40. Efflux of sphingolipids metabolically labeled with [1-3H]sphingosine, L-[3-3H]serine and [9,10-3H]palmitic acid from normal cells in culture.

Abstract

The membrane complex lipids of human fibroblasts and differentiated rat cerebellar granule cells in culture were metabolically radiolabeled with [1-3H]sphingosine, L-[3-3H]serine and [9,10-3H]palmitic acid. A relevant efflux of radioactive sphingolipids and phosphatidylcholine was observed from cells to the culture medium in the presence of fetal calf serum. This event was independent of the concentration and structure of the metabolic precursor administered to cells, and it was linearly time-dependent. The radioactive lipid patterns present in the medium were different from those present in the cells. Radioactive sphingomyelin and ganglioside GM3 containing short acyl chains were the main species present in the medium from human fibroblasts, while sphingomyelin and GD3 ganglioside in that from neuronal cells. In the absence of proteins in the culture medium, the efflux of complex lipids was much lower than in the presence of serum, and the patterns of released molecules were again different from those of cells. 41. Synthesis of GM1-Ganglioside Inner Ester

Abstract

A new procedure is described for preparing a GM1-ganglioside (GM1) inner ester, in which the sialic acid carboxyl group esterifies with the hydroxyl group at carbon-2 of the inner galactose unit. The reaction of lactonization consists of treatment of GM1 with dicyclohexylcarbodiimide in anhydrous dimethylsulfoxide. Under these conditions, GM1 inner ester is produced with almost quantitative yield (97%) and recovered from the reaction mixture by simple acetone precipitation. The involvement of a sialic acid carboxyl group in an ester linkage was proved by ammonolysis experiments, which gave rise to the amide of sialic acid, and by column chromatography on an ionexchange resin which retained GM1 but not the GM1 inner ester. The GM1 inner ester was analyzed for carbohydrate, long chain base and fatty acid composition by chemical and spectrometric methods. The inner galactose unit, involved in the ester linkage, was recognized by positive and negative ion fast Atom Bombardment Mass Spectrometry and the hydroxyl group at carbon-2, involved in the linkage, by proton Nuclear Magnetic Resonance spectroscopy.

Abstract

The complete definition of the chemical structure of GD1b-ganglioside (GD1b) lactone isolated from human brain has been given by means of spectrometric and spectroscopic analyses. GD1hlactone contains a single ester linkage involving the external sialic acid carboxyl group and the C-9 hydroxyl group of the internal sialic acid unit. A synthetic lactone of GD1bprepared treating GD1bwith glacial acetic acid characterized in the same way showed an identical chemical structure.

43. Association of gangliosides to fibroblasts in culture: A study performed with GM1[14C]-labelled at the sialic acid acetyl group

Abstract

The preparation of a GM1-ganglioside (GM1) [14C]-labelled in the sialic acid residue is reported. This can be obtained by re-N-acetylation in the presence of [1-14C]-acetic anhydride, of a GM1 derivative de-N-acetylated specifically on the sialic acid residue by alkaline hydrolysis of GM1 with tetramethylammonium hydroxide. The radiolabelled GM1 is utilized to investigate the binding properties and the mode of interaction of GM1 with cultured fibroblasts. Three different forms of association (one “serum-removable”, one “trypsin-removable” and one “trypsin-stable”) have been recognized to occur in a way that depended on cell culture conditions (presence or absence of fetal calf serum), ganglioside concentration (from, 5×10−9M to 10−4M) and incubation time (up to 24 h). Some metabolic modifications of GM1 during the period of high cell viability were also investigated.

44. Preparation of the tritiated molecular forms of gangliosides with homogeneous long chain base composition

Abstract

Gangliosides GM2, GM1and GD1bwere radiolabelled at C-6 of the terminal galactose orN-acetylgalactosamine by the galactose oxidase/[3H]NaBH4method; gangliosides GM2, GM1, Fuc-GM1and GD1awere radiolabelled at C-3 of the long chain base by the 2,3-dichloro-5,6-dicyanobenzoquinone/[3H]NaBH4method. By application of an original HPLC procedure, eight different molecular species were prepared from each labelled ganglioside. Each of these species was characterized by the presence of one of the following long chain bases:erythroC18 sphingosine,threoC18 sphingosine,erythroC18 sphinganine,threoC18 sphinganine,erythroC20 sphingosine,threoC20 sphingosine,erythroC20 sphinganine andthreoC20 sphinganine. From GD1bonly the species containing theerythroforms of long chain bases were obtained. The individual molecular species were more than 99% homogeneous and had a radiopurity better than 99%. The molecular species of the same ganglioside, radiolabelled at C-3 of the long chain base, had identical specific radioactivity, namely 1.17, 1.25, 0.85 and 1.28 Ci/mmol for GM2, GM1, Fuc-GM1and GD1arespectively. The molecular species of the same ganglioside, radiolabelled at C-6 of terminal galactose orN-acetylgalactosamine, had similar specific radioactivity, namely 1.34–1.40, 1.44–1.51, 1.37–1.44 Ci/mmol for GM2, GM1and GD1brespectively.

Abstract

Since the 1980s, it has been known that the administration of ganglioside GM1 to cultured cells induced or enhanced neuronal differentiation. GM1 mechanism of action relies on its direct interaction and subsequent activation of the membrane tyrosine kinase receptor, TrkA, which naturally serves as NGF receptor. This process is mediated by the sole oligosaccharide portion of GM1, the pentasaccharide β-Gal-(1-3)-β-GalNAc-(1-4)-[α-Neu5Ac-(2-3)]-β-Gal-(1-4)-β-Glc. Here we detailed the minimum structural requirements of the oligosaccharide portion of GM1 for mediating the TrkA dependent neuritogenic processing. By in vitro and in silico biochemical approaches, we demonstrated that the minimal portion of GM1 required for the TrkA activation is the inner core of the ganglioside's oligosaccharide β-Gal-(1-3)-β-GalNAc-(1-4)-[α-Neu5Ac-(2-3)]-β-Gal. The addition of a sialic acid residue at position 3 of the outer galactose of the GM1 oligosaccharide, which forms the oligosaccharide of GD1a, prevented the interaction with TrkA and the resulting neuritogenesis. On the contrary, the addition of a fucose residue at position 2 of the outer galactose, forming the Fucosyl-GM1 oligosaccharide, did not prevent the TrkA-mediated neuritogenesis., (© 2023. Abstract

The procedures for the preparation of radioactive and photoactivable ganglioside derivatives have been continuously developed from 1989, when for the first time the synthesis of photoactivable tritium labeled GM1 ganglioside was presented. We described previously the synthesis of photoactivable derivatives of GM3 and GM1 gangliosides, tritium-labeled at acetyl group of sugar units, and of photoactivable GM1 and GD1b gangliosides, tritium-labeled at position 6 of the external galactose. These procedures are reviewed in detail in the present paper. The use of these ganglioside derivatives to study the ganglioside-protein interactions and to identify proteins that specifically interact with gangliosides (including GPI-anchored proteins of the outer membrane leaflet, proteins anchored to the cytoplasmic side of the plasma membrane through a fatty acyl chain, transmembrane proteins, and soluble cytoplasmic proteins) is discussed.
