminium plates using mixture of Tetrahydrofuran: Methanol:Ethyl acetate:Ammonia (2:1.5:5.5:0.6 v/v/v/v) as mobile phase and densitometric evaluation of spots was carried out at 377 nm using CAMAG TLC Scanner-3 with win CATS 1.4.1 version software. The experimental factors like band width of spot, chamber saturation time, slit width, solvent front migration etc. were critically studied and optimum conditions were developed. Results: The drug was satisfactorily resolved with Rf value 0.34 ± 0.01. The accuracy and reliability of the proposed method was determined by evaluating various validation parameters like linearity (100-1000 ng/band), precision (intra-day RSD 0.05-0.10 %, inter-day RSD 0.04-0.13%), accuracy (98.26-100.60%) and specificity according to ICH guidelines. The drug was also subjected to acid, alkali, oxidation, thermal and photochemical degradation studies and the method was found to be effective in separating the drugs from their degradation products. Conclusion: The proposed method is simple, accurate, precise and may be used as a cost-effective quality control tool for routine analysis of Lornoxicam as bulk drug and in tablet formulation. Background: Paracetamol and lornoxicam in combined tablet dosage form are available in the market. This combination is used to treat inflammatory diseases of the joints, osteoarthritis and sciatica. Spectrophotometric and high performance liquid chromatography (HPLC) methods have been reported for their simultaneous estimation in tablet dosage form in specific solvent. This paper presents simple, accurate and reproducible spectrophotometric method for simultaneous determination of paracetamol and lornoxicam in tablet dosage form in different dissolution media. The reported method is helpful in determination of paracetamol and lornoxicam during dissolution study. Materials and Methods: Simple, sensitive, accurate and economical spectrophotometric method based on an absorption correction equation was developed for the estimation of paracetamol and lornoxicam simultaneously in tablet dosage form in different dissolution media at different pH. Results: Paracetamol showed absorption maxima at 243 nm in 0.1N HCland phosphate buffer pH 6.8, while lornoxicam showed absorption maxima at 374 nm in 0.1N HCland phosphate buffer pH 6.8. The linearity was obtained in the concentration range of 4-12 µg/ml for paracetamol and 4-16 µg/ml for lornoxicam. Discussion: The concentrations of the drugs were determined by an absorption correction equation method. The results of analysis have been validated statistically by recovery studies. Aim: To develop a simple, precise, rapid and accurate HPTLC method for the simultaneous estimation of Lornoxicam (LOR) and Thiocolchicoside (THIO) in bulk and pharmaceutical dosage forms. Materials and Methods: The separation of the active compounds from pharmaceutical dosage form was carried out using methanol:chloroform:water (9.6:0.2:0.2 v/v/v) as the mobile phase and no immiscibility issues were found. The densitometric scanning was carried out at 377 nm. The method was validated for linearity, accuracy, precision, LOD (Limit of Detection), LOQ (Limit of Quantification), robustness and specificity. Results: The Rf values (±SD) were found to be 0.84 ± 0.05 for LOR and 0.58 ± 0.05 for THIO. Linearity was obtained in the range of 60-360 ng/band for LOR and 30-180 ng/band for THIO with correlation coefficients r² = 0.998 and 0.999, respectively. The percentage recovery for both the analytes was in the range of 98.7-101.2 %. Conclusion: The proposed method was optimized and validated as per the ICH guidelines. [ABSTRACT FROM AUTHOR]

Aim: The objective of the study was to develop simple RP-HPLC method for the simultaneous determination of paracetamol and lornoxicam without prior separation. Materials and Methods: In this method, Kromasil C8 (250 mm, 4.6 mm, 5 μm) column was used. The mobile phase used was methanol:phosphate buffer (60:40, v/v, pH 6.4), at flow rate of 1 ml min-1. UV detection was monitored at 302 nm. Results: Calibration graphs were established in the range of 1-150 μg ml-1 and 0.5-100 μg ml-1 for paracetamol and lornoxicam, respectively. The average retention time for paracetamol and lornoxicam was found to be 3.15 ± 0.03 min and 5.25 ± 0.06 min, respectively. The detection limit and quantitation limit for paracetamol are 0.19 μg ml-1 and 0.59 μg ml-1 and for lornoxicam 0.10 μg ml-1 and 0.31 μg mL-1, respectively. The intraday and interday precision expressed as percent relative standard deviation were below 2%. The mean recovery of paracetamol and lornoxicam was found to be in the range of 99.03-101.2%. Conclusion: The validated HPLC method was found to be rapid, precise and accurate and can be readily utilized for analysis of paracetamol and lornoxicam in bulk and in pharmaceutical formulations. A simple, precise, reliable, rapid and reproducible reversed phase-high-performance liquid chromatography method was developed and validated for the simultaneous estimation of Paracetamol (PCM) and Lornoxicam (LOX) present in tablet dosage forms. Chromatographic separation achieved isocratically on Luna C18 column (5 µm, 150 x 4.60 mm) and methanol/phosphate buffer (60:40, v/v, pH 7.0) as mobile phase, at a flow rate of 1 ml/min. Detection was carried out at 260 nm. Parameters such as linearity, precision, accuracy, recovery, specificity and ruggedness are studied as reported in the ICH guidelines. The retention times for PCM and LOX was found to be 2.06±0.013and 4.38±0.07 min, respectively. Linearity for PCM and LOX was in the range of 10-50 µg/ml and 8-40 µg/ml, respectively. The mean recoveries obtained for LOX and PCM were 100±0.16 and 99.50±0.43%, respectively, and relative standard deviation (RSD) was less than 2. The correlation coefficients for all components are close to 1. The RSDs for three replicate measurements in three concentrations of samples in tablets are always less than 2%.
