Lactation physiology is a process that is only partly understood. Proteomics techniques have shown to be useful to help advance the knowledge on lactation physiology in human and rodent species but have not been used as major tools for dairy cows, except for mastitis. In this paper, advanced non-targeted proteomics techniques (Filter aided sample preparation and NanoLC-Orbitrap-MS/MS) were applied to study the milk fat globule membrane and milk serum fraction, resulting in the identification of 246 proteins. Of these, 23 transporters and enzymes were related to lipid synthesis and secretion in mammary gland and their functions are discussed in detail. The identification of these intracellular transporters and enzymes in milk provides a possibility of using milk itself to study lipid synthesis and secretion pathways. This full-scale scan of milk proteins by using non-targeted proteomic analysis helps to reveal the important proteins involved in lipid synthesis and secretion for further examination in targeted studies.

Emulsions were prepared with 20% soy oil and different concentrations of lactoferrin, and tested at pH values from 3 to 7·5. The stability of the emulsions decreased as the pH got closer to the isoelectric point of the protein. A concentration of 1% lactoferrin was determined to be sufficient to provide full coverage of the emulsion droplets. Lactoferrin-stabilized emulsions were then prepared in water at pH 6·6 and their behaviour when added to reconstituted milk was studied. It was observed that lactoferrin emulsions were stable when reconstituted in milk, but they showed aggregation when diluted in milk serum alone. The destabilization was caused by shielding of the charges on the surface of the oil droplets. Stabilization in milk occurred due to interactions at the interface with other soluble proteins. In fact, when β-lactoglobulin or sodium caseinate were added to the serum, stability of the emulsion droplets was restored, indicating that these proteins were able to adsorb at the interface and aid in the stabilization. Since ζ-potential measurements did not show significant overall charge on the emulsion droplets, this suggests that the stabilization forces are not only electrostatic in nature, but that there are other mechanisms at play.

The objective was to investigate the changes occurring in the activities of the enzymes lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and aspartate aminotransferase (AST) in sheep and goat milk as a result of subclinical intramammary infections (IMI) and to evaluate the use of these enzymes for the diagnosis of subclinical IMI in dairy sheep and goats. A total of 206 samples of sheep milk and 162 samples of goat milk, obtained from equal udder halves, were used in the study. For each species they were divided into two groups: a no-infection group and a subclinical infection group. Activities of LDH, ALP and AST were significantly higher in the subclinical infection group than in the no-infection group (Pv. 120·91±4·41; ALP: 2773·43±105·18 v. 2189±94·24; AST: 29·57±0·74 v. 17·32±0·46) and goats (LDH: 354·07±13·33 v. 103·79±3·75; ALP: 311·13±25·74 v. 137·24±19·62; AST: 27·59±6·42 v. 15·87±0·45). The activity of LDH was identified as indicator for subclinical IMI in both sheep and goats. The optimum cut-off values for LDH activity, offering the highest diagnostic sensitivity (DSn) and diagnostic specificity (DSp), determined by receiver operating characteristic (ROC) analysis, were at 197 U/l, 185 U/l and 197 U/l for sheep, goats and both species, respectively. DSn for sheep, goats and both species at these cut-off values was 92·8%, 98·2% and 94·0%, whereas DSp was 95·4%, 96·3% and 96·3%, respectively. It was concluded that the determination of LDH activity in milk serum is a sensitive and reliable method for the detection of subclinical IMI in dairy sheep and goats.

The objective of the present study was to determine the effect of high pressure (HP) processing (200, 450 and 650 MPa) at various temperatures (20, 40 and 55 degrees C) on the total plasmin plus plasminogen-derived activity (PL), plasminogen activator(s) (PA) and cathepsin D activities and on denaturation of major whey proteins in bovine milk. Data indicated that transfer of both PL and PA from the casein micelles to milk serum occurred at all pressures utilized at room temperature (20 degrees C). In addition to the transfer of PL and PA from micelles, there were reductions in activities of PL (16-18%) and PA (38-62%) for the pressures 450 and 650 MPa, at room temperature. There were synergistic negative effects between pressure and temperature on residual PL activity at 450 and 650 MPa and on residual PA activity only at 450 MPa. Cathepsin D activity in the acid whey from HP-treated milk was in general baroresistant at room temperature. The residual activity of cathepsin D decreased significantly at 650 MPa and 40 degrees C and at the pressures 450 and 650 MPa at 55 degrees C. Synergistic negative effects on the amount of native beta-lactoglobulin were observed at 450 and 650 MPa and on the amount of native alpha-lactalbumin at 650 MPa. There were significant correlations between enzymatic activities (PL, PA and cathepsin D) and the residual native beta-lactoglobulin and alpha-lactalbumin in bovine milk. In conclusion, HP significantly affected the activity of indigenous proteolytic enzymes and whey protein denaturation in bovine milk. Reduction in activity of indigenous enzymes (PL, PA and cathepsin D) and transfer of PL and PA from the casein to milk serum induced by HP is expected to have a profound effect on cheese yield, proteolysis during cheese ripening and quality of UHT milk during storage.

