Oligodendrocyte and neural precursor cells (OPCs and NPCs, respectively) in the central nervous system (CNS) have diverse roles in development and homeostasis. During development, precursors build the CNS. In adulthood, they maintain their ability to proliferate and generate differentiated progeny, indicating their tremendous potential to regenerate and repair injured or degenerated CNS. How can we utilize this capability? Cross-talk between neurons and OPCs may hold some clues. Neurons communicate with OPCs via two mechanisms: 1) paracrine secretion of ligands, and 2) neuronal activity and bona fide synapses with OPCs. Intriguingly, OPCs express receptors for chemokines, which are small signalling molecules produced by various cells, including neurons. In addition to inducing chemotaxis, chemokines also regulate cell proliferation, survival and differentiation. In this review, we will summarize the roles of neuronally secreted chemokines and their documented ability to directly regulate the diverse functions of OPCs and NPCs in the developing as well as adult normal and injured CNS. We will focus on the following neuronal chemokines: CCL2, CCL3, CCL20, CCL21, CXCL1, CXCL8, CXCL9, CXCL10, CXCL11, CXCL12 and CX3CL1. We will discuss the implications for neuronal chemokine signalling in OPCs and NPCs not only in developmental myelination and adult CNS regeneration, but also in cognition, behavior, neuroinflammation and neuronal function., (Copyright © 2019 Elsevier B.V. While an injury to the central nervous system (CNS) in humans and mammals is irreversible, amphibians and teleost fish have the capacity to fully regenerate after severe injury to the CNS. Xenopus laevis has a high potential to regenerate the brain and spinal cord during larval stages (47-54), and loses this capacity during metamorphosis. The optic nerve has the capacity to regenerate throughout the frog's lifespan. Here, we review CNS regeneration in frogs, with a focus in X. laevis, but also provide some information about X. tropicalis and other frogs. We start with an overview of the anatomy of the Xenopus CNS, including the main supraspinal tracts that emerge from the brain stem, which play a key role in motor control and are highly conserved across vertebrates. We follow with the advantages of using Xenopus, a classical laboratory model organism, with increasing availability of genetic tools like transgenesis and genome editing, and genomic sequences for both X. laevis and X. tropicalis. Most importantly, Xenopus provides the possibility to perform intra-species comparative experiments between regenerative and non-regenerative stages that allow the identification of which factors are permissive for neural regeneration, and/or which are inhibitory. We aim to provide sufficient evidence supporting how useful Xenopus can be to obtain insights into our understanding of CNS regeneration, which, complemented with studies in mammalian vertebrate model systems, can provide a collaborative road towards finding novel therapeutic approaches for injuries to the CNS., (Copyright © 2016 Elsevier Ireland Ltd. Physiological (spontaneous) and reactive (reparative) regenerative processes are fundamental part of life and greatly differ among the different animals and tissues. While spontaneous regeneration naturally occurs upon cell attrition, reparative regeneration occurs as a consequence of tissue damage. Both spontaneous and reparative regeneration play an important role in maintaining the normal equilibrium of the central nervous system (CNS) as well as in promoting its repair upon injury. Cells play a critical role in reparative regeneration as regenerating structures (cells or tissues) depend on the proliferation without (de)differentiation of parenchymal cells surviving to the injury, proliferation of stem (progenitor) cells resident in the injured tissue, dedifferentiation of mature cells in the remaining tissue, or by the influx of stem cells originating outside the damaged tissue. Considering the central role of stem and progenitor cells in regeneration, a spur of experimental stem cell-based transplantation approaches for tissue (e.g. CNS) repair has been recently generated. This review will focus on the therapeutic efficacy of different sources of somatic stem cells - and in particular on those of neural origin - in promoting CNS repair in a chronic (auto)immune-mediated inflammatory disorder such as multiple sclerosis.

