Using the INFOGEST in vitro digestion protocol adapted to carotenoids, the impact of additional rabbit gastric lipase (RGL) on the hydrolysis extent of β-cryptoxanthin esters was evaluated for the first time, and compared with the addition of porcine cholesterol esterase (CEL). Both the modifications increased the hydrolysis of (all-E)-β-cryptoxanthin esters from mandarin and peach pulps, although the outcomes were different. Addition of RGL consistently increased the average hydrolysis extent from 55.2% to 59.5% in mandarin pulp and from 22.7% to 48.8% in peach pulp (p 0.05). The addition of CEL produced lower hydrolysis extents, i.e., 58.5% in mandarin (not statistically significant) and 28.4% in peach (p0.05), compared to those obtained with RGL. The hydrolysis extent positively correlated with the carotenoid ester concentration in both matrices. Bioaccessibility values were higher in mandarin pulp (range 32-34%) compared to those in peach pulp (range 16-21%), and were associated with the hydrolysis extent of the carotenoid esters during digestion. Addition of RGL and CEL produced no significant (p 0.05) effect on the overall carotenoid bioaccessibility values of mandarin, while positively affected those in peach. Altogether these results corroborate that the hydrolysis extent of xanthophyll esters limits bioaccessibility. Additionally, hydrophobicity of the carotenoid inversely correlates with micellarization, as free (all-E)-xanthophylls micellarized in a higher extent compared to (all-E)-β-carotene and xanthophyll esters. The new information of our results is that the addition of rabbit gastric lipase substantially contributes to the hydrolysis of β-cryptoxanthin esters from fruit pulps, and consequently, to increase carotenoid bioaccessibility, being even more effective than CEL.

In fermented milks inoculated with two thermophilic strains (Lactobacillus bulgaricus and Streptococcus thermophilus), guabiroba pulp (Campomanesia xanthocarpa O. Berg) was added in different concentrations: 5% (I5 sample) and 10% (I10 sample), compared to a control sample, with no pulp addition. In these fermented milks, Bifidobacterium BB-12 was added and the samples were submitted to a progressive gastrointestinal simulation in vitro. The cells count was performed, including the survival rates for all the progressive steps of the simulated digestion. Total phenolic content (TPC) and antioxidant activity analysis by FRAP (Ferric Reducing Antioxidant Power) and DPPH (2,2-diphenyl-1-picrylhydrazyl) were performed in all the gastrointestinal steps. Before and during the entire gastrointestinal tract, the Bifidobacterium BB-12 count was 8–9 log CFU g−1, above the recommended for a probiotic product, with a highlight in intestinal colon steps. The loss and waste of food is a matter of great concern, leading to a multifaceted problem with negative economic, social, and environmental impacts as addressed in the UN Sustainable Development Goals number 2: zero hunger. The wine, fruit juice and vegetable oil processing industries generate significant amounts of wastes and side streams containing potentially valuable bioactive compounds. Some of them are plant secondary phenolic metabolites that offer remarkable health benefits (as antioxidants and anti-inflammatory compounds). One of the current challenges is the recovery of such bioactive compounds from residual matrices for further applications in food, pharmaceutical and cosmetic industries. Within this framework and in the scope of the Green Chemistry concept, one of the current challenges is to find eco-efficient techniques for the recovery of bioactive compounds. In this context, neoteric solvents are considered a greener alternative to traditional solvents, as the latter are more harmful to human and animal health, and environment. This overview focuses on recent advances in the use of hydrophobic neoteric solvents, i.e. ionic liquids, eutectic solvents, and bio-based solvents, for liquid-liquid extraction of phenolic compounds from liquid agri-food matrices.

