The Asia-Pacific region is renowned to harbor nearly half of the global hot spots of biodiversity. Accordingly, many endemic species of boletes have already been recorded from this geographic region. However, the majority of the specific descriptions of reported boletoid species follow classical concepts of taxonomy, and by comparison only a few taxa have been corroborated by modern molecular techniques. In this study, we focused on specimens in a new clade uncovered by our previous studies. By careful reexamination of macroscopic and microscopic characters of Boletus granulopunctatus, originally described from Japan, and Xerocomus mcrobbii, originally described from New Zealand, we discovered a new genus and species Amoenoboletus miraculosus from Sabah, Malaysia. In addition, three new combinations in Amoenoboletus are proposed, and a dichotomous key to species in the genus is provided. The phylogenetically close relationship among Amoenoboletus species suggests a tight geographic correlation in the Asia-Pacific region.

Within the family Entolomataceae, many reports of Entoloma and Rhodocybe have been published, but few reports on Clitopilus and allied genera are available. In this study, Clitopilus, Clitocella, and Clitopilopsis from China were studied by morphological and phylogenetic methods. Portions of nuc 28S rDNA (28S), RNA polymerase II second largest subunit (rpb2), translation elongation factor 1-alpha (tef1), and ATPase subunit 6 (atp6), were employed to elucidate the relationships of Clitopilus and allies. Results indicate that Clitopilus should be divided into three sections, namely, sect. Clitopilus, sect. Scyphoides, and sect. Crispi, the last of which is newly described. In total, 17 species of Clitopilus, Clitocella, and Clitopilopsis are found in China; 14 species belong to Clitopilus, of which 5 are new: C. yunnanensis, C. brunniceps, C. rugosiceps, C. sinoapalus, and C. umbilicatus. Two species of Clitocella, including one new species, Clitocella orientalis, are described. One new species of Clitopilopsis, namely, Clitopilopsis albida, is proposed.

Dipterocarp forests are a typical and widespread type of vegetation in tropical lowlands of southeast Asia that harbor a high diversity of ectomycorrhizal fungi, including boletes. Based on molecular and morphological characters, a unique bolete found in Singapore associated with the dipterocarp Hopea odorata was proven to represent a new species in the proposed new genus Spongispora. Phylogenetic analyses of five loci indicate that Spongispora is nested in the subfamily Leccinoideae of the Boletaceae, most closely related to an inclusive clade of Leccinum, Leccinellum, Octaviania, Rossbeevera, and Turmalinea. However, genetic distances between Spongispora and genera in Leccinoideae are mostly higher than that between any two known genera in this subfamily, which supports the proposal of a new genus. Spongispora temasekensis is characterized by a whitish to pale yellow hymenophore that stains brown where injured, coarsely reticulate stipe, interwoven trichodermial pileipellis, and broadly elliptical to ovoid basidiospores with sponge-like ornamentation perforated by irregular clefts, cracks, and warts under scanning electron microscopy. Morphological descriptions, illustrations, and comparisons with allied taxa are made, and a key to the genera of the subfamily Leccinoideae is provided.

Some Amanita specimens collected from Malaysia are critically investigated by morphological examination and molecular analysis of two gene fragments, the nuc rDNA partial 28S (28S) gene and the internal transcriber spacer (ITS1-5.8S-ITS2 = ITS) regions. Six phylogenetic species of Amanita section Caesareae are recognized among the studied collections. One of them is described as new, A. malayensis. Four of the phylogenetic species correspond with existing morphology-based taxa: A. aporema, A. javanica, A. princeps, and A. similis. The remaining species is not described because of the paucity of material. Detailed descriptions and the distribution of these southeastern Asian species are provided, along with a key to the species of section Caesareae from Malaysia.

Three new and one previously described species of Physalacria (Physalacriaceae, Agaricales) are reported from China. Specimens of two additional species described from Malaysia and North America were studied for comparison. Placements of these species were corroborated based on morphological observations and molecular evidence from partial sequences of the nuc rDNA internal transcribed spacer regions (ITS) and the 28S D1-D3 region, and genes for translation elongation factor 1-α (tef1α) and the second largest subunit of RNA polymerase II (rpb2). These new species of Physalacria distributed in subtropical China were found on rotten wood of broadleaf trees or bamboo and possess stipitate-capitate basidiomata with four-spored basidia, clamp connections and smooth, inamyloid basidiospores. To facilitate studies of the genus in Asia, a key is provided for all Physalacria species reported from this region.

Although taxonomic knowledge on Tricholoma (Agaricales, Basidiomycota) is fairly comprehensive in northwest Europe, knowledge of the global diversity and distribution of Tricholoma spp. is still sparse. In this study, the diversity and distribution of some Tricholoma spp. are analyzed by morphological and molecular methods based on 70 collections from Yunnan, China, 45 from central Europe, 32 from Colorado, USA, 9 from Japan, and 3 from Ukraine. A Holarctic distribution is suggested for several species, based on collections and nuc rDNA internal transcribed spacer ITS1-5.8S-ITS2 (ITS) sequences. Six species new to science are formally described from Yunnan: five in existing sections, Tricholoma forteflavescens, T. olivaceoluteolum, T. melleum, T. olivaceum, and T. sinoportentosum, and one, T. muscarioides, in the newly described section Muscaria alongside several previously described species. Additional putatively new species cannot be formally described because they lack sufficient material. Tricholoma foliicola is recognized as a species of the genus Gerhardtia.