In this study we investigated the reference frames used to plan arm movements. Specifically, we asked whether the body axis, visual cues and graviception can each play a role in defining "up" and "down" in the planning and execution of movements along the vertical axis. Horizontal and vertical pointing movements were tested in two postures (upright and reclined) and two visual conditions (with and without vision) to identify possible effects of each of these cues on kinematics of movement. Movements were recorded using an optical 3D tracking system and analysis was conducted on velocity profiles of the hand. Despite a major effect of gravity, our analysis shows an effect of the movement direction with respect to the body axis when subjects were reclined with eyes closed. These results suggest that our CNS takes into account multimodal information about vertical in order to compute an optimal motor command that anticipates the effects of gravity.

Jet propulsion fuel-8 (JP-8) is the predominant fuel for military land vehicles and aircraft used in the US and NATO. Occupational exposure to jet fuel in military personnel has raised concern for the health risk associated with such exposure in the Department of Defense. Clinical studies of humans chronically exposed to jet fuel have suggested both neurotoxicity and neurobehavioral deficits. We utilized rat neurobiology U34 array to measure gene expression changes in whole brain tissue of rats exposed repeatedly to JP-8, under conditions that simulated possible occupational exposure (6 h/day for 91 days) to JP-8 vapor at 250, 500, and 1000 mg/m(3), respectively. Our studies revealed that the gene expression changes of exposure groups can be divided into two main categories according to their functions: (1). neurotransmitter signaling pathways; and (2). stress response. The implications of these gene expression changes are discussed.

We examined directional sensitivities in the dendritic activity of the identified giant interneurons (GIs) in the cricket, using in vivo Ca(2+) imaging during different directional air-current stimuli. Air current stimulus evoked action potential burst and quick Ca(2+) increase in GI. The stimulus direction of the maximal Ca(2+) responses corresponded to that of the maximal voltage response. However, the shapes of the directional tuning curves based on the Ca(2+) responses for each dendritic branch were different from the overall tuning curve based on spike counts for the cell. Moreover, different dendritic branches displayed distinct directional sensitivity profiles to the air-current stimuli. We propose that postsynaptic activities will influence the local Ca(2+) signals in the distal dendrites, and produce the difference in directional sensitivity of the dendritic Ca(2+) response.

It is known that benzodiazepines have a hypotensive effect, but the mechanism has not been well elucidated yet. To clarify whether this effect is due to central or peripheral mechanism, we administered 5 mg of diazepam or saline intravenously to healthy volunteers and assessed the change in blood pressure, heart rate, muscle sympathetic nerve activity and heart rate variability. After diazepam administration, systolic and mean blood pressure decreased significantly. Muscle sympathetic nerve activity was also significantly reduced but heart rate did not change, whereas the variables of spectral analysis of heart rate variability did not show significant change. We concluded that the hypotensive effect of diazepam in human is mainly due to the central mechanism.

We studied how the central nervous system (CNS) organizes outputs of effectors in a redundant motor task. During four-finger ramp force production, finger forces show positive covariations across trials at low forces, which turn into negative covariations at a critical force value (F(CR)). Subjects performed such tasks with different target amplitudes and durations of the ramp. F(CR) showed significant linear relations to the rate of force change. The slopes of the relations varied across subjects corresponding to a critical time (T(CR)) ranging from 0.13 to 0.84 s. Across subjects, T(CR) showed no relation to maximal force production; T(CR) increased with the ramp duration. We conclude that the CNS needs a certain time to organize stabilization of total force by a negative covariation among finger forces.

The electrical properties of neurons are produced by the coordinated activity of ion channels. K+ channels play a key role in shaping action potentials and in determining neural firing patterns. Small conductance Ca2+-activated K+ (SK(Ca)) channels are involved in modulating the slow component of afterhyperpolarization (AHP). Here we examine whether rat type 2 SK(Ca) (rSK2) channels can affect the shape of the action potential and the neural firing pattern, by overexpressing rat SK2 channels in Aplysia neuron R15. Our results show that rSK2 overexpression decreased the intra-burst frequency and changed the regular bursting activity of neurons to an irregular bursting or beating pattern in R15. Furthermore, the overexpression of rSK2 channels increased AHP and reduced the duration of the action potential. Thus, our results suggest that ectopic SK(Ca) channels play an important role in regulating the firing pattern and the shape of the action potential.

