The genus Stevia comprises approximately 200 species, which are distributed in North and South America, and are representative of the species diversity of the Asteraceae in the New World. We reconstructed the phylogenetic relationships using sequences of ITS and cpDNA and estimated the divergence times of the major clade of this genus. Our results suggested that Stevia originated in Mexico 7.0–7.3 million years ago (Mya). Two large clades, one with shrub species and another with herb species, were separated at about 6.6 Mya. The phylogenetic reconstruction suggested that an ancestor of Stevia was a small shrub in temperate pine–oak forests and the evolutionary change from a shrub state to a herb state occurred only once. A Brazilian clade was nested in a Mexican herb clade, and its origin was estimated to be 5.2 Mya, suggesting that the migration from North America to South America occurred after the formation of the Isthmus of Panama. The species diversity in Mexico appears to reflect the habitat diversity within the temperate pine–oak forest zone. The presence of many conspecific diploid–polyploid clades in the phylogenetic tree reflects the high frequency of polyploidization among the perennial Stevia species.

Zanthoxylum ailanthoides Siebold & Zucc. is one of the most frequently encountered pioneer trees in Japanese warm-temperate evergreen oak forests. Our previous study in one region of Japan suggested high levels of population differentiation and putative natural selection acting on one of the nuclear loci analyzed. Here, we extend our analysis to study the genetic structure of 10 populations of Z. ailanthoides across Japan using 9 simple sequence repeat (SSR) loci for a better understanding of its genetic structure. First, the southernmost population (Kagoshima) in the samples was found to have the highest genetic diversity, suggesting there was a glacial refugium at or near the location of the population. Second, relatively strong genetic differentiation was found among populations, and there was a positive correlation between genetic distances and geographic distances (Mantel test; P < 0.001). Based on this information, we analyzed nucleotide variation at the putatively selected locus homologous to the gene encoding the ADP-glucose pyrophosphorylase large subunit (agpL). Despite the strong genetic differentiation among populations suggested by the SSR loci, the agpL locus was monomorphic in almost all populations analyzed. The results of this study strongly supported the possibility of a selective sweep at or near the agpL locus.

Similar distribution ranges shared by closely related plant species may have been shaped through different migration histories if those species have differing habitat preference. To test this hypothesis, phylogeographical patterns and population genetic structures were compared between two sister Viola species: Viola eizanensis preferring woodland and V. chaerophylloides var. sieboldiana preferring grassland, both being native to the Japanese Archipelago. Amplified fragment length polymorphism (AFLP) was used for phylogenetic reconstruction, together with Bayesian ancestry analysis, AMOVA, analysis of genetic diversity statistics, and analysis of the relative contribution of each population to total diversity. The results indicated that V. eizanensis had two distinct lineages occurring in the western and eastern part of Japan, but such lineages are not distinct in V. chaerophylloides var. sieboldiana. Both species exhibited the low genetic diversity and high between-population differentiation typical of selfing plants. In V. chaerophylloides var. sieboldiana, one particular population made a significantly higher contribution to the total heterozygosity (H T), whereas in V. eizanensis, no population was identified as making a particularly higher contribution to H T. These findings suggest that V. eizanensis had been isolated in two large glacial refugia, whereas populations of V. chaerophylloides var. sieboldiana were restricted to a single small refuge. Different light requirements between these two closely related species probably caused these differing responses to climatic change during the last ice age.

Reproductive failure of backcross could play important roles in determining the evolutionary outcome of hybridization. However, such studies have been somewhat fewer than those of the F1-producing cross and F1 x F1 cross. We conducted hand-pollination backcross experiments using Hemerocallis fulva, H. citrina, and their F1 hybrids. Seed set per flower of the backcrosses reduced to about 50% of the control cross, irrespective of ovule parent species. This symmetrical seed set reduction on the backcross might be caused by the death of backcross embryo as a result of Dobzhansky-Muller incompatibility by recessive alleles.

were examined by sequencing the internal transcribed spacers (ITS) of nuclear ribosomal DNA and restriction site analysis of chloroplast DNA. Molecular data provided strong evidence that (1) this genus originated in North America, (2) the genus diverged into three morphological species groups, Eutrochium, Traganthes and Uncasia in North America, and (3) one of the North American Uncasia lineages migrated into temperate Europe and eastern Asia over the Bering land bridge. The estimated divergence times support a late Miocene to early Pliocene migration from North America to Eurasia via the Bering land bridge. A European species was sister to all of the eastern Asian species examined. The disjunct distribution pattern of the genus Eupatorium is incongruent with the classical Arcto-Tertiary geoflora concept.

(Juncaginaceae). Germination tests were carried out at three salinity levels (0,200,400 mM NaCl in which seeds were exposed to increasing- or decreasing temperatures. Effects of moist-chilling pretreatment (stratification) in 0,200 and 400 mM NaCl on seed germination at 0,200 and 400 mM NaCl, respectively, were also examined. Under the highest salinity condition (400 mM NaCl), no germination was observed. The seeds germinated very well (88%) in fresh-water after 5-month moist-chilling pretreatment. Longer moist-chilling pretreatment resulted in higher germination percentages. Moist chilling pretreatment in 0,200 and 400 mM NaCl for 5 month enhanced germination percentages to 10% (in 400 mM NaCl) –88% (in 0 mM NaCl).

