The anomeric arylthio group and the hydroxyl-protecting groups of thioglycosides were optimized to construct carbohydrate building blocks for automated electrochemical solution-phase synthesis of oligoglucosamines having 1,4-β-glycosidic linkages. The optimization study included density functional theory calculations, measurements of the oxidation potentials, and the trial synthesis of the chitotriose trisaccharide. The automated synthesis of the protected potential N,N,N-trimethyl-d-glucosaminylchitotriomycin precursor was accomplished by using the optimized building block.

A new iterative one-pot sequential method for the solution-phase synthesis of oligosaccharides has been devised on the basis of the electrochemical oxidation of a propagating thioglycoside terminus to generate the corresponding triflate, followed by the reaction with a thioglycoside building block having a free hydroxyl group. A practical automated synthesizer was developed for the method and was effectively used for assembling up to six thioglycoside building blocks to synthesize partial structures of poly-β-D-(1-6)-N-acetylglucosamine.

Pauson-Khand reactions were achieved at ambient temperature without any additive using a photochemical flow microreactor. The efficiency of the reaction was better than that in a conventional batch reactor, and the reaction could be operated continuously for 1 h.

Oxidative hydroxylation of toluene derivatives via alkoxysulfonium ion intermediates was achieved by integration of anodic oxidation and hydrolysis to give benzyl alcohols which are also susceptible to oxidation. Alkenes were also oxidized to give 1,2-diols without overoxidation. Glycosyl sulfonium ions, which serve as persistent glycosyl cation equivalents, were prepared by the addition of diorganosulfides to an electrochemically generated glycosyl triflate. Low-temperature and variable-temperature NMR studies were performed to reveal the structure, stability, and reactivity of glycosyl sulfonium ions. The glycosyl sulfonium ions could be used as storable intermediates for reactions with various glycosyl acceptors including thioglycosides to give the corresponding disaccharides.

Carbolithiation of conjugated enynes with aryllithiums generated by Br-Li exchange reaction followed by reaction of the allenyllithium compounds with chlorosilanes was carried out in a microflow system to obtain various allenylsilanes in good yields.

Selective monolithiation of dibromobiaryls, such as 2,2'-dibromobiphenyl, 4,4'-dibromobiphenyl, 2,7-dibromo-9,9-dioctylfluorene, 2,2'-dibromo-1,1'-binaphthyl, and 5,5'-dibromo-2,2'-bithiophene, with 1 equiv of n-butyllithium followed by the reaction with electrophiles was achieved using a microflow system by virtue of fast micromixing and precise temperature control. Sequential introduction of two different electrophiles based on this method was also achieved using a microflow system composed of four micromixers and four microtube reactors.

"Cation pools" of diarylcarbenium ions have been generated by the oxidative C-H bond dissociation of diarylmethanes using anodic oxidation. "Diarylcarbenium ion pools" thus generated react with various nucleophiles, such as allylsilanes, ketene silyl acetals, and aromatic compounds. The reductive homocoupling of the "diarylcarbenium ion pool" has been achieved. The dimer thus obtained also serves as a precursor of the "diarylcarbenium ion pool" via oxidative C-C bond dissociation. [reaction: see text]

[reaction: see text] A novel three-component cyclization using allenylboronate ester is described. A three-component assembly of allenylboronate ester, aryl iodides, and stabilized carbon nucleophiles took place in the presence of palladium catalyst, furnishing functionalized alkenylboronate esters with high regio- and stereoselectivity. A Rh-catalyzed cyclization of the resultant three-component products then afforded interesting carbocyclic frameworks efficiently.

[reaction: see text] (2-Pyridyl)allyldimethylsilanes were found to be novel pyridyl transfer reagents in palladium-catalyzed cross-coupling reactions of various aryl iodides in the presence of silver(I) oxide as an activator.

[reaction: see text] We have developed the homocoupling reaction of alkenyl(2-pyridyl)silanes mediated by CuI and CsF, in which the strong directing effect of the 2-pyridyl group was observed. The homocoupling reaction was successfully integrated with the Mizoroki-Heck reaction of vinyl(2-pyridyl)silane with aryl halides, enabling a rapid and stereoselective synthesis of multisubstituted butadienes. From a relatively small compound library, it was possible to detect a number of fluorescent butadienes with a wide range of fluorescence color variations (blue to reddish-orange).

[reaction: see text] An N-acyliminium ion pool was found to undergo cycloaddition reaction with a variety of dienophiles such as alkenes and alkynes. A concerted mechanism seems to be most likely for alkyl-substituted alkenes as suggested by the DFT calculations, whereas a stepwise mechanism plays the major role for aryl-substituted alkenes. It is also noteworthy that the present study demonstrates the potential of the combination of the cation pool method and the micromixing in both mechanistic and synthetic aspects.

[formula: see text] The generation of (2-PyMe2Si)2CHLi was easily accomplished by the deprotonation of (2-PyMe2Si)2CH2 using n-BuLi in Et2O. Thus generated (2-PyMe2Si)2CHLi was found to react with a variety of aldehydes and ketones to give the corresponding vinylsilanes in extremely high yields with complete stereoselectivities.