The present study examined the correlation between the level of activity neuron B52 and the transition from protraction to retraction phases of buccal motor patterns (BMPs) and the termination of the BMPs. The level of activity in B52 during the protraction phase was positively correlated with the duration of that phase. A second burst of activity in B52 was associated with the termination of the retraction phase. An apparent monosynaptic inhibitory connection from B52 to B64, may mediate the effects of B52. The first burst of activity in B52 delays the onset of activity in B64, thereby prolonging the protraction phase, and the second burst inhibits activity in B64, thereby terminating the retraction phase. These results suggest that activity in B52 may contribute to switching between ingestion-like and rejection-like BMPs by regulating both phase transition and termination of BMPs.

This study demonstrated that expiratory resistive loading (ERL) induced an increase in force fluctuation during a unilateral, submaximal isometric contraction of the non-dominant left vastus lateralis (VL), but did not effect force fluctuation during complex bilateral contractions. The increase in force fluctuation in the unilateral left VL contraction during ERL was not accompanied by alterations of average force production, motor unit activation (median power frequency) or airflow rate when compared to the bilateral contraction. Inspiratory RL (IRL) did not significantly affect force fluctuation in unilateral or bilateral contractions. The results concur with previous reports of ERL, but not IRL, effecting VL function and suggest that patients with obstructive diseases may also be vulnerable to reduced fine motor control.

During gait initiation (GI), decoupling of the center of mass and center of pressure allows the center of mass to fall forwards. Subjects initiated gait rapidly before and after tibial nerve block of the tibial nerve. Static single limb stability, stance limb ground reaction forces, electromyogram and temporal data were measured. It was hypothesized that postural stability would decrease post-block and that this would affect the kinetic and temporal properties of GI. Subjects had significantly decreased postural stability post-block and changes in normal gait kinetics, however, no changes were noted in ground reaction forces or relative temporal data of the GI task. The finding that GI was unaffected by diminished single leg postural instability suggests that GI is a pre-programmed task.

Ingestive and egestive behaviors in Aplysia are generated by motor neurons and interneurons chiefly located in the buccal ganglion, but cerebral ganglion neurons appear to contribute to both ingestive and egestive motor programs. We investigated if the cerebral ganglion input to the buccal ganglion is necessary for the generation of buccal ingestive and egestive behaviors in free-moving animals. We confirmed a prior study that showed that animals with lesions of the cerebro-buccal connectives (CBCs) do not exhibit rhythmic biting following seaweed stimulation of the lips, but do show swallowing of seaweed inserted into the buccal cavity. We found that CBC-lesioned animals also exhibited rejection of a tube inserted into the buccal cavity and esophagus. The programs for swallowing and rejection behaviors were similar to those observed before lesioning the CBCs, although the rate of swallowing was slower. These results suggest that the cerebral input to the buccal ganglion is necessary for generating biting responses, but is not required for producing swallowing or rejection responses.

The aim of the study was to understand: (i), how far anticipatory postural adjustments (APA) participate in the sequences of a complex task; (ii), how, in the sagittal plane, horizontal and vertical progressions of the center of gravity (CG) are managed to achieve the prescribed goal. A posturo-kinetics sequence is identified during which forward CG progression is controlled in terms of a center of pressure (CP)-CG interaction. CP shifts and horizontal CG accelerations are correlated in timing and amplitude. There exists a control of whole body progression during the impulse, implying a braking of forwards movement. Vertical and horizontal CG accelerations are inter-dependent, and backward CP shift duration is related to its forward displacement. During the vertical jump initiation, whole body vertical propulsion and forward fall are linked. To increase impulse amplitude, initial whole body disequilibrium is lengthened and forward CG acceleration is greater. APA programming is probably elaborated on the basis of cognitive evaluation of the antero-posterior disturbance.

