The interrelationships between silage preference of ruminants and compounds that may affect forage choice are not yet fully enlightened. Analysis of the forage metabolome in addition to conventional chemical analysis and preference trials can provide new insights. Six silage treatments each of alfalfa (AL) and red clover (RC), with different dry matter concentrations (222–391 g/kg), silage additives, and intended addition of soil, were produced in quadruplicate to obtain a range of qualities. After 120 d of ensiling, silages were sampled for chemical analysis, vacuum-packed, and refrigerated for subsequent preference trials with goats. Within 21 d, each possible combination of 2 silages and an AL hay that served as control (n = 21) was presented to goats (Saanen-type wethers, n = 8, body weight 105 ± 2.7 kg) for 3 h for ad libitum intake. Comparisons among means for 3-h dry matter intake (DMI) for forages offered in choice situations were made using variance analysis, including terms for treatment and animal and the Waller-Duncan k-ratio t-test to separate means. The most preferred and avoided treatments of AL and RC silage amounted to 863, 858, 226, and 282 g DMI/3 h, respectively. To further explore relations between silage composition and preference, a metabolome analysis of the most preferred and most avoided AL and RC treatments were conducted. Metabolites (all low molecular weight molecules) were analyzed by a nontargeted metabolite profiling in the range of 50–1,700 Da. Metabolites showing the most distinct difference between preferred and avoided silages were identified by partial least squares discriminant analysis. In the 2 selected treatments of each plant species (those that were most different in forage preference), more than 6,400 compounds were detected and 2,010 were identified. Between preferred and avoided treatments, 934 of the detected compounds differed in RC and 1,860 in AL, of which 475 were altered in both plant species (251 were reduced and 186 were increased; only 38 behaved contrarily, meaning that they were increased in one substrate and decreased in the other). The database provides a useful foundation for the approach of explaining silage preference by ruminants.

We evaluated the effects of altering the dietary ratio of palmitic (C16:0; PA) and oleic (cis-9 C18:1; OA) acids on production responses of cows with a wide range of milk production (32 to 65 kg/d) in a crossover design experiment with a preliminary period. Thirty-two multiparous Holstein cows (144 ± 54 d in milk) were assigned randomly to a treatment sequence. Treatments were diets supplemented with fatty acid (FA) blends (1.5% of diet dry matter) that provided 80% C16:0 + 10% cis-9 C18:1 (PA) and 60% C16:0 + 30% cis-9 C18:1 (PA+OA). The corn silage and alfalfa-based diets contained 20.0% forage neutral detergent fiber (NDF), 28.5% starch, and 17.1% crude protein. Treatment periods were 21 d with the final 5 d used for data and sample collection. Treatment did not affect dry matter intake (DMI), milk yield, energy-corrected milk (ECM), body weight, or body weight change. The PA+OA diet increased total, 16-carbon, and 18-carbon FA digestibility compared with the PA diet. Compared with PA+OA, PA increased fat yield (1.97 vs. 1.91 kg/d) and protein yield (1.61 vs. 1.55 kg/d). The PA diet also increased the yield of de novo (448 vs. 428 g/d) and mixed (749 vs. 669 g/d) milk FA and decreased the yield of preformed FA (605 vs. 627 g/d) compared with PA+OA. Interactions were detected between treatment and preliminary milk yield for DMI, total FA intake, 16-carbon FA intake, ECM, 3.5% fat-corrected milk (linear interaction), and a tendency for milk yield (linear interaction); lower-producing cows (45 kg/d) had increased DMI and ECM on the PA diet, whereas higher-producing cows (55 kg/d) had increased DMI and ECM on the PA+OA diet. A linear interaction was detected between treatment and preliminary milk yield for mixed milk FA yield (linear interaction) and a tendency for de novo milk FA yield (linear interaction). Our results demonstrate that feeding a fat supplement containing more cis-9 C18:1 replacing C16:0 increased production responses (DMI, milk yield, and ECM) in higher-producing cows, but decreased production responses in lower-producing cows.

The present study was intended to evaluate the effect of forage source (alfalfa hay; ALF vs. corn silage; CS) along with a supplemental fat source (soybean oil; SO vs. rumen-inert palm fatty acids; PF) on growth performance, nutrient digestibility, and ruminal fermentation in dairy calves. Forty-eight new-born Holstein female calves (3 d old) were assigned to one of 4 treatments: (1) alfalfa hay with soybean oil (ALF-SO); (2) alfalfa hay with palm fatty acids (ALF-PF); (3) corn silage with soybean oil (CS-SO); (4) corn silage with palm fatty acids (CS-PF). Starter diets had equal amounts of forage (100 g/kg dry matter; DM) and fat source (30 g/kg DM). Calves were fed a constant amount of milk (d 1 to 63) and had ad libitum access to water and starters (d 1 to 83). The lowest and greatest starter intakes during the preweaning period occurred in ALF-SO and CS-PF, respectively. This coincided with forage × fat source interaction for average daily gain (ADG) during preweaning. The forage source affected total DM intake and ADG over the entire period, body weight (BW) at weaning, and final BW with greater values in calves that received CS compared with ALF. The concentrations of total short-chain fatty acids and butyrate were increased, whereas concentration of acetate and acetate:propionate ratio were decreased in the rumen of calves fed CS compared with ALF. Feeding CS increased urinary excretion of allantoin and, as a trend, total purine derivatives (PD) and estimated microbial protein synthesis in comparison with ALF. The fat source affected starter intake, ADG, and BW postweaning with the highest values in PF. The digestibility of neutral detergent fiber, crude protein and, as a trend, organic matter were higher in calves fed PF compared with SO. Calves fed PF had lower ruminal ammonia-N concentration and urinary N excretion and greater urinary excretion of allantoin and total PD. Calves receiving SO had a lower ruminal protozoa population. In conclusion, supplementing starter diets with CS and PF is superior to ALF and SO. Interaction of the positive effects of CS and PF on performance underlines that concurrent supplementation of CS with PF is especially recommendable in young calves before weaning.

Eight lactating Italian Friesian cows were housed in individual respiration chambers in a repeated Latin square design to determine their dry matter intake (DMI) and their milk and methane production, as well as to collect the total feces and urine to determine the N and energy balances. Four diets, based on the following forages (% of dry matter, DM), were tested: corn silage (CS, 49.3), alfalfa silage (AS, 26.8), wheat silage (WS, 20.0), and a typical hay-based Parmigiano Reggiano cheese production diet (PR, 25.3 of both alfalfa and Italian ryegrass hay). The greatest DMI was observed for cows fed PR (23.4 vs. 20.7 kg/d, the average of the other 3 diets). The DM digestibility was lower for PR (64.5 vs. 71.7%, the average of the other diets). The highest ash-free neutral detergent fiber digestibility values were obtained for CS (50.7%) and AS (47.4%). In the present study, no differences in milk production were observed between diets, although PR showed a higher milk yield trend. The highest milk urea N concentration (mg/dL) was found for the cows fed the WS diet (13.8), and the lowest was observed for the cows fed AS (9.24). The highest milk urea N concentration for the cows fed WS was also correlated with the highest urinary N excretion (g/d), which was found for the cows fed that same diet (189 vs. 147 on average for the other diets). The protein digestibility was higher for the cows fed the CS and WS diets (on average 68.5%) than for the cows fed AS and PR (on average 57.0%); dietary soybean inclusion was higher for CS and WS than for AS and PR. The rumen fermentation pattern was affected by the diet; the cows fed the PR diet showed a higher rumen pH and decreased propionate production than those fed CS, due to the lower nonfiber carbohydrate content and higher ash-free neutral detergent fiber content of the PR diet than the CS diet. Feeding cows with PR diet increased the acetate:propionate ratio in comparison with the CS diet (3.30 vs. 2.44 for PR and CS, respectively). Cows fed the PR diet produced a greater daily amount of methane and had a greater methane energy loss (% of digestible energy intake) than those fed the CS diet (413 vs. 378 g/d and 8.67 vs. 7.70%), but no differences were observed when methane was expressed as grams per kilogram of DMI or grams per kilogram of milk. The PR diet resulted in a smaller net energy for lactation content than the CS diet (1.36 vs. 1.70 Mcal/kg of DM for the PR and CS diets, respectively). Overall, our research suggests that a satisfactory milk production can be attained by including different high-quality forages in balanced diets without any negative effect on milk production or on the methane emissions per kilogram of milk.

Tall fescue might be an alternative to timothy in northeastern North America because of its tolerance of recurring drought periods and its good summer regrowth, but is not always considered as an option in dairy rations because of its possible lack of palatability. The objective of this study was to evaluate the effects on the performance of lactating dairy cows of (1) replacing timothy silage by tall fescue silage, offered as sole forage in the diet or in combination with alfalfa silage, and (2) feeding tall fescue as silage (35% dry matter, DM) or haylage (55% DM). Experimental diets with a forage-to-concentrate ratio of 70:30 were (1) 100% timothy silage (TS); (2) 100% tall fescue silage (TFS); (3) 55:45 timothy:alfalfa silages (TS + AS); (4) 55:45 tall fescue:alfalfa silages (TFS + AS); and (5) 100% tall fescue haylage (TFH). Fifteen Holstein cows in mid-lactation (5 fitted with a rumen fistula) were randomly assigned to treatments in a triple 5 × 5 Latin square design with treatment periods of 21 d. Preplanned contrasts were timothy versus tall fescue silages, sole grass species versus grass-alfalfa, interaction between sole grass species and grass-alfalfa, and TFS versus TFH. Grass species did not affect dry matter intake (DMI) or milk yield and fat concentration. Milk protein concentration was not affected by grass species when offered in combination with alfalfa, but it was higher with the TS diet than the TFS diet when offered as sole forages. Adding alfalfa to either tall fescue or timothy silage resulted in greater DMI and milk yield, but lower milk fat concentration, than when the grass silages were the sole forage in the diet. The molar proportion of propionate in the rumen was greater when cows were fed diets with tall fescue silage compared with timothy silage, which resulted in a lower acetate-to-propionate ratio. Milk fat concentrations of fatty acids from microbial origin, namely branched-chain fatty acids, were greater when grass silage, and especially timothy silage, were fed as sole forages rather than with alfalfa silage. Feeding TFH rather than TFS caused a decrease in DMI and tended to lower milk protein concentration, but did not affect milk yield. A more fibrolytic fermentation profile was observed in rumen of cows fed TFH compared with TFS, as indicated by the increase in the molar proportion of acetate and the higher acetate-to-propionate ratio in rumen fluid, and a concomitant increase in branched-chain fatty acid concentration in milk fat. Tall fescue as silage or haylage is a valuable alternative to timothy silage for lactating dairy cows.

Previous research has demonstrated that feeding canola meal (CM) improves milk production and N utilization by lactating dairy cows when replacing solvent-extracted soybean meal (SBM). The objective of the present study was to evaluate whether CM would improve milk yield and components and N utilization, compared with SBM, at different ratios of alfalfa silage (AS) to corn silage (CS) fed to lactating dairy cows. Twenty-four multiparous Holstein cows averaging, at the beginning of the study (mean ± SD), 2.8 ± 0.9 parity, 684 ± 56 kg of BW, 102 ± 41 DIM, and 49 ± 4 kg milk/d, and 24 primiparous cows averaging (mean ± SD) 565 ± 46 kg of BW, 123 ± 30 DIM, and 40 ± 4 kg milk/d were blocked by parity and DIM. A cyclic changeover design with 4 replications of 2 blocks of treatments of 6 cows was used in an arrangement with 4 28-d periods. Dietary treatments were arranged in a 3 × 2 factorial design of 3 proportions of AS to CS as forage source (HAS = high AS, 50% AS to 10% CS; MAS = medium AS, 30% AS to 30% CS; LAS = low AS, 10% AS to 50% CS) and 2 protein supplements (CM vs. SBM). Diets were formulated to contain [dry matter (DM) basis]: 60% forage, 8 to 15% high-moisture corn, 2 to 5% soy hulls, 1.3% mineral-vitamin premix, 16% crude protein, and 31 to 33% NDF. Data from the last 2 weeks of each period were used to compute mean milk yield and composition, and efficiencies of feed conversion, for each cow in each period. Data for the other variables were collected during the last week of each period. All data were analyzed using the MIXED procedure of SAS (SAS Institute Inc., Cary, NC). Regardless of the forage source, replacing SBM with CM improved yields of milk, milk protein, and solids-not-fat. Moreover, milk urea nitrogen concentration and urinary excretion of total N (g/d) and urea N (% of total urinary N) decreased when CM replaced SBM. An interaction effect occurred between forage source and protein supplements for apparent total-tract digestibility, and, overall, this effect was due to small differences in ingredient and chemical compositions of the diets. In addition, these differences had a minor effect on cow performance. Yields of milk and milk components were greatest for cows fed 50% CS, intermediate for 30% CS, and lowest for 10% CS, indicating that, under the conditions of the present study, cows fed 50% CS in the diet (DM basis) had greater production compared with those fed 50% AS.

