Cerebellar transcranial direct current stimulation (ctDCS) modulates cerebellar cortical excitability in a polarity-dependent manner and affects inhibitory pathways from the cerebellum. The cerebellum modulates spinal reflex excitability via the vestibulospinal tract and other pathways projecting to the spinal motor neurons; however, the effects of ctDCS on the excitability of spinal motor neurons and vestibulospinal tract remain unclear. The experiment involved 13 healthy individuals. ctDCS (sham-ctDCS, anodal-ctDCS, and cathodal-ctDCS) was applied to the cerebellar vermis at 2 mA with an interval of at least 3 days between each condition. We measured the maximal M-wave (Mmax) and maximal H-reflex (Hmax) in the right soleus muscle to assess the excitability of spinal motor neurons. We applied galvanic vestibular stimulation (GVS) for 200 ms at 100 ms before tibial nerve stimulation to measure Hmax conditioned by GVS (GVS-Hmax) and calculated the change rate of Hmax by GVS as the excitability of vestibulospinal tract. We measured the Mmax, Hmax, and GVS-Hmax before, during, and after ctDCS in the sitting posture. No main effects of tDCS condition, main effects of time, or interaction effects were observed in Hmax/Mmax or the change rate of Hmax by GVS. It has been suggested that ctDCS does not affect the excitability of spinal motor neurons and vestibulospinal tract, as measured by neurophysiological methods, such as the H-reflex, in healthy individuals in a sitting posture. Effect of ctDCS on other descending pathways to spinal motor neurons, the neurological mechanism of tDCS and the cerebellar activity during the experiment may have contributed to these results. Therefore, we need to investigate the involvement of the cerebellum in Hmax/Mmax and the change rate of Hmax by GVS under different neuromodulation techniques and postural conditions. Motoneuron excitability is possible to measure using H-reflex and V-wave responses. However, it is not known how the motor control is organized, how the H-reflex and V-wave responses modulate and how repeatable these are during dynamic balance perturbations. To assess the repeatability, 16 participants (8 men, 8 women) went through two, identical measurement sessions with ~ 48 h intervals, where maximal isometric plantar flexion (IMVC) and dynamic balance perturbations in horizontal, anterior–posterior direction were performed. Soleus muscle (SOL) neural modulation during balance perturbations were measured at 40, 70, 100 and 130 ms after ankle movement by using both H-reflex and V-wave methods. V-wave, which depicts the magnitude of efferent motoneuronal output (Bergmann et al. in JAMA 8:e77705, 2013), was significantly enhanced as early as 70 ms after the ankle movement. Both the ratio of M-wave-normalized V-wave (0.022–0.076, p < 0.001) and H-reflex (0.386–0.523, p < 0.001) increased significantly at the latency of 70 ms compared to the latency of 40 ms and remained at these levels at latter latencies. In addition, M-wave normalized V-wave/H-reflex ratio increased from 0.056 to 0.179 (p < 0.001). The repeatability of V-wave demonstrated moderate-to-substantial repeatability (ICC = 0.774–0.912) whereas the H-reflex was more variable showing fair-to-substantial repeatability (ICC = 0.581–0.855). As a conclusion, V-wave was enhanced already at 70 ms after the perturbation, which may indicate that increased activation of motoneurons occurred due to changes in descending drive. Since this is a short time-period for voluntary activity, some other, potentially subcortical responses might be involved for V-wave increment rather than voluntary drive. Effects of low-threshold afferents from the anterior (DA), middle (DM) and posterior parts of the deltoid (DP) on the excitability of biceps brachii (BB) motoneurons in humans were studied. We evaluated the effects on individual motor units and motoneuron pool using a post-stimulus time-histogram (PSTH) and an electromyogram-averaging (EMG-A) methods, respectively, in 11 healthy human subjects. Electrical conditioning stimulation was delivered to the axillary nerve branch innervating DA (DA nerve), DM (DM nerve) and DP (DP nerve) with the intensity below the motor threshold. In the PSTH study, stimulation to the DA, DM and DP nerves produced a significant peak (facilitation) in 26/40 (65%), 28/47 (59%) and 0/32 (0%) of BB motor units, respectively. Since the central latency of the facilitation from the DA and DM nerves was 0.1 ± 0.3 and 0.1 ± 0.2 ms (mean ± S.D.) longer than that of the homonymous monosynaptic Ia facilitation of BB, respectively, the facilitation thus being compatible with monosynaptic path. In the EMG-A study, stimulation to the DA and DM nerves produced a significant peak (facilitation) for the BB motoneuron pool in all the subjects, whereas stimulation to the DP nerve produced no effect on BB. The facilitation diminished by vibration stimulation, and the suppression lasted for 30–40 min after removal of the vibration. Therefore, group Ia afferents should be responsible for the facilitation. These findings suggest that monosynaptic facilitation mediated by group Ia afferents from the DA and DM nerves to BB motoneurons exists in humans. [ABSTRACT FROM AUTHOR]