The blackberry fruit ( Rubus spp.) presents an attractive color ranging from purple red to blue, due to the high content of bioactive compounds, such as anthocyanins. Bioactive compounds have antioxidant properties, acting as free radical scavengers and are also frequently linked to the prevention of cardiovascular and other chronic diseases. Due to native-form anthocyanins instability against environmental stress, bioactive compounds are not always as effective in improving human health as they could be. Therefore, the production of powders using adjuvants is a promising approach for encapsulating anthocyanins-rich pastes, in order to improve their stability and application. Thus, the aim of the present study was to evaluate the influence of adjuvants on the physical properties of the powder, and on anthocyanin content and antioxidant activity of the pastes and freeze-dried milk-blackberry pulp mixtures. The pastes formulated were of milk-blackberry pulp (25%:75%, (weight/weight ( w /w)) and of milk-blackberry pulp-adjuvant (25%:70%:5%, (w/w/w)), and the adjuvants employed were maltodextrins 10DE and 20DE and Capsul®. The addition of maltodextrin 20DE and Capsul® resulted in pastes with high anthocyanins content (451.6 ± 80.0 μg/g and 453.1 ± 26.0 μg/g, respectively), similar to the paste without adjuvants (568.6 ± 3.4 μg/g). Also, maltodextrin 20DE and Capsul® showed a high protection capacity of the anthocyanins present on the powders (around 96% of anthocyanins retention). When no adjuvants were used, the anthocyanins retention was very low (40.1%). The freeze-drying process maintained the antioxidant capacity of the bioactive compounds and the powders obtained showed good quality with low moisture content and hygroscopicity.

Prawn allergy is one of the most common food-borne allergies and current prevention is by avoidance. This review paper summarised different methodologies for the extraction, identification and quantification of prawn protein allergens, reported in various research studies. Following extraction, allergenic components have been analysed using well-established methodologies, such as SDS-PAGE, Immunoblotting, ELISA, CD Spectroscopy, HPLC, DBPCFC, SPT etc. Moreover, the preference towards Aptamer-based technique for allergenicity analysis has also been highlighted in this review paper. The summary of these methodologies will provide a reference platform for present and future research directions.

With two compounds first discovered in quinoa, an entire novel family of betalain pigments derived from dopamine is obtained and characterized. Betalains are nitrogenous water-soluble pigments and bioactive molecules with health-promoting effects and nutraceutical potential. It was assumed that all betalains contained betalamic acid as a structural unit derived from l-dihydroxyphenylalanine (l-DOPA). However, hitherto ignored compounds derived from dopamine have recently been discovered in nature. Here an entire family of betalains is described as decarboxylated pigments where 6-decarboxy-betalamic acid is the chromophoric and structural unit. This paper shows for the first time the production, purification and characterization of color and fluorescent properties of this novel family of pigments. Antioxidant and anti-aging effects of the just discovered betalains were tested in vivo using the animal model Caenorhabditis elegans. Some of them presented extraordinary properties, being glutamic acid-6-decarboxy-betaxanthin the most fluorescent molecule among both families of betalains. Methionine sulfoxide-6-decarboxy-betaxanthin is described as the most potent betalain in the reduction of oxidative stress in vivo in C. elegans (99.5 % at 25 µM) and dopa-6-decarboxy-betaxanthin increased the lifespan of the animal model up to 7.0 % at 25 µM. These results open new research lines in the search for molecules from plants with health-promoting properties and bioactivities.

Demand for honeydew honey (HDH) is growing, both from consumers and the food industry, due to its potential antimicrobial, anti-inflammatory, and antioxidant properties. However, information on the chemical profile of HDH remains scant, particularly on New Zealand honeydew honey (NZHDH). This paper aims to provide a comprehensive chemical analysis of NZHDH produced from honeybees, which feed on nectar exuded by the scale insect Ultracoelostoma assimile. This insect feeds on most species of the Southern Beech (Nothofagus spp.). The proximate composition, mineral profile, sugar profile, phenolic profile, amino acid profile, and antioxidant activity of NZHDH was determined and compared to results for HDH and other varieties of honey previously reported in literature. It was determined that the antioxidant activity of NZHDH is comparable to several studies on Manuka honey, which is widely considered as the "gold standard" for its antioxidant activity. This demonstrated that NZHDH indeed had excellent antioxidant properties. This study was the first time that a quantitative amino acid profile has been produced for NZHDH. The major amino acids were proline, L-aspartic acid, L-glutamic acid, L-alaine, and L-phenylalanine. The major phenolic compounds were pinocembrin, abscisic acid, and pinobanksin. Of the phenolic compounds quantified in this study, only p-hydroxybenzoic acid has previously been quantified in NZHDH.

