The phospholipid fatty acid composition of the sponge Amphimedon complanata was reinvestigated, and the 2-methoxy-13-methyltetradecanoic acid, 2-methoxy-14-methylpentadecanoic acid, and 2-methoxy-13-methylpentadecanoic acid were identified for the first time in nature. Structure characterization was accomplished by means of gas chromatographic retention times and gas chromatography-mass spectrometry. These acids could have originated from bacteria in symbiosis with the sponge.

The lipid composition of a Bacillus sp., isolated from Lake Pomorie in Bulgaria, was unusual and consisted of 26 different fatty acids between C12 and C26, with anteiso C15-C17 saturated fatty acids predominating. The furan fatty acid, 10,13-epoxy-11-methyloctadeca-10,12-dienoic acid, was also identified, a new finding for this genus. The hydrocarbons consisted of 30 different monounsaturated hydrocarbons, between C25 and C30, with the iso-iso, iso-anteiso, anteiso-anteiso, iso-normal, and anteiso-normal methyl branching for odd-numbered chains, and the iso-iso, iso-anteiso, iso-normal, and anteiso-normal methyl branching for even-numbered chains. The double bond positions in these hydrocarbons were determined by dimethyl disulfide derivatization followed by GC-MS, and the double-bond cis configuration was confirmed by infrared spectroscopy. Some previously unknown hydrocarbons in bacteria, such as (Z)-3,21-dimethyl-9-tricosene, (Z)-3,21-dimethyl-10-tricosene, (Z)-2,24-dimethyl-11-pentacosene, and (Z)-2,25-dimethyl-13-hexacosene were identified. Sterols were detected and were based on the sitosterol nucleus.

The fatty acid composition of a Pseudomonas sp. (Alteromonas) and its host, the dinoflagellate Ostreopsis lenticularis, vectors in ciguatera fish poisoning, has been studied. The major fatty acids in O. lenticularis were 16:0, 20:5n-3, and 22:6n-3, but 18:2n-6, 18:3n-3, and 18:n-3 were also identified. In contrast to other dinoflagellates, 1 8:5n-3 was not detected in O. lenticularis. Even-chain fatty acids such as 9-16:1, 11-18:1, and 13-20:1 predominated in the Pseudomonas sp. from O. lenticularis, but 1 6-20% of (E)-11-methyl-12-octadecenoic acid was also identified. The chirality of the latter was confirmed by total synthesis (28% overall yield) starting from oxacyclotridecan-2-one. The fatty acid compositions of two other Pseudomonas species, from the palytoxin-producing zoanthids Palythoa mamillosa and P. caribdea, were also studied and were similar to that of the Pseudomonas sp. from O. lenticularis. The possibility of using some of these fatty acids as chemotaxonomic lipids in identifying marine animals that consume toxic dinoflagellates or zoanthids is discussed.

The fatty acid composition of a new strain of Vibrio alginolyticus, found in the alga Cladophora coelothrix, was studied. Among 38 different fatty acids, a new fatty acid, 9-methyl-10-hexadecenoic acid and the unusual 11-methyl-12-octadecenoic acid, were identified. Linear alkylbenzene fatty acids, such as 10-phenyldecanoic acid, 12-phenyldodecanoic acid and 14-phenyltetradecanoic acid, were also found in V. alginolyticus. The alga contained 43% saturated fatty acids, and 28% C16-C20 polyunsaturated fatty acids of the n-3 and n-6 families.

The fatty acid composition of the tunicate Botryllus schlosseri and of two bacterial strains found within the tunicate, namely Vibrio parahaemolyticus and of an associated but previously unreported gram positive cocci were studied. The polyunsaturated fatty acids 6,9,12-octadecatrienoic acid, 5,8,11,14,17-eicosapentaenoic acid, and 4,7,10,13,16,19-docosahexaenoic acid were particularly abundant in B. schlosseri and were not detected in the two bacterial strains found in the tunicate. The iso/anteiso pair, 13-methyltetradecanoic acid and 12-methyltetradecanoic acid, were the principal fatty acids in the gram positive cocci, and the 9- and 11-hexadecenoic acids were particularly abundant in V. parahaemolyticus. The diunsaturated fatty acid 9,12-octadecadienoic acid was also shown to be present in V. parahaemolyticus. The fatty acid composition of a third bacterial strain, characterized as either a Pseudomonas or an Alteromonas species, and shown to be present only in the sea water from the Black Sea and not in B. schlosseri, is also reported. This is the first investigation on fatty acids from Black Sea bacteria.

The phospholipid fatty acid composition of the Calcarean sponge Leucosolenia canariensis was studied, and no delta 5,9 fatty acids were detected. These results are in contrast to the phospholipids from sponges belonging to the class Demospongiae where delta 5,9 fatty acids are predominant. Odd branched-chain fatty acids between 17 and 19 carbons accounted for 26% of the total fatty acids of L. canariensis, while straight-chain fatty acids between 16 and 22 carbons accounted for 61% of the total fatty acid composition. The sterol composition of L. canariensis is also reported, and only delta 5,7,22 sterols were observed.

