Phagocytosis and killing of microorganisms by reactive oxygen radicals are important defence mechanisms of the immune system and it was shown that human monocytes (MO) are heterogeneous in exerting these functions. Previously, we described that human peripheral blood MO consist of a major subset of Fcγ-receptor-I (CD64)-positive cells exhibiting low accessory cell capacity but high phagocytic activity, and a minor subset of CD64-negative cells with dendritic cell (DC)-like high T cell accessory cell capacity but low phagocytic capacity. Recently, we could show that each subset itself further differs in the expression of the Fcγ-receptor-III (CD16) and T cell accessory activities resulting in four different subsets: two CD16 + subsets (CD64 + or CD64 - ) with high T cell stimulation capacity and two CD16 - subsets (CD64 + or CD64 - ) with low accessory activities. In the present study we demonstrate that these subsets also differ in their ability to phagocytose opsonized bacteria ( S. aureus and E. coli ) and in the generation of reactive oxygen species. Both CD64 + subsets (CD16 + or CD16 - ) exhibit high phagocytic activity accompanied by intracellular superoxide induction. Luminal-dependent (mainly myeloperoxidase (MPO)-mediated) chemiluminescence (CL) response to latex and FMLP (formylmethionylleucylphenylalanine) was also high in these cell populations. Phagocytic activity and modest CL response was shown in CD64 - /CD16 + but not in CD64 - /CD16 - cells, indicating that each subset except for CD64 - /CD16 - cells may engulf bacteria and exhibit MPO activity. Taken together, these data demonstrate further heterogeneity of peripheral blood MO in both, phagocytic activity and generation of reactive oxygen species indicating differences between the four subsets in this kind of defence mechanisms against pathogens.

Three different anti-CD26 monoclonal antibodies (mAbs) are described, which specifically inhibited proliferation of human T lymphocytes after stimulation with PHA, tetanus toxoid or soluble anti-CD3 mAb.Anti-CD26 mAbs induced in T cells a dose-dependent shift of the maximum of DNA synthesis, which was due to a transitory arrest of cells in the cell cycle.This cell cycle arrest was found to occur in the late phase of G 1 since the expression of the T cell activation markers CD25-, CD71-, and HLA-DR-positive cells was the same in anti-CD26 mAb-containing and control cultures.Propidium iodide staining further confirmed the assumption that the arrest occurs in the first round of the cell cycle before S phase cells were detectable.Because the cells were arrested before consuming IL-2 in the S phase, we observed in accumulation of IL-2 in anti-CD26 mAb-containing cultures, whereas IFN-gamma production by PHA-activated T lymphocytes was reduced.These data indicate that CD26 is involved in the processes of T cell activation and proliferation.

Agonistic and antagonistic properties of LPS and partial structures in the induction of cytokines are reviewed. Studies on structure-activity relationships of LPS and lipid A with human mononuclear cells reveal that S- and notably R-form LPS are very potent cytokine inducers. Synthetic E. coli lipid A is somewhat less active, whereas synthetic S. minnesota-type lipid A is significantly less active. Pentaacylated forms of lipid A are less potent than hexaacylated forms, and tetraacylated synthetic precursor Ia and bisacylated disaccharides and monosaccharides are completely inactive, indicating that a structure-dependent hierarchy of LPS and lipid A partial structures determines the monokine-inducing capacity in human mononuclear cells. Precursor Ia is a potent LPS antagonist. The mechanism of its inhibitory activity is shown to be due to competitive binding to cellular binding sites (receptors). Proinflammatory and antiinflammatory cytokines, receptor antagonists, and soluble cytokine receptors influence the cytokine-inducing activity of LPS, suggesting a complex regulatory network.