Modulation of a Hoffmann (H)-reflex following transcranial magnetic stimulation (TMS) has been used to assess the nature of signals transmitted from cortical centers to lower motor neurons. Further characterizing the recruitment and time-course of the TMS-induced effect onto the soleus H-reflex adds to the discussion of these pathways and may improve its utility in clinical studies. In 10 healthy controls, TMS was used to condition the soleus H-reflex using TMS intensities from 65 to 110% of the resting motor threshold (RMT). Early facilitation [− 5 to − 3 ms condition-test (C-T) interval] was evident when TMS was 110% of RMT (P < 0.05). By comparison, late facilitation (+ 10 to + 20 ms C-T interval) was several times larger and observed over a wider range of TMS intensities, including 65–110% of RMT. The early inhibition (− 3 to − 1 ms C-T interval) had a low TMS threshold and was elicited over a wide range of intensity from 65% to 95% of RMT (all P < 0.05). A second inhibitory phase was seen ~ 4 ms later (+ 1 to + 4 ms C-T intervals) and was only observed for a TMS intensity of 95% of RMT (P < 0.05). The present findings reaffirm that subthreshold TMS strongly modulates soleus motor neurons and demonstrates that distinct pathways can be selectively probed at discrete C-T intervals when using specific TMS intensities. Action observation (AO) and motor imagery (MI) are potential methods to improve various motor functions. AO and MI facilitate corticospinal excitability. Additionally, the combination of AO and MI (AO + MI) facilitates greater corticospinal excitability than AO alone. However, it is unclear whether AO + MI facilitates the excitability of spinal motor neurons better than AO alone. Thus, in this study, we compared the excitability of spinal motor neurons between AO + MI and AO conditions using F-waves. We recruited healthy adult volunteers (n = 31) for F-wave analysis. We recorded F-waves during relaxation to determine the baseline level (Rest), followed by measurement during AO and AO + MI. To counterbalance the order effect, 15 participants performed AO first, while the others performed AO + MI first. F-wave persistence was significantly increased during AO and AO + MI compared to Rest. The F-wave/M-wave amplitude ratio was significantly increased during AO + MI compared to Rest. The present results suggest that the combination of AO and MI facilitates the excitability of spinal motor neurons compared to Rest, whereas AO alone does not. [ABSTRACT FROM AUTHOR]

This is the response to Prof. Tűrker on his note "challenging" our hypothesis assuming the possibility of two modes of spiking (direct and delayed) evoked by Ia-afferent electrical stimulation in human firing motoneurones. Besides, we touch on some common questions about the most relevant, functionally significant, estimations of motoneuronal firing behaviour in human motor control. In particular, in this context, the question arises: what is more important estimation, motoneurone spiking, itself, or a value of the corresponding interspike interval of the firing motoneurone? This is a note challenging the claim by Kudina and Andreeva's recent publication in Experimental Brain Research. In that publication, Kudina and Andreeva (Exp Brain Res 239:719–730, 2021) put forward a new idea about discovering two spiking modes in human motoneurons. We suggest that what they have shown in their publication maybe is the motor unit firing indicating the end of a net synaptic potential. We reason this challenge from our previous publication in the same journal. It was shown in previous studies that endurance training enhanced excitability of rat spinal motoneurons. However, the influence of the training on the spinal interneurons remains unclear. In this study, we investigated the training effects on spinal interneurons in dorsal and ventromedial area in mice (P42–P50). The electrophysiological properties of the interneurons were recorded from spinal cord slices (T13-L6) by whole-cell patch-clamp recording. The interneurons could be classified into three types based on their response to step currents: single spike (type 1), phasic firing (type 2), and tonic firing (type 3) in both control and trained mice. Interneurons collected from control mice possessed rheobase of 11.3 ± 6.0 pA and voltage threshold (Vth) of − 37.3 ± 4.7 mV. Treadmill training reduced the rheobase by 4.8 ± 1.5 pA and Vth by 3.1 ± 1.2 mV (P < 0.05). Furthermore, the training effects were dependent on the distribution and types of the interneurons. Treadmill training hyperpolarized Vth and decreased rheobase in ventromedial interneurons, while the significant change was observed only in the action potation height of the interneurons in dorsal horn. Treadmill training also hyperpolarized Vth and increased input resistance in type 3 interneurons, but none of these changes was shown in type 1 and 2 interneurons. Bath application of 5-HT (10−20 μM) increased the neuronal excitability in both control and trained mice. Serotonin had similar effect on membrane properties of the interneurons collected from both groups. This study suggested that treadmill