The distribution of C14-C22 fatty acids in the phosphatidylethanolamines isolated from the sponges Agelas sp. and Spongia tampa was investigated. Selective changes with phospholipase A2 (from Agkistrodon halys blomhoffii) followed by thin-layer chromatographic separation of the resulting lysophosphatidylethanolamines and free fatty acids and subsequent methylation with HCl/MeOH and diazomethane, respectively, revealed that the 5,9-octadecadienoic acid and the 9,12-octadecadienoic acid present showed no preference for either position sn-1 or sn-2 in these phosphatidylethanolamines. The other saturated and unsaturated fatty acids with chains between 14 and 22 carbons long were also found to be equally distributed between positions sn-1 and sn-2 in the phosphatidylethanolamines in these sponges. The results contrast with what is known about the distribution in most mammalian phospholipids, such as the phosphatidylcholines from human erythrocytes, where octadecadienoic acid typically occupies the sn-2 position.

The very long-chain fatty acids, (5E,9Z)-6-bromo-5,9-tetracosadienoic, (5E,9Z)-6-bromo-23-methyl-5,9-tetracosadienoic, (5E,9Z)-6-bromo-5,9-pentacosadienoic and (5E,9Z)-6-bromo-24-methyl-5,9- pentacosadienoic acids, were identified in the phospholipids (mainly phosphatidylethanolamine) of the sponge Agelas sp. Structure elucidation was accomplished by means of mass spectrometry and chemical transformations, including deuteration with Wilkinson's catalyst. All of the sterols from the sponge had the delta 5,7 nucleus, with 24-methylcholesta-5,7,22-trien-3 beta-ol (ergosterol) and 24-ethyl-cholesta-5,7, 22-trien-3 beta-ol being the most abundant.

The Caribbean sponge Smenospongia aurea revealed the presence of six novel branched alpha-hydroxy fatty acids: 2-hydroxy-17-methyloctadecanoic acid, 2-hydroxy-21-methyldocosanoic acid, 2-hydroxy-22-methyltricosanoic acid, and 2-hydroxy-22-methyltetracosanoic acid, 2-hydroxy-24-methylpentacosanoic acid, and 2-hydroxy-23-methylpentacosanoic acid. These novel alpha-hydroxy fatty acids were associated with phosphatidylethanolamine. The sponges Aplysina lacunosa and Aplysina fistularis also contained considerable amounts of alpha-hydroxy fatty acids, the very long-chain 5,9,23-tricontatrienoic acid (30:3), and phytanic acid. The sterol composition of the three sponges was also studied. It indicated that A. lacunosa and A. fistularis contained large amounts of aplysterol and verongulasterol, while S. aurea did not show any of these sterols. The results are discussed in terms of the taxonomy of the species.

The novel (5Z)-2-methoxy-5-hexadecenoic acid (1) was identified in the phospholipids of the sponge Tethya crypta while the also novel acid (6Z)-2-methoxy-6-hexadecenoic acid (2) was found in the phospholipids of the Caribbean sponge Spheciospongia cuspidifera. The methoxy-fatty acids were mainly associated with phosphatidylethanolamine. The double bond positions were determined by gas chromatography/mass spectrometry on the corresponding dimethyl-disulfide adducts and the double bond stereochemistry was ascertained by gas chromatography/Fourier transform infrared spectroscopy. The fatty acid composition of the two sponges is reported.

The novel fatty acids (Z)-6-nonadecenoic acid (1) and (Z)-17-pentacosenoic acid (2) were characterized in the sponge Geodia gibberosa. These fatty acids were mainly found in phosphatidylethanolamine and phosphatidylcholine.

The first trans fatty acids from a sponge have been isolated from Plakortis halichondroides. The sponge was shown to contain the iso acids (5E,9E)-19-methyl-5,9-eicosadienoic, (5E,9E)-20-methyl-5,9-heneicosadienoic and (5E,9E)-21-methyl-5,9-docosadienoic acid, as well as the anteiso acids (5E,9E)-19-methyl-5,9-heneicosadienoic and (5E,9E)-20-methyl-5,9-docosadienoic acid together with the straight chain (5E,9E)-5,9-docosadienoic acid. The acids were shown by gas chromatography, Fourier transform infrared and 13C nuclear magnetic resonance to contain the trans configuration. An eicosadienoic acid, namely (6E,14E)-6,14-eicosadienoic acid, and 12-methyl-5-octadecenoic acid were also identified in a sponge for the first time. The fatty acids were shown to be the principal constituents of phosphatidylethanolamine and phosphatidylcholine. No sterols were found in the sponge. The results presented in this work should be helpful in taxonomy of Homoscleromorpha.

The phospholipid fatty acids from the sponge Spheciospongia cuspidifera were studied revealing the presence of the rare 10-octadecenoic acid (10-18:1) and a new 2-methoxyhexadecenoic acid. The phospholipid fatty acids from Thalysias juniperina were also studied revealing the presence of the hitherto unreported 18-hexacosenoic acid (18-26:1). These results tend to indicate that the biosynthetic pathway from 10-18:1 to 18-26:1 may be operative in nature. The phospholipid mixture from the sponges was also analyzed by 31P-NMR and shown to mainly consist of phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol, and phosphatidylglycerol. Phosphatidylcholine was not found in the sponges analyzed in this work.