The fungal genus Strobilurus belongs to Physalacriaceae and contains approximately 11 species worldwide. Species of this genus grow and reproduce on cones of various conifers, seed pods or fruits of Magnolia and Liquidambar, and branches and wood of conifers. Previous studies focused mainly on samples from Europe and North America. And no genus-specific phylogenetic analysis has been carried out to date. The monophyly, degree of species diversity and substrate specificity, and overall distribution patterns are addressed here using morphological and molecular evidence. The authors collected samples of Strobilurus from much of its known distribution ranges and carried out morphological observations and multilocus phylogenetic analyses using five molecular markers. The results show that Strobilurus is a monophyletic group but may exclude one species, S. ohshimae. A total of 13 species was identified, with two, S. orientalis and S. pachycystidiatus, described as new from China. Several species were shown to be specific to certain substrates, whereas a few less so. Biogeographic analyses indicated that historical exchanges of species between East Asia, Europe, and North America, later vicariance events, and substrate specificity have contributed jointly to diversification of Strobilurus.

Species of Pulveroboletus (Boletaceae, Boletales) in China are investigated on the basis of morphology and molecular phylogenetic analyses of DNA sequences from nuc rDNA region encompassing the internal transcribed spacers 1 and 2, along with the 5.8S rDNA (ITS) and nuc 28S rDNA D1-D2 domains (28S) and sequences from the translation elongation factor 1-α gene (TEF1). Nine species are recognized in the country, three of them are described as new: P. flaviscabrosus, P. rubroscabrosus, and P. subrufus, five of them are previous described taxa: P. brunneopunctatus, P. brunneoscabrosus, P. macrosporus, P. reticulopileus, and P. sinensis, the remaining one is tentatively named P. cf. ridleyi, which will be further studied in the future. A key to the Chinese taxa of the genus is provided. Unexpectedly, the current study did not identify disjunct populations of Pulveroboletus species in East Asia and North/Central America.

Veloporphyrellus is a genus known from North and Central America, southeastern Asia, and Africa. Because species of this genus are phenotypi- cally similar to some taxa in several genera, such as Boletellus, Leccinum, Strobilomyces, Suillus and Tylopi- lus s.l. belonging to Boletales, its phylogenetic disposition has never been addressed. We analyzed four DNA regions, the nuclear ribosomal LSU and tef- 1a, and the mitochondrial mtSSU and atp6 genes, to investigate the phylogenetic disposition of Velopor- phyrellus. Although the monophyly of the genus and its systematic placement within the Boletaceae was well supported, its relationship to other genera was not resolved. Morphologically Veloporphyrellus is distinguished from other boletoid genera by the combination of the pinkish or grayish pink hymeno- phore, the membranous veil hanging on the pilea margin, the trichoderm-like pileus covering and the smooth basidiospores. Five species, including two new species and two new combinations, are described and illustrated. A key to the species of Veloporphyrellus also is provided.

Lethal amanitas (Amanita sect. Phalloideae) cause many casualties worldwide. Recent molecular phylogenetic studies revealed diverse lethal Amanita spp. in China. Here a 5-gene phylogeny (nuc rDNA region encompassing the internal transcribed spacers 1 and 2 with the 5.8S rDNA, the D1–D3 domains of nuc 28S rDNA, and partial RNA polymerase II second largest subunit, translation elongation factor 1-α and β-tubulin genes) is used to investigate the phylogenetic lineages and species delimitation in this section. Thirteen species are recognized, including four new species, namely A. griseorosea, A. molliuscula, A. parviexitialis, and A. subfuliginea. They are documented with morphological, multigene phylogenetic, and ecological evidence, line drawings, and photographs and compared with similar species. A key to the Chinese lethal Amanita species is provided.

