Supercritical fluid chromatography (SFC) holds the potential to become an orthogonal method to HPLC/UHPLC in xenobiotic metabolism studies, due to its outstanding capacity to simultaneously separate highly similar (as HPLC) and physicochemically different analytes (problematic using HPLC). Paucity of guideline-conform validation, however, has been a major obstacle to clinical application of SFC, even in cases where biotransformation yields chemically dissimilar metabolites that require more than one HPLC method for comprehensive analysis. Here, a method based on supercritical fluid chromatography coupled to single quadrupole MS detection was developed to simultaneously quantify the divisive analgesic flupirtine and its acidic and basic metabolites, represented by 4-fluorohippuric acid (4-FHA) and the active metabolite D-13223 respectively, using custom-made synthetic internal standards. Experimental data on the fundamental retention mechanisms under supercritical conditions, indicating the importance of halogen and π-π-bonding for specific retention on polysaccharide-based stationary-phases, is discussed. Compared to previous HPLC methods, the novel method offers higher versatility in terms of the target metabolite range (addressing both acidic and basic metabolites within a singular method), faster analysis (7.5 min), and compliance with green chemistry principles. Validation was performed according to EMA criteria on bioanalytical method validation, demonstrating selectivity, carry-over, calibration curve parameters (LLOQ, range, and linearity), within- and between-run accuracy and precision, dilution integrity, matrix effect and stability. For proof-of-concept, the SFC method was applied to clinical samples of human urine obtained after single intravenous (100 mg), single oral (100 mg), and repeated oral administration (400 mg). Flupirtine, D-13223, and 4-FHA could be quantified, shedding light on the extent of oxidative flupirtine metabolism in humans in the context of the unresolved biotoxification that has led to the withdrawal of specific neuronal KV7 openers.

Ethyleneamines have been produced and commercialized for decades in the chemical industry for a diverse range of applications. The presence of amine functional groups provides them opportunity to adsorb onto surfaces which can make them a very challenging sample matrix to analyze using separation techniques. In the present report, a new aqueous SEC-RI method, which enables MWD characterization of higher ethyleneamines, is described. The sample preparation was based on the dilute-and-shoot methodology. A surface-modified SEC column with positively charged groups attached to the stationary phase was used. The mobile phase composition (salt concentration, pH) was optimized to suppress interaction between the ethyleneamines and the packing material. Very symmetrical peak shapes were achieved for low MW monodisperse ethyleneamines despite their high primary amine content. MWD calculations were conducted using conventional narrow standard calibration with partial linear extrapolation of the calibration curve. The narrow standards were of the same chemistry as the samples of interest. Consequently, the standard components display a consistent behaviour towards the column packing as the sample components which makes the present method more robust and the interpretation of the quantitative results more convenient. A rapid and quantitative analytical method for volatile chlorinated hydrocarbons and benzene, such as chloroethylene, dichloromethane, and carbon tetrachloride, in soil samples was developed using a needle-type extraction device packed with activated carbon-based adsorbents. The soil sample of 10 g was placed in a glass cartridge and fixed with quartz wool to make sure not to move during the extraction process. After the internal standard solution was spiked into the soil sample, the cartridge was sealed with a polytetrafluoroethylene adapter, and the needle extraction device was used to collect the headspace gas. Analytes were purged from the soil sample and collected on the adsorbent, while pure nitrogen gas was introduced to the soil sample during the sample collection. The sampling time was approximately 10 min for 100 mL of gas sampling. After the sampling process, the extraction needle was directly inserted into a heated injection port of gas chromatography equipped with flame ionization detector (GC-FID) or that hyphenated to mass spectrometry (GC-MS). The extracted VOCs were thermally desorbed and simultaneously introduced to a GC capillary column for subsequent separation and determination. The limit of detection and limit of quantification of all the examined analytes was less than 20 and 60 ng/10 g soil in GC-FID and less than 2.5 and 7.5 ng/10 g soil in the case of GC-MS. Linear calibration curves were obtained up to 10 μg/10 g soil for all the analytes. The relative standard deviations of the ratio of analyte peak area and internal standard peak area were less than 10%. The recovery and sensitivity of analytes confirmed in the proposed method were compared with that obtained with a standard method conventionally employed.

The measurement of VOSCs in complex matrices is challenging due to their volatile and reactive nature. A straightforward method using headspace chromatography was developed for routine analyses of organosulfur compounds in a high saline liquid matrix with a pH of 8.4. In this work, a simple and effective strategy for the determination of 12 active compounds of Atractylodes macrocephala Koidz. (AM) was proposed by using high performance liquid chromatography-diode array detection (HPLC-DAD) combined with alternating trilinear decomposition (ATLD) algorithm. Utilizing the "second-order advantage", three common problems in HPLC could be resolved, namely baseline drifts, peak overlaps, and unknown interferences. 12 compounds were rapidly eluted within 12.5 min, and the average spiked recoveries were 80.8-109.9%. The figures of merit reflected the feasibility of the proposed method. Compared with the results of the traditional univariate calibration method based on HPLC-UV technique, the proposed strategy further verified the reliability and simplicity of the mathematical separation. On this basis, partial least squares-discriminant analysis (PLS-DA) was applied to discriminate 113 AM samples from different geographical origins, and variable importance in projection (VIP) was used to further screen the main differential components that affect the regional division of AM. A series of results show that the AM samples from the three regions have obviously different clustering trends. Overall, the strategy is expected to provide a scientific basis for the modern research of medicinal materials, and it is also conducive to the clinical use and market supervision of AM.