training increased excitability of spinal interneurons of the mice and thus would make the spinal motor system easier to generate locomotion. The output from a motor nucleus is determined by the synaptic input to the motor neurons and their intrinsic properties. Here, we explore whether the source of synaptic inputs to the motor neurons (cats) and the age or post-stroke conditions (humans) may change the recruitment gain of the motor neuron pool. In cats, the size of Ia EPSPs in triceps surae motor neurons (input) and monosynaptic reflexes (MSRs; output) was recorded in the soleus and medial gastrocnemius motor nerves following graded stimulation of dorsal roots. The MSR was plotted against the EPSP thereby obtaining a measure of the recruitment gain. Conditioning stimulation of sural and peroneal cutaneous afferents caused significant increase in the recruitment gain of the medial gastrocnemius, but not the soleus motor neuron pool. In humans, the discharge probability of individual soleus motor units (input) and soleus H-reflexes (output) was performed. With graded stimulation of the tibial nerve, the gain of the motor neuron pool was assessed as the slope of the relation between probability of firing and the reflex size. The gain in young subjects was higher than in elderly subjects. The gain in post-stroke survivors was higher than in age-matched neurologically intact subjects. These findings provide experimental evidence that recruitment gain of a motor neuron pool contributes to the regulation of movement at the final output stage from the spinal cord and should be considered when interpreting changes in reflex excitability in relation to movement or injuries of the nervous system. We used transcranial magnetic stimulation (TMS) of motor cortex, including a novel four-pulse superconditioning (TMSsc) paradigm, in repeated examinations of motor-evoked potentials (MEPs) in eight subjects with motor neuron disease (MND), including seven with amyotrophic lateral sclerosis (ALS). The goals were: (1) to look for evidence of cortical hyperexcitability, including a reduction in short-interval intracortical inhibition (SICI); and (2) to examine the utility of using TMSsc for quantifying upper motor neuron function during MND progression. Testing of abductor pollicis brevis (APB) and tibialis anterior (TA) muscles bilaterally was carried out every 3 months in MND subjects for up to 2 years; results were compared to those from a cohort of 15 control subjects. Measures of SICI were not significantly different between control and MND subjects for either APB or TA muscles. Other measures of cortical excitability, including TMS threshold and MEP amplitude, were consistent with lowered cortical excitability in MND subjects. Certain combinations of superconditioning TMS were capable of causing stronger inhibition or facilitation of MEPs compared to dual-pulse TMS, for both APB and TA target muscles. Moreover, there were multiple cases in which target muscles unresponsive to strong single-pulse TMS, whether at rest or when tested with an active contraction, showed an MEP in response to TMSsc optimized for facilitation. Our findings suggest that a multi-faceted neurophysiologic protocol for examining upper motor neuron function in MND subjects might benefit from inclusion of TMSsc testing. [ABSTRACT FROM AUTHOR]

Effects of low-threshold afferents from the flexor digitorum superficialis (FDS) to the flexor carpi radialis (FCR), flexor carpi ulnaris (FCU) and extensor carpi ulnaris (ECU) motoneurons were examined using a post-stimulus time-histogram (PSTH) and electromyogram-averaging (EMG-A) methods in seven healthy human subjects. Electrical conditioning stimulation to the median nerve branch innervating FDS with the intensity immediately below the motor threshold was delivered. In the PSTH study, the stimulation produced a trough (inhibition) in 19/44 (43%) of FCR and 17/41 (41%) of FCU motor units. Remaining motor units received no facilitatory and inhibitory effects. The central latency of the inhibition was 1.1 ± 0.6 ms (mean ± SD) and 0.6 ± 0.4 ms longer than that of the homonymous monosynaptic Ia facilitation of FCR and FCU, respectively. In the EMG-A study, the stimulation produced a trough (inhibition) in EMG-A of FCR and FCU in all the seven subjects. Amount of the inhibition was 14.5 ± 3.8% (FCR) and 17.9 ± 2.5% (FCU). Since the inhibition diminished after withdrawal of tonic vibration stimuli to the FDS muscle belly, group Ia afferents should be responsible for the inhibition. The stimulation did not produce facilitatory or inhibitory effect on ECU motoneurons in both the PSTH and EMG-A studies. These findings suggest that group Ia afferents from FDS inhibit excitability of motoneurons supplying FCR and FCU through an oligo (di- or tri-) synaptic path in the spinal cord. The effect of body posture on the human soleus H-reflex via electrical stimulation of the tibial nerve at the popliteal fossa was studied. All parameters that may influence the reflex were controlled stringently. H-reflexes were elicited in three different body postures while keeping the level of background muscle activation to a minimum. The H-reflex