This study was conducted in order to investigate biological compatibility of a thin and flexible hydroxyapatite (HAP) paper which consists of ultralong hydroxyapatite nanowires. Circular‐shaped cranial bone defects with a diameter of 8.8 mm were prepared to expose the dura maters in Wistar rats. The similar‐sized, circular‐shaped HAP paper was placed at the bottom of the bone defects. After 2, 4, and 8 weeks, the rats were sacrificed, and the experimental sections were examined by micro‐CT scanning and histological observation. The HAP paper covered with fibrous tissues showed no inflammatory cell infiltration, and their thicknesses decreased over time. Tartrate‐resistant acid phosphatase‐positive osteoclast‐like cells were induced around the edges of the HAP paper along with the exfoliation of the HAP paper. The newly‐formed bones were observed in the bone‐defected areas, either with a direct contact with the HAP paper or through thin fibrous tissues. The HAP paper‐induced osteoblast differentiation was confirmed since the alkaline phosphatase activities were detected on the surfaces of the HAP paper. These results indicated that the HAP paper may induce osteogenesis without causing any harmful effects. Paper‐based microfluidics devices can create a new healthcare model. Cellulose is carbohydrate polymer biocompatible and hydrophilic. These characteristics enhance the development of user‐friendly diagnostic devices, but the link between paper manufacturing process and performance of the devices is still unclear. Previous studies focused on either commercial papers or lab papers from wood‐cellulose fibers, with different basis‐weight. This work introduces the effect of refining process and lab paper from nonwood‐cellulose fibers, focusing on sisal fibers to overcome the aforementioned challenge. Structural characteristics of paper, such as basis‐weight and degree of refining, are optimized and correlated with blood typing test resolution. Unrefined sisal paper of 50 g/m2 and 100 g/m2 basis‐weight exhibit a higher gray intensity level than refined paper, and also maximal capillary rise and a pore size suitable for blood grouping tests. This study presents an enhanced paper‐based analytical device (PAD) for forward and reverse group blood typing. The proposed PAD uses a novel methodology, which provides highly reliable results on a fully cellulose based device. The PAD was printed on different cellulose substrates. These substrates were made of different cellulose fibers (sisal and eucalyptus), different grammages, refining steps, and wet additive content. Best parameters were chosen to achieve high reliability on both forward and reverse blood typing. The substrates were patterned with five hydrophilic channels and two hydrophobic areas. For reverse blood typing, the hemoagglutination reaction took place on the hydrophobic surface of the paper before being transferred to the paper web, where together with the forward blood typing tests were all washed with saline solution to read the results by elution. This device allows direct read‐out of results; the stains show were agglutination happens. Different blood types were in full agreement between the reverse and forward method and in agreement with traditional methods. The time and simplicity of this methodology confirmed its utility. A thin plastic‐like film separated from the epidermis of Cereus hildmannianus has excellent tensile strength, resistance to water and high antimicrobial activity and supports the growth of mouse fibroblast cells. Cactuses are one of the most under explored plant species with high potential for food, materials, pharmaceutical and other applications. Although studies have shown the ability of cactuses to be used for food, as a source for fibers, as reinforcement for composites and other applications, the role of individual layers and their properties has been studied to a limited extent. In this paper, a thin translucent layer was separated from the epidermis of C. hildmannianus and studied for its composition, structure and properties. The layer is composed of about 73% cellulose and 2% lignin and morphologically, shows surface with uneven and serrated edges. Films with length of up to 36 cm, strength of 6.8 MPa and elongation of 2.5% could be peeled from the cactus suggesting their suitability for food packaging and other applications. X‐ray diffraction patterns and FTIR spectrums indicated that the films are similar to that of cellulose and major thermal degradation occurred above 280°C. Compared to standards, the cactus films showed about 41% and 44% inhibition against gram positive and gram negative bacteria and 67% inhibition of the common fungal strain (A. niger). Films showed high stability in water and to common chemicals. When used as substrates for mouse fibroblast cell growth, no cytotoxicity was observed and the cactus peel supported the attachment and proliferation of cells demonstrating potential to be used as a biomaterial for tissue engineering applications. Titanium and its alloys are used to make dental implants because of its low density, high strength, and corrosion resistance. This paper describes the development of a potential biomaterial Ti‐10Nb by powder metallurgy utilizing four different compaction pressures and analyses its microstructural, physical, mechanical, electrochemical, biological, and tribological behavior under various situations. The alloys were fabricated using four different compaction pressures, that is, 600, 650, 700, and 750 MPa, and sintered in a vacuum atmosphere at 1000°C for 1.5 h. The density of the samples was measured using Archimedes principle. X‐ray diffraction and scanning electron microscopy equipped with energy dispersive spectroscopy were used to investigate the phase composition and microstructure, and a profilometer was used to examine the surface roughness of various samples. Vickers hardness tester was used to evaluate hardness, and a universal testing machine was used for compression testing. Corrosion and wear behavior were examined using a potentiostat and a Bio‐Tribometer, respectively. This Ti‐10Nb alloys consist of α + β phase, and have 16% highest porosity in sample compacted at 600 MPa. The samples compacted at 750 MPa achieved highest hardness, yield strength, compressive strength, and elastic modulus of 450 ± 29.72 HV, 718.22 ± 16.37 MPa, 1543.59 ± 24.37 MPa, and 41.27 ± 3.29 GPa, respectively. In addition, it also possesses highest corrosion and wear resistance with lowest icorr of 0.3954 ± 0.008 μA/cm2 and wear volume of (31.25 ± 0.206) × 10−3 mm3. These results indicate that the developed alloys have a variety of desirable properties, including high hardness, adequate compressive strength, good corrosion and wear resistance, apatite‐forming capability, and a low elastic modulus, which is advantageous for avoiding stress shielding. Tissue engineering is a promising approach for generating or repairing living tissues. The development of innovative biomaterials for tissue engineering has the potential to address the unmet clinical needs in certain applications. However, before these biomaterials can be used in clinical settings, they must undergo preclinical testing to ensure safety and performance. The chicken chorioallantoic membrane (CAM) assay is a preferred screening tool for studying biocompatibility, angiogenesis, and inflammation induced by biomaterials owing to ethical and economic considerations. This CAM‐based platform increased the throughput of biomaterial testing for tissue engineering before in vivo testing. In this paper, we discuss the advantages of the CAM model. We also provided a step‐by‐step guide for implementing the CAM model in a research laboratory, along with tips and tricks for successfully running CAM assays. Finally, we present examples of biomaterials screened using CAM assays. CAM assay is a powerful in vivo model for assessing the angiogenic potential of tissue‐engineered scaffolds. Alginates are salts of alginic acid derived mainly