Evodiae Fructus is used as a traditional Chinese medicine for the treatment of several kinds of diseases with its bioactive constituents. In this study, a capillary electrochromatography-mass spectrometry (CEC-MS) method was developed to determine three bioactive compounds including evodiamine, rutaecarpine and limonin in Evodiae Fructus fruit. Home-developed monolithic columns with methyl-vinylimidazole functionalized organic polymer monolilth as stationary phases were used in CEC-MS with excellent separation selectivity and high efficiency. The CEC-MS methods provided 4–16 folds improvement of LODs when compared with CEC-UV method. The conditions, which could affect separation efficiency and detection sensitivity, were optimized. Under optimum conditions, baseline separation with high detection sensitivity was obtained. A comprehensive Collision Cross Section (CCS) library was obtained via Travelling Wave Ion Guide mobility measurements through direct infusion (DI). The library consists of CCS and Mass Spectral (MS) data in negative and positive ElectroSpray Ionisation (ESI) mode for 463 and 479 endogenous metabolites, respectively. For both ionisation modes combined, TWCCSN2 data were obtained for 542 non-redundant metabolites. These data were acquired on two different ion mobility enabled orthogonal acceleration QToF MS systems in two different laboratories, with the majority of the resulting TWCCSN2 values (from detected compounds) found to be within 1% of one another. Validation of these results against two independent, external TWCCSN2 data sources and predicted TWCCSN2 values indicated to be within 1–2% of these other values. The same metabolites were then analysed using a rapid reversed-phase ultra (high) performance liquid chromatographic (U(H)PLC) separation combined with IM and MS (IM-MS) thus providing retention time (tr), m/z and TWCCSN2 values (with the latter compared with the DI-IM-MS data). Analytes for which TWCCSN2 values were obtained by U(H)PLC-IM-MS showed good agreement with the results obtained from DI-IM-MS. The repeatability of the TWCCSN2 values obtained for these metabolites on the different ion mobility QToF systems, using either DI or LC, encouraged the further evaluation of the U(H)PLC-IM-MS approach via the analysis of samples of rat urine, from control and methotrexate-treated animals, in order to assess the potential of the approach for metabolite identification and profiling in metabolic phenotyping studies. Based on the database derived from the standards 63 metabolites were identified in rat urine, using positive ESI, based on the combination of tr, TWCCSN2 and MS data.

Specifying effective analytical techniques and methods can be very challenging when a large number of sample streams need to be analyzed with high frequency. Combining those requirements with the need to monitor multiple components over a wide range of concentrations makes at-line analytical approaches preferred by many analytical scientists. Traditionally, at-line analytical support requires sample preparation and operator time. Recently developed technologies enabled automated sample preparation tools coupled with gas chromatography; thus, eliminating sample preparation steps and increasing productivity. Recently a commercial micro-reactor was introduced that can be combined with a flame ionization detector, providing the ability to quantify components without the need to perform a standard calibration, which saves significant time and materials. In this manuscript, we describe a unique analytical capability that combines automated sample preparation and gas chromatography with flame ionization detection and universal carbon response to provide high flexibility and accuracy when there is minimal information about the unknowns or reference materials are not available. Some of the challenges and performance monitoring techniques for this technology are also discussed in this manuscript.

The coated direct inlet probe (CDIP) is a new laboratory-made low-cost technology developed from a direct inlet probe (DIP), which has the advantage of quick enrichment/cleanup of an analyte from liquid samples. A capillary probe is coated with hydroxy-terminated polydimethylsiloxane (OH-PDMS), divinylbenzene (DVB), and β-cyclodextrin (β-CD) by a sol-gel method. This probe can be directly coupled with a commercialized atmospheric-pressure chemical ionization (APCI) ion source and high-resolution mass spectrometry, which are widely applicable, reliable, and durable. The ability to perform quantitative analyses with the use of a stable-isotope-labeled internal standard (SIL-IS) was tested by using different concentrations of acenaphthylene (ACY), acenaphthene (ACP), fluorene (FLR), fluoranthene (FLT), phenanthrene (PHE), and benzo[a]pyrene (B[a]P). Calibration curves with a coefficient of determination of R2 ≥ 0.9982 for different polycyclic aromatic hydrocarbons (PAHs) were obtained. A limit of detection (LOD) of 0.008–0.04 ng mL−1 for PAHs was determined. The entire workflow is solvent-free and can be completed in less than 5 min, which demonstrates the advantages of this technique for quantitative analysis.

The development of fast and precise measurement techniques for process analytical technology is important to operate chemical processes safely and efficiently. For quantitative measurements of multiple components at a trace level, often gas chromatographic methods are used which have a response time of several minutes or of up to one hour. For fast changing processes, this can be too slow for efficient control. For reducing the dead time of a control loop by increasing the measurement frequency, a multiplexing gas chromatography (mpGC) technique for a chromatographic system exhibiting a systematic non-linear response has been developed. For mpGC, superimposed chromatograms are measured by injecting consecutive samples before all components of previous samples have eluted from the column. The deconvolution of a superimposed chromatogram yields a computed chromatogram which is an average over the single chromatograms forming the superimposed chromatogram. Such a computed chromatogram typically shows so called correlation noise depending on the degree by which the single chromatograms forming the superimposed chromatogram will differ from each other (non-linear response). A technique is presented to calibrate the convolution matrix in order to suppress correlation noise introduced by systematic errors of the chromatographic system. The remaining correlation noise in the computed chromatogram is then exclusively caused by changing concentrations in the sample stream. For the method presented here, the sample is injected five times during the run time of a single chromatogram. The computed chromatogram is obtained three times within this timespan while representing each time an averaged chromatogram over the last five injections. Therefore, the sample throughput is increased by a factor of three compared to conventional GC.

Mechanistic modeling of chromatography has been around in academia for decades and has gained increased support in pharmaceutical companies in recent years. Despite the large number of published successful applications, process development in the pharmaceutical industry today still does not fully benefit from a systematic mechanistic model-based approach. The hesitation on the part of industry to systematically apply mechanistic models can often be attributed to the absence of a general approach for determining if a model is qualified to support decision making in process development. In this work a Bayesian framework for the calibration and quality assessment of mechanistic chromatography models is introduced. Bayesian Markov Chain Monte Carlo is used to assess parameter uncertainty by generating samples from the parameter posterior distribution. Once the parameter posterior distribution has been estimated, it can be used to propagate the parameter uncertainty to model predictions, allowing a prediction-based uncertainty assessment of the model. The benefit of this uncertainty assessment is demonstrated using the example of a mechanistic model describing the separation of an antibody from its impurities on a strong cation exchanger. The mechanistic model was calibrated at moderate column load density and used to make extrapolations at high load conditions. Using the Bayesian framework, it could be shown that despite significant parameter uncertainty, the model can extrapolate beyond observed process conditions with high accuracy and is qualified to support process development.

