Of the approximately one million described insect species, ground beetles (Coleoptera: Carabidae) have long captivated the attention of evolutionary biologists due to the diversity of defensive compounds they synthesize. Produced using defensive glands in the abdomen, ground beetle chemicals represent over 250 compounds including predator-deterring formic acid, which has evolved as a defensive strategy at least three times across Insecta. Despite being a widespread method of defense, formic acid biosynthesis is poorly understood in insects. Previous studies have suggested that the folate cycle of one-carbon (C1) metabolism, a pathway involved in nucleotide biosynthesis, may play a key role in defensive-grade formic acid production in ants. Here, we report on the defensive gland transcriptome of the formic acid-producing ground beetle Harpalus pensylvanicus. The full suite of genes involved in the folate cycle of C1 metabolism are significantly differentially expressed in the defensive glands of H. pensylvanicus when compared to gene expression profiles in the rest of the body. We also find support for two additional pathways potentially involved in the biosynthesis of defensive-grade formic acid, the kynurenine pathway and the methionine salvage cycle. Additionally, we have found an array of differentially expressed genes in the secretory lobes involved in the biosynthesis and transport of cofactors necessary for formic acid biosynthesis, as well as genes presumably involved in the detoxification of secondary metabolites including formic acid. We also provide insight into the evolution of the predominant gene family involved in the folate cycle (MTHFD) and suggest that high expression of folate cycle genes rather than gene duplication and/or neofunctionalization may be more important for defensive-grade formic acid biosynthesis in H. pensylvanicus. This provides the first evidence in Coleoptera and one of a few examples in Insecta of a primary metabolic process being co-opted for defensive chemical biosynthesis. Our results shed light on potential mechanisms of formic acid biosynthesis in the defensive glands of a ground beetle and provide a foundation for further studies into the evolution of formic acid-based chemical defense strategies in insects.

Hydrocyanic acid (HCN) is a well-known defensive allomone in the chemical arsenal of millipedes in the order Polydesmida. The presence of HCN in the headspace vapor of adult Xystocheir dissecta (Wood, 1867), a common millipede from the San Francisco Bay Area, was traced by laser desorption/ionization-mass spectrometry (LDI-MS). To accomplish this, the headspace vapor surrounding caged, live millipedes was allowed to diffuse passively over gold-nanoparticle (AuNP) deposits placed at various distances from the emitting source. The stainless steel plates with AuNP deposits were removed and irradiated by a 355-nm laser. The gaseous ions generated in this way were detected by time-of-flight mass spectrometry. The intensity of the mass spectrometric peak detected at m/z 249 for the Au(CN)2- complex anion was compared to that of the residual Au- signal (m/z 197). Using this procedure, HCN vapors produced by the live millipedes could be detected up to 50 cm away from the source. Furthermore, the addition of H2O2, as an internal oxygen source for the gold cyanidation reaction that takes place in the AuNP deposits, significantly increased the detection sensitivity. Using the modified H2O2 addition procedure, HCN could now be detected at 80 cm from the source. Moreover, we found a decreasing intensity ratio of the Au(CN)2-/Au- signals as the distance from the emitting source increased, following an exponential-decay distribution as predicted by Fick's law of diffusion. Ponerine ants are known to contain mixtures of pyrazines in their mandibular glands. We analyzed the mandibular gland contents of four ponerine species (Odontomachus chelifer, O. erythrocephalus, O. ruginodis, and O. bauri) by gas chromatography coupled with mass spectrometry, and found that each species contains specific mixtures of trisubstituted alkylpyrazines among other volatiles. Attempts to identify alkylpyrazines solely by mass spectral interpretation is unrealistic because spectra of positional isomers are indistinguishable. To avoid misidentifications, we synthesized a large number of reference compounds and compared their mass spectral and gas chromatographic properties with those present in the Odontomachus species under investigation. Most of the compounds identified were 2-alkyl-3,5-dimethylpyrazines. Interestingly, when the third substituent was an isopentyl group, the two methyl groups were found to be located at the 2 and 5 ring positions. Using our data, we recognized several misidentifications in previous publications.

Tiglic, 2-methylbutyric, and ethacrylic acids are found in the pygidial gland defensive fluid of many carabid beetles. By injecting a deuterium-labeled precursor into the carabid beetle Pterostichus (Hypherpes) californicus, and analyzing the defensive fluid by gas chromatography/mass spectrometry, we were able to demonstrate that tiglic and ethacrylic acids are biosynthesized from isoleucine via 2-methylbutyric acid. Moreover, we observed that the injection of L-isoleucine induces an increased production of tiglic acid in P. californicus. A strong primary kinetic isotope effect was found to operate in the dehydrogenation step of 2-methylbutyric acid to tiglic and ethacrylic acids. Consequently, ethacrylic acid was found to preferentially accumulate the deuterium labeling from [2,3,4,4-(2)H(4)]isoleucine during our biosynthetic experiments.

