Ethnopharmacological Relevance: Phytolacca acinosa Roxb. (PAR) is a Traditional Chinese Medicinal (TCM) plant with a broad global distribution encompassing 35 species, four of which are found in the People's Republic of China. It occupies a significant role in both Oriental and American traditional medicine, employed in treating a range of conditions such as edema, inflammation, dermatitis, and rheumatism. PAR is also used as a molluscicide and for addressing tumors and bronchitis. The plant is documented in the Chinese Pharmacopoeia and has a longstanding history in TCM, particularly for its diuretic properties and in treating ailments such as edema, swelling, and ulcers. Notably, PAR has demonstrated potent inhibitory effects against the A549 human lung cancer cell line, underscoring its potential in contributing to the development of novel cancer therapeutics., Aim of the Study: The research aims to elucidate the active components of PAR and their mechanisms in treating hepatocellular carcinoma (HCC)., Materials and Methods: Employing network pharmacology, this study predicted the principal active compounds and key targets of PAR. A holistic methodology incorporating biological network analysis, transcriptomics sequencing, molecular docking, and molecular dynamics (MD) simulations was utilized to forecast the effects of PAR on HCC, with empirical evidence supporting these findings., Results: Network pharmacology identified xanthomicrol as the foremost active compound in PAR. The tumor-suppressive functions of PAR, as indicated by KEGG pathway analysis and transcriptomics sequencing, predominantly occur via the PI3K/AKT pathway. Molecular docking and dynamics simulations demonstrated the high affinity of xanthomicrol towards TNF, MMP9, PPARG, KDR, and MMP2. In vivo experiments verified the efficacy of xanthomicrol in curtailing HCC tumor growth, while in vitro assessments revealed its substantial impact on the proliferation and apoptosis of HepG2 and HCCLM3 cells. Moreover, the study indicates that xanthomicrol may modulate the expression of TNF, MMP9, PPARG, KDR, and MMP2 in HCC cells and inhibit the activation of the PI3K/AKT pathway., Conclusions: Xanthomicrol, a principal active component of PAR, has been identified to impede the growth of HCC by targeting the PI3K/Akt/MMP9 pathway. Berries from Phytolacca dodecandra L'Herit. (endod in Amharic) offer a readily available molluscicide to control schistosomiasis. Parts of the endod plant have been used as a detergent and as traditional medicine for centuries in Ethiopia. An interview survey was performed in the highlands of Ethiopia to provide information on the distribution of the plant, people's traditional use of it, their perception of the plant, and the potential for increased production and use of endod as a soap for indirect control of schistosomiasis. People of all ages report that they are familiar with the plant and its detergent and medicinal uses. The plant is largely disappearing from unprotected areas due to land clearing. Younger people appear to use endod as a soap whenever it is available. Older women prefer commercial soap and consider endod to be associated with poor people. Common medicinal uses include treatment of skin itching (ringworm), abortion, gonorrhea, leeches, intestinal worms, anthrax and rabies. Two thirds of the people express interest in cultivating endod for personal use if supplied with rooted cuttings. Increased cultivation of endod and use of berries for washing might be possible if information about schistosomiasis and its control is disseminated among people. Preference for commercial soap and lack of land for cultivation are major obstacles for increasing the availability and use of endod.

The methanolic extract of the berries of Phytolacca tetramera, an Argentinean species submitted to a great anthropic impact, and therefore in critic risk of extinction, not previously studied, showed antifungal activity against opportunistic pathogenic fungi. Through fractionation of the extract followed by agar dilution assays, three monodesmosidic triterpenoid saponins have been isolated from the butanolic extract of P. tetramera. The structures were established as phytolaccosides: B [3-O-beta-D-xylopiranosyl-phytolaccagenin], E [3-O-beta-D-glucopyranosyl-(1-->4)-beta-D-xylopiranosyl-phytolaccagenin]. and F [3-O-alpha-L-rhamnopyranosyl-(1-->2)-beta-D-glucopyranosyl-(1-->2)-beta-D-xylopyranosyl-phytolaccagenic acid]. The three saponins belong to the olean-type triterpenoid saponins, with 28,30 dicarboxylic groups and an olefinic double bond on C-12. Phytolaccosides B and E but not phytolaccoside F, showed antifungal activities against a panel of human pathogenic opportunistic fungi. Phytolaccoside B was the most active compound and showed the broadest spectrum of action. The most sensitive fungus was Trichophyton mentagrophytes., (Copyright 2002 Elsevier Science Ireland Ltd.)
