Malaria, a parasitic disease of the tropics, is one of the major cause of morbidity and mortality, particularly in the tropics. Following the increased resistance of the primary causative parasites, Plasmodium sp, to the mainstream drug, artemisinin combination therapies (ACTs), combating malaria incidences, morbidity and mortality have remained elusive. Novel pyrano-benzodioxepin derivatives (DHA-PABA and DHA-LEVO) were synthesized and characterized using Fourier transform infrared (FT-IR) and nuclear magnetic resonance (NMR) spectrometries. The compounds were subjected to standard in vivo antimalarial screening (using chloroquine-sensitive strain) in mice, and the toxicity was also determined using a standard assay. The observed elevation in serum alkaline phosphatase and acid phosphatase activity in the untreated and the group administered lower doses of DHA-LEVO is an indication of the hepatic stage of the parasite in the experimental animal, which is accompanied by significant perturbation in the membrane of the hepatocyte leading to leakage of this enzyme out of the liver cells. The semisynthetic pyrano-benzodioxepin derivatives act rapidly by clearing the parasite load from the blood. The novel pyrano-benzodioxepin candidates containing endoperoxide functionality hold promise in the pursuit of new monotherapy drug candidates against the virulent strain of the plasmodium.

Peptic ulcer disease affects many people globally. With the increasing resistance to some orthodox antibiotics such as Clarithromycin and Metronidazole, it is important that new acceptable, safer and effective therapies are developed to manage this disease. Various herbal medicines have been used traditionally for the remedy of peptic ulcer disease (PUD), however scientific information with regards to their anti-peptic ulcer both in-vivo and in-vitro as well as clinical studies supporting their use is still inadequate. The Centre for Plant Medicine Research, (CPMR) Mampong-Akuapem, Ghana manufactures three herbal Products namely Enterica, Dyspepsia and NPK 500 capsules which are currently used for the remedy of PUD as a triple therapy at its out-patient clinic with promising effects. The aim of this review is to gather information from literature on the anti-ulcer properties, pharmacological, phytochemical constituents and related activities of herbal plants used at the CPMR for formulation of the triple herbal therapy. This review may, provide some scientific bases for the use of Enterica, Dyspepsia and NPK 500 capsules in the management of Peptic ulcer at the CPMR out-patient clinic. Methods Organization for the review involved the on and/or offline search for information from available literature using electronic data and scientific research information resources such as PubMed, Science Direct and Google scholar. Results In this review, fifteen ethno-medicinal plants used for the formulation of Enterica, Dyspepsia and NPK capsules have been discussed, presenting the description of the plants, composition and pharmacological activity. Interpretation Tables with the summary of reviewed medicinal plants with their anti-ulcer models and inference on possible mechanisms of action were drawn up. The mechanism(s) of action of individual plants and products (Enterica, Dyspepsia and NPK 500 capsules) must be further investigated and established experimentally in-vitro in addition to in-vivo pharmacological and clinical activity studies to confirm their use in the remedy of PUD.

Fowl adenoviruses (FAdVs) are a large group of viruses of different serotypes. They are responsible for inclusion body hepatitis, adenoviral gizzard erosion, and hepatitis hydropericardium syndrome. The present study presents a comprehensive overview of FAdVs in Egypt, with a focus on the epidemiological features of virus serotypes across the country. We conducted molecular investigation of multiple FAdV species based on the genetic signature of hypervariable regions 1–4 in the loop1 (L1) region of the hexon gene. Epidemiologically, the Nile Delta governorates showed high positivity of FAdVs, which were more commonly found in broilers than in layers. Genetically, species D and serotype 8a/E dominated, and the findings also revealed the emergence of new FAdV serotypes 1, 3, and 8b. The comparative analysis of hypervariable regions in the L1 region of the hexon gene revealed variables specific to each virus serotype. In silico predictions of L1 region revealed variations in the molecular structure and predicted the antigenic epitopes which may affect the cross-antigenicity between the different FAdV species and serotypes., Fowl adenovirus, Loop1, HVRs, Egypt.

Diabetes is a metabolic disorder which is characterised by high levels of blood glucose. Most of the oral drugs available today for the treatment of diabetes are associated with various side-effects. Herbal medicines are considered relatively safer alternatives and Gymnema sylvestre (GS) is one such known traditional medicinal plant widely used for the treatment of diabetes. In our previous work, we isolated active triterpene glycosides (TG) from Gymnema sylvestre (GS) and screened for yeast α-glucosidase inhibitory activity in vitro. The present study aims to use in silico techniques to understand and predict the inhibitory role of the isolated triterpene glycosides (TG); Gymnemic acid I, IV, VII and gymnemagenin against disaccharidase enzymes. enzyme kinetic analysis using Lineweaver-Burk plot indicated that TG competitively inhibited yeast α-glucosidase at IC50 concentration with Ki 0.0028 μM. TG also exhibited significant inhibitory activity against mammalian sucrase and maltase respectively, compared to control. Practical applications The molecular docking simulation reveals that TG is capable of docking well with crystallographic structures of the selected enzyme targets. Inhibition of α-glucosidases could delay the absorption of glucose in the blood during post-meal digestion. Thus the current study highlights the dietary intervention of TG towards the selected enzyme targets, thus making TG a potential nutraceutical candidate towards management of blood glucose., Gymnema sylvestre; Antidiabetic; in silico docking; Dissacharidases; Inhibition.

Consistent intake of legumes has been correlated with decreased possibility of developing colorectal cancer (CRC) due to the content of some phytochemicals like polyphenols. Bambara groundnut (BGN) is an underutilized crop with a rich nutritional profile, but have not been exploited for its nutraceutical and medicinal benefits. In this study, total polyphenol, flavonoid (flavonol and flavanol) content, antioxidant activity and cytotoxicity/antiproliferative properties of 70% ethanolic extracts of whole BGN, cotyledon and seed coat on Caco-2 and HT-29 colon cancer cells were evaluated. Seed coat had a significantly (p < 0.05) higher composition of total polyphenol, flavonol and flavan-3-ol (flavanol) compared to whole seed and cotyledon. Antioxidant activity determined with ferric reducing antioxidant power (FRAP), 2,2- azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and oxygen radical absorbance capacity (ORAC) assays, showed that seed coat with higher polyphenolic content had significantly (p < 0.05) greater antioxidant activity. BGN fractions demonstrated cytotoxic and antiproliferative effects against HT-29 and Caco-2 colon cancer cells in a dose-dependent manner, with seed coat and whole seed exhibiting greater cytotoxicity and higher antiproliferative activity and colon cancer cell inhibition. Extracts of the cotyledon also showed cytotoxic activity and hindered cancer cell growth/division but to a significantly (p < 0.05) lower magnitude. BGN parts indicated a greater cytotoxic effect and potential to slow down Caco-2 colon cancer cell growth and division over HT-29. This result provides new knowledge on the possible health benefits of BGN, as well as the potential for product development and may influence its consumption and utilisation.

Background and objective Intercropping is one of the sustainable intensification methods which reduces the demand for increased land resources. However, growing of two or more crops as an intercrop is not business as usual but it needs extensive knowledge about the nature of component crops to grow in harmony. Hence selection and evaluation of compatible legume species for intercropping with finger millet in the study area is necessary. Materials and methods A field experiment was conducted at two major finger millet producing areas of northwestern Ethiopia. Four legumes crops such as (sweet lupine, cowpea, haricot bean, and soybean) intercropped with finger millet. Results The analysis of variance showed that intercropping of finger millet with different legumes had yield advantage in both districts as compared to component crops. Land equivalent ratio was more than unity for all treatments in both districts except for finger millet intercropped with soybean at South Achefer district. At Guangua finger millet + cowpea and finger millet + soybean showed 27% and 12% yield advantage, respectively and also finger millet + cowpea preferred first by farmers for its high grain yield, soil fertility improvement and its suitability for animal feed. At South Achefer finger millet + sweet lupine and finger millet + cowpea showed 38% and 35% yield advantage respectively and also finger millet + sweet lupine preferred first by farmers for its high grain yield to make stew as a protein source, soil fertility improvement and its suitability for human and animal feed. Conclusion Therefore intercropping, finger millet + cowpea and finger millet + soybean at Guangua and finger millet + sweet lupine and finger millet + cowpea at South Achefer preferred by farmers and also had yield advantage as compared to component crops. Hence finger millet + cowpea and finger millet + soybean recommended for Guangua and similar agro-ecologies and finger millet + sweet lupine and finger millet + cowpea for South Achefer and similar agro-ecologies as 1st, 2nd option respectively., Farmers’ preference; Finger millet; Legumes; Land equivalent ratio; Multiple cropping.