Because of their antimicrobial properties, essential oils and their components have been suggested as alternatives to other antimicrobials (e.g., monensin) that are commonly fed to ruminants to improve nutrient utilization and enhance feed efficiency and milk performance. In this study, we evaluated the potential of oregano oil and its main component (carvacrol) as rumen modifiers. For this purpose, 8 ruminally cannulated lactating dairy cows (92 ± 11 d in milk, 36.5 ± 7.6 kg of milk yield, and 703 ± 74 kg of body weight) were used in a double 4 × 4 Latin square (28-d periods). Cows were fed 1 of the 4 following treatments: (1) control (CTL, no additive); (2) monensin [MON, 24 mg/kg of dry matter (DM)]; (3) oregano oil (ORE, 50 mg/kg of DM); and (4) carvacrol (CAR, 50 mg/kg of DM). Cows were fed (ad libitum intake) a total mixed ration consisting of 60% forages (corn silage and alfalfa silage) and 40% concentrates, on a DM basis. Feeding ORE and CAR had no effect on nutrient total-tract apparent digestibility, N utilization, rumen fermentation (i.e., pH, ammonia, volatile fatty acids), protozoa counts, or milk performance. Feeding MON increased the molar proportion of propionate and tended to increase total-tract apparent digestibility of crude protein. None of the feed additives evaluated affected enteric methane production (491 g/d, 21.1 g/kg of DM intake, 6.14% of gross energy intake on average). Milk fatty acid composition was not changed by ORE or CAR, but MON increased the proportion of trans-10 18:1, an intermediate of ruminal biohydrogenation. Thus, when included at 50 mg/kg of dietary dry matter, neither oregano oil nor carvacrol favorably altered rumen fermentation, improved nutrient utilization or milk performance, or mitigated enteric methane emissions in dairy cows.

This study evaluated the effects of feeding diets that were formulated to contain similar proportions of undigested neutral detergent fiber (uNDF) from forage, with wheat straw (WS) substituted for corn silage (CS), alfalfa hay (AH), or both. The diets were fed to lactating dairy cows and intake, digestibility, blood metabolites, and milk production were examined. Thirty-two multiparous Holstein cows (body weight = 642 ± 50 kg; days in milk = 78 ± 11 d; milk production = 56 ± 6 kg/d; mean ± standard deviation) were used in a randomized block design with 6-wk periods after a 10-d covariate period. Each period consisted of 14 d of adaptation followed by 28 d of data collection. The control diet contained CS and AH as forage sources (CSAH) with 17% of dietary dry matter as uNDF after 30 h of incubation (uNDF30). Wheat straw was substituted for AH (WSCS), CS (WSAH), or both (WSCSAH) on an uNDF30 basis, and beet pulp was used to obtain similar concentrations of NDF digestibility after 30 h of incubation (NDFD30 = 44.5% of NDF) across all diets. The 4 diets also contained similar concentrations of net energy for lactation and metabolizable protein. Dry matter intake was greatest for WSCS (27.8 kg/d), followed by CSAH (25.7 kg/d), WSCSAH (25.2 kg/d), and WSAH (24.2 kg/d). However, yields of milk, 3.5% fat-corrected milk (FCM), and energy-corrected milk did not differ, resulting in higher FCM efficiency (kg of FCM yield/kg of dry matter intake) for WSAH (1.83) and WSCSAH (1.79), followed by CSAH (1.69) and WSCS (1.64). Milk protein percentage was greater for CSAH (2.84%) and WSCS (2.83%) than for WSAH (2.78%), and WSCSAH (2.81%) was intermediate. The opposite trend was observed for milk urea nitrogen, which was lower for CSAH (15.8 mg/dL), WSCS (15.8 mg/dL), and WSCSAH (17.0 mg/dL) than for WSAH (20 mg/dL). Total-tract NDF digestibility and ruminal pH were greater for diets containing WS than the diet without WS (CSAH), but digestibility of other nutrients was not affected by dietary treatments. Cows fed WSAH had less body reserves (body weight change = -13.5 kg/period) than the cows fed the other diets, whereas energy balance was greatest for those fed WSCS. The results showed that feeding high-producing dairy cows diets containing different forage sources but formulated to supply similar concentrations of uNDF30 while maintaining NDFD30, net energy for lactation, and metabolizable protein constant did not influence milk production. However, a combination of WS and CS (WSCS diet) compared with a diet with CS and AH improved feed intake, ruminal pH, total-tract NDF digestibility, and energy balance of dairy cows.

Alfalfa has a lower fiber digestibility and a greater concentration of degradable protein than grasses. Dairy cows could benefit from an increased digestibility of alfalfa fibers, or from a better match between nitrogen and energy supplies in the rumen. Alfalfa cultivars with improved fiber digestibility represent an opportunity to increase milk production, but no independent studies have tested these cultivars under the agroclimatic conditions of Canada. Moreover, decreasing metabolizable protein (MP) supply could increase N use efficiency while decreasing environmental impact, but it is often associated with a decrease in milk protein yield, possibly caused by a reduced supply of essential AA. This study evaluated the performance of dairy cows fed diets based on a regular or a reduced-lignin alfalfa cultivar and measured the effect of energy levels at low MP supply when digestible His (dHis), Lys (dLys), and Met (dMet) requirements were met. Eight Holstein cows were used in a double 4 × 4 Latin square design, each square representing an alfalfa cultivar. Within each square, 4 diets were tested: the control diet was formulated for an adequate supply of MP and energy (AMP_AE), whereas the 3 other diets were formulated to be deficient in MP (DMP; formulated to meet 90% of the MP requirement) with deficient (94% of requirement: DMP_DE), adequate (99% of requirement: DMP_AE), or excess energy supply (104% of requirement; DMP_EE). Alfalfa cultivars had no significant effect on all measured parameters. As compared with cows receiving AMP_AE, the dry matter intake of cows fed DMP_AE and DMP_EE was not significantly different but decreased for cows fed DMP_DE. The AMP_AE diet provided 103% of MP and 108% of NEL requirements whereas DMP_DE, DMP_AE, and DMP_EE diets provided 84, 87, and 87% of MP and 94, 101, and 107% of NEL requirements, respectively. In contrast to design, feeding DMP_EE resulted in a similar energy supply compared with AMP_AE, although MP supply has been effectively reduced. This resulted in a maintained milk and milk component yields and improved the efficiency of utilization of N, MP, and essential AA. The DMP diets decreased total N excretion, whereas DMP_AE and DMP_EE diets also decreased milk urea-N concentration. Reducing MP supply without negative effects on dairy cow performance is possible when energy, dHis, dLys, and dMet requirements are met. This could reduce N excretion and decrease the environmental impact of milk production.

In the semi-arid highlands of central Mexico, triticale (× Triticosecale L.) is emerging as an alternative, less water-demanding forage crop than alfalfa for dairy cattle. Studies reported here were aimed at evaluating triticale hay (TH) relative to alfalfa hay (AH) for lactating cow performance, apparent digestibility, N partition, and ruminal degradation kinetics of dry matter (DM), crude protein (CP), and neutral detergent fiber (NDF). Study 1 was conducted on a privately owned farm. Four barns were used to conduct 4 replicated 3 × 3 Latin squares (1 barn = 1 square), where each barn included 3 pens (experimental units) receiving 1 of 3 dietary treatments. Each pen had 62 Holstein dairy cows. All diets included a forage-to-concentrate ratio of 42:58 (DM basis), which is typical for intensive dairy farms of the region. Dietary treatments were formulated to replace AH with TH on a CP basis, and included (DM basis) 15.1% AH and 0% TH, 9.0% AH and 7.4% TH, and 0% AH and 16.4% TH. Diets were iso-energetic (1.64 Mcal of net energy for lactation/kg of DM) and iso-nitrogenous (17.9% CP). Pen-level DM intake and milk production were from all cows in the pen, but pen-level milk composition, apparent digestibility, and N partitioning were from 8 cows (observational units) randomly selected in each pen. Orthogonal contrasts were used to determine linear and quadratic effects of increasing TH from 0 to 7.4, and 16.4% of dietary DM. Although DM intake was not affected, there was a tendency for CP intake to decline linearly and for NDF intake to increased linearly as TH replaced AH in the diet. Milk production declined linearly by 0.077 kg/d for each additional percentage unit of TH in the diet, which amounted to a 3.5% decline when TH replaced AH entirely. However, no effect was observed on energy-corrected milk production because of a compensatory linear effect of increasing milk fat concentration with the incorporation of TH in the diet. Total-tract NDF digestibility tended to increase linearly by 18.5%, but no differences were detected for urinary urea-N excretion and for N utilization estimated as milk N/(fecal N + urinary N + milk N). Study 2 was an in situ trial conducted to determine the degradation kinetics of AH and TH used in study 1. In spite of differences in degradation kinetics parameters for DM, CP, and NDF, only NDF effective ruminal degradation tended to be greater for TH than AH. Replacing AH with TH at the level typically found in intensive dairy farms of the semi-arid regions of Mexico had minimal effects on milk production and N utilization.

The purpose of this study was to optimize the conditions of a previously proposed enzymatic method used to estimate in situ undigested neutral detergent fiber (uNDF). We used a multi-step enzymatic approach, in which samples were first solubilized in NaOH solutions as a preincubation (PreInc) phase. After rinsing, samples were incubated (24 h at 39°C) in a buffered solution (pH 6) containing hemicellulase, cellulase, and Viscozyme L enzymes (Sigma-Aldrich s.r.l., Milan, Italy), followed by incubation (24 h at 39°C) in a buffered solution (pH 5) containing xylanase. Two sets of experiments were performed: a calibration trial (that tested different PreInc conditions on 9 selected forages) and a validation trial (that verified the results by testing multiple samples of 6 different forage types and a group of fibrous by-products). In the calibration trial, samples (300 mg in Ankom F57 filter bags; Ankom Technology Corp., Fairport, NY) were preincubated at 39°C in a 0.1 M NaOH solution for 90, 180, or 240 min, or in 0.2, 0.5, 1.0, or 2.0 M NaOH solution for 90 min. The results indicated that the best PreInc method, in terms of intra-laboratory repeatability and estimation of reference in situ values, was 90 min in a 0.2 M NaOH solution. Thus, we used this PreInc condition to determine enzymatic uNDF of 257 samples in the validation trial. Although the selected method generally had good accuracy in predicting in situ uNDF, inconsistencies were noted for certain forage types. Overall, when enzymatic uNDF was used to predict the in situ uNDF of all samples, the regression was satisfactory (intercept = 7.098, slope = 0.920, R2 = 0.73). The regression models developed for alfalfa hays, corn silages, and small grain silages had also acceptable regression performances and mean square error of prediction (MSEP) values, and the main sources of MSEP variation were error due to incomplete (co)variation and random error. Even when R2 values were >0.70, the MSEP value of the regression model for grass hays was 149.55, and that for nonforage fibrous feeds was 155.16. Although enzymatic uNDF partially overestimated the in situ uNDF, particularly in grass silages, the proposed procedure seems to be promising for accurately predicting in situ uNDF, because it generally had good repeatability and provided satisfactory estimates of in situ uNDF.

