Commercial cultivation of eukaryotic microalgae has so far employed a unicellular form of species only (e.g., Chlorella pyrenoidosa, Dunaliella salina, and Haematococcus pluvialis). In this study, we assessed the feasibility of using the filamentous eukaryotic microalga Klebsormidium sp. LGX80 as a new cultivar for biomass and lipid production. The effects of different forms and concentrations of nitrogen on growth and lipid production of Klebsormidium sp. LGX80 were studied by using a glass column (ø4.5 × 60 cm) photobioreactor under laboratory conditions. Growth and lipid production of the new strain were further evaluated in an outdoor pilot‐scale tubular photobioreactor. The results showed that when supplied with urea as a source of nitrogen Klebsormidium sp. LGX80 yielded a final biomass concentration of 8.49 ± 0.10 g · L−1 in which a cellular lipid content was 59.2 ± 0.4% DW. Under such conditions, the biomass and lipid productivities were 471.7 ± 5.9 and 248.1 ± 0.0 mg · L−1 · d−1, respectively. Fatty acid analysis revealed that the main fatty acids of Klebsormidium sp. LGX80 were palmitic acid (C16:0), linoleic acid (C18:2ω6), and linolenic acid (C18:3ω3), of which linoleic acid (C18:2ω6) accounted for up to 67.5 ± 0.1% of total fatty acids. When grown outdoors in a 13,000‐L tubular photobioreactor with an initial nitrogen concentration of 3 mM urea, Klebsormidium sp. LGX80 reached the highest biomass concentration of 2.63 ± 0.09 g · L−1 with the cells containing 38.0 ± 0.5% lipids (% DW), resulting in the volumetric biomass and lipid productivities of 147.2 ± 3.6 and 37.9 ± 0.9 mg · L−1 d−1, respectively. The results of light:dark cycle experiment showed that a durative and prolonged light irradiation hindered the biosynthesis of chlorophyll a and chlorophyll b in the cells, but promoted the carotenoid accumulation. These results suggested that Klebsormidium sp. LGX80 can be a potential oleaginous filamentous microalga for commercial production of microalgal oils. [ABSTRACT FROM AUTHOR]

Although desmids typically inhabit freshwater environments characterized by low amounts of nutrients and low salinity, several desmid species have been recorded in eutrophic waters, indicating their adaptation to elevated pollution and conductivity. This study aimed to determine whether desmids could be used for remediation of moderately saline aquaculture wastewater (AWW) from a fish farm situated in the southeast of Sweden. Fourteen desmid strains isolated from different climates (tropical to polar) and trophic conditions (oligotrophic to eutrophic) were cultivated in diluted AWW and we estimated their growth rates, biomass, nutrient removal efficiency, chlorophyll fluorescence parameters and cellular C, N and P quotas. Despite being grown at moderate salinity, unfavourable N:P ratio, and relatively low light/temperature regime the eutrophic strains, Cosmarium humile, Cosmarium laeve and a meso‐oligotrophic species Cosmarium impressulum, completely absorbed nitrate and phosphate from AWW media after 7 d, indicating their potential for remediation of fish effluents in colder climates. These species, along with the typical eutrophic species, Cosmarium meneghinii and Staurastrum chaetoceras, had biomass in the range 0.45–1.19 g · L−1 while maximum growth rates ranged from 0.36 to 0.51 · d−1, similar to published rates for several fast‐growing green microalgae cultivated in various AWW types. Tropical desmids had distinctly high values of saturating irradiance (Ik > 1,000 µmol photons · m−2 · s−1), and, along with eutrophic desmids, had high potential electron transport (rETRmax > 155 rel. units). Hence, the desmids studied demonstrated inherent photophysiological responses corresponding to their climate and trophic origin under the suboptimal growth conditions. [ABSTRACT FROM AUTHOR]

Our aim was to develop a novel medium for the large‐scale production of protein‐rich Arthrospira with potential applications as a biofertilizer. The novel culture medium, termed as FM‐II, was formulated using low‐cost commercial chemicals and specifically designed to improve protein production. Both Arthrospira platensis and Arthrospira maxima were produced using FM‐II and Arnon medium, which was used as a control. Photosynthetic status of the cells, which was checked by measuring chlorophyll fluorescence, biomass dry weight and protein content, was assessed daily. Arthrospira platensis had higher biomass and protein productivities than A. maxima when cultured in both control and FM‐II media. Incorporation of varied micronutrients