PubMedID: 22802394 Arguably more mycophiles hunt true morels (Morchella) during their brief fruiting season each spring in the northern hemisphere than any other wild edible fungus. Concerns about overharvesting by individual collectors and commercial enterprises make it essential that science-based management practices and conservation policies are developed to ensure the sustainability of commercial harvests and to protect and preserve morel species diversity. Therefore, the primary objectives of the present study were to: (i) investigate the utility of the ITS rDNA locus for identifying Morchella species, using phylogenetic species previously inferred from multilocus DNA sequence data as a reference; and (ii) clarify insufficiently identified sequences and determine whether the named sequences in GenBank were identified correctly. To this end, we generated 553 Morchella ITS rDNA sequences and downloaded 312 additional ones generated by other researchers from GenBank using emerencia and analyzed them phylogenetically. Three major findings emerged: (i) ITS rDNA sequences were useful in identifying 48/62 (77.4%) of the known phylospecies; however, they failed to identify 12 of the 22 species within the species-rich Elata Subclade and two closely related species in the Esculenta Clade; (ii) at least 66% of the named Morchella sequences in GenBank are misidentified; and (iii) ITS rDNA sequences of up to six putatively novel Morchella species were represented in GenBank. Recognizing the need for a dedicated Webaccessible reference database to facilitate the rapid identification of known and novel species, we constructed Morchella MLST (http://www.cbs.knaw. nl/morchella/), which can be queried with ITS rDNA sequences and those of the four other genes used in our prior multilocus molecular systematic studies of this charismatic genus. © 2012 by The Mycological Society of America.

Species of the genus Retiboletus (Boletaceae, Boletales) in China are investigated based on morphology and phylogenetic analyses of DNA sequences from nuc rDNA internal transcribed spacer (ITS) and partial 28S regions and sequences from the translation elongation factor 1-a gene (tef1a). Six lineages are recovered among the collections studied. Five of these are documented and presented in the present paper, including three new species and two new combinations. The remaining species is not described due to the paucity of material. The specimens from China identified as "R. ornatipes" or "R. retipes" are in fact R. sinensis or R. kauffmanii, those labeled "R. griseus" are either R. fuscus or R. pseudogriseus A key to all known taxa of the genus is provided. Phylogenetic relationships of taxa within Retiboletus are partially resolved. A preliminary biogeographical analysis shows that allied species of Retiboletus between eastern Asia and North/Central America are common but there are no Retiboletus species common to both continents. Species of Retiboletus in Japan and southern China are conspecific or closely related.

The genus Leucoagaricus has been well studied in Europe. However, species diversity of Leucoagaricus in Asia remains poorly known, especially in the mountains of southwestern China, a hot spot for biodiversity. Based on morphological characters and molecular data, four new species are described, La. asiaticus, La. subcrystallifer, La. subpurpureolilacinus and La. truncatus. Detailed morphological descriptions, drawings of microstructures for novel taxa and comparisons with closely allied taxa are provided. Phylogenetic analyses inferred from internal transcribed spacer (ITS) and region 6-7 of the gene for the second largest subunit of RNA polymerase II (rpb2) sequences show that the novel taxa are nested within a well-supported clade jointly formed by members of Leucoagaricus section Rubrotincti and subgenus Sericeomyces.

The diversity of root-associated fungi associated with four ectomycorrhizal herbaceous species, Kobresia capillifolia, Carex parva, Polygonum macrophyllum and Potentilla fallens, collected in three sites of alpine meadows in southwestern China, was estimated based on internal transcribed spacer (ITS) rDNA sequence analysis of root tips. Three hundred seventy-seven fungal sequences sorted to 154 operational taxonomical units (sequence similarity of ≥ 97% across the ITS) were obtained from the four plant species across all three sites. Similar taxa (in GenBank with ≥ 97% similarity) were not found in GenBank and/or UNITE for most of the OTUs. Ectomycorrhiz a made up 64% of the fungi operational taxonomic units (OTUs), endophytes constituted 4% and the other 33% were unidentified root-associated fungi. Fungal OTUs were represented by 57% basidiomycetes and 43% ascomycetes. Inocybe, Tomentella/Thelophora, Sebacina, Hebeloma, Pezizomycotina, Cenococcum geophilum complex, Cortinarius, Lactarius and Helotiales were OTU-rich fungal lineages. Across the sites and host species the root-associated fungal communities generally exhibited low host and site specificity but high host and sampling site preference. Collectively our study revealed noteworthy diversity and endemism of root-associated fungi of alpine plants in this global biodiversity hotspot.

Amanita altipes and A. parvipantherina are members of section Amanita, while A. orientifulva and A. liquii are representatives of section Vaginatae. They are compared with similar species and illustrated with line drawings.

The genus Singerocybe (Tricholomataceae, Agaricales, Basidiomycota) has been the subject of controversy since its proposal in 1988. Its taxonomic foundation, species circumscription and geographical distribution have not yet been examined with molecular sequence data. In this study phylogenetic analyses on this group of fungi were conducted based on collections from Europe, eastern Asia, southern Asia, North America and Australia, with four nuclear markers, ITS, nrLSU, tefl-alpha and rpb2. Molecular phylogenetic analyses, together with morphological observations, strongly support Singerocybe as a monophyletic group and identify the vesicles in the pileal and stipe cuticle as a synapomorphy of this genus. Seven species are recognized in the genus, including one new species and four new combinations. Clitocybe trogioides and Clitocybe trogioides var. odorifera are synonyms of Singerocybe humilis and Singerocybe alboinfundibuliformis respectively. Most of these species are geographically restricted in their distributions. Furthermore our study expands the distribution range of Singerogbe from the North Temperate Zone to Australia (Tasmania) and tropical southern Asia.
