This experiment evaluated the effects of roughage levels and supplemental fat on intake, growth performance, health, and physiological responses of newly received finishing cattle during 58-d receiving period. A total of 72 crossbred steers (initial body weight [BW] = 200 ± 13 kg) were used in a randomized complete block design with a 2 × 2 factorial arrangement of treatments, consisting of two roughage levels (wheat hay at 30% [R30] or 60% [R60]; dry matter [DM] basis) and two levels of supplemental fat (yellow grease at 0% [−FAT; no additional fat] or 3.5% [+FAT]; DM basis). Upon arrival, calves were individually weighed, blocked by off-truck shrunk BW, and assigned to 24 soil-surfaced pens (three calves per pen). Shrunk BW was also collected on day 58 for the calculation of average daily gain (ADG). Throughout the study, calves were assessed for bovine respiratory disease (BRD). Effects of roughage level × supplemental fat interaction were only observed for diet particle size distribution and estimated physically effective neutral detergent fiber (pe NDF) of diets (P ≤ 0.10). Adding fat to R60 diets tended to increase the percentage of particles retained in the 8-mm screen (P = 0.06) and the estimated pe NDF (P = 0.10), but did not affect R30 diets. Dietary roughage level did not affect DM intake (DMI; P = 0.85). Calves-fed R30 tended to have greater ADG and final BW than calves-fed R60 (P ≤ 0.08). Gain efficiency (gain:feed ratio; G:F) was greater for calves-fed R30 than calves-fed R60 (P = 0.01). Dietary roughage level did not affect morbidity and mortality (P ≥ 0.11). Supplemental fat did not affect DMI (P = 0.6) but tended (P = 0.09) to increase ADG compared to –FAT diets. The G:F was greater for calves-fed +FAT than –FAT (P = 0.03). The +FAT diet tended (P = 0.10) to increase the number of retreatments against BRD compared to −FAT, although the total number of antimicrobial treatments required to treat sick calves (P = 0.78) and the mortality rate (P = 0.99) were not affected by supplemental fat. Feeding +FAT diet tended (P ≤ 0.09) to increase plasma concentration of cortisol and immunoglobulin-G compared to –FAT. Grazing management and N-fertilizer have been reported to improve tropical forage productivity and quality, however, their effect on methane emission of grazing animals remains uncertain. Therefore, this study aimed to assess the effects of increasing application rates of nitrogen (N) fertilization of Marandu palisadegrass under continuous stocking on intake, digestibility, nitrogen balance, and enteric methane emissions of Nellore growing bulls. We hypothesized that changes in the forage nutritive value caused by N fertilization of pastures combined with adequate grazing management (e.g. greater crude protein [ CP ] and digestibility) would lead to an increase in animal productivity (e.g. greater average daily gain [ ADG ] and gain per area), and then, to a decrease in methane emission intensity. Treatments consisted of different annual application rates of nitrogen fertilization: 0, 75, and 150 kg N/ha using ammonium nitrate (32% N) as the nitrogen source. The experimental design was completely randomized, with three treatments and four replications (12 paddocks). Intake, digestibility, N balance, and methane emissions were measured in eight animals per treatment. CP intake, digestibility and N balance increased linearly with the increase in N fertilization (P < 0.05). In addition, stocking rate (SR) and ADG linearly increased from 1.75 animal unit (AU = 450 kg)/ha and 0.62 kg/d (0 kg N/ha) to 3.75 AU/ha and 0.82 kg/d (150 kg N/ha), respectively. Individual methane emissions nor methane emission intensity were affected by treatment with an average of 164.7 g/d and 199.7 g/kg ADG (P > 0.05). Annual N fertilization with ammonium nitrate between 75 and 150 kg N/ha in palisadegrass pastures under continuous stocking enhances animal performance per unit area yet not affecting neither methane production nor intensity. With most of the student attrition occurring early in undergraduate educational programs (Braunstein et al. 1997) it is necessary to interest and motivate students early on. The demographics of animal science students have shifted to students with minimal background in food producing animals. This presents a unique challenge as the current student population represents a diverse array of backgrounds and prior experiences. As a result, students enroll in undergraduate animal science programs with various expectations for their undergraduate degree and a focus primarily on careers in veterinary medicine. To engage all students, interest and motivation need to be generated. This review will use motivational frameworks as outlined by the self-determination theory, expectancy value theory, and interest, to explain the impact of the proposed solutions. Active learning classroom strategies are linked to increased knowledge compared with traditional, passive classrooms (Wells et al. 2019). Active learning shifts from a traditional teaching model to a student-centered model, which transitions instructors to facilitators of learning. This review summarizes current proposed pedagogies that have been researched in animal science classrooms such as experiential learning, flipped classrooms, hands-on animal experience, undergraduate research experiences, mentorship opportunities, capstone experiences, service-learning experiences, team-based learning, and cooperative learning. In this study, the efficacy of different nonstarch polysaccharide (NSP) enzyme sources on wheat ingredients and wheat basal diets in vitro were evaluated by simulating the avian digestive tract. In Exp. 1, pH level was increased from 2.0 to 8.0 by simulating the avian digestive tract. The relative enzyme activities of xylanase A, B, and C and β-glucanase X at pH 3.0–3.5 were higher (P < 0.05) than those at pH 2.0 or 7.0–8.0. The optimal pH levels of 3.5 and 7.0 were screened by simulating the proventriculus and small intestine, respectively to evaluate the efficacy of NSP enzyme on wheat sources. In Exp. 2, wheat 1 contained the highest content of NSP fractions and the lowest digestibility in vitro dry matter (IVDMD) and energy (IVED) in wheat samples. Therefore, wheat 1 was selected for hydrolysis research under different NSP enzyme sources and levels (1,500, 4,500, 13,500, 40,500, 121,500 U xylanase/kg and 250, 500, 1,000, 2,000, 4,000 U β-glucanase/kg) in vitro. The hydrolysis of wheat on the basis of the released reducing sugar content was determined by xylanase sources A > B > C (P < 0.05) and β-glucanase sources of X > Y (P < 0.05). On the basis of the hydrolysis, the optimum dose of xylanase A and β-glucanase X were 40,500 U/kg and 2,000 U/kg, respectively. Subsequently, the completely randomized designs involving 2 NSP enzymes treatments × 2 endogenous digestive enzymes treatments (Exp. 3), as well as 2 wheat basal diets × 2 NSP enzymes treatments (Exp. 4) were used to evaluate the efficacy of NSP enzymes on dietary nutrient digestibility. The addition of NSP enzymes (40,500 U xylanase A/kg and 2,000 U β-glucanase X/kg) increased the IVDMD and IVED of wheat 1 without endogenous enzymes (P < 0.05), while the IVDMD and IVED of wheat 1 with endogenous enzyme were only slightly increased (P > 0.05). The addition of NSP enzymes could increase the IVDMD and IVED of corn–wheat–soybean meal diet (P < 0.05), but had no effect on those of wheat–cottonseed meal rapeseed meal diet (P > 0.05). In conclusion, xylanase and β-glucanase additions could effectively eliminate the adverse effects on wheat and wheat basal diets at the optimal pH levels of 3.5 and 7.0 by simulating the proventriculus and small intestine parts, respectively. The objective was to determine the effects of injectable vitamin E (VE) before or after transit on feedlot cattle receiving performance, health, and blood parameters. Angus × Simmental steers (n = 196; body weight [ BW ] = 163 ± 29 kg) were utilized in a randomized complete block design. Steers were blocked by BW and randomly assigned to 1 of 3 treatments: intramuscular injections of saline pre- and post-transit (CON), intramuscular injections of VE (2,000 mg d-α-tocopherol) pre-transit and saline post-transit (PRE), or intramuscular injections of saline pre-transit and VE (2,000 mg d-α-tocopherol) post-transit (POST). Pre-transit injections were administered on day 0, and steers were transported on day 7 for approximately 4 h (348 km). After arrival, steers were fed a common corn silage-based diet in GrowSafe bunks. Final BW tended to be greater (P = 0.08) for CON steers compared with POST steers while PRE steers were intermediate. From days 7 to 63, treatment affected average daily gain (ADG) with PRE and CON steers exhibiting (P = 0.04) greater ADG compared with POST steers. Dry matter intake (DMI), water intake, and gain to feed from days 7 to 63 were not affected (P ≥ 0.17) by treatment. Day 0 serum α-tocopherol concentrations were considered marginal (2.3 ± 0.2 mg/l). A treatment × day interaction (P < 0.01) was observed for serum α-tocopherol concentrations. Serum α-tocopherol concentrations were greatest for PRE steers on day 7 (prior to and post-transit), but greater for POST steers on dys 10 and 14. Plasma ferric-reducing antioxidant potential concentrations increased (P = 0.04) for POST steers compared with CON steers and PRE steers being intermediate. Plasma non-esterified fatty acids (NEFA) concentrations exhibited a treatment × day interaction (P = 0.04) with CON and POST steers being 16% and 14% greater than PRE steers on day 14, respectively. On day 21, NEFA concentrations were greatest for POST steers compared with PRE steers and CON steers being intermediate. There was no main effect (P ≥ 0.14) of treatment on the number of bovine respiratory disease morbidity treatments. Hair cortisol concentrations were decreased (P < 0.01) 14 days after transit for PRE and POST steers compared with CON steers. Overall, injectable VE administered before or after transit increased serum tocopherol concentrations while reducing stress, but did not improve the growth performance of beef steers during the receiving phase. Eighty-four Angus crossbred heifers (13 ± 1 mo of age, 329.5 ± 61.92 kg of body weight [BW]) were used in a generalized randomized block design with a 2 × 2 factorial arrangement of treatments. The factors evaluated were: 1) diet type (whole plant sorghum silage [ SS ] vs. byproducts-based [ BP ]), and 2) feed additive: Aspergillus oryzae prebiotic (AOP; 2 g/d) vs. Negative control (CTL; 0 g/d), resulting in four treatments: sorghum silage-control (SC), sorghum silage-AOP (SA), byproducts-control (BC), and byproducts-AOP (BA). Heifers were stratified by body weight (BW), randomly assigned to treatments (21 heifers per treatment) and housed in 12 pens equipped with two GrowSafe feed bunks each to measure individual dry matter intake (DMI). After a 14-d adaptation, BW was measured every 14 d for 56 d. Chewing activity was monitored through collar-mounted HR-Tags (heat-related tags). Following the performance