Analyses of pygidial gland contents of two species of a previously uninvestigated family of beetles (Trachypachidae) by Gas Chromatography-Mass Spectrometry (GC-MS) revealed that their chemistry is similar to that reported from many members of the family Carabidae. Nevertheless, the composition of defensive gland fluids of the two species Trachypachus slevini and T. gibbsii differs sufficiently to distinguish between the two species solely on the basis of their defensive chemistry. The major components of T. slevini glandular fluid are methacrylic, tiglic, and octanoic (= caprylic) acids, together with the hydrocarbon (Z)-9-pentacosene. In contrast, the glandular contents of T. gibbsii contain a rather unique mixture of polar and nonpolar compounds, the principal constituents of which are methacrylic and ethacrylic acids (= 2-ethylacrylic acid), together with 2-phenylethanol, 2-phenylethyl methacrylate, 2-phenylethyl ethacrylate, and (Z)-9-pentacosene.

The secretion of the paracloacal glands of the American crocodile (Crocodylus acutus) contains over 80 lipophilic compounds, including saturated and unsaturated long-chain alcohols along with their formic, acetic, and butyric acid esters, and several isoprenoids. Most of these compounds were identified on the basis of mass spectra, obtained by GC-MS. In addition, identification of the major components was supported by infrared spectra obtained by GC-FTIR. Major differences are indicated in the composition of the paracloacal gland secretion of C. acutus and that of another crocodylid, the African dwarf crocodile (Osteolaemus tetraspis).

The host plants of the native American butterfly, Pieris napi oleracea, include most wild mustards. However, garlic mustard, Alliaria petiolata, a highly invasive weed that was introduced from Europe, appears to be protected from this insect. Although adults will oviposit on the plant, most larvae of P. n. oleracea do not survive on garlic mustard. We used feeding bioassays with different larval stages of the insect to monitor the isolation and identification of two bioactive constituents that could explain the natural resistance of this plant. A novel cyanopropenyl glycoside (1), alliarinoside, strongly inhibits feeding by first instars, while a flavone glycoside (2), isovitexin-6"-D-beta-glucopyranoside, deters later instars from feeding. Interestingly, the first instars are insensitive to 2, and the late instars are little affected by 1. Furthermore, differential effects of dietary experience on insect responses suggest that 1 acts through a mechanism of post-ingestive inhibition, whereas 2 involves gustatory deterrence of feeding.

In wind-tunnel bioassays, dispensers loaded with 1 μg of the synthetic major component (3E,8Z,11Z)-3,8,11-tetradecatrienyl acetate (TDTA) of the sex pheromone emitted by Tuta absoluta (Meyrick) females were found to be highly attractive to conspecific males. Field experiments were conducted to evaluate the efficacy of five trap designs. The best trap, baited with 100 μg of the synthetic sex pheromone caught on average 1200 males per trap per night, while those baited with virgin females caught only 201 males. The male response to this pheromone is restricted to the same early-morning time window during which females exhibit calling behavior. The high biological activity of the synthetic pheromone suggests that it could be useful for pest monitoring and in mating disruption.

Members of a wild group of wedge-capped capuchin monkeys (Cebus olivaceus) intentionally anoint themselves with millipedes (Orthoporus dorsovittatus). Chemical analysis revealed these millipedes secrete two benzoquinones, compounds known to be potently repellent to insects. We argue that the secretion that rubs off on the monkeys in the course of anointment provides protection against insects, particularly mosquitoes (and the bot flies they transmit) during the rainy season. Millipede secretion is so avidly sought by the monkeys that up to four of them will share a single millipede. The anointment must also entail risks, since benzoquinones are toxic and carcinogenic. We suggest that for capuchins the immediate benefits of anointment outweigh the long-range costs.

Field evaluations demonstrate that the addition of the minor pheromone component (3E,8Z)-tetradecadien-1-yl acetate to the major component (3E,8Z,11Z)-tetradecatrien-1-yl acetate does not significantly increase the trap catches of Tuta absoluta males in the field. The triene acetate itself is highly attractive, catching about 869 ± 255 males per trap in three consecutive nights. The addition of two isomers of the minor component (3E,8Z)-tetradecadien-1-yl acetate, (3E,11Z)-tetradecadienyl-yl acetate and (8Z,11Z)-tetradecadien-1-yl acetate, to the major component (3E,8Z,11Z)-tetradecatrien-1-yl acetate also did not significantly alter the number of the males caught in the traps.