The quantification of preservatives in cosmetics has attracted great attentions for their controversial and widespread use. HPLC is a prevailing method for preservatives determination among various analytical methods. However, it takes long time to fully separate these compounds because of the complexity of cosmetic matrices. In this study, a fast and green HPLC-DAD strategy assisted with second-order multivariate calibration methods based on alternating trilinear decomposition (ATLD) and multivariate curve resolution-alternating least squares (MCR-ALS) was developed for the simultaneous determination of eight selected preservatives in complex facial mask samples. This appealing strategy proved to be a useful tool for eliminating unknown interferences in complex matrices without complete separation, which benefited from the “second-order advantages” and thus made the determination of the eight analytes in facial mask samples shorten to 8.2 min under a fast elution program. In particular, for the first time, we focused on the applicability of ATLD method for modeling of HPLC-DAD data with severe signal overlapping and slight time shifts. The spiked recovery values were in the range of 71.4–124.6%, and the RMSEP and REP values ranged from 0.07 to 2.4 μg mL−1 and 1.3–14.5%, respectively, indicating that the ATLD method could provide satisfactory prediction. The resolved spectral profiles and concentration values were compared with those obtained by the MCR-ALS method, an excellent tool for modeling of data deviating from trilinearity. Both qualitative and quantitative results from the two methods were consistent with each other, which evidenced the competence of ATLD method in handling HPLC-DAD data with severe signal overlapping and slight time shifts.

[EN] The quantification of the enantiomeric fraction (EF) during the biodegradation process is essential for environmental risk assessment. In this paper the enantioselective biodegradation of ibuprofen, IBU, and ketoprofen, KET, two of the drugs most consumed, was evaluated. Biodegradation experiments were performed in batch mode using a minimal salts medium inoculated with an activated sludge (collected from a Valencian Waste Water Treatment Plant) and supplemented with the racemate of each compound. The inoculum activity was verified using fluoxetine as reference compound. The experimental conditions used (analyte concentration and volume of inoculum) were chosen according to OECD guidelines. In parallel, the optical density at 600 nm was measured to control the biomass growth and to connect it with enantioselectivity. Two RPLC methods for chiral separations of IBU and KET using polysaccharides-based stationary phases were developed. Novel calculations and adapted models, using directly the chromatographic peak areas as dependent variable, were proposed to estimate significant parameters related to the biodegradation process: biodegradation (BD) and EF values at given time, half-life times of (R)- and (S)-enantiomers, number of days to reach a complete BD and the minimum EF expected. The modelled BD and EF curves fitted adequately the data (R-2 > 0.94). The use of these new equations provided similar results to those obtained using concentration data. However, the use of chromatographic peak areas data, eliminates the uncertainty associated to the use of the calibration curves. The results obtained in this paper indicate that an enantiorecognition towards 1BU enantiomers by the microorganisms present in the activated sludge used in this study occurred, being the biodegradation of (R)-IBU higher than that of (S)-IBU. The present study describes a rapid and effective capillary electrophoresis (CE) method for the enantioseparation of pindolol using single-isomer octa(6-O-sulfo)-γ-cyclodextrin. The complexation parameters were determined under neutral and high pH conditions to identify optimal separation conditions using a theoretical model. Baseline separation of pindolol enantiomers was achieved within 6 min in a sodium/MOPS buffer, pH 7.2, with a selector concentration of 6 mM. The method was validated according to the ICH guidelines using imidazole as an internal standard. Low limits of detection and quantification were found, specifically 1.2 μ g / m L and 4 μ g / m L (0.6 μ g / m L and 2 μ g / m L per enantiomer), respectively. The calibration curves showed good linearity, with a coefficient of determination R 2 ≥ 0.999 over a 5 – 55 μ g / m L concentration range and over a 50 – 300 μ g / m L concentration range of the racemic mixture. The relative standard deviations (%RSD) of intra-day and inter-day precision were lower than 8% at LOQ level, lower than 3% at 50 μ g / m L level and lower than 1.5% at 300 μ g / m L level. Accuracy ranged from 95 to 103% (106% at LOQ level). The proposed method was successfully tested on a medical formulation of Visken® Sandoz intravenous solution and Visken® Teofarma pills for oral use.

An efficient mathematical tool for the design and scaling up of protein chromatography is suggested, in which the model parameters can be determined quickly over a wide operating space without large material investments. The design method is based on mathematical modelling of column dynamics and moment analysis. The accuracy of the dynamic models that are most frequently used for simulations of chromatographic processes is analyzed, and possible errors that can be generated using the moment analysis are indicated. The so-called transport dispersive model was eventually employed for the process simulations. The model was modified to account for the protein dispersion in void volumes of chromatographic systems. The manner of the model calibration was suggested, which was based on a few chromatographic runs and verified over a wide space of the operating parameters, including composition and flow rate of the mobile phase, column dimensions, residence time, and mass loading. The model system for the study was ion-exchange chromatography. The analysis was performed based on the elution profiles of basic fibroblast growth factor 2 and lysozyme, on two different IEX media.

Oligo- and polysaccharides, especially fructooligosaccharides and inulin, are increasingly used as food ingredients. They are reported to stimulate the growth of bifidobacteria and are therefore used as prebiotics. As mono-, di-, oligo- and polysaccharides can together be present in food, an HPTLC method was developed for analyzing all in a single development. This method was used for both, effective food screening and quantification on the same plate. After a minimal sample preparation (aqueous extraction, dilution and filtration) and application, the samples were separated on an HPTLC plate silica gel 60 with acetonitrile - water 4:1 (V/V), containing 3.6 mM natural product reagent A. For a good reproducibility of the separation, the control of the layer activity was recommended. For up to 20 samples in parallel, the HPTLC analysis took 45 min (2.3 min/sample) with running costs of ca. 7 Euro (0.35 Euro/sample). Determination coefficients of the calibration curves were obtained between 0.9980 and 0.9998. The high degree of automation proved the method to be robust and suited for routine analysis, especially in food control. The newly developed method was successfully tested with 4 different prebiotic food and 60 naturally degraded inulin samples. A minimal sample preparation along with the determination of the intact inulin and FOS allowed the evaluation of the natural inulin degradation profile, as shown for naturally degraded inulin samples. This fast analysis might also be of interest in other fields that study, for example, plant breeding, edible insects, functional feed and metabolic processes.

This study describes the use of a magnetic multi-walled carbon nanotube composite for the preconcentration of nine parabens (methyl, ethyl, propyl, isopropyl, butyl, isobutyl, pentyl, phenyl and benzylparaben) from water and urine samples after in situ acetylation. The enriched extracts were obtained by dispersive magnetic solid phase extraction (DMSPE) and analysed by gas chromatography with mass spectrometry (GC-MS). Several parameters affecting the adsorption (extraction time, magnetic material mass and ionic strength) and desorption (desorption solvent nature and volume, and desorption time) DMSPE steps were investigated. Matrix-matched calibration was recommended for quantification of the samples. Linearities in the 0.5-150 ng mL-1 range were obtained, depending on the compound. Under the optimal experimental conditions, the limits of detection ranged between 0.03 and 2.0 ng mL-1, depending on the sample matrix and the paraben congener. None of the samples analysed contained the above mentioned parabens. The method was validated with two water samples (harbour and wastewater treatment plant) and two urine samples spiked at two concentration levels. Recoveries in the 81-119% range were obtained.