from sea algae of the genus brown algae. They are also synthesized by some bacteria. They belong to negatively charged polysaccharides exhibiting some rheological properties. High plasticity and the ability to modify the structure are the reasons for their application in numerous industries. Moreover, when in contact with the living tissue, they do not trigger an immune response, and for this reason they are the most often tested materials for medical applications. The paper discusses the latest applications, including 3D bioprinting, drug delivery systems, and sorptive properties. Recognizing alginates as biomaterials, it emphasizes the necessity for precise processing and modification to industrialize them for specific uses. In recent years, cancerous cases have increased remarkably worldwide, and metastasis is the leading cause of death. Therefore, research on the early detection of cancer and metastasis has expanded to aid successful cancer treatment. Here in this paper, at the first step, an electrospun nanofibrous membrane (NFM) with a core‐shell structure was fabricated from PCL and HA to achieve cancer cell capturing (about 75% of cells). On the other hand, hyaluronic acid (HA)‐functionalized graphene quantum dots (GQDs) were used to detect captured cancer cells on NFM through the changes in photoluminescence intensity. Therefore, CD44 receptor‐HA interaction is the main principle used for both entrapment and detection of cancer cells. Results demonstrated the GQD‐HA fluorescent intensity of solution decreased through the increase of the captured cancer cell numbers on NFM, which is related to the more adsorption of GQD nanocomposites to the CD44 receptors. In contrast, this intensity for noncancerous cells was steady with any cell concentrations. This difference shows the system's remarkable selectivity and specificity, which can be crucial in fluorescent imaging for accurate cancer diagnosis. [ABSTRACT FROM AUTHOR]

This paper presents the results of an experimental and computational study of the effects of laser‐induced heating provided by magnetite nanocomposite structures that are being developed for the localized hyperthermic treatment of triple‐negative breast cancer. Magnetite nanoparticle‐reinforced polydimethylsiloxane (PDMS) nanocomposites were fabricated with weight percentages of 1%, 5%, and 10% magnetite nanoparticles. The nanocomposites were exposed to incident Near Infrared (NIR) laser beams with well‐controlled powers. The laser‐induced heating is explored in: (i) heating liquid media (deionized water and cell growth media [Leibovitz L15+]) to characterize the photothermal properties of the nanocomposites, (ii) in vitro experiments that explore the effects of localized heating on triple‐negative breast cancer cells, and (iii) experiments in which the laser beams penetrate through chicken tissue to heat up nanocomposite samples embedded at different depths beneath the chicken skin. The resulting plasmonic laser‐induced heating is explained using composite theories and heat transport models. The results show that the laser/nanocomposite interactions decrease the viability of triple‐negative breast cancer cells (MDA‐MB‐231) at temperatures in the hyperthermia domain between 41 and 44°C. Laser irradiation did not cause any observed physical damage to the chicken tissue. The potential in vivo performance of the PDMS nanocomposites was also investigated using computational finite element models of the effects of laser/magnetite nanocomposite interactions on the temperatures and thermal doses experienced by tissues that surround the nanocomposite devices. The implications of the results are then discussed for the development of implantable nanocomposite devices for localized treatment of triple‐negative breast cancer tissue via hyperthermia. Infection associated with tissue‐contacting biomedical devices is a compelling clinical problem initiated by the microbial colonization of the device surface. Among the possible sources of contaminating bacteria is the operating room (OR) itself, where viable bacteria in the atmosphere can sediment onto a device surface intraoperatively. We have developed an aerosolizing system that can reproducibly spray small quantities of aerosolized bacteria onto a surface to mimic OR contamination. This paper describes the design of the system and characterizes key aspects associated with its operation. The area density of sprayed bacteria is on the order of 102/cm2. Using titanium (Ti) alloy coupons as test substrates contaminated by staphylococci, we quantify the fraction of bacteria that are well adhered to the substrate, those that can be removed by sonication, and those that are not recovered after spraying. Despite the relatively low levels of surface contamination, we furthermore show that such a model is able to demonstrate a statistically significant reduction in colonization of Ti coupons modified by antimicrobial quaternary ammonium compounds relative to unmodified controls. This paper presents in vitro studies of the sustained release of Annona muricata leaf extracts (AME) from hybrid electrospun fibers for breast cancer treatment. Electrospun hybrid scaffolds were fabricated from crude AME extracts, poly(lactic‐co‐glycolic acid)/gelatin (PLGA/Ge) and pluronic F127. The physicochemical properties of the AME extract and scaffolds were studied. The antiproliferative effects of the scaffolds were also assessed on breast cancer (MCF‐7 and MDA‐MB‐231) and non‐tumorigenic breast (MCF10A) cell lines. Scanning electron microscope micrographs revealed a random network of micro‐ and submicron fibers. In vitro drug release profiles, governed by quasi‐Fickian diffusion at pH 7.4 and non‐Fickian super case II at pH 6.7, showed initial burst AME release from the PLGA/Ge‐AME and PLGA/Ge‐F127/AME fibers at pH 7.4, and burst release from PLGA/Ge‐F127/AME (not observed from PLGA/Ge‐AME) at pH 6.7. Then, a slower, sustained release of the remaining AME from the fibers, attributed to the onset of degradation of the PLGA/Ge backbone, was observed for the next 72 hr. The cumulative release of AME was 89.33 ± 0.73% (PLGA/Ge‐AME) and 51.17 ± 7.96% (PLGA/Ge‐F127/AME) at pH 7.4, and 9.27 ± 2.3% and 73.5 ± 4.5%, respectively, at pH 6.7. Pluronic F127 addition increased the drug loading capacity and prolonged the sustained AME release from the fibers. The released AME significantly inhibited the in vitro growth of the breast cancer cells more than the non‐tumorigenic cells, due to the induction of apoptosis, providing evidence for using pluronic F127‐containing electrospun fibers for sustained and localized AME delivery to breast cancer cells. The synergistic tumor therapy in single nanoplatform has always been the goal for high efficacy tumor treatment while still remains great challenge. This paper reports a versatile nanotheranostic platform enlisting magnetic iron oxide nanoparticles, polydopamine (PDA), gold nanocages (Au nanocage) and metal organic framework (MOF, MIL101‐NH2) in order to achieve synergistic chemothermal tumor therapy both in vitro and in vivo. The prepared magnetic photothermal nanoparticles (MPNPs) exhibit high drug loading capacity (31.34 mg/g), superior photo‐thermal capacity (11.5°C enhancement in 180 s), low bio‐toxicity, good magnetic resonance with a low dosage of 22 μg/g, as well as high antitumor efficacy in vivo. The international standard ISO 23317:2014 for the in vitro testing of inorganic biomaterials in simulated body fluid (SBF) uses TRIS buffer to maintain neutral pH. In our previous papers, we investigated the interaction of a glass–ceramic scaffold with TRIS and HEPES buffers. Both of them speeded up glass–ceramic dissolution and hydroxyapatite (HAp) precipitation, thereby demonstrating their unsuitability for the in vitro testing of highly reactive biomaterials. In this article, we tested MOPS buffer (3‐[N‐morpholino] propanesulfonic acid), another amino acid from the group of "Goods buffers". A highly reactive glass–ceramic scaffold (derived from