We investigated the effect of reconstitution of alfalfa hay on starter feed intake, nutrient digestibility, growth performance, rumen fermentation, selected blood metabolites, and health criteria of dairy calves during the pre- and postweaning periods. A total of 20 newborn male Holstein calves (3 d of age; 40.3 ± 1.30 kg of body weight; ±SE) were assigned randomly to 1 of 2 treatments, a starter feed containing either 10% dry (AH) or reconstituted alfalfa hay (RAH), each consisting of 10 calves. Alfalfa hay was reconstituted with water 24 h before feeding to achieve a theoretical dry matter content of 20%. Both starter feeds had the same ingredients and nutrient compositions but differed in their dry matter content (91.2 and 83.8% dry matter for AH and RAH, respectively). Calves were weaned on d 50 and remained on the study until d 70. All calves had free access to fresh and clean drinking water and the starter feed at all times. During the study period, the average maximum temperature-humidity index was 73.8 units, indicating no degree of environmental heat load for dairy calves. Starter feed dry matter intake, total dry matter intake, and body weight (at weaning and at the end of the trial) were unaffected by treatment. Nutrient intake (except for total ether extract intake) increased during the postweaning period compared with the preweaning period. Average daily gain and feed efficiency were unchanged between treatments. Calves had higher average daily gain and skeletal growth during the postweaning period; however, feed efficiency was lower during the post- versus preweaning period. Calves fed RAH gained more hip width and body barrel compared with calves fed AH during the preweaning and all studied periods, respectively. Rectal temperature was similar between treatments, but feeding RAH decreased fecal score and general appearance score during the preweaning period. Apparent total-tract nutrient digestibility was not affected by reconstitution of alfalfa hay; however, reconstitution increased total-tract digestibility of neutral detergent fiber during the postweaning period. Ruminal fluid pH, and concentrations and profile of total volatile fatty acids were unchanged between treatments. Molar concentration of propionate and acetate to propionate ratio increased and decreased, respectively, during the postweaning period. Reconstitution of alfalfa hay did not affect concentrations of glucose, β-hydroxybutyrate, blood urea N, and albumin, and albumin to globulin ratio during the studied periods; however, reconstitution increased concentration of blood total protein during the overall period. Calves had higher concentrations of blood glucose and globulin during the preweaning and β-hydroxybutyrate during the postweaning period. Overall, reconstitution of alfalfa hay did not interact with calf phase (pre- vs. postweaning) to affect dry matter intake, growth performance, and metabolic indications of rumen development (measured as ruminal volatile fatty acids and selected blood metabolites), but improved health-related variables (fecal score and general appearance score) during the preweaning period.

The objective of this study was to evaluate the effects of a fermentation byproduct on rumen fermentation and microbial yield in high producing lactating dairy cattle. Eight ruminally cannulated multiparous Holstein cows averaging (mean ± standard deviation) 60 ± 10 d in milk and 637 ± 38 kg of body weight were assigned to 1 of 2 treatment sequences in a switchback design. Treatment diets contained (dry matter basis) 44% corn silage, 13% alfalfa silage, 12% ground corn, and 31% premix containing either a control mix of urea and wheat middlings (CON) or a commercial fermentation byproduct meal (Fermenten, Arm and Hammer Animal Nutrition, Princeton, NJ) at 3% diet inclusion rate (EXP). Diets were formulated to be isonitrogenous and isocaloric, with similar levels of neutral detergent fiber and starch. The trial consisted of three 28-d experimental periods, where each period consisted of 21 d of diet adaptation and 7 d of data and sample collection. Omasal nutrient flows were determined using a triple-marker technique and double-labeled 15N15N-urea. The EXP diet provided 18 g/d more nonammonia N versus the CON diet, representing 3.0% of total N intake. Energy-corrected milk yield (41.7 and 43.1 kg/d for CON and EXP, respectively), milk fat, and protein yield and content did not differ between treatments. Total dry matter intake was similar between treatments (25.5 and 26.4 kg/d for CON and EXP, respectively). Ammonia N concentration and pool size in the rumen was greater in cows fed the EXP diet. No differences were observed in rumen or total-tract dry matter, organic matter, or neutral detergent fiber digestibility. Ruminal degradation of feed N was 15% lower in cows fed EXP diets, resulting in differences in omasal N flows. Results demonstrated the fermentation byproduct meal had a sparing effect on degradable feed protein, but did not increase microbial N flow from the rumen.

The objective of this study was to evaluate the effect of a fermentation by-product on rumen function, microbial yield, and composition and flows of nutrients from the rumen in high-producing lactating dairy cattle. A triple-marker technique and double-labeled 15N15N-urea were used to were used to measure protozoal, bacterial, and nonmicrobial omasal flow of AA. Rumen pool sizes and omasal flows were used to determine digestion parameters, including fractional rates of carbohydrate digestion, microbial growth, and yield of microbial biomass per gram of degraded substrate. Fermentation by-product inclusion in EXP diets increased microbial N and amino acid N content in microbes relative to microbes from CON cows fed the urea control. Microbial AA profile did not differ between diets. Daily omasal flows of AA were increased in EXP cows as a result of decreased degradation of feed protein. The inclusion of the fermentation by-product increased nonmicrobial AA flow in cows fed EXP versus CON. Average protozoal contribution to microbial N flow was 16.8%, yet protozoa accounted for 21% of the microbial AA flow, with a range of 8 to 46% for individual AA. Cows in this study maintained an average rumen pool size of 320 g of microbial N, and bacterial and protozoal pools were estimated at 4 different theoretical levels of selective protozoa retention. Fractional growth rate of all microbes was estimated to be 0.069 h-1, with a yield of 0.44 g of microbial biomass per gram of carbohydrate degraded. Results indicated that fermentation by-product can increase omasal flow of AA while maintaining adequate rumen N available for microbial growth and protein synthesis. Simulations from a developmental version of the Cornell Net Carbohydrate and Protein System indicated strong agreement between predicted and observed values, with some areas key for improvement in AA flow and bacterial versus protozoal N partitioning.

The digestion of neutral detergent fiber treated with amylase and sodium sulfite and ash corrected (aNDFom) has been described as a single digestible pool and a fraction undigested in ruminants. Further, most models that predict rate and extent of digestion of aNDFom in the rumen assume first-order processes, in which the rates of digestion and passage are proportional to the pool size of aNDFom consumed and digested. Data exist demonstrating that the undigested fraction of aNDFom is not well described by a fixed coefficient and varies by maturity and agronomic growing conditions of the plant. Thus, our objective was to improve the prediction of digestible aNDFom (pdNDF) and to quantify, using a minimum number of fermentation time points, 2 pools of digestible aNDFom, pdNDF1 and pdNDF2, and their respective rates. Based on fermentations from 0 to 240 h among 34 forages (grasses, conventional and brown midrib corn silages, and alfalfas), 3 pools were described by aNDFomt = pdNDF1 × e-k1(t-L) + pdNDF2 × e-k2(t-L) + uNDF, where aNDFomt is the residue at time t; L is the lag; k1 is the rate of digestion of pdNDF1; k2 is the rate of digestion of pdNDF2; and uNDF the unavailable NDF on an aNDFom basis. A nonlinear estimation allowed the computation of the pool size and respective digestion rates. Using 3 time points from the digestion curve, 30, 120, and 240 h, as the fermentation endpoints to represent uNDF, we optimized the same model in Vensim (Ventana Simulation Environment; Ventana Systems Inc., Belmont, MA) to obtain rates and pool sizes of aNDFom. In addition, the same optimization was also performed with 2 timepoints and a forage type-specific range for uNDF. Parameters (with and without uNDF) obtained per forage using Vensim were compared by fitting kinetics data from the nonlinear calculations, using coefficients of determination and residual mean squares at convergence for ranking purposes for the whole equation and mean squared prediction errors for specific parameters. The highest coefficient of determination (0.98) and lowest mean square prediction error [0.0927 (NDF-1)2] were obtained when using 48, 120, and 240 h of aNDFom residues or when using 30 and 120 h and a range for the forage-specific uNDFom. Correlations were in all cases consistently high for all kinetic parameters, ranging from 0.76 to 0.99. Results demonstrated that an adequate description of the heterogeneity of aNDFom disappearance was possible without multiple fermentation time points. The equation was fit to all data generated; however, because of the variable nature of pool sizes and rates, forage-specific equations should be developed for better estimations of the forage specific pool sizes and uNDF estimation. This study further describes the heterogeneous nature of aNDFom disappearance and provides an approach for estimating the individual pool sizes and rates of digestion for application for diet formulation.

This study compared dry matter (DM) predictions of 3 handheld near-infrared spectrophotometer (NIRS) units (Moisture Tracker, Digi-Star Inc., Fort Atkinson, WI) to conventional oven drying at 60°C using 2 alfalfa and 2 corn silages. In addition, on-farm DM methods [microwave, Koster tester (Koster Moisture Tester Inc., Brunswick, OH), and food dehydrator methods] were also compared. Corn and alfalfa silages (1,600 g) obtained from the University of Wisconsin Dairy Cattle Center (DCC) and the Arlington Research Station (ARS) were analyzed for DM daily for 20 d. Two NIRS calibration methods were also tested within each unit. The DM predicted from the factory-preset calibrations was NIRf. The adjusted DM prediction was NIRa, where the average difference between oven-dried and NIRf determined on duplicate forage samples for 3 d before the experiment was used as a bias adjustment for all subsequent DM determinations. The average predicted DM from the 20 scans was recorded as the forage DM. The process was repeated 3 times with each NIRS unit. Two 100-g subsamples of each forage were also oven-dried for 48 h at 60°C daily in a forced-air oven. Oven DM of ARS and DCC alfalfa silages were 37.3 ± 1.1% and 48.5 ± 1.9%, respectively (mean ± standard deviation). Oven DM of ARS and DCC corn silages were 34.7 ± 1.2% and 37.4 ± 0.5%, respectively (mean ± standard deviation). Dry matter determinations from NIRf were on average 3.5 units higher than the oven DM values. The NIRa DM predictions were on average 1.7 DM units lower than the oven DM values. Additionally, differences among the 3 NIRf probe results were detected (43.1, 40.7, and 41.3% DM, respectively), but all other results were similar between probes. Determinations of DM by the microwave and food dehydrator were also similar with the 60°C, 48-h oven method, whereas the Koster tester was lower than the oven. The handheld NIRS units more accurately predicted DM content of the alfalfa silage but were not as accurate with corn silages when the factory preset calibrations were corrected for bias.

Dietary lecithin is a source of choline. Our objective was to evaluate the effects of dietary deoiled soy lecithin feeding on circulating choline, choline metabolites, and the plasma phospholipid profile in lactating dairy cows fed fractionated palm fatty acids. In a split-plot Latin square design, 16 Holstein cows (160 ± 7 d in milk; 3.6 ± 1.2 parity) were randomly allocated to a main plot receiving a corn silage and alfalfa haylage-based diet with palm fat containing either moderate or high palmitic acid content at 1.75% of ration dry matter (moderate and high palmitic acid containing 72 or 99% palmitic acid in fat supplement, respectively; n = 8/palm fat diet). Within each palm fat group, deoiled soy lecithin was top-dressed at 0, 0.12, 0.24, or 0.36% of ration dry matter in a replicated 4 × 4 Latin square design with 14-d experimental periods. A 14-d covariate period was used to acclimate cows to palm fat feeding without lecithin supplementation. Blood sampling occurred during the final 3 d of each experimental period. Plasma choline and choline metabolites were quantified using liquid chromatography and mass spectrometry. Plasma phospholipids were profiled using time-of-flight mass spectrometry. Whereas no effects of treatments were detected for plasma choline or methionine, lecithin feeding increased the plasma concentrations of choline metabolites trimethylamine N-oxide and dimethylglycine (24 and 11%, respectively). Plasma phosphatidylcholine (PC) and sphingomyelin (SM) concentrations increased with deoiled lecithin feeding (e.g., PC 16:0/22:6 and SM d18:1/18:3). Lecithin supplementation also increased plasma lysophosphatidylcholine (LPC) concentrations (e.g., LPC 18:0) while reducing plasma phosphatidylethanolamine (PE) concentrations (e.g., PE 16:0/20:5). Although increases in microbial-derived trimethylamine N-oxide suggest gastrointestinal lecithin degradation, elevations in plasma dimethylglycine, PC, LPC, and SM suggest that choline availability was improved by lecithin feeding in cows, thus supporting enhanced endogenous phospholipid synthesis.

Dietary supplementation of alfalfa hay or calf starter during the preweaning period was beneficial to the gastrointestinal development in dairy calves and lambs. In the present study, we designed 2 experiments using weaning with calf starter and alfalfa hay to investigate the diet-ruminal microbiome-host crosstalk in yak calves by analyzing the ruminal microbiota and rumen epithelial transcriptome. During the preweaning period, supplementation with either alfalfa hay or the starter significantly promoted animal growth and organ development in yak calves, including increases in body weight, body height, body length, chest girth, and development of liver, spleen, and thymus. These improvements could be attributed to increased dry matter intake, rumen fermentation, and development. Butyrate concentration increased in yak calves fed alfalfa hay or the starter, which could further promote ruminal epithelium development. Using 16S rRNA gene amplicon sequencing, we determined that butyrate-producing genera were increased by the supplementation with alfalfa hay or the starter. Transcriptomic analysis of the rumen epithelia revealed that the PI3K-Akt signaling pathway, which is critical in mediating many aspects of cellular function such as cell growth, was upregulated in response to alfalfa hay or the starter supplementation. The starter supplementation also increased the jejunal α-amylase activity, whereas alfalfa hay supplementation reduced the ileal α-amylase activity. Furthermore, the co-supplementation of both the starter and alfalfa hay reduced intestinal α-amylase activity. The starter increased ruminal propionate concentration, whereas alfalfa hay exhibited the opposite trend. The observed opposite effects of the starter and alfalfa hay on rumen propionate concentration corresponded with up- and downregulation, respectively, of the ruminal cholecystokinin involved in pancreatic secretion pathway, and thereby increased and decreased pancreatic α-amylase activity. In conclusion, both alfalfa hay and the starter could promote the growth and ruminal epithelial development of yak calves. The starter and alfalfa hay also differentially affected the intestinal α-amylase activities due to their different chemical components and different effects on ruminal fermentation, especially the ruminal propionate production.

