Perhalogenated boron clusters derived from B

A new type of water-soluble anchor dyes, that is, dyes which carry an auxiliary unit for strong binding to macrocyclic host molecules, has been synthesized. It consists of 7-nitrobenzofurazan (NBD) as a dye and the dodecaborate cluster (B12H11R) as a dianionic, globular, and purely inorganic anchoring group for cyclodextrins (Ka10(5) M(-1)). The synthesized dodecaborate-substituted dyes show marked changes in their photophysical properties (UV-vis and fluorescence) upon complexation with cyclodextrins (β-CD and γ-CD), such that the resulting host·dye complexes (1:1 stoichiometry) present sensitive reporter pairs for indicator displacement applications.

Kinetic solvent effects on hydrogen abstractions involving C-H donors (dienes, ethers, alkylbenzenes) have been corroborated by experiment and theory (UB3LYP/6-311++G**, polarized continuum model). To single out the effect of solvent polarity, rate constants for scavenging of the cumyloxyl radical and fluorescence quenching of 2,3-diazabicyclo[2.2.2]oct-2-ene were obtained in binary aprotic mixtures of ethylacetate and acetonitrile. Polar solvents result in a selective stabilization of the reactants (see TOC graphic), which results in slower rate constants.

The derivatization of 3-amino-9-ethylcarbazole with a diamino-alkyl anchor affords a fluorescent dye suitable for indicator displacement from cucurbituril macrocycles. The novel compound 1 shows, due to a complexation-induced pKa shift, a large and predictable dual fluorescence response (100-fold increase at 375 nm and 9-fold decrease at 458 nm) upon supramolecular encapsulation and a strong affinity for cation-receptor macrocycles, in particular cucurbit[6]uril (CB6). A direct application is presented by monitoring the enzymatic activity of lysine decarboxylase.

Kinetic solvent effects on hydrogen abstractions, namely, acceleration in nonpolar solvents, have been presumed to be restricted to O-H hydrogen donors. We demonstrate that also abstractions from C-H and even Sn-H bonds by cumyloxyl radicals and n,pi*-excited 2,3-diazabicyclo[2.2.2]oct-2-ene are fastest in the gas phase and nonpolar solvents but slowest in acetonitrile. Accordingly, solvent effects on hydrogen abstractions are more general, presumably due to stabilization of the reactive oxygen or nitrogen species in polar solvents.

[reaction: see text] The fluorescence quenching of 2,3-diazabicyclo[2.2.2]oct-2-ene (DBO) by nucleotides has been studied. The quenching mechanism was analyzed on the basis of deuterium isotope effects, tendencies for exciplex formation, and the quenching efficiency in the presence of a molecular container (cucurbit[7]uril). Exciplex-induced quenching appears to prevail for adenosine, cytidine, and uridine, while hydrogen abstraction becomes competitive for thymidine and guanosine. Compared to other fluorescent probes, DBO responds very selectively to the type of nucleotide.

[reaction: see text] Photolysis of hydroxyacetophenone and thermolysis of the corresponding dioxetane afford spiroiminodihydantoin rather than 4,8-dihydro-4-hydroxy-8-oxo-2'-deoxyguanosine (4-HO-8-oxodG) through the oxidation of 2'-deoxyguanosine (dG) by triplet-excited hydroxyacetophenone and the peroxyl radicals derived thereof by alpha cleavage and subsequent oxygen trapping. The structure of the spiroiminodihydantoin is assigned by the SELINQUATE NMR technique, which unequivocally establishes the spirocyclic connectivity.

Photoactivable (“caged”) fluorescent dyes and probes are crucial for temporally and spatially resolved tracer experiments, e.g., in cell biology or fluid physics. The thiadiazolidinedione 1 represents a new class of caged fluorophore. Upon UV-irradiation it releases in a rapid photoreaction with high quantum yield the azoalkane 2. Longer wavelength excitation of 2 to the singlet excited state results in strong and long-lived fluorescence with maximum intensity at 425 nm. It has been demonstrated that one single uncaging laser pulse suffices for time-resolved or steady-state detection of the fluorescence.

The rates of acid hydrolysis of N-benzoyl-cadaverine (1), mono-N-(tert-butoxy)carbonyl cadaverine (2), and benzaldoxime (3) with binding motifs for cucurbit[6]uril (1,2) and cucurbit[7]uril (1,3) were investigated in the absence and presence of these hosts. Significant rate enhancements (up to a factor of ca. 300 for the hydrolysis of 3) were observed. Competitive inhibition due to encapsulation of added cadaverine and the successful use of sub-stoichiometric amounts of macrocycle confirmed the function of cucurbiturils in promoting acid hydrolysis.