curve relative to the M-wave curve did not change significantly in any of the body postures. However, the maximal H-reflex amplitude significantly increased in the prone position compared with the sitting (p = 0.02) and standing positions (p = 0.01). The background level of electrical activity of the soleus muscle did not significantly change during varying body postures. Together, these findings indicate that the effectiveness of the spindle primary afferent synapse on the soleus motor neuron pool changes significantly in prone position as compared to sitting and standing positions. Given that we have controlled the confounding factors excluding the head position relative to the gravity and the receptors that may be differentially activated at varying body postures such as the proprioceptors, it is concluded that the tonic activity from these receptors may presynaptically interfere with the effectiveness of the spindle primary afferent synapses on the soleus motor neurons. Direct and indirect corticospinal pathways to finger muscles may play a different role in control of the upper extremity. We used transcranial magnetic stimulation (TMS) and coherence analysis to characterize the corticospinal drive to the first dorsal interosseous (FDI) and abductor pollicis brevis (APB) when active during a precision and power grip task. In experiment 1, single motor units were recorded during precision grip and power grip in 20 adults (25.2 ± 7.1 years). Post-stimulus time histograms (PSTH) were obtained following TMS. In experiment 2, coherence and cross-correlation analysis of the FDI and APB surface EMG were used to investigate the temporal organization of corticospinal drive during precision grip and power grip in 15 adults (27.4 ± 8.1 years). We found no significant differences in PSTH peak onset (26.6 ± 1.9 vs. 26.7 ± 2.0 ms, p = 0.75), maximal peak (27.4 ± 1.9 vs. 27.4 ± 1.9 ms, p = 1.0) or peak duration (2.3 ± 1.1 vs. 2.3 ± 1.0 ms, p = 0.75) for the 11 recovered motor units during precision grip and power grip. Also, no significant difference in coherence or the width of the synchronization peaks during precision grip (7.2 ± 3.7 ms) and power grip (7.9 ± 3.1 ms) could be observed (p = 0.59). The short duration of peaks elicited in the PSTH of single motor units following TMS and central synchronization peaks of voluntarily activated motor units during precision and power grip suggests that the direct corticospinal pathway (the corticomotoneuronal system) is equally involved in the control of both tasks. This study investigated the impact of attention on the sensory and motor actions during postural recovery from underfoot perturbations in young and older adults. A dual-task paradigm was used involving disjunctive and choice reaction time (RT) tasks to auditory and visual stimuli at different delays from the onset of two types of platform perturbations (rotations and translations). The RTs were increased prior to the perturbation (preparation phase) and during the immediate recovery response (response initiation) in young and older adults, but this interference dissipated rapidly after the perturbation response was initiated (<220 ms). The sensory modality of the RT task impacted the results with interference being greater for the auditory task compared to the visual task. As motor complexity of the RT task increased (disjunctive versus choice) there was greater interference from the perturbation. Finally, increasing the complexity of the postural perturbation by mixing the rotational and translational perturbations together increased interference for the auditory RT tasks, but did not affect the visual RT responses. Vision plays an important role in postural control, and visual perception of the gravity-defined vertical helps maintaining upright stance. In addition, the influence of the gravity field on objects' motion is known to provide a reference for motor and non-motor behavior. However, the role of dynamic visual cues related to gravity in the control of postural balance has been little investigated. In order to understand whether visual cues about gravitational acceleration are relevant for postural control, we assessed the relation between postural sway and visual motion congruent or incongruent with gravity acceleration. Postural sway of 44 healthy volunteers was recorded by means of force platforms while they watched virtual targets moving in different directions and with different accelerations. Small but significant differences emerged in sway parameters with respect to the characteristics of target motion. Namely, for vertically accelerated targets, gravitational motion (GM) was associated with smaller oscillations of the center of pressure than anti-GM. The present findings support the hypothesis that not only static, but also dynamic visual cues about direction and magnitude of the gravitational field are relevant for balance control during upright stance. It is known that the median preoptic nucleus (POMe) sends dense projections to the subfornical organ (SFO). However, the functional significance of these projections have not been well discussed. In this electron microscopic study, we investigated