Statistical tests regarding correlations between changes in chromosome number, total chromosome length, mean chromosome length, karyotypic asymmetry and chromosome length heterogeneity and changes in habit were performed based on the matK molecular phylogenetic tree. The reductions in chromosome number and total chromosome length, and the increases in mean chromosome length, chromosome length heterogeneity and karyotypic asymmetry were found to be correlated with the change in habit from perennial to annual. A reduction in total chromosome length is favored to shorten the mitotic cell cycle and to produce smaller cells conducive to more rapid development of smaller annuals under the time-limited environment. Stepwise dysploidal reductions in chromosome number were achieved through the translocation of large chromosome segments onto other chromosomes, followed by the loss of a centromere, resulting in one fewer linkage group and one fewer haploid chromosome. The correlations between the dysploidal reduction in chromosome number and the increases in mean chromosome length, length heterogeneity and asymmetry in karyotype can be attributed to this mode of chromosomal change. These changes occurred independently in several different lineages in Brachyscome.

Similarity among the floral scents of hawkmoth-pollinated plants was investigated with headspace samplings. Six of seven plant species belonging to different families were found to be rich in isoprenoids, among which linalool was the most common compound. Linalool showed rhythmicity with a nocturnal increase inLonicera japonica. These findings suggest that linalool is a common attractant for nocturnal hawkmoths. However, the composition of other isoprenoids, benzenoids and fatty acid derivatives varied markedly among the plant species examined. There was a significant correlation between species composition of flower-visiting hawkmoths and specific floral scents, suggesting that attractiveness to each hawkmoth species is dependent upon floral scent.

Few observations have been made on temporal changes in the siring success of flowers in the male stage. In this study, we estimated both male and female contributions to fitness for 21 plants of protandrous andromonoeciosHeracleum lanatum with differing dates of first flowering. The results of multiple regression analysis showed that total male fitness significantly increases with the advance of the first-flowering date but does not depend upon plant size, whereas female fitness increases with plant size but does not depend upon the first-flowering date. We also showed that the earlier-flowering plants have more late-blooming male flowers in their secondary umbels. Based on these results, we suggest that polymorphism of the early- and late-bloomers may be maintained by frequency-dependent selection through temporally changing male reproductive success.

We investigated the extent of genetic differentiation among populations of fujihatazao,Arabis serrata along an altitudinal gradient at Mt. Fuji in Shizuoka Prefecture. This species is a perennial plant, widely distributed in Japan forming small isolated populations. However, at Mt. Fuji, this species constitutes a large population distributed from 1440 to 2400 m altitude. A total of 411 individuals were sampled from ten subpopulations. Eighteen loci were detected on eleven enzyme systems. Eleven loci were monomorphic and seven loci were polymorphic with a mean of 2.11 alleles per loci. Nei's genetic distance (mean 0.01) and genetic identity (mean 0.968) were very similar among populations indicating a low genetic differentiation. The total genetic diversity (H T ) estimated for the polymorphic loci was, in average, 0.396. The mean gene differentiation (GST=0.091) was very low. Gene frequency of seven polymorphic loci was analyzed by spatial autocorrelation methods based on Moran's indexes. Only Pgi-3 exhibited a significant negative autocorrelation (−0.160;P

To examine whether floral and post-pollination isolation develops independently or not, we conducted a crossing experiment between Hemerocallis fulva and Hemerocallis citrina that shows large floral divergence adapted for diurnal and nocturnal pollinators that have been believed to be fully cross-fertile. Flowers of the two species from sympatric populations were hand-pollinated with conspecific pollen from the same population (control), interspecific pollen from the same area (sympatric cross), and interspecific pollen from the different area (allopatric cross). After capsule dehiscence, the fruit set, seed set per fruit and seed set per flower were determined among three cross categories. The seed sets per flower were 32 and 77% lower in sympatric and allopatric crosses than in the control when H. fulva was the pollen recipient. There was no difference in three reproductive measures among the cross categories when H. citrina was the pollen recipient. This finding indicates that post-pollination isolation does exist between H. fulva and H. citrina, although it is partial, asymmetric, and weakened in sympatry. Our result suggests that floral and post-pollination isolation may develop independently, and reinforcement may not be a general phenomenon in plants.

Time of flower anthesis in a day is thought to evolve in response to the time of pollinator activities. We studied blooming and withering time in natural populations of daylily (Hemerocallis fulva), nightlily (Hemerocallis citrina) and their hybrids, and also in an artificially obtained array of the F1 hybrids. Blooming time of H. fulva varied from 4:30 to 7:30 and H. citrina varied from 16:30 to 20:30. In a natural hybrid population, blooming time and withering time showed discontinuous bimodal distribution in spite that morphological traits of flowers showed continuous unimodal variation. Most F1 hybrids showed diurnal flowering. These findings indicate that only a few genes have strong phenotypic effect on the determination of flowering time in Hemerocallis, and suggest that the evolution from a H. fulva-like ancestor to H. citrina was not a continuous process by accumulation of minute mutations.

To examine the evolution of gynodioecy under biparental inbreeding, we developed a new theoretical model and tested it by estimating paramaeters incorporated in the model in a gynodioecious population ofChionographis japonica var.kurohimensis. The model and the results showed biparental inbreeding affects the evolution of gynodioecy, but the effect is very small.