Nitrobenzene, a potentially harmful compound found in tobacco smoke, has been largely excluded from prior analysis due to difficulties with quantification. Quantifying harmful compounds in cigarette smoke is useful to compare products, to examine the impact of design parameters on delivery, and to help estimate exposures. A sensitive high-throughput method has been developed for quantifying nitrobenzene in machine-generated mainstream cigarette smoke using isotope dilution gas chromatography-tandem mass spectrometry (ID-GC-MS/MS). This method has sufficient sensitivity to measure vapor phase nitrobenzene concentrations in the low nanogram range, with a 418 pg/cig method limit of detection. Precision estimates from two quality control cigarette products resulted in percent relative standard deviations of 11.5% and 14.9%; product variability estimates from 13 cigarette products resulted in percent relative standard deviations ranging from 2.8% to 16.9%. Nitrobenzene in the machine-generated, mainstream smoke from 15 cigarette products are reported and range from 18 to 38 ng/cig under the Health Canada Intense smoking regimen.

With mass spectrometric detection in liquid chromatography, co-eluting impurities affect the analyte response due to ion suppression/enhancement. Internal standard calibration method, using co-eluting stable isotope labelled analogue of each analyte as the internal standard, is the most appropriate technique available to correct for these matrix effects. However, this technique is not without drawbacks, proved to be expensive because separate internal standard for each analyte is required, and the labelled compounds are expensive or require synthesising. Traditionally, standard addition method has been used to overcome the matrix effects in atomic spectroscopy and was a well-established method. This paper proposes the same for mass spectrometric detection, and demonstrates that the results are comparable to those with the internal standard method using labelled analogues, for vitamin D assay. As conventional standard addition procedure does not address procedural errors, we propose the inclusion of an additional internal standard (not co-eluting). Recoveries determined on human serum samples show that the proposed method of standard addition yields more accurate results than the internal standardisation using stable isotope labelled analogues. The precision of the proposed method of standard addition is superior to the conventional standard addition method.

The size-exclusion separation of a water-soluble polyelectrolyte polymer, sodium polystyrene sulfonate (NaPSS), was demonstrated on common reversed-phase (C(18,) C(4), phenyl, and cyano) and hydrophilic interaction chromatography (HILIC) columns. The effect of common solvents – acetonitrile (ACN), tetrahydrofuran (THF), and methanol (MeOH), used as mobile phase modifiers – on the elution of NaPSS and the effect of column temperature (within a relatively narrow range corresponding to typical chromatographic conditions, i.e., 10 °C–60 °C) on the partition coefficient, K(SEC), were also investigated. Non-size-exclusion chromatography (non-SEC) effects can be minimized by the addition of an electrolyte and an organic modifier to the mobile phase, and by increasing the column temperature (e.g., to 50 °C or 60 °C). Strong solvents such as THF and ACN are more successful in the reduction of such effects than is the weaker solvent MeOH. The best performance is seen on medium polarity and polar stationary phases, such as cyanopropyl- and diol-modified silica (HILIC), where the elution of the NaPSS polyelectrolyte is by a near-ideal SEC mechanism. Hydrophobic stationary phases, such as C(18), C(4), and phenyl, require a higher concentration of a strong solvent modifier (THF) in the mobile phase to reduce non-SEC interactions of the solute with the stationary phase.

Quantitative High Performance Thin Layer Chromatography (HPTLC) requires the application of several standards to each plate, reducing the number of actual samples that can be analyzed in a single run. Using pure standard compounds and a selective detection method, the standards for quantitation can be applied besides – a cote – the chromatography area. This frees the sample application space to accommodate the maximal number of sample on each plate. Also, analysis time is spent exclusively on samples, drastically shortening the effective analysis time per sample and increasing sample throughput. Using this new calibration approach, the sample capacity of regular HPTLC methods can be increased or their scope be extended by an additional quantitative analysis. As a limitation, changes to the distribution of samples and standards within the plate as well as interferences from matrix compounds must be observed. We demonstrate the feasibility of this method by complementing an HPTLC method with a quantitative analysis of total anthocyanin content in colored wheat varieties. The quantitation was validated and compared to the conventional photometric analysis. As outcome, the additional photometric analysis could be replaced and rendered unnecessary, saving time, effort and equipment. This approach could also be employed to quantify highly retained substances, which are usually inaccessible for quantitative analysis.

In this report, we put forward an experimental method to determine the ionic strength of an aqueous solution. To this end, we have developed a theory of ionic strength I expressed in terms of the retention ratios in field-flow fractionation (FFF) as I 1 / 2 = κ ( 1 − R ) / ( 1 − R o ) − e − 1 / 2 . Here R is a measured retention ratio using an FFF technique, for instance, sedimentation FFF (SdFFF), and Ro is the sterically-corrected standard retention ratio as given by the standard retention theory (SRT) for a latex particle system of diameter d . For a standard latex system with known d (or Ro) and I, we can construct a linear calibration of I 1 / 2 against ( 1 − R ) / ( 1 − R o ) . Therefore, if we measure the retention ratio R of a carrier liquid of which ionic strength is of interest, then we will be able to estimate the ionic strength from the calibration curve thus built. In this paper, we have demonstrated the relation of I 1 / 2 with respect to ( 1 − R ) / ( 1 − R o ) for the polystyrene latex systems of which information on R, Ro, and I is available from Ref. [1] .

In this study, a QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) method, optimized by a 23 full factorial design, was developed for the determination of 72 pesticides in plant parts of carrot, corn, melon, rice, soy, silage, tobacco, cassava, lettuce and wheat by ultra-high-performance liquid chromatographic tandem mass spectrometry (UHPLC-MS/MS). Considering the complexity of these matrices and the need of use calibration in matrix, a new calibration approach based on single level standard addition in the sample (SLSAS) was proposed in this work and compared with the matrix-matched calibration (MMC), the procedural standard calibration (PSC) and the diluted standard addition calibration (DSAC). All approaches presented satisfactory validation parameters with recoveries from 70 to 120% and relative standard deviations≤20%. SLSAS was the most practical from the evaluated approaches and proved to be an effective way of calibration. Method limit of detection were between 4.8 and 48μgkg-1 and limit of quantification were from 16 to 160μgkg-1. Method application to different kinds of plants found residues of 20 pesticides that were quantified with z-scores values≤2 in comparison with other calibration approaches. The proposed QuEChERS method combined with UHPLC-MS/MS analysis and using an easy and effective calibration procedure presented satisfactory results for pesticide residues determination in different crop plants and is a good alternative for routine analysis.