Filter bags facilitate the measurement of amylase-treated neutral detergent fiber (aNDF) and in vitro (IV) undigested aNDF (uNDF) by eliminating the transfer of residues from beakers into filtration crucibles. The objectives of this study were to (1) determine effects of filter bags on recovery of aNDF and (2) evaluate effects of filter bags on IV uNDF. For study 1, 6 samples each of grass hay (GR), alfalfa (AL), and corn silage (CS) were selected. Large standard deviations (SD) of ash-free aNDF (aNDFom) for samples in each forage type indicated compositional diversity (15.1, 7.45, and 12.9% of DM for GR, AL, and CS, respectively), and starch SD for CS was 16.4% of DM. Samples were weighed into Berzelius beakers or filter bags [25-µm pores (F57) or 6-µm pores (F58); Ankom Technology, Macedon, NY] for measurement of aNDF and aNDFom. All samples were extracted with neutral detergent, thermostable α-amylase, and sodium sulfite, and then soaked in boiling water and then acetone. Residues from beakers were filtered through a sea sand-covered GF/D filter (Whatman, Marlborough, MA) in Gooch crucibles (CR). Filter bags were extracted in a pressurized chamber at 100°C. The aNDF values did not differ between F57 and CR, but F58 was greater than CR for CS and AL. For GR, F58 was greater than CR for aNDFom. For study 2, diverse samples with large SD of aNDFom (20.7, 7.45, and 12.9% of DM for GR, AL, and CS, respectively) were weighed as loose powder into medium bottles (LS) or F57 bags, which were weighted to prevent floating. Blended ruminal fluid from 3 steers fed a 30% aNDFom diet was used as inoculum. Three samples of 1 forage type were randomly assigned to 1 of 6 IV runs using both treatments (LS and F57), and 3 bottles of each sample-treatment combination were removed after 12 h and 2 were removed after 120 h to measure uNDF. For LS, residues were extracted as in study 1 for CR. For F57, bags were rinsed in cold water and extracted as described in study 1. After 12 h, uNDF of F57 was greater than LS in CS, AL, and overall types. Ash-free uNDF (uNDFom) after 12 h of F57 was greater than LS in CS and overall types. After 120 h, F57 was greater than LS for uNDF of CS, but no differences were detected for uNDFom. The SD of uNDFom, but not uNDF, was higher after 12 and 120 h for F57 compared with LS. From 6 to 96 h, overall gas production of F57 was less than LS, and F57 was less than LS for CS from 3 to 96 h. Overall, LS gave greater maximum and faster rates of gas production than F57, as did AL and CS, but lag did not vary. Results indicate that filter bags affected aNDF and aNDFom measurement and inhibited fermentation for some materials.

The objective of this study was to determine effects of various forages and live yeast culture on intake, growth, nutrient digestibility, and ruminal fermentation of weaned dairy calves. Holstein calves (n = 45) were randomly assigned to 2 × 3 factorial treatments: live yeast culture or no yeast and alfalfa haylage (AH), corn silage (CS), or grass hay (GH). Calves were weaned at 6 wk of age, housed individually, and studied from 7 to 16 wk of age. Rations, consisting of an 18% crude protein texturized grower (yeast or no yeast) and assigned forage, were offered as separate components until 9 wk of age. After 9 wk, diets were offered as a total mixed ration (TMR). Concentrate intake was capped at 2.25 kg of dry matter (DM)/d, and forage was offered ad libitum. The TMR contained equal forage neutral detergent fiber (8 ± 0.5%) on a DM basis for each basal diet. Calves were fed TMR to limit concentrate intake, and additional forage was offered ad libitum after 8 h if the entire TMR allotment was consumed. Total fecal collection (12 calves) was conducted for 4 d at 11 and 15 wk of age. Feeds and feces were evaluated for DM, neutral detergent fiber, acid detergent fiber, and starch to calculate digestibility. On the last day of fecal collection, rumen samples were collected to evaluate pH and volatile fatty acid (VFA) profile. Metabolizable energy and DM intake was least for calves consuming GH compared with other forages. Forage intake (% of DM intake) increased as calves aged (AH = 20 to 44.4%, CS = 24.5 to 37.6%, GH = 11.3 to 32.3% at 10 and 16 wk of age, respectively). Calves on CS had the greatest average daily gain and empty body weight gain, and calves on GH had the least. Calves on GH tended to have the lowest final body weight. There were no differences in structural growth. Digestibility of DM decreased with age. Fiber digestibility was least for calves on CS, and starch digestibility was least for calves on AH. Mean and minimum rumen pH increased with age. Forage offered changed VFA profile and there was an interaction between yeast and forage on VFA profile. These results indicated that recently weaned calves perform well on AH, CS, or GH and have minimal benefit from yeast supplementation. Feeding GH reduced weight gain, but all calves achieved a level of gain to meet growth goals for breeding and freshening. Furthermore, the ability to consume large portions of the ration as forage allows for more economical diets to be fed.

Conceptually, there are 2 feeding strategies for avoiding over-conditioning, which can be problematic for gravid dairy heifers that have reduced dietary energy density requirements relative to younger animals: (1) diluting the ad libitum-fed diet with low-energy forages; or (2) offering a diet of greater nutrient density but intentionally restricting the DM available for consumption (limit-feeding). Our objectives for this study were to evaluate the effects of feedbunk restriction and feed push-up frequency on the growth performance of gravid Holstein dairy heifers. A total of 128 Holstein heifers (434 ± 46.7 kg) were enrolled in the trial. Heifers were blocked by weight, and assigned to 1 of 16 identical research pens (4 pens/weight block; 8 heifers/pen), where the mean initial body weight (BW; ± SD) for the 4 blocks were 491 ± 19.0, 450 ± 16.5, 419 ± 10.6, 374 ± 23.0 kg. Within each block, a 2 × 2 factorial arrangement of treatments was assigned; treatments consisted of feedbunk access [full (FUL) or restricted (RES] and feed push-up frequency [1.5- or 3.0-h intervals]. The RES treatment was applied by covering 2 of the 8 head-locking feed gates in assigned pens with plywood partitions, thereby creating a feedbunk-stocking rate of 133%. A total mixed ration diet composed of alfalfa haylage (60.5%), corn silage (38.0%), and mineral (1.5%) was offered once daily for 91 d; daily feed allotments (overall mean = 9.11 kg dry matter (DM)/d) were generally consumed entirely within 9 h of feeding. Nutrient intakes were not affected by push-up frequency or the interaction of main effects, but all intakes were affected by feedbunk access, except for DM and neutral detergent fiber expressed as a percentage of BW (overall means = 1.93 and 0.80%, respectively). In each case, intakes for FUL were greater than those observed for RES; for DM intake, this amounted to a difference of 0.20 kg/d between those main-effect treatments. After 91 d, heifers without feedbunk restriction exhibited greater final BW, but total gain and average daily gain differed only numerically between FUL and RES. Under the conditions of this trial, heifers were blocked by weight, such that BW were relatively uniform within each pen, and head-locking feed gates were used, which also provided some protection from adjacent aggressive heifers. These results suggest heifers can exhibit acceptable growth performance on high-forage diets in a limit-feeding program that includes moderate feedbunk restriction provided other forms of stress are minimized.

Feeding fat to lactating dairy cows may reduce methane production. Relative to cellulose, fermentation of hemicellulose is believed to result in less methane; however, these factors have not been studied simultaneously. Eight multiparous, lactating Jersey cows averaging (±SD) 98 ± 30.8 d in milk and body weight of 439.3 ± 56.7 kg were used in a twice-replicated 4 × 4 Latin square to determine the effects of fat and hemicellulose on energy utilization and methane production using a headbox-type indirect calorimetry method. To manipulate the concentration of fat, porcine tallow was included at either 0 or 2% of the diet dry matter. The concentration of hemicellulose was adjusted by manipulating the inclusion rate of corn silage, alfalfa hay, and soybean hulls resulting in either 11.3 or 12.7% hemicellulose (dry matter basis). The resulting factorial arrangement of treatments were low fat low hemicellulose (LFLH), low fat high hemicellulose (LFHH), high fat low hemicellulose (HFLH), and high fat high hemicellulose (HFHH). Neither fat nor hemicellulose affected dry matter intake, averaging 16.2 ± 1.18 kg/d across treatments. Likewise, treatments did not affect milk production, averaging 23.0 ± 1.72 kg/d, or energy-corrected milk, averaging 30.1 ± 2.41 kg/d. The inclusion of fat tended to reduce methane produced per kilogram of dry matter intake from 24.9 to 23.1 ± 1.59 L/kg, whereas hemicellulose had no effect. Increasing hemicellulose increased neutral detergent fiber (NDF) digestibility from 43.0 to 51.1 ± 2.35%. Similarly, increasing hemicellulose concentration increased total intake of digestible NDF from 6.62 to 8.42 ± 0.89 kg/d, whereas fat had no effect. Methane per unit of digested NDF tended to decrease from 64.8 to 49.2 ± 9.60 L/kg with increasing hemicellulose, whereas fat had no effect. An interaction between hemicellulose and fat content on net energy balance (milk plus tissue energy) was observed. Specifically, increasing hemicellulose in low-fat diets tended to increase net energy balance, but this was not observed in high-fat diets. These results confirm that methane production may be reduced with the inclusion of fat, whereas energy utilization of lactating dairy cows is improved by increasing hemicellulose in low-fat diets.

Groundwater depletion is one of the most pressing issues facing the dairy industry in arid regions. One strategy to improve the industry's drought resilience involves feeding drought-tolerant forage crops in place of traditional forage crops such as alfalfa and corn silage. The objective of this study was to assess the productivity of lactating dairy cows fed diets with teff hay (Eragrostis tef) as the sole forage. Teff is a warm-season annual grass native to Ethiopia that is well adapted to drought conditions. Nine multiparous Holstein cows (185 ± 31 d in milk; mean ± standard deviation) were randomly assigned to 1 of 3 diets in a 3 × 3 Latin square design with 18-d periods (14 d acclimation and 4 d sampling). Diets were either control, where dietary forage consisted of a combination of corn silage, alfalfa hay, and native grass hay, or 1 of 2 teff diets (teff-A and teff-B), where teff hay [13.97 ± 0.32% crude protein, dry matter (DM) basis] was the sole forage. All 3 diets were formulated for similar DM, crude protein, and nonfiber carbohydrate concentrations. Control and teff-A were matched for concentrations of neutral detergent fiber (NDF) from forage (18.2 ± 0.15% of DM), and teff-B included slightly less, providing 16.6% NDF from forage. Dry matter intake, milk and component production, body weight, body condition score, as well as DM and NDF digestibility were monitored and assessed using mixed model analysis, with significance declared at P < 0.05. Treatment had no effect on dry matter intake (28.1 ± 0.75 kg/d). Similarly, treatment had no effect on milk production (40.7 ± 1.8 kg/d). Concentrations of milk fat (3.90 ± 0.16%) and lactose (4.68 ± 0.07%) were also unaffected by treatment. Teff-A and teff-B increased milk protein concentration compared with the control (3.07 vs. 3.16 ± 0.09%). Treatment had no effect on energy-corrected milk yield (43.4 ± 1.3 kg/d), body weight, or body condition score change. Additionally, treatment had no effect on total-tract DM or NDF digestibility. Results from this study indicate that teff hay has potential to replace alfalfa and corn silage in the diets of lactating dairy cattle without loss of productivity.