The high decaying tendency of date palm juice limited its manifold application. Here, we developed a preservation technique for concentrated juice and formulated ready to serve (RTS) drinks. The collected date palm juice was heated at 85 °C for 5 min; scum was removed and preserved in the sterilized glass bottle. After that, this pasteurized juice was concentrated to 9%, 12%, 15%, 18% and 21% total soluble solids (TSS) levels and treated with 200 ppm of potassium metabisulfite (KMS) and stored at refrigerated temperature (4 °C). Among them, 18% TSS showed overall acceptability to sensory evaluations for colour, flavour, sweetness, TSS, acidity and microbial load. In contrast, heat treated date palm juice (9% TSS) was used to formulate RTS drinks containing 10%, 20%, 30% and 40% date palm juice, were preserved in sterilized bottles and stored at 30 °C. Results showed that RTS drinks containing 30% date palm juice secured the best colour, flavour, sweetness, TSS, acidity, microbial load and overall acceptability. Furthermore, no significant change was observed in TSS, acidity, microbial load up to three and six months for concentrated juice and RTS drinks, respectively.

Six rhizobia-like-bacterial strains in total, secluded from the root and stem nodules of various leguminous plants were characterized for growth promoting ability on ICCV 2 variety of chickpea. Bacterial strains showed production of IAA, NH3, siderophore, HCN, ACC deaminase, hydrolytic enzyme production such as chitinase, amylase, protease, lipase, β-1, 3-glucanase and solubilization of nutrients such as phosphate, zinc and potassium. However the performance of PGP traits characterized in-vitro varied among the six bacterial strains. The sequences of 16S rRNA gene of bacterial strains IHSR, IHRG, IHAA, IHGN-3, IHCP-1 and IHCP-2 showed maximum identity with Rhizobium sp., Rhizobium tropici, Rhizobium multihospitium, Mesorhizobium sp., Burkholderia cepacia and Rhizobium pusense. In plate culture conditions the bacterial strains changed the colour of media (NFB) from green to blue and showed amplification of nifH gene by PCR, and also enhanced nodule formation in chickpea under greenhouse conditions, which explains their nitrogen fixing ability. Scanning electron microscopy studies of chickpea roots showed colonization by all the six bacterial strains in solo and by consortium (IHRG þ IHGN-3). Under greenhouse conditions, chickpea plants inoculated with different strains showed improvement in plant height, number of branches, total chlorophyll, nodule number, nodule weight, shoot weight, root weight, root volume and root surface area at 30 and 45 days after sowing (DAS) over the uninoculated control plants. It was also observed at the crop maturity stage all the bacterial strains inoculated separately enhanced pod number, seed number and total NPK compared to uninoculated control plants. This study suggests that bacteria associated with root and stem nodules can be a promising resource to enhance nodulation, PGP and crop yields in chickpea.

Trypanosoma brucei brucei causes animal trypanosomiasis in several vertebrates and human African trypanosomiasis. Previous studies have only explored the incidence, clinical symptoms, haematology and biochemical changes associated with the disease. The behavioral manipulation hypothesis posits that parasites alter the behavior of host to increase the reproductive abilities of such parasites. Hence, the present study was carried out to investigate changes in behavior and cognition following experimental infection of T. brucei brucei in rat model. This study involved two groups of animals (uninfected control and T. brucei infected) with 8 rats per group. After confirmation of parasitaemia in the infected rats both groups were assessed to investigate if infection led to behavioral alterations and neuropathological changes using the open field, social interaction and forelimb suspension tests. Immunohistochemistry was performed on brain tissues using glial fibrillary acidic protein and anticalbindin-D28k, antibodies. We demonstrated that T. brucei infection triggered a significant decrease in exploratory activity, anxiety-like behavior, altered recognition of social novelty and reduced hanging latency in the hanging wire test. Immunohistochemistry revealed significant astrocytosis, loss of dendritic spines and reduction of Purkinje cell layer of the cerebellum. These results demonstrate that T. brucei infection induce signs of anxiety, impaired motor co-ordination with degeneration and loss of Purkinje cells.

Determining a suitable stocking density of fish for an ambient condition is very important for economic benefit in cage aquaculture, which is not yet tested for many species in Sylhet district of Bangladesh. Therefore, current research was conducted in order to explore the effect of various stocking densities on growth and production performances of mono-sex Nile tilapia cage aquaculture in an open running water body, the Gurukchi River. Considering maximizing economic benefit, it is the first instance of such research in the Sylhet district of Bangladesh. In the three treatments (T1, T2, and T3), fingerlings were stocked at 40, 60 and 80 fish/m3, respectively with initial weights of 39.51 ± 0.91, 39.61 ± 0.71 and 38.54 ± 0.57g, respectively. Fish were fed with commercial floating pellet feed at 8-4% of their body weight. The results showed that growth performance of Nile tilapia significantly decreased with increasing stocking density. The mean total yields were 13.25 ± 0.48, 18.43 ± 0.88 and 22.76 ± 0.63 kg/m3 in T1, T2 and T3, respectively, which showed significant variations (p < 0.05) among treatments. The benefit-cost ratio analysis revealed that T1 (1.512 ± 0.022) and T2 (1.499 ± 0.063) were significantly (p < 0.05) higher than T3 (1.191 ± 0.071), with no significant differences observed between T1 and T2. Notably, a significantly higher (p < 0.05) net profit was observed in T2 than in T1 and T3. Overall, 60 fish/m3 stocking density was the best stocking density for commercial cage aquaculture of tilapia in a riverine environment of the north-eastern Bangladesh.

Herbal remedies have been used in many cultures for decades to treat illnesses. These medicinal plants have been found to contain various phytochemical compounds that can help to cure mild to severe illnesses. The inadequacies of conventional medicines and their unusual side effects sparked a determined search for alternative natural therapeutic agents. Another reason for this hunt could be the availability and fewer side effects of natural products. T. arjuna is widely used in traditional medicine to alleviate various diseases like relieving pain, ameliorating diabetes, mitigating inflammation, and back-pedaling of depression. In this study, the ethanolic extract of T. arjuna possesses a promising effect on the animal model (p < 0.05/p < 0.01) as an antihyperglycemic, analgesic, anti-inflammatory, and antidepressant agent, but in a dose-dependent manner. The lower dose of T. arjuna was found to be capable of reversing the disturbed physiological state at a significant level (p < 0.05). However, a higher dose of T. arjuna exerts better therapeutic effects for those diseases. This animal study aims to evaluate the anti-diabetic, anti-depressant, and anti-inflammatory properties of T. arjuna compared to conventional marketed drugs. We will perform an in-silico study for active constituents of T. arjuna against their proposed targets and look for the molecular cascade on their claimed pharmacological properties. This study shows that different doses of T. arjuna bark extracts give similar therapeutic responses compared with established marketed drugs in managing hyperglycemia, stress-induced depression, and inflammation. Besides, our docking study reveals that flavonoids and triterpenoid active constituents of T. arjuna play an important role in its usefulness. Ramie is one of the long natural fiber has strong mechanical properties. To improve the quality of ramie fiber, this study developed a bio degumming method from superior isolates of pectinolytic fungi Rhizopus sp. and optimization of raw fiber based on harvesting time. The results of the pectinolytic fungi selection were used as a bio degumming bio starter under optimum conditions of pH and temperature. Also, fiber material harvested at 50-day and 60- day to obtain an increase in the physical quality. The bio degummed fiber was analysed to determine the tenacity and fineness, the functional groups contained, thermal analysis, moisture regain and content, material polymers, and degree of crystallinity. Based on the results, the finest ramie properties with 50-day harvested fiber are as follows: strength 24.54 ± 0, 02 g/tex, elongation 12.04 ± 2,90%, fineness 1.33 ± 0,17 tex, moisture regain 8.23 ± 0,18 %, and moisture content 8.96 ± 0,21 %. Ramie fibers at initial conditions and after bio degumming at 50 and 60-day harvested had the same pattern of thermal stability. The dyeability test showed that the degummed 60-day harvested fibers has the greatest dye fixing ability. In traditional medicine, Ocimum gratissimum (scent leaf) is used in the treatment of various diseases such as diabetes, cancer, inflammation, anaemia, diarrhoea, pains, and fungal and bacterial infections. The present study reviewed the phytochemicals, essential oils, and pharmacological activities of O. gratissimum. The bioactive compounds extracted from O. gratissimum include phytochemicals (oleanolic acid, caffeic acid, ellagic acid, epicatechin, sinapic acid, rosmarinic acid, chlorogenic acid, luteolin, apigenin, nepetoidin, xanthomicrol, nevadensin, salvigenin, gallic acid, catechin, quercetin, rutin, and kaempfero) and essential oils (camphene, β-caryophyllene, α- and β-pinene, α-humulene, sabinene, β-myrcene, limonene, 1,8-cineole, trans-β-ocimene, linalool, α- and δ-terpineol, eugenol, α-copaene, β-elemene, p-cymene, thymol, and carvacrol). Various in vivo and in vitro studies have shown that O. gratissimum and its bioactive constituents possess pharmacological properties such as antioxidant, anti-inflammatory, anticancer, hepatoprotective, antidiabetic, antihypertensive, antidiarrhoeal, and antimicrobial properties. This review demonstrated that O. gratissimum has a strong preventive and therapeutic effect against several diseases. The effectiveness of O. gratissimum to ameliorate various diseases may be attributed to its antimicrobial and antioxidant properties as well as its capacity to improve the antioxidant systems. However, despite the widespread pharmacological activities of O. gratissimum, further experiments in human clinical trial studies are needed to establish effective and safe doses for the treatment of various diseases.