period, apparent total tract digestibility was measured in 40 heifers, using indigestible neutral detergent fiber as a marker. Heifers fed with the BP diets had greater DMI (2.92% vs. 2.59% of BW, P < 0.01) and average daily gain (ADG ; 1.16 vs. 0.68 kg, P ≤ 0.01) than heifers fed with SS diets. Compared with BP-fed animals, heifers consuming the SS diets had 23 more visits/d to the feed bunks (P ≤ 0.01), consumed 53% less dry matter on each visit (P ≤ 0.01), and spent 39% more min chewing/d and 63% more min chewing/kg of DMI (P ≤ 0.01). However, chewing measured in min/kg of neutral detergent fiber intake was not affected by treatment (average 111.3 min/kg of NDF intake). Feeding AOP improved gain:feed (GF) by 15% in BP-fed heifers (0.120 vs. 0.104 kg/kg; P < 0.05). Inclusion of AOP increased organic matter digestibility (OMD) in SS diets (55.88% vs. 49.83%; P < 0.01), whereas it decreased OMD in BP diets (61.67% vs. 65.77%; P < 0.05). In conclusion, ADG and GF of BP-fed heifers was greater than SS-fed heifers, and GF was greater with AOP supplementation in BP-fed heifers. Improvement in GF in BP-fed heifers was likely not related to differences in nutrient digestibility as AOP inclusion did not enhance digestibility in the BP diet. Additionally, the effects of the AOP inclusion appear to be diet-dependent, where the 15% improvement in GF by AOP occurred in heifers fed with the more fermentable diet. This study investigated the effects of injectable vitamin C (VC) at weaning and prior to transit on growth performance and immune function in early-weaned beef steers. On day 0, 91 Angus × Simmental steers (92 ± 4 kg) were weaned (65 ± 11 d of age), given vaccination boosters, blocked by age, and randomly assigned to weaning (WEAN) treatments: intramuscular injections (20 mL per steer) of VC (250-mg sodium ascorbate per mL; 5 g per steer) or saline (SAL). From days 0 to 48, steers were housed at the Dixon Springs Agricultural Center (Simpson, IL) in pens (six pens; N = 14 to 16 steers per pen) equipped with two to three Vytelle bunks to measure individual daily feed disappearance. On day 49, half of the steers in each WEAN treatment were randomly assigned to an additional injection treatment (20 mL per steer) of VC or SAL prior to transport (TRANS). After administering pretransit injections, all steers were loaded onto a commercial livestock trailer with equal representation of treatments across compartments. Steers were transported for 6 h (approximately 480 km) to the Illinois Beef and Sheep Field Laboratory (Urbana, IL). Upon arrival, steers were sorted into pens (six pens; N = 13 to 17 steers per pen) with 2 Vytelle bunks per pen. Steers were weighed on days 0, 1, 14, 48, 49, 64, 78, 106, and 107. Blood was collected (WEAN = 24 steers per treatment; TRANS = 12 steers per treatment) on days 0, 1, 2, 14, 49 (pre- and posttransit), 50, and 51. Data were analyzed using the MIXED procedure of SAS 9.4 with fixed effects of age block, WEAN, TRANS, and WEAN × TRANS. Plasma ascorbate concentrations were greater (WEAN × time P < 0.01) on days 1 and 2 for steers that received VC at weaning. Similarly, for steers that received VC on day 49 pretransit, ascorbate concentrations were greater (TRANS × time P = 0.04) on days 49 posttransit, 50, and 51. Treatments did not affect (P ≥ 0.13) body weight, average daily gain, or gain to feed throughout the trial. Serum Bovine Viral Diarrhea Virus type 1 and 2 antibody titers on days 14 and 51 were not affected (P ≥ 0.32) by treatment. Injectable VC administered to early-weaned beef steers at the time of weaning or pretransit increased plasma ascorbate concentrations but did not improve growth performance or antibody response to vaccination booster. Five ruminally and duodenally cannulated red angus steers (n = 5; initial body weight [ BW ] = 542 kg, SD = 40 kg) were used in a three-period Youden square design consisting of three 21-d periods, three treatments, and five steers (one or two steers per treatment within each period) to evaluate the effect of feeding hempseed cake on ruminal fermentation parameters, organic matter (OM) intake, total tract nutrient digestion, and nitrogen (N) balance in steers fed finishing diets. The control (CON) diet contained 75% dry-rolled corn, 20% corn silage, and 5% supplement (DM basis). The dried corn distillers grains plus solubles (DDGS) and hempseed cake (HEMP) diets contained 55% dry-rolled corn, 20% corn silage, 20% dried corn distillers grains plus solubles or hempseed cake, and 5% supplement (DM basis). Total ruminal volatile fatty acid concentration was greater (P < 0.01) in steers fed the HEMP diet than in steers fed the DDGS or CON diets. Ruminal fluid pH was not influenced (P = 0.93) by treatment. Organic matter intake tended (P = 0.07) to be greater and OM total tract digestibility was less (P = 0.03) in steers fed the HEMP diet compared with steers fed the DDGS or CON diets. Ruminal true and total tract apparent N digestibility was greater (P < 0.01) in steers fed the HEMP diet than steers fed the DDGS or CON diets. Duodenal flow of essential, nonessential, and total amino acids was not influenced (P ≥ 0.37) by dietary treatment, but the lack of response was likely because ruminally degradable protein (RDP) supply exceeded the RDP requirement. Steers fed the HEMP diet had greater (P < 0.01) N retention (g/d) than steers fed the DDGS diet, which was greater (P < 0.01) than steers fed the CON diet, suggesting that feeding hempseed cake improved utilization of N. Although inclusion of hempseed cake decreased total tract OM digestibility compared with dried corn distillers grains or corn, improvements in N utilization suggest that hempseed cake could be a useful alternative feed ingredient for finishing cattle diets. Ducks will access water to maintain feather condition and exhibit natural water-related behaviors such as wet preening. Providing water to ducks commercially is challenging as it may reduce litter and air quality leading to higher duck mortality or illness. This research aimed to measure the behavioral and welfare impacts of water provision via a misting system for commercial Pekin grower ducks in Victoria, Australia. Seven grower flocks were observed (four misted and three nonmisted in open-sided sheds) during May and November 2021. From 26 until 33 d of age, treatment ducks were provided 1 h of misting with shed curtains closed in both treatment and control sheds. At the start and end of the misting application period, external health and welfare measures were taken directly on the ducks via transect walks throughout each shed and catch-and-inspect observations on a sample of 150 ducks from each shed. Video recordings were also made of the misted and nonmisted ducks for 3 h representing time periods prior to, during, and after the 1-h misting across all sheds for all 8 d of the treatment period. Observations were made of all behavior that ducks exhibited at 10-min scan sample intervals across four cameras per shed, totaling 4,198 scans across the seven sheds. General linear mixed models showed the misting application predominantly had impacts on the patterns of behavioral change across the treatment time periods between the misted and nonmisted ducks rather than increasing or decreasing the overall expression of specific behaviors (interaction terms all P ≤ 0.003). The misted ducks increased drinking, tail wagging, and walking, and reduced preening, rooting litter, sitting, and stretching during misting relative to what they showed prior. The nonmisted ducks showed less sitting and more panting during misting relative to prior. Pearson's Chi-square tests showed some differences between the treatment groups in feather cleanliness on the back and wings (both P < 0.0001), likely resulting from pre-existing differences between sheds in blood from pin feathers. Most welfare indicators showed no positive or negative effect of the misting treatment. These results indicate overhead misting does affect duck behavior to some degree without compromising their welfare, but further research with larger water droplet sizes resulting in greater accumulation of surface water or extended durations of misting may lead to greater effects. This study was conducted to determine greenhouse gas (GHG) emissions, water consumption, land use, as well as nitrogen (N), phosphorus (P), and carbon (C) balance of five diet formulation strategies and feeding programs for growing-finishing pigs (25–130 kg body weight) in the three spatially explicit geographic regions where the majority of U.S. pork production occurs. Feeding programs evaluated consisted of 1) standard corn-soybean meal (CSBM) diets, 2) CSBM containing 15% corn distillers dried grains with solubles (DDGS), 3) CSBM with 8.6% thermally processed supermarket food waste (FW), 4) low crude protein CSBM diets supplemented with synthetic amino acids (SAA), and 5) CSBM with phytase enzyme (PHY) added at 600 FTU (phytase units)/kg of diet. An attributional Life Cycle Assessment approach using a highly specialized, spatially explicit Food System Supply-Chain Sustainability (FoodS3) model was used to quantify GHG emissions, water consumption, and land use of corn, soybean meal, and DDGS based on county level sourcing. The DDGS, FW, and SAA feeding programs had less estimated N and P intake and excretion than CSBM, and the PHY feeding program provided the greatest reduction in P excretion. The FW feeding program had the least overall GHG emissions (319.9 vs. 324.6 to 354.1 kg CO2 equiv./market hog), land use (331.5 vs. 346.5 to 385.2 m2/market hog), and water consumption (7.64 vs. 7.70 to 8.30 m3/market hog) among the alternatives. The DDGS feeding program had the greatest GHG emissions (354.1 kg CO2 equiv./market hog) among all programs but had less impacts on water consumption (7.70 m3) and land use (346.5 m2) per market hog than CSBM and PHY. The SAA feeding program provided a 6.5–7.4% reduction in land use impacts compared with CSBM and PHY, respectively. Regardless of feeding program, the Midwest had the least contributions to GHG emissions and land use attributed to feed and manure among regions. Water consumption per market hog associated with feeding programs was much greater in the Southwest (59.66–63.58 m3) than in the Midwest (4.45–4.88 m3) and Mid-Atlantic (1.85–2.14 m3) regions. Results show that diet composition and U.S. geographic region significantly affect GHG emissions, water consumption, and land use of pork production systems, and the potential use of thermally processed supermarket food waste at relatively low diet inclusion rates (<10%) can reduce environmental impacts compared with other common feeding strategies. Dried distillers' grains, coproducts from the ethanol industry, may provide sustainable ingredients for pet food. Due to new post-fermentation separation techniques, corn-fermented protein (CFP) is higher in protein and lower in fiber compared with traditional dried distillers' grains, increasing its appeal for inclusion into pet food. Therefore, the objectives of this study were to determine