IntroductionTropical cloud forest ecosystems are expected to face reduced water inputs due to climatic changes.MethodsHere, we study the ecophysiological responses of trees and epiphytes within in an Asian cloud forest to investigate the contributions of rainfall, fog, and soil to leaf water in 60 tree and 30 vascular epiphyte species. We measured multiple functional traits, and δ2H, and δ18O isotope ratios for leaf water, soil water, rainfall, and fog in the wettest (July) and driest (February) months. Using a Bayesian stable isotope mixing model, we quantified the relative contributions of soil water, fog, and rainfall to leaf water.Results and discussionRainfall contributes almost all the leaf water of the epiphytes in July, whereas fog is the major source in February. Epiphytes cannot tap xylem water from host trees, and hence depended on fog water when rainfall was low. Most of leaf water was absorbed from soil water in July, while fog was an important source for leaf water in February despite the soil moisture content value was high. In February, lower temperatures, along with reduced photosynthesis and transpiration rates, likely contributed to decreased soil water uptake, while maintaining higher soil moisture levels despite the limited rainfall. These contrasting contributions of different water sources to leaf water under low and high rainfall and for different plant groups outline the community-level ecophysiological responses to changes in rainfall. While direct measurements of water flux, particularly in roots and stems, are needed, our results provide valuable insights on tropical cloud forest hydrology under scenarios of decreased fog immersion due to climatic changes.

IntroductionA critical issue in tropical forests is that anthropogenic deforestation (i.e., mining) degrades the integrity of its ecosystem. Reforestation with appropriate native plant species helps to alleviate these detrimental impacts. A protocol to select appropriate plant species for this purpose currently lacks efficacy and timeliness.MethodsWe provided a trait-based protocol to quickly and effectively select native plant species for mining reforestation. A 0.2-km2 area of Baopoling (BPL) at Hainan Island, China, was used as a study site, which has been severely degraded by 20 years of limestone mining for cement production. First, we identified the tree species in nearby undisturbed tropical forests, followed by evaluating the similarities in functional traits of the most dominant one (target species) and 60 local candidate native plant species (candidate species) whose saplings can be purchased from a local market.Results and discussionThis dataset was used in our trait-based protocol, and only within 1 month, we successfully selected eight plant species which are very similar to target species from the 60 candidate species. We also quantified whether the eight selected plant species were indeed suitable for sustained reforestation by testing their effects on landscape and also their survival rate and recruitment ability after using them to perform reforestation in BPL from 2016 to 2023. Finally, these eight plant species are indeed suitable for reforestation due to their huge influences on a significant shift from originally degraded landscape (comprising only barren rocks) to a forest landscape totally and also their high survival rate (90%–97%) and ability for natural recruitment after 7 years’ reforestation in BPL. Thus, we anticipate that this protocol would be integral to species selection during reforestation of tropical mining areas.

Rapidly global urbanization and economic growth in the past several decades have resulted in a sharp contraction of arable areas worldwide. However, food supply requirements are quickly increasing due to higher living standards and larger populations. Therefore, food crises are still a major threat to human society. The conflict between farmland areas and the increasing need for essential supplies is becoming acuter in China. Therefore, we suggest that a novel strategy would address the issue, in which temporal and spatial arrangement of wheat sowing dates would be highly focused.

Breeders agree that leaf senescence is a favorable process for wheat seed yield improvement due to the remobilization of leaf nutrients. However, several studies have suggested that staying green may be an important strategy for further increasing wheat yields. In this study, we performed a comparative transcriptome analysis between wheat cultivars CN17 and CN19 after heading and also measured photosynthetic parameters, chlorophyll (Chl) contents, and antioxidant enzyme activities at various time points after heading. The physiological and biochemical indexes revealed that CN17 exhibited a functionally stay-green phenotype while CN19 did not. We identified a total of 24,585 and 34,410 differential expression genes between genotypes at two time-points and between time-points in two genotypes, respectively, and we also found that 3 (37.5%) genes for leaf senescence, 46 (100%) for photosynthesis – antenna protein, 33 (70.21%) for Chl metabolism and 34 (68%) for antioxidative enzyme activity were upregulated in CN17 compared with CN19 during leaf senescence, which could be regulated by the differential expression of SAG39 (senescence-associated gene 39), while 22 (100%) genes for photosynthesis – antenna proteins, 6 (46.15%) for Chl metabolism and 12 (80%) for antioxidative enzyme activity were upregulated in CN17 compared with CN19 before the onset of leaf senescence. Here, we further clarified the expression profiles of genes associated with a functional stay-green phenotype. This information provides new insight into the mechanism underlying delayed leaf senescence and a new strategy for breeders to improve wheat yields.

