Theranostic sutures are derived from innovative ideas to enhance wound healing results by adding wound diagnostics and therapeutics to typical sutures by functionalizing them with additional materials. Here, we present a new direct electrospinning method for the fast, continuous, inexpensive, and high-throughput production of versatile nanofibrous-coated suture threads, with precise control over various essential microstructural and physical characteristics. The thickness of the coating layer and the alignment of nanofibers with the thread's direction can be adjusted by the user by varying the spooling speed and the displacement between the spinneret needle and thread. To show the flexibility of our method for a range of different materials selected, gelatin, polycaprolactone, silk fibroin, and PEDOT:PSS (poly(3,4-ethylene dioxythiophene):poly(styrene sulfonate)) were the resultant nanofibers characterized by scanning electron microscopy (SEM) imaging and conductivity tests. In a series of in vitro and ex vivo tests (pig skin), sutures were successfully tested for their flexibility and mechanical properties when used as weaving and knotting sutures, and their biocompatibility with a keratinocyte cell line. For temperature-based drug-releasing tests, two fluorescent molecules as drug models with high and low molecular weight, namely fluorescein isothiocyanate-dextran (20 kDa) and rhodamine B (470 Da), were used, and their steady release with incremental increase of temperature to 37 °C over 120 min was seen, which is appropriate for bacterial treatment drugs. Given the advantages of the presented technique, it seems to have promising potential to be used in future medical applications for wound closure and bacterial infection treatment via a temperature-triggered drug release strategy.

The emergence of viral nanotechnology over the preceding two decades has created a number of intellectually captivating possible translational applications; however, the in vitro fate of the viral nanoparticles in cells remains an open question. Herein, we investigate the stability and lifetime of virus-like particle (VLP) Qβ-a representative and popular VLP for several applications-following cellular uptake. By exploiting the available functional handles on the viral surface, we have orthogonally installed the known FRET pair, FITC and Rhodamine B, to gain insight of the particle's behavior in vitro. Based on these data, we believe VLPs undergo aggregation in addition to the anticipated proteolysis within a few hours of cellular uptake.

Development of fluorescence enhancement (FE) platforms based on ZnO nanorods (NRs) has sparked considerable interest, thanks to their well-demonstrated potential in chemical and biological detection. Among the multiple factors determining the FE performance, high-order waveguide modes are specifically promising in boosting the sensitivity and realizing selective detection. However, quantitative experimental studies on the influence of the NR diameter, substrate, and surrounding medium, on the waveguide-based FE properties remain lacking. In this work, we have designed and fabricated a FE platform based on patterned and well-defined arrays of vertical, hexagonal prism ZnO NRs with six distinct diameters. Both direct experimental evidence and theoretical simulations demonstrate that high-order waveguide modes play a crucial role in FE, and are strongly dependent on the NR diameter, substrate, and surrounding medium. Using the optimized FE platform, a significant limit of detection (LOD) of 10-16 mol L-1 for Rhodamine-6G probe detection is achieved. Especially, a LOD as low as 10-14 g mL-1 is demonstrated for a prototype biomarker of carcinoembryonic antigen, which is improved by one order compared with the best LOD ever reported using fluorescence-based detection. This work provides an efficient path to design waveguiding NRs-based biochips for ultrasensitive and highly-selective biosensing.

An abnormal pH microenvironment results from the development of tumors, and also affects the therapeutic efficiency of anti-tumor drugs. In this work, a Förster resonance energy transfer (FRET)-based theranostic fluorescent nanoprobe was constructed for simultaneous ratiometric pH sensing and tumor-targeted photodynamic therapy. Based on the FRET process between rhodamine B and protoporphyrin IX (PpIX), the fabricated nanoprobe exhibited excellent pH responsiveness in both solutions and live cells with the ratiometric fluorescence changes. Moreover, this ratiometric pH fluorescent nanoprobe also possessed the capability for pH-responsive singlet oxygen (1O2) generation under light irradiation, guiding robust photodynamic therapy in a pH-dependent manner. Benefiting from the enhanced permeability and retention (EPR) effect, the nanoprobe could significantly inhibit tumor growth and metastasis via targeted photodynamic therapy in vivo. This work presents a novel paradigm for precise tumor theranostics by ratiometric pH fluorescence imaging-guided photodynamic therapy.

By combining a superwettable interface with a nanodendritic gold structure, we have fabricated a superwettable nanodendritic gold substrate for direct SERS detection of multiple concentrations of miRNAs. The nanodendritic gold substrate provides numerous hotspots for Raman signal enhancement, and the superwettable interface ensures the immobilization of droplets in superhydrophilic microwells, which hold great potentials for applications in disease diagnostics.

DNA origami nanostructures are a versatile tool to arrange metal nanostructures and other chemical entities with nanometer precision. In this way gold nanoparticle dimers with defined distance can be constructed, which can be exploited as novel substrates for surface enhanced Raman scattering (SERS). We have optimized the size, composition and arrangement of Au/Ag nanoparticles to create intense SERS hot spots, with Raman enhancement up to 10(10), which is sufficient to detect single molecules by Raman scattering. This is demonstrated using single dye molecules (TAMRA and Cy3) placed into the center of the nanoparticle dimers. In conjunction with the DNA origami nanostructures novel SERS substrates are created, which can in the future be applied to the SERS analysis of more complex biomolecular targets, whose position and conformation within the SERS hot spot can be precisely controlled.

The key to ultrabright fluorescent nanomaterials is the control of dye emission in the aggregated state. Here, lipophilic rhodamine B derivatives are assembled into nanoparticles (NPs) using tetraphenylborate counterions with varied fluorination levels that should tune the short-range dye ordering. Counterion fluorination is found to drastically enhance the emission characteristics of these NPs. Highly fluorinated counterions produce 10-20 nm NPs containing >300 rhodamine dyes with a fluorescence quantum yield of 40-60% and a remarkably narrow emission band (34 nm), whereas, for other counterions, aggregation caused quenching with a weak broad-band emission is observed. NPs with the most fluorinated counterion (48 fluorines) are ∼40-fold brighter than quantum dots (QD585 at 532 nm excitation) in single-molecule microscopy, showing improved photostability and suppressed blinking. Due to exciton diffusion, revealed by fluorescence anisotropy, these NPs are efficient FRET donors to single cyanine-5 acceptors with a light-harvesting antenna effect reaching 200. Finally, NPs with the most fluorinated counterion are rather stable after entry into living cells, in contrast to their less fluorinated analogue. Thus, the present work shows the crucial role of counterion fluorination in achieving high fluorescence brightness and photostability, narrow-band emission, efficient energy transfer and high intracellular stability of nanomaterials for light harvesting and bioimaging applications.

