Sugar substrates are rich in organic compounds. However, its concentration may vary depending on the culture. Thus, the objective of this work was to evaluate the composition of sugarcane and sorghum juices using the UVvis technique. An evaluation of the composition of the substrates was carried out and a pre-inoculum with YPSAC 5% medium, where 0.10 grams of FT858 yeast was inoculated and kept incubated at 30°C for 24h at 250rpm. The cells were recovered by centrifugation, resulting in a concentration of 10 mg mL-1 of wet mass that was inoculated into 50 mL of the previously prepared fermentation medium with a concentration of 22°Brix and pH 5.0. Aliquots were taken for evaluation of molecular absorption spectra using a spectrophotometer (Global Trade Technology). These substrates are composed of water and fermentable sugars. The evaluation of the molecular absorption spectra showed a difference in the profile of the substrates as well as in the subsequent hours of fermentation, with a decrease in the intensity of the peaks being observed that may be related to the absorption of nutrients by the yeast. vis shows promise for this type of analysis. The state of Mato Grosso do Sul (Brazil) has a rich biodiversity, with varied biomes and many native species. Simultaneously, the plants in the region have been studied for their chemical and biological potential, including photoprotective properties and applications in phytocosmetics. Edible plants have potential application based on the presence of photoprotective and antioxidant compounds in their chemical composition. In this context, the aim was to review systematically studies on the photoprotective action of native food plants from Mato Grosso do Sul in the Scopus database, delimiting for articles published between 2000 and 2020. Only 4.08% of the species presented articles exploring the photoprotective activity, distributed in 10 articles. The extracts evaluated were varied, with different formulations. 40% of the articles addressed ethanolic extracts, 20% hydroethanolic extracts, and 40% vegetable fixed oil. The lowest sun protection factor reported was for the aqueous extract of the fruit pulp of Hymenaea martian Hayne (SPF 0.66) and the highest was for a formulation using Schinus terebinthifolius Radi extracts added to Lanette cream (SPF 32.40). Casearia sylvestris var. lingua (Cambess.) Eichler is widely used in traditional medicine to treat diseases. Simultaneously, the growing search for phytocosmetics has culminated in the exploration of plant extracts. In this context, this study aimed to obtain the chemical composition and antioxidant and photoprotective potential of the infusion of C. sylvestris var. lingua leaves. The techniques of UV/Vis spectroscopy, gas chromatography with mass spectrometry detector (GC-MS) and liquid chromatography with diode array detector (LC-DAD) were used to get the results. The total phenolic content in the infusion of leaves from C. sylvestris var. lingua was 101.57 mg GAE g-1, flavonoids 50.37 mg RE g-1 and tannins 1.12 mg TAE g-1. Quercetin, ferulic acid, gallic acid, ellagic acid, caffeic acid, β-sitosterol, lupeol, lupeol acetate, stigmasterol and campesterol were identified and quantified in the samples. The infusion of C. sylvestris var. lingua leaves has potential for application in phytocosmetics and sunscreens, with a sun protection factor of 8.65 ± 0.45, a UVA/UVB ratio of 1.16 and a critical wavelength of 373. Coconut oil has more than 90% of saturated fatty acids. Of this percentage, about 47% of lauric acid, an important saturated acid that gives great stability to oxidation, in addition to benefic properties such as anti-inflammatory. Sunflower oil also presents important properties for daily balance of the body, however, the high amount of unsaturated fatty acids provides low oxidative stability. Knowing the nutritional values of both raw materials under study, the aim of this study was to mix them in two different proportions: 50% sunflower + 50% coconut (blend 1) and 70% sunflower + 30% coconut (blend 2), in order to evaluate the antioxidant potential and chemical composition, by analysis of fatty acids content; antioxidant activity (DPPH); phenolic compounds; oxidative stability, by accelerated oxidation test, Rancimat method. The acidity values for both pure oils and blends are within the values established by Anvisa and Codex Alimentarius, and phenolic compounds were proportional to the amount of sunflower oil incorporated into the coconut oil. The values of oil stability index (OSI), determined by Rancimat, were high, especially for coconut oil, and in the