The sea cucumber Holothuria scabra is both an economically important species in Asian countries and an emerging experimental model for research studies in regeneration and medicinal bioactives. Growth factors and their receptors are known to be key components that guide tissue repair and renewal, yet validation of their presence in H. scabra has not been established. We performed a targeted in silico search of H. scabra transcriptome data to elucidate conserved growth factor family and receptor genes. In total, 42 transcripts were identified, of which 9 were validated by gene cloning and sequencing. The H. scabra growth factor genes, such as bone morphogenetic protein 2A (BMP 2A), bone morphogenetic protein 5-like (BMP5-like), neurotrophin (NT) and fibroblast growth factor 18 (FGF18), were selected for further analyses, including phylogenetic comparison and spatial gene expression using RT-PCR and in situ hybridization. Expression of all genes investigated were widespread in multiple tissues. However, BMP 2A, BMP5-like and NT were found extensively in the radial nerve cord cells, while FGF18 was highly expressed in connective tissue layer of the body wall. This work investigated the antifungal, cytotoxic and LPS-induced anti-inflammatory effects of five Vachellia species (V. karroo, V. kosiensis, V. sieberiana, V. tortalis and V. xanthophloea). The antifungal activity of the aqueous-methanolic extracts were performed using the broth dilution method against four non-albicans Candida species (C. glabrata, C. auris, C. tropicalis and C. parapsilosis). The cytotoxic and anti-inflammatory effects of the extracts were evaluated on African green monkey Vero kidney cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay and the 2ʹ,7ʹ-dichlorofluorescin diacetate (H2DCF-DA) method. The fourier-transform infrared spectroscopy (FTIR) and Q Exactive plus orbitrap™ Ultra-high-performance liquid chromatography-mass spectrometer (UHPLC-MS) analysis was conducted to evaluate phytochemical constituents of the extracts. The plant extracts selected in this study displayed potency against the Candida species tested, with MIC values ≤0.62 mg/mL for V. karroo, V. kosiensis and V. xanthophloea. A dose-dependent cell viability was observed on Vero cells with all extracts showing LC50 values >20 μg/mL. Extracts tested at 10 μg/mL elicited a significant decrease in lipopolysaccharide (LPS)-induced reactive oxygen species (ROS) in Vero cells with V. sieberiana, V. tortilis, V. karroo, V. kosiensis and V. xanthophloea displaying inhibitory percentages of 35%, 32%, 55%, 52% and 49%, respectively. Characterisation of functional groups representing compounds in the extracts demonstrated the presence of different classes of compounds of the aliphatic, sugar and aromatic types. The Q Exactive plus orbitrap™ mass spectrometer enabled tentative identification of three major compounds in the extracts, including epigallocatechin, methyl gallate and quercetin amongst others. Based on the mass spectrometer results, it is postulated that quercetin found mostly in active extracts of V. karroo, V. xanthophloea, and V. kosiensis may be responsible for the observed antifungal and anti-inflammatory activity. Recently, the spread of pharmaceuticals and personal care products (PPCPs) in the aquatic environment has steadily increased. In this study, phytoremediation technology, using an ornamental plant (Alternanthera spp.), was investigated to improve the removal of acetaminophen (AC) and methylparaben (MP) from a synthetically prepared wastewater. Three exposure lines (AC-line, MP-line and control-line) were performed with a total of 26 subsurface-horizontal constructed wetlands (SSH-CWs) that operated in batch feeding mode. The influence of plants in addition to the initial spiking concentration (20, 60 and 100 mg/L) of AC and MP on the removal efficiency was evaluated throughout the 35-days experiments. The highest removal efficiencies for AC and MP were 88.6% and 66.4%, respectively, achieved in the planted CWs; whereas only 29.7% and 21.9% were achieved in the control CWs for AC and MP, respectively. The results confirmed the role of Alternanthera spp. for accelerating the removal of AC and MP from synthetically contaminated wastewater in CWs., Phytoremediation; Constructed wetlands; Phytotoxicity; Alternanthera spp; Pharmaceuticals and personal care products.

Foraminifera are unicellular organisms and play a pivotal role in the marine material cycles. Past observations have shown that the species Elphidium excavatum is the most common foraminifera in the Baltic Sea. Feeding experiments showed that the food uptake and thus the turnover of organic matter are influenced by changes of physical parameters (e.g., temperature, salinity). Since many areas of the Baltic Sea are strongly affected by anthropogenic activity and are strongly contaminated by heavy elements from shipping in the past, this study examined the effect of heavy elements pollution on the food uptake of the most common foraminiferal species of the Baltic Sea, E. excavatum which was a subject of several previous studies. Therefore, Baltic Sea seawater was enriched with metals at various levels above normal seawater levels and the uptake of 13C- and 15N-labelled phytodetritus was measured by isotope ratio mass spectrometry. For each combination of metal type, concentration and time point 20 individuals of E. excavatum (three replicates) were fed with the green algae Dunaliella tertiolecta. The effect of dose parameters was measured in a two-way analysis of variance. Significant differences of food uptake were observable at different types and levels of heavy elements in sea water. Even a 557-fold increase in the Pb concentration did not affect food uptake, whereas strong negative effects were found for higher levels of Zn (144 and 1044-fold) and especially for Cu (5.6 and 24.3-fold). In summary it can be stated, that an increase in the heavy elements pollution in the Kiel Fjord will lead to a significant reduction in the turnover of organic matter by foraminifera such as E. excavatum., Foraminifera, Feeding experiments, Heavy metal, Toxic metal, Metabolism.

This aim of this study was to evaluate the effects of pretreatments and temperature on the hot air drying characteristics of hog plum fruits. Hog plum fruits were pretreated with olive oil/K2CO3 or sunflower oil/K2CO3 at 28 °C and olive oil/NaOH cum blanching at 96 °C for 15s, hot water at 96 °C for 15s, and dried in a hot air drier at 50, 60, and 70 °C. Mathematical models were used to fit the data of drying and rehydration kinetics. Results showed that increase in temperature reduced drying time, increased effective diffusivity and shrinkage. Sunflower oil aided chemical pretreated sample had the shortest drying time (780 min) and highest effective diffusivity (6.3 × 10−8 m2/s) at 60 °C, faster rehydration ability at 60 °C, highest retention rate for ascorbic acid (15 %), phenolic content (29 %), and antioxidant activity (12.3 %), while olive oil aided chemical (K2CO3) pretreated sample had the shortest drying time at 50 °C (990 min) and 70 °C (600 min), lowest shrinkage (48.5 %), slower rehydration capacity at 40 °C, and lowest colour change (ΔE = 11.5). Modified Henderson and Pabis and Vega-Galvez were superior to other fitting models in predicting the drying and rehydration kinetics. Sunflower oil/K2CO3 pretreatment could help improve the drying and quality characteristics of hog plum.