Bioglass®) was exposed to SBF under static–dynamic conditions for 13/15 days. The kinetics and morphology of the newly precipitated HAp were studied using two different concentrations of (PO4)3− ions in SBF. The pH value and the SiIV, Ca2+, and (PO4)3− concentrations in the SBF leachate samples were measured every day (AAS, spectrophotometry). The glass–ceramic scaffold was monitored by SEM/EDS, XRD, WD‐XRF, and BET before and after 1, 3, 7, 11, and 13/15 days of exposure. As in the case of TRIS and HEPES, the preferential dissolution of the glass–ceramic crystalline phase (Combeite) was observed, but less intensively. The lower concentration of (PO4)3− ions slowed down the kinetics of HAp precipitation, thereby causing the disintegration of the scaffold structure. This phenomenon shows that the HAp phase was predominately generated by the presence of (PO4)3− ions in the SBF, not in the glass–ceramic material. This paper presents three families of semi‐interpenetrating polymer network (SIPN) hydrogels based on an ester‐based polyurethane (EBPU) and hydrophilic monomers: N,N‐dimethylacrylamide (NNDMA), N‐vinyl pyrrolidone (NVP) and acryloylmorpholine (AMO) as potential materials for keratoprosthesis, orthokeratology and mini‐scleral lens application. Hydrogels sheets were synthesized via free‐radical polymerization with methods developed in‐house. SIPN hydrogels were characterized for their equilibrium water content, mechanical and surface properties. Three families of optically clear SIPN‐based hydrogels have been synthesized in the presence of water with >10% of composition attributable to EBPU. Water contents of SIPN materials ranged from 30% to 70%. SIPNs with ≤15% EBPU of total composition showed little influence to mechanical properties, whereas >15% EBPU contributed significantly to an increase in material stiffness. In the hydrated state, SIPNs with ≤15% EBPU of total composition show little difference in polar component (γp) of surface free energy, whereas for >15% EBPU there is a decrease in γp. The EBPU SIPN hydrogels display complementary material properties for keratoprosthesis, orthokeratology, and mini‐scleral applications. © 2018 The Authors. There are a number of papers discussing medical and mechanical aspects of ventral hernia management. Despite intensive work on the problem understanding, recurrences of the sickness still happen too often. For that reason, new aspects of the problem must be considered. In this article, a change in the abdominal implant's stiffness is discussed, which is caused by cyclic loading. Such loading influence abdominal implant, for example, while patient is coughing or exercising. For the first time, this stiffness change is quantitatively described for a selected knitted mesh. The influence of mesh stiffness changes on repaired hernia persistence is studied. Then, ex vivo experiments on a repaired hernia model under cyclic pressure loading are performed. Finally, numerical simulations of the experiments are made in which stresses and forces in the system are calculated. The two following cases are considered. In the first case, the mesh has its baseline stiffness, and in the second case, the mesh is preconditioned by former loading; thus, it is stiffened. Reaction forces at the mesh fixation points appear to be approximately twofold higher in the second case than in the first case. That may be a reason for a fixation damage in operated hernia system. The results presented shed new light on the necessary strength of mesh fixation in the abdominal wall. In this paper, we reported the results of our efforts in developing DCPA/nanosilica composite orthopedic cement. It is motivated by the significances of DCPA and silicon in bone physiological activities. More specifically, this paper examined the effects of various experimental parameters on the properties of such composite cements. In this work, DCPA cement powders were synthesized using a microwave synthesis technique. Mixing colloidal nanosilica directly with synthesized DCPA cement powders can significantly reduce the washout resistance of DCPA cement. In contrast, a DCPA-nanosilica cement powder prepared by reacting Ca(OH)2, H3PO4 and nanosilica together showed good washout resistance. The incorporation of nanosilica in DCPA can improve compressive strength, accelerate cement solidification, and intensify surface bioactivity. In addition, it was observed that by controlling the content of NaHCO3 during cement preparation, the resulting composite cement properties could be modified. Allowing for the development of different setting times, mechanical performance and crystal features. It is becoming increasingly accepted that to understand and model the bacterial colonization and infection of abiotic surfaces requires the use of a biofilm model. There are many bacterial colonizations by at least two primary species, however this is difficult to model in vitro. This study reports the development of a simple mixed‐species biofilm model using strains of two clinically significant bacteria: Staphylococcus aureus and Pseudomonas aeruginosa grown on nanoporous polycarbonate membranes on nutrient agar support. Scanning electron microscopy revealed the complex biofilm characteristics of two bacteria blending in extensive extracellular matrices. Using a prototype wound dressing which detects cytolytic virulence factors, the virulence secretion of 30 single and 40 mixed‐species biofilms was tested. P. aeruginosa was seen to out‐compete S. aureus, resulting in a biofilm with P. aeruginosa dominating. In situ growth of mixed‐species biofilm under prototype dressings showed a real‐time correlation between the viable biofilm population and their associated virulence factors, as seen by dressing fluorescent assay. Abstract: Transplantation of autologous cultured melanocytes as cell suspension has been used for the treatment of vitiligo. The recipient site is often prepared by laser‐mediated dermabrasion. Such procedures encounter disadvantages including prolonged transplantation duration, unsecured cell adherence to lesional skin and potential scarring. To improve this, here we propose a method by preparing recipient sites before transplantation by psoralen and ultraviolet A (PUVA)‐induced sunburn followed by transplanting cells with a chitosan‐based melanocyte spheroid patch. We evaluated the method in nude mice. Application of methoxsalen‐soaked filter paper on skin for 30 min followed by ultraviolet A exposure induced controlled sunburn blisters in 2 days. Upon transplantation, the blister roof could be quickly peeled off by a waxing patch. The chitosan membrane on which melanocytes were precultured into multicellular spheroids was transplanted with cells facing the skin. The chitosan patch adhered well to skin and secured the contact of melanocytes with the recipient site. One day later, melanocyte spheroids already detached from the chitosan membrane and adhered to the recipient skin. Our results suggest that the combination of chitosan‐based melanocyte spheroid patch with epidermal ablation by PUVA‐induced sunburn reaction can be a feasible method to facilitate melanocyte transplantation. © 2018 Wiley Periodicals, Inc. Abstract: This paper studied the influence of two common sterilization techniques, ethylene oxide (EO) and gamma irradiation (GI), on the corrosion rate of four Mg‐based materials in CO2‐bicarbonate buffered Hanks’ solution. The four materials were: high‐purity (HP)‐Mg, ZE41, ultra‐high purity (XHP)‐Mg, and XHP‐ZX00. The corrosion rate was measured through mass loss (Pm) and hydrogen evolution (PH). Two‐way analysis of variance (ANOVA) was conducted to assess the effect of the sterilization techniques on the corrosion rates across the four materials. The ANOVA analyzed the variables of (1) material, (2) sterilization condition (EO, GI, and an unsterilized control group), and (3) the interaction between these two independent variables. Neither sterilization technique (EO and GI) significantly influenced the