Weeds have been a major threat in agriculture for several generations as they lead to decreases in productivity and cause significant economic losses. Parasitic plants are a specific type of weed causing losses in crops of great relevance. A new strategy has emerged in the fight against parasitic plants, which is called 'suicidal germination' or the 'honey-pot strategy'. Regarding the problem of weed control from an ecological point of view, it is interesting to investigate new natural compounds with allelopathic activity with the aim of developing new natural herbicides that can inhibit the growth of weeds without damaging the environment. Safflower crops have been affected by parasitic plants and weeds and, as a consequence, the secondary metabolites exuded by safflower roots have been studied. The sesquiterpene lactone dehydrocostuslactone was isolated and characterised, and the structurally related costunolide was identified by UHPLC-MS/MS in safflower root exudates. These sesquiterpene lactones have been shown to stimulate germination of Phelipanche ramosa and Orobanche cumana seeds. In addition, these compounds were phytotoxic on three important weeds in agriculture, namely Lolium perenne, Lolium rigidum and Echinochloa crus-galli. The exudation of the strigolactones solanacol and fabacyl acetate have also been confirmed by UHPLC-MS/MS. The study reported here contributes to our knowledge of the ecological role played by some secondary metabolites. Pale swallow-wort ( Vincetoxicum rossicum) and black swallow-wort ( V. nigrum) are two invasive plant species in the northeastern United States and eastern Canada that have undergone rapidly expanding ranges over the past 30 years. Both species possess a highly bioactive phytotoxin -(−) antofine in root tissues that causes pronounced inhibition in laboratory bioassays of native plant species co-located in habitats where swallow-wort is found. To further evaluate the allelopathic potential of -(−) antofine, we: determined its concentration in young plant tissues; used in situ approaches to assess antofine stability, potential activity of degradation products, activity in sterile and nonsterile soil; and determined accumulation and concentration in hydroponic cultivation and field collected soil samples. Extracts of seeds and young seedlings were found to have approximately 2-3 times the level of -(−) antofine in comparison to root extracts of adult plants. Breakdown products of antofine accumulated rapidly with exposure to light, but more slowly in the dark, at ambient temperatures, and these products did not retain biological activity. Extraction efficiencies of control soil spiked with -(−) antofine were low but easily detectable by HPLC. Soil samples collected over two growing seasons at four different sites where either pale swallow-wort or black swallow-wort populations are present were negative for the presence of -(−) antofine. Dose response curves using sterile and nonsterile soil spiked with -(−) antofine demonstrated a requirement for at least 20-55 × greater -(−) antofine concentrations in soil to produce similar phytotoxic effects to those previously seen in agar bioassays with lettuce seedlings. Sterile soil had a calculated EC of 686 μM (250 μg/g) as compared to nonsterile soil treatments with a calculated EC of 1.88 mM (640 μg/g). When pale swallow-wort and black swallow-wort adult plants were grown in hydroponic cultivation, −(−) antofine was found in root exudates and in the growing medium in the nM range. The concentrations in exudate were much lower than that needed for biological activity (μM) although they might be an underestimate of what may accumulate over time in an undisturbed rhizosphere. Understanding allelopathy has been hindered by the lack of methods available to monitor the dynamics of allelochemicals in the soil. Previous work has demonstrated the feasibility of using polydimethylsiloxane (PDMS) microtubing (silicone tubing microextraction, or STME) to construct sampling devices to monitor the release of lipophilic allelochemicals from plant roots. The objective of this study was to use such sampling devices to intensively monitor thiophene fluxes beneath marigolds over several weeks to gain insight into the magnitude of temporal and spatial heterogeneity in these fluxes. Marigolds were grown in rhizoboxes (20.5 x 20.5 x 3.0 cm) with 16 individual STME samplers per box. Thiophene sampling and HPLC analysis began 45 days after planting. At the end of the study, roots around each sampler were