SUMMAARYThe content and composition of protein in milk samples from creameries in south-west Scotland were determined over a period of 12 months. The composition of the whole casein was expressed in terms of αsl-, β-, κ-, αs2- and γ-caseins, and that of the total milk serum protein in terms of β-lactoglobulins (β-lg), α-lactalbumins, bovine serum albumin, and a mixture of immunoglobulins, proteose-peptone component 3 and lactoferrin (IPL). Concentrations of the individual caseins varied appreciably and for most, concentration was closely correlated with and showed the same seasonal pattern as total casein concentration. Concentrations of the milk serum proteins also varied but only those of β-lg and the IPL fraction were closely correlated with that of total milk serum protein and seasonal trends were not marked. Relative amounts of the individual proteins, on the other hand, showed smaller variations and so throughout the experimental period the milks contained a protein complex of comparatively constant composition. Because of this comparative constancy it would appear that seasonal variations in milk properties in south-west Scotland are unlikely to be determined to a major extent by milk protein composition, but could be more affected by protein concentration.

SummaryThe strength of binding of the individual caseins and the nature of the bonding within bovine casein micelles were examined through dissociation of the micelles by dialysis of skim milk either against phosphate-free buffers containing 3 or 6 mm-CaCl2, or against buffers that were nearly saturated with respect to micellar calcium phosphate, but which had a free Ca2+ concentration in the range 0·4–5·9 mm. Dissociation was followed by ultracentrifuging the dialysed milks and determining the partition of the total and the individual caseins between the pellet and serum. During dialysis against the phosphate-free buffers both colloidal Ca and Pi in the milks decreased and about 30 % of the Pi could be removed without significant casein dissociation. With further loss of Pi, however, increasing dissociation occurred and the proportions of the individual caseins retained in the casein pellet were in the order αs2- > αs1- > β- ≈ κ-casein. Dialysis against the calcium phosphate buffers resulted in no loss of colloidal Pi but colloidal Ca increased with the free Ca2+ concentration of the buffer. Little change in the casein partition occurred in the presence of more than 1 mm free Ca2+, but serum casein increased markedly at lower levels, and the strength of binding of the individual caseins in the pelleted casein was in the order αs2-> αs1- > β- > κ-casein. In both types of buffer, dissociation is considered to occur through the breaking of linkages between the caseins and inorganic constituents. Analysis of the amino acids in a calcium phosphate-rich material obtained after exhaustive proteolytic digestion of casein micelles suggests that these linkages involve the phosphate centres of the caseins.

SummaryThree cationic materials markedly reduced the rennet clotting time of casein micelle suspensions, the efficacy of each being primarily dependent on the charge and the amount absorbed by the micelles. The reduction in coagulation time was unaffected by components of the milk serum other than salts. No enzymic action by lysozyme on casein micelles was detected. All materials acted by the same mechanism, increasing the affinity of rennet for the micelles and accelerating the aggregation phase. The results indicated that coagulation occurs by specific interactions between micelles modified by rennet.

Resonances from previously unknown constituents such as phosphocreatine were also observed. When the pH-dependence of inorganic phosphate,N-acetylglucosamine-1 -phosphate, glycerophosphorylcholine and gly-cerophosphorylethanolamine was examined it was observed that the resonance of inorganic phosphate overlapped that ofN-acetylglucosamine-1-phosphate around neutral pH. This is the most probable explanation as to why this constituent was not observed in earlier31P-NMR studies on milk.

SUMMARYCasein micelles fractionated on controlled pore glass (CPG-10/3000) were shown to be stable by recycling experiments. Only minor effects on the size distribution of the casein micelles were found after heating skim-milk to 100 °C for 10 min, or freeze-drying skim-milk at – 70 °C followed by resuspension in the synthetic milk serum of Jenness & Koops (1962). The heating caused some whey proteins (β-lactoglobulin) to enter the micelle fractions while the freeze-drying caused some of the largest micelles to disrupt. In colloidal calcium phosphate-free skim-milk prepared according to Pyne & McGann (1960) all the micelles appeared to dissociate into monomeric caseins.

