Ribonuclease T(1) treatment of 30S ribosomes of Escherichia coli converts a large region at the 3' OH end of 16S ribosomal ribonucleic acid (rRNA) to low-molecular-weight RNA. The final 25 nucleotides at the 3' terminus of the molecule emerge relatively intact, whereas most of the region "upstream," for about 150 nucleotides, is converted to oligonucleotides. Identical enzyme treatment generates a fragment of about 60 nucleotides from the middle of 16S rRNA (section D'). To determine whether there are similar sequences in other bacteria, which occupy similar accessible surface locations, we treated 30S ribosomes from Azotobacter vinelandii and Bacillus stearothermophilus with RNase T(1). In each case, a fragment of RNA about 25 nucleotides in length containing the 3' OH end of 16S rRNA and a fragment of about 60 nucleotides in length similar, but not identical, in oligonucleotide composition to section D' of E. coli 16S rRNA were obtained from nuclease-treated 30S ribosomes. These data indicate that, although the primary structure at the 3' end and the middle (section D') of the various 16S rRNA's is not completely conserved, their respective conformations are conserved. A number of identical oligonucleotides were found in the low-molecular-weight fraction obtained from RNase T(1)-treated E. coli, A. vinelandii, and B. stearothermophilus 30S ribosomes. These results show that identical RNase T(1)-sensitive sequences are present in all three bacteria. Hydrolysis of these regions leads to the production of the fragments 25 and 60 nucleotides in length.

Chemical analysis indicated that D-glucose is tha major neutral monosaccharide present in the microcysts of a range of gram-negative bacteria. Varying amounts of other neutral sugars were found. The glucose was mainly present as a glucan that could be extracted from microcysts of representative strains with alkali or mild acid treatment. The glucan could be identified as an alpha-1,3-linked polymer on the basis of (i) periodate resistance of the extracted polymer and the material present in microcysts; (ii) lectin agglutination of the microcysts; (iii) lectin precipitation of the extracted glucans; and (iv) susceptibility of the glucan either in the walls or after extraction to a specific alpha-1,3-glucanase from Aspergillus nidulans, yielding glucose as the sole hydrolysis product. The galactosamine found in microcysts of Myxococcus xanthus by other workers is clearly a component of another polymer, distinct from the glucan. The presence of an alpha 1,3-linked glucan, common to microcyst walls of various bacterial genera, probably contributes to the rigidity of the walls of these forms and, inter alia, to their resistance to ultrasonic treatment. Preliminary experiments indicate that the gulcan is discarded on germination of the microcysts rather than being broken down by specific enzymes.

The high-potential 4Fe-4S* center ofAzotobacter vinelandii ferredoxin I has been titrated potentiometrically by a reductive procedure. The absorbance decrease at 510 nm accompanying the reduction of the high-potential center titrated with an Em of 320 mV (n = 1). The low-potential 4Fe-4S* center was titrated by using the absorbance decrease at 410 nm to monitor its reduction. This center exhibited an Em of -424 mV (n = 1).

The properties of Azotobacter vinelandii deoxyribonucleic acid (DNA) and folded chromosomes were studied and compared to those of Escherichia coli as a standard. Based on melting temperature and buoyant density measurements, the guanosine + cytosine content of purified A. vinelandii DNA was 65%, whereas that of E. coli DNA was 50%. The results of renaturation studies showed that the unique DNA sequence lengths of the two organisms were similar with Cot1/2 values of 7.3 +/- 0.4 mol.s/liter and 7.5 +/- 0.3 mol.s/liter, respectively, for A. vinelandii and E. coli. Folded chromosomes of A. vinelandii sedimented in a centrifugal field at a rate identical to those derived from E. coli, 1,600 to 1,700S. Based on the DNA content per cell and the mass of a single genome, A. vinelandii contains at least 40 chromosomes per cell.

Azotobacter vinelandii was found to form novel lipid compounds when encystment was initiated by 0.2% beta-hydroxybutyrate. An examination of these compounds led to the isolation and characterization of 5-n-heneicosylresorcinol, 5-n-tricosylresorcinol, and their galactoside derivatives.