Akebia trifoliata is a perennial climbing woody liana plant with a high potential for commercial exploitation and theoretical research. Similarly, microsatellites (simple sequence repeats, SSRs) also have dual roles: as critical markers and as essential elements of the eukaryotic genome. To characterize the profile of SSRs and develop molecular markers, the high-quality assembled genome of A. trifoliata was used. Additionally, to determine the potential transferability of SSR loci, the genomes of Amborella trichopoda, Oryza sativa, Vitis vinifera, Arabidopsis thaliana, Papaver somniferum, and Aquilegia coerulea were also used. We identified 434,293 SSRs with abundant short repeats, such as 290,868 (66.98%) single-nucleotide repeats (SNRs) and 113,299 (26.09%) dinucleotide repeats (DNRs) in the A. trifoliata genome. 398,728 (91.81%) SSRs on 344,283 loci were physically mapped on the chromosomes, and a positive correlation (r = 0.98) was found between the number of SSRs and chromosomal length. Additionally, 342,916 (99.60%) potential SSR markers could be designed from the 344,283 physically mapped loci, while only 36,160 could be viewed as high-polymorphism-potential (HPP) markers, findings that were validated by PCR. Finally, SSR loci exhibited broad potential transferability, particularly DNRs such as the “AT/AT” and “AG/CT” loci, among all angiosperms, a finding that was not related to the genetic divergence distance. Practically, we developed a whole set of effective, polymorphic, and physically anchored molecular markers and found that, evolutionarily, DNRs could be responsible for microsatellite origin and protecting gene function.

Akebia trifoliata is an important multiuse perennial plant that often suffers attacks from various pathogens due to its long growth cycle, seriously affecting its commercial value. The absence of research on the resistance (R) genes of A. trifoliata has greatly limited progress in the breeding of resistant varieties. Genes encoding proteins containing nucleotide binding sites (NBSs) and C-terminal leucine-rich repeats (LRRs), the largest family of plant resistance (R) genes, are vital for plant disease resistance. A comprehensive genome-wide analysis showed that there were only 73 NBS genes in the A. trifoliata genome, including three main subfamilies (50 coiled coil (CC)-NBS-LRR (CNL), 19 Toll/interleukin-1 receptor (TIR)-NBS-LRR (TNL) and four resistance to powdery mildew8 (RPW8)-NBS-LRR (RNL) genes). Additionally, 64 mapped NBS candidates were unevenly distributed on 14 chromosomes, most of which were assigned to the chromosome ends; 41 of these genes were located in clusters, and the remaining 23 genes were singletons. Both the CNLs and TNLs were further divided into four subgroups, and the CNLs had fewer exons than the TNLs. Structurally, all eight previously reported conserved motifs were identified in the NBS domains, and both their order and their amino acid sequences exhibited high conservation. Evolutionarily, tandem and dispersed duplications were shown to be the two main forces responsible for NBS expansion, producing 33 and 29 genes, respectively. A transcriptome analysis of three fruit tissues at four developmental stages showed that NBS genes were generally expressed at low levels, while a few of these genes showed relatively high expression during later development in rind tissues. Overall, this research is the first to identify and characterize A. trifoliata NBS genes and is valuable for both the development of new resistant cultivars and the study of molecular mechanisms of resistance.

Soybean cyst nematode (SCN) has devastating effects on soybean production, making it crucial to identify genes conferring SCN resistance. Here we employed next-generation sequencing-based bulked segregant analysis (BSA) to discover genomic regions, candidate genes, and diagnostic markers for resistance to SCN race 4 (SCN4) in soybean. Phenotypic analysis revealed highly significant differences among the reactions of 145 recombinant inbred lines (RILs) to SCN4. In combination with euclidean distance (ED) and Δsingle-nucleotide polymorphism (SNP)-index analyses, we identified a genomic region on Gm11 (designated as rhg1-paralog ) associated with SCN4 resistance. Overexpression and RNA interference analyzes of the two candidate genes identified in this region ( GmPLAC8 and GmSNAP11 ) revealed that only GmSNAP11 significantly contributes to SCN4 resistance. We developed a diagnostic marker for GmSNAP11 . Using this marker, together with previously developed markers for SCN-resistant loci, rhg1 and Rhg4 , we evaluated the relationship between genotypes and SCN4 resistance in 145 RILs and 30 soybean accessions. The results showed that all the SCN4-resistant lines harbored all the three loci, however, some lines harboring the three loci were still susceptible to SCN4. This suggests that these three loci are necessary for the resistance to SCN4, but they alone cannot confer full resistance.