the presence of synapses between POMe-derived axon terminals and SFO neurons that project to the paraventricular hypothalamic nucleus (PVN). After injection of a retrograde tracer, wheat germ agglutinin-conjugated horseradish peroxidase-colloidal gold complex, into the PVN, many labeled neurons were found in the SFO. In contrast, after injection of an anterograde tracer, biotinylated dextran amine, in the POMe, abundant labeled axon varicosities were observed in the SFO. Using electron microscopy, synapses were identified between retrogradely labeled dendrites and cell bodies, and anterogradely labeled axon terminals, indicating that POMe neurons innervate SFO neurons projecting to the PVN. The possibility that POMe neurons play multiple roles in the neuronal circuit responsible for vasopressin release and/or cardiovascular regulation is also discussed. [ABSTRACT FROM AUTHOR]

Transcranial magnetic stimulation (TMS) has been first described 30 years ago, and since then has gained enormous attention by neurologists, psychiatrists, neurosurgeons, clinical neurophysiologists, psychologists, and neuroscientist alike. In the early days, it was primarily used to test integrity of the corticospinal tract. Beyond further developments of TMS in diagnostics, mapping and monitoring of the motor system, major other applications expanded into using TMS as research tool in the cognitive neurosciences, and as therapeutic tool in neurological and psychiatric disease by virtue of inducing long-term change in excitability and connectivity of the stimulated brain networks. This mini-review will highlight these developments by reviewing the 10 most frequently cited TMS publications. Despite the tremendous popularity and success of TMS as a non-invasive technique to stimulate the human brain, several aims remain unresolved. This review will end with highlighting those 10 most frequently cited papers that have been published in 2014-2016 to indicate the currently hottest topics in TMS research and major avenues of development. Speeded responses to an external cue are slower when the cue interrupts preparation to perform the same or a similar action in a self-paced manner. To explore the mechanism underlying this 'cost of intention', we examined whether the size of the cost is influenced by the nature of the external cue. Specifically, we assessed whether the cost of intention is different for movements made in response to an imitative cue (an on-screen hand movement) compared to those made in response to a symbolic cue. Consistent with previous reports, externally cued responses were significantly slower on trials where participants were preparing to perform an internally driven movement later in the trial. Also as predicted, simple response times to the imitative cue were faster than those made to the symbolic cue. Critically, the cost of intention was similar for each cue type, suggesting that preparing an intentional action influenced responses cued by the symbolic and imitative cues to a similar degree. These findings suggest that the nature of the external cue does not influence the response time delay associated with concurrent intentional preparation. Midbrain superior colliculus (SC) contains a variety of neuronal types, influencing a rich spectrum of functions beyond gaze orienting. Here, we report on a novel class of SC neurons in the rhesus monkey ( Macaca mulatta) that are activated by an unexpected perturbation in a goal-directed arm-movement task. One monkey subject reached for and pressed an illuminated target on a working panel upon a visual go-signal, while maintaining visual fixation elsewhere. On 50 % of trials, a task perturbation occurred-the working panel abruptly and unexpectedly moved against the subject's hand after he pressed the target. During the performance, we recorded single SC neurons and found neurons activated exclusively for the task perturbation. These perturbation neurons were localized in the deep lateral zone of the SC, were silent during non-perturbed trials, did not appear to respond to task-irrelevant stimuli, and they had intriguingly long neuronal latencies. If the perturbation neurons' activity relates to the hand-target contact, it may reflect the saliency of an unexpected sensation, i.e. a sensation that is not self-induced and thus cannot be predicted on a basis of the monkey's motor program. Based on H-reflex data, spinal mechanisms are proposed to be responsible for the first 50-80 ms of the transcranial magnetic stimulation (TMS)-induced silent period. As several methodological issues can compromise H-reflex validity as a measure of motoneuron excitability, this study used transmastoid stimulation to elicit cervicomedullary motor evoked potentials (CMEPs) during the silent period. Eleven subjects made 1-3 visits which involved 32 or 44 brief (~3 s) isometric elbow flexor contractions at 25 % of maximal torque. During each contraction, transmastoid stimulation was delivered in isolation to elicit an unconditioned CMEP and at interstimulus intervals (ISIs) ranging from 50 to 150 ms after TMS to elicit a conditioned CMEP. Stimulus intensities for TMS and transmastoid stimulation were set