Unique cyclic compounds were found in the lipid fraction of Azotobacter vinelandii cysts. In addition to two major molecular species which had already been identified, 5-n-alkylresorcinol and its galactoside derivative, five other molecular species (two alkyl side chain homologs of each) were isolated, and their structures were established by infrared, ultraviolet, nuclear magnetic resonance, and mass spectroscopy. These 10 compounds were 6-n-heneicosylresorcylic acid methyl ester and 6-n-tricosylresorcylic acid methyl ester, 5-n-(2-hydroxy)heneicosylresorcinol and 5-n-(2-hydroxy-tricosylresorcinol, 5-n-heneicosyl-4-acetylresorcinol and 5-n-tricosyl-4-acetylresorcinol, 6-n-heneicosyl-4-hydroxypyran-2-one and 6-n-tricosyl-4-hydroxypyran-2-one, and 6-(2-oxotricosyl)-4-hydroxy-pyran-2-one and 6-(2-oxopentacosyl)-4-hydroxypyran-2-one.

The bacterioferritin from Azotobacter vinelandii exhibits properties which in ferritins from other sources are attributed to the heteropolymeric nature of the holoprotein. The native bacterioferritin displayed multiple bands on isoelectric focusing gels. On discontinuous sodium dodecyl sulfate-polyacrylamide gels, there were two subunit polypeptides of approximate Mr 21,000 and 23,000. These molecular weights were corroborated by gel filtration experiments. Peptide maps produced by partial trypsin digestion and electrophoresis showed no detectable differences between the subunits. Similarities to well-characterized mammalian ferritins and apparent anomalies in two commonly applied electrophoretic procedures are discussed.

Cyclopropane fatty acids constitute 25% of the phospholipid acyl groups in cysts of Azotobacter vinelandii. These are lost by dilution during germination when the synthesis of the fatty acids characteristic of vegetative cell phospholipids commences.

d(-) Lactate oxidation in Azobacter vinelandii strain O is readily carried out by the membrane bound enzyme that concentrates in the electron transport fraction (R(3)). This oxidation in the R(3) fraction is not dependent on externally added nicotinamide adenine dinucleotide, flavine adenine dinucleotide, or flavine mononucleotide. Phenazine methosulfate, O(2), and menadione all served as good electron carriers, and the oxidation of lactate was limited to the d(-) stereoisomer. Of all the alpha-hydroxyacids examined, only d(-) lactate and d(-) alpha-hydroxybutyrate were oxidized by the R(3) fraction. Paper chromatographic studies revealed that the 2,4-dinitrophenylhydrazine derivative formed from d(-) lactate oxidation was pyruvate. Pyruvate, in turn, could be further decarboxylated nonoxidatively by the R(3) fraction. Spectral studies revealed that both the R(3) flavoprotein and cytochrome (a(2), a(1), b(1), c(4), and c(5)) components were reduced by d(-) lactate. The d(-) lactic oxidase activity was sensitive to electron transport inhibitors, i.e., chlorpromazine, antimycin A, 2-n-heptyl-4-hydroxyquinoline-N-oxide, rotenone, dicumarol, and cyanide, and to a small extent to 4,4,4-trifluoro-1-(2-thienyl)-1,3-butane-dione (TFTB) and Amytal. The d(-) lactic phenazine methosulfate and menadione reductases were sensitive only to dicumarol and TFTB. Chlorpromazine was found to be a highly specific inhibitor of d(-) lactic oxidase activity, 50% inhibition occurring at 6.6 x 10(-6)m.

Cysts of Azotobacter vinelandii were ruptured with 3.0 mm ethylenediaminetetraacetate in 0.05 m tris(hydroxymethyl)aminomethane buffer (pH 7.8) and fractionated by differential centrifugation in density gradients of sucrose or Ficoll (polysucrose) at 10,000 x g for 2 hr at 4 C. The average densities of the cyst exine, the cyst central body, the vegetative cell, and the intact cyst were found to be 1.13, 1.26, 1.28, and 1.31 g/cm(3) in sucrose, and 1.08, 1.12, 1.13, and 1.17 g/cm(3) in Ficoll, respectively. These data were utilized to devise procedures for the isolation of cyst components. Cyst exine appeared as a multilayered structure in ultrathin sections. This same sheetlike structure was seen when exines were subjected to detergent treatment, metal-shadowed, and examined in an electron microscope. The exine was lysed by trypsin and was resistant to lysozyme, indicating that protein may be the principal structural material of the outer cyst coat.