The core-shell structured mesoporous silica nanomaterials (MSNs) are experiencing rapid development in many applications such as heterogeneous catalysis, bio-imaging and drug delivery wherein a large pore volume is desirable. We develop a one-pot method for large-scale synthesis of brain-like mesoporous silica nanocomposites based on the reasonable change of the intrinsic nature of the -Si-O-Si- framework of silica nanoparticles together with a selective etching strategy. The as-synthesized products show good monodispersion and a large pore volume of 1.0 cm(3) g(-1). The novelty of this approach lies in the use of an inorganic-organic hybrid layer to assist the creation of large-pore morphology on the outermost shell thereby promoting efficient mass transfer or storage. Importantly, the method is reliable and grams of products can be easily prepared. The morphology on the outermost silica shell can be controlled by simply adjusting the VTES-to-TEOS molar ratio (VTES: triethoxyvinylsilane, TEOS: tetraethyl orthosilicate) as well as the etching time. The as-synthesized products exhibit fluorescence performance by incorporating rhodamine B isothiocyanate (RITC) covalently into the inner silica walls, which provide potential application in bioimaging. We also demonstrate the applications of as-synthesized large-pore structured nanocomposites in drug delivery systems and stimuli-responsive nanoreactors for heterogeneous catalysis.

Extensive efforts have been devoted to the development of a new biophotonic system using near infrared (NIR) nano-agents for non-invasive cancer diagnosis and therapy. Here, we developed a simple synthesis reaction of ligands, hydrazine, and iron(ii) chloride to fabricate Fe3O4 cluster-structured nanoparticles (CNPs) with interesting NIR photonics and high magnetization (Ms: 98.3 emu g(-1) and proton relaxivity r2: 234.6 mM(-1) s(-1)). These Fe3O4 CNPs exhibited optical absorption and reflection over all wavelengths, showing a U-shape absorption band with a low absorbance at a range of 750-950 nm and a progressive evolution in the second near infrared region. Strengthening of the scattering effect by incubating Fe3O4 CNPs with HeLa cells was observed when optical contrast enhancement was performed in an optical coherence tomography (OCT) microscope system with a laser light source at 860 nm. Using a 1064 nm laser at a low power density (380 mW cm(-2)) to excite the Fe3O4 CNPs (375 ppm[Fe]) led to a rise in the water temperature from 25 °C to 58 °C within 10 min. Finally, we present the first example of magnetomotive OCT cellular imaging combined with enhanced photothermal therapy using Fe3O4 CNPs and applying a magnetic field, which is promising for preclinical and clinical trials in the future.

We demonstrate the successful construction of fluorescently labeled magnetic antibody-like nanoparticles (ANPs) via a facile one-step surface-initiated in situ molecular imprinting approach over silica coated magnetite (Fe3O4@SiO2) core-shell nanocomposites. The as-prepared ANPs had a highly compact structure with an overall size of 83 ± 5 nm in diameter and showed excellent aqueous dispersion stability. With the predetermined high specificity to the target protein and high biocompatibility, the ANPs enabled rapid, efficient, selective and optically trackable sequestration of target proteins within living cells. This work represents the first example of fully artificially engineered multifunctional ANPs for the intracellular protein-sequestration without disruption of the cells. The established approach may be further extended to generate ANPs for various proteins of interest and provide useful tools for related biological research and biomedical applications.

Coordination nanoparticles (CNPs) are becoming attractive platforms for chemical sensing applications because their unique adjustable properties offer the opportunity to design various luminescent nanoprobes. Here, we present a CNP-based fluorescent nanoprobe, in which fluorophores (rhodamine B, RB) and quenchers (methylene blue, MB) were spontaneously enfolded by coordination networks self-assembled of adenine, biphenyl-4,4'-dicarboxylic acid (BDA) and zinc ions. The aggregation of fluorophores and quenchers in CNPs resulted in a quenched state fluorescence of RB. RB and MB could be released from CNPs in the presence of phosphate, which triggered the fluorescence of RB. On the basis of recognition-driven disassembly principle, a novel turn-on fluorescent probe for the determination of PO4(3-) with a wide response range (0.5-50 μM) has been successfully applied in the detection of phosphate in human serum samples. This work not only develops a probe for phosphate but also provides a general strategy for designing nanoprobes or nanocarriers towards various targets by altering organic linkers or metal ions.

DNA nanotechnology has touched the epitome of miniaturization by integrating various nanometer size particles with nanometer precision. This enticing bottom-up approach has employed small DNA tiles, large multi-dimensional polymeric structures or more recently DNA origami to organize nanoparticles of different inorganic materials, small organic molecules or macro-biomolecules like proteins, and RNAs into fascinating patterns that are difficult to achieve by other conventional methods. Here, we are especially interested in the self-assembly of nanomaterials that are potentially attractive elements in the burgeoning field of nanophotonics. These materials include plasmonic nanoparticles, quantum dots, fluorescent organic dyes, etc. DNA based self-assembly allows excellent control over distance, orientation and stoichiometry of these nano-elements that helps to engineer intelligent systems that can potentially pave the path for future technology. Many outstanding structures have been fabricated that are capable of fine tuning optical properties, such as fluorescence intensity and lifetime modulation, enhancement of Raman scattering and emergence of circular dichroism responses. Within the limited scope of this review we have tried to give a glimpse of the development of this still nascent but highly promising field to its current status as well as the existing challenges before us.