SummaryBy resuspending casein micelles in whey and dialysate it is shown that the role of whey proteins in the ethanol (EtOH)-induced coagulation of skim-milk is minimal. Experiments involving the interchange of milk sera indicated that the position of the EtOH stability/pH profile along the pH axis was governed by the diffusible constituents of the milk serum phase. The identities of those serum components governing the shape and position of the EtOH stability/pH profile were investigated. The addition of Ca2+ caused a shift in the entire profile to higher pH. Reduction of the available Ca2+ by addition of EDTA (up to 5 ran) shifted the profile to lower pH. The addition of phosphate (up to 5 mM) or citrate (up to 1 mM) had no effect on the profile, though higher concentrations of citrate (up to 5 mMi) caused slight shifts to lower pH. When equimolar amounts of Ca and phosphate were added, the system showed a shift in profile approximately equivalent to that of the free Ca introduced. Increasing the ionic strength of a milk by the addition of NaCl did not shift the profile, but decreased the maximum EtOH stability of the high pH arm of the sigmoidal profile. The EtOH stability/pH profile retained the same sigmoidal shape in all cases.

SUMMARYThe effects of temperature, dilution, dialysis and the presence of heparin on the stability of lipoprotein lipase (LPL) in milk, skim-milk, milk serum and casein micelles were investigated. At 4 and 20 °C milk serum was the source of the least stable LPL and casein was that of the most stable. There was little difference between LPL stability in milk and skim-milk at these temperatures, or between serum and casein LPL at 50 °C. Heparin (5 µg/ml) increased stability although the effect was less for casein LPL than for serum LPL. A 40-fold dilution of serum LPL with either simulated milk ultrafiltrate (SMUF) or 0·01 M-Tris-Cl pH 8·3 increased the loss of serum LPL, but not of casein LPL. Dialysis of skim-milk against deionized water or SMUF increased stability at 4 or 20 °C but not at 37 °C. LPL activity was more stable in diluted samples of dialysed skim-milk than in diluted samples of the same milk which had not been dialysed. Dialysis against deionized water increased lipolysis but against SMUF it did not increase. Solutions prepared by dialysing water against some milks were found to inhibit lipolysis and this effect was overcome by heparin. The possibility that milk serum contains a factor which influences LPL stability is discussed.

SUMMARYHigh performance gel chromatography of the casein micelles disaggregated by 6 m-urea was carried out on a TSK-GEL G4000SW column using 6 M-urea synthetic milk serum as the effluent. The eluate was divided into two fractions. Fast eluted fraction 1 decreased from 67·5 to 57·3% on reduction of casein micelles and was not observed in reduced colloidal phosphate-free casein micelles. Fraction 1 from reduced casein micelles contained 1·7 times as much Ca and Pibound to casein as did whole casein micelles, while fraction 2 essentially contained only bound Ca. These facts indicated that fraction 1 of reduced casein micelles consisted of the casein aggregates cross-linked by colloidal Ca phosphate (CCP). Polyacrylamide-gel electrophoresis showed that fraction 1 of reduced casein micelles contained more αs1- and αs2-caseins and less β-casein than whole micellar casein. No κ-casein was detected in fraction 1 of reduced casein micelles. It is suggested that the ester phosphate groups of casein are the sites for interaction with CCP.

SummaryTwo possible hypotheses for the mechanism of milk clotting were tested. The results obtained constituted strong evidence against one and suggested that the second is improbable. Milk was separated into a 5-fold-concentrated casein micelle suspension and milk serum. Pre-renneting of the serum did not reduce the rate of clotting on subsequent addition to the micelle suspension whether or not the conditions were such that the para-κ-casein became extensively aggregated. Washing of casein micelles up to 3 times with milk dialysate at 23°C extracted very little casein from the micelles and did not increase the clotting time of micelles resuspended to about the same concentration as in milk. The results appear to constitute decisive evidence against the hypothesis of milk clotting proposed by Parry & Carroll (1969). S-carboxymethyl-κ-casein, S-carboxymethyl-κ-casein containing 2·5 dimethylaminonaphthalene sulphonyl residues per mole, and rennin-treated dimethylaminonaphthalene sulphonated-S-carboxymethyl-κ-casein all bound Ca to the same extent at 30°C and pH 6·5, over the range 0·5–15·3 mM-CaCl2. This adds support to existing evidence that milk clotting does not involve formation of Ca bridges between casein micelles.

SUMMARYIt has been shown that there is a time-dependent transfer of β-casein and the milk serine proteinase system from micelles to milk serum with change of temperature from 38 to 4 °C. It has been established that the γ-caseins can be formed by proteolytic degradation of β-casein. By a simple extraction technique, the very hydrophobic γ-casein fraction was separated from stored milks (26 and 4 °C) and estimated quantitatively. The results showed that the proteolytic degradation of β-casein is faster at 4 °C than at room temperature and this can be explained by the immobilization of enzyme and substrate at the micelle surface at higher temperatures (26 °C).The results indicate that irreversible changes during cooling for short periods do not cause problems in milk processing, but the formation of γ-caseins and phosphopeptides may influence the technological properties of raw milk stored for more than 48 h.