into FM‐II formulation did not improve biomass productivity. Maximum biomass dry weight in FM‐II and control medium was 2.9 and 2.5 g · L−1, respectively. Total protein content of the biomass ranged between 55% and 65%, suggesting potential for being used in the development of high‐value agricultural products. As some nutrients were discarded unused, the initial content of phosphates and bicarbonates was reduced by 75% and 50%, respectively, without affecting the process productivity. Results reported herein could promote the production and utilization of Arthrospira platensis by significantly reducing productions costs and therefore increasing the feasibility of the process. Harmful cyanobacterial blooms (CyanoHABs) are a rapidly proliferating global problem, threatening the use and sustainability of our freshwater resources. In recent decades, the United States, China, and other developed and developing countries threatened by CyanoHAB expansion have established collaborative efforts aimed at mitigating and managing this environmental and human health problem. However, an escalating negative political climate and restrictive policies on scientific exchange threaten these efforts. In this Perspective, I point to progress that has been made to counter the CyanoHAB problem on U.S.–Chinese fronts through our collaborations, which have been mutually beneficial from research and academic perspectives. Much like global efforts now needed to control pandemics, we are all "in the same boat" when to comes to countering the threat CyanoHABs pose for drinkable, swimmable, and fishable freshwater supplies and human health. To test the hypothesis that rotifers release one or more chemical microalgal growth inhibitors in addition to devouring the microalgal cells, the effects of different concentrations of filtered, bacteria‐free, rotifer culture filtrate (RCF) on the growth and physiological parameters of Chlorella sp., and the response of Chlorella sp. at different starting cell densities to 10% RCF, were studied. The results show that RCF significantly decreased Chlorella cell densities during the incubation, suggesting that rotifers release some chemical(s) that inhibit microalgal cell growth. Chlorella cell densities decreased with increasing RCF concentration. Increasing the initial cell density of Chlorella dispersed the inhibitory chemical(s) present in 10% RCF over more cells, reducing their effect. The results confirm that the action of the chemical(s) released by rotifers on microalgal cell growth was dependent on both the RCF concentration and the exposure time. They also demonstrate that ≥10% RCF significantly inhibited photosynthesis and respiration, which would account for some of the decreased Chlorella cell growth in the presence of RCF. Calculations based on the data indicate that the rotifer‐derived chemical(s) released hourly from each rotifer inhibits growth by 45.5 microalgal cells in addition to the rotifer predation, with a 48 h LC50 value of 18.8% RCF. The domesticated marine microalga Diacronema lutheri is of great interest for producing various highly valuable molecules like lipids, particularly long‐chain polyunsaturated fatty acids (LC‐PUFA). In this study, we investigated the impact of phosphorus (P) and nitrogen (N) starvation on growth, carbon fixation (photosynthetic activity) and partitioning, and membrane lipid remodeling in this alga during batch culture. Our results show that the photosynthetic machinery was similarly affected by P and N stress. Under N starvation, we observed a much lower photosynthetic rate and biomass productivity. The degradation and re‐use of cellular N‐containing compounds contributed to triacylglycerol (TAG) accumulation. On the other hand, P‐starved cells maintained pigment content and a carbon partitioning pattern more similar to the control, ensuring a high biomass. Betaine lipids constitute the major compounds of non‐plastidial membranes, which are rich in eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids. Dunaliella salina is well known for its ability to accumulate large amounts of β‐carotene. Myo‐inositol (MI) enhances the biomass production of D. salina, but the underlying mechanisms were unclear. The present study showed that the concentration of exogenous MI decreased gradually and reached a constant level at the 4th day of cultivation. MI enhanced the contents of total colored carotenoids and the activity of photosystem II. Metabolic profiles were significantly changed after the addition of exogenous MI, as revealed by multivariate statistical