Fucoxanthin (FUC) is a hydrophobic carotenoid that has a protective effect on the colon. To exert the beneficial effects of FUC in the colon and expand its application in functional food, FUC was encapsulated in carboxymethyl konjac glucomannan (CMKGM)-coated gliadin nanoparticles (Gli-CMKGM NPs) in this paper. Gli-CMKGM NPs were prepared at pH 5.0 with Gli/CMKGM mass ratio of 1:1. The formation of Gli-CMKGM NPs was associated with hydrogen bonding, hydrophobic interactions and electrostatic attractions. Additionally, Gli-CMKGM NPs exhibited good stability to pH, salt, heating and storage. The results showed that FUC had been successfully encapsulated in Gli-CMKGM NPs, and the encapsulation efficiency of FUC-Gli-CMKGM NPs was significantly higher than that of uncoated FUC-Gli NPs. FUC-Gli-CMKGM NPs had a nano-spherical structure, and embedded FUC in Gli-CMKGM NPs improved their stabilities to photodegradation and thermal degradation. Furthermore, in vitro release and in vivo organ distribution studies showed that FUC-Gli-CMKGM NPs had an excellent colon targeting function. Overall, our findings illustrated the promise of CMKGM-coated Gli NPs for constructing targeted delivery systems for FUC.

The oral environment is an essential part of the human microbiome. The consumption of probiotic products may improve the oral microbiota and reduce the risk of diseases. This paper presents a bibliometric and critical review of randomized clinical trials (RCTs) that used probiotics to analyze oral parameters in humans. RCTs carried out with no age, gender, and ethnicity restrictions and published in the pre-COVID-19 period were included. Furthermore, the utilization of probiotic dairy products to improve oral health is discussed. The bibliometric review demonstrated that 'Microbiology,' 'Dental caries,' and 'Streptococcus mutants' were the most highlighted keywords. Furthermore, Sweden and India have the highest number of publications. The most prevalent outcomes were 'salivary parameters,' 'periodontal disease,' and 'dental caries.' The most used vehicles for probiotic administration were pharmaceutical formulas and dairy products. The administration of probiotic dairy products could modify the oral microbiota (reductions in S. mutans counts), influence the caries development and periodontal disease in children, adolescents, adults, and the elderly, and improve gingival health. The main probiotic dairy products investigated were milk, fermented milk, yogurt, kefir, curd, and cheese. Lacticaseibacillus paracasei SD1 was the most used probiotic culture. The studies demonstrated that the probiotic effect lasted 2-4 weeks after discontinuing consumption. However, the results depended on the subject type, study design, probiotic strain and concentration, and dairy product type. In conclusion, probiotic dairy products are promising alternatives to improve oral health.

Food spoilage and waste, human and animal poisoning, and even death caused by foodborne microorganisms remain extensive concerns in food safety. The global demand for functional, eco-friendly, and efficient antimicrobial food packaging is increasing. However, the bacteriostatic or bactericidal effects of most conventional food packaging display limited action, and their major components are petrochemical materials (non-renewable, non-biodegradable, and not environmentally friendly), and the current target microorganisms easily acquire drug-resistant. Therefore, the development of more effective, sustainable and safe antimicrobial materials has become a research hotspot in food packaging. Among legumes, broad bean (Vicia faba L.) has received particular attention due to its nutraceutical, functional and economic importance. The plant-derived microRNAs (miRNAs), as novel dietary functional components, are found to regulate the expression of endogenous mRNAs in vertebrates. To understand the role of broad bean miRNAs in the regulation of muscle texture, we investigated the miRNA-mRNA network in the established crisp grass carp model fed with broad bean. We identified various miRNAs that potentially improved muscle texture; miR-101b-3p activated Wnt signaling and satellite cell proliferation; miR-126-3p, miR-29a and miR-148b promoted hyperplasia by targeting muscle structure genes (tln2, TPM1, etc.); miR-152-5p and miR-185 regulated collagen expression via Smads signaling pathways; and miR-146a and miR-371-3p increased reactive oxygen species (ROS) by suppressing cat and prdx6. Among these changes, at least the Wnt signal activation was driven by broad bean-derived miR-101b-3p. This paper was conducted to investigate the cross-kingdom regulatory effects of broad bean miRNA on muscle structure and provide basic data for the development and application of broad bean.