We have investigated the release of tumor necrosis factor-alpha (TNF-alpha) by human mononuclear cells (MNC) and isolated human monocytes/macrophages stimulated with S- and R-form lipopolysaccharide (LPS), natural lipid A, and natural and synthetic partial structures thereof. We found that LPS of Salmonella minnesota (S. min.) Rb2, which represents a partial structure of wildtype LPS of Salmonella abortus equi (S.a.e.) lacking the O-chain and parts of the outer core region, was the most active inducer of all substances tested, even more active than the wildtype LPS. Lipid A also induced the production of TNF-alpha by monocytes/macrophages but was less active than wildtype LPS. The natural Escherichia coli (E. coli) type hexaacyl lipid A (compound 506) was more active than the natural S. min. type heptaacyl lipid A (compound 516). The 1- and 4'-monodephospho partial structures (compounds 505 and 504) of E. coli lipid A were less active and represented the smallest structures tested that were able to induce TNF-alpha release by monocytes/macrophages. Synthetic tetraacyl lipid A precursor Ia of E. coli lipid A, lacking non-hydroxylated fatty acids (compound 406), and the monosaccharide precursor lipid X did not induce the release of TNF-alpha in MNC or isolated monocytes/macrophages. This might indicate that the ability of a lipid A structure to induce the release of TNF-alpha is closely connected with the conditions to be at least hexaacylated and/or to contain hydroxylated fatty acids. These results demonstrate a structure-dependent hierarchy of LPS and natural or synthetic partial structures in their capacity of inducing TNF-alpha release by monocytes/macrophages.

We examined the effect of interleukin 2 (IL 2) on the capacity of human monocytes to secrete reactive oxygen species triggered via Fc-gamma receptor (Fc gamma R) function as measured by chemiluminescence (CL). IL 2 did not activate highly purified (hp) monocytes to respond to Fc gamma R-mediated phagocytic stimulation with an enhanced respiratory burst activity, unless low numbers of T lymphocytes had been cocultured with hp monocytes. The degree of monocyte activation in peripheral blood mononuclear cells (PBMC) by IL2 was dependent on T lymphocytes to monocytes ratios as well as on IL 2 concentrations. Optimal Fc gamma R function was detected with IL 2 concentrations of 60 to 600 U/ml and one to two T lymphocytes per monocyte, whereas higher cellular ratios were suppressive. Supernatants from IL 2-treated PBMC contained interferon-gamma (IFN-gamma) and monocyte activating factor (MAF) activity (= enhanced monocytic Fc gamma R mediated CL). The secretion of both cytokine activities was strongly enhanced by a cooperative function of monocytes. The correlation of IL 2-induced secretion of IFN-gamma and MAF activity was striking; however, monoclonal anti-IFN-gamma antibodies failed to abrogate IL 2-stimulated and lymphocyte-dependent monocyte activation. Although IL 2 had no direct monocyte activating effect, pretreatment of hp monocytes with IL 2 led to a monocyte priming: subsequent coculture with autologous control T lymphocytes enhanced the monocyte Fc gamma R-mediated CL response. The priming of monocytes by IL 2 was dependent on the interaction of IL 2 with the monocytic IL 2 receptor, as shown by inhibition experiments with anti-IL 2 receptor monoclonal antibodies. Thus, we observed an IL 2-driven monocyte/T lymphocyte interaction leading to an increased Fc gamma R-mediated monocytic respiratory burst activity and to the secretion of a soluble MAF activity, but there were no detectable amounts of IFN-gamma.

A fibroblast proliferation assay was developed for the detection of interleukin 1 (IL 1).Proliferation was measured by thymidine incorporation and by staining of cellular proteins with crystal violet. Response of fibroblasts was optimal at cell numbers of 4,000 to 9,000 cells/ culture and an incubation period of four days. Serum content of the culture medium, ranging from 1 to 10 % fetal calf serum (FCS), enhanced the proliferative response in a concentrationdependent manner, while higher concentrations of FCS did not lead to further increase. Both detection methods were equally suitable for the measurement of IL 1 biological activity in purified and crude preparations. In contrast to the conventional thymocyte comitogenic assay, the fibroblasts in this assay did not proliferate in response to IL 2 or IL 6. Fibroblasts were weakly stimulated by recombinant (rec) tumor necrosis factor (rec TNF-α); they did, however, not proliferate in response to mitogens, lipopolysaccharide, rec granulocyte-macrophage colony stimulating factor (rec GM-CSF), macrophage-CSF, rec interferon-γ, insulin or transferrin. The detection of IL 1 activity by crystal violet staining of human dermal fibroblasts was easier and faster than by measurement of thymidine incorporation of fibroblasts or mouse thymocytes; without loss of sensitivity, the sample capacity of the IL 1 assay could be enhanced, and the use of experimental animals was avoided.