corrosion rate as measured by Pm (p < 0.84) nor PH (p < 0.08). This result was consistent across the four materials tested, as there was no interaction between the test variables of material and sterilization condition for Pm (p < 0.49) or PH (p < 0.27). As neither EO nor GI influenced the corrosion rates, either of these techniques warrants consideration for use on Mg‐based medical implants and devices. © 2017 Wiley Periodicals, Inc. Abstract: To systematically review the literature to analyze the current trends and future perspectives of dental pulp capping materials through an analysis of scientific and technological data. This study is reported in accordance with the PRISMA Statement. Nine databases were screened: PubMed (MedLine), Lilacs, IBECS, BBO, Web of Science, Scopus, SciELO, Google Scholar, and The Cochrane Library. Additionally, the following patent applications were searched online in Questel Orbit (Paris, France), USPTO, EPO, JPO, INPI, and Patentscope databases. A total of 716 papers and 83 patents were included. Calcium hydroxide was the main type of material studied, especially for direct pulp capping, followed by MTA. Patents related to adhesives or resins increased from 1998 e 2008, while in the last years, a major increase was observed in bioactive materials (containing bioactive proteins), materials derived from MTA (calcium silicate, calcium phosphate and calcium aluminate‐based cements) and MTA. It was possible to obtain a scientific and technological overview of pulp capping materials. MTA has shown favorable results in vital pulp therapy that seem to surpass the disadvantages of calcium hydroxide. Recent advances in bioactive materials and those derived from MTA have shown promising results that could improve biomaterials used in vital pulp treatments. © 2017 Wiley Periodicals, Inc. The aim of this paper is to quantify the adhered fibronectin (FN; by adsorption and/or grafting) and the exposure of its cell adhesive motifs (RGD and FNIII7-10) on poly(ethyl acrylate) (PEA) copolymers whose chemical composition has been designed to increase wettability and to introduce acid functional groups. FN was adsorbed to PEA, poly(ethyl acrylate- co-hydroxyethyl acrylate), poly(ethyl acrylate- co-acrylic acid), and poly(ethyl acrylate- co-methacrylic acid) copolymers, and covalently cross-linked to poly(ethyl acrylate- co-acrylic acid) and poly(ethyl acrylate- co-methacrylic acid) copolymers. Amount of adhered FN and exhibition of RGD and FNIII7-10 fragments involved in cell adhesion were quantified with enzyme-linked immunosorbent assay tests. Even copolymers with a lower content of the hydrophilic component showed a decrease in water contact angle. In addition, FN was successfully fixed on all surfaces, especially on the hydrophobic surfaces. The importance of decellularized medical products has significantly increased during the last years. In this paper, we evaluated the effects of selected physical and procedural decellularization (DC) factors with the aim to systematically assess their influence on DC results. 72 porcine aortic walls (AW) were divided into three groups and exposed to a DC solution for 4 h and 8 h, either continuously or in repeated cycles. The AW were rocked (90bpm), whirled (10 l/min), sonicated (120W, 45 kHz) or exposed to a combination of these treatments, followed by 10 washing cycles. Defining successful DC as removal of nuclei while keeping an intact extracellular matrix (ECM), we equalized the efficiency to the penetration depth (PD), obtained by DAPI fluorescence and H&E staining. Additionally, we performed scanning electron microscopy (SEM), Pentachrome and Picrosirius-Red staining. Results showed that significantly higher DC depths are achieved on outer compared to inner surfaces (61 ± 7%; p < 0.001). Furthermore, the PD showed a high time dependency for all samples. An international standard (ISO: 23317:2014) exists for the in vitro testing of inorganic biomaterials in simulated body fluid (SBF). This standard uses TRIS buffer to maintain neutral pH in SBF, but in our previous paper, we showed that the interaction of a tested glass-ceramic material with TRIS can produce false-positive results. In this study, we evaluated whether the HEPES buffer, which also belongs to the group of Good´s buffers, would be more suitable for SBF. We compared its suitability in two media: SBF with HEPES and demineralized water with HEPES. The tested scaffold (45S5 bioactive glass-based) was exposed to the media under a static-dynamic arrangement (solutions were replaced on a daily basis) for 15 days. Leachate samples were collected daily for the analysis of Ca2+ ions and Si (AAS), (PO4)3- ions (UV-VIS), and to measure pH. The glass-ceramic scaffold was analyzed by SEM/EDS, XRD, and WD-XRF before and after 0.3, 1, 3, 7, 11, and 15 days of exposure. Our results confirmed the rapid selective dissolution of the glass-ceramic crystalline phase ( Combeite) containing Ca2+ ions due to the presence of HEPES, hydroxyapatite supersaturation being reached within 24 h in both solutions. These new results suggest that, like TRIS, HEPES buffer is not suitable for the in vitro testing of highly reactive inorganic biomaterials (glass, glass-ceramics). Background: Due to degradation and metal dissolution during articulation of metal joint replacements the chemical periprosthetic environment may change. The aim was to establish whether metal replacements cause the local changes in pH and elevated metal concentrations. Methods: pH was measured on samples from 167 patients: native hip and knee osteoarthritic joints, joints with hip and knee replacements revised for aseptic or septic reasons. pH of synovial fluid and periprosthetic tissue was measured perioperatively using a microelectrode and pH indicator papers for removed metal components. Metal concentrations were measured in 21 samples using inductively coupled plasma mass spectrometry. Results: The mean pH value of synovial fluid at native osteoarthritic joints ( n = 101) was 7.78 ± 0.38. The mean pH value of synovial fluid at revision aseptic operation ( n = 58) was 7.60 ± 0.31, with statistically significant difference ( p = 0.002) compared to native osteoarthritic joints. The mean pH value of synovial fluid at revision septic operation ( n = 8) was 7.55 ± 0.25, with statistically significant difference ( p = 0.038) compared to native osteoarthritic joints. Measurements in tissue and at stems were not reliable. In the majority of samples taken at revision increased levels of cobalt and chromium were measured. Conclusion: A small but statistically significant difference was observed in the pH of synovial fluid between natural joints with degenerative diseases and joints treated with metal replacements. Objectives The release of magnesium ions (Mg2+) from titanium surfaces has been shown to boost the initial biological response of peri-implant bone and to increase the biomechanical strength of osseointegration. The objective of the present paper was to investigate if the initial improvement in osseointegration would influence the bone remodeling also during the maturation stage of bone healing. Methods: Titanium implants were coated with mesoporous titania layers and either loaded with Mg2+ (test group) or left untreated (control group). The implants were inserted in the tibiae of 10 New Zealand White rabbits. Osseointegration was assessed after 6 weeks by means of biomechanical testing (RTQ), non-decalcified histology and histomorphometry (BIC%, BA%, NBA%). The expression of genes involved in the bone formation and remodeling was quantified using qPCR. Results: Mg2+ releasing mesoporous titania coatings showed, on average, higher removal torques and histomorphometrical outcomes (RTQ: 17.2 Ncm vs. 15 Ncm; BIC: 38.8% vs. 32.1%; BA%: 71.6% vs. 64%; NBA% 62.5% vs. 54% for the tests vs the controls); however, the differences were not statistically significant. Three osteogenic markers, osteocalcin (OC), collagen 1 alpha 1 (COL1A1), and alkalin phosphatase (ALPL), were respectively 2-fold, 1.53-fold, and 1.13-fold up-regulated in the control group compared to the test. The expression of COL1A1 was particularly high in both groups, while the biomarkers for remodeling and inflammation showed a low expression in both groups. Membranes with zeolites are promising for performing blood dialysis because zeolites can eliminate uremic toxins through molecular sieving. Although the size and the shape of zeolite particles can potentially influence the performance of the membranes with respect of creatinine uptake level, it is not clear what sizes and shapes lead to better performance. In this paper, we carry out experiments to answer this question. Spherical microparticle 840, spherical nanoparticle P-87 and rod-like nanoparticle P-371 zeolites were chosen to be used in all the experiments. Their creatinine uptake levels were first measured as powders in creatinine solutions with different concentrations, volumes and adsorption times. Then, nanofibrous membranes with zeolites were electrospun and their ability to adsorb creatinine was measured and compared against their respective powders' creatinine uptake level. The experiment shows that the zeolites have similar creatinine uptake ability as powders. However, they have significantly different creatinine uptake ability after being incorporated inside the membranes. Spherical microparticle 840 in the membrane presented the best creatinine uptake ability, at 8957 µg g−1, which was half of its powders'. On the other hand, P-87 presented largely decreased, while P-371 presented even lower creatinine uptake ability in membranes when compared to respective powders'. In this paper, we report the in vitro biocompatibility and cellular interactions of a chitosan/dextran-based (CD) hydrogel and its components as determined by mutagenicity, cytotoxicity, cytokine/chemokine response, and wound healing assays. The CD hydrogel, developed for postsurgical adhesion prevention in ear, nose, and throat surgeries, was shown by previously published experiments in animal and human trials to be effective. The hydrogel was synthesized from the reaction between succinyl chitosan (SC) and oxidized dextran (DA). Cytotoxicity was assessed in an xCELLigence system and cytokine/chemokine responses were measured by ELISA in human macrophage, nasopharyngeal epithelial, and dermal fibroblast cells. A wound healing model utilized nasopharyngeal epithelial cells. CD hydrogel and DA were nonmutagenic in the Ames test. CD hydrogel showed moderate cytotoxicity for the cell lines, DA being the cytotoxic component. Some inhibition of wound healing occurred due to the cytotoxic nature of DA. Cells cultured with CD hydrogel showed no increase in TNF-α, IL-10, and IL-8 levels. It is hypothesized that the cytotoxicity of DA is moderated when reacted with SC and that CD hydrogel inhibits unwanted fibroblastic invasion preventing scarring and adhesions. Together with the previously published human and animal trial data, the results indicate CD hydrogel is biocompatible in the setting of endoscopic sinus surgery. This study evaluated the cytotoxicity of experimental adhesive systems (EASs) on odontoblast-like cells. Paper discs ( n = 132) were impregnated with 10 µL of each EAS-R1, R2, R3, R4, and R5 (in an ascending order of hydrophilicity), followed by photoactivation. R1 and R2 are nonsolvated hydrophobic blends, R3 represents a simplified etch-and-rinse adhesive system, and R4 and R5 represent simplified self-etch adhesive systems. Discs were immersed in Dulbecco's modified Eagle's medium for 24 h to obtain eluates applied on MDPC-23 cell cultures. No material was applied on discs used as control (R0). Cell viability [3-(4,5-dimethythiazol-2-yl)−2,5-diphenyl tetrazolium bromide assay], total protein (TP) production, alkaline phosphatase (ALP) activity, type of cell death, and degree of monomer conversion Fourier transform infrared (%DC-FTIR) were evaluated. Data were analyzed by Kruskal-Wallis and Mann-Whitney tests (α = 0.05). Considering R0 (control) as having 100% of cell viability, R1, R2, R3, R4, and R5 reduced the metabolic activity of cells by 36.4, 3.1, 0.2, 21.5, and 65.7%, respectively, but only R1 and R5 differed from R0. Comparing with R0, lower TP production was observed for R1, R4, and R5, while ALP activity decreased for R1 and R5. Necrotic cell death was predominant for all EASs, but only R1, R4, and R5 differed from R0. Only R5 presented a different apoptotic cell death ratio from R0. R1 presented the lowest %DC (ca. 37%), whereas R4 and R5 presented the highest (ca. 56%). Peptides containing the sequence of arginine-glycine-aspartate (RGD), a famous adhesion moiety, can specifically conjugate integrins in cell membranes, and are usually applied to enhance cell adhesion after linking to solid substrates in tissue engineering or to nanoparticles in targeting delivery. This paper reveals, however, that free RGD peptides can assist in preventing tissue adhesion by blocking focal adhesion between cells and surfaces of barrier devices. In order to avoid a rapid peptide loss after straightforward injection of a peptide solution, we employed a thermosensitive injectable hydrogel composed of a biodegradable block copolymer poly(ε-caprolactone- co-lactide)-poly(ethylene glycol)-poly(ε-caprolactone- co-lactide) (PCLA-PEG-PCLA) to encapsulate peptides cyclo(-RGDfK-). A sustainable release for one week was achieved in vitro. The rabbit model of sidewall defect and bowel abrasion was selected to examine the in vivo anti-adhesion efficacy. It reveals a significant reduction of postoperative peritoneal adhesion in the group of RGD-loaded PCLA-PEG-PCLA hydrogels. This study evaluated the influence of organic acids present in the oral biofilm on the microtensile bond strength (μTBS) of adhesive systems to human dentin. Sixty occlusal dentin surfaces were wet ground with 600 grit SiC abrasive paper and divided into four groups according to the adhesive systems: Scotchbond Multipurpose (SMP), Adper Single Bond 2, Adper Scotchbond SE (ASE), and Clearfill SE Bond (CSE). After the adhesive systems were applied, a block of resin composite was built up on the dentin surfaces. After 24 h storage in distilled water at 37°C, the teeth were perpendicularly cut to obtain beams (1 mm2). For each adhesive system, the beams were divided into three groups according to storage media: artificial saliva (AS); propionic acid (PA), and lactic acid (LA). After 7 days storage at 37°C, the beams were submitted to μTBS testing. The μTBS ranged from 36.0 ± 1.6 (ASE-PA) to 52.5 ± 1.2 (CSE-AS). For all adhesive systems, the μTBS values after storage in PA were lower than those in AS. Except for the SMP, the values of μTBS after storage in LA were lower than those in AS. The adhesive ASE presented the lowest values of μTBs in the three media. Electrospun fibrous mats have gained popularity in bioengineering over the past decade, but few papers detail their degradative mechanisms. To address this, blends of hydrophobic poly(ε-caprolactone) (PCL) and hydrophilic PGA-PCL-PGA triblock copolymer were electrospun into aligned fibrous mats to assess the copolymers' mechanical and degradative properties. Increased hydrophilic triblock content led to enhanced morphological uniformity of fiber, tightening of fiber diameters, increased storage and Young's modulus, and decreased elongation. The corresponding decrease in hydrophobic PCL content led to faster hydrolytic degradation rate, as reflected by enhanced decrease in mass, molecular weight, and modulus loss at 25, 37, and 45°C. The activation energy for hydrolytic degradation for 15:85 PCL: triblock copolymer was approximately half that of 85:15 PCL: triblock copolymer. Detailed examination of fiber morphology and crystallinity revealed initial surface erosion followed by the evolution of crystalline lamellar stacks and bulk degradation at 37°C. Because of the high surface to volume and short diffusion length scale of the small diameter fibers, surface