Sarcopenia is having a serious impact on highly aged populations. A Japanese traditional medicinal remedy containing Schisandra fruits and Astragalus roots recently showed effective treatment of sarcopenia and thus needed to be investigated further. This current study selected Schisandra cauliflora fruits for investigation of their chemical constituents and evaluation of their antisarcopenia effects. Dried S cauliflora fruits were ultrasonically extracted with methylene chloride. This extract was purified by a combination of chromatographic techniques. The isolated compounds were characterized by nuclear magnetic resonance spectroscopy, electronic circular dichroism, and high resolution electrospray ionization mass spectrometry methods. Antisarcopenia activities of the isolated compounds were evaluated by their effects on the viability and proliferation of C2C12 skeletal muscle cells. Seven dibenzocyclooctadiene lignans, including 2 that had been previously undescribed, cauliflorins A and B (1 and 2), ananolignan C (3), 6- epi-ananolignan D (4), ananolignan F (5), ananonin H (6), and ananonin J (7) were isolated from the methylene chloride extract of the fruits of S cauliflora. The C2C12 cells treated with compounds 1-7 (5 µM) exhibited proliferation in the range of 75.7% ± 7.4% to 98.2% ± 11.7%, compared to 100.0% ± 6.0% of the untreated cells. Two previously undescribed and 5 known dibenzocyclooctadiene lignans were isolated from S cauliflora fruits. All the isolated compounds were inactive on the proliferative enhancement of C2C12 skeletal muscle cells.

Seven phenolic compounds, including a new phenylethanoid, named passifoside (1), and 6 previously described compounds, syringaresinol (2), berchemol (3), threo-guaiacylglycerol (4), p-hydroxybenzaldehyde (5), 3,4,5-trimethoxyphenyl- O- β-D-glucopyranoside (6), and trans- p-coumaric acid (7), were identified from the aerial parts of Passiflora foetida. Their chemical structures were elucidated from HR-ESI-MS and NMR spectral data, and comparison with the literature. Compound 2 inhibited nitric oxide production in lipopolysaccharide-activated RAW264.7 cells, showing an IC50 value of 9.45 ± 0.33 µM. The other compounds showed either weak or no significant activity compared to L-NMMA, which was used as a positive control.

Phytochemical study of the methanolic extract of the stems and leaves of Strychnos vanprukii led to the isolation of three new (1-3) and four known (4-7) compounds. Their chemical structures were elucidated to be (+)-lyoniresinol 3 α-O- β-D-allopyranoside (1), (-)-lyoniresinol 3 α-O- β-D-allopyranoside (2), 2 α-[(3,5-dimethoxy-4-hydroxy)-benzoyl]-(-)-lyoniresinol-3 α-O- β-D-glucopyranoside (3), (+)-lyoniresinol 3 α-O- β-D-glucopyranoside (4), (-)-lyoniresinol 3 α-O- β-D-glucopyranoside (5), palicoside (6), and rutin (7) by means of detailed HR-ESI-MS, 1D and 2D NMR spectroscopic analysis. In addition, the absolute configurations of 1–3 were determined by experimental Circular Dichroism (CD) spectra. This is the first report of compounds 5 and 7 from S. vanprukii. Compounds 1 - 7 showed inhibitory effects on NO production in LPS stimulated RAW 264.7 cells with IC50 values of 58.50 ± 2.30, 67.70 ± 2.23, 54.44 ± 1.26, 71.58 ± 3.59, 74.47 ± 2.61, 83.62 ± 2.25, and 47.57 ± 4.01 µM, respectively, compared to a value of 43.24 ± 2.41 µM for the positive control compound, NG-monomethyl-L-arginine (L-NMMA).

A new sterol (1) and 8 known (2-9) compounds were isolated from the methanol extract of the Vietnamese marine sponge Halichondria panicea (Pallas, 1766) by various chromatographic methods. Their chemical structures were determined as 5 α,6 α-epoxy-(22 E)-ergosta-3 β,7 α-diol-8,22,25-triene (1), (22 E)-ergosta-7,22,25-triene-3 β,5 α,6 β-triol (2), 3 β-hydroxycholest-5ene-7-one (3), cholesterol sulfate (4), 3-formamidotheonellin (5), para-hydroxybenzylideneacetone (6), indole-3-carbaldehyde (7), thymidine (8), and adenosine (9) by using a combination of HR-ESI-MS, 1D and 2D NMR spectral data, as well as by comparison with the previous literature. This is the first report of these compounds from H panicea. Compounds 1 and 2 exhibited cytotoxic effects on Hep-G2, SK-Mel-2, LU-1, and MCF-7 cell lines with IC50 values of 9.27 ± 1.25, 11.34 ± 1.18, 15.45 ± 1.47, and 10.85 ± 1.19 µM (for 1), and 6.45 ± 0.77, 9.74 ± 1.03, 7.08 ± 0.64, 5.61 ± 0.39 µM (for 2), compared to the positive control compound, ellipticine, 1.60 ± 0.37, 2.53 ± 0.51, 1.82 ± 0.20, and 2.17 ± 0.66 µM, respectively.