We analysed the co-ordination of the elevation angles of the thigh (alpha(t)), shank (alpha(s)) and foot (alpha(f)) during walking in 19 adults and 21 children (aged 11--144 months), including the very first unsupported steps in four. Cross-correlation functions (CCF) maturation of pairs of elevation angles was quantified by a global error parameter (Et((CCF))) reflecting the difference between particular CCF value of toddlers and mean adult value (Ea((CCF))). During the very first step, Et((CCF)) could be five times higher than Ea((CCF)). With walking experience, Et((CCF)) for both alpha(t)-alpha(s) and alpha(s)-alpha(f) pairs evolved following a biexponential profile, with a fast time constant below 6 months. Adult-like CCF parameters were reached earlier for alpha(s)-alpha(f) than alpha(t)-alpha(s), indicating disto-proximal maturation of the temporal co-ordination of the lower limb segments in human locomotion.

Recent debate has focused upon the issue of whether general principles and laws of movement coordination may be derived without reference to anatomical, mechanical and physiological mechanisms. It has been proposed that self-generated movement involves the interaction of biomechanical and neuromuscular constraints. Biomechanical constraints are usually considered of as arising from the pendular dimensions of the limb or limb segments whereas neuromuscular constraints are commonly associated with nervous and metabolic control processes. The present study aims to investigate the interplay between these two different constraints on bimanual pattern stability and attentional demands. Five subjects were asked to execute an anti-phase coordination pattern (180 degrees of relative phase), while gradually increasing the frequency of oscillation and changing the rotational inertia of the joysticks. Frequency manipulation was expected to affect the neuromuscular constraints. Inertial manipulation was expected to affect biomechanical constraints. Attentional demands, reflecting the central cost associated with the maintenance of the coordination pattern was assessed using a dual-task paradigm. The results showed that: (1) increasing the oscillation frequency altered both coordination dynamics and attentional demands associated with the maintenance of bimanual coordination patterns; (2) manipulation of rotational inertia of the joysticks also altered pattern stability (standard deviation of relative phase) and coordination dynamics (i.e. the number of phase transition and the before transition), but these alterations were not paralleled by a change in attentional demands.

To examine whether the postural balance is influenced by the degree of anxiety, body sway during orthostatic standing while gazing at a visual target was examined in college students. Students, physically and mentally healthy, were divided into two groups according to the degree of state anxiety; high anxiety group (HA) and low anxiety group (LA). A fast Fourier transform analysis of the postural sway in antero-posterior axis showed that frequency components of 0.02-0.21 Hz, reflecting vestibular inputs, were 16% greater and those of 2.02-10.0 Hz, reflecting somatosensory inputs, were 24% smaller in HA. These differences between HA and LA were abolished when the eyes were closed. It is concluded that the interactions of visual inputs with vestibular and somatosensory inputs are influenced by anxiety.

The aim of this study was to develop protocols for computer imaging of the thoraco-abdominal ganglion of Drosophila from serial semi-thin sections, in which specific neurons were stained and related to neuropilar structures. The central projections of a subset of transgenically labelled sensory neurons were revealed by immunohistochemistry, while Nomarski optics were used to show motor neuron targets in the neuropil. Digital photomicrographs of each section were aligned and the resultant image stacks rendered into three-dimensional (3D) images that can be rotated in real time. The result is a detailed, in-depth visualization of labelled neurons at a resolution comparable with that in confocal reconstructions, which also allows investigation of their relationships with other components of the neuropil.

The hypothesis that during self-moved target tracking, the eye-arm co-ordination control system uses an internal model of the arm dynamics was tested. The contribution of arm proprioception to this model was also assessed. Subjects (nine healthy adults and one deafferented subject) were requested to make forearm movements and visually track an arm-driven target. Unexpected changes in mechanical properties of the manipulandum were used to modify the dynamical conditions of arm movement. The smooth pursuit gain (SPG) was computed before and during the perturbation. Results showed a decrease of SPG during perturbation in control subjects only. We propose that an internal model of the arm dynamics may be used to co-ordinate eye and arm movements, and arm proprioception may contribute to this internal model.