Analyses of resting cells of Azotobacter vinelandii revealed that numerous phospholipids were present that did not concentrate in the membranous R(3) fraction which carried out electron transport function.

Azotobacter vinelandii, in late exponential growth phase, encysts when the glucose in the medium is replaced with beta-hydroxybutyrate. A final cell division then occurs without apparent deoxyribonucleic acid (DNA) synthesis, resulting in a reduction from two to one nucleoids per cell and a final DNA content of 3.2 x 10(-14) g per cell. This is also the DNA content per cyst. A beta-hydroxybutyrate dehydrogenase is derepressed by the addition of the inducer and is identical to the enzyme in acetate-grown cells in its pH optimum, Michaelis constant for substrate, temperature-activity response, and mobility during electrophoresis in acrylamide gel.

The ribosomal proteins from nine species of prokaryotes have been compared by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. The stained gels were scanned spectrophotometrically, the weight and number average molecular weights were calculated, and the detailed distribution of the proteins as a function of molecular weight was determined. By all of these criteria, the ribosomal proteins from all the species closely resembled each other, despite differences in the pattern of protein bands by conventional disc-gel electrophoresis. Therefore, it is suggested that the structural requirements for the assembly of ribosomal subunits have imposed limitations on the evolution of the ribosomal proteins and that their size has been highly conserved.

Extraction with pentane virtually abolished reduced nicotinamide adenine dinucleotide oxidase activity in small particles from Azotobacter vinelandii, but activity was largely restored by added ubiquinone.

Poly-beta-hydroxybutyrate (PHB) from various representative strains of the genera Azotobacter, Beijerinckia, and Derxia was isolated and characterized. During growth in shake culture, with glucose as a carbon and energy source, and molecular nitrogen as a nitrogen source, increase in dry weight appeared linear, and PHB formed a constant percentage of the dry weight. In a medium containing 1% (w/v) glucose, PHB declined with the onset of the stationary phase of growth; with 2% (w/v) glucose, an increase in PHB content during stationary phase was noted in the case of some strains, before a subsequent decline. The decrease in PHB as a percentage of dry cellular weight (not of total amount present in the culture) during growth of some strains with 2% as opposed to 1% (w/v) glucose may be ascribed to a greater production of capsular polysaccharide. PHB content could not be used as a taxonomic criterion. Strain differences were as great as or greater than species differences. The only strain of Beijerinckia fluminensis obtained contained PHB, but it could not be grown on the nitrogen-free medium used. Two species of the genus Azotomonas, reported to be aerobic, nonsymbiotic nitrogen-fixers, did not grow on the nitrogen-free medium used and did not produce PHB during growth with a combined nitrogen source.

Cysts of Azotobacter vinelandii ATCC 12837 were germinated by exposure to 3.0 mm ethylenediaminetetraacetic acid (EDTA)-tris(hydroxymethyl)aminomethane buffer at pH 7.8, and their outer coats (exines) were purified by differential and isopycnic centrifugation. Electron micrographs of exine showed it to consist of multilayers of a three-membered sheet structure whose thickness was 7.0 to 7.5 nm. The inner, less electron-dense layer (intine) was also prepared from cysts by EDTA treatment, centrifugation, concentration, and dialysis. The exine consisted of 32% carbohydrate, 28% protein, 30% lipid, and 3.2% ash, with the ash comprised of 1.62% calcium, 0.02% magnesium, and 0.34% phosphorus. The amino acid composition of exine was similar to that of gram-negative bacterial cell walls. The intine consisted of 44% carbohydrate, 9.1% protein, 37% lipid, and 4.1% ash, with the ash comprised of 2.45% calcium, 0.02% magnesium, and 0.38% phosphorus. The carbohydrates of both exine and intine contained glucose, mannose, xylose, and rhamnose. Glucosamine and galactosamine were found only in the exines. The fatty acids consisted of normal, iso, and anteiso saturated fatty acids with 10 to 18 carbon atoms and mono-unsaturated C(11), C(16), and C(18) fatty acids. The exines contained mostly bound lipid, but intines contained primarily free lipid.