Four new acyclic diterpenes, siegetalises A-D (1-4), were isolated from the aerial parts of Siegesbeckia orientalis. Their chemical structures were elucidated by extensive analysis of high-resolution electrospray ionization mass spectrometry and nuclear magnetic resonance spectral data. The effects of the isolated compounds on the activity of xanthine oxidase were evaluated by the oxidative reaction with xanthine as a substrate. At a concentration of 50 µM, compounds 1-4 exhibited xanthine oxidase inhibitory activity at levels of 13.59% ± 0.51%, 19.64% ± 1.54%, 17.45% ± 1.26%, and 21.36% ± 1.40%, respectively.

Five conjugated polyene ketones (1-5) were isolated from the methanol extract of the marine sponge Clathria ( Thalysias) reinwardti (Vosmaer, 1880) living in the coastal waters of Vietnam. Their structures were determined to be 8-(2′,3′,4′-trimethylphenyl)-6-methyl-oct-3( E),5( E),7( E)-trien-2-one (1), 13-apoastaxanthinone (2), 9-apoastaxanthinone (3), 2,3-dehydro-4-oxo- β-ionone (4), and 4-(2′,3′,4′-trimethylphenyl)-but-3( E)-en-2-one (5), by extensive analysis of high-resolution electron spray ionization mass spectrum (HR-ESI-MS), one-dimensional, and two-dimensional (2D) nuclear magnetic resonance (NMR) spectra, as well as by comparison of the spectral data with those reported in the literature. Compound 1 was new, compounds 2 to 4 were isolated from nature for the first time, and the chemical structure as well as the NMR assignments, of 5 were indicated by 2D NMR for the first time. Additionally, compound 5 exhibited cytotoxic activity against the human cancer cells SK-LU-1, SK-Mel-2, MCF-7, and Hep-G2 with half-minimal inhibitory concentration (IC50) values of 15.12 ± 3.43, 17.41 ± 2.83, 33.12 ± 3.39, and 34.38 ± 3.52 µM, respectively, but displayed only a weak cytotoxic effect on the normal HEK-239A cells (IC50 64.67 ± 3.67 µM). Compound 5 also significantly increased Caspase-3 activity in SK-LU-1 cells at concentrations of 10, 15, and 20 µM.

A total of 4 thiazinedione derivatives including 1 new thiazinedione glycoside (1) and 3 known compounds (2-4) were isolated from the fruits of Xanthium strumarium L. Their chemical structures were determined as 7-hydroxymethyl-8,8-dimethyl-4,8-dihydrobenzo[1,4]-thiazine-3,5-dione-11- O-[ β-D-glucopyranosyl-(1→6)- O-β-D-glucopyranoside] (1), 7-hydroxymethyl-8,8-dimethyl-4,8-dihydrobenzo[1,4]-thiazine-3,5-dione-11- O-[ β-D-apiofuranosyl-(1→6)- O- β-D-glucopyranoside] (2), xanthiside (3), and xanthiazone (4) by extensive nuclear magnetic resonance spectroscopic and high-resolution electron spray ionization mass spectrum analysis and by comparison of the spectral data with those reported in the literature. Compounds 3 and 4 exhibited cytotoxic activity against lung carcinoma (SK-LU-1), human breast carcinoma (MCF-7), hepatocellular carcinoma (HepG2), and skin melanoma (SK-Mel-2) cell lines with half-maximal inhibitory concentration (IC50) values ranging from 27.0 ± 1.1 to 43.2 ± 1.8 µM.

Seven compounds (1-7) were isolated from the marine sponge Aaptos aaptos living in the Vietnamese sea. Their structures were determined as 2 hours, 5 H,7 H,9 H-9 S-hydroxy-imidazo[1,5- α]pyridine-1,3-dione (1), 3-([9-methylhexadecyl]oxy)propane-1,2-diol 2, 2,3-dihydro-2,3-dioxoaaptamine (3), indol-3-aldehyde (4), methyl indole-3-carboxylate (5) 4-hydroxy-5-(indole-3-yl)−5-oxo-pentan-2-one (6), and thymidine (7) by extensive analysis of HR-ESI-MS, 1D, and 2D NMR spectral data, as well as by comparison of the spectral data with those reported in the literature. In addition, the absolute configuration of 1 was determined from the experimental ECD spectrum and comparison of this with the theoretical ECD calculations using the TDDFT method. Compounds 1 and 2 were isolated from nature for the first time. Compound 3 induced cytotoxic activity against SK-LU-1, MCF-7, HepG2, and SK-Mel-2 cell lines with IC50 values of 41.27 ± 2.63, 40.70 ± 2.65, 34.31 ± 3.43, and 36.63 ± 1.40 µM, respectively.