The objective of the present work is development of joint approximate diagonalization of eigenmatrices (JADE) as a member of independent component analysis (ICA) family, for the analysis of gas chromatography-mass spectrometry (GC-MS) and comprehensive two-dimensional gas chromatography-mass spectrometry (GC×GC-MS) data to address incomplete separation problem occurred during the analysis of complex sample matrices. In this regard, simulated GC-MS and GC×GC-MS data sets with different number of components, different degree of overlap and noise were evaluated. In the case of simultaneous analysis of multiple samples, column-wise augmentation for GC-MS and column-wise super-augmentation for GC×GC-MS was used before JADE analysis. The performance of JADE was evaluated in terms of statistical parameters of lack of fit (LOF), mutual information (MI) and Amari index as well as analytical figures of merit (AFOMs) obtained from calibration curves. In addition, the area of feasible solutions (AFSs) was calculated by two different approaches of MCR-BANDs and polygon inflation algorithm (FACPACK). Furthermore, JADE performance was compared with multivariate curve resolution-alternating least squares (MCR-ALS) and other ICA algorithms of mean-field ICA (MFICA) and mutual information least dependent component analysis (MILCA). In all cases, JADE could successfully resolve the elution and spectral profiles in GC-MS and GC×GC-MS data with acceptable statistical and calibration parameters and their solutions were in AFSs. To check the applicability of JADE in real cases, JADE was used for resolution and quantification of phenanthrene and anthracene in aromatic fraction of heavy fuel oil (HFO) analyzed by GC×GC-MS. Surprisingly, pure elution and spectral profiles of target compounds were properly resolved in the presence of baseline and interferences using JADE. Once more, the performance of JADE was compared with MCR-ALS in real case. On this matter, the mutual information (MI) values were 1.01 and 1.13 for resolved profiles by JADE and MCR-ALS, respectively. In addition, LOD values (μg/mL) were respectively 1.36 and 1.24 for phenanthrene and 1.26 and 1.09 for anthracene using MCR-ALS and JADE which showed outperformance of JADE over MCR-ALS.

Matrix effects are probably the Achilles heel of most quantitative liquid chromatography mass spectrometry (LC-MS) methods based on electrospray ionization. This work reports the evaluation of matrix effects in challenging matrices such as food extracts, human urine or wastewater at different dilution factors using nanoflow liquid chromatography-high resolution mass spectrometry (LC-MS). For this purpose, a suite of representative low-molecular weight compounds such as pesticides, drugs of abuse, sport drugs or environmental contaminants were selected. The approach is based on the use of reversed-phase C18 nano columns furnished with an integrated emitter tip. The nanoflow LC system was combined with full-scan high resolution mass spectrometry using a HRMS (orbitrap) instrument operated at a resolution of 70000. The sensitivity achieved with this configuration enables the implementation of high dilution factors (e.g. 1:20, 1:50 or beyond). When combining nanoflow LC-MS analysis with such high dilution factors (e.g. 1:50), signal suppression was negligible in most cases, so that matrix-matched standards may eventually be skipped, simplifying laboratory workflows by using external calibration in demanding applications such as drug analysis in urine, environmental contaminants in wastewater or pesticide testing in food, thus, eliminating the need for standard addition, matrix-matched calibration or isotopically-labelled standards.

A new approach for the calibration of SEC-light scattering (SEC-LS) setups is proposed, which requires solely the molar mass of a reference polymer. Neither the specific refractive index increment of the calibrant nor of the analyte is required. Comparison of the molar masses derived in different solvents for a large number of chemically different polymers shows that the new approach yields the same molar masses as if molar masses were derived using dn/dc to calibrate the light scattering setup. The approach therefore allows easier determination of molar masses by SEC-LS.

Few articles are reported for the simultaneous separation and sensitive detection of the kynurenine pathway (KP) metabolites. This work describes a capillary electrochromatography-mass spectrometry (CEC-MS) method using acrylamido-2-methyl-1-propanesulfonic acid (AMPS) functionalized stationary phase. The AMPS column was prepared by first performing silanization of bare silica with gamma-maps, followed by polymerization with AMPS. The CEC-MS/MS methods were established for six upstream and three downstream KP metabolites. The simultaneous separation of all nine KP metabolites is achieved without derivatization for the first time in the open literature. Numerous parameters such as pH and the concentration of background electrolyte, the concentration of the polymerizable AMPS monomer, column length, field strength, and internal pressure were all tested to optimize the separation of multiple KP metabolites. A baseline separation of six upstream metabolites, namely tryptophan (TRP), kynurenine (KYN), 3-hydroxykynurenine (HKYN), kynurenic acid (KA), anthranilic acid (AA), and xanthurenic acid (XA), was possible at pH 9.25 within 26 min. Separation of six downstream and related metabolites, namely: tryptamine (TRPM), hydroxy‑tryptophan (HTRP), hydroxyindole-3 acetic acid (HIAA), 3-hydroxyanthranilic acid (3-HAA), picolinic acid (PA), and quinolinic acid (QA), was achieved at pH 9.75 in 30 min. However, the challenging simultaneous separation of all nine KP metabolites was only accomplished by increasing the column length and simultaneous application of internal pressure and voltage in 114 min. Quantitation of KP metabolites in commercial human plasma was carried out, and endogenous concentration of five KP metabolites was validated. The experimental limit of quantitation ranges from 100 to 10,000 nM (S/N = 8–832, respectively), whereas the experimental limit of detection ranges from 31 to 1000 nM (S/N = 2–16, respectively). Levels of five major KP metabolites, namely TRP, KYN, KA, AA, and QA, and their ratios in patient plasma samples previously screened for inflammatory biomarkers [C-reactive protein (CRP) and tumor necrosis factor-alpha (TNF-α)] was measured. Pairs of the level of metabolites with significant positive correlation were statistically evaluated.