Background Ocular toxoplasmosis is an infection caused by Toxoplasma gondii. In South America, the clinical course of ocular toxoplasmosis is more severe than in Europe and North America because virulent strains of the parasite are present. Ocular toxoplasmosis is the leading cause of posterior uveitis and retinochoroiditis in Colombia, requiring timely and appropriate treatment. However, there is no standardized therapy protocol based on economic studies for the country. Purpose To compare the cost-effectiveness of four first-line treatment regimens for active ocular toxoplasmosis in immunocompetent adults in Colombia, using the number of averted therapeutic failures as the outcome. Methods We performed an economic and cost-effectiveness analysis to compare four first-line treatment regimens for ocular toxoplasmosis from the perspective of a third-party payer (Colombian General System of Social Security in Health). A decision analysis tree was used over a 24-week time horizon, considering only direct costs. Additionally, we performed a discrete sensitivity analysis and a probabilistic sensitivity analysis with 10,000 iterations in the Monte Carlo simulation. Platycosides, saponins contained in balloon flower, which have been used as food health supplements for respiratory diseases, have diverse pharmacological effects. Platycosides exhibit better pharmacological activity by hydrolyzing their own sugars. However, to date, there have been no studies on the production of deglucosylated platycodin D suitable for food applications. In this study, Pluszyme 2000P, which was derived from Aspergillus niger, a food-grade microorganism, was used to completely convert platycoside E into deglucosylated platycodin D. For an efficient and economical production of deglucosylated platycodin D, the productivity was improved approximately 2.4 times by application of high hydrostatic pressure and the discarded balloon flower leaf was used as a substrate. As a result, deglucosylated platycodin D was produced with the highest concentration (3.49 mg/mL) and productivity (581.7 mg/L/h) reported so far. Aedes (Ae.) albopictus is an important vector for many pathogens. Previous studies have revealed a role for midgut bacteria during pathogen infection in mosquitoes; however, studies of Ae. albopictus midgut bacteria are limited. We examined the diversity of midgut bacteria in female laboratory-colonized and field-collected Ae. albopictus. A total of 31 bacterial genera were identified representing 10 and 28 genera of laboratory-colonized and field-collected Ae. albopictus, respectively. The predominant bacterial genera in the laboratory-colonized Ae. albopictus were Staphylococcus and Micrococcus, whereas the bacterial diversity in the field-collected Ae. albopictus exhibited a higher proportion of Rhizobium and Agrobacterium as the dominant genera. However, only Staphylococcus showed a significant difference between laboratory-colonized and field-collected Ae. albopictus. The midgut bacterial species were identified from 30 laboratory-colonized Ae. albopictus mosquitoes. A total of 16 bacterial species were identified and the predominant bacterial species was Micrococcus luteus, followed by Staphylococcus epidermidis and Agrobacterium tumefaciens. Field mosquitoes were collected from the Sing Buri, Chumphon, and Yala Provinces of Thailand. The midgut bacterial species identified from the 10 Ae. albopictus collected from the Sing Buri Province included Bacillus subtilis, Staphylococcus haemolyticus, Staphylococcus hominis, and Serratia marcescens. Serratia marcescens was the only bacteria identified from this area. Midgut bacterial species were identified from 40 filed-collected Ae. albopictus from Chumphon Province. A total of 25 bacterial species were identified and the predominant species were Enterobacter cloacae, Micrococcus luteus, and Providencia rettgeri. Only 15 bacterial species were identified from the mosquitoes collected from Chumphon Province. A total of 18 bacterial species were identified from 30 Ae. albopictus collected from Yala Province and the predominant species were Rhizobium pusense and Agrobacterium tumefaciens. Only 12 bacterial species were found in mosquitoes collected from Yala Province. These findings indicate changes in the midgut bacteria population in Ae. albopictus from various locales, which may result from variability in the blood-meal source, diet, or habitat. A comprehensive survey of the midgut bacteria community prevalence in wild populations is critical for not only gaining a better understanding of the role of this bacterium in shaping the microbial community in Ae. albopictus, but also for informing current and future mosquito and disease control programs.

Palm grass (Curculigo recurvata) is an ethnomedicinally important herb reported to have significant medicinal values. The present study aimed to evaluate the antidepressant and anxiolytic activities of a methanol extract of C. recurvata rhizome (Me-RCR) through different approaches. The antidepressant and anxiolytic properties of Me-RCR were assessed by using elevated plus maze (EPM), hole-board (HBT), tail suspension (TST), and forced swimming (FST) tests in Swiss Albino mice. The in-depth antioxidative potential of Me-RCR was also evaluated through DPPH radical scavenging activity, ferric-reducing power capacity, total phenolic, flavonoid, flavonol, and antioxidant content analysis. Computational investigations were performed using computer-aided methods for screening the anxiolytic, antidepressant, and antioxidative activities of the selected lead molecules. Treatment with Me-RCR (200 and 400 mg/kg, b.w.) notably increased the number of open arm entries and the time spent in the EPM test. In the HBT, Me-RCR exhibited significant anxiolytic activity at a dose of 200 mg/kg, whereas similar activity was observed at 400 mg/kg in the EPM test. Me-RCR significantly decreased the immobility time in a dose-dependent manner in both TST and FST. The IC50 for DPPH and reducing power capacity assay were found to be 18.56 and 193 μg/mL, respectively. Promising outcomes were noted for the determination of total phenolics, flavonoids, flavonols, and antioxidant capacity. In the case of computer-aided studies, nyasicoside showed promising binding energy for antidepressant and anxiolytic activities, whereas isocurculigine demonstrated promising effects as an antioxidant. Scientific research into developing new antimicrobials from plants continues to be an interesting area for many scientists. This is because the resistance of microorganisms to anti-infective agents has affected a wide range of conditions, some of which are life-threatening. This study aimed to investigate the antimicrobial properties of Cnestis ferruginea (CF). Powdered roots of Cnestis ferruginea were extracted with petroleum ether (CFP), ethyl acetate (CFE) and methanol (CFM). The antimicrobial and microbial resistance modifying activity profiles of the extracts were studied against Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 4853, Staphylococcus aureus ATCC 25923, clinical strains of Methicillin-Resistant Staphylococcus aureus, Streptococcus pyogenes, Klebsiella pneumonia, Staphylococcus epidermidis, Proteus mirabilis and Candida albicans. CFP and CFE showed no activity against the test organisms. CFM had mean zones of growth inhibition in the range of 11.0 ± 0.5 to 22.17 ± 0.24 mm against the test organisms. The MIC of CFM was within the range of 0.31 and 5.0 mg/mL, with MBC/MFC range of 2.5–20.0 mg/mL. The time-kill kinetics studies showed CFM is a static agent. At sub-inhibitory concentrations, CFM was able to increase the susceptibility of the test organisms to standard antibiotics from the range of 1–8 folds. CFM reduced the formation of biofilms from 100% to 56.59%, 62.33%, 65.89% and 71.88% against K. pneumonia, S. aureus, E. coli and P. aeruginosa, respectively. In this study, we have isolated and characterized proteolytic soil bacteria and their alkaline protease. Based on 16S rRNA sequence analysis, 12 isolates with the highest protease activity were classified as B. subtilis and B. cereus groups. B. subtilis D9 isolate showing the highest protease activity was selected for in vitro and in silico analysis for its ِِAKD9 protease. The enzyme has a molecular mass of 48 kDa, exhibiting optimal activity at 50 °C pH 9.5, and showed high stability till 65 °C and pH 8–11 for 1 h. Fe3+‏ stimulated, but Zn2+ and Hg2+ strongly inhibited the protease activity. Also, the maximum inhibition with PMSF indicated serine protease-type of AKD9 protease. AkD9 alkaline serine protease gene showed high sequence similarity and close phylogenetic relationship with AprX serine protease of B. subtilis isolates. Functional prediction of AKD9 resulted in the detection of subtilase domain, peptidase_S8 family, and subtilase active sites. Moreover, prediction of physicochemical properties indicated that AKD9 serine protease is hydrophilic, thermostable, and alkali-halo stable. Secondary structure prediction revealed the dominance of the coils enhances AKD9 activity and stability under saline and alkaline conditions. Based on molecular docking, AKD9 showed very promising binding affinities towards casein substrate with expected intrinsic proteolytic activities matching our obtained in vitro results. Background Tuberculosis (TB) and drug-resistant TB (DR-TB) continue to persist as a serious public health challenges in Ghana. Although several research has evaluated the drug resistance of Mycobacterium tuberculosis complex (MTBc) strains across the country, there is a paucity of data on its magnitude as well as the various lineages circulating in the Eastern region of Ghana. Objective This study therefore evaluated the distribution of the various lineages of MTBc in the Eastern region of the country and the associated drug resistance. Materials and methods One hundred and forty-three (143) patients with pulmonary TB attending the Eastern Regional Hospital, Koforidua/Ghana were included in the study. The BACTEC MGIT 960 tube media was used for both sputum culture and drug susceptibility of streptomycin (STR), isoniazid (INH), rifampicin (RIF) and Ethambutol (ETH). Isolates were initially typed using IS6110, followed by large sequence polymorphisms analysis and spoligotyping. Results The majority [108 (75.5%)] of the 143 patients were male gender and the 45–54 years [46 (32.2%)] age range had the highest frequency. Forty-one (28.7%) of the 143 isolates were IS6110 negative. Of the 102 spoligotyped isolates, the main sub-lineages included 45 (44.1%) Cameroon and 23 (22.5%) Ghana. SITs 61 and 53 represented the major cluster with 22/102 (21.6%) and 13/102 (12.7%) isolates respectively, while 59/65 (90.8%) isolates belonged to Lineage 4 with 27/65 (41.5%) LAM10_CAM. MDR-TB occurred in 26/79 (32.9%) isolates, and was not associated with neither gender [20/58 (34.5%) male vs 6/21 (28.6%) female, OR = 1.31; 95%CI, 0.44–3.92; p = 0.624)] nor age. No association was found between MDR-TB and the major sub-lineages [8/25 (32%) Cameroon (OR = 0.94; 95%CI, 0.34–2.59; p = 0.920) and 5/11 (45.5%) Ghana (OR = 1.87; 95%CI, 0.51–6.80; p = 0.489)], or previously treated [8/23 (34.8%), OR = 0.89; 95%CI, 0.32–2.48; p = 0.823)] patients. Conclusion Despite the serious threat posed by MDR in the study area, no sub-lineage was shown to be associated with drug resistance. Nonetheless, a sustained surveillance of drug resistance pattern is advocated. A lower proportion of M. africanum was observed in the Eastern region of Ghana and will require further evaluation.