Milk powders are commonly used for a variety of food products in which among others the milk proteins add to the properties of the products. Processing of milk can, depending on the processing parameters, change the size and structure of the proteins. These changes can be difficult to measure due to the polydispersity of milk components, which makes it a challenge to obtain direct information about the individual proteins. In this paper, the results from an investigation of casein micelle size,size distribution, and structure in reconstituted skim milk and the comparison with raw and pasteurized skim milk are reported. The investigation used asymmetrical flow field-flow fractionation (AF4) in combination with online UV, multi-angle light scattering (MALS), and refractive index (RI) detection and the results were confirmed by transmission electron microscopy (TEM). The results show that there is a difference in casein micelle size distribution between the differently processed milk samples. The casein micelles of the reconstituted milk were found to have a z-average radius of gyration of 72 nm and the casein micelles in the raw and pasteurized skim milk were 58 and 62 nm respectively. The AF4 and TEM data suggest that the cause of the larger casein micelle size is a layer of aggregated whey proteins associated with the casein micelles surface. Moreover, the TEM investigation showed that a larger proportion of the casein micelles are aggregated in reconstituted milk compared to raw and fresh skim milk. Investigation of the effect of reconstitution time shows that the amount of aggregated casein micelles decreases during the first 20 min of reconstitution. The results show that the AF4-method can provide detailed insights into the reconstitution process and properties of different milk samples. Hence, it can be used as a reference or validation for more indirect methods to track the reconstitution of milk powders.

Understanding potential cross-cultural sensory differences in the perception of Irish dairy products is important for key markets such as the USA and China. As most Irish dairy products are produced from pasture derived milk, this study investigated the impact of pasture and non-pasture diets on the cross cultural sensory perception of skim milk powder (SMP) in Ireland, USA and China. SMP was produced from cows fed outdoors on ryegrass (GRS), ryegrass/white clover (CLV), and indoors on trial mixed rations (TMR). SMP samples were evaluated by Irish (n = 78), USA (n = 100) and Chinese (n = 106) consumers using an identical hedonic sensory acceptance test in Ireland, USA and China. Optimized Descriptive Profiling (ODP) was performed using trained assessors familiar with dairy products in Ireland (n = 25) and China (n = 22), and traditional descriptive analysis was undertaken by a trained panel (n = 7) in the USA. Volatile analysis was undertaken on each SMP sample. Hedonic assessment found that USA consumers preferred SMP derived from TMR, and Irish consumers preferred SMP from either GRS or CLV. Chinese consumers perceived SMP samples differently to the USA and Irish consumers, but preference was not influenced by diet. Both Irish and Chinese trained assessors found it more difficult to discern differences between GRS or CLV SMP, but could differentiate TMR SMP. Irish assessors preferred GRS and CLV SMP. Chinese and Irish assessors had different preferences for many attributes. Trained USA panelists found significant differences, exclusively associating pasture based diets with “cowy/barny” and “cardboard/wet paper” attributes and more intense “grassy/hay” attributes than in TMR SMP. The abundance of ten volatile compounds differed significantly based on diet with acetoin derived from carbohydrate metabolism at much greater abundance in TMR SMP. This study found that sensory perception and volatile profiles of SMP were influenced by diet and differences in sensory perception existed between the three cultural groups. Irish and USA sensory responses aligned with familiarity of dairy products derived from pasture and non-pasture diets, respectively, and Chinese sensory responses differed to Irish and USA responses likely reflecting their lack of familiarity with dairy products.