mixtures, the increase in oxidative stability was proportional to the amount of coconut oil added to sunflower oil. Yeasts are exposed to stressing agents in the fermentation process. The study aims to analyze the production of ethanol and intracellular glycerol, as well as to evaluate the consumption of amino acids by yeast FT-858 in different culture conditions. The sugarcane and sorghum broth were used, at 22ºBx and pH of 5.0. A 2% YPD medium was pre-inoculated, and 0.10 g of lyophilised yeast were inoculated, incubated at 30 °C for 12 hours at 250 rpm. Cells were recovered by centrifugation and reinoculated in the fermentation medium at 30 °C and 40 °C at 250 rpm, this procedure was conducted with cell recycles, and 4 mL aliquots were withdrawn for analysis. Quantification of glycerol was accomplished by cell lysis and enzymatic kit of triglycerides, ethanol by gas chromatography and amino acid analysis by high performance liquid chromatography. The results showed that intracellular glycerol production and ethanol concentration were inversely proportional due to stress factors. The highest amount of amino acids available was in sorghum broth, and the most assimilated by the yeast were: serine, arginine, alanine and tryptophan and the high temperature accelerated their consumption of them. The light source can cause ultrastructural and morphological changes in yeast and affect your metabolism. Thus, this work has the objective of evaluating the influence of white and ultraviolet light on the fermentative performance of FT-858 yeast in different culture conditions. Yeast cells were grown in Yeast Extract-Peptone-Dextrose (YPD 2%) in which 0.10 g of yeast were added and incubated on a shaker for 10 h at 30 °C at 250 rpm, subsequently washed and centrifuged in solution saline. The biomass harvested was used in the fermentation experiments that were readily performed under white and ultraviolet light with temperature control. Aliquots were withdrawn for cell viability analysis and ethanol production. Yeast showed better performance under white light at 28 °Brix concentration at 30 °C. Ultraviolet light associated with high temperature and substrate concentration affected yeast physiology by directly interfering with viability and ethanol production. [ABSTRACT FROM AUTHOR]

Brazil, which has a great territorial extension, with fertile soils and availability of water and, in this way, can produce a diversity of vegetal products with bioenergetics potential. Thus, this study aims to evaluate the technological characteristics of saccharine substrates of direct fermentation for ethanol production as well as to analyze the fermentative profile of FT-858 yeast in different culture conditions. For this, a bibliographic survey was carried out comparing the technological qualities of the analyzed biomasses. To evaluate the fermentative capacity, the saccharine substrates were used at concentrations of 18 and 32°Brix at 30 °C for 24 hours in test tubes containing Durhan tubes. For the analysis of the fermentation profile the yeasts were grown in 2% YPD medium and the biomass obtained was used in the saccharine substrates of direct fermentation for 50 hours. The saccharine substrates presented potential for ethanol production: sugar cane and saccharine sorghum. The yeast FT-858 presented fermentation capacity in the different substrates in the concentrations of Brix analyzed, presenting a better fermentative performance in sugarcane broth and the highest concentrations of ethanol occurring in the times of 10 and 15 hours of fermentation for both substrates. During ethanol production, yeasts are subjected to a number of stress factors - among them high substrate concentrations and high temperatures - while displaying an ability to adapt to the adverse conditions found during fermentation. This study evaluated the metabolic profile exhibited by Saccharomyces cerevisiae Catanduva-1 during fermentation. Yeast cells were cultured in 2% YPD medium, incubated in a shaker at 30 °C and 250 rpm for 10 h, and subsequently centrifuged and washed in saline solution for biomass harvesting. Fermentation experiments were performed at 18, 22, and 25°Bx sterilized at 120 °C for 20 min. Biomass was reinoculated into wort and incubated in a shaker at 30 °C and 40 °C at 250 rpm. Evaluation of fermentation parameters revealed better fermentative performance at 30 °C for all substrate concentrations investigated. Under the test conditions, cellular stress was observed at 40 °C. A superior ethanol production profile was obtained at 22°Bx and 30 °C. Glycerol accumulation was highest at 25°Bx and 40 °C.