In this study, cerumen, a non-conventional biological secretion, was examined as an alternative matrix for forensic analyzis. A fully validated analytical UPLC-MS/MS method was developed for the detection and quantification of the most prevalent psychoactive illicit drug globe wide, Δ9-tethrahydrocannabinol, commonly known as THC, and four major cannabinoids found in cannabis Sativa. The method was validated, and standard external calibration curves were established with correlation coefficients > 0.99. A validated experimental procedure, along with a direct extraction of cannabinoids with acidified acetonitrile resulted in a short total analyzis time and a good extraction efficiency for all the analytes under study. LOD and LOQ values were determined to be 0.01-0.08 pg/mg and 0.04-0.23 pg/mg, respectively. To prove applicability of the proposed assay, volunteers were selected, and cerumen samples were examined for cannabis. The analyzis by use of UPLC-MS/MS indicated that all samples were positive, reporting recent cannabis abuse. Surprisingly, both THC and Cannabinol (CBN) were detected, and quantification was possible in 75% of the cases.

Measurement of chiral thiol compounds such as glutathione (GSH), cysteine (Cys), and homocysteine (Hcy) in human serum plays an important role in the early diagnosis and warning of cardiovascular disease, neurodegenerative disease, and cancer. We developed a novel chiral mass spectrometry derivatization reagent, (R)-(5-(3-isothiocyanatopyrrolidin-1-yl)-5-oxopentyl) triphenylphosphonium (NCS-OTPP), with triphenylphosphine (TPP) as a basic structure carrying a permanent positive charge for the diastereomeric separation of chiral thiol compounds by ultrahigh-performance liquid chromatography coupled to quadrupole-Orbitrap high resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS). A novel method was developed for simultaneous determination of three kinds of chiral thiol compounds based on the NCS-OTPP derivatization method. Three kinds of chiral thiol compounds on a YMC Triart C18 (2.0 × 150 mm, 1.9 μm) column with Rs were 1.56–1.68. The protonated precursor to product ion transitions monitored for GSH was m/z 780.16→747.24/473.18, Cys was m/z 594.20→561.18/473.18, and Hcy was m/z 608.21→575.19/473.18. An excellent linearity for all the analytes with correlation coefficients ≥ 0.9995 and suitable precision with inter-day and intra-day coefficients of variation RSDs was 0.83–4.06% and 0.95–3.11%. Satisfactory accuracy with recoveries between 83.73 and 103.35% was observed. The limit of detection (S/N = 3) was 2.4–7.2 fmol. Furthermore, the method was successfully applied to the simultaneous determination of three kinds of free and total thiol compounds in serum from 10 healthy volunteers at normal and stress states.

A three column-switching high-performance liquid chromatography (HPLC) using an electrochemical detector (ECD) equipped with a diamond electrode was established to determine 3-hydroxypropylmercapturic acid (3-HPMA) in urine. An extracted urine sample was consecutively fractionated using a strong anion-exchange column (first column) and a C8 column (second column) via a switching valve before application on an Octa Decyl Silyl (ODS) column (third column), followed by ECD analysis. The% recovery of 3-HPMA standard throughout the three-column process and limit of detection (LOD) were 94±1% and 0.1pmol, respectively. A solid phase extraction step is required for the sensitive analysis of 3-HPMA in urine by column-switching HPLC-ECD despite a decreased% recovery (55%) of urine sample spiked with 100pmol of 3-HPMA. To test the utility of our column-switching HPLC-ECD method, 3-HPMA levels of 27 urine samples were determined, and the correlation between HPLC-ECD and LC-Electrospray ionization (ESI)-MS/MS method was examined. As a result, the median values of μmol 3-HPMA/g Creatinine (Cre) in urine obtained by column-switching HPLC-ECD and LC-MS/MS were 2.19±2.12μmol/g Cre and 2.13±3.38μmol/g Cre, respectively, and the calibration curve (y=1.5171x-1.007) exhibited good linearity within a defined range (r2=0.907). These results indicate that the combination of column-switching HPLC and ECD is a powerful tool for the specific, reliable detection of 3-HPMA in urine.

Despite their advantages, there is concern that single-use systems used in biopharmaceutical manufacture might release potentially toxic substances during standard unit operations that negatively impact cell growth. Characterization of the extractables profile for single-use systems is necessary to know which compounds potentially become leachables under operational cell culture conditions. A key issue in the design of extractables studies is the composition of the model solvent, in particular its pH and polarity. In this study, a new approach, based on design of experiments (DoE), has been applied to determine the composition of the model solvent for extractable profiling of single-use bags (SUBs). Particular focus was placed on the determination of the degradation products of the antioxidant Irgafos 168®, due to evidence that some of these degradation products have cytotoxic effects on CHO cells. Results indicated that 2-propanol:water is the most appropriate solvent for the extraction of highly hydrophobic compounds with polar groups and/or acid-base properties from SUBs. The described DoE approach simplifies the number of experiments, evaluates all possible solvent water mixtures to select the best extraction solvent based on polarity, establishes the influence of each variable and provides information about variable interaction, which represents an important improvement over current best practice. The developed approach was applied to seven SUBs from different vendors and production dates facilitating the identification of potentially non-satisfactory films for cultivation of CHO cell lines under process conditions.

Flame ionized detector has been such widely applied in chemical analysis since its great invention and the discovery of chem-ionization. Thanks to the excellent contribution of Sternberg and the successors in this field, effective carbon number concept (ECN) can make the analysis calibration greatly simplified especially when authentic standard substances are unavailable or in the complicated case such as petrochemical industry and biomass processing. To supplement the ECN rule in multi-ethers, this work determined relative response factors of poly(oxymethylene) dimethyl ethers experimentally, and developed a probabilistic P1-P2 effective carbon number model (P-ECN) for multi-ethers compounds. showed this method could improve the precision of quantitative analysis for poly(oxymethylene) dimethyl ethers and could predict relative response factors of other ethers with similar structure to a degree. LOD for each DMMn monomer ranged between 0.4-0.7 ng, and MDL ranged between 2 and 4 ug/mL. In the practical quantitation of diluted samples at level of 10 ug/mL, the relative standard deviation was less than 5%. Practical ethers-fuel blend with complex composition also was quantified with errors less than 3%.