the effects of increasing levels of CFP on stool quality, apparent total tract digestibility (ATTD), and palatability in adult cats. Four extruded diets were fed to 11 adult cats in an incomplete 4 × 4 replicated Latin square design. The control diet contained 15% soybean meal (0C) and CFP was exchanged for soybean meal at either 5%, 10%, or 15% (5C, 10C, 15C). Cats were fed each dietary treatment for 9-d adaption followed by 5-d total fecal collection. Feces were scored on a 1 to 5 scale, with 1 representing liquid diarrhea and 5 representing hard pellet-like (Carciofi et al. 2008). A fecal score of 3.5 to 4 was considered ideal. Titanium dioxide was added to all diets (0.4%) as a marker to estimate digestibility. Data were analyzed using a mixed model in SAS (version 9.4, SAS Institute, Inc. Cary, NC) with treatment as a fixed effect and cat and period as random effects. Fecal dry matter percent and dry fecal output were greater (P < 0.05) at elevated levels of CFP. Stool scores were maintained (P > 0.05) throughout treatments (average; 4). Dry matter, organic matter, crude protein, and gross energy ATTD decreased when cats were fed 15C. There was no difference in ATTD of fat or total dietary fiber among treatments. For palatability assessment, cats preferred 5C over 0C but had no preference with increased CFP inclusion. Rumen methanogenesis results in the loss of 6% to 10% of gross energy intake in cattle and globally is the single most significant source of anthropogenic methane (CH4) emissions. The purpose of this study was to analyze greenhouse gas traits recorded in a commercial feedlot unit to gain an understanding into the relationships between greenhouse gas traits and production traits. Methane and carbon dioxide (CO2) data recorded via multiple GreenFeed Emission Monitoring (GEM), systems as well as feed intake, live weight, ultrasound scanning data, and slaughter data were available on 1,099 animals destined for beef production, of which 648 were steers, 361 were heifers, and 90 were bulls. Phenotypic relationships between GEM emission measurements with feed intake, weight traits, muscle ultrasound data, and carcass traits were estimated. Utilization of GEM systems, daily patterns of methane output, and repeatability of GEM system measurements across averaging periods were also assessed. Methane concentrations varied with visit number, duration, and time of day of visit to the GEM system. Mean CH4 and CO2 varied between sex, with mean CH4 of 256.1 g/day ± 64.23 for steers, 234.7 g/day ± 59.46 for heifers, and 156.9 g/day ± 55.98 for young bulls. A 10-d average period of GEM system measurements were required for steers and heifers to achieve a minimum repeatability of 0.60; however, higher levels of repeatability were observed in animals that attended the GEM system more frequently. In contrast, CO2 emissions reached repeatability estimates >0.6 for steers and heifers in all averaging periods greater than 2-d, suggesting that cattle have a moderately consistent CO2 emission pattern across time periods. Animals with heavier bodyweights were observed to have higher levels of CH4 (correlation = 0.30) and CO2 production (correlation = 0.61), and when assessing direct methane, higher levels of dry matter intake were associated with higher methane output (correlation = 0.31). Results suggest that reducing CH4 can have a negative impact on growth and body composition of cattle. Methane ratio traits, such as methane yield and intensity were also evaluated, and while easy to understand and compare across populations, ratio traits are undesirable in animal breeding, due to the unpredictable level of response. Methane adjusted for dry matter intake and liveweight (Residual CH4) should be considered as an alternative emission trait when selecting for reduced emissions within breeding goals. The objective of this work was to evaluate the use of a Torula yeast (TY) on diet processing, palatability, and total tract nutrient digestibility in extruded feline diets. Four dietary treatments were compared, differing by protein source: TY, pea protein concentrate (PP), soybean meal (SM), and chicken meal (CM). Diets were produced using a single-screw extruder under similar processing conditions. Palatability assessment was conducted as a split plate design where both first choice and intake ratio (IR) were determined. Apparent total tract digestibility (ATTD) of nutrients was estimated using Titanium dioxide as an indigestible marker. During diet production, specific mechanical energy of TY and SM (average of 187 kJ/kg) was greater (P < 0.05) than for PP (138 kJ/kg); however, CM was similar to all treatments (167 kJ/kg). Kibble diameter, piece volume, and sectional expansion ratio were greatest for TY (P < 0.05). Additionally, both bulk and piece density were lowest (P < 0.05) for TY. Kibble hardness was lower for TY and SM (P < 0.05; average of 2.10 Newtons) compared to CM and PP (average of 2.90 Newtons). During the palatability trial, TY was chosen first a greater number of times than CM (P < 0.05; 36 vs. 4, respectively), but differences were not found between TY and PP (25 vs. 15, respectively) or TY and SM (24 vs. 16, respectively). Cats had a greater IR (P < 0.05) of TY compared to CM and PP (0.88 and 0.73, respectively). However, there was no difference in preference between TY and SM. ATTD of dry matter (DM) and organic matter (OM) was greater (P < 0.05) for CM (87.43% and 91.34%, respectively) than other treatments. Both DM and OM ATTD of TY were similar (P < 0.05) to PP and SM (average of 86.20% and average of 89.76%, respectively). Ash ATTD was greater (P < 0.05) for cats fed TY and SM (average of 37.42%), intermediate for PP (32.79%), and lowest for CM (23.97%). Crude protein (CP) ATTD of TY was similar to all other treatments (average of 89.97%), but fat ATTD was lower (P < 0.05; 92.52%) than other treatments (93.76% to 94.82%). Gross energy ATTD was greater (P < 0.05) for CM than TY (90.97% vs. 90.18%, respectively); however, TY was similar to PP and SM (average of 90.22%). Total dietary fiber ATTD was similar between TY and CM (average of 66.20%) and greater (P < 0.05) than PP and SM (average of 58.70%). Maternal care is essential to optimally support survival of the offspring. During evolution of mammalian species, different phenotypes have evolved in relation to gestation length, number, size, and maturation stage of the offspring at parturition, as well as colostrum and milk composition. The aim of the present review is to describe relationships between placental function and colostrum and milk composition in different mammalian species. Species covered in this article include humans, rabbits, rodents (rat and mouse), carnivores (cats and dogs), and a variety of ungulate species (cattle, sheep, goats, pigs, and horses). Species-specific aspects are elucidated with a special focus on the transfer of passive immunity. In this regard, the structure and thus the capability of the placenta to transport immunoglobulins from maternal to fetal circulation in utero dictates the necessity of the passive transfer of immunity via colostrum. Consequently, species with exclusive postpartal transfer of immunity such as in all ungulate species have greater immunoglobulin G concentrations in colostrum than species with a prepartal transfer in utero, where especially immunoglobulin A with its local immune function in the gastrointestinal tract is present in colostrum (e.g. rabbit and human). In terms of the nutritional purpose, suckling frequency is an important factor determining the gross composition of colostrum as well as in the mature milk of these species. Milk of nidicolous animals with long intervals in-between suckling events contains more fat than milk of nidifugous animals with constant access to their mother. However, the importance of colostrum and milk consumption for newborn animals and human babies goes beyond nutrition and the transfer of immunity. Numerous bioactive components such as growth factors, hormones, and oligosaccharides are enriched in colostrum and transition milk, which support the development of the intestinal tract and local immune system. The ability of a diet or an ingredient to satisfy the indispensable amino acid (IAA) requirements of an individual is a reflection of protein quality (PQ). The concept of PQ is gaining recognition in the pet food industry as a way to identify candidate ingredients for diet formulation. The objective of this report was to use IAA digestibility data from swine and cecectomized rooster assays to generate digestible IAA scores (defined herein as DIAAS -like values) to predict the PQ of ingredients used in dog diets. However, as PQ equation development relies on a reference IAA profile, which is intended to be based on the physiological requirements of a specific population, we sought to generate DIAAS-like values using IAA requirements established by the National Research Council (NRC) as well as practical IAA recommendations presented by the Association of American Feed Control Officials (AAFCO) and European Pet Food Industry Federation (FEDIAF), to assess how these profiles may affect PQ. In total, 30 animal (75 unique inputs) and 27 plant ingredients (94 unique inputs) satisfied all inclusion criteria to be used in the final data set. Ingredients were initially categorized as animal or plant, and further categorized based on AAFCO Official Common and Usual Names and Definitions of Feed Ingredients to allow for additional, more distinct comparisons to be made. Data were analyzed using PROC GLIMMIX in SAS, with ingredient reference as a random effect, and ingredient category, regulatory body, and life stage as fixed effects. As expected, differences were observed in DIAAS-like values for nearly all ingredients and ingredient categories when determined using NRC, AAFCO, or FEDIAF IAA requirements or recommendations as the reference pattern. Moreover, applying reference patterns based on NRC adult maintenance IAA requirements consistently produced the lowest DIAAS-like values. Ultimately, while future studies assessing PQ should utilize NRC minimal requirements, individual ingredient and ingredient category differences in DIAAS-like values when using AAFCO and FEDIAF recommendations underpin the different regulatory approaches to establishing dietary nutrient recommendations that exist globally and support the need for harmonization of dietary recommendations. For baled silages, production of clostridial fermentation products can be exacerbated by exceeding normal moisture targets (45% to 55%), and/or by the application of dairy slurry before harvest. Our objectives were to test a microbial inoculant as a mitigant of clostridial products in high-moisture, grass-legume (52% ± 13.8% cool-season grasses, 44.0% ± 14.0% legumes [predominately alfalfa]) baled silages in swards that were fertilized with dairy slurry. A secondary objective was to examine the effects of bale moisture and inoculation on the aerobic stability of these fermented silages following exposure to air. After the