analyzed by HPLC. Results confirmed the tremendous spatial and temporal heterogeneity in thiophene production seen in our previous studies. STME probes show that thiophene concentrations generally increase over time; however, these effects were sampling-port specific. When sampling ports were monitored at 12 h intervals, fluxes at each port ranged from 0 to 2,510 ng day. Fluxes measured over daylight hr averaged 29 % higher than those measured overnight. Fluxes were less than 1 % on average of the total thiophene content of surrounding roots. While the importance of such heterogeneity, or 'patchiness', in the root zone has been recognized for soil nutrients, the potential importance in allelopathic interactions has seldom been considered. The reasons for this variability are unclear, but are being investigated. Our results demonstrate that STME can be used as a tool to provide a more finely-resolved picture of allelochemical dynamics in the root zone than has previously been available. This review examines the state of the field for chemically mediated interactions involving marine angiosperms (seagrasses, mangroves, and salt marsh angiosperms). Small-scale interactions among these plants and their herbivores, pathogens, fouling organisms, and competitors are explored, as are community-level effects of plant secondary metabolites. At larger spatial scales, secondary metabolites from marine angiosperms function as reliable cues for larval settlement, molting, or habitat selection by fish and invertebrates, and can influence community structure and ecosystem function. Several recent studies illustrate the importance of chemical defenses from these plants that deter feeding by herbivores and infection by pathogens, but the extent to which allelopathic compounds kill or inhibit the growth of competitors is less clear. While some phenolic compounds such as ferulic acid and caffeic acid act as critical defenses against herbivores and pathogens, we find that a high total concentration of phenolic compounds within bulk plant tissues is not a strong predictor of defense. Residual chemical defenses prevent shredding or degradation of plant detritus by detritivores and microbes, delaying the time before plant matter can enter the microbial loop. Mangroves, marsh plants, and seagrasses remain plentiful sources of new natural products, but ecological functions are known for only a small proportion of these compounds. As new analytical techniques are incorporated into ecological studies, opportunities are emerging for chemical ecologists to test how subtle environmental cues affect the production and release of marine angiosperm chemical defenses or signaling molecules. Throughout this review, we point to areas for future study, highlighting opportunities for new directions in chemical ecology that will advance our understanding of ecological interactions in these valuable ecosystems. Abstract  The difficulties of monitoring allelochemical concentrations in soil and their dynamics over time have been a major barrier to testing hypotheses of allelopathic effects. Here, we evaluate three diffusive sampling strategies that employ polydimethylsiloxane (PDMS) sorbents to map the spatial distribution and temporal dynamics of root-exuded thiophenes from the African marigold, Tagetes erecta. Solid phase root zone extraction (SPRE) probes constructed by inserting stainless steel wire into PDMS tubing were used to monitor thiophene concentrations at various depths beneath marigolds growing in PVC pipes. PDMS sheets were used to map the distribution of thiophenes beneath marigolds grown in thin glass boxes. Concentrations of the two major marigold thiophenes measured by these two methods were extremely variable in both space and time. Dissection and analysis of roots indicated that distribution of thiophenes in marigold roots also was quite variable. A third approach used 1 m lengths of PDMS microtubing placed in marigold soil for repeated sampling of soil without disturbance of the roots. The two ends of the tubing remained out of the soil so that solvent could be washed through the tubing to collect samples for HPLC analysis. Unlike the other two methods, initial experiments with this approach show more uniformity of response, and suggest that soil concentrations of marigold thiophenes are affected greatly even by minimal disturbance of the soil. Silicone tube microextraction gave a linear response for α-terthienyl when maintained in soils spiked with 0–10 ppm of this thiophene. This method, which