Cadmium stress disrupts plant-microbial interactions and reduces plant growth and development. In plants, the tolerance to stress can be increased by inoculation with endophytic microorganisms. The aim of this study was to investigate the distribution of endophytic fungi in various plant organs of barley and soybean and evaluate their Cd removal ability. Two hundred fifty-three fungal strains were isolated from various organs of barley (Hordeum vulgare cv Arna) and soybean (Glycine max cv Almaty). The colonization rate ranged from 13.6% to 57.3% and was significantly higher in the roots. Ten genera were identified: Fusarium, Penicillium, Aspergillus, Metarhizium, Beauveria, Trichoderma, Rhodotorula, Cryptococcus, Aureobasidium and Metschnikowia. Twenty-three fungal strains have a Cd tolerance index from 0.24 to 1.12. Five strains (Beauveria bassiana T7, Beauveria bassiana T15, Rhodotorula mucilaginosa MK1, Rhodotorula mucilaginosa RH2, Metschnikowia pulcherrima MP2) with the highest level of Cd tolerance have minimum inhibitory concentrations from 290 to 2400 μg/ml. These fungi were able to remove Cd up to 59%. The bioaccumulation capacity ranged from 2.3 to 11.9 mg/g. Bed bugs are common urban pests. Unlike many other blood-feeding human ectoparasites, bed bugs are not known to be vectors of human infectious diseases, but clinical and epidemiological studies to directly interrogate this link have been limited. Here, we aimed to determine whether bed bugs were associated with infectious diseases in a set of infested patients presenting to emergency departments (ED) in the greater Cleveland, OH area. We performed a retrospective case-control study involving 332 ED patients with bed bugs and 4,952 control patients, seen from February 1, 2011, through February 1, 2017. Cases and controls were matched by age, sex, and the presenting ED. Additionally, data were adjusted for ≥20 sociodemographic variables, triage data, and comorbidities in multivariable regression analyses. Seventeen laboratory values, ten different ED and inpatient diagnoses, chest radiographs, infectious disease consults, and blood cultures were examined. The odds of bed bug infestation were significantly higher for patients that had positive blood cultures, had blood cultures growing coagulase-negative Staphylococcus, were diagnosed with pneumonia, were diagnosed with cellulitis, received an infectious disease consult, received a chest radiograph, and had higher percentages of eosinophils in the blood (P < .05 for all). Heavy metal contamination is a universal concern due to health risks associated with metal pollution. Soil contamination by heavy metals is known to affect microbial activities at elevated concentrations adversely. However, indigenous soil bacterial populations' response to added heavy metal and metal combinations is poorly understood. Microbes prevailing in the soil are the driving factors. Their properties are recognized as sensitive indicators of soil quality and health. Moreover, these microscopic organisms are accountable for the fertility and aeration of the soil that forms fundamental aspects of soil function. The current study was performed to explore the diversity of bacterial species in heavy metal polluted roadside soil. The roadside soil samples were collected from diverse sites and processed for physicochemical properties, microbial characterization, and heavy metals distribution in the selected locations. Serial dilution and spread plate techniques were used for the isolation of bacterial species. The 16S-rRNA gene sequencing identified bacterial species in roadside soil as Bacillus drentensis (MK217088), Bacillus safensis (MK774729), Bacillus haynesii (MK192808), Bacillus subtilis (MK217089), and Bacillus cereus (MK801278). In addition, the 16S rRNA sequences of isolated bacterial strains were aligned to generate a phylogenetic tree. The degree of persistence in daily PM25 and O3 in the ten most populated US cities, namely New York, Los Angeles, Chicago, Houston, Phoenix, Philadelphia, San Antonio, San Diego, Dallas and San Jose is examined in this work. We employ a methodology based on fractional integration, using the order of integration as a measure of the degree of persistence. Using data for the time period from January 1, 2019 to December 31, 2020, our results indicate that fractional integration and long memory features are both present in all the examined cases, with the integration order of the series being constrained in the (0, 1) interval. The introduction of water hyacinth poses a serious threat to economic viability of Lake Tana and its environments. This study aimed to capture the spatial coverage of water hyacinth and its effect on water loss in Lake Tana using quantitative research methods. Four satellite images representing each season of 2019 were downloaded from USGS. In addition, pan evaporation data were taken from the National Meteorological Agency. ArcGis 10.4, Envi 5.3, Qgis 3.12.1 plug in CSP and Excel used to manage land use land cover classification and water loss estimation analysis. The seasonal coverage of water hyacinth was 15.35, 4.14, 11.82 and 13.59 km2 in winter, autumn, summer and spring 2019 respectively representing 0.63 and 0.17 percent of the Lake as a maximum and minimum coverage. The mean daily evaporation of Lake Tana was 5.14 mm/day, but this increased to 18.85 mm/day due to the presence of water hyacinth. The mean net daily water loss due to water hyacinth at Lake Tana was 0.14 km2 while 52,62 km3 in 2019. The study concludes that water hyacinth caused enormous negative impact on the water volume reduction in Lake Tana. Management of the Lake Tana environment and control of the water hyacinth weed are recommended to sustain the Lake., Evapotranspiration, Lake Tana, spatial cover, water hyacinth, water loss.

The objective of this research is to assess the effect of enzymatic treatment of guava puree on the physicochemical parameters of the juice. Pectinases from Aspergillus niger were applied to the puree at 43 ± 3 °C under constant stirring. Enzyme concentrations used were: 0.033 % (w/w), 0.055% (w/w), 0.078 % (w/w) and 0.1 % (w/w). For each enzyme concentration, the treatment times were varied from 3 – 90 min. Physicochemical parameters of raw puree and enzymatically treated juice were determined. These were: viscosity, pH, electric conductivity, protein and polyphenol content, galacturonic acid content, color, TSS, and antioxidant capacity. Particle distribution, homogeneity of raw puree and juice samples dried extracts were assessed using a Field Emission Scanning Electron Microscopy (FESEM). A 91% viscosity decrease was recorded for each enzyme concentration after 3 min of enzyme reaction. That drecrase was accompanied by an increase in galacturonic acid content with increasing depectinization factors. Enzyme treatment of guava puree led to a decrease in pH, protein and polyphenol contents and an increase in conductivity and color. Analysis of FESEM images of guava samples bestowed a decrease in particle size, a scattering of particles in the medium, an increase in continuous phase proportion and an improvement of sample homogeneity with increasing values of processing parameters, due to the breaking-down of bigger particles and the solubilization during depectinization., Pectinases, hydrolysis, guava pulp, juice, FESEM