As a healthy food and traditional Chinese medicine, sagittaria sagittifolia L. has been used for a long history. Nevertheless, reports on the bioactivity and chemical characterization of S. sagittifolia L. polysaccharides are still very rare. In this paper, ultrasound-assisted method (UAE) was used to extract S. sagittifolia L. polysaccharides, after alcohol precipitation and column chromatography isolation, the structural characteristics, antioxidant and immunological activities of the purified polysaccharide (SPU60-W) were preliminarily investigated. The results indicated SPU60-W (16.62 kDa) was a pyranoid polysaccharide containing α-glycosidic bond composed of mannose, xylose, and glucose with a molar ratio of 2.69: 2.04: 95.27. It consisted of slender wormlike strands, which may involve some degree of aggregation of helices, as well as a small proportion of irregular spherical structures. Furthermore, antioxidant activity analysis showed that SPU60-W possess excellent hydroxyl and ABTS radical scavenging activity comparable to vitamin C (Vc), and moderate DPPH radical scavenging activity. Immunity tests suggested that SPU60-W significantly promoted the proliferation, phagocytosis and NO production of mouse macrophage RAW264.7. According to this study, SPU60-W might be utilized as a potent antioxidant and immunomodulator in food and medicinal industry.

Both the Maillard reaction (MR) and thermal treatment influence the nutritional value of milk. In this paper, the capability of polyphenolic berry extract (PBE) to inhibit MR in an ultra-high temperature (UHT) treated milk was investigated. Total polyphenol content and antioxidant capacity of blueberry (BE) and raspberry extracts (RE) were also tested. A gas chromatography-mass spectrometry (GC–MS) method was developed to monitor the MR product N e-(carboxymethyl)-L-lysine (CML) and L-lysine (LYS). PBE was added to milk at 0.05 and 0.1% w/v prior to UHT processing. Data revealed that formation of CML was significantly reduced (23.4 ± 5.1%) by addition of 0.1% w/v BE. The final concentrations of LYS measured following the addition of PBE prior to thermal treatment were statistically similar to the control milk which was not subjected to thermal processing. Additionally, the metabolic profile of milk samples was investigated by GC–MS and visualised using ‘FancyTiles’.

Emojis can be used to explore food-evoked emotions in order to provide information that can support the product development and marketing decisions. This study aimed to evaluate consumers' acceptance, purchase intent and emotional responses to milk beverages, with and without kefir added, before and after these consumers were informed about the products' composition (0%, 15%, 30% and 50% m/v) and health claims toward kefir (blind and informed tests, respectively). Emotional responses were assessed by emoji use within a RATA questionnaire in order quantify the perceived significance of the emojis chosen. In the informed test, the consumers' perception of the sensory attributes of the milk beverages, such as their perception of an acid taste in added kefir beverages was shown to have changed. Overall, participants attributed significantly higher acceptance and purchase intent scores to added kefir beverages after they had been informed on its health benefits. In addition, expressions of positive emotion increased when participants were exposed to stimuli related to health benefits of kefir (15%, 30% and 50% m/v), while negative expressions of emotion decreased. The provided information of kefir modified valence and arousal in subjects, and it can be said that to 30% of kefir can be added to yogurt without compromising its sensory acceptability. Thus, health benefits alone cannot improve product acceptance, since participants found a 50% addition of kefir to be unpleasant when tasted during a blind test. Mixed beverages may present a probiotic beverage alternative for consumers who dislike kefir milk, but want to include it in their diets. The implications of liking and purchase intent and how they are linked to emotions are discussed in this paper as well.