In cycling cells, microtubule assembly is initiated at the centrosome and requires the centrosomal protein gamma-tubulin. Previously, it was reported that gamma-tubulin is present at the centrosome of cervical ganglion cells undergoing axonal growth, but not in the axons or dendrites. We find that although gamma-tubulin is present at the centrosomes of neurons just beginning to extend processes, it is not associated with centrosomes in hypothalamic and cortical neurons on which functional synaptic connections have formed. In contrast, another centrosomal protein, pericentrin, is associated with the centrosome at all stages. These results suggest that centrosomal microtubule nucleation is required for early stages of neurogenesis to supply sufficient microtubule polymer to support rapid axonal growth, but is not required for maintenance of axonal microtubules in synaptically coupled neurons.

The effects of changes in gamma-aminobutyric acid (GABA) metabolism or inhibitory processes was studied in the perforant path-dentate gyrus synapses in rat cortico-hippocampal slices, and in the monosynaptic-reflex circuit in isolated newborn, rat spinal cord. GABA metabolism was modulated by pharmacological block of either the anabolic enzyme glutamate decarboxylase (GAD) or the catabolic enzyme GABA transaminase (GABA-T). The results support the notion that GABA concentration determines the efficacy of inhibition in these regions of the central nervous system (CNS).

MEF2D, a member of myocyte-specific enhancer binding factor 2 (MEF2) gene family, was shown by Northern blot hybridization to be strongly expressed in the head portion of mouse embryos at later stages of ontogenesis, in the cerebellum and the cerebrum of adult mice, in cultured cell lines of neuronal origin, and in skeletal and cardiac muscles. During ontogenesis, MEF2D transcripts were detected by in situ hybridization in the olfactory bulb, entorhinal cortex, pyriform cortex, and hippocampus, in Purkinje and granule cells, and in large neurons in both the ventral and dorsal horns of spinal cord. Adult mice continued to express MEF2D in these limited areas of the central nervous system. Thus, MEF2D seems to be involved in either the differentiation process or the function of these neurons.

Olfactory neuron recovery and axon growth was studied in 12-24 month old hamsters after unilateral olfactory nerve transection. At recovery times ranging from 4 to 126 days olfactory nerve regeneration and axon reconnection to the olfactory bulb were examined by anterograde horseradish peroxidase (HRP) neurohistochemistry and electron microscopy. Nerve transection produced immediate retrograde neuron degeneration and there was no HRP reaction product in the bulb at 4 days post transection. By day 35, centrally growing olfactory axons had reached the bulb. Axons formed glomeruli smaller than those in the unoperated control bulb and they were not always confined to the glomerular layer of the bulb. Some animals showed robust fiber growth with axon fascicles penetrating the different layers of the bulb and forming ectopic glomeruli along their path. Second order bulb neurons contained wheat germ agglutinin HRP reaction product, indicating that transneuronal transport had occurred. Electron microscopy confirmed transneuronal transport between olfactory axons and second order bulbar neurons. These results show that the capacity for olfactory neuron recovery and reconnection persists in the hamster well into old age.

Although several beta-carboline alkaloids display hallucinogenic properties in humans, their mechanism of action remains unclear. To ascertain their influence on central dopaminergic transmission, in this study we investigated the facilitating effect of low doses of various beta-carbolines on L-DOPA (250 mg/kg)-induced stereotypy in mice. Harmaline (0.075, 0.15 and 0.35 mg/kg) and harmine (0.15, 0.35 and 0.75 mg/kg) powerfully enhanced the degree of stereotypy, whereas 6-methoxy-harmalan, 6-methoxy-harman and harman were far less potent, augmenting stereotypy only at much higher doses (3 mg/kg). 6-Methoxy-tetrahydro-beta-carbolin (6-MeO-THBC) had only a weak effect at the dose of 3 mg/kg and tetrahydro-beta-carbolin (THBC) had no effect up to the dose of 3 mg/kg. The Ca(2+)-channel blocker nimodipine (2.5 mg/kg) only slightly antagonized harmaline (0.15 mg/kg) facilitation of L-DOPA-induced stereotypy.