first-cutting was removed, three manure treatments were applied as a whole-plot factor: 1) control (no manure); 2) slurry applied immediately to stubble (63,250 L/ha); or 3) slurry applied after a 1-wk delay (57,484 L/ha). An interactive arrangement of bale moisture (64.1% or 48.4%) and inoculation (yes or no) served as a subplot term in the experiment. The inoculant contained both homolactic (Lactococcus lactis 0224) and heterolactic (Lactobacillus buchneri LB1819) bacteria. The experimental design was analyzed as a randomized complete block with four replications, and the study included 48 experimental units (1.2 × 1.2-m round bales). Total fermentation acids were affected (P ≤ 0.021) by slurry application strategies, but this was likely related to inconsistent bale moisture across slurry-application treatments. Concentrations of butyric acid were low, and there were no detectable contrasts comparing manure treatments (mean = 0.05%; P ≥ 0.645). Bale moisture affected all measures of fermentation, with bales made at 64.1% moisture exhibiting a more acidic final pH (4.39 vs. 4.63; P < 0.001), less residual water-soluble carbohydrates (2.1% vs. 5.1%; P < 0.001), as well as greater lactic acid (4.64% vs. 2.46%; P < 0.001), acetic acid (2.26% vs. 1.32%; P < 0.001), and total fermentation acids (7.37% vs. 3.97%; P < 0.001). Inoculation also reduced pH (4.47 vs. 4.56; P = 0.029), and increased acetic acid (1.97% vs. 1.61%; P < 0.001) and 1,2-propanediol (1.09% vs. 0.72%; P < 0.001) compared to controls. During a 34-d aerobic exposure period, maximum surface bale temperatures were not affected (P ≥ 0.186) by any aspect of treatment, likely due to the prevailing cool ambient temperatures; however, yeast counts were numerically lower in response to greater (P < 0.001) production of acetic acid that was stimulated by both high bale moisture and inoculation. Twenty-four gilts (PIC 337 × 1050, PIC Genus, Hendersonville, TN) with an initial body weight (BW) of 33.09 ± 1.33 kg were used to investigate the effects of benzoic acid (BA) and a Bacillus-based direct-fed microbial (DFM) on the nutrient metabolism and manure gas emissions of growing pigs. Pigs were blocked by BW, placed into metabolism stalls, and randomly assigned to one of four dietary treatments: basal control (PC), low nitrogen (NC), PC plus 0.3% BA (PC+BA; VevoVitall, DSM Nutritional Products), and PC plus 0.3% BA and 0.025% DFM (PC+BA+DFM; PureGro, DSM Nutritional Products). Pigs were fed a common diet from day 0 to 14, and the experimental diets were fed in two phases (day 14 to 28 and day 28 to 53). The experiment consisted of four collection periods, with each period subdivided into two subperiods to collect samples for gas emissions and nutrient balance. Firstly, manure samples were collected for 72 h. Twice daily, urine and feces were weighed, and urine pH was measured. After each period, manure was subsampled and taken to the lab to measure gas emissions. Secondly, urine and feces were quantitatively collected for 96 h to allow for measurement of nutrient digestibility (ATTD) and retention. Data were analyzed as repeated measures in SAS 9.4 (SAS Inst., Cary, NC) with fixed effects of treatment, collection period, and block. Pig was the experimental unit, and results were considered significant at P ≤ 0.05 and a tendency at 0.05 < P ≤ 0.10. Pigs fed PC+BA had the greatest ADG compared to pigs fed NC (P = 0.016), with intermediate ADG for pigs fed PC or PC+BA+DFM (P ≥ 0.148). The ATTD of dry matter, gross energy, P, and N did not differ between treatments (P ≥ 0.093). However, the ATTD of Ca was reduced in pigs fed PC+BA+DFM compared to pigs fed PC+BA (P = 0.012). Pigs fed PC+BA or NC excreted less urinary N compared to PC and PC+BA+DFM (P ≤ 0.034), which contributed to greater nitrogen retention in PC+BA compared to PC (P = 0.016). Furthermore, decreased manure pH from pigs fed PC+BA or NC resulted in lower ammonia (NH3 ) emissions compared to pigs fed PC+BA+DFM or PC. There was no effect of dietary treatment on manure hydrogen sulfide, methane, or carbon dioxide emissions. In conclusion, supplementing 0.3% BA improved N retention and reduced manure pH and NH3 emissions, similar to feeding pigs low N, but improved the ADG of pigs when compared to feeding a low N diet. Poor outcomes reflect low performance during the farrowing and lactation periods and unanticipated sow removals. Since the period around farrowing has the highest risk for sow health issues, monitoring of sows in that time-period will improve both welfare and productivity. The aim of this study was to identify the most relevant risk factors for predicting poor outcomes and the implication for sow welfare. Identifying these factors could potentially enable management interventions to decrease incidences of compromised welfare or poor performance. Data from 1,103 sows sourced from two nucleus herds were recorded for a range of variables investigated as potential predictors of poor outcomes in the farrowing house. Poor outcomes (scored as binary traits) reflected three categories in a sow’s lifecycle: farrowing, lactation, and removals. Univariate logistic regression was used to identify predictors in the first instance. Predictors from univariate analyses were subsequently considered together in multi-variate models. The least square means representing predicted probabilities of poor outcomes were then reported on the observed scale. Several predictors were significant across two different environments (farms) and for all three categories. These predictors included feed refusal (lack of appetite), crate fit, locomotion score, and respiration rate. Normal appetite compared to feed refusals reduced the risk of farrowing failure (13.5 vs. 22.2%, P = 0.025) and removals (10.4 vs. 20.4%, P < 0.001). Fit in the crate was significant (P < 0.001) for farrowing and lactation outcomes, and was more informative than parity. Sows with sufficient space had two to three times reduced risk of poor outcomes compared to restrictive crates relative to sow dimensions. Sows with good locomotion score pre-farrowing had two to three times less risk of farrowing failure (P = 0.025) and reduced piglet mortality (P < 0.001), weaned two piglets more relative to affected sows (P < 0.001), and were less likely to be removed before weaning (3.24 vs. 12.3%, P = 0.014). Sows with higher respiration rates had a significantly (P < 0.001) reduced risk of poor farrowing outcomes. This study demonstrated it is possible to predict poor outcomes for sows prior to farrowing, suggesting there are opportunities to decrease the risk of poor outcomes and increase overall sow welfare. Beef cattle consume more tall fescue [Lolium arundinaceum (Schreb.) Darbysh.] than any other perennial cool-season grass in North America. Tall fescue forms a mutualistic relationship with the fungus Epichloë coenophiala (Morgan-Jones and W. Cams) C.W. Bacon & Schardl, comb. nov.; these plants are often referred to as endophyte-infected tall fescue. Agronomically, endophyte infection is good; endophyte- infected plants resist biotic and abiotic stresses better than do endophyte-free plants. However, tall fescue plants infected with a common toxic endophyte produce the ergot alkaloids that cause tall fescue toxicosis. Several agronomic practices can eliminate or reduce the effects of tall fescue toxicosis. Renovating tall fescue fields infected with a common toxic endophyte to new cultivars infected with a novel (or nontoxic) endophyte eliminates the problem for that field. Although the cost to renovate currently exceeds US$600/ha, most economic analyses show it to be a good long-term investment, especially on fields with high forage production potential. Less effective, but popular, methods of dealing with tall fescue toxicosis include adding clovers (Trifolium spp.) or other forage species to the pastures to dilute the effects of the toxins, avoiding the use of high rates of nitrogen fertilizers, feeding supplements, and rotating cattle to warm-season forages in summer. Also, the alkaloid concentration in forage can be reduced by making it (drying) into hay. Recent research shows that animal tolerance to tall fescue toxicosis is possible, although there is still much to learn about this topic. Tall fescue toxicosis is likely a reality in the beef industry for some time to come. We contend that renovation of the most productive fields to cultivars infected with a novel endophyte coupled with the prospect of using tolerant livestock in scenarios where renovation is not possible will give innovative producers a competitive edge. A major role of the dairy cow is to convert low-quality plant materials into high-quality protein and other nutrients for humans. We must select and manage cows with the goal of having animals of the greatest efficiency matched to their environment. We have increased efficiency tremendously over the years, yet the variation in productive and reproductive efficiency among animals is still large. In part, this is because of a lack of full integration of genetic, nutritional, and reproductive biology into management decisions. However, integration across these disciplines is increasing as the biological research findings show specific control points at which genetics, nutrition, and reproduction interact. An ordered systems biology approach that focuses on why and how cells regulate energy and N use and on how and why organs interact through endocrine and neurocrine mechanisms will speed improvements in efficiency. More sophisticated dairy managers will demand better information to improve the efficiency of their animals. Using genetic improvement and animal management to improve milk productive and reproductive efficiency requires a deeper understanding of metabolic processes throughout the life cycle. Using existing metabolic models, we can design experiments specifically to integrate data from global transcriptional profiling into models that describe nutrient use in farm animals. The advent of high-throughput technologies to study an animal's genome, proteome, and metabolome (i.e., "omics" tools) constituted a setback to the use of reductionism in livestock research. More recent development of "next-generation sequencing" tools was instrumental in allowing in-depth studies of the microbiome in the rumen and other sections of the gastrointestinal tract. Omics, along with bioinformatics, constitutes the foundation of modern systems biology, a field of study widely used in model organisms (e.g., rodents, yeast, humans) to enhance understanding of the complex biological interactions occurring within cells and tissues at the gene, protein, and metabolite level. Application of systems biology concepts is ideal for the study of interactions between nutrition and physiological state with tissue and cell metabolism and function during key life stages of livestock species, including the transition from pregnancy to lactation, in utero development, or postnatal growth. Modern bioinformatic tools capable of discerning functional outcomes and biologically meaningful networks