Microorganisms in clay barriers could affect the long-term performance of waste containers in future deep geological repositories (DGR) for used nuclear fuel through production of corrosive metabolites (e.g., sulfide), which is why clay materials are highly compacted: to reduce both physical space and access to water for microorganisms to grow. However, the highly compacted nature of clays and the resulting low activity or dormancy of microorganisms complicate the extraction of biomarkers (i.e., PLFA, DNA etc.) from such barriers for predictive analysis of microbial risks. In order to overcome these challenges, we have combined culture- and 16S rRNA gene amplicon sequencing-based approaches to describe the functional diversity of microorganisms in several commercial clay products, including two different samples of Wyoming type MX-80 bentonite (Batch 1 and Batch 2), the reference clay for a future Canadian DGR, and Avonlea type Canaprill, a clay sample for comparison. Microorganisms from as-received bentonites were enriched in anoxic 10% w/v clay microcosms for three months at ambient temperature with addition of 10% hydrogen along with presumable indigenous organics and sulfate in the clay. High-throughput sequencing of 16S rRNA gene fragments indicated a high abundance of Gram-positive bacteria of the phylum Firmicutes (82%) in MX-80 Batch 1 incubations. Bacterial libraries from microcosms with MX-80 Batch 2 were enriched with Firmicutes (53%) and Chloroflexi (43%). Firmicutes also significantly contributed (70%). Sequence analysis revealed presence of the bacterial families Peptostreptococcaceae, Clostridiaceae, Peptococcaceae, Bacillaceae, Enterobacteriaceae, Veillonellaceae, Tissierellaceae and Planococcaceae in MX-80 Batch 1 incubations; Bacillaceae, along with unidentified bacteria of the phylum Chloroflexi, in MX-80 Batch 2 clay microcosms, and Pseudomonadaceae, Hydrogenophilaceae, Bacillaceae, Desulfobacteraceae, Desulfobulbaceae, Peptococcaceae, Pelobacteraceae, Alcaligenaceae, Rhodospirillaceae in Canaprill microcosms. Exploration of potential metabolic pathways in the bacterial communities from the clay microcosms suggested variable patterns of sulfur cycling in the different clays with the possible prevalence of bacterial sulfate-reduction in MX-80 bentonite, and probably successive sulfate-reduction/sulfur-oxidation reactions in Canaprill microcosms. Furthermore, analysis of potential metabolic pathways in the bentonite enrichments suggested that bacteria with acid-producing capabilities (i.e., fermenters and acetogens) together with sulfide-producing prokaryotes might perhaps contribute to corrosion risks in clay systems. Herbal medicines are currently being adopted as alternatives to orthodox medicines for the management of drug-resistant and emerging multidrug-resistant microbial strains of various diseases, including typhoid fever. A herbal decoction, MA 001, manufactured by the Centre for Plant Medicine Research (CPMR), has been used for the treatment of typhoid fever for at least two decades in Ghana with desirable outcomes. MA 001 is formulated from Citrus aurantifolia, Spondias mombin, Latana camara, Bidens pilosa, Trema occidentalis, Psidium guajava, Morinda lucida, Vernonia amygdalina, Persea americana, Paulina pinnatta, Momordia charantia and Cnestis ferruguinea medicinal plants. The low palatability and compliance to treatment due to the bulky nature of the decoction poses challenges in its optimum use. This study sought to design and formulate the therapeutic components of the aqueous herbal decoction of MA 001 into an optimal solid dosage form of effervescent granules to improve the delivery of MA 001 as well as increase patient compliance and convenience of product handling. The methods involved pre-formulation studies on the suitability of effervescent vehicles, formulation and evaluation of effervescent granules for drug excipient interactions using high performance liquid chromatography analysis. The objective of the current study was to evaluate the biochemical and immunological responses of tilapia, Oreochromis niloticus due to heavy metals pollution. Histomorphological alterations in the liver and kidney suggested tissue damages due to this polluted water exposure. The brain acetylcholinesterase (AChE) as an indicator of neurotoxicity was significantly (P < 0.01) decreased after 10 days exposure of fish to heavy metal contained river water, while plasma glutamate oxalacetate transaminase and plasma glutamate pyruvate transaminase were significantly increased (P < 0.01). Moreover, superoxide dismutase, catalase, glutathione peroxidase, glutathione S-transferase enzyme activities, as well as reduced glutathione and malondialdehyde levels were significantly increased in heavy metals contained river water treated fish compared to the control. Additionally, glucose level and blood serum Ca2+ concentrations were significantly (P < 0.01) decreased in fish exposed to heavy metal contained river water compared to the control. Hematological indices such as Hemoglobin, RBC, WBC, MCV etc. of polluted river water treated fish were significantly (P < 0.01) different in comparison to that of control fish. The cytokines i.e. IL-1β, IL-6, and TNF-α level were significantly (P < 0.01) increased in the fish exposed to heavy metals contained river water in comparison to that of control fish. Traditional fermented buffalo milk gel; Meekiri, is a popular buffalo milk-derived product in Sri Lanka. Predominantly, it is produced using the back-slopping (adding a small amount of the previous fermentate) technique, following the life-long traditions available at the cottage level. Hence, diverse and unclassified starter cultures are likely to be established across the varying geographical regions of Meekiri production. In the present study, we aimed to elucidate the diversity of lactic acid bacteria (LAB) and their characteristics including probiotic properties from major Meekeri production areas (n = 22) in Sri Lanka. Lactic acid bacteria was isolated from locally produced Meekiri samples (n = 23) and characterized based on morphological, biochemical, physiological profiles and potential of probiotic properties. The isolates revealed five different colony and cell morphologies and were classified as heterofermenters, homofermenters and facultative heterofermenters based on CO2 production using glucose. None of the isolates showed the ability to grow either at 5 °C or 0 °C, while 71 % and 100 % survival of the isolates were observed at 15 °C and 45 °C, respectively. Amplified ribosomal DNA restriction analysis (ARDRA) primarily grouped the isolates into three distinct clusters based on their DNA banding patterns. Subsequently, 16S rRNA sequencing of isolates revealed the presence of four species namely, Limosilactobacillus fermentum (n = 18), Latilactobacillus curvatus (n = 2), Lactobacillus acidophilus (n = 2) and Lactiplantibacillus plantarum (n = 1) and in the phylogenetic analysis, it was represented by four distinctive clades. All the isolated species demonstrated promising probiotic potential with antibiotic sensitivity, antimicrobial properties, bile acid tolerance and acid tolerance. In conclusion, traditional back-slopping Meekiri in Sri Lanka contains diverse LAB, with a negligible geographical variation at species-level. Pectinase is an enzyme having a broad industrial and commercial application. However, higher production costs may be the major constraint for the wide-scale application of pectinase. Therefore, researchers are trying to reduce the pectinase production cost for subsequent application in the industrial processes by using a unique substrate and optimizing the fermentation medium components and process conditions. The main purpose of the current study was to optimize medium composition for pectinase production using Aspergillus niger-ATCC 1640 in the solid-state fermentation. The Response Surface Methodology (RSM) was performed to evaluate the effects of variables, specifically the concentrations of Satkara peel, urea, (NH4)2PO4, NH4NO3, KH2PO4, ZnSO4, and MgSO4.7H2O on pectinase production in the solid substrate. Firstly, a two-factorial design, Plackett-Burman design (PBD) was applied to screen the variables that significantly influenced the pectinase production. After finding the critical variables, 15 experimental runs were carried out using a Box-Behnken design (BBD) to derive a statistical model for optimizing the concentrations of the selected variables. The PBD model revealed that Satkara peel, urea, and (NH4)2SO4 significantly affected the pectinase production. RSM results indicated that the predicted response for pectinase production was in good agreement with experimental data (R2 = 0.9836). Under the optimized condition of Satkara peel (8.4 g/L), urea (0.5 g/L), and (NH4)2SO4 (2.7 g/L), the pectinase activity was predicted to be 0.6178 μmol/mL. In the present study, the experimental pectinase production achieved 0.6045 μmol/mL. The study revealed that optimization through RSM could improve the pectinase production from Satkara peel.

This study was designed to evaluate the cytotoxic and analgesic potential of methanol extracts of five wild mushrooms available in the University of Chittagong, Bangladesh. The acetic acid-induced writhing method was used for the analgesic activity, while cytotoxicity was tested using brine shrimp lethality bioassay. In silico molecular docking and ADME/T study have been employed by using Schrodinger v11.1, BIOVIA Discovery Studio 2020 and online tool (AdmeSAR) respectively. The methanol extracts of Daldinia concentrica, Trametes lactinea, Fomitopsis cajanderi and Daedaleopsis confragosa exhibited a significant (p < 0.001) decrease in the number of writhing when compared to the control group. Except for Lentinus squarrosulus at 200 mg/kg body weight, the remaining mushroom extracts showed equal to or above 50 % inhibition of writhing. Daldinia concentrica showed the lowest LC50 values with 0.63 μg/mL, while Daedaleopsis confragosa showed the highest LC50 values of 2.33 μg/mL, indicating decisive cytotoxic action all mushrooms extracts. Considering the secondary metabolites, daldinan A and fomlactone A were found the most promising myco-compounds in analgesic and cytotoxic molecular docking studies. Besides, all the selected metabolites meet the rule of Lipinski's drug-likeliness. These results concluded that each mushroom extracts except Lentinus squarrosulus possess a potential analgesic. Stemona collinsiae exhibits insecticidal resistance against various pests and insect vectors. However, insecticidal activity of S. collinsiae roots has not been tested for some insect vectors, including the American cockroach, Periplaneta americana. The synanthropic insect P. americana is a reservoir of pathogenic and non-pathogenic microorganisms and a cause of infectious diseases and cockroach allergy. This important vector transmits microorganisms to animals and humans to cause vector-borne diseases. This research involved detection of the nymphicidal and adulticidal activities of S. collinsiae root extracts against P. americana through oral administration. The effects of hexane, dichloromethane, ethanol, and water crude extracts were tested on final instar nymphs and adult P. americana. After P. americana ingested bait containing hexane and dichloromethane crude extracts, signs of toxicity occurred, such as hind leg shaking, whole-body tremor, immobility, abdomen swelling, and death. At 48 h, the nymphs and adult P. americana that ingested dichloromethane crude extract-containing bait showed corrected mortality of 65%–100% and 20%–100%, respectively. Whereas none of the nymphs and adult P. americana that ingested the water crude extract-containing bait died (0% corrected mortality). When we dissected alimentary canals of the dead P. americana that had ingested dichloromethane and hexane crude extract-containing baits, the foreguts were found to be swollen. TLC analysis showed the dichloromethane and hexane crude extracts contained the alkaloid didehydrostemofoline and unknown fluorescent substances. Phytochemicals from crude extracts were detected in extracts of dissected alimentary canals using thin-layer chromatography, and didehydrostemofoline alkaloid and unknown fluorescent substances were found in cockroaches that ingested dichloromethane- and hexane-containing baits. The cause of death of P. americana may be attributed to alkaloids and synergistic effects of other substances in S. collinsiae root extract. Mechanisms of action might include several pathways involved in nervous system function. Thus, dichloromethane and hexane crude extracts can be developed as alternative active ingredients in a natural insecticide for cockroach control.