We examined expression and production of interleukin-3 (IL-3) mRNA and IL-3 protein in mouse primary cultured neurons and glia by the reverse transcription and polymerase chain reaction and a bioassay using an IL-3-dependent cell line. IL-3 mRNA was demonstrated mainly in hippocampal neurons but not in glia, while a small but definite production of bioactive IL-3 was detected in septal and hippocampal neuronal cultures. Thus, endogenous IL-3 might be produced by certain neurons in situ.

Fictive locomotion was evoked by stimulation of the mesencephalic locomotor region (MLR) in immobilized, vagotomized and decerebrate cats. The coherence between heart beat fluctuation and efferent discharges of the hindlimb nerve was used to evaluate the strength of the coupling between the cardiac and locomotor rhythms during MLR-elicited fictive locomotion. This study demonstrated that there was a locomotor-cardiac coupling of central origin.

Pseudorabies virus was injected into the wall of the urinary bladder and, following incubation times of 2, 3 and 4 days, central nervous tissue was processed immunohistochemically for the presence of virus. Longer incubation times resulted in more extensive spread of the virus. Infected neurons were initially found in the spinal cord (mainly lumbosacral) and, after longer survival times, in raphe nuclei, reticular area, pontine micturition center, locus coeruleus, red nucleus, hypothalamus, preoptic, and cortical areas. These data define a multisynaptic circuit of neurons whose ultimate output influences urinary bladder function.

The pharmacology of excitatory amino acid receptors on an identified neurone in the insect central nervous system was investigated using single electrode recording techniques. Application of kainate, domoate and quisqualate elicited large, slow depolarizations with a rise time of approximately 4 min and recovery time of 23 min. Concentration-response curves were constructed giving an order of potency, domoate greater than quisqualate greater than kainate, and fitted curves demonstrated quisqualate to be a possible partial agonist, compared to domoate and kainate. Agonist responses were insensitive to the antagonists CNQX and picrotoxin and the vertebrate receptor-subtype selective agonists AMPA and trans-ACPD did not elicit any response, suggesting a novel type of excitatory amino acid receptor present on neurones in the insect central nervous system.

In the present paper we collected evidence for the occurrence of D2-like dopamine receptors on the cell bodies of the neuroendocrine growth hormone-producing cells (GHCs) in the central nervous system (CNS) of the snail Lymnaea stagnalis. Measurements of the membrane potential of GHCs in situ as well as isolated GHCs revealed that stimulation of these dopamine receptors results in a hyperpolarization. Although immunohistochemical analysis of the CNS of L. stagnalis clearly revealed the occurrence of dopamine containing cells and nerve fibers, no projections of dopamine immunopositive fibers to the GHC cell bodies could be observed. By using HPLC with electrochemical detection we found that the blood concentration of dopamine in L. stagnalis is in the range of concentrations hyperpolarizing GHCs in vitro (0.1-10 microM). On the basis of these findings it is proposed that dopamine is involved in hormonal communication in the CNS of L. stagnalis.

Extracellular unit recordings performed on 113 spontaneously discharging neurons in the substantia innominata-nucleus of the horizontal limb of the diagonal band region revealed that 46 (41%) were transsynaptically discharged or inhibited following stimulation of the main and accessory olfactory bulb (MOB, AOB) and the prepyriform cortex (PPC). A significantly greater number of cells were responsive to MOB and PPC than to AOB shocks. Eight (7%) neurons were antidromically invaded following MOB volleys. These findings suggest that the MOB has functional reciprocal connections with the 'ventral striatum', a region which can function, presumably as a nodal point for convergence of visual, gustatory and olfactory information relevant to feeding and drinking behavior.