Traditionally, gas chromatography – mass spectrometry (GC/MS) analysis has used a targeted approach called selected ion monitoring (SIM) to quantify specific compounds that may have adverse health effects. Due to method limitations and the constraints of preparing duplicate samples, the information that could be obtained from separately collecting the full scan chromatogram of the sample has often been sacrificed. However, a hybrid technique called synchronous SIM/scan mode that switches back and forth between the two acquisition methods has become available from equipment manufacturers that maintains the accuracy and sensitivity of SIM for targeted analysis while also providing the full scan chromatogram for discovery of non-target compounds. We have explored the value and performance of this new technology using calibration data and real-world breath samples from a joint EPA/NIOSH collaboration that assessed the safety of firefighters’ protective gear during controlled structural burns. Collecting field samples is costly and must be performed strategically to ensure that time points and replicates are accurate and representative of the intended population. This is especially difficult, if not impossible, to accomplish with firefighters who are working under volatile conditions. The synchronous SIM/scan method decreases the number of field samples that need to be collected by half and reduces error in trying to recreate time points since a breath sample from a single sorbent tube can be used to collect both the SIM and scan data simultaneously. This work demonstrates the performance of the technology using calibration data. As a practical demonstration of the method, we investigate thirty-six firefighter breath samples, document organic compounds of interest, and identify additional non-target compounds.

Shuang-huang-lian powder injection (SHLPI) is a traditional Chinese medicine injection (TCMI) frequently used in the clinical treatment of faucitis, bronchitis, and other viral and bacterial infections of upper respiratory tract. However, its allergenic reactions, being the main adverse effects (AEs) of SHLPI, have been a serious problem of its clinical safety. This problem has not been solved due to short of methods for detecting haptens in complex TCMIs. In this study, an on-line high-performance liquid chromatography-diode-array detector-mass spectrometry combined with bovine serum albumin-fluorescence detector (HPLC-DAD-MS-BSA-FLD) system was established for the first time, validated and applied for identification of haptens in SHLPI. Fourteen of 35 identified compounds showed BSA binding activity, and they were six flavonoids, six caffeoylquinic acids (CQAs), and two phenylethanoid glycosides. The structure-activity relationships of 10 active components were studied, and their ability of sensitization together with that of two CQAs were further verified by ELISA assay. It was found that 10 compounds had sensitization, and flavonoids showed stronger sensitizability than CQAs while the diCQAs were slightly stronger than caffeoylquinic acids. The system was validated using 3-CQA as a positive control, and was proved to have good reproducibility, stability, precision (RSD 0.9993). This online system is fast, sensitive and efficient for screening haptens in traditional Chinese medicine injection (TCMI), provides a new approach to reveal the chemical basis of haptens in TCMIs.

Three quantification methodologies, namely calibration with internal standard (Cal-IS, non-weighted), weighted calibration with internal standard (wCal-IS) and isotope pattern deconvolution (IPD) have been used for the determination of testosterone in urine by LC-MS/MS. Uncertainty has been calculated and compared for the three methodologies through intra- and inter-laboratory reproducibility assays. IPD showed the best performance for the intra-laboratory reproducibility, with RSD and combined uncertainty values below 4% and 9% respectively. wCal-IS showed similar performance, while Cal-IS where not constant and clearly worse at the lowest concentration assayed (2 ng/mL) reaching RSD values up to 16%. The inter-laboratory assay indicated similar results although wCal-IS RSD (20%) was higher than IPD (10%) and Cal-IS get worse with RSD higher than 40% for the lowest concentration level. Uncertainty budgets calculated for the three procedures revealed that intercept and slope were the most important factors contributing to uncertainty for Cal-IS. The main factors for wCal-IS and IPD were the volumes of sample and/or standard measured. Thorough product understanding is one of the basic tenets for successful implementation of Quality by Design (QbD). Complexity encountered in analytical characterization of biotech therapeutics such as monoclonal antibodies (mAbs) requires novel, simpler, and generic approaches towards product characterization. This paper presents a methodology for implementation of QbD for analytical method development. Optimization of an analytical cation exchange high performance liquid chromatography (CEX-HPLC) method utilizing a sigmoidal gradient has been performed using a hybrid mechanistic model that is based on Design of experiment (DOE) based studies. Since sigmodal gradients are much more complex than the traditional linear gradients and have a large number of input parameters (five) for optimization, the number of DOE experiments required for a full factorial design to estimate all the main effects as well as the interactions would be too large (243). To address this problem, a mechanistic model was used to simulate the analytical separation for the DOE and then the results were used to build an empirical model. The mechanistic model used in this work is a more versatile general rate model in combination of modified Langmuir binding kinetics. The modified Langmuir model is capable of modelling the impact of nonlinear changes in the concentration of the salt modifier. Further, to get the input and output profiles of mAb and salts/buffers, the HPLC system, consisting of the mixer, detectors, and tubing was modelled as a sequence of dispersed plug flow reactors and continuous stirred tank reactors (CSTR). The experimental work was limited to calibration of the HPLC system and finding the model parameters through three linear gradients. To simplify the optimization process, only three peaks in the centre of the profile (main product and the adjacent acidic and basic variants) were chosen to determine the final operating condition. The regression model made from the DoE data yielded a R2>0.97 which made it possible to predict and choose the design space where the optimal resolution between the acidic/main peaks and the basic/main peaks could be achieved (>1.2 and >2.5, respectively). The optimal operating condition was validated using experimental runs and was found to give the same resolution as what was predicted by the simulation. The proposed approach aims to significantly reduce the time required for method optimization as well as the extent of experimentation.

In this work, we present a novel concept for a gas chromatography detector utilizing an atmospheric pressure chemical ionization which is initialized by a dielectric barrier discharge. In general, such a detector can be simple and low-cost, while achieving extremely good limits of detection. However, it is non-selective apart from the use of chemical dopants. Here, a demonstrator manufactured entirely from fused silica capillaries and printed circuit boards is shown. It has a size of 75 × 60 × 25 mm 3 and utilizes only 2W of power in total. Unlike other known discharge detectors, which require high-purity helium, this detector can theoretically be operated using any gas able to form stable ion species. Here, purified air is used. With this setup, limits of detection in the low parts-per-billion range have been obtained for acetone.

The paper describes a new online quantitative electrophoretically mediated microanalysis (EMMA) for use in immunoassays based on the unique drop-by-drop introduction of a sample by means of an inkjet for capillary electrophoresis (CE). Plugs of a fluorescein-labeled antibody (Anti-humanIgG-DyL550) and human IgG were alternately injected into a capillary using the inkjet, followed by the merging of the plugs and the subsequent immune reaction. The antigen-antibody complex that was formed in the merged zone was then separated by CE. As a proof-of-concept, the method was used to determine human IgG. As a result, both the consumption of the reaction solution and the analysis time were significantly reduced. The method showed a wide linear range (10–2000 ng mL−1, R2 = 0.9912) of calibration and the detection limit (5 ng mL−1) was substantially lower than that by for conventional methods.

