Industrial-scale thermal separation processes have contributed greatly to the rise in carbon dioxide emissions. Porous materials, such as metal-organic frameworks (MOFs), can potentially reduce these emissions by achieving nonthermal chemical separations through the physical adsorption of targeted species with high selectivity. Hydrogen-bonded organic frameworks (HOFs), self-assembled from strategically pre-designed molecular tectons with complementary hydrogen-bonding patterns, are rapidly evolving into a novel and important class of porous materials. In addition to their common features shared with other functionalized porous materials constructed from modular building blocks, the intrinsically flexible and reversible H-bonding connections endow HOFs with straightforward purification procedures, high crystallinity, solution processability, and recyclability. These unique advantages of HOFs have attracted considerable attention across a broad range of fields, including gas adsorption and separation, catalysis, chemical sensing, and electrical and optical materials. However, the relatively weak H-bonding interactions within HOFs can potentially limit their stability and potential use in further applications. To that end, this Perspective highlights recent advances in the development of chemically and thermally robust HOF materials and systematically discusses relevant design rules and synthesis strategies to access highly stable HOFs.

The hydrolytic loss of coding bases from cellular DNA is a common and unavoidable reaction. The resulting abasic sites can undergo β-elimination of the 3’-phosphoryl group to generate a strand break with an electrophilic α,β-unsaturated aldehyde residue on the 3’-terminus. The work reported here provides evidence that the thiol residue of the cellular tripeptide glutathione rapidly adds to the alkenal group on the 3’-terminus of an AP-derived strand break. The resulting glutathionylated adduct is the only major cleavage product observed when β-elimination occurs at an AP site in the presence of glutathione. Formation of the glutathionylated cleavage product is reversible but, in the presence of physiological concentrations of glutathione, the adduct persists for days. Biochemical experiments provided evidence that the 3’-phosphodiesterase activity of the enzyme apurinic/apyrimidinic endonuclease (APE1) can remove the glutathionylated sugar remnant from an AP-derived strand break to generate the 3’OH residue required for repair via base excision or single-strand break repair pathways. The results suggest that a previously unrecognized 3’glutathionylated sugar remnant – and not the canonical α,β-unsaturated aldehyde end group – may be the true strand cleavage product arising from β-elimination at an abasic site in cellular DNA. This work introduces the 3’glutathionylated cleavage product as the major blocking group that must be trimmed to enable repair of abasic site-derived strand breaks by the base excision repair or single-strand break repair pathways.

One of the most widely employed strategies to produce chiral molecules involves the asymmetric hydrogenation of functionalized olefins using rhodium catalysts. Despite their excellent performance, the exact identity of the active Rh species is still ambiguous as each site may plausibly feature one or two phosphorus ligands. In this work, we used a sequential postsynthetic modification approach to successfully incorporate single-site Rh species into a zirconium-based metal-organic framework comprised of chiral spinol-based ligands. These Rh species feature one phosphorus ligand per Rh, which contrasts with the molecular analogue that contains two phosphorus ligands per Rh site. Following extensive characterization of the Rh-monophosphorus material using techniques including solid-state NMR and extended X-ray absorption fine-structure (EXAFS) spectroscopy, we studied their catalytic performance in the asymmetric hydrogenations of enamides and α-dehydroamino acid esters and observed excellent yields and enantioselectivities (up to 99.9% ee). Notably, the Rh-monophosphorus catalyst is 5 times more active than the homogeneous Rh-biphosphorus control, which we attributed to the higher activity of the single-site Rh-monophosphorus species and the confined MOF cavities that can enrich reactants. In addition, we observed a unique topology-dependent behavior in which linker expansion leads to the formation of a novel Zr-MOF with a distinct 4,8-connected net that cannot be phosphorylated, presumably due to intense tensile strain and steric repulsion present within this framework. Elpasolite- and cryolite-type oxyfluorides can be regarded as superstructures of perovskite and exhibit structural diversity. While maintaining a similar structural topology with the prototype structures, changes in the size, electronegativity, and charge of cation and/or anion inevitably lead to structural evolution. Therefore, the nominal one-to-one relation suggested by a doubled formula of perovskite does not guarantee a simple 2-fold superstructure for many cases. Herein, the commensurately modulated perovskite-like K3TiOF5 was refined at 100 K from single-crystal X-ray diffraction data by using a pseudotetragonal subcell with lattice parameters of a = b = 6.066(2) A and c = 8.628(2) A. The length of the modulation vector was refined to 0.3a* + 0.1b* + 0.25c*. In the commensurate supercell of K3TiOF5, the B-site Ti4+ and K+ cations in [TiOF5]3- and [KOF5]6- octahedral units were found to be significantly displaced from the average atomic positions refined in the subcell. The displacements of the K+ cations are ±0.76 A, and those for the Ti4+ cations are approximately ±0.13 A. One- and two-dimensional solid-state 19F NMR measurements revealed two tightly clustered groups of resonances in a ratio of ca. 4:1, assigned to equatorial and axial fluorine, respectively, consistent with local [TiOF5]3- units. S/TEM results confirmed the average structure. Electronic structure calculations of the idealized I4mm subcell indicate the instability to a modulated structure arises from soft optical modes that is controlled by the octahedrally coordinated B-site potassium ions in the cryolite-type structure.