We investigated the interactive effects of forage source and forage particle size (PS) as a free-choice provision on growth performance, rumen fermentation, and behavior of dairy calves fed texturized starters. Forty-eight Holstein calves (42 ± 3 kg of body weight) were randomly assigned (n = 12 calves per treatment) in a 2 × 2 factorial arrangement of treatments with the factors of forage source [alfalfa hay (AH) and wheat straw (WS)] and forage PS [(AH: medium = 1.96 mm or long = 3.93 mm) and (WS: medium = 2.03 mm or long = 4.10 mm), as geometric mean diameters]. The treatments were (1) AH with medium PS (AH-MPS), (2) AH with long PS (AH-LPS), (3) WS with medium PS (WS-MPS), and (4) WS with long PS (WS-LPS). Regardless of forage PS, the preweaning starter intake, dry matter intake, metabolizable energy intake, weaning body weight, and forage intake were greater for AH calves than WS calves. Average daily gain, average daily gain/metabolizable energy intake, feed efficiency, and final body weight of the calves did not differ among groups. An interaction of forage source and forage PS influenced acetate, propionate, and acetate-to-propionate ratio in the rumen on d 35, with the greatest acetate proportion and acetate-to-propionate ratio, but the least propionate proportion for AH-MPS calves than the other calves. The total volatile fatty acid concentration and the rumen proportions of propionate (d 70), butyrate (d 35), and valerate (d 35) were greater in AH-MPS calves than in AH-LPS calves. Calves fed AH had greater total volatile fatty acid concentration (d 35 and 70) and propionate proportion (d 70), but lesser ruminal proportions of butyrate (d 35 and 70), valerate (d 35 and 70), and acetate-to-propionate ratio (d 70) compared with calves fed WS. The ruminal valerate proportion (d 70) was greatest in WS-MPS calves than the other calves. An interaction of forage source and forage PS influenced preweaning standing time and starter eating time, with the least standing time for WS-MPS calves and the greatest eating starter time for AH-LPS calves. Calves fed AH spent less time for rumination, but devoted more time to non-nutritive oral behaviors than WS calves. Calves fed forage with long PS spent more time for rumination, eating forage, and spent less time lying and non-nutritive oral behaviors than medium PS. In conclusion, forage source and PS interacted, affecting behavior and rumen fermentation when calves were fed texturized starters. In addition, a desirable ruminal pH in dairy calves can be obtained with texturized starters.

The first objective of this study was to examine effects of adding Escherichia coli O157:H7 with or without chemical or microbial additives on the bacterial diversity and composition of alfalfa silage. The second objective was to examine associations between the relative abundance of known and unknown bacterial species and indices of silage fermentation quality. Alfalfa forage was harvested at 54% dry matter, chopped to a theoretical length of cut of 19 mm, and ensiled in quadruplicate in laboratory silos for 100 d after the following treatments were applied: (1) distilled water (control); (2) 1 × 10 5 cfu/g of E. coli O157:H7 (EC); (3) EC and 1 × 10 6 cfu/g of Lactobacillus plantarum (EC+LP); (4) EC and 1 × 10 6 cfu/g of Lactobacillus buchneri (EC+LB); and (5) EC and 0.22% propionic acid (EC+PA). After 100 d of ensiling, the silage samples were analyzed for bacterial diversity and composition via the Illumina MiSeq platform (Illumina Inc., San Diego, CA) and chemically characterized. Overall, Firmicutes (74.1 ± 4.86%) was the most predominant phylum followed by Proteobacteria (20.4 ± 3.80%). Relative to the control, adding E. coli O157:H7 alone at ensiling did not affect bacterial diversity or composition but adding EC+LP or EC+LB reduced the Shannon index, a measure of diversity (3.21 vs. 2.63 or 2.80, respectively). The relative abundance of Firmicutes (69.2 and 68.8%) was reduced, whereas that of Proteobacteria (24.0 and 24.9%) was increased by EC+LP and EC+PA treatments, relative to those of the control (79.5 and 16.5%) and EC+LB (77.4 and 18.5%) silages, respectively. Compared with the control, treatment with EC+LP increased the relative abundance of Lactobacillus, Sphingomonas, Pantoea, Pseudomonas , and Erwinia by 426, 157, 200, 194, and 163%, respectively, but reduced those of Pediococcus, Weissella , and Methylobacterium by 5,436, 763, and 250%, respectively. Relative abundance of Weissella (9.19%) and Methylobacterium (0.94%) were also reduced in the EC+LB silage compared with the control (29.7 and 1.50%, respectively). Application of propionic acid did not affect the relative abundance of Lactobacillus, Weissella , or Pediococcus. Lactate concentration correlated positively (r = 0.56) with relative abundance of Lactobacillus and negatively (r = −0.41) with relative abundance of Pediococcus . Negative correlations were detected between ammonia-N concentration and relative abundance of Sphingomonas (r = −0.51), Pantoea (r = −0.46), Pseudomonas (r = −0.45), and Stenotrophomonas (r = −0.38). Silage pH was negatively correlated with relative abundance of Lactobacillus (r = −0.59), Sphingomonas (r = −0.66), Pantoea (r = −0.69), Pseudomonas (r = −0.69), and Stenotrophomonas (r = −0.50). Future studies should aim to speciate, culture, and determine the functions of the unknown bacteria detected in this study to elucidate their roles in silage fermentation.

Feeding trials were conducted with lactating cows and growing lambs to quantify effects of replacing dietary alfalfa silage (AS) with red clover silage (RCS) on nutrient utilization. The lactation trial had a 2 × 4 arrangement of treatments: AS or RCS fed with no supplement, rumen-protected Met (RPM), rumen-protected Lys (RPL), or RPM plus RPL. Grass silage was fed at 13% of dry matter (DM) with AS to equalize dietary neutral detergent fiber (NDF) and crude protein contents. All diets contained (DM basis) 5% corn silage and 16% crude protein. Thirty-two multiparous (4 ruminally cannulated) plus 16 primiparous Holstein cows were blocked by parity and days in milk and fed diets as total mixed rations in an incomplete 8 × 8 Latin square trial with four 28-d periods. Production data (over the last 14 d of each period) and digestibility and excretion data (at the end of each period) were analyzed using the MIXED procedure of SAS (SAS Institute Inc., Cary, NC). Although DM intake was 1.2 kg/d greater on AS than RCS, milk yield and body weight gain were not different. However, yields of fat and energy-corrected milk as well as milk content of fat, true protein, and solids-not-fat were greater on AS. Relative to AS, feeding RCS increased milk and energy-corrected milk yield per unit of DM intake, milk lactose content, and apparent N efficiency and reduced milk urea. Relative to AS, apparent digestibility of DM, organic matter, NDF, and acid detergent fiber were greater on RCS, whereas apparent and estimated true N digestibility were lower. Urinary N excretion and ruminal concentrations of ammonia, total AA, and branched-chain volatile fatty acids were reduced on RCS, indicating reduced ruminal protein degradation. Supplementation of RPM increased intake, milk true protein, and solids-not-fat content and tended to increase milk fat content. There were no silage × RPM interactions, suggesting that RPM was equally limiting on both AS and RCS. Supplementation of RPL did not influence any production trait; however, a significant silage × RPL interaction was detected for intake: RPL reduced intake of AS diets but increased intake of RCS diets. Duplicated metabolism trials were conducted with lambs confined to metabolism crates and fed only silage. After adaptation, collections of silage refusals and excreta were made during ad libitum feeding followed by feeding DM restricted to 2% of body weight. Intake of DM was not different when silages were fed ad libitum. Apparent digestibility of DM, organic matter, NDF, and hemicellulose was greater in lambs fed RCS on both ad libitum and restricted intake; however, acid detergent fiber digestibility was only greater at restricted intake. Apparent and estimated true N digestibility was substantially lower, and N retention was reduced, on RCS. Results confirmed greater DM and fiber digestibility in ruminants and N efficiency in cows fed RCS. Specific loss of Lys bioavailability on RCS was not observed. Based on milk composition, Met was the first-limiting AA on both silages; however, Met was not limiting based on production and nutrient efficiency. Depressed true N digestibility suggested impaired intestinal digestibility of rumen-undegraded protein from RCS.

This study aimed to examine the temporal (hourly within a day and daily over the long term) effects of monensin on CH4 emissions, ruminal fermentation, and in situ alfalfa degradation in dairy goats during dietary monensin supplementation by controlling the confounding effects of feed intake and ambient temperature. Six ruminally cannulated dairy goats were used, and they were housed in environmental chambers and fed a restricted amount of ration throughout the experiment. The experiment included a baseline period of 20 d followed by a treatment period of 55 d with 32 mg of monensin/d. During the whole experiment, CH4 production was measured every 5 d, whereas fermentation characteristics and in situ alfalfa degradation were analyzed every 10 d. The CH4-depressing effect of monensin was time dependent on the duration of treatment, highly effective at d 5 but thereafter decreased gradually until d 55 even though CH4-suppressing effect still remained significant. The decreasing effects of monensin on ruminal acetate proportion and acetate to propionate ratio also faded over days of treatment, and the acetate proportion returned up to the pre-supplementation level on d 50. Monensin supplementation elevated ruminal propionate proportion and decreased the effective ruminal degradability of alfalfa NDF, but both measurements tended to recover over time. The postprandial increase rate of hourly CH4 emissions was reduced, whereas that of propionate proportion was enhanced by monensin supplementation. However, the postprandial responses to monensin in CH4 emission rates, ruminal VFA profiles, and in situ degradation kinetics declined with both hours after feeding and days of treatment. Our results suggest that the CH4-suppressing effect of monensin supplementation in goats was attributed to reductions in both ruminal feed degradation and acetate to propionate ratio, but those reductions faded with time, hours after feeding, and days of treatment.

Double cropping and increasing crop diversity could improve dairy farm economic and environmental sustainability. In this experiment, corn silage was partially replaced with 2 alternative forages, brown midrib-6 brachytic dwarf forage sorghum (Sorghum bicolor) or fall-grown oat (Avena sativa) silage, in the diet of lactating dairy cows. We investigated the effect on dry matter (DM) intake, milk yield (MY), milk components and fatty acid profile, apparent total-tract nutrient digestibility, N utilization, enteric methane emissions, and income over feed cost. We analyzed the in situ DM and neutral detergent fiber disappearance of the alternative forages versus corn silage and alfalfa haylage. Sorghum was grown in the summer and harvested in the milk stage. Oats were grown in the fall and harvested in the boot stage. Compared with corn silage, neutral detergent fiber and acid detergent fiber concentrations were higher in the alternative forages. Lignin content was highest for sorghum silage and similar for corn silage and oat silage. The alternative forages had less than 1% starch compared with the approximately 35% starch in the corn silage. Ruminal in situ DM effective degradability was similar, although statistically different, for corn silage and oat silage, but lower for sorghum silage. Diets with the alternative forages were fed in a replicated 3 × 3 Latin square design experiment with three 28-d periods and 12 Holstein cows. The control diet contained 44% (DM basis) corn silage. In the other 2 diets, sorghum or oat silages were included at 10% of dietary DM, replacing corn silage. Sorghum silage inclusion decreased DM intake, MY, and milk protein content but increased milk fat and maintained energy-corrected MY similar to the control. Oat silage had no effect on DM intake, MY, or milk components compared to the control. The oat silage diet increased apparent total-tract digestibility of dietary nutrients, except starch, whereas the sorghum diet slightly decreased DM, organic matter, crude protein, and starch digestibility. Cows consuming the oat silage diet had higher milk urea N and urinary urea N concentrations. Milk N efficiency was decreased by the sorghum diet. Diet did not affect enteric methane or carbon dioxide emissions. This study shows that oat silage can partially replace corn silage at 10% of the diet DM with no effect on MY. Intensive dairy goat production in the Mediterranean basin is based on imported conventional ingredients to be included in concentrates. Fourteen Murciano-Granadina goats in the middle of the third lactation were allocated into 2 groups of 7 animals each fed, respectively, a control diet based on alfalfa hay and concentrate in a 40:60 ratio, and a diet in which the concentrate included tomato fruits, citrus pulp, brewer's grain and brewer's yeast (T100CBY) to study the effect of diet on nutrient utilization, ruminal fermentation, purine derivatives excretion in urine, milk yield and composition, and methane emissions. No effect of the diet on total dry matter intake was observed. Digestibility of neutral detergent fiber and acid detergent fiber were higher for T100CBY compared with the control diet. The N in feces and urine was lower and balance and retained N were higher in animals fed T100CBY than the control diet. Milk protein N and energy were similar for both diets. Metabolizable energy per energy intake and metabolizable energy per digestible energy were higher and energy in methane was lower with diet T100CBY than with the control. Milk yield and composition were not affected by diet, with the exception of protein, casein, and total solids, which were higher for diet T100CBY than the control. Diet T100CBY promoted less saturated fatty acids and higher mono- and polyunsaturated fatty acids in milk than the control diet. Diet T100CBY produced less methane and NH3 concentration in the rumen, higher propionate, and a lower acetate-to-propionate ratio without an effect on the volatile fatty acid concentration. The concentrate with by-products did not affect urinary excretion of total purine derivatives, reduced feeding costs, and increased profit margin by 14 and 16% compared with the control. The mixture of tomato fruits, citrus pulp, brewer's grain, and brewer's yeast could replace 47% of conventional ingredients (corn, wheat bran, sunflower meal, and soy flour) in the concentrate of the dairy goat diet, reducing feeding cost and methane production, leading to a healthier fatty acids profile in milk without compromising nutrient utilization or milk yield.

