In this work, the phytochemical analysis of Teucrium chamaedrys L. collected in Italy was reported. Eight compounds were isolated and identified by means of classical column chromatography and spectroscopic techniques, such as NMR and MS. In detail, these compounds were: verbascoside (1), forsythoside b (2), samioside (3), alyssonoside (4), harpagide (5), 8-O-acetyl-harpagide (6), cirsiliol (7) and β-arbutin (8). The presence of these compounds, in particular iridoids and phenyl-ethanoid glycosides, has a chemotaxonomic relevance and results to be in perfect accordance with the current botanical classification of the species. In addition, it provides a phytochemical rationale for the use of this particular plant in the ethno-pharmacological field. Conversely, it is worth of mention the absence of potentially toxic components, unlike to what observed in other species of the genus which can no longer be used for ethno-medicinal purposes.

Since its first isolation in 1844, usnic acid [2,6-diacetyl-7,9-dihydroxy-8,9b-dimethyl-1,3(2H,9bH)-dibenzo-furandione] has become the most extensively studied lichen metabolite and one of the few that are commercially available. Lichens belonging to usnic acid-containing genera have been used as crude drugs throughout the world. There are indications of usnic acid being a potentially interesting candidate for such activities as anti-inflammatory, analgesic, healing, antioxidant, antimicrobial, antiprotozoal, antiviral, larvicidal and UV protection. However, some studies reported the liver toxicity and contact allergy. Thus, further studies are needed to establish the efficacy and safety of usnic acid.

In this work, we report the study on the composition in primary and secondary metabolites of the polar and less-polar fractions obtained from the fruiting bodies of Suillus bellinii (Inzenga) Kuntze collected from Central Italy. Thirteen compounds were identified through a series of classical chromatographic methods and through spectroscopic techniques (nuclear magnetic resonance (NMR) and mass spectrometry (MS)), namely suillin (1), ergosterol (2), glutamic acid (3), isoleucine (4), leucine (5), threonine (6), tyrosine (7), valine (8), D-mannitol (9), sucrose (10), D-sorbitol (11), fumaric acid (12) and alanine (13). These results allowed us to make specific considerations about the possible applications of this mushroom, which vary from medicine, due to the high cytotoxic and antitumor properties of suillin (1), to nutritional values, due to the presence of essential nutrients as amino acids and pre-vitamin D2 (ergosterol), together with low-calories sweeteners, confirming the close association between the identified components and the health-promoting properties of this species.

The redox properties of the hydroethanol extract (EE) and its ethyl acetate (EAF) and hydromethanol (HMF) fractions obtained from Abarema cochliacarpos (Gomes) Barneby & Grimes stem bark were evaluated. EAF had the highest total phenol content (848.62 ± 78.18 mg g−1), while EE showed the highest content of catechin (71.2 µg g−1). EE, EAF and HMF exhibited the highest levels of antioxidant activity at 100 and 1000 µg mL−1 when the non-enzymatic antioxidant potential was evaluated by the total reactive antioxidant potential, total antioxidant reactivity and nitric oxide scavenging assays. In addition, EAF and HMF showed SOD-like activity. The results for EE, EAF and HMF in this study showed that A. cochliacarpos (Gomes) Barneby & Grimes stem bark have redox properties and may be able to help the endogenous enzymatic and non-enzymatic systems to keep the redox balance.

The phytochemical analysis of the extracts of Linaria vulgaris, has allowed to underline an iridoidic pattern similar to that of the other Linaria plants, with the presence of antirrinoside, antirride, 6-beta-idrossiantirride, 10-beta-glucosilaucubina and a new iridoidic compound, whose structure was demonstrated to be that of 4-carboxy-boonein.

The phenyl propanoid fraction of Teucrium subspinosum Pourrette ex Willd. (Lamiaceae) from Baleari Island, was examined together with the same fraction of T. subspinosum from Sardinia. In T. subspinosum, besides verbascoside as main component, a new phenyl-propanoid-glycoside was isolated and identified by (1)H- and (13)C-NMR spectra analysis. Comparison with molecular composition of T. marum confirmed the differences between these species and evidenced their endemic character.

In this study, we report the first phytochemical analysis of polar fraction of Pedicularis kerneri Dalla Torre growing in Dolomites, Italy. Several iridoid glucosides were isolated, namely aucubin (1), monomelittoside (2), plantarenaloside (3), euphroside (4), mussaenosidic acid (5) and 8-epiloganic acid (6), showing a composition in accordance with previous study on this genus. The studied samples, collected from Dolomites, presented a chemotype already recognised in species from North America, characterised by euphroside (4) and aucubin (1) as main components, but the main character was the presence of monomelittoside (2) never reported in this genus. The identification of verbascoside (7), leucosceptoside A (9) and echinacoside (10) complete the systematic framing of this species since is ascertained the co-occurrence of phenylethanoid glycosides with iridoids in Lamiales species.

The monoterpenoid composition of Verbascum conocarpum and Scrophularia trifoliata, both endemic plants of Sardinia, was examined. The main chemotaxonomic markers of Scrophulariaceae, the iridoids aucubin, verbascoside and catalpol, were isolated.

Teucrium marum and T. subspinosum have been extracted with different polarity solvents, each crude extract was analysed with (1)H-NMR to compare the overall metabolic component. The polar extracts showed antioxidant activity in vitro. Verbascoside and arabinosyl-verbascoside were isolated and identified. Their concentrations were in connection to the antioxidant activity.

The phytochemical analysis of iridoidic fraction of the aerial parts of Vinca sardoa and Vinca difformis, gave the identification of loganic acid in V. difformis as unique iridoid glucoside, while loganin is also present in V. sardoa. The two taxa highly differ in alkaloids, so the chemotaxonomic classification in two different species is supported.

The phytochemical study of aerial parts of Viburnum veitchii C.H. Wright, a deciduous species originary of Central China, led to the isolation of three salicin derivative bis-glucosides, i.e. henryoside (1), and the new structures 2′b-acetyl-3′b-(3-methylbutyryl)-henryoside (2) and 2′b,6′b-diacetyl-3′b-(3-methylbutyryl)-henryoside (3). In addition, luteolin, lonicerin, robustaflavone and eugenyl-β-D-glucopyranoside were also isolated.

Glaucium flavum collected in Sardinia was studied using a phytochemical approach in order to evaluate its alkaloid composition and obtain a comparison with the alkaloid contents of the same species in populations of other geographic proveniences. In fact, different chemoecotypes of G. flavum have been identified, on the basis of their particular content and composition in alkaloids, in accordance with the different distribution areas. The analysis showed that Sardinian G. flavum contains a homogeneous alkaloid pattern of aporphyne type, significantly different from those reported for populations from other parts of Europe.

The phytochemical study of Euphrasia genargentea, a rare species only present in Sardinia, led to the identification of iridoid glucosides, i.e. aucubin, catalpol, mussaenosidic acid and melampyroside, which allowed chemotaxonomic considerations on the genus. On the basis of iridoid distribution in the genus, E. genargentea does not show any particular analogy with other Italian Euphrasia spp. This study is also important considering the severe risk of extinction of E. genargentea.

The monoterpenoid fractions of three endemic Galium ssp. (Rubiaceae) from Sardinia Island were examined and compared with the iridoidic pattern yet known in Galium species. This comparison evidenced theirs endemic characters. In particular, in G.corsicum and in G. glaucophyllum loganic acid was isolated and identified for the first time in Galium genus. In G. schmidii a rare iridoid is present, 10-hydroxy-loganin, whose presence in this genus was evidenced only in G. mollugo and loganin isolated for the first time.