analysis. The metabolites could be categorized into four groups based on the relative levels in different samples. Exogenous MI increased the levels of most detected sugars, amino acids, and total saturated and unsaturated fatty acids. Based on the physiological and metabolic analyses, a hypothetical growth‐promoting model that MI promotes the growth of D. salina TG by increasing the levels of key metabolites and possibly enhancing photosynthesis, was proposed. The red macroalgae Hydropuntia cornea, Gracilariopsis longissima and Halopithys incurva were cultured for 14 d under laboratory conditions, in enriched seawater with a high nutrient content (N‐NH4+ and P‐PO43−) and two radiation regimes: PAR (400–700 nm) and PAB (280–700 nm). The UV radiation effects under high availability of nutrients on growth, photosynthetic pigments (chlorophyll a, carotenoids and phycobiliproteins), photosynthetic activity and biochemical composition were studied. Maximum quantum yield (Fv/Fm) was not significantly different among the PAR and PAB treatments during the experiment. However, the maximum electronic transport rate (ETRmax) increased over time, showing the highest values in PAR for H. incurva and H. cornea, whereas for G. longissima it was found in PAB. Photosynthetic efficiency (αETR) decreased over time in the first two species, but increased in G. longissima. Saturation irradiance (EkETR) and maximum nonphotochemical quenching (NPQmax) increased in PAB with time up to 80% and 30%, respectively, indicating a photosynthetic acclimatization like that of sun‐type algae. Five MAAs were identified in all species using high performance liquid chromatography (HPLC). The total content of MAAs increased over time, being 30% higher in H. incurva, 40% in G. longissima and 50% in H. cornea in PAB than in the PAR treatment. Finally, the antioxidant activity was also higher in the PAB treatment. All of the species presented an effective mechanism of photoprotection based on the accumulation of photoprotective compounds with antioxidant activity, as well as a high dissipation of excitation energy (high NPQmax). A novel freshwater strain of Coccomyxa elongata (MZ–Ch64) was isolated from the Zaporizhia region, Ukraine. The identification was based on the phylogenetic analysis of SSU rDNA gene and ITS1–5.8S rDNA–ITS2 region and predicted secondary structure of the ITS2. Phylogenetic analysis placed this strain in the Coccomyxa group, within the class Trebouxiophyceae. The novel strain MZ–Ch64 formed a strongly supported lineage closest with C. elongata. The MZ–Ch64 strain differed from the morphological description of the species by the size of vegetative cells and absence of small mucilaginous caps at one end of the cell. A number of experiments with different concentrations of phosphate and nitrate were conducted to evaluate changes in the resulting fatty acid profiles and biomass productivity. The fatty acid profile and total fatty acids varied significantly under different nutrient deficiencies. The dominant fatty acid during cultivation on standard BBM medium, as well as in phosphorus‐depleted conditions, was oleic acid (to 48.0%–54.6% of total fatty acids). Absence of nitrogen alone, and absence of both nitrogen and phosphorus, led to an increase of palmitic acid (to 24.7%–25.6%), cis‐7‐hexadecenoic acid (to 14.8%) and α‐linolenic acid (to 9.1%–10.1%) in comparison with the control sample. The greatest variation was found for oleic acid (31.9%–54.6%). Thus, this strain can be considered as a potential producer of oleic acid or cis‐7‐hexadecenoic and α‐linolenic acids for biotechnological applications. [ABSTRACT FROM AUTHOR]

The present study investigated the effect of different culture conditions on the vegetative growth of a new species, Haematococcus alpinus (strain LCR‐CC‐261f) using airlift photobioreactors. The influence of culture medium, aeration rates, CO2 concentration in air‐gas mixture, temperature, light intensities, and wavelengths were investigated to achieve sustainable high cell density cultures. Growth parameters were determined by fitting the data to a form of the logistic equation that included a lag phase. The shear‐sensitive vegetative cells favored lower aeration rates in the photobioreactors. MLA medium increased to 40 mM nitrate produced high density cultures. Temperatures between 12°C and 18°C, 3% (v/v) CO2 concentration and a narrow photon flux density ranging between 37 and 48 μmol photons · m−2 · s−1 were best suited for growth. The wavelength of the light source also impacted growth and a