We have carried out a combined experimental and simulation study identifying the key physical and optical parameters affecting the presence and degree of fluorescence intensification measured on zinc oxide nanorod (ZnO NR) ends. Previously, we reported on the highly localized, intensified, and prolonged fluorescence signal measured on the NR ends, termed fluorescence intensification on NR ends (FINE). As a step towards understanding the mechanism of FINE, the present study aims to provide insight into the unique optical phenomenon of FINE through experimental and simulation approaches and to elucidate the key factors affecting the occurrence, degree, and temporal stability of FINE. Specifically, we examined the effect of the length, width, and growth orientation of single ZnO NRs on the NR-enhanced biomolecular emission profile after decorating the NR surfaces with different amounts and types of fluorophore-coupled protein molecules. We quantitatively and qualitatively profiled the biomolecular fluorescence signal from individual ZnO NRs as a function of both position along the NR long axis and time. Regardless of the physical dimensions and growth orientations of the NRs, we confirmed the presence of FINE in all ZnO NRs tested by using a range of protein concentrations. We also showed that the manifestation of FINE is not dependent on the spectroscopic signatures of the fluorophores employed. We further observed that the degree of FINE is dependent on the length of the NR with longer NRs showing increased levels of FINE. We also demonstrated that vertically oriented NRs exhibit much stronger fluorescence intensity at the NR ends and a higher level of FINE than the laterally oriented NRs. Additionally, we employed finite-difference time-domain (FDTD) methods to understand the experimental outcomes and to promote our understanding of the mechanism of FINE. Particularly, we utilized the electrodynamic simulations to examine both near-field and far-field emission characteristics when considering various scenarios of fluorophore locations, polarizations, spectroscopic characteristics, and NR dimensions. Our efforts may provide deeper insight into the unique optical phenomenon of FINE and further be beneficial to highly miniaturized biodetection favoring the use of single ZnO NRs in low-volume and high-throughput protein assays.

Upconversion nanophosphors (UCNPs) are extremely useful for analytical applications, since they display a high signal-to-noise ratio, and their photobleaching can be ignored. Herein, a novel upconversion nanocomposite composed of β-cyclodextrin (β-CD) derivative modified UCNPs and rhodamine B (RB) was prepared for the detection of cholesterol (Cho). The upconversion luminescence (UCL) emission can serve as a Cho-sensing signal by an effective fluorescence resonance energy transfer (FRET) process, using UCNPs as the donor and RB as the quencher. The sensor for Cho detection in human serum shows excellent sensitivity and selectivity, which has the potential for clinical applications in the analysis of other biological and environmental samples.

We present herein two colour encoded silica nanospheres (2nanoSi) for the fluorescence quantitative ratiometric determination of trypsin in humans. Current detection methods for cystic fibrosis diagnosis are slow, costly and suffer from false positives. The 2nanoSi proved to be a highly sensitive, fast (minutes), and single-step approach nanosensor for the screening and diagnosis of cystic fibrosis, allowing the quantification of trypsin concentrations in a wide range relevant for clinical applications (25-350 μg L(-1)). Furthermore, as trypsin is directly related to the development of cystic fibrosis (CF), different human genotypes, i.e. CF homozygotic, CF heterozygotic, and unaffected, respectively, can be determined using our 2nanoSi nanospheres. We anticipate the 2nanoSi system to be a starting point for non-invasive, easy-to-use and cost effective ratiometric fluorescent biomarkers for recessive genetic diseases like human cystic fibrosis. In a screening program in which the goal is to detect disease and also the carrier status, early diagnosis could be of great help.

Monodisperse multifunctional MnFe2O4/dye/silica core/shell nanoparticles have been designed and developed. The magnetic cores act as nano-heaters in biological systems under RF field excitation and the encapsulated dyes work as local temperature probes. The silica shells enable the water-solubility and biocompatibility of the NPs and protect the encapsulated fluorophores from photobleaching.

PEGylated magneto-plasmonic nanoparticles with a hollow or semi-hollow interior have been successfully synthesized and their physico-chemical characteristics have been investigated. The hollow interior space can be used to store drugs or other molecules of interest whereas magnetic characterization shows their potential as contrast agents in magnetic resonance imaging (MRI) applications. In addition, their plasmonic characteristics in the near infrared (NIR) region make them efficient in photothermal applications producing high temperature gradients after short irradiation times. We show that by controlling the etching conditions the inner silica shell can be selectively dissolved to achieve a hollow or semi-hollow interior without compromising the magnetic or plasmonic characteristics of the resulting nanoparticles. Magnetic measurements and transmission electron microscopy observations have been used to demonstrate the precise control during the etching process and to select an optimal concentration of the etching reagent and contact time to preserve the inner superparamagnetic iron oxide-based nanoparticles and the plasmonic properties of the constructs. Drug loading capabilities were also evaluated for both semi-hollow and as-synthesized nanoparticles using Rhodamine B isothiocyanate as a model compound. The nanoparticles produced could be potentially used as "theranostic" nanoparticles with both imaging capabilities and a dual therapeutic function (drug delivery and hyperthermia).

Super-hydrophobic surfaces are bio-inspired interfaces with a superficial texture that, in its most common evolution, is formed by a periodic lattice of silicon micro-pillars. Similar surfaces reveal superior properties compared to conventional flat surfaces, including very low friction coefficients. In this work, we modified meso-porous silicon micro-pillars to incorporate networks of metal nano-particles into the porous matrix. In doing so, we obtained a multifunctional-hierarchical system in which (i) at a larger micrometric scale, the super-hydrophobic pillars bring the molecules dissolved in an ultralow-concentration droplet to the active sites of the device, (ii) at an intermediate meso-scale, the meso-porous silicon film adsorbs the low molecular weight content of the solution and, (iii) at a smaller nanometric scale, the aggregates of silver nano-particles would measure the target molecules with unprecedented sensitivity. In the results, we demonstrated how this scheme can be utilized to isolate and detect small molecules in a diluted solution in very low abundance ranges. The presented platform, coupled to Raman or other spectroscopy techniques, is a realistic candidate for the protein expression profiling of biological fluids.

In this paper, Au/CuS composites were fabricated by a two-step method based on a facile solvothermal approach combined with the in situ reduction. It was demonstrated that the Au/CuS composite not only exhibited excellent peroxidase-like catalytic activity in the oxidation of the typical peroxidases (o-phenylenediamine and diaminobenzidine), but also showed promising SERS performance with remarkable sensitivity and high reproducibility. Based on these properties, the bi-functional Au/CuS composite was employed both as a catalyst for degrading a pollutant (Rhodamine 6G) and a SERS substrate for real-time monitoring of the degradation process quantitatively.