and bulk degradation may both contribute to the hydrolytic degradative behavior of these electrospun fibrous mats. Statement of Problem: Clinicians use occlusal indicators to identify tooth contacts. However, the reliability of these indicators has been questioned. At times occlusal contacts are not identified or false positive occlusal contacts are observed. Purpose of Study: This study was designed to compare the number and size of occlusal indicator marks from both thick and thin occlusal indicator materials with different loads. Materials and Methods: Ivorine casts were articulated and mounted on an Hana Mate nonadjustable articulator. Loads of 100N, 150N, and 200N were applied with Accufim (25 μm thick, Parkell, Farmingdale, NY) and Articulating Paper (60 μm thick, G.E. Rudischauer Dental Articulating Paper, Brooklyn, NY) as the occlusal indicators. A fresh piece of indicator was used for each trial. Comparisons were made of the number and size of the contacts for both the thick and thin occlusal indicators at the different loads. Results: Observation of the marks recorded with the thicker occlusal indicator demonstrated both a greater number marks and a larger size to the marks when compared to the thinner Accufilm, p ≤ 0.02–0.0001. Silane chemistry has emerged as a powerful tool for surface modification, offering a versatile means to enhance the properties of various substrates, such as dental implant abutment materials. In this study, we investigated the stability of the 3‐aminopropyldiisopropylethoxysilane (APDS) layer on yttria‐partially stabilized zirconia (Y‐TZP) surfaces after mechanical, acid, and thermal treatment in order to simulate fluctuations within the oral cavity. To accomplish that, the viability of human gingival fibroblasts on APDS‐modified surfaces after applied treatment strategies was assessed by live/dead staining. Moreover, the hydrolysis stability and enzymatic degradation resistance of crosslinked fibronectin to the APDS layer was examined by immunostaining and western blot. The results revealed that the applied modifications were not affected by the different treatment conditions and could withstand the fluctuations in the oral cavity. Furthermore, crosslinked fibronectin on silanized Y‐TZP was stable against hydrolysis over 21 days and enzymatic degradation. Recent studies show good osteoinductive properties of polyurethanes modified with polyhedral oligomeric silsesquioxanes (POSS). In this work, three types of POSS; propanediolisobutyl‐POSS (PHI‐POSS), disilanolisobutyl‐POSS (DSI‐POSS), and octahydroxybutyl‐POSS (OCTA‐POSS) were chemically incorporated into linear polyurethane based on an aliphatic isocyanate, hexamethylene diisocyanate (HDI), to obtain new nanohybrid PU‐POSS materials. The full conversion of POSS was confirmed by Fourier transform infrared spectroscopy (FTIR‐ATR) spectra of the model reactions with pure HDI. The materials obtained were investigated by FTIR, SEM‐EDS, and DSC. The DSC studies showed the thermoplasticity of the obtained materials and apparently good recovery. 30‐day immersion in SBF (simulated body fluid) revealed an increase in the rate of deposition of hydroxyapatite (HAp) for the highest POSS loadings, resulting in thick layers of hydroxyapatite (~60–40 μm), and the Ca/P ratio 1.67 (even 1.785). The structure and properties of the inorganic layer depend on the type of POSS, the number of hard segments, and those containing POSS, which can be tailored by changing the HDI/poly(tetramethylene glycol) (PTMG) ratio. Furthermore, the obtained composites revealed good biocompatibility, as confirmed by cytotoxicity tests conducted on two cell lines; normal human dermal fibroblasts (NHDF) and primary human osteoblasts (HOB). Adherent cells seeded on the tested materials showed viability even after a 48‐h incubation. After this time, the population of viable, and proliferating cells exceeded 90%. To obtain bone allografts that are safe for transplantation, several processing steps for decellularization and decontamination have to be applied. Currently available processing methods, although well‐established, may interfere with the biomechanical properties of the bone. High hydrostatic pressure (HHP) is known to devitalize tissues effectively while leaving the extracellular matrix intact. However, little is known about the inactivation of the contaminating microorganisms by HHP. This study aims to investigate the ability of high‐pressure decontamination and to establish a treatment protocol that is able to successfully inactivate microorganisms with the final goal to sterilize bone specimens. Using Escherichia coli (E. coli) as a model organism, HHP treatment parameters like temperature and duration, pressurization medium, and the number of treatment cycles were systematically adjusted to maximize the efficiency of inactivating logarithmic and stationary phase bacteria. Towards that we quantified colony‐forming units (cfu) after treatment and investigated morphological changes via Field Emission Scanning Electron Microscopy (FESEM). Additionally, we tested the decontamination efficiency of HHP in bovine cancellous bone blocks that were contaminated with bacteria. Finally, two further model organisms were evaluated, namely Pseudomonas fluorescens as a Gram‐negative microorganism and Micrococcus luteus as a Gram‐positive representative. A HHP protocol, using 350 MPa, was able to sterilize a suspension of stationary phase E. coli, leading to a logarithmic reduction factor (log RF) of at least −7.99 (±0.43). The decontamination of bone blocks was less successful, indicating a protective effect of the surrounding tissue. Sterilization of 100% of the samples was achieved when a protocol optimized in terms of treatment temperature, duration, pressurization medium, and number and/or interval of cycles, respectively, was applied to bone blocks artificially contaminated with a suspension containing 104 cfu/mL. Hence, we here successfully established protocols for inactivating Gram‐negative model microorganisms by HHP of up to 350 MPa, while pressure levels of 600 MPa were needed to inactivate the Gram‐positive model organism. Thus, this study provides a basis for further investigations on different pathogenic bacteria that could enable the use of HHP in the decontamination of bone grafts intended for transplantation. This in vivo study evaluated the bone healing response around endosteal implants with varying surface topography/chemistry in a preclinical, large transitional model induced with metabolic syndrome (MS) and type‐2 diabetes mellitus (T2DM). Fifteen Göttingen minipigs were randomly distributed into two groups: (i) control (normal diet, n = 5) and (ii) O/MS (cafeteria diet for obesity induction, n = 10). Following obesity induction, five minipigs from the obese/metabolic syndrome (O/MS) group were further allocated, randomly, into the third experimental group: (iii) T2DM (cafeteria diet + streptozotocin). Implants with different surface topography/chemistry: (i) dual acid‐etched (DAE) and (ii) nano‐hydroxyapatite coating over the DAE surface (NANO), were placed into the right ilium of the subjects and allowed to heal for 4 weeks. Histomorphometric evaluation of bone‐to‐implant contact (%BIC) and bone area fraction occupancy (%BAFO) within implant threads were performed using histomicrographs. Implants with NANO surface presented significantly higher %BIC (~26%) and %BAFO (~35%) relative to implants with DAE surface (%BIC = ~14% and %BAFO = ~28%, p <.025). Data as a function of systemic condition presented significantly higher %BIC (~28%) and %BAFO (~42%) in the control group compared with the metabolically compromised groups (O/MS: %BIC = 14.35% and %BAFO = 26.24%, p <.021; T2DM: %BIC = 17.91% and %BAFO = 26.12%, p <.021) with no significant difference between O/MS and T2DM (p >.05). Statistical evaluation considering both factors demonstrated significantly higher %BIC and %BAFO for the NANO surface relative to DAE implant, independent of systemic condition (p <.05). The gain increase of %BIC and %BAFO for the NANO compared with DAE was more pronounced in O/MS and T2DM subjects. Osseointegration parameters were significantly reduced in metabolically compromised subjects compared with healthy subjects. Zirconia implants are gaining attention as a viable alternative to titanium implants due to their comparable osseointegration development, improved soft tissue adaptation, and enhanced aesthetics. An encouraging avenue for improving zirconia implant properties involves the potential application of bioactive coatings to their surfaces. These coatings have shown potential for inducing hydroxyapatite formation, crucial for bone proliferation, and improving implant mechanical properties. This study aimed to evaluate the effect of coating zirconia implants with two bioactive glasses, 45S5 and BioK, on osteogenesis in vitro and osseointegration in vivo. Zirconia samples and implants were prepared using Zpex zirconia powder and blocks, respectively. The samples were divided into three groups: polished zirconia (ZRC), zirconia coated with 45S5 bioglass (Z + 45S5), and zirconia coated with BioK glass (Z + BK). Coatings were applied using a brush and sintered at 1200°C. Chemical analysis of the coatings was carried out using x‐ray diffraction and Fourier Transform Infrared Spectroscopy. Surface topography and roughness were characterized using scanning electron microscopy and a roughness meter. In vitro experiments used mesenchymal cells from Wistar rat femurs, and the coated zirconia implants were found to promote cell viability, protein synthesis, alkaline phosphatase activity, and mineralization, indicating enhanced osteogenesis. In vivo experiments with 18 rats showed positive results for bone formation and osseointegration through histological and histomorphometric analysis and a push‐out test. Improvement of mechanical properties of injectable tissue engineering scaffolds is a current challenge. The objective of the current study is to produce a highly porous injectable scaffold with improved mechanical properties. For this aim, cellulose nanocrystals‐reinforced dual crosslinked porous nanocomposite cryogels were prepared using chemically crosslinked methacrylated gelatin (GelMA) and ionically crosslinked hyaluronic acid (HA) through the cryogelation process. The resulting nanocomposites showed highly porous structures with interconnected porosity (>90%) and mean pore size in the range of 130–296 μm. The prepared nanocomposite containing 3%w/v of GelMA, 20 w/w% of HA, and 1%w/v of CNC showed the highest Young's modulus (10 kPa) and excellent reversibility after 90% compression and could regain its initial shape after injection by a 16‐gauge needle in the aqueous media. The in vitro results demonstrated acceptable viability (>90%) and migration of the human chondrocyte cell line (C28/I2), and chondrogenic differentiation of human adipose stem cells. A two‐month in vivo assay on a rabbit's ear model confirmed that the regeneration potential of the prepared cryogel is comparable to the natural autologous cartilage graft, suggesting it is a promising alternative for autografts in the treatment of cartilage defects. This study emphasizes the development of a multifunctional biomaterial ink for wound healing constructs. The biomaterial ink benefits from Aloe vera's intrinsic biocompatible, biodegradable, antioxidant, antimicrobial, anti‐inflammatory, and immunomodulatory attributes, thus alleviating the need for supplementary substances employed to combat infections and stimulate tissue regeneration. Moreover, this biomaterial ink seeks to address the scarcity of standardized printable materials possessing adequate biocompatibility and physicochemical properties, which hinder its widespread clinical adoption. The biomaterial ink was synthesized via ionic crosslinking to enhance its rheological and mechanical characteristics. The findings revealed that Aloe vera substantially boosted the hydrogel's viscoelastic behavior, enabling superior compressive modulus and the extrusion of fine filaments. The bioprinted constructs exhibited desirable resolution and mechanical strength while displaying a porous microstructure analogous to the native extracellular matrix. Biological response demonstrated no detrimental impact on stem cell viability upon exposure to the biomaterial ink, as confirmed by live/dead assays. These outcomes validate the potential of the developed biomaterial ink as a resource for the bioprinting of wound dressings that effectively foster cellular proliferation, thereby promoting enhanced wound healing by leveraging Aloe vera's inherent properties. Silicone rubber (SR), a common medical‐grade polymer used in medical devices, has previously been modified for nitric oxide (NO) releasing capabilities. However, the effects of material properties such as film thickness on NO release kinetics are not well explored. In this study, SR is used in the first analysis of how a polymer's thickness affects the storage and uptake of an NO donor and subsequent release properties. Observed NO release trends show that a polymer's thickness results in tunable NO release. Nontraumatic osteonecrosis of the femoral head (ONFH) is a refractory condition that commonly results in femoral head collapse and degenerative arthritis of the hip. In the early stages, surgical procedures for hip preservation, including core decompression (CD), have been developed to prevent progressive collapse of the femoral head. Optimization of bone regeneration and biological augmentation may further enhance the therapeutic efficacy of CD for ONFH. Thus, combining CD with cell‐based therapy has recently been proposed. In fact, patients treated with cell‐based therapy using autologous bone marrow concentrate demonstrate improved survivorship of the femoral head, compared with conventional CD alone. Preclinical research studies to investigate adjunctive therapies for CD often utilize the rabbit model of corticosteroid‐induced ONFH. Mesenchymal stem cells (MSCs) are known to promote osteogenesis and angiogenesis, and decrease inflammation in bone. Local drug delivery systems have the potential to achieve targeted therapeutic effects by precisely controlling the drug release rate. Scaffolds can provide an osteoconductive structural framework to facilitate the repair of osteonecrotic bone tissue. We focused on the combination of both cell‐based and scaffold‐based therapies for bone tissue regeneration in ONFH. We hypothesized that combining CD and osteoconductive scaffolds would provide mechanical strength and structural cell guidance; and that combining CD and genetically modified (GM) MSCs to express relevant cytokines, chemokines, and growth factors would promote bone tissue repair. We developed GM MSCs that overexpress the anti‐inflammatory, pro‐reconstructive cytokines platelet‐derived growth factor‐BB to provide MSCs with additional benefits and investigated the efficacy of combinations of these GM MSCs and scaffolds for treatment of ONFH in skeletally mature male New Zealand white rabbits. In the future, the long‐term safety, efficacy, durability, and cost‐effectiveness of these and other biological and mechanical treatments must be demonstrated for the patients affected by ONFH. Bifunctional tissue engineering constructs promoting osteogenesis and angiogenesis are essential for bone regeneration. Metal ion‐incorporated scaffolds and fibrin encapsulation attract much attention due to low cost, nontoxicity, and tunable control over ion and growth factor release. Herein, we investigated the effect of Cu.nHA/Cs/Gel scaffold and fibrin encapsulation on osteogenic and angiogenic differentiation of Wharton's jelly mesenchymal stem cells (WJMSCs) in vitro and in vivo. Cu‐laden scaffolds were synthesized using salt leaching/freeze drying and were characterized using standard techniques. WJMSCs were isolated from the human umbilical cord and characterized. WJMSCs with or without encapsulating in fibrin were seeded