high cell density of 9.6 × 105 cells · mL−1 was achieved using a mixture of red and blue compared to warm white, red, or blue LEDs. Microbial volatiles have a significant impact on the physiological functions of prokaryotic and eukaryotic organisms. Various ketones are present in volatile mixtures produced by plants, bacteria, and fungi. Our earlier results demonstrated the inhibitory effects of soil bacteria volatiles, including ketones, on cyanobacteria. In this work, we thoroughly examined the natural ketones, 2‐nonanone and 2‐undecanone to determine their influence on the photosynthetic activity in Synechococcus sp. PCC 7942. We observed for the first time that the ketones strongly inhibit electron transport through PSII in cyanobacteria cells in vivo. The addition of ketones decreases the quantum yield of primary PSII photoreactions and changes the PSII chlorophyll fluorescence induction curves. There are clear indications that the ketones inhibit electron transfer from QA to QB, electron transport at the donor side of PSII. The ketones can also modify the process of energy transfer from the antenna complex to the PSII reaction center and, by this means, increase both chlorophyll fluorescence quantum yield and the chlorophyll excited state lifetime. At the highest tested concentration (5 mM) 2‐nonanone also induced chlorophyll release from Synechococcus cells that strongly indicates the possible role of the ketones as detergents. Nanoparticles have unique properties that make them attractive for use in industrial and medical technology industries but can also be harmful to living organisms, making an understanding of their molecular mechanisms of action essential. We examined the effect of three different sized poly(isobutyl‐cyanoacrylate) nanoparticles (iBCA‐NPs) on the unicellular green alga Chlamydomonas reinhardtii. We found that exposure to iBCA‐NPs immediately caused C. reinhardtii to display abnormal swimming behaviors. Furthermore, after one hour, most of the cells had stopped swimming and 10%–30% of cells were stained with trypan blue, suggesting that these cells had severely impaired plasma membranes. Observation of the cyto‐ultrastructure showed that the cell walls had been severely damaged and that many iBCA‐NPs were located in the space between the cell wall and plasma membrane, as well as inside the cytosol in some cases. A comparison of three strains of C. reinhardtii with different cell wall conditions further showed that the cell mortality ratio increased more rapidly in the absence of a cell wall. Interestingly, cell mortality over time was essentially identical regardless of iBCA‐NP size if the total surface area was the same. Furthermore, direct observation of the trails of iBCA‐NPs indicated that the first trigger was their contact with the cell wall, which is most likely accompanied by the inactivation or removal of adsorbed proteins from the cell wall surface. Cell mortality was accompanied by the overproduction of reactive oxygen species, which was detected more readily in cells grown under constant light rather than in the dark. [ABSTRACT FROM AUTHOR]

Ulva spp. dominates green tides around the world, which are occurring at an accelerated rate. The competitive nitrogen assimilation efficiency in Ulva is suggested to result in ecological success against other seaweeds. However, molecular characterization of genes involved in nitrogen assimilation has not been conducted. Here, we describe the identification of the nitrate reductase ( NR) gene from a green seaweed Ulva prolifera, an alga which is responsible for the world's largest green tide in the Yellow Sea. Using rapid amplification of cDNA ends and genome walking, the NR gene from U. prolifera ( Up NR) was cloned, which consisted of six introns and seven exons encoding 863 amino acids. According to sequence alignment, the NR in U. prolifera was shown to possess all five essential domains and 21 key invariant residues in plant NRs. The GC content of third codon position of Up NR (82.75%) was as high as those of green microalgae, and the intron number supported a potential loss issue from green microalga to land plant. Real-time quantitative PCR results showed that Up NR transcript level was induced by nitrate and repressed by ammonium, which could not be removed by addition of extra nitrate, indicating that U. prolifera preferred ammonium to nitrate. Urea would not repress NR transcription by itself, while it weakened the induction effect of nitrate, implying it possibly inhibited nitrate uptake rather than nitrate reduction.