complement the ever-increasing ability to generate large molecular, microbial, and metabolite data sets. Simultaneous visualization of the complex intertissue adaptations to physiological state and nutrition can now be discerned. Studies to understand the linkages between the microbiome and the absorptive epithelium using the integrative approach are emerging. We present examples of new knowledge generated through the application of functional analyses of transcriptomic, proteomic, and metabolomic data sets encompassing nutritional management of dairy cows, pigs, and poultry. Published work to date underscores that the integrative approach across and within tissues may prove useful for fine-tuning nutritional management of livestock. The predominant cool-season forage in the southeastern United States is the tall fescue cultivar Kentucky 31 (KY31). Kentucky 31 possesses an endophyte (Epichloë coenophiala), which produces a family of toxins called ergot alkaloids. These toxins negatively affect the physiology of animals on consumption and result in the syndrome known as fescue toxicosis. Currently, the United States annually produces approximately 11.4 billion kg of beef, of which 25% originates in the southeastern region of the United States where forage systems frequently are tall fescue based. Cattle within this forage system exhibit reduced gains and reproductive performance. The result is a reduction in the nation's beef supply with annual revenue losses recently estimated at approximately US$1 billion. Our hypothesis is that exposure to these ergot alkaloids in conjunction with limited availability of nutrients decreases bull semen quality and fertility. Although the literature is clear that these toxins affect BW, body temperature, blood flow, hair growth, and female reproduction in cattle, their effect on bull reproduction and the mechanisms through which the toxins act are not well defined. Six studies published from 2004 to 2015 assessed bull growth, body composition, and semen quality of young beef bulls exposed to ergot alkaloids. If semen quality or fertility is altered, the mechanisms involved may be either direct effects of ergot alkaloids through neurotransmitter receptors or indirect effects such as inhibiting the release of prolactin (PRL). The possible effects of ergot alkaloids or PRL require establishing the presence or absence of dopamine, adrenergic, serotonin, or PRL receptors in the testis, epididymis, and sperm cell of the bull. The objective of this review is to relate our findings to the few previous studies conducted that evaluated the impact of fescue toxicosis on bull reproduction and to propose possible mechanisms of action for lowered semen quality. Forages are the base source of nutrition for any cow-calf operation. Forage types vary based on soil type and climate. Tall fescue (Lolium arundinaceum) is the most commonly used cultivated grass for grazing beef cattle in the United States. This cool-season perennial is easily established; is resistant to drought, insects, and nematodes; and has the ability to withstand heavy grazing pressure. Most tall fescue varieties are infected with the endophyte fungus (Neotyphodium coenophialum) that is essential for the plant's survival but detrimental to cattle performance. Ergot alkaloids are the generally accepted toxic agents produced by the fescue endophyte. Cattle that consume forages infected with this endophyte can develop fescue foot, fat necrosis, or fescue toxicosis. It is estimated that the beef industry loses over US$500 million annually due to fescue toxicosis through heat stress, reduced weight gain, suppressed appetite, and decreased reproductive performance. Other symptoms include a retained or rough hair coat and increased body temperature, which can be detrimental when animals are located in hot and or humid environments. Different forages and forage systems, feed additives, and animal management strategies have been tested through the years allowing the use of tall fescue in beef production systems while minimizing the adverse effects. An animal genetics approach needs to identify and select animals less susceptible to the ergot alkaloids. Research in this area reports that different cattle within the same herd can respond differently when grazing tall fescue, and evidence exists that breed type may also play a role in genetic tolerance to the negative effects on performance. Some studies have looked at the potential of identifying genetic markers that may assist in the selection of more resistant animals. From these studies, there is evidence that genetic variation does exist for resistance to the ergot alkaloids present when grazing endophyte-infected tall fescue. Forage management coupled with animal genetic selection could allow for more efficient use of tall fescue as a significant forage source in beef cattle systems. The objective of this study was to evaluate the acaricidal effect of 2 compounds extracted from Eysenhardtia polystachya in vitro, namely coatlines and matlalines. Each extract was obtained by aqueous extraction in a nitrogen atmosphere. Engorged adult females of an amitraz-resistant strain of Rhipicephalus (Boophilus) microplus were used to produce larvae and to carry out the adult immersion test (AIT), while larvae were assayed using the larval packet test (LPT). After exposure to coatlines or matlalines, the mortality of larvae and engorged females, as well as the reproductive parameters of adult ticks were assessed. Coatlines were not lethal against larvae and did not decrease reproductive parameters. Conversely, matlalines showed a 90 to 100% efficacy against adults and larvae at all concentrations tested and a reduction of 76.41 to 80.64% oviposition and a 48.02 to 54.86% reduction in egg hatchability. Therefore, the acaricidal activity of matlalines was more efficient. Further studies are required to elucidate both the mode of action of matlalines as well the structure-activity relationships responsible for the observed differential efficacy between these 2 related isoflavans against the cattle tick. Very low-protein (VLP) diets are beneficial for reducing the nitrogen excretion from swine production, but these diets depress the performance of pigs while supplemented with first four limiting amino acids (FFL; i.e. Lys, Met, Thr and Trp). The objective of this study was to investigate the effect of supplementation of Val and Ile above NRC (2012) levels on performance, thermal radiation and blood metabolites of pigs fed with VLP diets. Three-week-old weaned barrows (n = 40) were assigned to: positive control (PC): normal protein diet; negative control (NC): VLP diet containing FFL at NRC levels; HV: NC with Val above NRC; HI: NC with Ile above NRC; HVI: NC with both Val and Ile above NRC. After 5 weeks of data collection, blood samples were collected. Univariate GLM with Tukey post-hoc test was used to analyze the data (SPSS). Pigs fed with NC had a decreased average daily gain (ADG) and average daily feed intake (ADFI) compared with PC (P< 0.05). HVI pigs had less ADG than PC, but greater ADG than NC (P< 0.05). ADFI of pigs fed with HVI was similar to PC and it was greater than NC (P< 0.05). HVI pigs had a higher thermal radiation than NC (P< 0.05). While HVI pigs had greater blood glucose than PC and NC at baseline (P< 0.05), no differences among these groups were observed on postprandial blood glucose concentration. HVI pigs had less blood triglycerides than PC. All groups increased the blood cholesterol compared with PC (P< 0.05). There were no differences in blood urea nitrogen (BUN) for NC vs. PC; however, pigs fed with HVI reduced the BUN compared with NC (P< 0.05). In conclusion, supplementation of Val and Ile greater than NRC into VLP diets partially restored the performance and fully recovered the ADFI, reduced the blood triglycerides and BUN, and improved the glucose tolerance. Digital dermatitis (DD) is an infectious skin disease and a major cause of lameness that signifcantly impacts cattle productivity and welfare. However, DD does not always result in lameness and lameness scoring systems are not specifc to hoof pathologies. Digital dermatitis detection protocols could be improved by including gait traits most related to DD. The aims of this study were to 1) determine the association between DD M-stage (“M” for Mortellaro), locomotion, and gait traits: arched back (AB), asymmetric gait (AG), head bobbing (HB), tracking up (TU), and reluctance to bear weight (WB), and 2) determine which traits are most associated with DD. Cattle (n = 480) from three feedlots were enrolled. Locomotion score (LS) and gait traits were assessed as cattle walked four strides along a dirt alleyway. Next, cattle were restrained in a chute, each hind foot lifted, and DD M-stage (absent, active, or chronic) determined. The association between presence of DD, LS, and gait traits were scored independently (n = 291). For both LS and gait the lowest score represents normal and the highest score severely altered. Digital dermatitis presence was associated with higher LS (P < 0.001). Odds ratios (ORs) for cattle with DD being lame or moderately to severely lame were 8.0 (P < 0.001) and 10.1 (P < 0.001) times more than cattle without lesions. Cattle with active lesions had the greatest odds of being lame (OR = 9.4; P < 0.001). Digital dermatitis presence was associated with all gait traits (P < 0.001), where AG (OR = 5.5; P < 0.001) and WB (OR = 5.8; P < 0.001) had the greatest OR for classifying cattle with DD as having altered gait. The OR for cattle with active lesions having altered gait was greatest for WB which was 6.0 (P < 0.001) times greater than cattle without lesions. The OR for cattle with chronic lesions having altered gait was greatest for AG being 6.5 (P < 0.001) times more than cattle without lesions. All gait traits had low sensitivity (Se) for detecting cattle with DD and varied from 6.7% to 55.8%. Locomotion score (Se 55.8%) and AG (Se 44.2%) were most predictive with positive predictive values of 76.6% and 74.3%, respectively. Specifcity for all traits ranged from 94.1% for LS to 98.4% for WB with negative predictive values of 72.1% and 68.9%, respectively. This experiment was designed to examine the effects of a dietary supplementation of polysaccharides-rich noni (Morinda citrifolia L.) fruit extract (NFP) on the anti-oxidant enzyme activities, cytokines level, and expression of corresponding genes in blood of cashmere goats. Twelve castrated, 2-yr-old male cashmere goats (45.44 ± 3.30 kg of BW ± SD) were used in a 2 × 2 crossover design: the basal diet with or without (CON) supplementation of NFP at 4 g per kg DM (0.4%). Each period lasted for 29 d, including 1 wk for diet transition, 20 d for adaptation, and the last 2 d for sampling. The results showed that NFP supplementation increased (P < 0.05) the levels of nitric oxide, interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α), and the activities of catalase (CAT), glutathione peroxidase (GPx), thioredoxin reductase (TrxR), and total superoxide dismutase (T-SOD) in serum. The expressions of CAT, GPx4, TrxR, SOD1, IL-6, and TNF-α genes were upregulated (P < 0.05), whereas the levels of malondialdehyde (P = 0.015) and reactive oxygen species (P = 0.051) in serum were reduced. The body weight gain of goats was increased (P = 0.006) with a nonsignifcant increase of feed intake with NFP supplementation. The objective of this study was to evaluate the effects of two rumen-native microbial feed supplements (MFS) on milk production, milk composition, and feed effciency. A total of 90 multiparous cows between 40 and 60 d in milk were enrolled in a randomized block design study. Within each block (baseline milk yield), cows were randomly assigned to: control (no microbial feed supplementation), MFS1 (0.33 g/kg total mixed ration [TMR] of an MFS containing a minimum of Clostridium beijerinckii at 2 × 106 CFU/g and Pichia kudriavzevii at 2 × 107 CFU/g), or MFS2 (0.33 g/kg TMR of a MFS containing a minimum of C. beijerinckii at 2 × 106 CFU/g, P. kudriavzevii at 2 × 107 CFU/g, Ruminococcus bovis at 2 × 107 CFU/g, and Butyrivibrio fbrisolvens at 2 × 107 CFU/g). Cows were housed in a single group and fed the study diets ad libitum for 270 d. Individual milk yield was recorded using electronic milk meters, and milk fat and protein were measured using optical in-line analyzers at each of two daily milkings. Treatment and treatment by time effects were assessed through multiple linear regression analyses. Treatment effects were observed for milk and energy-corrected milk (ECM) yields, milk fat and protein yields and concentrations, dry matter intake (DMI), and feed effciency; those effects were conditional to time for milk yield, DMI, and feed effciency. Overall, milk, ECM, fat, and protein yields were higher for MFS2 compared with control cows (+3.0, 3.7, 0.12, and 0.12 kg/d, respectively). Compared with MFS1, milk yield was higher and protein yield tended to be higher for MFS2 cows (+2.9 and 0.09 kg/d, respectively). In contrast, MFS1 cows produced 0.17 and 0.08 units of percentage per day more fat and protein than MFS2 cows, and 0.07 units of percentage per day more protein than control cows. Dry matter intake and feed effciency were higher for MFS2 cows compared with MFS1 cows (+1.3 kg/d and 0.06, respectively), and feed effciency was higher for MFS2 cows compared with control cows (+0.04). Where observed, treatment by time effects suggest that the effects of MFS2 were more evident as time progressed after supplementation was initiated. No effects of microbial supplementation were observed on body weight, body condition score, somatic cell count, or clinical mastitis case incidence. The aim of this experiment was to investigate the effects of dietary crude protein (CP) contents and crystalline amino acids (CAA) supplementation patterns on growth performance, intestinal histomorphology, and immune response in weaned pigs under clean (CSC) or unclean sanitary conditions (USC). A total of 144 weaned pigs (6.35 ± 0.63 kg body weight) were assigned to 6 treatments in a 3 × 2 factorial arrangement based on CP content and sanitary conditions using a randomized complete block design, giving 8 replicates per treatment with 3 pigs per pen. Pigs were fed one of three diets for 21 d: one high CP (HCP ; 22%) and two low CP (LCP ; 19%) diets supplemented with 9 indispensable AA or only 6 AA (Lys, Met, Thr, Trp, Val, and Ile) as CAA. The CSC room was washed weekly, whereas the USC room had sow manure spread in the pens and was not washed throughout the experiment. Body weight and feed disappearance were recorded weekly. Blood was sampled from 1 pig per pen weekly, and the same pig was euthanized for jejunal tissues sampling on day 21. Pigs raised under USC had reduced (P < 0.05) average daily gain (ADG) and gain to feed ratio (G:F) in week 2, but contrary results that greater (P < 0.05) ADG and G:F were found in pigs under USC in week 3. Overall, there was an interaction where G:F did not differ between HCP and LCP under CSC, however, LCP decreased (P < 0.05) G:F compared to HCP under USC. Pigs fed the HCP diet had higher (P < 0.05) fecal scores than those fed the LCP diets throughout the experiment. Pigs fed the LCP had higher (P < 0.05) villus height to crypt depth ratio than those fed the HCP. An interaction was observed where goblet cell density in the jejunum was higher (P < 0.05) in pigs fed LCP than HCP under CSC, but no difference was found between HCP and LCP under USC. Different CAA supplementation patterns did not influence both growth performance and histomorphology. Pigs raised under USC had greater (P < 0.05) plasma interleukin (IL)-10 and IL-6 concentrations and reduced (P < 0.05) plasma tumor necrosis factor-alpha concentration. Also, the LCP diets resulted in a greater (P < 0.05) plasma IL-10 concentration. In conclusion, overall growth performance did not differ between HCP and LCP under CSC, but LCP diets reduced G:F under USC. Feeding growing-finishing pigs supplemental fat is a common practice in the swine industry and can result in improved feed efficiency and reduced feed intake; however, dietary lipids also play a key role in determining pork quality. Objectives of the study were to evaluate the effects of feeding graded levels of high oleic soybean oil (HOSO) on loin and belly quality. A total of 288 pig raised in two separate blocks (144 pigs each) were assigned to one of four diets containing either 25% dried distiller's grains with solubles (DDGS), 2% high oleic soybean oil (HOSO 2 ), 4% high oleic soybean oil (HOSO 4 ), or 6% high oleic soybean oil (HOSO6). Following the conclusion of the feeding trial, 144 pigs were slaughtered at the University of Illinois Meat Science Laboratory. Following fabrication, loins were collected for the evaluation of fresh quality measurements and color stability. Belly quality and fatty acid composition were evaluated using skin-on natural fall bellies. There were no differences (P ≥ 0.11) in pH, visual color, lightness (L *), drip loss, or WBSF among dietary treatments. However, visual marbling was increased (P ≤ 0.01) in loin chops from pigs fed HOSO4 and HOSO6 treatments compared with chops from pigs fed the DDGS dietary treatment. Additionally, loin chops were more red (a *) (P ≤ 0.01) from pigs fed HOSO diets when compared with pigs fed DDGS. Extractable lipid was decreased (P ≤ 0.01) in fresh loin chops from pigs fed DDGS and HOSO2 diets compared with pigs fed HOSO6. There were no differences (P ≥ 0.75) in trained sensory tenderness, juiciness, or flavor for loin chops from pigs fed different dietary treatments. Pork fatty acid composition was altered by dietary HOSO inclusion, with pigs fed DDGS having (P ≤ 0.01) the greatest concentration of C16:0 and was decreased with increasing levels of HOSO inclusion. Inversely, the percentage of C18:1 n -9 was least (P ≤ 0.01) in pigs fed DDGS and increased with increasing levels of HOSO inclusion. Pigs fed DDGS produced wider (P ≤ 0.03) and thinner (P ≤ 0.04) bellies with reduced flop distance compared with pigs fed HOSO diets. Overall, HOSO diets did not negatively affect fresh loin quality or sensory traits of loin chops. The objective of this study was to examine the effects of diet energy density on ranking for dry matter intake (DMI), residual feed intake (RFI), and greenhouse gas emissions. Forty-two mature, gestating Angus cows (600 ± 69 kg body weight [ BW ]; body condition score [ BCS ] 5.3 ± 1.1) with a wide range in DMI expected progeny difference (−1.38 to 2.91) were randomly assigned to two diet sequences; forage then concentrate (FC) or concentrate then forage (CF). The forage diet consisted of long-stem native grass hay plus protein supplement (HAY; 1.96 Mcal ME/kg DM). The concentrate diet consisted of 35% chopped grass hay and 65% concentrate feeds on a dry matter basis (MIX; 2.5 Mcal ME/kg DM). The GreenFeed Emission Monitoring system was used to determine carbon dioxide (CO 2 ), oxygen (O 2 ), and methane (CH 4 ) flux. Cow performance traits, ultrasound back fat and rump fat, feed DMI, and gas flux data were analyzed in a crossover design using a mixed model including diet, period, and sequence as fixed effects and pen and cow within sequence as random effects. For all measured traits excluding DMI, there was a diet × sequence interaction (P < 0.05). The correlation between MIX and HAY DMI was 0.41 (P = 0.067) and 0.47 (P = 0.03) for FC and CF sequences, respectively. There was no relationship (P > 0.66) between HAY and MIX average daily gain (ADG), regardless of sequence. Fifty-seven percent of the variation in DMI was explained by metabolic BW, ADG, and BCS for both diets during the first period. During the second period, the same three explanatory variables accounted for 38% and 37% of the variation in DMI for MIX and HAY diets, respectively. The negative relationship between BCS and DMI was more pronounced when cows consumed the MIX diet. There was no relationship between MIX and HAY RFI, regardless of sequence (P > 0.18). During the first period, correlations for CO2, CH4, and O2 with MIX DMI were 0.69, 0.81, and 0.56 (P ≤ 0.015), respectively, and 0.76, 0.74, and 0.64 (P < 0.01) with HAY DMI. During the second period, correlations for CO2, CH4, and O2 with MIX DMI were 0.62, 0.47, and 0.56 (P ≤ 0.11), respectively. However, HAY DMI during the second period was not related to gas flux (P > 0.47). Results from this experiment indicate that feed intake of two energy-diverse diets is moderately correlated while ADG while consuming the two diets is not related. Our previous study found that dietary nucleotide supplementation, including adenosine 5ʹ-monophosphate (AMP), could increase AMP content in sow milk and promote piglet growth, but its effects on placental efficiency and piglet vitality remain unknown. This experiment aimed to investigate the effects of dietary AMP or its metabolite adenosine (ADO) supplementation on sow reproductive performance and placental angiogenesis. A total of 135 sows with a similar farrowing time were blocked by backfat and body weight (BW) at day 65 of gestation and assigned to one of three dietary treatment groups (n = 45 per treatment): basal diet, basal diet supplemented with 0.1% AMP or 0.1% ADO, respectively. Placental analysis and the characteristics of sows and piglets unveiled that compared with control (CON) group, AMP or ADO supplementation could improve sow placental efficiency (P < 0.05) and newborn piglet vitality (P < 0.05), increase piglet birth weight (P < 0.05), and reduce stillbirth rate (P < 0.05). More importantly, AMP or ADO supplementation could increase the contents of AMP, ADO, and their metabolites in placentae (P < 0.05). Meanwhile, AMP or ADO supplementation could also increase placental vascular density (P < 0.05) and the expression of vascular endothelial growth factor A (P < 0.05), as well as promote the migration and tube formation of porcine iliac artery endothelial cells (P < 0.05). The microbial composition resemblance among individuals in a group can be summarized in a square covariance matrix and fitted in linear models. We