is experimentally simple and uses inexpensive materials, should be broadly applicable to the measurement of non-polar root exudates, and thus provides a means to test hypotheses about the role of root exudates in plant-plant and other interactions. Abstract  Ferulic acid, in the form of feruloyl CoA, occupies a central position as an intermediate in the phenylpropanoid pathway. Due to the allelopathic function, its effects were tested on root growth, H2O2 and lignin contents, and activities of cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.195) and peroxidase (POD, EC 1.11.1.7) from soybean (Glycine max (L.) Merr.) root seedlings. Three-day-old seedlings were cultivated in half-strength Hoagland's solution (pH 6.0), with or without 1.0 mM ferulic acid in a growth chamber (25°C, 12/12 hr light/dark photoperiod, irradiance of 280 μmol m−2 s−1) for 24 or 48 hr. Exogenously supplied ferulic acid induced premature cessation of root growth, with disintegration of the root cap, compression of cells in the quiescent center, increase of the vascular cylinder diameter, and earlier lignification of the metaxylem. Moreover, the allelochemical decreased CAD activity and H2O2 level and increased the anionic isoform PODa5 activity and lignin content. The lignin monomer composition of ferulic acid-exposed roots revealed a significant increase of guaiacyl (G) units. When applied jointly with piperonylic acid (an inhibitor of the cinnamate 4-hydroxylase, C4H), ferulic acid increased lignin content. By contrast, the application of 3,4-(methylenedioxy) cinnamic acid (an inhibitor of the 4-coumarate:CoA ligase, 4CL) with ferulic acid did not. Taken together, these results suggest that ferulic acid may be channeled into the phenylpropanoid pathway (by the 4CL reaction) and, further, may increase the lignin monomer amount solidifying the cell wall and restricting the root growth. Abstract  Methyl jasmonate (MeJA) and methyl salicylate (MeSA) are important signaling molecules that induce plant defense against insect herbivores and microbial pathogens. We tested the hypothesis that allelopathy is an inducible defense mechanism, and that the JA and SA signaling pathways may activate allelochemicals release. Exogenous application of MeJA and MeSA to rice (Oryza sativa L.) enhanced rice allelopathic potential and led to accumulation of phenolics, an increase in enzymatic activities, and gene transcription of phenylalanine ammonia-lyase (PAL) and cinnamate 4-hydroxylase (C4H), two key enzymes in the phenylpropanoid pathway. Aqueous extracts of the leaves of rice IAC165, a putative allelopathic variety, treated with MeSA (5 mM) or MeJA (0.05 mM), showed increased inhibitory effects (25 and 21%, respectively) on root growth of barnyardgrass (Echinochloa crus-galli L.), and increased inhibitory effects (18 and 23%, respectively) on shoot growth. Aqueous extracts from leaves of Huajingxian 1 rice, a putative nonallelopathic variety treated with MeJA and MeSA, caused 63 and 24% inhibition of root growth in barnyardgrass seedlings. The root exudates of both IAC165 and Huajingxian 1 plants treated with MeJA and MeSA for 48 hr also showed significant increases in their inhibitory effects on root growth of barnyardgrass seedlings. At the four-leaf stage, levels of 3,4-hydroxybenzoic acid, vanillic acid, coumaric acid, and ferulic acid that accumulated in the leaves were 5.3-, 31.3-, 2.2-, and 1.7-fold higher in response to MeJA exposure, and 3.3-, 13.1-, 2.0-, and 2.2-fold higher in response to MeSA. Treatments of MeSA and MeJA enhanced the PAL activity in the rice leaves up to 52.3 and 80.1%, respectively, whereas C4H activity was increased by 40.2 and 67%. Gene transcription of PAL and C4H in rice leaves significantly increased after the plants were subjected to treatment with MeJA and MeSA. Laboratory screening studies were conducted to evaluate the allelopathic potential of fine leaf fescues. Of the seven accessions selected from prior field evaluations for weed-suppressive ability, all inhibited root growth of large crabgrass and curly cress in laboratory assays. Grown in agar as a growth medium and in the presence of living rescue seedlings for 14 or 21 days, test species were sensitive depending on the fescue cultivars. Growth inhibition increased when rescue was grown for increasing periods of time in agar. Seedling rescues produced significant quantities of bioactive root exudates, which were released into the agar medium. Bioactive root exudates were extracted from living fescue roots by using