Defense against oxidative stress in mammals includes the regeneration of the major thiol reductants glutathione and thioredoxin by glutathione reductase and thioredoxin reductase (TrxR), respectively. In contrast, Drosophila, and possibly insects in general, lacks glutathione reductase and must rely solely on the TrxR system. The mammalian TrxRs described so far are selenoproteins that utilize NADPH to reduce protein as well as nonprotein substrates in mitochondria and cytoplasm of cells. We show that a single Drosophila gene, Trxr-1, encodes non-selenocysteine-containing cytoplasmic and mitochondrial TrxR isoforms that differ with respect to their N termini. We generated transcript-specific mutants and used in vivo approaches to explore the biological functions of the two enzyme variants by introducing the corresponding transgenes into different Trxr-1 mutants. The results show that, although the two TrxR isoforms have similar biochemical properties, their biological functions are not interchangeable.

Three highly purified endosomal fractions from rat liver were used to purify and characterize a major protein of endosomal membranes. Intravenously injected ligands, which are taken up via receptor-mediated endocytosis, accumulate first in the fraction of intermediate density, the compartment of uncoupling of receptors and ligands. The high density membranous fraction is highly enriched in a receptor recycling compartment. The endosomal fraction of lowest density is composed of multivesicular bodies, which appear to be the immediate prelysosomal compartment. The most prominent membrane protein of these endosomes is one of 68 kDa, as revealed by silver and Coomassie Brilliant Blue staining of SDS-gel electrophoretograms. This protein dominates profiles obtained from purified membranes of the compartment of uncoupling of receptors and ligands, multivesicular bodies, and receptor recycling compartment, but is greatly reduced in those obtained from plasma membranes and lysosomes. The 68-kDa protein was purified from endosomes and digested with trypsin, and cleavage products were analyzed by protein sequencing. The tryptic fragments of the endosomal 68-kDa protein share 96% identity with corresponding sequences of mouse annexin VI and 91% identity with sequences of human annexin VI. Using immunoblots, high concentrations of annexin VI with an apparent molecular mass of 68 kDa were detected in endosomal membranes by specific antiserum to annexin VI. Significant amounts of annexin VI were also detected in Golgi membranes. Yet, the concentration was substantially lower than that of the three endosomal fractions. The association of annexin VI with endosomal membranes is calcium-dependent, as revealed by the complete solubilization from endosomal membranes by EGTA. Incubation of intact endosomes with Pronase leads to a complete degradation of annexin VI without any detectable disintegration of proteins localized on the luminal surface of endosomal membranes. Evidently, annexin VI is localized on the cytoplasmatic leaflet of the membrane of endosomes and may be of significance for their intracellular trafficking.

Hepatocytes rapidly internalize epidermal growth factor (EGF) and transferrin by receptor-mediated endocytosis. Both EGF and its receptor are thought to be targeted for destruction in lysosomes, leading to down-regulation of the receptor, whereas transferrin, after unloading iron within the cell, is thought to recycle to the cell surface bound to its receptor. Previously, we isolated three endosomal fractions from livers of estradiol-treated rats and examined their roles in cellular trafficking of low density lipoproteins (LDL) and the LDL receptor, which cycles constitutively (Belcher, J. D., Hamilton, R. L., Brady, S. E., Hornick, C. A., Jackle, S., Schneider, W. J., and Havel, R. J., Proc. Natl. Acad. Sci. U. S. A. (1987) 84, 6785-6789). In the current study we have taken advantage of the distinct trafficking of the EGF receptor and transferrin to evaluate further the functions of these endosome fractions. Intravenous injection of a saturating amount of EGF into estradiol-treated rats induced internalization of a single population of EGF receptors, which rapidly accumulated in the endosome fraction of intermediate density ("compartment of uncoupling of receptor and ligand" (CURL)) and subsequently in the low density endosome fraction (multivesicular bodies (MVBs)). The high density endosome fraction, whose membranes contain a high concentration of recycling receptors (designated receptor-recycling compartment (RRC)), failed to accumulate EGF receptors after injection of EGF. In livers of rats not given exogenous EGF, EGF receptors were found in small but comparable concentrations in RRC, CURL, and MVB membranes, consistent with other evidence that targeting of the EGF receptor to lysosomes is mediated by ligand-induced phosphorylation. Transferrin also accumulated first in CURL and later in MVBs, but it also accumulated rapidly in the RRC fraction, consistent with the proposed function of this fraction in receptor recycling. Since transferrin is not degraded during its endocytic cycle, these observations indicate that apotransferrin and its receptor recycle from late endosomes (MVBs) located at the apical pole of hepatocytes, as well as from early endosomes near the sinusoidal pole.

Proteins from small amounts of biological material are separated at high resolution by micro two-dimensional gel electrophoresis. Under nonequilibrium conditions, proteins are focused in 10-microliters capillaries and separated on ultrathin slab gels. Our device allows simultaneous focusing of up to 10 samples which can be processed in the second dimension on a single gel. Several hundred protein spots from less than a total of 300 ng of protein are resolved on an area the size of a postage stamp. A single spot containing less than 10 pg of protein or 1 cpm can be detected by silver staining or autoradiography, respectively. Our system allows inexpensive, simultaneous, rapid analysis of proteins when only a limited amount of biological material is available for investigation. A detailed description of the equipment, the methods, and the characteristics of this microanalysis are presented.