to elicit a silent period of ~200 ms and an unconditioned CMEP of 15, 50, or 85 % of the maximal compound muscle action potential ( M ), respectively. At all ISIs and intensities of transmastoid stimulation, the conditioned CMEP was significantly smaller than the unconditioned CMEP ( p < 0.001). However, suppression of the conditioned CMEP was significantly less at 85 % compared to 15 or 50 % M ( p = 0.001). Contrary to published H-reflex data, the conditioned CMEP did not recover within 50-80 ms, remaining significantly suppressed at the longest ISI tested (150 ms). These data suggest the spinal portion of the TMS-evoked silent period is considerably longer than reported previously. Transmastoid stimulation, unlike peripheral nerve stimulation, does not impact proprioceptive inflow to motoneurons. Hence, relative to the H-reflex, the CMEP will be subjected to greater afferent-mediated disfacilitation and inhibition due to the TMS-induced muscle twitch. [ABSTRACT FROM AUTHOR]

Although there still is conflicting evidence whether schizophrenia is a neurodegenerative disease, cognitive changes in schizophrenia resemble those observed during normal aging. In contrast to extensively demonstrated deficits in explicit learning, it remains unclear whether implicit sequence learning is impaired in schizophrenia and normal aging. Implicit sequence learning was investigated using a computerized drawing task, the 'implicit pattern learning task (IPLT)' in 30 stable patients with schizophrenia, 30 age-matched controls and 30 elderly subjects on two consecutive days and after 1 week (sessions 1, 2 and 3). Fixed sequence trials were intermixed with random trials, and sequence learning was assessed by subtraction of the response time in fixed sequence trials from random trials. Separate analyses of response times and movement accuracy (i.e., directional errors) were performed. Explicit sequence knowledge was assessed using three different awareness tasks. All groups learned equally during sessions 1 and 2. In session 3, control subjects showed significantly larger learning scores than patients with schizophrenia ( p = .012) and elderly subjects ( p = .021). This group difference is mainly expressed in movement time and directional errors. Patients with schizophrenia demonstrated less subjective sequence awareness, and both patients with schizophrenia and elderly subjects had less explicit sequence recall. Explicit recall was positively correlated with task performance in all groups. After a short 24 h interval, all subjects showed similar improvements in implicit sequence learning. However, no benefit of prior task exposure 1 week later was observed in patients with schizophrenia and elderly subjects compared to controls. As patients with schizophrenia and elderly both display less explicit sequence recall, the control group superiority after 1 week could be explained by an explicit learning component. The few patients with schizophrenia and elderly subjects who had some sequence recall could possibly utilize this explicit knowledge to improve their task performance but did this by distinct mechanisms. Motor responses to unexpected external perturbations require the adjustment of the motor commands driving the ongoing activity. Strategies can be learned with practice to compensate for these unpredictable perturbations (e.g., externally induced slips and trips). It has been hypothesized that response improvements reflect the adaptation of motor commands through updates of an internal model. This hypothesis may be nuanced when a pre-existing motor response could be used. In that case, since a relatively adequate response is known, only the timing of the command needs to be determined. If so, then it could be inferred that the timing of movement initiation and the specific sequence of motor commands can be dissociated. Previously, we quantified the benefits of cuing vs. learning on recovery motor responses resulting from a trip induced by the abrupt stop of one side of a split belt treadmill. Trip occurrence was randomized within a series of strides. Two groups of young adults participated to two distinct experiments (learning, cuing). In the learning experiment, trip recovery improved progressively from the 4th to the 8th trial to reach an 'adapted response'. In the cuing experiment, trip recovery was immediate (from 1st trial). Expanding from these results, the aim of the present work was to differentiate the processes underlying the generation of motor compensation strategies in response to an external perturbation under time uncertainty. A supplementary analysis revealed that 'cued' responses were kinematically similar to the 'adapted response' and remained invariant regardless of cue lead time (250, 500 ms before trip) and application location of the cue (arm, trunk, lower leg). It is posited that all responses (cued and non-cued) are the expression of a pre-existing motor program derived from life experiences. Here, the cue significantly reduces time uncertainty and adaptation consists primarily in resolving time uncertainty based on the trial-by-trial learning of the stochastic property of trip occurrence in order to reduce the