The first alpha-hydroxy fatty acids from a marine sponge, namely 2-hydroxydocosanoic and 2-hydroxytricosanoic, were identified in the Caribbean sponge Amphimedon compressa. These acids were found to occur in phosphatidylethanolamine and phosphatidylserine and constituted 52% of the total fatty acid mixture of this sponge. The long chain fatty acids 5,9,23-nonacosatrienoic (29:3) and 5,9,23-tricontatrienoic (30:3), as well as a new tetratricontatetraenoic (34:4) acid, were also found in A. compressa.

The phospholipid fatty acids from the Caribbean sponge Ectyoplasia ferox were studied. The novel fatty acids 25-methyl-5,9-heptacosadienoic (1) and 26-methyl-5,9-heptacosadienoic (2) were identified in 3.4 and 2.0% abundance, respectively, representing the longest set of delta 5,9 iso and anteiso acids yet isolated from a marine sponge. The new acid 10,13-dimethyltetradecanoic (3), the unusual acid 15-methyl-11-hexadecenoic (4) and the also novel acid 9-methyl-11-hexadecenoic (5) were also identified in E. ferox. The principal sterols isolated from E. ferox were 24-ethylcholest-5-en-3 beta-ol (46%) and 24(R)-methylcholesta-5,22-dien-3 beta-ol (14%).

The fatty acid 5,9-hexadecadienoic was identified in the sponge Chondrilla nucula. This is the shortest fatty acid with the delta 5,9 unsaturation yet isolated from a marine sponge.

The novel fatty acid 7-methyl-8-hexadecenoic (1) was identified in the marine spongeDesmapsama anchorata. Other interesting fatty acids identified were 14-methyl-8-hexadecenoic (2), better known through its methyl ester as one of the components of the sex attractant of the female dermestid beetle, and the saturated fatty acid 3-methylheptadecanoic (3), known to possess larvicidal activity. The main phospholipid fatty acids encountered inD. anchorata were palmitic (16∶0), behenic (22∶0) and 5,9-hexacosadienoic acid (26∶2), which together accounted for 50% of the total phospholipid fatty acid mixture.

The alpha-hydroxy fatty acids 2-hydroxy-eicosanoic (1) acid, 2-hydroxyheneicosanoic (2) acid, 2-hydroxydocosanoic (3) acid, 2-hydroxytetracosanoic (4) acid, 2-hydroxy-23-methyl-tetracosanoic acid and 2-hydroxypentacosanoic (5) acid were isolated from the Caribbean sponges Verongula gigantea and Aplysina archeri. The very long chain fatty acids 5,9-nonacosadienoic acid (29:2) and 5,9,23-tricontatrienoic acid (30:3) were also identified together with the iso-prenoid fatty acid 3,7,11,15-tetramethylhexadecanoic (phytanic) acid that seems to be common in the Aplysinidae. A. archeri contained an extremely long chain fatty acid tentatively characterized as dotricontaenoic (32:1) acid. These acids were found to occur in phosphatidylserine, phosphatidylinositol, phosphatidylethanolamine, phosphatidylcholine and traces of phosphatidylglycerol.

The burrowing sponges Anthosigmella varians and Spheciospongia vesparium were found to be rich in the isoprenoid phospholipid fatty acid 4,8,12-trimethyltridecanoic (5.2% and 23%, respectively, of the total fatty acid composition), while the burrowing sponge Chondrilla nucula and the demosponge Agelas dispar contained the acid 3,7,11,15-tetramethylhexadecanoic (13.8% and 8.6%, respectively, of the total phospholipid fatty acid composition). No other isoprenoid fatty acid was found, and the two acids described in this work did not occur concomitantly in the same sponge.

The phospholipid fatty acids from the sponge Euryspongia rosea were studied revealing the presence of the new 6,11-eicosadienoic (20:2) acid and the rare 6,11-octadecadienoic acid (18:2). The isolation of these 6,11-dienoic acids reveals the presence of new biosynthetic possibilities in sponges and suggests the presence of an active delta 6 desaturase in E. rosea. Other acids isolated from E. rosea include 12-eicosenoic (20:1) and 17-tetracosenoic acid (24:1). The major phospholipids encountered in the sponge were phosphatidylethanolamine (PE), phosphatidylserine (PS), and phosphatidylcholine (PC).

The rare phospholipid fatty acids 3,7,11-trimethyldodecanoic (1), 5,9-hexadecadienoic (2) and 12-methyl-hexadecanoic (3) were identified in the marine spongeXestospongia muta. Branched fatty acids inX. muta accounted for 35% of the total fatty acid mixture. It was observed that the occurrence of the 5,9-hexadecadienoic acid (2) coincides with the complete absence of the very long chain fatty acid 5,9-hexacosadienoic. The acid 5,9,19-octacosatrienoic seems to be found in mostXestospongia species.