Crystalline silicon nanotubes (Si NTs) provide distinctive advantages as electrical and biochemical analysis scaffolds through their unique morphology and electrical tunability compared to solid nanowires or amorphous/non-conductive nanotubes. Such potential is investigated in this report. Gate-dependent four-probe current-voltage analysis reveals electrical properties such as resistivity to differ by nearly 3 orders of magnitude between crystalline and amorphous Si NTs. Analysis of transistor transfer characteristics yields a field effect mobility of 40.0 cm(2) V(-1) s(-1) in crystalline Si NTs. The hollow morphology also allows selective inner/outer surface functionalization and loading capability either as a carrier for molecular targets or as a nanofluidic channel for biomolecular assays. We present for the first time a demonstration of internalization of fluorescent dyes (rhodamine) and biomolecules (BSA) in Si NTs as long as 22 μm in length.

This study deals with the exploration of NixCo₁-xFe₂O₄ (x = 0.0, 0.2, 0.4, 0.6, 0.8, 1.0) ferrite nanoparticles as catalysts for reduction of 4-nitrophenol and photo-oxidative degradation of Rhodamine B. The ferrite samples with uniform size distribution were synthesized using the reverse micelle technique. The structural investigation was performed using powder X-ray diffraction, high-resolution transmission electron microscopy, energy dispersive X-ray and scanning tunneling microscopy. The spherical particles with ordered cubic spinel structure were found to have the crystallite size of 4-6 nm. Diffused UV-visible reflectance spectroscopy was employed to investigate the optical properties of the synthesized ferrite nanoparticles. The surface area calculated using BET method was found to be highest for Co₀.₄Ni₀.₆Fe₂O₄ (154.02 m(2) g(-1)). Co₀.₄Ni₀.₆Fe₂O₄ showed the best catalytic activity for reduction of 4-nitrophenol to 4-aminophenol in the presence of NaBH4 as reducing agent, whereas CoFe₂O₄ was found to be catalytically inactive. The reduction reaction followed pseudo-first order kinetics. The effect of varying the concentration of catalyst and NaBH₄ on the reaction rates was also scrutinized. The photo-oxidative degradation of Rhodamine B, enhanced oxidation efficacy was observed with the introduction of Ni(2+) in to the cobalt ferrite lattice due to octahedral site preference of Ni(2+). Almost 99% degradation was achieved in 20 min using NiFe₂O₄ nanoparticles as catalyst.

Graphene has recently emerged as a novel material in the biomedical field owing to its optical properties, biocompatibility, large specific surface area and low cost. In this paper, we provide the first demonstration of the possibility of using light to remotely trigger the release of drugs from graphene in a highly controlled manner. Different drugs including chemotherapeutics and proteins are firmly adsorbed onto reduced graphene oxide (rGO) nanosheets dispersed in a biopolymer film and then released by individual millisecond-long light pulses generated by a near infrared (NIR) laser. Here graphene plays the dual role of a versatile substrate for temporary storage of drugs and an effective transducer of NIR-light into heat. Drug release appears to be narrowly confined within the size of the laser spot under noninvasive conditions and can be precisely dosed depending on the number of pulses. The approach proposed paves the way for tailor-made pharmacological treatments of chronic diseases, including cancer, anaemia and diabetes.

Pre-formed silver-boron nanoparticles of 22 nm form pearl-like necklace nanostructures with interparticle junctions of less than 10 nm length in the matrix of polyethylene glycol (8000 Da). The silver necklace nanostructure is stable at 37 °C or 70 °C and also inside a live cell medium. A polyethylene glycol matrix with a shorter chain length (1000 Da) does not protect the nanoparticles against attraction, and random aggregates are formed. Silver necklace nanostructures exhibit strong Raman enhancement by more than ∼10(9) which is much higher than for silver-citrate or random silver-boron aggregates. The polymeric matrix of 8000 Da contributes strongly to the electromagnetic field enhancement and removes the chemical contribution to the surface Raman scattering increase. The stable interparticle junctions act as local hot spots for strong Raman scattering signals collected from live fibroblasts and allow systematic in situ studies.

pH is an important control parameter for maintenance of cell viability and tissue functions. pH monitoring provides valuable information on cell metabolic processes and the living environment. In this study, we prepared dual pH-sensitive, fluorescent dye-loaded polypeptide nanoparticles (DPNs) for ratiometric sensing of pH changes in living cells. DPNs contain two types of dyes: N-(rhodamine B) lactam cystamine (RBLC), an acid activatable fluorescent dye with increased fluorescence in an acidic environment, and fluorescein isothiocyanate (FITC), a base activatable fluorescent dye with enhanced fluorescence in an alkaline environment. Hence, DPNs exhibited a dual response signal with strong red fluorescence and weak green fluorescence under acidic conditions; in contrast, they showed strong green fluorescence and almost no red fluorescence under alkaline and neutral conditions. The favorable inverse pH responses of the two fluorescent dyes resulted in ratiometric pH determination for DPNs with an optimized pH-sensitive range of pH 4.5-7.5. Quantitative analysis of the intracellular pH of intact MCF-7 cells has been successfully demonstrated with our nanosensor. Moreover, single acid activatable fluorescent dye doped polypeptide nanoparticles that only contained RBLC can distinguish tumor tissue from normal tissue by monitoring the acidic extracellular environment.

We report the design and synthesis of a nano-container consisting of mesoporous silica nanoparticles with the pore openings covered by "snap-top" caps that are opened by near-IR light. A photo transducer molecule that is a reducing agent in an excited electronic state is covalently attached to the system. Near IR two-photon excitation causes inter-molecular electron transfer that reduces a disulfide bond holding the cap in place, thus allowing the cargo molecules to escape. We describe the operation of the "snap-top" release mechanism by both one- and two-photon activation. This system presents a proof of concept of a near-IR photoredox-induced nanoparticle delivery system that may lead to a new type of photodynamic drug release therapy.

We report on a synthetic approach to produce self-supported flexible surface-enhanced Raman scattering (SERS) active membranes consisting of polyamide (PA) nanofibers grafted with vertical Ag-nanosheets, via a combinatorial process of electrospinning PA-nanofiber membranes, assembling Au-nanoparticles on the PA-nanofibers as seeds for subsequent growth of Ag-nanosheets, and electrodepositing Ag-nanosheets on the electrospun PA-nanofibers. As a high density of Ag-nanosheets are vertically grown around each PA-nanofiber in the three-dimensional (3D) networked PA-nanofiber membranes, homogeneous nano-scaled gaps between the neighboring Ag-nanosheets are formed, leading to a high density of 3D SERS "hot spots" within the Ag-nanosheet-grafted PA-nanofiber membranes. The Ag-nanosheet-grafted PA-nanofiber membranes demonstrate high SERS activity with excellent Raman signal reproducibility for rhodamine 6G over the whole membrane. For a SERS-based trial analysis of polychlorinated biphenyls (PCBs, a kind of global environmental hazard), the 3D SERS substrate membranes are modified with mono-6-β-cychlodextrin to effectively capture PCB molecules. As a result, not only a low concentration down to 10(-6) M is reached, but also two congeners of PCBs in their mixed solution are identified, showing promising potential in SERS-based rapid detection of trace organic pollutants such as PCBs in the environment.