Ligand-induced protein degradation has emerged as a compelling approach to promote the targeted elimination of proteins from cells by directing these proteins to the ubiquitin-proteasome machinery. So far, only a limited number of E3 ligases have been found to support ligand-induced protein degradation, reflecting a dearth of E3-binding compounds for proteolysis-targeting chimera (PROTAC) design. Here, we describe a functional screening strategy performed with a focused library of candidate electrophilic PROTACs to discover bifunctional compounds that degrade proteins in human cells by covalently engaging E3 ligases. Mechanistic studies revealed that the electrophilic PROTACs act through modifying specific cysteines in DCAF11, a poorly characterized E3 ligase substrate adaptor. We further show that DCAF11-directed electrophilic PROTACs can degrade multiple endogenous proteins, including FBKP12 and the androgen receptor, in human prostate cancer cells. Our findings designate DCAF11 as an E3 ligase capable of supporting ligand-induced protein degradation via electrophilic PROTACs.

Racemates have recently received attention as nonlinear optical and piezoelectric materials. Here, a machine-learning-assisted composition space approach was applied to synthesize the missing M = Ti, Zr members of the Δ,Λ-[Cu(bpy)2(H2O)]2[MF6]2·3H2O (M = Ti, Zr, Hf; bpy = 2,2'-bipyridine) family (space group: Pna21). In each (CuO, MO2)/bpy/HF(aq) (M = Ti, Zr, Hf) system, the polar noncentrosymmetric racemate (M-NCS) forms in competition with a centrosymmetric one-dimensional chain compound (M-CS) based on alternating Cu(bpy)(H2O)22+ and MF62- basic building units (space groups: Ti-CS (Pnma), Zr-CS (P1), Hf-CS (P2/n)). Machine learning models were trained on reaction parameters to gain unbiased insight into the underlying statistical trends in each composition space. A human-interpretable decision tree shows that phase selection is driven primarily by the bpy:CuO molar ratio for reactions containing Zr or Hf, and predicts that formation of the Ti-NCS compound requires that the amount of HF present be decreased to raise the pH, which we verified experimentally. Predictive leave-one-metal-out (LOO) models further confirm that behavior in the Ti system is distinct from that of the Zr and Hf systems. The chemical origin of this distinction was probed via fluorine K-edge X-ray absorption spectroscopy. Pre-edge features in the F1s X-ray absorption spectra reveal the strong ligand-to-metal π bonding between Ti(3d - t2g) and F(2p) states that distinguishes the TiF62- anion from the ZrF62- and HfF62- anions.

The development of adsorbents with molecular precision offers a promising strategy to enhance storage of hydrogen and methane─considered the fuel of the future and a transitional fuel, respectively─and to realize a carbon-neutral energy cycle. Herein we employ a postsynthetic modification strategy on a robust metal-organic framework (MOF), MFU-4l, to boost its storage capacity toward these clean energy gases. MFU-4l-Li displays one of the best volumetric deliverable hydrogen capacities of 50.2 g L-1 under combined temperature and pressure swing conditions (77 K/100 bar → 160 K/5 bar) while maintaining a moderately high gravimetric capacity of 9.4 wt %. Moreover, MFU-4l-Li demonstrates impressive methane storage performance with a 5-100 bar usable capacity of 251 cm3 (STP) cm-3 (0.38 g g-1) and 220 cm3 (STP) cm-3 (0.30 g g-1) at 270 and 296 K, respectively. Notably, these hydrogen and methane storage capacities are significantly improved compared to those of its isoreticular analogue, MFU-4l, and place MFU-4l-Li among the best MOF-based materials for this application.