The objective of this study was to investigate the effects of 2 alfalfa hays differing in undigested neutral detergent fiber content and digestibility used as the main forage source in diets fed to high producing cows for Parmigiano-Reggiano cheese production. Diets were designed to have 2 different amounts of undigestible NDF [high (Hu) and low (Lu)], as determined by 240-h in vitro analysis (uNDF240). Alfalfa hay in vitro digestibility [% of amylase- and sodium sulfite-treated NDF with ash correction (aNDFom)] at 24 and 240 h was 40.2 and 31.2% and 53.6 and 45.7% for low- (LD) and high-digestibility (HD) hays, respectively. The 4 experimental diets (Hu-HD, Lu-HD, Hu-LD, and Lu-LD) contained 46.8, 36.8, 38.8, and 30.1% of alfalfa hay, respectively, 8.6% wheat straw, and 35.3% corn (50% flake and 50% meal; DM basis). Soy hulls and soybean meal were used to replace hay to balance protein and energy among diets. Eight multiparous Holstein cows (average milk production = 46.0 ± 5.2 kg/d, 101 ± 38 d in milk, and 662 ± 42 kg of average body weight) were assigned to a 4 × 4 Latin square design, with 2 wk of adaptation and a 1-wk collection period. Dry matter and water intake, rumination time, ruminal pH, and milk production and composition were measured. Diets and feces were analyzed for NDF on an organic matter basis (aNDFom), acid detergent fiber, acid detergent lignin, and uNDF240 to estimate total-tract fiber digestibility. Dry matter intake and rumination times were higher in HD diets compared with LD diets, regardless of forage amount. Rumination time was constant per unit of dry matter intake but differed when expressed as a function of uNDF240, aNDFom, or physically effective NDF intake. No differences were found among treatments on average ruminal pH, but the amount of time with pH

Two lactation trials were conducted comparing the feeding value of silages made from birdsfoot trefoil (BFT, Lotus corniculatus L.) that had been selected for low (BFTL), medium (BFTM), and high (BFTH) levels of condensed tannins (CT) to an alfalfa silage (AS) when fed as the principal forage in total mixed rations. Diets also included corn silage, high-moisture shelled corn, soybean meal, soy hulls, and supplemental fat. In trial 1, 32 lactating Holstein cows were blocked by days in milk, assigned to treatment sequences in 8 balanced 4 × 4 Latin squares, and fed 50% dietary dry matter from AS or 1 of 3 BFT silages containing 0.6, 1.2, or 1.7% CT. Diets averaged 17.5 to 19.5% crude protein and 26% neutral detergent fiber on a dry matter basis. Data were collected over the last 2 wk of each 4-wk period. Intakes were 1.3 to 2.8 kg of dry matter/d greater on BFT than on AS and cows gained 0.5 kg of body weight/d on BFT diets while losing 0.14 kg of body weight/d on the AS diet; this resulted in greater milk per dry matter intake (DMI) on AS. Linear effects indicated true protein yield and milk urea nitrogen declined with increasing CT concentration and quadratic effects indicated DMI, energy-corrected milk, and fat yield were increased at intermediate CT concentration. True protein yield and apparent N-efficiency were greater, and milk urea nitrogen lower, on all BFT diets than on AS. In trial 2, 50 lactating Holstein cows were fed a covariate AS diet for 2 wk and then blocked by parity and days in milk and randomly assigned to 1 of 5 diets that were fed continuously for 12 wk. Diets contained (dry matter basis) 48% AS, 16% AS plus 32% of 1 of 3 BFT silages with 0.5, 0.8, or 1.5% CT, or 48% of an equal mixture of each BFT silage. Diets averaged 16.5% crude protein and 30% neutral detergent fiber. Intake and milk yield tended to be lower on AS than BFT, but body weight gains averaged 0.6 kg/d on all diets. Cows fed any of the BFT silages had reduced milk urea nitrogen and ruminal ammonia and reduced urinary N excretion. Feeding the BFT mixture reduced concentrations of milk true protein and milk urea nitrogen and depressed apparent nutrient digestibility. Among diets containing the individual BFT silages, linear reductions in DMI and yield of milk, fat, true protein, lactose, and SNF were observed with increasing CT concentration. By contrast, a previous trial with the same BFT populations showed that substituting BFTH silage containing 1.6% CT for AS in rations containing 60% silage dry matter had no effect on intake, increased yield of milk, energy-corrected milk and milk components, elevated protein use-efficiency, but with a more modest reduction in milk urea nitrogen and urinary N excretion. Silage analyses suggested that the inconsistent responses among trials were related to growth environment or ensiling effects that altered tannin-protein interactions in BFT silage. Differences in diet formulation among trials may have also influenced responses. Results from the current and previous trials indicate further work is needed to identify optimum tannin levels in forages.

Our objectives were to evaluate technical lignins for their antifungal properties against 3 molds and 1 yeast causing hay spoilage, and their ability to preserve ground high-moisture alfalfa hay nutritive value in vitro. In experiment 1, 8 technical lignins and propionic acid (PRP; positive control) were tested at a dose of 40 mg/mL. The experiment had a randomized complete block design (RCBD, 4 runs) and a factorial arrangement of 3 molds × 10 additives (ADV). The effects of the ADV on yeast were evaluated separately with a RCBD. Sodium lignosulfonate (NaL) and PRP were the only treatments with 100 ± 2.8% inhibition of fungi. In experiment 2, the minimum inhibitory concentration (MIC) for selected lignins and PRP were determined. At pH 4, NaL had the lowest MIC across the molds (20-33.3 mg/mL) and magnesium lignosulfonate (MgL) for the yeast (26.7) among the lignins. However, PRP had MIC values that were several-fold lower across all fungi (1.25-3.33). In experiment 3, a RCBD (5 blocks) with a 3 (ADV; NaL, MgL, and PRP) × 4 (doses: 0, 0.5, 1, and 3% wt/wt fresh basis) factorial arrangement of treatments was used to evaluate the preservative effects of ADV in ground high-moisture alfalfa hay inoculated with a mixture of the fungi previously tested and incubated under aerobic conditions in vitro. After 15 d, relative to untreated hay (14.9), dry matter (DM) losses were lessened by doses as low as 1% for NaL (3.39) and 0.5% for PRP (0.81 ± 0.77%). The mold count was reduced in both NaL at 3% (3.92) and PRP as low as 0.5% (3.94) relative to untreated hay (7.76 ± 0.55 log cfu/fresh g). Consequently, sugars were best preserved by NaL at 3% (10.1) and PRP as low as 0.5% (10.5) versus untreated (7.99 ± 0.283% DM), while keeping neutral detergent fiber values lower in NaL (45.9) and PRP-treated (45.1) hays at the same doses, respectively, relative to untreated (49.7 ± 0.66% DM). Hay DM digestibility was increased by doses as low as 3% for NaL (67.5), 1% MgL (67.0), and 0.5% PRP (68.5) versus untreated hay (61.8 ± 0.77%). The lowest doses increasing neutral detergent fiber digestibility relative to untreated hay (23.3) were 0.5% for MgL and PRP (30.5 and 30.1, respectively) and 1% for NaL (30.7 ± 1.09% DM). Across technical lignins, NaL showed the most promise as a potential hay preservative. However, its effects were limited compared with PRP at equivalent doses. Despite not having an effect on preservation, MgL improved DM digestibility by stimulating neutral detergent fiber digestibility. This study warrants further development of NaL under field conditions.

The objective of this study was to evaluate the effects of a commercially available fermentation by-product in a diet containing adequate rumen-degradable protein (RDP) on milk performance, intake, and total-tract nutrient digestion in lactating dairy cattle. Primiparous (n = 48) and multiparous (n = 144) lactating dairy cattle were stratified by milk production and randomly allocated into 12 pens containing 4 primiparous and 12 multiparous animals each. Cattle averaged 118 d in milk and 712 kg of body weight at trial start. Treatment diets, on a dry matter (DM) basis, consisted of 42% corn silage, 13% alfalfa hay and silage, 20% grain corn, and 25% protein premix containing either soybean meal, wheat middlings, and urea (SBM+U), soybean meal and fermentation by-product (SBM+F), or soybean meal and rumen-protected soybean meal (RP-SBM). All 3 diets provided a similar level (DM basis) of neutral detergent fiber analyzed using α-amylase and sodium sulfite and corrected for ash content (31%), crude protein (CP, 14.9%), starch (26%), and metabolizable energy (2.7 Mcal/kg), and differed in sources of RDP. The trial consisted of a 2-wk adaptation and covariate period during which all cows were fed the RP-SBM diet and covariate measurements were taken. Pens were then randomly allocated to treatments, and weekly measurements of milk production, intake, body weight, and condition score were taken for 10 wk. All data were analyzed using the Proc Mixed procedure in SAS (SAS Institute Inc., Cary, NC). Increased DM intake was observed for cows fed SBM+F compared with cows fed SBM+U and RP-SBM (28.3 vs. 26.9 and 26.7 kg/d, respectively). Cows fed SBM+F produced more energy-corrected milk (45.3 kg/d) compared with cows fed SBM+U and RP-SBM (43.6 and 43.7 kg/d, respectively). Milk protein yield was also increased in cows fed SBM+F. No differences were observed with body weight or condition score gain throughout the trial. Apparent total-tract digestibility of fiber was decreased in cows fed SBM+F, likely as a result of increased intake. Responses are consistent with previous research in our laboratory that demonstrated a decrease in ruminal CP degradation, leading to an increase in metabolizable protein supply in the small intestine. The fermentation by-product might be useful in diets containing adequate amounts of RDP from soybean meal or alfalfa. The results from this experiment demonstrate beneficial milk performance responses to fermentation by-product when fed with a source of RDP.

Our objective was to determine the effects of replacing alfalfa silage (AS) neutral detergent fiber (NDF) with corn silage (CS) NDF at 2 levels of forage NDF (FNDF) on enteric methane (CH4), lactation performance, ruminal fluid characteristics, digestibility, and metabolism of N and energy in Holstein and Jersey cows. Twelve Holstein and 12 Jersey cows (all primiparous and mid-lactation) were used in a triplicated split-plot 4 × 4 Latin square experiment, where breed and diet formed the main and subplots, respectively. The 4 iso-nitrogenous and iso-starch dietary treatments were arranged as a 2 × 2 factorial with 2 levels of FNDF [19 (low FNDF, LF) and 24% (high FNDF, HF) of dry matter] and 2 sources of FNDF (70:30 and 30:70 ratio of AS NDF to CS NDF). Soyhull (non-forage NDF) and corn grain were respectively used to keep dietary NDF and starch content similar across diets. Total collection of feces and urine over 3 d was performed on 8 cows (1 Latin square from each breed). The difference in dry matter intake (DMI) between Holsteins and Jerseys was greater when fed AS than CS. Compared with Jerseys, Holstein cows had greater body weight (48%), DMI (34%), fat- and protein-corrected milk (FPCM; 31%) and CH4 production (22%; 471 vs. 385 g/d). However, breed did not affect CH4 intensity (g/kg of FPCM) or yield (g/kg of DMI), nutrient digestibility, and N partitioning. Compared with HF, LF-fed cows had greater DMI (10%), N intake (8%), and FPCM (5%), but they were 5% less efficient (both FPCM/DMI and milk N/intake N). Compared with HF, LF-fed cows excreted 11 and 17% less urinary N (g/d and % of N intake, respectively). In spite of lower (2.5%) acetate and higher (10%) propionate (mol/100 mol ruminal volatile fatty acids) LF-fed cows had greater (6%) CH4 production (g/d) than did HF-fed cows, most likely due to increased DMI, as affected mainly by the soyhulls. Compared with AS, CS-fed cows had greater DMI (7%) and FPCM (4%), but they were less efficient (5%), and CH4 yield (g/kg of DMI) was reduced by 8%. In addition, per unit of gross energy intake, CS-fed cows lost less urinary energy (15%) and CH energy (11%) than did AS-fed cows. We concluded that, in contrast to level and source of FNDF, breed did not affect digestive and metabolic efficiencies, and, furthermore, neither breed nor dietary treatments affected CH4 intensity. The tradeoff between CH4 and N losses may have implications in future studies assessing the environmental effects of milk production when approached from a whole-farm perspective.