response delay. To lift an object, the motor system must predict the weight of the object and use this information to program appropriate lifting forces. If this prediction is erroneous, people may assign blame for the error to either themselves or an external source-a process called credit assignment. In the present study, we explored the role of credit assignment on weight predictions during a lifting task. Participants were told that the EMG surface electrodes attached to their lifting hand were either part of a 'passive' system that recorded muscular activity, or part of an 'active' system that would apply energy to the muscle, influencing weight perception. Participants performed 90 lifts of the training blocks, followed by 10 lifts of a newly encountered larger test block. In between training and test trials, the experimenter turned off the recording system and removed the surface electrodes for participants in the 'active' group. For each lift, we determined the initial peak rate of change of vertical load force rate and load-phase duration, estimates of predicted object weight. Analysis of the first 10 training lifts and the last 10 training lifts revealed no effect of Active versus Passive EMG on weight predictions. However, after removing the EMG equipment, participants in the 'active' group failed to scale their predictive load forces in the same manner as those in the 'passive' condition when lifting a novel block. A specific feature of motor unit (MU) firing behaviour is rhythmic trains of single discharges at low rate resulting from the prolonged motoneuronal afterhyperpolarization. However, some MUs exhibit occasional doublets with uniquely short interspike intervals (2.5-20.0 ms). Motoneuronal delayed depolarization is commonly accepted to be doublet underlying mechanism. Apart from doublets, much scarcer MU triple discharges were described, but their mechanisms are disputable. The aim of the present study was to analyse MU triplet firing origin in healthy humans. MU triple discharges occasionally arising during gentle voluntary muscle contractions were compared with those arising in axons during motor nerve stimulation. Firing pattern was analysed in 109 MUs of four muscles: the tibialis anterior, the flexor carpi ulnaris, the abductor pollicis brevis, and the abductor digiti minimi. Our findings present evidence that during voluntary contractions two kinds of MU triplet firing can be occasionally observed: 'true' motoneuronal triplets (interspike intervals of 3.6-17.3 ms) with the delayed depolarization as the possible underlying mechanism and axonal triple discharges including the M-response and F-wave. The findings can be useful not only for understanding mechanisms of the very rare motoneuronal firing in healthy humans but also for estimation of pathological triplet firing origin. The quadriceps femoris (QF) muscle group plays an essential role in human movement, such as standing, walking and running. The ability to maintain a steady force during physical activity of the human lower limb is important for mobility, postural control and balance. Although prolonged mechanical vibration of the muscle-tendon unit can moderate the efficacy of synaptic input from Ia afferent onto the α-motor neuron pathway, the effect of prolonged tendon vibration on fluctuations of knee extensor force has received little attention. The purpose of the present study was to examine the effects of prolonged patellar tendon vibration on the force steadiness of the QF muscle. Nine healthy men performed a submaximal force-matching task involving isometric knee extension before and after patellar tendon vibration or quiet seated rest ( n = 7, control condition) for 30 min. The target force was 2.5, 10 and 30 % of maximal voluntary contraction (MVC). Surface electromyography (EMG) of the four QF synergists was recorded and normalized to EMG amplitude during the MVC. The knee extension force and the EMG amplitude of vastus medialis during the MVC were significantly reduced after the vibration, but did not significantly decrease in the control condition. Fluctuations of force and normalized EMG of individual QF muscles at each submaximal force level did not significantly change after the vibration. Handedness is a feature of human motor control that is still not fully understood. Recent work has demonstrated that the dominant and nondominant arm each excel at different behaviors and has proposed that this behavioral asymmetry arises from lateralization in the cerebral cortex: the dominant side specializes in predictive trajectory control, while the nondominant side is specialized for impedance control. Long-latency stretch reflexes are an automatic mechanism for regulating posture and have been shown to contribute to limb impedance. To determine whether long-latency reflexes also contribute to asymmetric motor behavior in the upper limbs, we investigated the effect of arm dominance on stretch reflexes during a postural task that required varying degrees of impedance control. Our results demonstrated slightly but significantly larger reflex responses in the biarticular muscles of the nondominant arm, as would be consistent with