Applications of mesoporous silica (m-SiO2) have suffered from its fragility (monolithic m-SiO2 easily collapses under compression) and limited internal molecular exchange through small channels. Previously reported hierarchical porous m-SiO2 structures containing interconnected macropores could improve adsorption properties, but they were still intrinsically fragile without sufficient mechanical strength to sustain deformation. Here, we embed a three-dimensional carbon nanotube (CNT) skeleton into m-SiO2 to fabricate bulk, robust sponges that can be compressed to large strains (60% volume reduction) repeatedly in both air and water. This is done by directly casting a uniform m-SiO2 layer with tunable thickness onto the surface of CNTs while maintaining the original network and open porous structure, resulting in a core-shell CNT@m-SiO2 hybrid sponge. By pumping fluid through the CNT@m-SiO2 sponges under cyclic compression, the adsorption rate and efficiency of dye molecules can be significantly enhanced due to the mesoporous coating on CNTs and enhanced fluid exchange throughout internal pores. The CNT@m-SiO2 sponges may be used as robust and flexible adsorption media, and chemical and biological sensors with high performance.

Single crystal TiOF2 nanocubes assembled into a mesoporous structure were synthesized by alcoholysis of TiF4 under solvothermal conditions, which displayed spectral response in the visible area owing to the intrinsic narrow energy band gap. Mechanism studies revealed that TiOF2 was formed via consecutive hydrolysis reactions and the H2O produced by condensation between two alcohols played a key role in determining the TiOF2 crystal growth and its transformation to anatase TiO2. The TiOF2 nanocube size could be easily adjusted by changing either alcoholysis time, or solvothermal temperature, or alcohol kind owing to the different H2O production rate and amount. The small-sized TiOF2 nanocubes with large surface area exhibited high activity in photocatalytic degradation of Rhodamine B (RhB) and 4-chlorophenol (4-CP) owing to the enhanced adsorption for reactant molecules and the reduced photoelectron-hole recombination rate. Meanwhile, they also showed strong durability since the mesoporous structure enhanced the stability against either the phase transformation from TiOF2 crystal to anatase TiO2 or the agglomeration of TiOF2 nanocubes.

Stable Cu nanoparticles (NPs) prepared using catechol containing dopamine-based linkers could generate reactive oxygen species (ROS) that can activate peroxidase enzymes and catalyze the degradation of fluorescent dye pollutants.

The objective of this study was to evaluate the targeted T1-magnetic resonance imaging (MRI), quantitative biodistribution and toxicity of aptamer (AS411) conjugated Mn3O4@SiO2 core-shell nanoprobes (NPs) in human cervical carcinoma tumor-bearing mice. The NPs were firstly prepared by encapsulating a hydrophobic Mn3O4 core within an amino functionalized silica shell. The fluorophore rhodamine (RB) was doped into the silica shell and the amphiphilic polymer poly(ethylene glycol) (PEG) was modified on the surface of the shell to improve its biocompatibility, then the aptamer AS411 was conjugated onto the end of the PEG chains as targeting ligands. The final NPs were abbreviated as Mn3O4@SiO2(RB)-PEG-Apt. By means of in vitro fluorescence confocal imaging and in vivo MRI, the NPs have been demonstrated to target cancer cells and prominent tumor aggregation effectively. The imaging results were further confirmed by a quantitative biodistribution study. In addition, histological, hematological and biochemistry analysis also proved the low toxicity of NPs in vivo. Our results showed the great potential of the Mn3O4@SiO2(RB)-PEG-Apt NPs could be used as a multifunctional nanoplatform for long-term targeted imaging and therapy of cancer.

The study of pathogen interactions with eukaryotic host cells requires the introduction of fluorescent probes to visualize processes such as endocytosis, intracellular transport or host cell manipulation by the pathogen. Here, three types of fluorescent nanoparticles (NPs), i.e. Rhodamine-labeled polymethacrylate (PMA) NPs, silica NPs and gold NPs, were employed to label the host cellular endolysosomal system and monitor manipulations by the pathogen Salmonella enterica. Using live cell imaging, we investigated the performance of NPs in cellular uptake, labeling of endocytic vesicles and lysosomes, as well as interaction with the pathogen. We show that fluorescent gold and silica, but not PMA NPs appropriately label host cell structures and efficiently track rearrangements of the host endosomal system by the activities of intracellular Salmonella. Silica NPs slightly aggregated and located in Salmonella-induced compartments as isolated dots, while gold NPs distributed uniformly inside such structures. Both silica and gold NPs exhibited no adverse impact on either host cells or pathogens, and are versatile tools for infection biology.

We report on the self-assembly of colloidal gold nanoparticles on a stretchable, elastomeric membrane, and the use of this membrane as a base substrate for far-field confocal Raman measurements. Surface-enhanced Raman scattering (SERS) enhancement for such a substrate was estimated as 10(6) to 10(7). Atomic force microscopy has been used to study the changes in nanoparticle topography when the membrane is stretched. The homogeneous strain defined by average relative motion of nanoparticles is approximately half the macroscopically-applied biaxial strain. The SERS intensity was maximized when the membrane was at rest (i.e. without stretch), and reduced as stretching was increased. Our measurements are consistent with theoretical and numerical SERS enhancements for the interstitial gap between two spheres. The data indicate that the resting gap between the spheres is 11 nm or 16 nm, using two theoretical models. This work represents progress towards particularly facile sample fabrication and in situ tuning techniques for SERS.

