Abstract

Membrane proteins that transport hydrophobic compounds have important roles in multi-drug resistance (1-3) and can cause a number of diseases (4,5), underscoring the importance of protein-mediated transport of hydrophobic compounds. Hydrophobic compounds readily partition into regular membrane lipid bilayers (6), and their transport through an aqueous protein channel is energetically unfavourable (3). Alternative transport models involving acquisition from the lipid bilayer by lateral diffusion have been proposed for hydrophobic substrates (3,4,7-12). So far, all transport proteins for which a lateral diffusion mechanism has been proposed function as efflux pumps. Here we present the first example of a lateral diffusion mechanism for the uptake of hydrophobic substrates by the Escherichia coli outer membrane long-chain fatty acid transporter FadL. A FadL mutant in which a lateral opening in the barrel wall is constricted, but which is otherwise structurally identical to wild-type FadL, does not transport substrates. A crystal structure of FadL from Pseudomonas aeruginosa shows that the opening in the wall of the β-barrel is conserved and delineates a long, hydrophobic tunnel that could mediate substrate passage from the extracellular environment, through the polar lipopolysaccharide layer and, by means of the lateral opening in the barrel wall, into the lipid bilayer from where the substrate can diffuse into the periplasm. Abstract

The development of selective ubiquitin-specific protease-7 (USP7) inhibitors GNE-6640 and GNE-6776, which induce tumour cell death and reveal differential kinetics of Lys-48 and Lys-63-linked ubiquitin chain depolymerization by USP7. Deubiquitinating enzymes remove the small modifier protein ubiquitin from target substrates regulating their stability. One such enzyme, USP7, is a potential target for anti-cancer therapy, as its inhibition would result in the degradation of the ubiquitinated oncoprotein MDM2, leading to reactivation of the tumour suppressor protein p53. However, selective inhibitors of USP7 have remained elusive. Here, Ingrid Wertz and team develop two USP7 inhibitors, providing structural insights into the mode of action of these compounds and demonstrating their toxicity towards tumour cells. Elsewhere in this issue, David Komander and colleagues independently report the identification of two small molecules that inhibit USP7 with high affinity and specificity both in vitro and within cells, also demonstrating their ability to inhibit tumour growth. The ubiquitin system regulates essential cellular processes in eukaryotes. Ubiquitin is ligated to substrate proteins as monomers or chains and the topology of ubiquitin modifications regulates substrate interactions with specific proteins. Thus ubiquitination directs a variety of substrate fates including proteasomal degradation1. Deubiquitinase enzymes cleave ubiquitin from substrates and are implicated in disease2; for example, ubiquitin-specific protease-7 (USP7) regulates stability of the p53 tumour suppressor and other proteins critical for tumour cell survival3. However, developing selective deubiquitinase inhibitors has been challenging4 and no co-crystal structures have been solved with small-molecule inhibitors. Here, using nuclear magnetic resonance-based screening and structure-based design, we describe the development of selective USP7 inhibitors GNE-6640 and GNE-6776. These compounds induce tumour cell death and enhance cytotoxicity with chemotherapeutic agents and targeted compounds, including PIM kinase inhibitors. Structural studies reveal that GNE-6640 and GNE-6776 non-covalently target USP7 12 A distant from the catalytic cysteine. The compounds attenuate ubiquitin binding and thus inhibit USP7 deubiquitinase activity. GNE-6640 and GNE-6776 interact with acidic residues that mediate hydrogen-bond interactions with the ubiquitin Lys48 side chain5, suggesting that USP7 preferentially interacts with and cleaves ubiquitin moieties that have free Lys48 side chains. We investigated this idea by engineering di-ubiquitin chains containing differential proximal and distal isotopic labels and measuring USP7 binding by nuclear magnetic resonance. This preferential binding protracted the depolymerization kinetics of Lys48-linked ubiquitin chains relative to Lys63-linked chains. In summary, engineering compounds that inhibit USP7 activity by attenuating ubiquitin binding suggests opportunities for developing other deubiquitinase inhibitors and may be a strategy more broadly applicable to inhibiting proteins that require ubiquitin binding for full functional activity.

Abstract

The data from a recently completed survey of the galactic plane with the SAS-3 modulation collimators provide precise (20 to 60 arcsec) celestial positions of galactic X-ray sources. Preliminary positions of 60-arcsec precision are reported for five sources. One of these led to the identification of the star, Gamma Cas, as an X-ray source, and the others lend substantial confidence to previously proposed optical identifications: 3U 0352+30 = X Per, 3U 1145-61 = HEN 715, GX301-2 = WRA977, and GX304-1 = MMV star. These identifications seem to establish the existence of a previously suggested class of Be-star X-ray emitters.

Abstract

The CD4 T-lymphocyte differentiation antigen is an essential component of the cell surface receptor for human immunodeficiency viruses (HIVs) causing AIDS (acquired immune deficiency syndrome) (refs 1–3). Peripheral blood lymphocytes of apes, New World and Old World monkeys express cell surface antigens homologous to CD4 of human T-helper lymphocytes. The cells of several of these species can be infected in short term culture with diverse strains of the type-1 or type-2 human immunodeficiency viruses (HIV-1 and HIV-2). HIV-1 is the prototype AIDS virus, and HIV-2 is the second type of AIDS virus, prevalent in West Africa4. Infection of the primate cells correlates with evolutionary conservation on CD4 of one particular epitope cluster, and is inhibited by treatment of the cells with monoclonal antibodies to this epitope. The capacity of HIV to replicate in simian cells may provide a means for evaluating antiviral drugs and vaccines.

Abstract

HERE I report some new and unexpected findings on the role of the pituitary in the reproductive cycle of the tammar wallaby, Macropus eugenii. My results suggest a pituitary suppression of the corpus luteum during the seasonal embryonic diapause found in this species. They may have some relevance to the hormonal control of the analogous seasonal delayed implantation of eutherian species such as the badger, Meles meles1; the roe deer, Capreolus capreolus2; and the black bear, Ursus americanus3.

Abstract

IN a previous study1 of the response of the ionospheric total electron content (TEC) data to magnetic storms, an average storm time behaviour of TEC for middle latitudes was deduced. On average TEC is enhanced during the first 24 h following the start of a storm, depressed for the next 24 h and near normal through the third 24 h. Another study2 has shown a strong diurnal component in TEC behaviour with the maximum enhancement occurring in the dusk sector (1600 to 1800 LT). The present study of disturbed TEC behaviour generally agrees with that reported above but in addition indicates a longitudinal effect in the behaviour of both the storm time and diurnal components of disturbed TEC.

As their name implies, prostaglandins were first identified in secretions of the male reproductive tract. In the early 1930s several workers1–3 showed that human semen could stimulate various smooth muscle preparations, and this activity was ascribed to an unidentified component which von Euler named ‘prostaglandin’. von Euler4,5 then found that a watery alcoholic extract of rhesus monkey seminal vesicles markedly lowered the blood pressure in the rabbit but, unlike human seminal vesicle extracts, it had little or no action on the isolated intestine and the uterus of the rat and other laboratory animals. He therefore postulated the existence of another substance in the monkey which he named ‘vesiglandin’ and which he suggested could be a component of human seminal ‘prostaglandin’.

WE report here work in progress on the measurement of the solubility of metals and metal oxides in pure water, in the temperature range 50°–550° C and at pressures up to 400 bars. The isobaric solubility of cupric oxide at 310 bars is reported here and compared with the 300 bar isobar for silica (quartz).