Plastic microchips have been broadly used as disposable microfluidic devices, but the poorly defined surface properties limit their application. Herein, we proved that an anionic polymer could be used as the background electrolyte (BGE) to provide a strong and stable cathodic electroosmotic flow (EOF) and modulate the electrophoretic behavior for efficient separation in relative thicker microchannels (∼75μm id). A cathodic EOF of ∼3.3×10-4cm2V-1s-1 was maintained using sodium polystyrene sulfonate (PSSNa) with a molecular weight of 5×105 as the BGE, which ensured fluorescein isothiocyanate labeled biogenic amines (BAs) appeared ahead of other components in the electropherograms obtained with microchips of cyclic olefin copolymer. Four selected BAs appeared within 50s and theoretical plate numbers of 8.0×105/m were achieved. The role of PSSNa was evaluated with streaming potential, dynamic light scattering, contact angle and atomic force microscopy. Its functionalities as surface modifier, viscosity regulator and pseudostationary phase were also confirmed. The proposed electrophoretic method was applied in the fast determination of BAs in fish meat samples.

In this work, we studied the combination of chemometric methods with chromatographic separations as a strategy applied to the analysis of enantiomers when complete enantioseparation is difficult or requires long analysis times and, in addition, the target signals have interference from the matrix. We present the determination of ibuprofen enantiomers in pharmaceutical formulations containing homatropine as interference by chiral HPLC-DAD detection in combination with partial least-squares algorithms. The method has been applied to samples containing enantiomeric ratios from 95:5 to 99.5:0.5 and coelution of interferents. The results were validated using univariate calibration and without homatropine. Relative error of the method was less than 4.0%, for both enantiomers. Limits of detection (LOD) and quantification (LOQ) for (S)-(+)-ibuprofen were 4.96 × 10−10 and 1.50 × 10−9 mol, respectively. LOD and LOQ for the R-(−)-ibuprofen were LOD = 1.60 × 10−11 mol and LOQ = 4.85 × 10−11 mol, respectively. Finally, the chemometric method was applied to the determination of enantiomeric purity of commercial pharmaceuticals. The ultimate goal of this research was the development of rapid, reliable, and robust methods for assessing enantiomeric purity by conventional diode array detector assisted by chemometric tools. International audience; The effect of the polydispersity of polystyrenes on the dispersion through silicas having different morphologies (fully porous, core-shell particles and monoliths) was investigated. The heights equivalent to a theoretical plate (HETP) of those columns were measured for a small molecule (toluene) and a series of polystyrenes of different sizes in non-adsorbing conditions. The different contributions to the total HETP including polydispersity were determined experimentally. The longitudinal diffusion and the mass transfer resistance term were obtained from peak parking experiments. The eddy dispersion was obtained from models and experiments. The effect of polydispersity on the HETP values (Hpoly) can thus be calculated from the total HETP by substraction of the other contributions. The results were compared to the Knox model which surestimates the Hpoly values for porous and core-shell particles which is usually explained by an overestimation of the polydispersity index (PDI) given by the manufacturer. The PDI of two polymers (P02, Mw= 690 g.mol−1 and P03, Mw=1380 g.mol−1) was verified by liquid chromatography by separating each fraction of the polymer on the silica columns by using adsorbing conditions which are obtained with a mixture of heptane and THF. The PDI obtained are comparable to the PDI given by the manufacturer meaning that the assumptions made by Knox are not entirely valid. A direct method is proposed in this paper in order to determine Hpoly. In this method the excess of spreading as compared with a polymer with only one size corresponding to the average size is studied assuming the polymer size distribution is gaussian. The Hpoly values obtained by the direct method are comparable to the experimental values.

Herein, a facile and practical aptamer-grafted ionic affinity monolith with mixed-mode mechanism was explored as a versatile platform for online specific recognition of polar and non-polar mycotoxins. The mixed-mode mechanism including molecular affinity adsorption (between aptamers and targets), hydrophilic interaction and ionic interaction (between stationary phase and targets) were adopted and provided a better flexibility in adjusting separation selectivity to reduce nonspecific adsorption with respect to the single mode. Preparation and characterization of aptamer-based affinity monoliths were investigated, The characterization of pore size distribution, Brunauer-Emmett-Teller (BET) surface area and the specificity and cross-reaction were also evaluated. As a result, the hydrophilic nature and negative charge on affinity monolith were obtained. Multiple interactions including aptamer affinity binding, hydrophilic interaction (HI) and ion exchange (IE) could be adopted for online selective extraction. Specific recognitions of polar ochratoxin A (OTA), non-polar zearalenone (ZEN) and aflatoxin B1 (AFB1) was fulfilled with LODs as 0.03, 0.05 and 0.05 μg/L, respectively. Applied to real cereals, good recoveries of the fortified OTA, AFB1 and ZEN were achieved as 92.6 ± 1.3% ~ 95.6 ± 1.3% (n=3), 93.9 ± 2.3% ~ 98.2 ± 3.4% (n=3) and 92.7 ± 2.0% ~ 96.9 ± 3.5% (n=3) in corn, wheat and rice, respectively. The results displayed that Apt-MCs with hydrophilic and ionic interaction mixed-mode mechanism were efficient enough and competent for the online recognition of mycotoxins in cereals.

To evaluate the endocannabinoid system in an animal model of Parkinson's disease, in-tube solid-phase microextraction (in-tube SPME) was directly coupled to a tandem mass spectrometry (MS/MS) system for determination of the endocannabinoids anandamide (AEA) and 2-arachidonoylglycerol (2-AG) in rat brain samples. In-tube SPME-which consisted of a microtube of restricted access material (RAM) with a hydrophilic diol external surface and a hydrophobic octyl inner surface-efficiently excluded (up to 95%) macromolecules from the biological samples and selectively pre-concentrated the analytes. In-tube SPME parameters, such as sample volume, mobile phases, flow rate, and pre-concentration time, were evaluated to improve the extraction efficiency and throughput performance. The selectivity of the in-tube SPME and MS/MS (MRM mode) techniques allowed them to be directly coupled online, which dismissed the need for the chromatographic separation step. The in-tube SPME-MS/MS method was validated and shown to be linear from 6.0 to 30.0 ng mL-1 for AEA and from 10.0 to 100.0 ng mL-1 for 2-AG; the intra- and inter-assay accuracy and precision were lower than 15%. Parallelism between the calibration curves constructed in the matrix and aqueous solution confirmed that there was no matrix effect. The method allowed endogenous concentrations of AEA and 2-AG to be determined in rat brain striatum from unilaterally 6-hydroxydopamine-lesioned animals.