In the present study, we report the design and synthesis of peptide-based-multi-walled carbon nanotubes (MWCNTs) to target mitochondria. Targeting these intracellular organelles might open the way to develop alternative systems to address diseases related to genetic mutations in mitochondrial (mt)-DNA, by delivering therapeutic oligonucleotides. The first step towards mitochondrial delivery of this type of nucleic acid was to target MWCNTs to mitochondria by covalent functionalization with a well-known endogenous mitochondrial targeting sequence (MTS). The subcellular localization of the conjugates, which were fluorescently labeled, in murine RAW 264.7 macrophages and human HeLa cells was then studied using different microscopy techniques, such as wide-field epifluorescence microscopy, confocal laser scanning microscopy (CLSM) and transmission electron microscopy (TEM). The localization of the MTS-MWCNT conjugates into mitochondria was further confirmed by analyzing the isolated organelles using TEM.

Silica-based luminescent nanoparticles (SiNPs) show promising prospects in nanomedicine in light of their chemical properties and versatility. In this study, we have characterized silica core-PEG shell SiNPs derivatized with PEG moieties (NP-PEG), with external amino- (NP-PEG-amino) or carboxy-groups (NP-PEG-carbo), both in cell cultures as well as in animal models. By using different techniques, we could demonstrate that these SiNPs were safe and did not exhibit appreciable cytotoxicity in different relevant cell models, of normal or cancer cell types, growing either in suspension (JVM-2 leukemic cell line and primary normal peripheral blood mononuclear cells) or in adherence (human hepatocarcinoma Huh7 and umbilical vein endothelial cells). Moreover, by multiparametric flow cytometry, we could demonstrate that the highest efficiency of cell uptake and entry was observed with NP-PEG-amino, with a stable persistence of the fluorescence signal associated with SiNPs in the loaded cell populations both in vitro and in vivo settings suggesting this as an innovative method for cell traceability and detection in whole organisms. Finally, experiments performed with the endocytosis inhibitor Genistein clearly suggested the involvement of a caveolae-mediated pathway in SiNP endocytosis. Overall, these data support the safe use of these SiNPs for diagnostic and therapeutic applications.

One-dimensional nanowire (NW) optical sensors have attracted great attention as promising nanoscale tools for applications such as probing inside living cells. However, achieving single molecule detection on NW sensors remains an interesting and unsolved problem. In the present paper, we investigate single-molecule detection (SMD) on a single SiNW based surface-enhanced Raman scattering (SERS) sensor, fabricated by controllably depositing silver nanoparticles on a SiNW (AgNP-SiNW). Both Raman spectral blinking and bi-analyte approaches are performed in aqueous solution to investigate SMD on individual SiNW SERS sensors. The results extend the functions of the SiNW sensor to SMD and provide insight into the molecule level illustration on the sensing mechanism of the nanowire sensor.

Uniform SiO2@CdS mesoporous nanospheres with an average diameter of 300 nm have been synthesized successfully by a facile process. The as-prepared mesoporous composite nanospheres have a BET-specific surface area of 640 m(2) g(-1) and an average pore size of 2.82 nm. The results demonstrated that more than 60% Rhodamine B (RhB) dye in solution (4.8 mg L(-1), 50 mL) could be removed by adsorption in the dark for 30 min using the as-prepared SiO2@CdS mesoporous nanospheres (40 mg). The as-prepared SiO2@CdS mesoporous nanospheres have a mesoporous nanostructure, suggesting a higher specific surface area and resulting in a strong adsorption ability. In addition, the mesoporous silica was decorated with ca. 5 nm CdS nanocrystals, which showed excellent photocatalytic activity under visible light and could rapidly remove most of the RhB molecules from a pollutant solution under visible light irradiation. Furthermore, the mesoporous SiO2@CdS nanospheres synthesized by the present protocol exhibited excellent antibacterial activity.

Closely packed, size-controllable and stable Au nanohorns (AuNHs) that are difficult to synthesize through pure chemical reduction are facilely synthesized using a microorganism-mediated method in the presence of hexadecyltrimethylammonium chloride (CTAC). The results showed that the size of the as-synthesized AuNHs could be tuned by adjusting the dosage of the Pichia pastoris cells (PPCs). The initial concentrations of CTAC, ascorbic acid (AA) and tetrachloroaurate trihydrate (HAuCl4·3H2O) significantly affected the formation of the AuNHs. Increasing the diameters of AuNHs led to a red shift of the absorbance bands around 700 nm in their UV-vis-NIR spectra. Interestingly, the AuNH/PPC composites exhibited excellent Raman enhancement such that rhodamine 6G with concentration as low as (10(-9) M) could be effectively detected. The formation process of the AuNHs involved the initial binding of the Au ions onto the PPCs with subsequent reduction by AA to form supported Au nanoparticles (AuNPs) based on preferential nucleation and initial anisotropic growth on the platform of the PPCs. The anisotropic growth of these AuNPs, which was influenced by CTAC and PPCs, resulted in the formation of growing AuNHs, while the secondary nucleation beyond the PPCs produced small AuNPs that were subsequently consumed through Ostwald ripening during the aging of the AuNHs. This work exemplifies the fabrication of novel gold nanostructures and stable bio-Au nanocomposites with excellent optical properties by combining microorganisms and a surfactant.

We present an innovative approach to the production of sub-100 nm hollow Au-SnO2 hybrid nanospheres, employing a low-cost, surfactant-free and environmentally friendly solution-based route. The hollow hybrid nanostructures were synthesized using a seed-mediated hydrothermal method, which can be divided into two stages: (1) formation of multicore-shell Au@SnO2 nanoparticles (NPs) and (2) thermal diffusion and ripening to form hollow Au-SnO2 hybrid NPs. The morphology, optical properties and formation mechanism were determined by a collection of joint techniques. Photocatalytic degradation of Rhodamine B (RhB) in the liquid phase served as a probe reaction to evaluate the activity of the as-prepared hollow hybrid Au-SnO2 NPs under the irradiation of both visible light and ultraviolet light. Significantly, the as-obtained Au-SnO2 hybrid nanostructures exhibited enhanced visible light or UV photocatalytic abilities, remarkably superior to commercial pure SnO2 products and P25 TiO2, mainly owing to the effective electron hole separation at the SnO2-Au interfaces and strong localization of plasmonic near-fields effects.