3,4-Dihydro-8-hydroxy-3-methylisocoumarin (mellein) and 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one are among the volatile constituents identified from the hindgut of the formicine ant Lasius (Dendrolasius) fuliginosus Mellein induces trail-following behavior in worker ants of this species and evokes electrophysiological responses from their antennae. The trail-following activity released by (R)-(−)-mellein is significantly higher than that elicited by its (S)-(+) antipode, or the racemic mixture. The above-mentioned pyranone is found also in the honeydew of aphids, the customary diet of this ant species. The pyranone also evokes some trail-following behavior, but its activity is far less pronounced than that of mellein. Apparently, extra pheromone components are present in the hindgut, since the activity of these two constituents, either individually or as a mixture, does not completely account for the total activity released by a hindgut extract containing similar amounts of these two compounds.

The sex attractant ofScrobipalpuloides absoluta females is a 90:10 mixture of (3E,8Z,11Z)-3,8,11-tetradecatrien-1-yl acetate and (3E,8Z)-3,8-tetradecadien-1-yl acetate. Tetradecadienyl acetates bearing 8Z,11Z; 3E,8Z; and 3E,11Z double bonds were synthesized by stereospecific procedures; the mass spectral and gas chromatographic properties of the 3E,8Z isomer were found to be congruent with those of the tetradecadienyl acetate fromS. absoluta. In wind tunnel bioassays, a 10:1 mixture of synthetic (3E,8Z,11Z)-3,8,11-tetradecatrien-1-yl acetate and (3E,8Z)-3,8-tetradecadien-1-yl acetate was highly attractive toS. absoluta males. Interestingly, the presence of (8Z,11Z)-8,11-tetradecadien-1-yl acetate (10%) inhibited the response to (3E,8Z,11Z)-3,8,11-tetradecatrien-1-yl acetate completely.

The cervical gland secretion of larvae ofSchizura unicornis andS. badia (Lepidoptera: Notodontidae) contains formic acid, acetic acid, and a mixture of lipophilic compounds, including 2-alkanones, 2-alkanols, and formates of the alkanols. InS. unicornis, the secretion also contains severalω-monounsaturated analogs of the alcohols, ketones, and formates. The absolute configuration of two of the alcohols (2-tridecanol and 2-pentadecanol) was established asS in both species. The larvae spray their secretion when physically disturbed, aiming it accurately in the direction of the offending agent.

The acceptance of Solanum surattenses as a host plant for the larvae of Manduca sexta was explained by the presence of feeding stimulants in foliage. Bioassay-guided fractionation of plant extracts resulted in the isolation of a highly active compound (1), which was identified as a furostan derivative {26-O-beta-D-glucopyranosyl-(25R)-furosta-5-ene-3-beta-yl-O-alpha-L-rhamnopyranosyl-(1''-2')-O-alpha-L-rhamnopyranosyl-(1'''-3'')-O-beta-D-glucopyranoside}. This compound has the same steroidal core substructure as that in a stimulant (indioside D) previously identified from potato foliage. However, the sugar substituents attached to the core are different.

The defensive spray of a single female of the rare carabid beetle,Helluomorphoides clairvillei (subfamily Carabinae, supertribe Lebiitae, tribe Helluonini). was found to contain a mixture of compounds, including carboxylic acids (formic, acetic), aliphatic esters (principally nonyl acetate), and hydrocarbons (principally decane). In a single series of discharges, the beetle ejected a total of about 1% of its body weight as formic acid (1.1 mg). Our findings demonstrate that characterization of secretory components from even minimal samples of secretion should be possible in many cases.

The defensive secretion ofTenebrio molitor contains a mixture of 2-methyl-1,4-benzoquinone andm-cresol. The phenol had not previously been detected in the secretion, although some investigators reported presence of 2-ethyl-1,4-benzoquinone as a second component. We failed to detect the latter quinone in secretion samples from three laboratory populations ofT. molitor.

The structure elucidation of sex pheromones of Lepidoptera by a solid-sample injection technique in conjunction with capillary gas chromatography is described. The applicability of this method in GC and GC-MS modes was demonstrated by reanalyzing the sex attractants of females ofOstrinia nubilalis andBombyx mori. The pheromone complex ofMamestra brassicae was reinvestigated and (Z)-9-hexadecenyl acetate and (Z)-11-hexadecenol were found in addition to already known pheromone components of this species. By using the solid-sample injection, the exact site of pheromone release could be determined inM. brassicae.

The secretion of the hypertrophied metapleural gland of the antCrematogaster deformis contains a mixture of phenols, consisting mainly of 3-propylphenol, 3-pentylphenol, 3,4-dihydro-8-hydroxy-3-methylisocoumarin (mellein), 5-propylresorcinol, and 5-pentylresorcinol. The secretion is released, as a repellent, when the highly vulnerable petiolar-postpetiolar region of the abdomen is attacked by enemy ants. In addition, small amounts of the secretion are released regularly to serve as an antiseptic, which is considered the original function of the gland. The secretion also has some insecticidal properties.
