The major lipids and antioxidant activities of Asterias rolleston gonad lipids were evaluated systematically. Major lipids of A. Rolleston gonad lipids were triacylglycerols (TAGs) and phospholipids (PLs). Total lipids were composed of 15.62% of polyunsaturated fatty acids (PUFAs), and 40.81% of monounsaturated fatty acids (MUFAs). The most abundant PUFA were C20:5n-3 (EPA) (6.28%) and C22:6n-3 (DHA) (5.80%). Predominantly composed of phosphatidylcholine (PC) and phosphatidylethanolamine (PE), polar lipids were rich in PUFAs and could contain up to 34.59% EPA and DHA, and PE and PI (phosphatidylinositol) were also found to be the main carriers of EPA and ARA (arachidonic acid) in polar lipids. The MUFA and PUFA of Sn -2 in TAG are 39.72% and 30.37%, respectively. A total of 64 TAG species were identified, with Eo-P-M, Eo-Eo-M, and M-M-Eo being the main TAGs components. • Fatty acid distribution is not uniform in fish oil/milkfat solid lipid particles. • Saturated fatty acids cover the outermost layer of lipid particles. • Better stability with higher milkfat due to transformation to β' during storage. • Raman spectroscopy can monitor fatty acids distribution in lipid particles. • Polymorphism in lipid particles is inevitable even with low storage temperatures. Saturated fatty acid-containing lipids, such as milkfat, may protect long chain polyunsaturated fatty acids in fish oil when blended together into solid lipid particles (SLPs). One of the main challenges of SLPs is structural polymorphism, which can lead to expulsion of the protected component during prolonged storage. To investigate this phenomenon, the change in thermal and crystalline behaviours, and fatty acid distribution, were analysed in SLPs of fish oil and milkfat during storage at different temperatures for up to 28 days. X-ray diffraction analysis showed changes in molten and crystalline states occurred even at –22 °C. Room temperature (21 °C) storage led to more than 45% molten state but SLPs retained their initial shape. Confocal Raman Spectroscopy of the SLPs showed the distribution of fatty acids was not uniform, with 10 μm outermost layer of predominantly saturated fatty acids likely responsible for the intact SLP shape and stability of the core. • High-value polar lipids from C. vulgaris were obtained using EtOH with UAE. • UAE and DCM showed the highest lipid yields and purity and EtOH the lowest yields. • The polar lipidome of DCM and UAE extracts was similar, with minor differences. • EtOH had lower efficiency for extraction of anionic lipids such as PI, SQDG and PG. • UAE and EtOH lipid extracts revealed antioxidant activity comparable to DCM. Glycolipids and phospholipids are the main reservoirs of omega polyunsaturated fatty acids in microalgae. Their extraction for the food industry requires food grade solvents, however, the use of these solvents is generally associated with low extraction yields. In this study, we evaluated the lipid extraction efficiency of food-grade ethanol, ultrasound-assisted ethanol (UAE) and dichloromethane/methanol (DCM) from Chlorella vulgaris cultivated under autotrophic and heterotrophic conditions. Yields of lipids, fatty acids (FA), and complex lipid profiles were determined by gravimetry, GC–MS, and LC-MS/MS, respectively. UAE and DCM showed the highest lipid yields with similar purity. The FA profiles were identical for all extracts. The polar lipidome of the DCM and UAE extracts was comparable, while the EtOH extracts were significantly different. These results demonstrated the effectiveness of UAE extraction to obtain high yields of polar lipids and omega-3 and -6-rich extracts from C. vulgaris that can be used for food applications. Abstract: Farmed New Zealand King Salmon (Oncorhynchus tshawytscha) was prepared according to common consumer techniques; raw, poached, steamed, microwaved, pan fried (no added oil), oven baked (no added oil) and deep fried (in sunflower oil). The fatty acid profile was investigated to determine the optimal preparation techniques to achieve both optimal sensory and nutritional qualities, in particular the levels of long chain polyunsaturated omega-3 fatty acids. The modified Bligh and Dyer method was used for lipid extraction and the Hartman and Lago method for FAMES preparation. Fatty acid composition was determined by gas chromatography. There were moisture and lipid losses during cooking amongst the different methods. The fatty acid profile showed only minor differences between the methods apart from an increase in PUFA in the deep fried salmon due to linoleic acid uptake from the frying oil. In all the cooking methods the omega-3 fatty acids were well preserved. However, deep fried showed the lowest amounts of omega-3 fatty acids. As the results showed good preservation of omega-3 fatty acids regardless of cooking method, there may be possible “internal protection” of omega-3 fatty acids in King Salmon that warrants future research.