Opuntia ficus-indica L. Mill cladodes are considered to be a source of an abundance of bioactive compounds. To identify a natural product that can be used in the chemoprevention and treatment of cancer, this study was conducted to produce an anticancer agent extracted from in vitro-derived cladodes of prickly pear cactus. Toward this goal, assays of seed germination and micropropagation revealed that the highest seed germination rate was 66% and that the highest shoot number per explant was obtained with benzyl adenine (BA) (2 mg/l) and kinetin (Kin) (1 mg/l) within 2 months, at 22.6. In addition, the maximum length of shoots was obtained with BA (3 mg/l) and Kin (0.5 mg/l), at 7.44 cm. The in vitro-derived cladode extract showed higher total phenolic and kaempferol contents than the in vivo-derived cladode extract (total phenolics 156.5 mg/g and 86 mg/g DW; kaempferol 2.807 mg/g and 1.304 mg/g DW, respectively). These remarkable results reflected the anticancer activity on the viability and proliferation/migration of PC3 prostate and mammary Mcf7-7 cells. In terms of cytotoxicity, the IC50 values on PC3 and Mcf7 cells were 5775.7 and 6311.3 μg/ml, respectively, showing dose-dependent increases. Meanwhile, from in vivo analyses of the plants, the IC50 values were 5927.93 and 6825.6 μg/ml, respectively, again showing dose-dependent increases., Opuntia ficus-indica L. Background and aim Microsporum canis (M. canis) is a dermatophyte fungal pathogen that causes ringworms. Cats are considered to be a dominant reservoir host enabling M. canis transmission to humans. The concerns of dermatophyte resistance were raised among the usage of antifungal drugs to treat the ringworm. This study aimed to evaluate the fungal activity of cyclic lipopeptides (CLPs) obtained from Bacillus subtilis (B. subtilis) as an alternative method for the inhibition of M. canis growth. Materials and methods The culture plate of M. canis from confirmed cats with ringworm infection was provided. The purification of CLP extract, fengycin, iturin A, and surfactin was carried out from B. subtilis by preparative thin-layer chromatography (PTLC) coupled with solid-phase extraction (SPE) methods. Half-maximal effective concentration (EC50) and agar well diffusion assays were performed to determine the efficacy of Bacillus CLP extract, fengycin, iturin A, and surfactin to inhibit the growth of M. canis isolated from cats. Results All purified Bacillus substances displayed antifungal activity to control the growth of M. canis when compared with 80% ethanol (control). EC50 values for CLP extract, fengycin, iturin A, and surfactin were 0.23, 0.05, 0.17, and 0.08 mg/mL, respectively. In agar well diffusion assay, the ability of CLP extract, fengycin, iturin A, and surfactin on fungal inhibition had no statistically significant difference at 24 and 48 h after treatment (p < 0.05). However, CLP extract showed a statistically significant difference on M. canis inhibition at 62.21% followed by surfactin with 59.04% at 72 h after treatment. Conclusion In vitro, Bacillus CLPs revealed an inhibitory effect on M. canis growth which is a zoonotic pathogen that causes ringworms. This study evaluated moisture loss, oil gain and porosity when frying pieces of yam (Dioscorea rotundata). The parallelepiped-shaped samples, approximately 1 × 1 × 4 cm, were subjected to frying temperatures of 145, 165 and 185 °C for 50, 150, 300, 450, or 600 s. Fick's law was used to determine the diffusion coefficient from the experiment data for the varieties 153 traditional Espino and 125 Brazilian Espino. The moisture loss in 153 traditional Espino was greater than in 125 Brazilian Espino. The diffusion coefficient and the activation energy were determined for both varieties, which were higher in the 153 traditional Espino variety. The porosity was expressed as a percentage and was also higher in the 153 traditional Espino variety.

Little is known about folk perception of pearl millet livestock fodder status across the Niger republic. Thus, this work assesses farmers' perceptions of the main pearl millet accessions fodder performance features in Niger. To that end, we: (i) Identify farmers' criteria for choosing pearl millet accessions fodder-profile, (ii) Analyze the farmer preferences for pearl millet accessions adapted to local cropping systems, and (iii) Analyze current cropping systems. Data have been derived from semi-structured surveys based on individual and focus group interviews of 508 stakeholders which were sampled from seven major ethnic groups across the eight country regions. Generated database was submitted to multivariate analysis in XLSTAT software. As results, the farmers' perception of a fodder profile may combine long and wide leaves, abundant tillers, slender stems, coupled to a large biomass and a plant capacity to regrow after grazing or mowing. Height accessions were agro-ecologically and socio-culturally chosen and valued by Farmers across the country, i.e. Haïni Kirey at Tillabéry, Niamey and Dosso, HKP and Guerguéra at Tahoua, Ankoutess at Agadez, Goudiss and Môro at Diffa, Batoukouché at Zinder, and HKP and Zongo at Maradi. Determination of heavy metal concentrations in vegetables and agricultural soils is crucial because high levels of heavy metals could affect soil quality, crop production and safe consumption of crops. A field study was conducted to determine the heavy metal concentrations and their transfer from agricultural soils to different parts (leaf, stem, and root) of Brassica juncea (L.) Czern. In addition, potential health risks of contamination in the vegetables grown in the field were evaluated. Acid digestion method USEPA 3050B in combination with ICP-OES were used to analyze heavy metal (Al, Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn) contents in both pre- and post-harvest soils and vegetable samples. Results showed that none of the heavy metals in soils had concentrations above the maximum safety limits based on the WHO, USEPA and CCME guidelines. Calculated metal transfer factor (MTF >1) showed B. juncea accumulated Cd, Co, Ni, Pb and Zn in leaves, stems and roots, but Cu and Mn, as well as Cr were only accumulated in stems and roots, respectively. There were variations in heavy metal contents between the different parts of B. juncea, but only Cd and Pb contents were above the maximum allowable limit recommended by FAO/WHO. PCA analysis was able to identify 4 major components corresponding to 38.38%, 28.98%, 14.39% and 10.67% of the total variance and PC1 was clearly associated to leaves of B. juncea. Based on the MTF values, only Cd was found to have a value of HRI >1 compared to the other heavy metals, implying potential health risk associated with long-term ingestion of the vegetable., Brassicaceae; DIM; EDEM; HRI; MTF; PCA

Hyalomma anatolicum is one of the most economically important ticks in the Sudan. It is the main vector of tropical theileriosis in cattle and other diseases in different species. The study aims to investigate the drop-off rhythms and survival periods of tick stages fed on crossbred calves. Larvae, nymphs and adults fed on crossbred calves (male, 3–6 months old, Friesians x Zebu) kept under zero grazing system. Drop-off rhythms of engorged stages were studied under field conditions while the survival periods of unfed stages were investigated under field and laboratory conditions (27 °C and 85% R.H.). Significantly high numbers of engorged larvae dropped at night between 20H:00 and 07H:00. (March, April and August). Engorged nymphs dropped late evening and early night 16H:00–20H:00. (September), while insignificant number of engorged females dropped between 10H:00 and 14H:00 (October). Survival durations of unfed H. anatolicum stages were longer under laboratory conditions than in the field. These were 99.36 ± 1.24 and 13.12 ± 0.68 days for larvae, 63 ± 1.33 and 16 ± 0.87 days for nymphs and 90 ± 3.6 and 45 ± 2.7 days for adults, respectively. Under field condition survival studies were conducted in May, August and December 2016 for larvae, March, May, August and December 2016 for nymphs and in October 2016 for Adults. In August, 70% of unfed larvae and nymphs survived for three weeks and 2.5 weeks, respectively, in May, 16% of unfed larvae survived for one week while no unfed nymphs survived for more than 4 days. This finding may indicate that larvae are more resistant to desiccation than nymphs. The abnormal changes on the alloscutum of H. anatolicum desiccated nymphs and females were observed being shrunken and truncated posteriorly, a finding which opens new avenues for further studies. For control strategies more studies are recommended on the dropping to allow ticks drop in an area not suitable for further development as one of the control strategies and the survival studies also is exploited in rotational grazing of animals.

