Abstract

The structure-based optimization of a series of estrogen receptor-beta (ERbeta) selective ligands is presented. Using ab initio quantum mechanics, it is demonstrated that certain functional groups incorporated during lead optimization are able to experience a differential interaction with ERalpha Met(sub 421) relative to ERbeta IIe(sub 373) based on both electrostatic and dispersive effects.