methylene chloride. Shoot tissue was extracted in water and the aqueous extract was partitioned against hexane, ethyl acetate, and methylene chloride. Extracts were tested for inhibitory activity on seedling growth as measured by inhibition of curly cress germination and radicle elongation. Root exudates were more toxic (70% inhibition) than shoot extracts (up 40% inhibition), when formulated at 0.25 mg/ml concentration. Light microscopy and transmission electron microscopy were utilized in an attempt to identify the cellular location of production of secondary products contained in bioactive root exudates. Ultrastructural analysis indicated that the exudate is produced in actively dividing tips of fibrous root cells. The mode of release of these exudates into the environment remains unknown. Macaranga tanarius is widely distributed in the abandoned lowlands of Taiwan where substantial amounts of leaves accumulate on the ground. A unique pattern of weed exclusion underneath trees is often found and thought to result from allelopathic interactions. Density-dependent phytotoxicity analysis of Lactuca sativa L. (lettuce) growing in soil mixed with the powder of M. tanarius leaves showed a significant deviation from the expected yield-density relationship. Lettuce growth was most suppressed in the low seed density experiment suggesting that the phytotoxins produced during leaf decomposition inhibit the growth of lettuce seedlings. Bidens pilosa and Leucaena leucocephala, growing in soil mixed with the leaf powder of M. tanarius were also suppressed. Aqueous leaf extracts were bioassayed against lettuce and B. pilosa, and exhibited a significant suppression in radicle growth. Compounds identified from leaves included nymphaeol-A (1), nymphaeol-B (2), nymphaeolC (3), quercetin (4), abscisic acid (ABA) (5), blumenol A (6), blumenol B (7), roseoside 11 (8), tanariflavanone A (9), and tanariflavanone B (10). ABA was the major growth inhibitor. At concentrations of 20 ppm, ABA suppressed lettuce germination, while at 120 ppm it inhibited the growth of Miscanthus floridulus, Chloris barbata, and Bidens pilosa. At 600 ppm, quercetin, blumenol A, and blumenol B, caused 20-25% inhibition of radicle and shoot growth of M. floridulus. The amount of ABA in M. tanarius leaves was approximately 3-5 µg g[sup -1] dry weight, significantly higher than previously reported. We conclude that the pattern of weed exclusion underneath stands of M. tanarius and its invasion into its adjacent grassland vegetation results from allelopathic interactions. [ABSTRACT FROM AUTHOR]

Artemisia tridentata ssp. tridentata is the dominant and defining shrub in the Great Basin Desert, with well-documented allelopathic tendencies that have generally been ascribed to its most abundantly released secondary metabolites. However, as a minor component, sagebrush releases a highly biologically active substance, methyl jasmonate (MeJA), which is known to function as both a germination inhibitor and promoter in laboratory studies. Nicotiana attenuata is a tobacco species native to the Great Basin Desert and grows in newly burned juniper-sagebrush habitats for 2-3 yr following a fire. With a combination of field and laboratory studies, we examined the role of MeJA release from sagebrush by both air and water transport in inhibiting N. attenuata seed germination. We demonstrated that sagebrush interacts allelopathically with the seed bank of N. attenuata through its release of MeJA. In the field, seeds buried 0-0 cm from sagebrush plants for 4 months in net bags had significantly reduced germination compared to seeds buried similarly but protected in plastic bags. Moreover, germination on soils collected lom underneath sagebrush plants was reduced by 60% compared to seeds placed on soils collected between sagebrush plants or outside of the sagebrush population. Exposure to A. tridentata seeds and seedlings did not affect N. attenuata germination, suggesting that established sagebrush plants only influence the tobacco's seed bank. In the laboratory, exposure of seeds to sagebrush emissions resulted in germination delays of up to 6 d. Exposure to volatile and aqueous MeJA also inhibited germination of N. attenuata seeds at quantities that are released naturally by sagebrush: 3.5 µg/hr and 1.12 µg/seed cup (56 ng/seed), respectively. A. tridentata seeds were significantly more resistant to MeJA, being inhibited at 336 µg MeJA (16.8 µg/seed), 300 times greater than the level of aqueous MeJA required to inhibit N. attenuata seeds.