Nondestructive estimation of fruit properties during their ripening stages ensures the best value for producers and vendors. Among common quality measurement methods, spectroscopy is popular and enables physicochemical properties to be nondestructively estimated. The current study aims to nondestructively predict tissue firmness (kgf/cm), acidity (pH level) and starch content index (%) in apples (Malus M. pumila) samples (Fuji var.) at various ripening stages using visible/near infrared (Vis-NIR) spectral data in 400–1000 nm wavelength range. Results show that non-linear regression done by an artificial neural network-cultural algorithm (ANN-CA) was able to properly estimate the investigated fruit properties. Moreover, the performance of the proposed method was evaluated for Vis-NIR data based on optimal NIR wavelength values selected by a genetic optimization tool. In the past, Aedes aegypti was present in Southern Europe. Although the mosquito was eradicated from the Mediterranean region, its regional ecotype survived the second half of the 20th century in the eastern Black Sea area. The aim of the study was to model the changes in the altering climatic suitability, ontogenetic development time and the survival rate of Aedes aegypti from first-stage larvae to adulthood in Southern Europe. The modelled present climatic suitability patterns of the mosquito show that large areas of the lower altitude Mediterranean regions, including the coastal areas of the Balkan Peninsula, South France, and large regions of the Apennines and the Iberian Peninsulas could be suitable for Ae. aegypti. The future (2041–2060 and 2061–2080) projections predict the potential northward shift of the northern occurrence of the species in the circum-Mediterranean and Black Sea areas. Both, the potential development time, and survival rate of Ae. aegypti in the late 19th and the early 20th century could be like in the present times along the Mediterranean coast. The current climatic conditions cannot explain the absence of the mosquito in wide areas of the Mediterranean and sub-Mediterranean ecoregions. The future models predict the notable increase in the development time and survival rate of the mosquito in the southern and central regions of Europe. In general, the container ports of the Alboran, Balearic, and Aegean seas seem to be the most suitable sites for the re-colonization of the mosquito, and such northern parts of the Mediterranean Sea like the Gulf of Lion, the Ligurian, and Adriatic Seas are in less extent.

The rapid rate of deforestation, combined with Ethiopia's rapid population growth, has resulted in increased energy scarcity, high greenhouse gas (GHG) emissions, and climate change. Biogas technology has recently been described as one of the most promising solutions to the problems of deforestation, energy supply, and climate change. Therefore, this study was aimed to investigate the potential of biogas technology in fuelwood saving and carbon emission reduction in the Central Rift Valley of Ethiopia. A multi-stage sampling procedure was employed to select a sample of 192 households (HH) (68 biogas adopters and 124 non-adopters). To conduct the Kitchen efficiency test, 25 test subjects were chosen at random from each category. The data were analyzed using descriptive statistics and an independent-sample t-test. The energy source for cooking and heating were fuelwood, biogas, animal dung, agricultural residue, and charcoal were the most common energy source for domestic use, accounting for 46.9%, 35.4%, 9.4 %, 5.2 %, and 3.1%, respectively. Kerosene lamps, battery cells, and tiny solar panels were utilized by 35 %, 32.4 %, and 18 % of respondents, respectively, for lighting. The most widely used woody plants for domestic energy usage were Eucalyptus camaldulensis and Eucalyptus saligna, according to the findings. The digester size of 6m3 was used in 93 (56.5%) of the 165 biogas plants, while 8m3 and 10m3 digesters were used in 69 (43.5%) and 3 (1.8%) of the plants, respectively. Hence, the annual fuelwood savings from using biogas technology per household was calculated to be 1856.78 kg per year, with an annual Carbon dioxide (CO2) emission reduction capacity of 2.75 tons per biogas plant. Consequently, it was projected that all functional biogas plants (111) would save around 305.25 tons of CO2e per year. Generally, Biogas has been proven to be a viable technique for reducing reliance on forest resources and mitigating climate change in general. The aim of this study was to assess the antifungal effectiveness of neutral electrolyzed water (NEW) to inhibit the spore germination of post-harvest fungi common in fruits, determine the required available chlorine concentration (ACC) of NEW and to compare it with copper oxychloride (CO) and sterile distilled water (SDW) in vitro. This study evaluated the biological effectiveness of NEW to inactivate pure cultures of 11 different fungi obtained from post-harvest tropical fruits with anthracnose, rottenness or necrosis symptoms. A conidial solution of 1 × 104 spores/mL per culture was prepared and treated with a low, medium and high ACC of NEW (pH 7.0 ± 0.05, 12, 33 and 53 mg/L of ACC and ORP of 850 mV), CO at 0.3 g/L, or sterile distilled water as a control, for 3-, 5- and 10-min contact time. Spore germination of Alternaria alternata, Botrytis cinerea, Cladosporium australiense, Colletotrichum gloeosporioides and C. siamense, Fusarium solani and F. oxysporum, and Lasiodiplodia theobromae was inhibited in 100% by NEW at 12, 33 and 53 ppm ACC; 3,5 and 10 min contact time. Aspergillus niger and A. tamarii required 53 mg/L ACC to inhibit 100% of spore germination. Due to the growing competitive challenges, athletes' mutation between clubs has emerged an area of interest within career development. However, studies aimed at analysing this specific process of adaptation to clubs that lead to success or failure in such career mutations are seldom. We developed a comprehensive understanding of the psychological mutation processes to clubs using a narrative level of McAdams’ model of personality. The qualitative method approach used in the Big Three narrative framework (McLean et al., 2019) and good life stories standards (McAdams, 1996) were applied to uncover the components of the narratives of twenty professional basketball players during mutation between clubs. The current study sort to test the links between these narrative components and athlete successful club to club mutation (CCM-successful). On average, the results showed that the participants experienced richer narratives as compared to general population t (17.08) = -1.48, p < .05. Narratives in the CCM-successful were also richer (Mean = 2.83, SD = .16) as compared to narratives of the CCM-unsuccessful (Mean = 1.63, SD, = .36). The overall club mutation success correlated positively with exploratory processing, meaning-making and agency narrative components. These results suggest that when athletes develop active attitudes seeking for solution and sharing their experience with others, they may build the conditions of adaptability that corresponds to a successful outcome for their mutation. A field experiment was carried out in Adiyo, Bonke, Chencha, Sodo zuriya, and North Ari districts, they are found in Southern Ethiopia, during the 2019 main production season. The objective of the experiment was to evaluate the effectiveness of the integrations of host resistance and application of fungicides with designated spray frequency on Fusarium head blight under natural epiphytotic conditions and to determine the management effects on yield and yield components of wheat. The treatments consisted of wheat cultivars (Shorima and Hidase) and fungicides (Propiconazole and Tebuconazole) with four spray frequencies, including an unsprayed one. The experiment was arranged in a split-split plot design with three replications. Wheat cultivars were assigned to the main plots and fungicides allotted to the sub-plots, while the spray frequencies were appointed to sub-sub plots. Results exhibited that integration of wheat cultivars and fungicides with spray frequencies significantly (p < 0.001) reduced disease pressure and increased grain yields across the locations. The highest disease severity (36.46%) and area under disease progress curve (AUDPC) (404.78%-day) and lowest grain yield (2.42 t ha−1) were recorded at North Ari. The lowest severity (7.70%), and AUDPC (130.26%-day), and the highest grain yield (6.68 t ha−1) were recorded in Bonke. The lowest severity (4.78 and 5.74%) and AUDPC (52.86 and 59.78%-day) were recorded from Shorima due to integrated use of Tebuconazole with three and two times spray frequencies, respectively. The highest grain yield of 5.30 t ha−1 was recorded on Shorima in combination with Tebuconazole with three times spray frequencies. The grain yield loss of 46.49% was computed on unsprayed plots of Hidase cultivar. Overall, the results exhibited a combination of moderately resistance wheat cultivars supplemented by fungicide with appropriate spray frequencies right at disease onset reduced disease pressure and increased grain yield. Planting of Shorima combined with Tebuconazole with three times spray frequencies was found effective in reducing FHB epidemics and increasing grain yield with the highest monetary advantage.