investigated eight approaches to create the matrix that quantified the resemblance between animals based on the gut microbiota composition. We aimed to compare the performance of different methods in estimating trait microbiability and predicting growth and body composition traits in three pig breeds. This study included 651 purebred boars from either breed: Duroc (n = 205), Landrace (n = 226), and Large White (n = 220). Growth and body composition traits, including body weight (BW), ultrasound backfat thickness (BF), ultrasound loin depth (LD), and ultrasound intramuscular fat (IMF) content, were measured on live animals at the market weight (156 ± 2.5 d of age). Rectal swabs were taken from each animal at 158 ± 4 d of age and subjected to 16S rRNA gene sequencing. Eight methods were used to create the microbial similarity matrices, including 4 kernel functions (Linear Kernel, LK ; Polynomial Kernel, PK ; Gaussian Kernel, GK ; Arc-cosine Kernel with one hidden layer, AK1), 2 dissimilarity methods (Bray-Curtis, BC ; Jaccard, JA), and 2 ordination methods (Metric Multidimensional Scaling, MDS ; Detrended Correspondence analysis, DCA). Based on the matrix used, microbiability estimates ranged from 0.07 to 0.21 and 0.12 to 0.53 for Duroc, 0.03 to 0.21 and 0.05 to 0.44 for Landrace, and 0.02 to 0.24 and 0.05 to 0.52 for Large White pigs averaged over traits in the model with sire, pen, and microbiome, and model with the only microbiome, respectively. The GK, JA, BC, and AK1 obtained greater microbiability estimates than the remaining methods across traits and breeds. Predictions were made within each breed group using four-fold cross-validation based on the relatedness of sires in each breed group. The prediction accuracy ranged from 0.03 to 0.18 for BW, 0.08 to 0.31 for BF, 0.21 to 0.48 for LD, and 0.04 to 0.16 for IMF when averaged across breeds. The BC, MDS, LK, and JA achieved better accuracy than other methods in most predictions. Overall, the PK and DCA exhibited the worst performance compared to other microbiability estimation and prediction methods. The current study shows how alternative approaches summarized the resemblance of gut microbiota composition among animals and contributed this information to variance component estimation and phenotypic prediction in swine. The objective of the present study was to evaluate the productive performance and grazing behavior of 25 primiparous and 44 multiparous Holstein cows in a pasture-based automatic milking system (AMS) while experiencing heat stress (defined as a temperature–humidity index [THI] ≥ 68). Productive traits were analyzed according to the THI from days 0, −1, −2, and −3 in relation to the milking day, and grazing behaviors (expressed as the percentage of daily observation time) were related to the average THI only on the day of observation. Milk yield was not associated with the THI on day 0, but a significant linear relationship was found with the THI on the three previous days, decreasing approximately 0.18 kg (primiparous) and 0.40 kg (multiparous) per THI unit increment. In contrast, for multiparous cows only, the milking frequency was positively associated with the THI on the day of evaluation but not on the previous days, increasing 0.01 milking/THI unit increments. Additionally, for each unit of THI increment, cows spent 0.14% more time standing, whereas they exhibited a decrease in grazing, lying, and ruminating behaviors time by 0.30%, 0.04%, and 0.70%, respectively, for both parities. In conclusion, milk loss was related to heat stress conditions from the previous days, but not milking frequency, which increased with the THI of the same milking day. The beneficial effect of elevated concentrations of copper (Cu) on growth performance of pigs has been already demonstrated; however, their mechanism of action is not fully discovered. The objective of the present experiment was to investigate the effects of including Cu from copper sulfate (CuSO4) or monovalent copper oxide (Cu2O) in the diet of growing pigs on oxidative stress, inflammation, gene abundance, and microbial modulation. We used 120 pigs with initial body weight (BW) of 11.5 ± 0.98 kg in 2 blocks of 60 pigs, 3 dietary treatments, 5 pigs per pen, and 4 replicate pens per treatment within each block for a total of 8 pens per treatment. Dietary treatments included the negative control (NC) diet containing 20 mg Cu/kg and 2 diets in which 250 mg Cu/kg from CuSO4 or Cu2O was added to the NC. On day 28, serum samples were collected from one pig per pen and this pig was then euthanized to obtain liver samples for the analysis of oxidative stress markers (Cu/Zn superoxide dismutase, glutathione peroxidase, and malondialdehyde, MDA). Serum samples were analyzed for cytokines. Jejunum tissue and colon content were collected and used for transcriptomic analyses and microbial characterization, respectively. Results indicated that there were greater (P < 0.05) MDA levels in the liver of pigs fed the diet with 250 mg/kg CuSO4 than in pigs fed the other diets. The serum concentration of tumor necrosis factor-alpha was greater (P < 0.05) in pigs fed diets containing CuSO4 compared with pigs fed the NC diet or the diet with 250 mg Cu/kg from Cu2O. Pigs fed diets containing CuSO4 or Cu2O had a greater (P < 0.05) abundance of genes related to the intestinal barrier function and nutrient transport, but a lower (P < 0.05) abundance of pro-inflammatory genes compared with pigs fed the NC diet. Supplementing diets with CuSO4 or Cu2O also increased (P < 0.05) the abundance of Lachnospiraceae and Peptostreptococcaceae families and reduced (P < 0.05) the abundance of the Rikenellaceae family, Campylobacter, and Streptococcus genera in the colon of pigs. In conclusion, adding 250 mg/kg of Cu from CuSO4 or Cu2O regulates genes abundance in charge of the immune system and growth, and promotes changes in the intestinal microbiota; however, Cu2O induces less systemic oxidation and inflammation compared with CuSO4. Precision feeding (PF) aims to provide the right amount of nutrients at the right time for each animal. Lactating sows generally receive the same diet, which either results in insufficient supply and body reserve mobilization, or excessive supply and high nutrient excretion. With the help of online measuring devices, computational methods, and smart feeders, we introduced the first PF decision support system (DSS) for lactating sows. Precision (PRE) and conventional (STD) feeding strategies were compared in commercial conditions. Every day each PRE sow received a tailored ration that had been computed by the DSS. This ration was obtained by blending a diet with a high AA and mineral content (13.00 g/kg SID Lys, 4.50 g/kg digestible P) and a diet low in AAs and minerals (6.50 g/kg SID Lys, 2.90 g/kg digestible P). All STD sows received a conventional diet (10.08 g/kg SID Lys, 3.78 g/kg digestible P). Before the trial, the DSS was fitted to farm performance for the prediction of piglet average daily gain (PADG) and sow daily feed intake (DFI), with data from 1,691 and 3,712 lactations, respectively. Sow and litter performance were analyzed for the effect of feeding strategy with ANOVA, with results considered statistically significant when P < 0.05. The experiment involved 239 PRE and 240 STD sows. DFI was similarly high in both treatments (PRE: 6.59, STD: 6.45 kg/d; P = 0.11). Litter growth was high (PRE: 2.96, STD: 3.06 kg/d), although it decreased slightly by about 3% in PRE compared to STD treatments (P < 0.05). Sow body weight loss was low, although it was slightly higher in PRE sows (7.7 vs. 2.1 kg, P < 0.001), which might be due to insufficient AA supply in some sows. Weaning to estrus interval (5.6 d) did not differ. In PRE sows SID Lys intake (PRE: 7.7, STD: 10.0 g/kg; P < 0.001) and digestible P intake (PRE: 3.2, STD: 3.8 g/kg; P < 0.001) declined by 23% and 14%, respectively, and feed cost decreased by 12%. Manure N from cattle contributes to nitrate leaching, nitrous oxide, and ammonia emissions. Measurement of manure N outputs on commercial beef cattle operations is laborious, expensive, and impractical; therefore, models are needed to predict N excreted in urine and feces. Building robust prediction models requires extensive data from animals under different management systems worldwide. Thus, the study objectives were to 1) collate an international dataset of N excretion in feces and urine based on individual observations from beef cattle; 2) determine the suitability of key variables for predicting fecal, urinary, and total manure N excretion; and 3) develop robust and reliable N excretion prediction models based on individual observation from beef cattle consuming various diets. A meta-analysis based on individual beef data from different experiments was carried out from a raw dataset including 1,004 observations from 33 experiments collected from 5 research institutes in Europe (n = 3), North America (n = 1), and South America (n = 1). A sequential approach was taken in developing models of increasing complexity by incrementally adding significant variables that affected fecal, urinary, or total manure N excretion. Nitrogen excretion was predicted by fitting linear mixed models with experiment as a random effect. Simple models including dry matter intake (DMI) were better at predicting fecal N excretion than those using only dietary nutrient composition or body weight (BW). Simple models based on N intake performed better for urinary and total manure N excretion than those based on DMI. A model including DMI and dietary component concentrations led to the most robust prediction of fecal and urinary N excretion, generating root mean square prediction errors as a percentage of the observed mean values of 25.0% for feces and 25.6% for urine. Complex total manure N excretion models based on BW and dietary component concentrations led to the lowest prediction errors of about 14.6%. In conclusion, several models to predict N excretion already exist, but the ones developed in this study are based on individual observations encompassing larger variability than the previous developed models. In addition, models that include information on DMI or N intake are required for accurate prediction of fecal, urinary, and total manure N excretion. Substantial economic losses in animal agriculture result from animals experiencing heat stress (HS). Pigs are especially susceptible to HS, resulting in reductions in growth, altered body composition, and compromised substrate metabolism. In this study, an artificial high-intensity sweetener and capsaicin (CAPS-SUC; Pancosma, Switzerland) were supplemented in combination to mitigate the adverse effects of HS on pig performance. Forty cross-bred barrows (16.2 ± 6 kg) were assigned to one of five treatments: thermal neutral controls (TN) (22 ± 1.2 °C; 38%–73% relative humidity) with ad libitum feed, HS conditions with ad libitum feed with (HS+) or without (HS−) supplementation, and pair-fed to HS with (PF+) or without supplementation (PF−). Pigs in heat-stressed treatments were