Interstrand DNA cross-links (ICLs) are cytotoxic because they block the strand separation required for read-out and replication of the genetic information in duplex DNA. The unavoidable formation of ICLs in cellular DNA may contribute to aging, neurodegeneration, and cancer. Here we describe the formation and properties of a structurally complex ICL derived from an apurinic/apyrimidinic (AP) site, which is one of the most common endogenous lesions in cellular DNA. The results characterize a cross-link arising from aza-Michael addition of the N(2)-amino group of a guanine residue to the electrophilic sugar remnant generated by spermine-mediated strand cleavage at an AP site in duplex DNA. An α,β-unsaturated iminium ion is the critical intermediate involved in ICL formation. Studies employing the bacteriophage ϕ29 polymerase provided evidence that this ICL can block critical DNA transactions that require strand separation. The results of biochemical studies suggest that this complex strand break/ICL might be repaired by a simple mechanism in which the 3’-exonuclease action of the enzyme apurinic/apyrimidinic endonuclease (APE1) unhooks the cross-link to initiate repair via the single-strand break repair pathway.

Organophosphorus nerve agents, a class of extremely toxic chemical warfare agents (CWAs), have remained a threat to humanity because of their continued use against civilian populations. To date, Zr(IV)-based metal organic framework (MOFs) are the most prevalent nerve agent hydrolysis catalysts, and relatively few reports disclose MOFs containing nodes with other Lewis acidic transition metals. In this work, we leveraged this synthetic tunability to explore how the identity of the transition metal node in the M-MFU-4l series of MOFs (M = Zn, Cu, Ni, Co) influences the catalytic performance toward the hydrolysis of the nerve agent simulant dimethyl (4-nitrophenyl)phosphate (DMNP). Experimental studies reveal that Cu-MFU-4l exhibits the best performance in this series with a half-life for hydrolysis of ∼2 min under these conditions. In contrast, both Ni- and Co-MFU-4l demonstrate significantly slower reactivity toward DMNP, as they both fail to surpass 30% conversion of DMNP after 1 h under analogous conditions. Further modification of the active site within Cu-MFU-4l is possible, and we found that although the identity of the anion coordinated to the Cu(II)-X (X = Cl-, HCOO-, ClO4-, NO3-) active site has little influence on the catalytic performance, reduction of the Cu(II) sites yields nodes that contain Cu(I) ions in a trigonal geometry with open metal sites, leading to remarkable catalytic activity with a half-life for hydrolysis less than 2 min. Computational studies indicate the Cu(I) sites exhibit stronger binding affinities than Cu(II) to both water and DMNP, which corroborates the experimental results.

MXenes, derived from layered MAX phases, are a class of two-dimensional materials with emerging applications in energy storage, electronics, catalysis, and other fields due to their high surface areas, metallic conductivity, biocompatibility, and attractive optoelectronic properties. MXene properties are heavily influenced by their surface chemistry, but a detailed understanding of the surface functionalization is still lacking. Solid-state nuclear magnetic resonance (NMR) spectroscopy is sensitive to the interfacial chemistry, the phase purity including the presence of amorphous/nanocrystalline phases, and the electronic properties of the MXene and MAX phases. In this work, we systematically study the chemistry of Nb MAX and MXene phases, Nb2AlC, Nb4AlC3, Nb2CTx, and Nb4C3Tx, with their unique electronic and mechanical properties, using solid-state NMR spectroscopy to examine a variety of nuclei (1H, 13C, 19F, 27Al, and 93Nb) with a range of one- and two-dimensional correlation, wide-line, high-sensitivity, high-resolution, and/or relaxation-filtered experiments. Hydroxide and fluoride terminations are identified, found to be intimately mixed, and their chemical shifts are compared with other MXenes. This multinuclear NMR study demonstrates that diffraction alone is insufficient to characterize the phase composition of MAX and MXene samples as numerous amorphous or nanocrystalline phases are identified including NbC, AlO6 species, aluminum nitride or oxycarbide, AlF3·nH2O, Nb metal, and unreacted MAX phase. To the best of our knowledge, this is the first study to examine the transition-metal resonances directly in MXene samples, and the first 93Nb NMR of any MAX phase. The insights from this work are employed to enable the previously elusive assignment of the complex overlapping 47/49Ti NMR spectrum of Ti3AlC2. The results and methodology presented here provide fundamental insights on MAX and MXene phases and can be used to obtain a more complete picture of MAX and MXene chemistry, to prepare realistic structure models for computational screening, and to guide the analysis of property measurements.
