This paper reports the development of Metal-amplified Density Assays, or MADAs – a method of conducting quantitative or multiplexed assays, including immunoassays, by using Magnetic Levitation (MagLev) to measure metal-amplified changes in the density of beads labeled with biomolecules. The binding of target analytes (i.e. proteins, antibodies, antigens) to complementary ligands immobilized on the surface of the beads, followed by a chemical amplification of the binding in a form that results in a change in the density of the beads (achieved by using gold nanoparticle-labeled biomolecules, and electroless deposition of gold or silver), translates analyte binding events into changes in density measureable using MagLev. A minimal model based on diffusion-limited growth of hemispherical nuclei on a surface reproduces the dynamics of the assay. A MADA – when performed with antigens and antibodies – is called a Density-Linked Immunosorbent Assay, or DeLISA. Two immunoassays provided a proof of principle: a competitive quantification of the concentration of neomycin in whole milk, and a multiplexed detection of antibodies against Hepatitis C virus NS3 protein and syphilis T. pallidum p47 protein in serum. MADAs, including DeLISAs, require, besides the requisite biomolecules and amplification reagents, minimal specialized equipment (two permanent magnets, a ruler or a capillary with calibrated length markings) and no electrical power to obtain a quantitative readout of analyte concentration. In this paper, we present a fluorescence activated sorter realized in a continuous flow microfluidic chip. Sorting is achieved by deflecting a focused particle stream with short acoustic bursts (2.5 ms), in a fluorescence activated configuration. The system utilizes two-dimensional acoustic pre-focusing, using a single actuation frequency, to position all particles in the same fluid velocity regime at flow rates up to 1.7 mL min−1. Particles were sorted based on their fluorescence intensities at throughputs up to 150 particles s−1. The highest purity reached was 80% when sorting at an average rate of 50 particles s−1. The average recovery of a sort was 93.2 ± 2.6%. The presented system enables fluorescence activated cell sorting in a continuous flow microfluidic format that allows aseptic integration of downstream microfluidic functionalities, opening for medical and clinical applications. This study describes the function, optimization, and demonstration of a new class of passive, low-cost microfluidic flow meters based on birefringent chitosan biomembranes analyzed by polarized microscopy. We subjected the membrane to dynamic flow conditions while monitoring the real-time response of its optical properties. We obtained figures of merit, including the linear response operating range (0 to 65 μL min−1), minimum response time (250 ms), sensitivity (2.03% × 10−3 μL−1 min), and minimum sensor longevity (1 week). In addition, possible sources of interference were identified. Finally, we demonstrate the membrane as a low-cost flow rate measurement device for the close loop control of a commercial pressure-driven pump. Preliminary experiments using a basic PID controller with the membrane-based flow rate measurement device showed that stable control could be achieved and the system could reach steady-state behavior in less than 15 seconds. In this work, we demonstrate a novel method for multi-domain analysis of properties of analytes in volumes as small as picoliters, combining electrochemistry and optical measurements. A microcavity in-line Mach–Zehnder interferometer (μIMZI) obtained in a standard single-mode optical fiber using femtosecond laser micromachining was able to accommodate a microelectrode and optically monitor electrochemical processes inside the fiber. The interferometer shows exceptional sensitivity to changes in the optical properties of analytes in the microcavity. We show that the optical readout follows the electrochemical reactions. Here, the redox probe (ferrocenedimethanol) undergoing reactions of oxidation and reduction changes the optical properties of the analyte (refractive index and absorbance) that are monitored using the μIMZI. Measurements have been supported by numerical analysis of both optical and electrochemical phenomena. On top of the capability of the approach to perform analysis on a microscale, the difference between oxidized and reduced forms in the near-infrared region can be measured using the μIMZI, which is hardly possible using other optical techniques. Lateral flow assays (LFAs) are rapid, inexpensive, easy-to-manufacture and -use tests widely employed in medical and environmental applications, particularly in low resource settings. Historically, LFAs have been stigmatized as having limited sensitivity. However, as their global usage expands, extensive research has demonstrated that it is possible to substantially improve LFA sensitivity without sacrificing their advantages. In this critical review, we have compiled state-of-the-art approaches to LFA sensitivity enhancement. Moreover, we have organized and evaluated these approaches from a system-level perspective, as we have observed that the advantages and disadvantages of each approach have arisen from the integrated and tightly interconnected chemical, physical, and optical properties of LFAs. This paper presents a rapid, ultra-low-cost approach to fabricate