exposed to a cyclical environmental temperature of 12 h at 35 ± 1.2 °C with 27%–45% relative humidity and 12 h at 30 ± 1.1 °C with 24%–35% relative humidity for 21 d. Supplementation (0.1 g/kg feed) began 7 d before and persisted through the duration of environmental or dietary treatments (HS/PF), which lasted for 21 d. Rectal temperatures and respiration rates (RR; breaths/minute) were recorded thrice daily, and feed intake (FI) was recorded daily. Before the start and at the termination of environmental treatments (HS/PF), a muscle biopsy of the longissimus dorsi was taken for metabolic analyses. Blood samples were collected weekly, and animals were weighed every 3 d during treatment. Core temperature (TN 39.2 ± 0.02 °C, HS− 39.6 ± 0.02 °C, and HS+ 39.6 ± 0.02 °C, P < 0.001) and RR (P < 0.001) were increased in both HS− and HS+ groups, but no difference was detected between HS− and HS+. PF− pigs exhibited reduced core temperature (39.1 ± 0.02 °C, P < 0.001), which was restored in PF+ pigs (39.3 ± 0.02 °C) to match TN. Weight gain and feed efficiency were reduced in PF− pigs (P < 0.05) but not in the PF+ or the HS− or HS+ groups. Metabolic flexibility was decreased in the HS− group (−48.4%, P < 0.05) but maintained in the HS+ group. CAPS-SUC did not influence core temperature or weight gain in HS pigs but did restore core temperature, weight gain, and feed efficiency in supplemented PF pigs. In addition, supplementation restored metabolic flexibility during HS and improved weight gain and feed efficiency during PF, highlighting CAPS-SUC's therapeutic metabolic effects. Skeletal muscle growth is largely dependent on the proliferation and differentiation of muscle-specific stem cells known as satellite cells (SC). Previous work has shown that dietary inclusion of the vitamin D3 metabolite, 25-hydroxycholecalciferol (25OHD3), also called calcidiol, can promote skeletal muscle growth in post-hatch broiler chickens. Improving vitamin D status of broiler breeder hens by feeding 25OHD3 in addition to vitamin D3 has also been shown to positively impact progeny. Yet, whether combined pre- and post-hatch supplementation with 25OHD3 produces an additive or synergistic SC-mediated, skeletal muscle growth response remains unanswered. To evaluate the effect of combined maternal and post-hatch dietary 25OHD3 supplementation on the growth and SC mitotic activity of the Pectoralis major (PM) muscles in broiler chickens, a randomized complete block design experiment with the main effects of maternal diet (MDIET) and post-hatch diet (PDIET) arranged in a 2 × 2 factorial treatment structure was conducted. From 25 to 36 wk of age, broiler breeder hens were fed 1 of 2 MDIET formulated to provide 5,000 IU D3 (MCTL) or 2,240 IU of D3 + 2,760 IU of 25OHD3 per kg of feed (M25OHD3). Their male broiler chick offspring (n = 400) hatched from eggs collected from 35 to 36 wk of age were reared in raised floor pens. Broilers were fed 1 of 2 PDIET formulated to provide 5,000 IU of D3 per kg of feed (PCTL) or 2,240 IU of D3 + 2,760 IU of 25OHD3 per kg of feed (P25OHD3). Muscle was collected at days 4, 8, 15, 22, and 29 and stored until immunofluorescence analysis. Data were analyzed as a 2-way ANOVA with SAS GLIMMIX. Dietary 25OHD3 was effectively transferred from hen plasma to egg yolks (P = 0.002) and to broiler progeny plasma (days 4 to 22; P ≤ 0.044). Including 25OHD3 in either MDIET or PDIET altered PM hypertrophic growth prior to day 29 (P ≥ 0.001) and tended to reduce Wooden Breast severity (P ≤ 0.089). Mitotic SC populations were increased in PM of MCTL:P25OHD3 and M25OHD:PCTL-fed broilers at d 4 (P = 0.037). At d 8, the PM mitotic SC populations were increased 33% by P25OHD3 (P = 0.054). The results of this study reveal that combined maternal and post-hatch 25OHD3 supplementation does not produce additive or synergistic effects on SC-mediated broiler muscle growth. However, vitamin D status improvement through dietary 25OHD3 inclusion in either the maternal or post-hatch diet stimulated broiler breast muscle growth by increasing proliferating SC populations. Several studies have evaluated the effects of the dietary application of exogenous alpha-amylase preparations (AMA) as a strategy to increase total tract starch digestibility (TTSD) and milk yield (MY) in dairy cows, but the results have been inconsistent. Thus, the objective of this study was to evaluate the effects of the dietary application of AMA on the performance, digestibility, and rumen fermentation of lactating dairy cows using a meta-analytic method. A total of 18 peer-reviewed manuscripts (N = 32 treatment comparisons) from 2003 to 2019 were systematically identified following the PRISMA method. The weighted raw mean differences between dietary AMA and control treatments were compared with a robust variance estimation. Likewise, diet characteristics like crude protein (CP) content, NDF content, starch content, days in milk (DIM), experimental design (Latin square and continuous), and AMA dose (0 to 732 Kilo Novo units [KNU]/kg TMR) were used as covariates in a meta-regression, subgrouping, and dose–response analysis. Compared to the control, dietary AMA increased (P < 0.05) DM digestibility (69.32% vs. 68.30%), TTSD (94.62% vs. 94.10%), milk protein concentration and yield (3.11% vs. 3.08%; 1.14 vs. 1.10 kg/d) and tended to increase (P = 0.09) fat-corrected milk (35.96 vs. 35.10 kg/d), but no effects were observed on DM intake (22.99 vs. 22.90 kg/d) and feed efficiency (1.50 vs. 1.48). Dietary AMA tended (P = 0.10) to reduce rumen pH (6.27 vs. 6.30). Both the enzyme dose and DIM strongly influenced (P < 0.05) the effects of AMA on digestibility and performance. The dose–response analysis revealed that feeding 600 KNU/kg to high-producing early lactation (< 70 DIM) dairy cows increased FCM and milk protein. Accounting for the type of experimental design was associated with a lower between-studies-variance among comparisons. Overall, this meta-analysis supports the hypothesis that dietary AMA supplementation is associated with a better lactational performance in dairy cows. The first objective was to investigate the effects of feeding rumen-protected methionine (RPM) during a heat stress (HS) challenge on abundance and phosphorylation of mechanistic target of rapamycin (mTOR)-related signaling proteins in mammary gland. The second objective was to investigate how HS and RPM may modulate the response of mammary gland explants to an inflammatory challenge using lipopolysaccharide (LPS). Thirty-two multiparous, lactating Holstein cows (184 ± 59 DIM) were randomly assigned to 1 of 2 environmental treatment groups, and 1 of 2 dietary treatments [TMR with RPM (Smartamine M; Adisseo Inc.; 0.105% DM as top dress) or TMR without RPM (CON)] in a crossover design. There were two periods with two phases per period. In phase 1 (9 d), all cows were in thermoneutral conditions (TN) and fed ad libitum. During phase 2 (9 d), group 1 (n = 16) cows were exposed to HS using electric heat blankets, whereas group 2 cows (n = 16) remained in TN but were pair-fed to HS counterparts to control for DMI decreases associated with HS. After a washout period (14 d), the study was repeated (period 2). Environmental treatments were inverted in period 2 (sequence), whereas dietary treatments remained the same. Mammary tissue was harvested via biopsy at the end of both periods. Tissue was used for protein abundance analysis and also for incubation with 0 or 3 μg/mL of LPS for 2 h and subsequently used for mRNA abundance. Data were analyzed using PROC MIXED in SAS. Analysis of protein abundance data included the effects of diet, environment and their interaction, and period and sequence to account for the crossover design. The explant data model also included the effect of LPS and its interaction with environment and diet. Abundance of phosphorylated mTOR and ratio of phosphorylated eukaryotic translation elongation factor 2 (p-EEF2) to total EEF2 in non-challenged tissue was greater with RPM supplementation (P = 0.04 for both) and in both cases tended to be greater with HS (P = 0.08 for both). Regardless of RPM supplementation, incubation with LPS upregulated mRNA abundance of IL8 , IL6 , IL1B , CXCL2 , TNF , NFKB1 , and TLR2 (P < 0.05). An environment × LPS interaction was observed for NFKB1 (P = 0.03); abundance was greater in LPS-treated explants from non-HS compared with HS cows. Abundance of CXCL2 , NFKB1 , NOS2 , NOS1 , and SOD2 was lower with HS (P < 0.05). Although LPS did not alter mRNA abundance of the antioxidant transcription factor NFE2L2 (P = 0.59), explants from HS cows had lower abundance of NFE2L2 (P  < 0.001) and CUL3 (P  = 0.04). Overall, RPM supplementation may alter mTOR activation in mammary tissue. Tenderness is considered as one of the most important quality attributes dictating consumers' overall satisfaction and future purchasing decisions of fresh beef. However, the ability to predict and manage tenderness has proven very challenging due to the numerous factors that contribute to variation in end-product tenderness. Proteomic profiling allows for global examination of differentially abundant proteins in the meat and can provide new insight into biological mechanisms related to meat tenderness. Hence, the objective of this study was to examine proteomic profiles of beef longissimus lumborum (LL) steaks varying in tenderness, with the intention to identify potential biomarkers related to tenderness. For this purpose, beef LL muscle samples were collected from 99 carcasses at 0 and 384 h postmortem. Based on Warner–Bratzler shear force values at 384 h, 16 samples with the highest (intermediate tender, IT) and lowest (very tender, VT) values were selected to be used for proteomic analysis in this study (n = 8 per category). Using tandem mass tag-based proteomics, a total of 876 proteins were identified, of which 51 proteins were differentially abundant (P < 0.05) between the tenderness categories and aging periods. The differentially identified proteins encompassed a wide array of biological processes related to muscle contraction, calcium signaling, metabolism, extracellular matrix organization, chaperone, and apoptosis. A greater (P < 0.05) relative abundance of proteins associated with carbohydrate metabolism and apoptosis, and a lower (P < 0.05) relative abundance of proteins involved in muscle contraction was observed in the VT steaks after aging compared with the IT steaks, suggesting that more proteolysis occurred in the VT steaks. This may be explained by the greater (P < 0.05) abundance of chaperonin and calcium-binding proteins in the IT steaks, which could have limited the extent of postmortem proteolysis in these steaks. In addition, a greater (P < 0.05) abundance of connective tissue proteins was also observed in the IT steaks, which likely contributed to the difference in tenderness due to added background toughness. The established proteomic database obtained in this study may provide a reference for future research regarding potential protein biomarkers that are associated with meat tenderness.