Two new acylated sucroses, (6- O-axetoxyl)- β-d-fructofuranosyl-(2→1)-(6- O-feruloyl- α- d-glucopyranoside (1) and (6- O-axetoxyl)- β-d-fructofuranosyl-(2→1)-(6- O-( E)- p-coumaroyl- α- d-glucopyranoside (2), and four known compounds, 2-(3′,4′-dihydroxyphenyl)-1-propanol-4′- O-[4′′′-hydroxy-3′′′,5′′′-dimethoxybenzoyl-(→6′′)- β-d-fructofuranoside (3), tryptophol glucoside (4), corchonoside C (5), and 2-hydroxy-5-(2-hydroxyethyl)phenyl β-d-fructofuranoside (6), were isolated from the roots of Canna indica L. (Cannaceae). Their structures were determined by extensive analysis of HR-ESI-MS, 1D, and 2D NMR spectral data, as well as by comparison of these with those reported in the literature. Antioxidant activity of compounds 1-6 were evaluated by peroxyl radical absorbance capacity assay. Compounds 1 and 3 neutralized high amounts of peroxyl radical. At a concentration of 1 µM, their ORACROO* values were 3.07 ± 0.15 and 4.27 ± 0.30, respectively, many times greater than that of trolox, which was used as an internal standard. At 10 µM, the peroxyl radical absorbance capacity of compounds 1 and 3 exhibited equivalents to 15.60 ± 0.22 and 24.91 ± 0.43 times the net protection by 1 µM of trolox.

Using various chromatographic methods, two new alkaloids, antidesoic acids A (1) and B (2) along with fourteen known compounds (3-16) were isolated from the leaves of Antidesma ghaesembilla Gaertn. Their chemical structures were elucidated by physical and chemical methods. All the isolated compounds were evaluated for their inhibitory activity on LPS-stimulated nitric oxide (NO) production in BV2 cells and RAW 264.7 macrophages. Bisflavone 8 significantly inhibited LPS-stimulated NO production in BV2 cells and RAW 264.7 macrophages with IC50 values of 5.4 and 8.0 μM, respectively. Compounds 1–3, 7, 10, 12, 14, and 16 showed moderate inhibitory activities with IC50 values ranging from 11.7 to 77.4 μM.

Two new steroidal alkaloid saponins, solanigrosides Q and R (1 and 2), together with four known compounds khasianine (3), solamargine (4), solasonine (5), and soladulcoside A (6) were isolated from the whole plants of Solanum nigrum. The structures of new compounds (1 and 2) were determined by interpretation of HR-ESI-MS, 1D and 2D NMR spectra, and chemical transformation. Compound 4 exhibited potent cytotoxic activity against MDA-MB-231, A549, Hep3B, PC3 human cancer cell lines with IC50 values of 1.86 ± 0.45, 2.24 ± 0.15, 0.78 ± 0.11, 5.13 ± 1.34 μM, respectively.

Using various chromatographic methods, one new lanostane triterpene, 3 α-hydroxylanosta-8,24 E-dien-26-oic acid (1), one new lignan, matairesinol 4'- O-β-D-glucopyranoside (2) along with five known compounds, ecdysone (3), 20-hydroxyecdysone (4), 20-hydroxyecdysone 2,3-monoacetonide (5), turkesterone (6), and polypodine B (7) were isolated from the leaves of Vitex trifolia L. Their structures were elucidated by 1D-, 2D-NMR spectroscopic analysis, HR-ESI-MS, CD, and by comparing with the NMR data reported in the literature.

One new tirucallane- type triterpene glycoside, antidesoside (1), along with two biflavones, podocarpusflavone A (2) and amentoflavone (3) and two megastigmane glycosides, byzantionoside B (4), and (6 S,9 R)-roseoside (5) were isolated from the methanol extract of Antidesma bunius leaves. Their structures were determined by spectroscopic methods and in comparison with the published data. Compounds 1 - 3 were found to show strong inhibitory effect of NO production in BV2 cells and RAW264.7 macrophages LPS-stimulated, with IC50 values ranging from 8.5 to 26.9 μM.