The aim of this study was to reveal the reconstitution dynamics of alfalfa microbiota and their contribution to the fermentation quality of Napier grass silages. Napier grass was harvested at approximately 20% dry matter content, chopped to a theoretical length of cut of 2 to 3 cm, and ensiled in laboratory polyethylene plastic bags (400 × 250 mm) for 30 d. The Napier grass was treated with the following: natural fermentation and distilled water (NG), inoculum of alfalfa microbiota (AM), gamma-ray irradiation and distilled water (IR), and gamma-ray radiation and alfalfa microbiota (IR+AM). Three milliliters of inoculum (containing 8.93 log cfu/mL lactic acid bacteria, 9.76 log cfu/mL Enterobacteriaceae, 5.94 log cfu/mL yeast, and 6.53 log cfu/mL mold) eluted from equivalent fresh alfalfa (450 g) was added to each silo of AM and IR+AM treatments, and 3 mL of distilled water was added to the silo of the NG and IR treatments. Three triplicate silos per treatment were opened on d 1, 3, 5, 7, 14, and 30 for sampling and analysis of fermentation quality and bacterial community. Relative to the NG silages, IR+AM silages exhibited a higher lactic acid concentration. The higher acetic acid concentration in NG than in IR+AM silages after 7 d of ensiling was attributed to the dominant genus of Leuconostoc (64.29-49.04%). Adding alfalfa microbiota to sterile Napier grass could increase ammonia-N concentration compared with NG and IR silages after 3 d of ensiling. Leuconostoc was the most predominant genus in NG silages, followed by Lactobacillus. Pediococcus had a greater relative abundance than the indigenous microorganisms and was exclusively found in AM and IR+AM silages, whereas Lactobacillus exhibited a slight increase after 30 d of ensiling (relative abundance in each silage: 6.29 vs. 3.82%, respectively). Lactobacillus was the predominant genus in IR silages since the onset of the ensiling. These results suggest that alfalfa microbiota affected bacterial community succession in Napier grass silages, which in turn affected the fermentation products. Adding alfalfa microbiota to sterile Napier grass could decrease acetic acid concentration compared with NG silages; however, it increased ammonia-N concentration compared with IR silages after 3 d of ensiling.

Nitrogen concentrations in feeds, feces, milk, and urine samples were measured using 2 analytical methods following different drying procedures. Ten samples of corn silage, alfalfa silage, and concentrates collected from 2017 to 2018 at Krauss Dairy Research Center, The Ohio State University (Wooster), were used. Our objective was to evaluate the effects of replacing either corn or alfalfa silage with tall fescue hay on total-tract neutral detergent fiber (NDF) digestibility and lactation performance in dairy cows. Twenty-four primiparous (75±35 d in milk) and 40 multiparous (68±19 d in milk) Holstein cows were blocked by parity and randomly assigned to 1 of 4 treatment groups in a pen equipped with 32 feeding gates to record intake by cow. Each gate was randomly assigned to 1 treatment group; thus, each cow had access to all 8 gates within the respective treatment and cow was the experimental unit. Treatments were formulated to replace either corn silage (CS) or alfalfa silage (AS) with tall fescue hay (TF) as follows (DM basis): 33% AS and 67% CS (control; 33AS67CS), 60% TF and 40% AS (60TF40AS), 60% TF and 40% CS (60TF40CS), and 33% TF and 67% CS (33TF67CS). The experiment was a 7-wk continuous lactation trial with a 2-wk covariate period. Milk production did not differ among treatments and averaged 40.4 kg/d. Fat yield and concentration and protein yield and concentration did not differ among treatments and averaged 1.58 kg/d, 3.94%, 1.28 kg/d, and 3.15%, respectively. Dry matter intake was greater for 33AS67CS (24.5 kg/d) compared with 60TF40CS (22.1 kg/d) and 33TF67CS (22.7 kg/d), and tended to be greater than 60TF40AS (23.2 kg/d). In vivo total-tract dry matter digestibility did not differ among treatments and averaged 66.2%. In vivo total-tract NDF digestibility was lower for 33AS67CS (37.8%) compared with 60TF40AS (44.4%) and 33TF67CS (45.3%), and similar to 60TF40CS (42.4%). In vivo total-tract NDF digestibility and an estimate of in situ total-tract NDF digestibility were similar between techniques across all treatment diets (42.3 vs. 42.6%, respectively). Inclusion of tall fescue grass hay increased the total-tract NDF digestibility of the diet and has the potential to replace corn silage and alfalfa silage and maintain milk production if economically feasible based on current market prices.

The objective of the study was to determine the effect of feeding sainfoin (SF; Onobrychis viciifolia) and birdsfoot trefoil (BT; Lotus corniculatus), 2 temperate climate forage legumes that contain condensed tannins (CT), on ruminal fermentation and N turnover in dairy cows. Six ruminally cannulated multiparous dairy cows (milk yield = 40 kg/d; 36 d in milk) were used in a replicated 3 × 3 Latin square design. All animals were fed basal diets containing 20% pelleted SF (223 g of CT/kg of dry matter), BT (30.3 g of CT/kg of dry matter), or alfalfa (AL) and concentrate to meet their predicted nutrient requirements. Each experimental period consisted of a 21-d adaptation period in a tiestall, followed by a 7-d collection period in metabolic crates, where feces and urine were collected quantitatively. During the 7-d period, milk yield was recorded daily and milk samples were taken at each milking. Blood, ruminal fluid, and papillae were sampled on d 2 and 5. The relative abundance of selected bacterial strains in ruminal fluid and the gene expression of transporter genes in the papillae were determined with quantitative PCR. Total volatile fatty acids and the abundance of the cellulolytic bacteria Prevotella spp. and Ruminococcus flavefaciens decreased with SF compared with AL. The relative gene expression of the monocarboxylate transporter 1 was increased with BT compared with AL and SF. Total yields of milk, milk fat, and milk protein were similar among treatments. The proportion of 18:3n-3 in milk fat was greater and those of 22:5n-3 and 22:6n-3 were lower with SF than with BT. The contents of urea N in blood (2.71, 3.45, and 3.90 mmol/L for SF, AL, and BT, respectively), milk (79.8, 100.1, and 110.9 mg/kg for SF, AL, and BT, respectively), and urine were lower with SF than with AL and BT, and a trend toward a lower ruminal ammonia content occurred with SF compared with BT. Intake and excretion of N with milk were similar among treatments, but urine N was lower with SF than with AL. The N excretion to N intake relation showed a shift in a part of urine N (17.5, 20.8, and 19.5% for SF, AL, and BT, respectively) to fecal N (45.2, 41.3, and 38.5% for SF, AL, and BT respectively) with SF compared with AL and BT. In conclusion, SF and BT differed in their effects on fermentation and milk fatty acid profile and SF also showed potential to decrease metabolic and environmental loads. Given its elevated content of carotenoids, alfalfa protein concentrates (APC) have the potential to prevent oxidation of milk enriched in polyunsaturated fatty acids. The effects of feeding APC or supplemental vitamin E on production, composition, and oxidative stability of milk enriched in polyunsaturated fatty acids were evaluated using 6 lactating Holstein cows (224±18d in milk) in a replicated 3×3 Latin square (21-d periods, 14d for adaptation). Treatment diets contained (dry matter basis) (1) 9% soybean meal (control, CTL); (2) 9% soybean meal + 300 IU of vitamin E/kg (VitE treatment); or (3) 9% APC (APC treatment). Cows received a continuous abomasal infusion of 450g/d of linseed oil. As a result, milk fat content of cis-9,cis-12 18:2 increased from 1.08±0.13 to 3.9±0.40% (mean ± SD), whereas cis-9,cis-12,cis-15 18:3 increased from 0.40±0.04 to 14.27±1.81% during the experimental period compared with the pretrial period. Milk yield tended to be higher for APC (14.7kg/d) compared with CTL (13.4kg/d), and was greater than that for VitE (13.0kg/d). Protein yield was higher in cows fed APC (518g/d) compared with VitE (445g/d) but was not different from that in cows fed CTL (483g/d). These effects resulted in improved milk N efficiency in cows fed APC (26.1% of N intake secreted in milk) compared with CTL (23.0%) and VitE (22.9%). Feeding APC increased milk fat content of lutein (252μg/g) compared with CTL (204μg/g) and VitE (190μg/g). Milk fat content of vitamin E was higher for APC (34.5μg/g) compared with CTL (19.0μg/g) and tended to be lower than that with VitE (44.9μg/g). Redox potential of fresh milk from cows fed APC (152mV) was similar to that of VitE (144mV), but lower than that of CTL (189mV). Treatments had no effect on fresh milk contents of dissolved oxygen (8.1±1.5mg/L), and conjugated diene hydroperoxides (2.7±0.5mmol/L). The concentrations of volatile lipid oxidation products (propanal, hexanal, hept-cis-4-enal, 1-octen-3-one) tended to be decreased by APC relative to CTL, whereas similar values were observed for VitE, except for hexanal, which was reduced by 40% in VitE. In conclusion, feeding APC to lactating dairy cows could serve as a source of dietary protein that improves dietary N utilization efficiency, and also as a preharvest technology to increase natural antioxidant levels in milk to limit oxidation.

The production of baled silage is attractive to producers because it offers advantages over dry hay, particularly by limiting risks associated with wet or unstable weather conditions. Our objectives were to test the effects of delayed wrapping on silage fermentation, storage characteristics, and the nutritive value of baled alfalfa silages. To accomplish this, large-round bales of alfalfa were wrapped in plastic film within 4h of baling (d 0), or after delays of 1, 2, or 3 d. A secondary objective was to evaluate a prototype bale wrap containing an O2-limiting barrier (OB) against an identical polyethylene wrap without the O2 barrier (SUN). Sixty-four 1.19×1.25-m bales of alfalfa were made from 4 field blocks at a mean moisture concentration of 59.1±4.3% with a mean initial wet bale weight of 473±26.4kg. Two bales per field block were assigned to each combination of bale wrap (SUN or OB) and wrapping time (0, 1, 2, or 3 d postbaling), and one bale of each pair was fitted with a thermocouple placed in the geometric center of each bale. All bales were sampled after a 97-d storage period. Internal bale temperatures, recorded at the time bales were wrapped, were greater for all bales with wrapping delays compared with bales wrapped on d 0 (54.9 vs. 34.9°C), and increased to a maximum of 63.9°C after a 3-d delay exhibiting a linear effect of time delay. Total silage fermentation acids (lactic, acetic, propionic, butyric, and isobutyric) were greatest when bales were wrapped on d 0 compared with all bales wrapped with time delays (4.64 vs. 2.26% of DM), and declined with linear and quadratic effects of wrapping delay. Total fermentation acids also were related quadratically to internal bale temperature by regression [y (% of DM)=0.0042x(2) - 0.50x + 17.1; R(2)=0.725]. Similar responses were observed for lactic acid, except that trends were linear, both for orthogonal contrasts evaluating length of wrapping delay, and in regressions on internal bale temperature [y (% of DM)=-0.046x + 3.5; R(2)=0.663]. Butyric acid also was detected, regardless of treatment, but was greatest within bales wrapped on d 0 compared with those with wrapping delays (0.99 vs. 0.38% of DM), and a similar response (0.68 vs. 0.52% of DM) was observed for NH3-N, suggesting that clostridial activity occurred during silage fermentation. Based on these results, silage fermentation characteristics and the nutritive value declined with time delays before wrapping, but responses were exacerbated when delays exceeded 1 d.