This work demonstrates a novel multifunctional nanofibrous mat for photocatalytic applications based on TiO2 nanocables functionalized by Ag nanoparticles and coated with a thin (~2 nm) graphitic shell. In this mat, which was realized by an electrospinning technique, each component serves a unique function: the carbon coating acts as both an adsorption material for capturing pollutants and as a charge-transfer material, the Ag nanoparticles act as a visible-light sensitizing agent and also as a charge-transfer material, finally the TiO2 nanocable mat acts as a UV sensitive photocatalytic matrix and as the flexible substrate for the other functional components. This multicomponent nanocable mat exhibits excellent photocatalytic activity under simulated solar irradiation for the degradation of model pollutants including RhB and phenol. The significant photocatalytic properties are attributed to the synergetic effect of the three functional components and the unique charge transport "freeway" property of the nanofibrous mat. In addition, the porous carbon coating infiltrated into the nanocable matrix endows the mat with excellent flexibility and enables robust, large-area (10 × 10 cm) fabrication, representing a significant advantage over previous brittle ceramic nanofibrous mat photocatalyst substrates. This study provides new insight into the design and preparation of an advanced, yet commercially practical and scaleable photocatalytic composite membrane material. The as-prepared photocatalytic mat might also be of interest in solar cell, catalysis, separation technology, biomedical engineering, and nanotechnology.

With growing interest in the photocatalytic performance of TiO2-graphene composite systems, the ternary phase of TiO2, graphene, and Ag is expected to exhibit improved photocatalytic characteristics because of the improved recombination rate of photogenerated charge carriers and potential contribution of the generation of localized surface plasmon resonance at Ag sites on a surface of the TiO2-graphene binary matrix. In this work, Ag-TiO2-reduced graphene oxide ternary nanocomposites were successfully synthesized by a simple solvothermal process. In a single-step synthetic procedure, the reduction of AgNO3 and graphene oxide and the hydrolysis of titanium tetraisopropoxide were spontaneously performed in a mixed solvent system of ethylene glycol, N,N-dimethylformamide and a stoichiometric amount of water without resorting to the use of typical reducing agents. The nanocomposites were characterized by X-ray diffraction, X-ray photoelectron spectroscopy, along with different microscopic and spectroscopic techniques, enabling us to confirm the successful reduction of AgNO3 and graphite oxide to metallic Ag and reduced graphene oxide, respectively. Due to the highly facilitated electron transport of well distributed Ag nanoparticles, the synthesized ternary nanocomposite showed enhanced photocatalytic activity for degradation of rhodamine B dye under visible light irradiation.

Here we report a controllable method based on electrodeposition to fabricate Ag nanodendrites (NDs) on a microwell patterned electrode. The microwell patterns on the ITO electrode are fabricated via the microcontact printing technique. By varying the microwell size and electrodeposition time, the morphology of metal deposits on the microwell patterned ITO electrode can be tuned from boulders to dendrites. At the edge of the microwells, the current density was strengthened, which incurs rapid nucleation. The nucleus develops into dendrites because of Mullins-Sekerka instability. However, only boulders were observed at the center of microwells. By reducing the size of the microwells, only NDs were fabricated due to the edge effect. On the basis of understanding the underlying mechanism for dendritic growth in a confined space, our method is used for fabricating other noble metal (Au, Pt) nanodendrites. The controllable synthesis of Au and Pt NDs indicates the universality of this method. Compared with Ag film obtained from electron beam evaporation, the as-prepared Ag NDs exhibit highly enhanced surface-enhanced Raman scattering (SERS) sensitivity when they are used to detect rhodamine 6G (R6G). This approach provides a very controllable, reliable and general way for space-confined fabricating the noble metal nanodendrite arrays which show great promise in catalysis, sensing, biomedicine, electronic and magnetic devices.

In this work we report, for the first time, on the preparation of ZnO nanorod-SnO2 nanoparticle (ZnO NR-SnO2 NP) heterostructures by a simple two-step thermal evaporation approach. Systematical characterization of the product reveals that the rutile SnO2 NPs, with a diameter of about 20 nm, are uniformly and tightly decorated on the entire ZnO NRs. Photoluminescence (PL) investigation on the ZnO NR-SnO2 NP heterostructures shows that they exhibit a significantly decreased UV emission compared with the bare ZnO NRs, revealing an efficient charge separation arising from the type-II band alignment. Enlightened by this merit, photocatalytic behavior of the synthesized heterostructures is studied, which shows a remarkably enhanced photodegradation performance of rhodamine B (RhB) in contrast to the pure ZnO NRs. We also carry out the stability test of the ZnO NR-SnO2 NP heterostructures and the result indicates an extremely high adhesion nature between the ZnO NR and the coated SnO2 NPs. This advantage endowed with the thermal evaporation approach can lead to an efficient spatial charge separation between the ZnO NR and the SnO2 NPs and thus effectively minimize the charge recombination along three-dimensional heterointerfaces, which makes such ZnO NR-SnO2 NP architectures highly promising for a wide range of photovoltaic and photocatalytic applications.

Analytical techniques based on surface-enhanced Raman scattering (SERS) suffer from a lack of reproducibility and reliability, thus hampering their practical applications. Herein, we have developed a SERS-active substrate based on a graphene oxide embedded sandwich nanostructure for ultrasensitive Raman signal readout. By using this novel Au@Ag NPs/GO/Au@Ag NPs sandwich nanostructure as a SERS substrate, the Raman signals of analytes were dramatically enhanced due to having plenty of hot spots on their surfaces and the unique structure of the graphene oxide sheets. These features make the sandwich nanostructured film an ideal SERS substrate to improve the sensitivity, reproducibility and reliability of the Raman readout. The sandwich nanostructure film can be applied to detect rhodamine-6G (R6G) with an enhancement factor (EF) of ∼7.0 × 10(7) and the pesticide thiram in commercial grape juice with a detection limit of as low as 0.1 μM (0.03 ppm), which is much lower than the maximal residue limit (MRL) of 7 ppm in fruit prescribed by the U.S. Environmental Protection Agency (EPA). The GO embedded sandwich nanostructure also has the ability to selectively detect dithiocarbamate compounds over other types of agricultural chemical. Furthermore, spiked tests show that the sandwich nanostructure can be used to monitor thiram in natural lake water and commercial grape juice without further treatment. In addition, the GO enhanced Raman spectroscopic technique offers potential practical applications for the on-site monitoring and assessment of pesticide residues in agricultural products and environments.