onto scaffolds, followed by differentiating into the osteogenic lineage for 7 and 21 days. Osteogenic and angiogenic differentiation were evaluated using real‐time polymerase chain reaction, western blot, and Alizarin red staining. Then, scaffolds were implanted into critical‐sized calvarial bone defects in rats and histological assessments were performed using hematoxylin/eosin, Masson's trichrome, and CD31 immunohistochemical staining at 4 and 12 weeks. The scaffolds had good physicochemical and biological characteristics suitable for cell attachment and growth. Cu and fibrin increased the expression of ALP, RUNX2, OCN, COLI, VEGF, and HIF1α in differentiated WJMSCs. Implanted scaffolds were also biocompatible and were integrated well with the host tissue. Increased collagen condensation, mineralization, and blood vessel formation were observed in Cu‐laden scaffolds. The fibrin‐encapsulated groups showed the highest collagen and cell densities, immune cell infiltration, and bone trabeculae. CD31‐positive cell population increased with fibrin encapsulation and seeding onto Cu‐laden scaffolds. Adding Cu to scaffolds and encapsulating cells in fibrin are promising methods that guide osteogenesis and angiogenesis cellular signaling, leading to better bone regeneration. Biomedical alloys, like many engineering alloys, have chemical or physical heterogeneities at the surface, and such heterogeneities can potentially act as sites for pit initiation. Alloys of particular interest are 316/316L (and 316LVM) stainless steel, nitinol, and CoCr alloys. This review focuses on the sites—generally inclusions—that have been associated with pitting in various studies of biomedical alloys in simulated physiological solutions. The effect of these sites is discussed in relation to factors such as type and size. For both 316/316L stainless steel and nitinol, pitting has been found to initiate at two different types of inclusions: sulfide and oxide inclusions in stainless steel, and carbide and oxide inclusions in nitinol. Sulfide inclusions tend to be the predominant sites for pitting on 316/316L stainless steel, while there is some evidence to suggest that carbide inclusions may be more effective than oxide inclusions for pitting on nitinol. CoCrMo alloys differ from the other two alloys in that, although particles can be present in the form of carbides, the carbides typically do not provide sites for pit initiation except possibly for alloys with a high‐C content, certain heat treatments, and when anodically polarized to high potentials. CoNiCrMo differs further in that TiN inclusions can be present in the vicinity of pits and might be associated with them, but irrespective of the initiation site, any pits are unlikely to grow because of repassivation. Strontium (Sr) has important functions in bone remodeling. Incorporating strontium‐doped α‐calcium sulfate hemihydrate (SrCSH) into poly(lactic‐co‐glycolic acid) (PLGA) fibrous scaffolds were expected to increase its bio‐activity and provide a potential material for bone tissue engineering. In the present study, Sr‐containing aligned PLGA/SrCSH fibrous scaffolds similar to the architecture of natural bone were prepared via wet spinning. CCK‐8 assay revealed that Sr‐containing scaffolds possessed better bioactivity and supported favorable cell growth effectively. The aligned PLGA/SrCSH fibers exerted a contact effect on cell attachment, inducing regular cell alignment and influencing a series of cell behaviors. Releasing of high concentration Sr from a‐PLGA/SrCSH scaffolds could induce high expression levels of BMP‐2, increase ALP activity and upregulate RUNX‐2 expression, and further promote the expressions of COL‐I and OCN and the maximum mineralization. This study demonstrated that Sr and ordered structure in a‐PLGA/SrCSH fibrous scaffolds could synergistically enhance the osteogenic differentiation of umbilical cord mesenchymal stem cells (UCMSCs) by regulating cell arrangement and expressions of osteogenic genes. Sterilization of structural bone allografts is a critical process prior to their clinical use in large cortical bone defects. Gamma irradiation protocols are known to affect tissue integrity in a dose dependent manner. Alternative sterilization treatments, such as supercritical carbon dioxide (SCCO2), are gaining popularity due to advantages such as minimal exposure to denaturants, the lack of toxic residues, superior tissue penetration, and minor impacts on mechanical properties including strength and stiffness. The impact of SCCO2 on the fracture toughness of bone tissue, however, remains unknown. Here, we evaluate crack initiation and growth toughness after 2, 6, and 24 h SCCO2‐treatment using Novakill™ and ethanol as additives on ~11 samples per group obtained from a pair of femur diaphyses of a canine. All mechanical testing was performed at ambient air after 24 h soaking in Hanks' balanced salt solution (HBSS). Results show no statistically significant difference in the failure characteristics of the Novakill™‐treated groups whereas crack growth toughness after 6 and 24 h of treatment with ethanol significantly increases by 37% (p =.010) and 34% (p =.038), respectively, compared to an untreated control group. In contrast, standard 25 kGy gamma irradiation causes significantly reduced crack growth resistance by 40% (p =.007) compared to untreated bone. FTIR vibrational spectroscopy, conducted after testing, reveals a consistent trend of statistically significant differences (p <.001) with fracture toughness. These trends align with variations in the ratios of enzymatic mature to immature crosslinks in the collagen structure, suggesting a potential association with fracture toughness. Additional Raman spectroscopy after testing shows a similar trend with statistically significant differences (p <.005), which further supports that collagen structural changes occur in the SCF‐treated groups with ethanol after 6 and 24 h. We engineered an in vitro model of bioartificial 3D bone organoid consistent with an anatomical and vascular microenvironment common to mammalian flat and short bones. To achieve this, we chose the decellularized–decalcified matrix of the adult male rat scapula, implemented with the reconstruction of its intrinsic vessels, obtained through an original intravascular perfusion with polylevolactic (PLLA), followed by coating of the PLLA‐fabricated vascularization with rat tail collagen. As a result, the 3D bone and vascular geometry of the native bone cortical and cancellous compartments was reproduced, and the rat tail collagen–PLLA biomaterial could in vitro act as a surrogate of the perivascular extracellular matrix (ECM) around the wall of the biomaterial‐reconstituted cancellous vessels. As a proof‐of‐concept of cell compatibility and site‐dependent osteoinductive properties of this bioartificial 3D construct, we show that it in vitro leads to a time‐dependent microtopographic positioning of rat mesenchymal stromal cells (MSCs), initiating an osteogenic fate in relation to the bone compartment. In addition, coating of PLLA‐reconstructed vessels with rat tail collagen favored perivascular attachment and survival of MSC‐like cells (mouse embryonic fibroblasts), confirming its potentiality as a perivascular stroma for triggering competence of seeded MSCs. Finally, in vivo radiographic topography of bone lesions in the human jaw and foot tarsus of subjects with primary osteoporosis revealed selective bone cortical versus cancellous involvement, suggesting usefulness of a human 3D bone organoid engineered with the same principles of our rat organoid, to in vitro investigate compartment‐dependent activities of human MSC in flat and short bones under experimental osteoporotic challenge. We conclude that our 3D bioartificial construct offers a reliable replica of flat and short bones microanatomy, and promises to help in building a compartment‐dependent mechanistic perspective of bone remodeling, including the microtopographic dysregulation of osteoporosis.