Nutritional status and glucose precursors are known regulators of gluconeogenic gene expression. Glycerol can replace corn in diets fed to dairy cows and use of glycerol is linked to increased rumen propionate production. The effect of dietary glycerol on the regulation of gluconeogenic enzymes is unknown. The objective of this study was to examine the effect of glycerol on expression of pyruvate carboxylase (PC), cytosolic and mitochondrial phosphoenolpyruvate carboxykinase (PEPCK-C and PEPCK-M), and glucose-6-phosphatase. Twenty-six multiparous Holstein cows were fed either a control diet or a diet where high-moisture corn was replaced by glycerol from -28 through +56 d relative to calving (DRTC). Liver tissue was collected via percutaneous liver biopsy at -28, -14, +1, +14, +28, and +56 DRTC for RNA analysis. Expression of PC mRNA increased 6-fold at +1 and 4-fold at +14 DRTC relative to precalving levels. Dietary glycerol did not alter expression of PC mRNA expression. Expression of PEPCK-C increased 2.5-fold at +14 and 3-fold at +28 DRTC compared with +1 DRTC. Overall, dietary glycerol increased PEPCK-C expression compared with that of cows fed control diets. The ratio of PC to PEPCK-C was increased 6.3-fold at +1 DRTC compared with precalving and tended to be decreased in cows fed glycerol. We detected no effect of diet or DRTC on PEPCK-M or glucose-6-phosphatase mRNA, and there were no interactions of dietary treatment and DRTC for any transcript measured. Substituting corn with glycerol increased the expression of PEPCK-C mRNA during transition to lactation and suggests that dietary energy source alters hepatic expression. The observed increase in PEPCK-C expression with glycerol feeding may indicate regulation of hepatic gene expression by changes in rumen propionate production.

Gravid heifers consuming high-quality forage diets are susceptible to excessive weight gains and overconditioning. One approach for controlling this problem is to dilute diets with low-energy forages, such as straw, that reduce the caloric density and dry matter intake (DMI) of that diet by heifers. These diluting agents are often sortable by dairy heifers, but previous visual evidence has suggested that eastern gamagrass haylage may be a nonsortable alternative. Our objectives were (1) to compare the growth performance of dairy heifers offered a high-quality forage diet (control) with diets containing 1 of 3 diluting agents [eastern gamagrass haylage (EGH), chopped wheat straw (WS), or chopped corn fodder (CF)]; and (2) evaluate sorting behaviors of heifers offered these forage diets. Holstein heifers (n=128) were stratified (32 heifers/block) on the basis of initial body weight (heavy, 560 ± 27.7 kg; medium-heavy, 481 ± 17.7 kg; medium-light, 441 ± 22.0 kg; and light, 399 ± 14.4 kg), and then assigned to 1 of 16 identical research pens (4 pens/block; 8 heifers/pen), where each of the 4 research diets were assigned to 1 pen within each block. Diets were offered in a 118-d feeding trial with heifers crowded to 133% of capacity at the feed bunk. Inclusion of low-energy forages was effective in reducing both diet energy density and DMI. Concentrations of physically effective fiber (pef) particles did not change during the 24-h period following feeding for either the control or EGH diets; however, this response for pef particles masked the competing (and cancelling) responses for individual large and medium particles, which heifers sorted with discrimination and preference, respectively. Sorting against pef particles was detected for WS, and much more severely for the CF diet. Sorting of forage particles by heifers could not be related to heifer performance. Compared with control (1.16 kg/d), average daily gains (ADG) were reduced by dilution in all cases, but were virtually identical between EGH (0.98 kg/d) and CF (0.97 kg/d), which exhibited no sorting and extensive sorting of pef, respectively. Furthermore, ADG for WS was approximately 0.2 kg/d less than EGH or CF, despite exhibiting sorting characteristics intermediate between EGH and CF. Diets diluted with low-energy forages were formulated to be isonitrogenous and isocaloric; within that context, WS was most effective in reducing DMI and maintaining ADG within typical recommendations for Holstein heifers.

This study aimed to evaluate intake, body growth, and the development of the rumen, mammary gland, and immune system in Holstein Friesian calves reared for 100 d on the commercially available feed FiberStart (conserved alfalfa, Medicago sativa; Fiber Fresh Feeds Ltd., Reporoa, New Zealand) and fed calf milk replacer (CMR) for either 56 or 91 d. Eighty calves (40 bulls and 40 heifer calves) were reared indoors in groups (n = 5 of the same sex/pen). All calves were fed 4 L of CMR/d (175 g/L of CMR) in 2 feeds/d for the first 10 d and then 1 feed/d until d 49 or 84. The calves were gradually weaned by d 56 (earlier weaned; n = 8 pens) and d 91 (later weaned; n = 8 pens). All calves were fed FiberStart ad libitum as the only solid feed source from d 1 to 100 of the study. Irrespective of treatment, all calves had similar body weights at d 0 (40.9 ± 3.0 kg) and d 49 (74.2 ± 5.1 kg) of the study. Calf sex had no effect on intake, growth, blood, and immune system parameters. Earlier-weaned calves consumed 18% more solid feed dry matter but had 16% lower body weight gain (28.9 vs. 38.5 kg, respectively) from d 56 to 100 relative to later-weaned calves, resulting in different body weight at 100 d (104 vs. 121 ± 1.3 kg). Although earlier-weaned calves could compensate for the loss of CMR dry matter and crude protein intake from d 56 to 100 by increasing forage intake, they were unable to compensate for the loss of energy from the CMR by increasing solid feed consumption. Plasma β-hydroxybutyrate concentrations were 52% greater in earlier-weaned calves than in later-weaned calves at d 84, indicating greater metabolic activity of the rumen wall. The duration of CMR feeding had no influence on humoral or cell-mediated immune functions of the calves, as evidenced by a lack of effect on antivaccine antibody responses as well as on immune gene expression. Earlier- versus later-weaned heifer calves had 5% lower mammary gland mass, indicating that greater energy supply increased mammary mass. The results of this experiment demonstrate the ability to artificially rear dairy calves on a conserved alfalfa as the only solid feed. Furthermore, earlier weaning off CMR promotes solid feed intake and an associated increase in blood β-hydroxybutyrate, an indicator of rumen development, but increasing the duration of CMR feeding improves growth and mammary gland mass by d 100. The implications of these findings on lifetime growth, health, and milk production in dairy heifers warrant further investigation.

Reducing enteric methane (CH4) production and improving feed conversion efficiency of dairy cows is of high importance. Residual feed intake (RFI) is one measure of feed efficiency, with low RFI animals being more efficient in feed conversion. Enteric CH4 is an important source of digestible energy loss in ruminants and, because research in beef cattle has reported a positive relationship between RFI and daily CH4 production, we hypothesized that low RFI dairy heifers, which are more feed efficient, would produce less CH4/d. We measured the daily methane production (g of CH4/d), methane yield [g of CH4/kg of dry matter intake (DMI)], and CH4 per kilogram of body weight (BW) gain for 56 heifers (20-22 mo old) in a 2 × 2 factorial arrangement: factors included 2 breeds (Holstein-Friesian and Jersey; n = 28/breed), with equal numbers of animals previously determined as being either high [+2.0 kg of dry matter (DM)/d] or low RFI (-2.1 kg of DM/d; n = 28/RFI category). All heifers were commingled and offered unrestricted access to the same diet of dried alfalfa cubes. Between RFI categories, heifers did not differ in BW or BW gain but low RFI heifers had 9.3 and 10.6% lower DMI and DMI/kg of BW, respectively, than high RFI heifers. Similarly, RFI category did not affect CH4/d or CH4/kg of BWg, but CH4/kg of DMI was higher in low RFI heifers because of their lower DMI. These results might reflect more complete digestion of ingested feed in the more efficient, low RFI heifers, consistent with previous reports of greater apparent digestibility of organic matter. Holstein-Friesian heifers were heavier and consumed more total DM than Jersey heifers, but breed did not affect DMI/kg of BW or BWg. Jersey heifers produced less CH4/d, but not CH4/kg of DMI or CH4/kg of BWg. We detected no interaction between breed and RFI category in any of the variables measured. In conclusion, differences in RFI in dairy heifers did not affect daily CH4 production (g/d); however, low RFI heifers had a greater CH4 yield (g/kg of DMI) on a high forage diet.

We investigated the effect of reconstitution of alfalfa hay on feeding and chewing behavior, sorting activity, and health status using 20 neonate Holstein male calves (3 d of age; 40.3 ± 1.30 kg of body weight) that were assigned randomly to 2 treatments, a starter feed containing either 10% dry (AH) or reconstituted alfalfa hay (RAH), each consisting of 10 calves. Alfalfa hay was reconstituted with water 24 h before feeding to achieve a theoretical dry mater (DM) content of 20%. Both starter diets had the same ingredients and nutrient compositions but differed in their DM content (90.9 vs. 66.6% DM for AH vs. RAH, respectively). Calves were weaned on d 50 and remained in the study until d 70. Reconstitution of alfalfa hay increased the percentage of particles retained on 8- and 1.18-mm sieves, but reduced feed materials retained on the bottom pan. Feeding RAH tended to increase meal frequency (preweaning) and increased meal duration (pre- and postweaning), and thereby increased time devoted to eating without affecting nutrient intake. Calves fed RAH tended to have lower eating rate during the pre- and postweaning periods. Tendencies to concurrent increase in rumination frequency and rumination duration in calves fed RAH increased time devoted to ruminating during the preweaning period; however, a tendency to longer rumination duration did not affect ruminating time during the postweaning period. Calves fed RAH spent less time resting and standing and more time drinking during the postweaning period. Non-nutritive oral behaviors were not affected by treatment during the pre- and postweaning period. Feeding RAH decreased sorting of particles retained on 8-mm sieve compared with AH; however, calves in both treatment groups sorted for particles retained on 8- and 1.18-mm sieves and against the feed materials retained on the bottom pan. Crude protein and neutral detergent fiber intakes of particles retained on the both 8- and 1.18-mm sieves increased in calves fed RAH versus AH, with no changes in intake of nutrients retained on the bottom pan. Calves fed AH were more susceptible to develop diarrhea (odds ratio = 2.02) and pneumonia (odds ratio = 4.74) and thereby had lower chances of being treated. We found no difference between treatment groups for frequency and medication days of diarrhea; however, calves fed RAH experienced fewer days with diarrhea. Furthermore, frequency and number of days with pneumonia and administration of medication were greater for calves fed AH compared with calves fed RAH. Blood concentrations of total protein and globulin tended to be higher in calves fed RAH compared with calves fed AH. Overall, feeding RAH increased time devoted to eating by increasing meal frequency and meal duration but did not affect feed intake due to lower eating rate. Calves in both treatment groups exhibited feed sorting at the extent to which they balanced intake of nutrients and met their nutritional needs. Calves, in general, were healthy, but it seems that feeding a diet containing reconstituted alfalfa hay has a potential health-promoting effect, possibly through reducing diet dustiness and improving immune status.

Gluconic acid is a carboxylic acid naturally occurring in plants and honey. In nonruminant animals, gluconic acid has been shown to increase gastrointestinal butyrate concentrations and improve growth performance, but a ruminant application remains undescribed. This experiment examined the effects of postruminal calcium gluconate (CaG) on milk production, fecal volatile fatty acid concentrations, and plasma metabolite concentrations in lactating dairy cows. Six rumen cannulated multiparous Holstein cows (60 ± 6 d in milk) were randomly assigned to 6 treatment sequences within a 6 × 6 Latin square design in which each experimental period consisted of 5 d of continuous postruminal infusion followed by a 2 d wash-out period. Test treatments included a negative control (CON; 0.90% NaCl wt/vol), positive control (Na-butyrate, 135 g/d), and 4 doses of CaG (44, 93, 140, and 187 g/d). Cows received a total mixed ration (31% corn silage, 28% alfalfa silage, 5% hay, 36% concentrate) with dry matter intake fixed (25.3 ± 1.7 kg/d) throughout the experiment. On d 5 of each infusion period, samples of milk, feces, and blood were collected from each animal. Calcium gluconate treatments increased milk fat concentration, and a tendency was observed for increased milk fat yield and energy-corrected milk yield above levels achieved by CON, with maximal treatment responses of 4.43% (CON 3.81%), 2.089 kg/d (CON 1.760 kg/d), and 51.8 kg/d (CON 47.1 kg/d), respectively. Concentrations of iso-butyric acid in feces were greater in cows infused with CaG (13.3 µmol/g) treatments compared with CON (9.7 µmol/g). Arterial concentrations of glucose and nonesterified fatty acids were lower (glucose: CaG 2.98 mmol/L, CON 3.29 mmol/L and nonesterified fatty acids: CaG 0.130 mmol/L vs. 0.148 mmol/L) and β-hydroxybutyrate higher (CaG 1.703 vs. CON 0.812) in cows infused with CaG than CON. Together, these results suggest that postruminal infusion of CaG may alter metabolic mechanisms to support a milk fat production response.