Conventional vapor-deposition techniques for coatings require sophisticated equipment and/or high-temperature resistant substrates. Therefore bio-inspired techniques for the fabrication of inorganic coatings have been developed in recent years. Inspired by the biology behind the formation of the intricate skeletons of diatoms orchestrated by a class of cationic polyamines (silaffins) we have used surface-bound spermine, a naturally occurring polyamine, to promote the fast deposition of homogeneous, thin and transparent biomimetic SnO2 coatings on glass surfaces. The bio-enabled SnO2 film is highly photoactive, i.e. it generates superoxide radicals (O2˙(-)) upon sunlight exposure resulting in a strong degradation of organic contaminants and a strong antimicrobial activity. Upon illumination the biomimetic SnO2 coating exhibits a switchable amphiphilic behavior, which - in combination with its photoactivity - creates a self-cleaning surface. The intrinsic self-cleaning properties could lead to the development of new protective, antifouling coatings on various substrates.

BSA protected gold nanoclusters (Au25) are attracting a great deal of attention due to their unique spectroscopic properties and possible use in biophysical applications. Although there are reports on synthetic strategies, spectroscopy and applications, little is known about their polarization behavior. In this study, we synthesized the BSA protected Au25 nanoclusters and studied their steady state and time resolved fluorescence properties including polarization behavior in different solvents: glycerol, propylene glycol and water. We demonstrated that the nanocluster absorption spectrum can be separated from the extinction spectrum by subtraction of Rayleigh scattering. The nanocluster absorption spectrum is well approximated by three Gaussian components. By a comparison of the emissions from BSA Au25 clusters and rhodamine B in water, we estimated the quantum yield of nanoclusters to be higher than 0.06. The fluorescence lifetime of BSA Au25 clusters is long and heterogeneous with an average value of 1.84 μs. In glycerol at -20 °C the anisotropy is high, reaching a value of 0.35. However, the excitation anisotropy strongly depends on the excitation wavelengths indicating a significant overlap of the different transition moments. The anisotropy decay in water reveals a correlation time below 0.2 μs. In propylene glycol the measured correlation time is longer and the initial anisotropy depends on the excitation wavelength. BSA Au25 clusters, due to long lifetime and high polarization, can potentially be used in studying large macromolecules such as protein complexes with large molecular weight.

We report a methodology developed to detect and track stable fluorescent-labeled nanoclay, in polymer-clay nanocomposite films, and in a contact solvent after migration testing. Fluorescein-5-maleimide (fluorescein) or tetramethylrhodamine-5-maleimide (rhodamine) was covalently bonded to organically modified montmorillonite (o-MMT). Fluorescein- and rhodamine-labeled nanoclay showed good thermal stability up to 220 °C and the rhodamine-labeled nanoclay remained stable at 250 °C. Confocal laser scanning microscopy was used to confirm the tagging and to detect the fluorescent-labeled nanoclays in various systems.

Porous BiOCl micro-flowers constructed from ultrathin nanosheets with nearly 100% {001} facets exposed were selectively prepared. The exposed {001} facets terminated with a high density of oxygen atoms and are not only favorable for the adsorption of the cationic dye RhB but also can accumulate the photogenerated electrons injected from excited RhB. These electrons can be captured by O(2) and transformed to reactive oxygen species, which possess a strong photooxidative ability to degrade the dye pollutants directly and easily. Therefore, the resultant BiOCl photocatalysts exhibit superior activity for indirect dye photosensitization degradation under visible light, with a rapid degradation rate and high photostability.

In general, the procedures for producing a high density hot spots structure should be stable, inexpensive, and easy to make. It still remains a grand challenge to assemble silver or gold nanoparticles (Au NPs) with well-defined hot spots for SERS detection. In this study, we present a very simple method for designing and fabricating a surface-enhanced Raman scattering (SERS) substrate with high density hot spots, using large area positively charged polyaniline (PANI) nanofibers as template to assemble negatively charged Au NPs. In order to obtain the optimized SERS-active substrates, different experiments to synthesize diverse Au/PANI with different sizes of Au NPs from about 50, 30 to 15 nm were carried out. The results revealed that the PANI nanofibers were fully coated by the ~15 nm Au NPs, forming a high density Au/PANI SERS substrate. The results evidence that we can obtain stable and sensitive SERS measurements.

Detection of Cu(2+) ions and study of their subcellular distribution in physiological processes are of considerable significance because of their potential environmental and biological applications. Some fluorescence based sensors have been developed for selective detection of Cu(2+) ions, based on organic fluorescent probes that specifically bind to Cu(2+) ions. However, these sensors are not suitable for detection in biological samples due to the short penetration depth of UV/visible light used to excite the fluorescent probes. The use of near-infrared (NIR) light can afford penetration depths of an order of magnitude greater than that of visible light, however, a material that can convert NIR light to visible light is required. A facile method has been developed for in-depth detection of Cu(2+) ions based on fluorescence upconversion. A mesoporous silica shell is coated on upconversion nanoparticles (UCNPs) and a Cu(2+) ion sensitive fluorescent probe, rhodamine B hydrazide, is incorporated into the mesoporous silica. Upon excitation by a NIR light, the UCNPs emit visible light to excite the Cu(2+)-sensitive fluorescent probe. Because of the unique optical properties of UCNPs and their ability to convert NIR light to visible light, this is a feasible method for sensitive and in-depth detection of Cu(2+) ions in a complex biological or environmental sample due to the low autofluorescence and the high penetration depth of NIR light.

We have developed simple and green electrochemistry to synthesize Ag nanostructures with high purity, good crystallinity and smooth surface for applications as super-SERS (surface-enhanced Raman scattering), SERS-active substrates and with highly effective antimicrobial activities. This synthesis takes place in a clean and slow reaction environment without any chemical additives, which ensures an ultrahigh active surface of the as-synthesized Ag nanostructures owing to their purity, good crystallinity and smooth morphology. Using this method, we synthesized nearly perfect Ag nanodendrites (NDs), which exhibit super-SERS sensitivity when they are used to detect the SERS spectra of rhodamine 6G at concentrations as low as 5 × 10(-16) M, and have an ultrahigh electromagnetic (EM) enhancement factor of the order of 10(13), breaking through the theoretical limit of EM enhancement. Meanwhile, the as-synthesized Ag NDs possess highly effective antimicrobial activities for Escherichia coli, Candida albicans and Staphylococcus aureus, which are over 10 times that of silver nanoparticles. Additionally, the basic physics and chemistry involved in the fabrication of Ag nanostructures are pursued. These investigations show that silver nanostructures with highly active surfaces can make the most of Ag nanostructures functioning as super-SERS-active substrates and multiple antibiotics.
