Graphical abstract Abstract This investigation reports a novel nanoGold-core multifunctional dendrimer for pulsatile chemo-, photothermal- and photodynamic- therapy of rheumatoid arthritis (RA). Architecturally, the nanocomposites comprised of a nanoGold (Au) at the focal whose surface is functionalized by hydroxy-terminated thiolated-dendrons following Au-thiol bond formation to produce nanoGold-core multifunctional dendrimer (Au-DEN). The surface hydroxyl groups of Au-DEN were then conjugated with methotrexate (MTX; a disease-modifying first line anti-rheumatic drug; DMARD; 74.29 ± 0.48% loading) to form Au-DEN-MTX-NPs (Particle size: 100.15 ± 28.36 nm; poly dispersibility index, PDI: 0.39 ± 0.02; surface zeta potential, ζ: −22.45 ± 1.06 mV). MTX was strategically selected to serve as an anti-rheumatic DMARD as well as a targeting ligand to attain selective localization of the formulation in arthritic tissue via folate receptors upregulated on arthritic tissues. The docking study was performed to confirm the viable binding efficiency of MTX towards β-folate receptors that are overexpressed on arthritic tissues taking folic acid as a reference standard. The IR780, a NIR active bioactive was also loaded in Au-DEN-MTX NPs to offer photothermal benefit upon irradiation with NIR laser (wavelength: 808 nm). The hypothesis was tested by elucidation of in vitro drug release profile, photothermal activity, cellular uptake (Fluorescence and confocal laser scanning microscopy; CLSM), cell viability assay (MTT protocol) and Intracellular reactive oxygen species (ROS) generation in mouse macrophage RAW264.7 cells and Lipopolysaccharide (LPS) activated RAW264.7 cells. Furthermore, the hemolytic toxicity and stability studies were also investigated to determine the blood compatibility as well as ideal storage condition of NPs. The outcome of this investigations presents developed multifunctional targeted NPs to be potential therapeutics for the improved treatment of RA. Graphical abstract Abstract A series of small-sized (about 2.0 nm) gold nanoparticles (AuNPs) with apparent lattice fringes are synthesized by NaBH 4 reduction of HAuCl 4 in the presence of stabilizing linear or dendronized 1,2,3-triazolyl-containing ferrocenyl polymers. These AuNPs show high catalytic activity for biphasic 4-nitrophenol (4-NP) reduction to 4-aminophenol (4-AP) by NaBH 4. The lower generation G1 dendronized polymers (DPs) is a more efficient stabilizer for AuNP catalytic activity in the reduction of 4-NP than linear G0 polymers or bulky G2 DPs. The G1 DP-stabilized AuNPs are robust and easily recyclable, and the catalyst is recovered and reused at least twenty times with progressive smooth AuNP size increase along with some decrease of catalytic activity. Abstract: Fabrication of Langmuir films at the air–water interface of four linear-dendritic block copolymers (LDBCs) is described. The LDBCs are composed of a linear hydrophilic chain of poly(ethylene glycol) (PEG) and the first four generations of hydrophobic aliphatic polyester dendrons functionalized at the periphery with cyanoazobenzene chromophores. Langmuir films of the LDBCs, coded as PEG-AZOn (n indicates the number of cyanoazobenzene units at the periphery of the dendritic block), have been characterized by a combination of surface pressure versus area per molecule isotherms, UV–vis reflection spectroscopy and Brewster angle microscopy. The observed PEG-AZOn Langmuir film behavior depends strongly on the hydrophilic/hydrophobic ratio. A typical transition, related to PEG chains desorption from the air–water interface into the water subphase is observed for all the LDBCs, except for PEG-AZO16. In addition, PEG-AZO2 and PEG-AZO4 show a second transition whose nature has been studied in detail. Azobenzene chromophore interactions have been shown to be relevant in the organization of PEG-AZOn (n =4, 8 and 16) Langmuir films. Moreover, for PEG-AZO16 the orientation of the azobenzene units has been determined, revealing the formation of a well organized structure of azobenzene moieties at the air–water interface. Abstract: Since we observed that dendron-assembled surface provided high single nucleotide polymorphism discrimination efficiency for DNA microarrays, and that the binding yield for streptavidin increased when biotin was immobilized on top of it, the nanoscale-controlled surface is examined for surface plasmon field-enhanced fluorescence spectroscopy (or SPFS). Firstly, a silica film was coated onto a gold substrate using the sol–gel technique, followed by the covalent immobilization of a layer of second-generation dendrons with a DNA catcher strand at their apex. The thickness of the inorganic interlayer () was effectively suppressing fluorescence quenching. Thus, the kinetics and affinity characteristics of DNA hybridization could be investigated very sensitively by SPFS. The kinetic rate constants found for DNA hybridization on the dendron-modified surface were larger than those reported for a streptavidin-modified surface by one order of magnitude, except for dissociation rate constant for a single mismatched case. In addition, we observed that the DNA on the cone-shaped linker maintained its capability to capture DNA target strands even after extended storage at ambient conditions. [Copyright &y& Elsevier]

Abstract: TiO2 nanoparticles were synthesized at ∼0 °C by hydrolyzing [(CH3)2CHO]4Ti in 1-propanol solutions of poly(amido amine) dendrons with a siloxy focal point and long alkyl chain spacers. Transmission electron microscopic photographs showed that TiO2 nanoparticle was 1–5 nm in size and protected by dendrons, when prepared at a mixing ratio 1:10 of Ti ion and dendron. At higher contents of Ti ion, TiO2 nanoparticles aggregated up to a maximum size of 90 nm, depending on the dendron generation (first to third). It was confirmed from X-ray photoelectron spectroscopy that Sih name="sbnd" />Ti covalent bond was formed in dendron-protected TiO2 nanoparticles. The ability of dendron-protected TiO2 nanoparticles as a photocatalyst for the photodegradation of 2,4-dichlorophenoxyacetic acid was higher than that of nonprotected nanoparticle and superior at higher generation. It was suggested that the dendrons protecting TiO2 nanoparticle have enough void volume to conserve guest molecules and behave effectively as a reservoir of guest molecules. [Copyright &y& Elsevier]

Interactions between poly(amidoamine) dendrimer (PAMAM)–gold nanocomposites and alkanethiols and between the former nanocomposites and thiol-modified poly(amidoamine) dendrons in ethyl acetate were investigated by adding alkanethiols, such as 1-propanethiol and 1,3-propanedithiol, and thiol-modified poly(amidoamine) dendrons, generations 0.5 and 2.5 (G0.5–SH and G2.5–SH). The PAMAM dendrimers with surface methyl ester groups used were generations 1.5 and 5.5 (G1.5 and G5.5). The mean particle sizes of PAMAM–gold nanocomposites were about 2.1 for G1.5 and 2.4 nm for G5.5. In both nanocomposite systems where 1-propanethiol and 1,3-propanedithiol were added, the mean particle size was about 4 nm, twice that of the systems where these thiols were not added. Increasing the addition of 1,3-propanedithiol made the average particle size smaller for both nanocomposites systems. To compare with alkanethiol, thiol-modified poly(amidoamine) dendron with a highly branched structure on one side was synthesized. Using G2.5–SH as a protective agent, dendron–gold nanocomposites with mean diameters of 3 to 4 nm were obtained. The difference in particle size was seen only when the combination of PAMAM–gold nanocomposites and thiol-modified dendron was less sterically dense, modified dendron (G0.5–SH). The mechanisms for morphology changes in the dendrimer–gold nanocomposites by the addition of these thiols are discussed. [Copyright &y& Elsevier]