The purpose of this paper is to investigate the trace metal content in edible biomonitors (i.e., mollusks) in the Beagle Channel (southern Patagonia) and to assess the human health risks associated with their consumption. Rationale: The monitoring breakdown structure (MBS) conceptual model was applied to four sampling campaigns (2005 → 2012) that collected 729 samples of Mytilus chilensis and Nacella magellanica. The composition of trace elements (Cd, Cr, Cu, Ni, Pb and Zn) in the mollusks was determined using graphite furnace (GFAAS) or flame atomic absorption spectrometry (FAAS). We compared the mean obtained values with the maximum levels (MLs) of each element established by international organizations. Then, based on semi-structured interviews, we calculated the estimated daily intake (EDI) of local residents and compared it with safety reference doses, i.e., the provisional tolerable daily intake (PTDI), provisional maximum tolerable daily intake (PMTDI), and tolerable daily intake (TDI), as well as the benchmark dose level lower confidence limit for Pb (BMDL01, a reference point (RP)/point of departure (POD). Moreover, to obtain information about the potential health risks of ingesting heavy metals (HMs) through mollusk consumption, we evaluated the target hazard quotient (THQ) and the hazard index (HI). Findings: For Cd and Pb, 65% and 40% of bivalves exceeded the MLs established by the Mercado Comun del Sur (Mercosur), respectively. Except for Cd in N. magellanica (i.e., 1.20 μg/kg/bw/day), EDI values were clearly lower than the safety reference doses. For Cr, Cu, Ni, Pb and Zn, mussels were safe for consumption and did not raise concerns for public health. Likewise, THQ values were well below one for most of the studied metals, indicating that the exposed human population is assumed to be safe. Occasional high consumers of mollusks from the most contaminated sites may be at some health risk. Originality: The food production system and the environment are complex systems; this is crucial to understand when we consider ecosystems as a food source (i.e., marine ecosystems). Here we consider edible biomonitors, that are organisms that can have a dual function. They are food, and at the same time, if properly calibrated, they can act as indicators of environmental quality. This study is the first to investigate relevant essential and non-essential trace metal content in two edible mollusks from the Beagle Channel in a long-term survey (2005 → 2012). The information variety was high; approximately thirteen thousand determinations were conducted to support the risk assessment for mollusk consumption. Other aspects connected with the health risks and the uncertainty factors related to the presence of essential and non-essential minerals in edible mollusks as well as the use of the MBS are also discussed.

This review presents up-to-date information about current research on nicarbazin, one of the most used anticoccidials in poultry production. The focus is to elucidate regulation concerning nicarbazin, limits for its residues in food, how maximum residue limits in different countries are calculated regarding edible chicken tissues and the possible implications in human health. Analytical methods to extract and quantify this residue, expressed as dinitrocarbanilide (DNC) are presented and discussed, including qualitative screening and quantitative/confirmatory analytical methods. Monitoring results and occurrence of DNC residues in chicken meat are discussed. Additionally, the causes of eventual chicken meat contamination and possible solutions to reduce or eliminate DNC residue in tissues are also presented. The paper concludes with perspectives, the current state of DNC residue analysis and suggestions for future research, especially considering the gap in the study of residue recycling effect due to continuous chicken litter use. Pectin, an abundant polysaccharide in the human diet, has structural characteristics and functional properties that are strongly dependent on the food matrix (e.g., origin, type, cultivar/variety, ripening stage, style and intensity of processing). These polysaccharides have a strong effect on lipid digestion, which is required for the liberation of carotenoids from emulsified lipid droplets in the gastrointestinal content and for the formation of micelles, in which the carotenoids must be incorporated before absorption. Only micellarized carotenoids can be absorbed and subsequently exert protective effects on human health. The alteration of lipolysis by pectin can occur through several mechanisms; however, they have not been linked directly to carotenoid micellarization. This paper provides an overview of the effects of the properties of pectin on the ion concentration in the digestive content, the viscosity of the digestive medium, the properties of the lipid droplet surfaces and lipase activity and analyzes the impact of these events on lipid digestion and subsequent carotenoid micellarization.