microfluidic devices using a polyolefin shrink film and a digital craft cutter. The shrinking process (with a 95% reduction in area) results in relatively uniform and consistent microfluidic channels with smooth surfaces, vertical sidewalls, and high aspect ratio channels with lateral resolutions well beyond the tool used to cut them. The thermal bonding of the layers results in strongly bonded devices. Complex microfluidic designs are easily designed on the fly and protein assays are also readily integrated into the device. In developing countries, there are often limited resources available to provide important medical diagnostics, which severely limits our ability to diagnose conditions and administer proper treatment, leading to high mortality rates for treatable conditions. Here, we propose a multiplex tool capable of density-based cell sorting via magnetic focusing in parallel with fluorescence imaging to provide highly specific clinical assays. While many cell sorting techniques and fluorescence microscopes generally are costly and require extensive user training, limiting accessibility and usability in developing countries, this device is compact, low-cost, and portable. The device can separate cells on the basis of density, which can be used to identify cell type and cell activity, and image the cells in either brightfield, darkfield, or fluorescent imaging modes using the built-in smartphone camera. The combination of these two powerful and versatile techniques – magnetic focusing and fluorescence imaging – will make this platform broadly applicable to a range of biomedical assays. Clinical applications include cell cytometry and immunocytochemistry-based assays in limited-resource settings, which can ultimately help to improve worldwide accessibility to medical diagnostics. This work introduces a method of continuous particle separation through standing surface acoustic wave (SSAW)-induced acoustophoresis in a microfluidic channel. Using this SSAW-based method, particles in a continous laminar flow can be separated based on their volume, density and compressibility. In this work, a mixture of particles of equal density but dissimilar volumes was injected into a microchannel through two side inlets, sandwiching a deonized water sheath flow injected through a central inlet. A one-dimensional SSAW generated by two parallel interdigital transducers (IDTs) was established across the channel, with the channel spanning a single SSAW pressure node located at the channel center. Application of the SSAW induced larger axial acoustic forces on the particles of larger volume, repositioning them closer to the wave pressure node at the center of the channel. Thus particles were laterally moved to different regions of the channel cross-section based on particle volume. The particle separation method presented here is simple and versatile, capable of separating virtually all kinds of particles (regardless of charge/polarization or optical properties) with high separation efficiency and low power consumption. We have demonstrated nanofabrication with commercialized cellulose acetate. Cellulose acetate is used for bulk nanofabrication and surface nanofabrication. In bulk nanofabrication, cellulose acetate reacts with an e-beam and permanent patterns are formed in it instead of being transferred to other substrates. We have studied the nano relief modulation performance of cellulose acetate before and after development. The depth of the nanopatterns is magnified after development, and is varied by exposing dosage and line width of the pattern. The thinnest 65 nm wide line is achieved in the bulk fabrication. We also demonstrate a binary phase Fresnel lens array which is directly patterned in a cellulose acetate sheet. Because of its unique mechanical and optical properties, cellulose is a good candidate for a template material for soft imprinting lithography. In the surface nanofabrication, cellulose acetate thin film spin-coated on silicon wafers is employed as a new resist for e-beam lithography. We achieved 50 nm lines with 100 nm pitches, dots 50 nm in diameter, and single lines with the smallest width of 20 nm. As a new resist of e-beam lithography, cellulose acetate has high resolution comparable with conventional resists, while having several advantages such as low cost, long stock time and less harmfulness to human health. Polyolefins are finding increased popularity in microfluidic applications due to their attractive mechanical, processing, and optical properties. While intricate features are typically realized in these thermoplastics by hot embossing and injection molding, such fabrication approaches are expensive and slow. Here, we apply our shrink-induced approach—first demonstrated with polystyrene ‘Shrinky-Dink’ sheets—to create micro- and nanostructures with cross-linked polyolefin thin films. These multi-layered films shrink by 95% and with greater uniformity than the Shrinky-Dinks. With such significant reduction in size, along with attractive material properties, such commodity films could find important applications in low cost microfluidic prototyping as well as in point-of-care diagnostics. In this technical note, we demonstrate the ability to rapidly and easily create unique microstructures, increase microarray feature density, and even induce self-assembled integrated metallic nanostructures with these shrink wrap films.