The effect on the fusimotor system of an injection of the gamma-aminobutyric acid (GABA) antagonist picrotoxin into the reticular part of the substantia nigra (SNR) was studied in muscle spindle primary and secondary endings of pretibial flexors in cats anaesthetized with ketamine. Changes in fusimotor action after drug administration were deduced from changes in bias and sensitivity to sinusoidal stretching of the receptor-bearing muscles when compared to the deefferented preparation. Intranigral injection of picrotoxin (2 micrograms) removed a tonic static fusimotor action from flexor muscle spindle primary endings without affecting the secondary endings in the same muscles. It is concluded that static gamma motoneurones of cat flexors are of more than one functional type, and that the central nervous control of the static fusimotor input to cat flexor muscle spindles has selective access to static bag2 fibres and nuclear chain fibres.

In the present investigation it is shown that a reflex change in the heart activity of Protophormia flies is evoked by olfactory stimulation with several volatile substances, and particularly with those which are repellent for blowflies. Among these, i-pentanal and hexanal vapours evoked a fast, highly persistent cardiac response, whereas in the case of hexanol and ammonia vapours the response resulted slower and could be suppressed as a function of repeated stimulation.

Chemosensory systems in the mammalian nasal cavity are said to be functional before birth. Fluorescent beads (1 micron, diameter), injected into the amniotic fluid surrounding E18 mouse fetuses, were sought in serial frozen sections of E19 animals. Beads were found in all regions of the nasal cavity except for the lumen of the vomeronasal organ (VNO) where the receptor neurons of the accessory olfactory system (AOS) are located. Thin plastic sections through the rostral aspect of the E19 nasal cavity revealed that the VNO canals, which provide stimulus access to the AOS, are not patent. Thus, this chemosensory system can be excluded as a potential channel of chemosensory information for the mouse fetus.

Following injections of horseradish peroxidase into the mediodorsal thalamic nucleus (MD), retrogradely labeled cells were found in various areas in the cat and rabbit. Among these, olfactory-related areas to which the olfactory bulb projects directly or indirectly via the piriform cortex were the olfactory tubercle, amygdala and insular cortex, while no labeled cells were detected in the piriform cortex and endopiriform nucleus in both species. These results indicate that the piriform cortex and endopiriform nucleus do not send their axons directly to the MD.

Recordings from the intact nervus terminalis with a hook electrode or from a stump of the divided nerve with a suction electrode show a tonic, irregular discharge of broad, low frequency spikes in ca. 4-6 units. These nerve impulses are efferent from the brain. The mean frequency of discharge is not influenced by various chemical, thermal, tactile, acoustic, photic, vibratory and electric field stimuli but is decreased by certain forms of mechanical stimuli, presumably acting on the lateral line organs of the lateral aspect of the head. We have not succeeded in recording from afferents. The nerve consists of greater than 1000 unmyelinated axons, mostly less than 1 micron, a very few greater than 1.5 micron in diameter; presumably the efferents recorded from were these larger fibers.

Neurotrophic effects (NTEs) of various brain regions of 4-week-old rats were examined in primary culture of rat embryonic cerebral hemispheres. Extracts of the hippocampus, brainstem and septal nucleus highly enhanced the survivability of neuronal cells and the division of non-neuronal cells by 9 days. The septohippocampal tract (fimbria fornix) was cut and the effect on the neurotrophic activity in the hippocampus was examined. The NTEs of hippocampal extracts remained unchanged 3 days after septal deafferentation, was significantly increased by 7 days, peaked at 14 days and returned to the basal level by 21 days.

Rats with one olfactory bulb removed when neonates and the second bulb removed when adults were tested on tone-light discrimination and odor detection tasks. On the neonatally operated side reconstituted olfactory receptor cell axons penetrated the frontal neocortex or portions of the anterior olfactory nucleus, and formed glomerular-like structures. On the adult operated side there was extensive scar formation which prevented in-growing sensory axons from contacting the brain. All experimental animals acquired the tone-light discrimination but failed to show any evidence of odor detection. These results indicate that reconstituted olfactory projections which terminate in the frontal neocortex or anterior olfactory nucleus do not support olfaction.

A method is presented allowing direct and continuous estimation of the excitability changes of single fiber terminal arborizations within the central nervous system. In essence, the method measures the current required to maintain a preset antidromic firing probability of the unit under study. This implies operation in a closed loop system controlled by a computer. With this technique one can accurately determine input-output curves of single units requiring relatively small current variations (0.15-2.1 microA) to change from zero to maximal probability of response. The method also allows measurement of the excitability changes produced by conditioning volleys to sensory nerves.

The possibility that axon terminals of the nervus terminalis in the goldfish retina regulate visual sensitivity was examined psychophysically. Fish were classically conditioned to respond in darkness to a diffuse red light conditioned stimulus. Bilateral ablation of the olfactory bulb and telencephalon had no significant effect on response threshold which was measured by a staircase method. Retinopetal nervus terminalis fibres thus appear to play no role in maintaining scotopic photosensitivity.

Stingrays swim with an active elevation-depression sequence of the pectoral fin resembling an extension-flexion sequence. During forward locomotion this sequence passes caudally along the pectoral fin. Immediately following high decerebration, stingrays are capable of locomotion, and the pattern of muscle activity closely resembles that of intact animals. Spontaneous and midbrain evoked rhythmic motoneuron discharges can be recorded in paralyzed decerebrated animals. In contrast to dogfish sharks, stringrays with high spinal transections do not locomote.

The effects of feeding or of exposure to food odors (without opportunity to feed) on dopamine metabolism in the prefrontal cortex have been investigated in rats food deprived for 24 h. Dopamine metabolism was assessed by measuring extracellular DOPAC levels by in vivo voltammetry with carbon fiber electrodes. Feeding in fasted rats was accompanied by an increase in extracellular cortical DOPAC levels which gradually returned to basal levels within 90 min after the onset of meal. A similar, though slightly less pronounced, increase in cortical dopamine metabolism was found in fasted rats exposed to food odors but not allowed to feed. These results indicate that feeding or the olfactory stimulation associated with food presentation causes an increase in mesocortical dopaminergic neuron activity.

Abstract The central nervous system preplans postural responses to successfully perform complex multi-joint movements. These responses have been termed anticipatory postural adjustments (APAs), and they constitute a general type of response to stabilize posture prior to movement initiation. APA sequences are elicited with shorter latency when a startling acoustic stimulus is applied, demonstrating their preplanned nature. Increasing task complexity using a simple reaction time (RT) paradigm has been shown to delay limb movement RT as a result of additional planning or sequencing requirements; however, the effect of task complexity on APA dynamics is unclear. The purpose of the present study was to investigate if task complexity modulates APA onset in a manner analogous to that observed in the primary effector. 13 participants completed 150 trials of simple (1-target) and complex (2- or 3-target) arm movements while standing on a force plate. Results indicated participants had significantly faster arm movement RTs in the simple versus the most complex condition. Similar to the primary effector, APA RTs were longer in the most complex (3-target) movement compared to both the 1-target and 2-target movements. Furthermore, APA excursion velocities were scaled to the complexity of the upcoming movement: the rate of APAs increased from simplest to most complex movements. These findings clearly demonstrate APAs are sensitive to task complexity, further elucidating their preplanned role in stabilizing posture which enables the successful completion of intended movements. The myelin sheath that insulates some axons in the central nervous system allows for faster signal conduction. Previously, axons were thought to be either unmyelinated or fully myelinated. Recent experimental work has discovered a new pattern of myelination (intermittent myelination) along axons in the mouse brain, in which long unmyelinated axon segments are followed by myelinated segments of comparable length. We use a computational model to explore how myelin distribution (in particular intermittent myelination) affects conduction velocity. We find that although fully myelinated axons minimize conduction velocity, varying the spatial distribution of a fixed amount of myelin along a partially myelinated axon leads to considerable variation in the conduction velocity for action potentials. Whether sodium ion channel number or sodium ion channel density is held constant as the area of the unmyelinated segments increases has a strong influence on the optimal pattern of myelin and the conduction velocity.