increased impedance control. These differences were attributed solely to higher levels of voluntary background activity in the nondominant biarticular muscles, indicating that feedforward strategies for postural stability may differ between arms. Reflex sensitivity, which was defined as the magnitude of the reflex response for matched levels of background activity, was not significantly different between arms for a broad subject population ranging from 23 to 51 years of age. These results indicate that inter-arm differences in feedforward strategies are more influential during posture than differences in feedback sensitivity, in a broad subject population. Interestingly, restricting our analysis to subjects under 40 years of age revealed a small increase in long-latency reflex sensitivity in the nondominant arm relative to the dominant arm. Though our subject numbers were small for this secondary analysis, it suggests that further studies may be required to assess the influence of reflex lateralization throughout development. In two experiments using a center-out task, we investigated kinesthetic-motor and auditory-motor integrations in 5- to 12-year-old children and young adults. In experiment 1, participants moved a pen on a digitizing tablet from a starting position to one of three targets (visuo-motor condition), and then to one of four targets without visual feedback of the movement. In both conditions, we found that with increasing age, the children moved faster and straighter, and became less variable in their feedforward control. Higher control demands for movements toward the contralateral side were reflected in longer movement times and decreased spatial accuracy across all age groups. When feedforward control relies predominantly on kinesthesia, 7- to 10-year-old children were more variable, indicating difficulties in switching between feedforward and feedback control efficiently during that age. An inverse age progression was found for directional endpoint error; larger errors increasing with age likely reflect stronger functional lateralization for the dominant hand. In experiment 2, the same visuo-motor condition was followed by an auditory-motor condition in which participants had to move to acoustic targets (either white band or one-third octave noise). Since in the latter directional cues come exclusively from transcallosally mediated interaural time differences, we hypothesized that auditory-motor representations would show age effects. The results did not show a clear age effect, suggesting that corpus callosum functionality is sufficient in children to allow them to form accurate auditory-motor maps already at a young age. Many activities of daily living involve multi-digit, voluntary rotational manipulations of grasped objects. Yet, only a few studies have focused on coordination of individual fingertip forces during such tasks. The objective of this study was to investigate individual digit contributions to a three-digit task in which an object was rotationally manipulated against gravity. Center of mass was varied through the use of containers shaped like a water bottle, pint glass, and cocktail glass, from which subjects poured fluid carefully into a nearby receptacle. The center of mass of the grasped object changed continuously as fluid was poured out. Self-selected digit placement and contributions of fingertip forces to rotational manipulation were dependent upon anticipated center of mass location associated with container shape. The thumb resisted the rotation of the top-heavy, cocktail glass container until 79 % of the pouring phase had elapsed, but actively assisted the rotation of the less challenging containers. More directly opposing the thumb, the index finger contributed more to grasp stability. The middle finger contributed more to rotation of the container for pouring. It was found that the thumb, index, and middle fingers acted in unison temporally, but contributed independently to the grip forces and stabilizing moments throughout the dynamic, rotational manipulation task. The goal of this study is to assess how reflexes and intrinsic properties contribute to low-back stabilization and modulate with conditions. Upper body sway was evoked by anterior-posterior platform translations, while subjects were seated with a restrained pelvis and free upper body. Kinematic analysis of trunk translations and rotations illustrated that a fixed rotation point between the vertebrae L4 and L5 adequately captures lumbar bending up to 5 Hz. To investigate the motor control modulation, the conditions varied in vision (eyes open or closed), task instruction ( Balance naturally or Resist perturbations by minimizing low-back motions), and perturbation bandwidth (from 0.2 up to 1, 3 or 10 Hz). Frequency response functions and physiological modeling parameters showed substantial modulation between all conditions. The eyes-open condition led to trunk-in-space behavior with additional long-latency visual feedback and decreased proprioceptive feedback. The task instruction to resist led to trunk-on-pelvis stabilization behavior, which was achieved by higher co-contraction levels and increased reflexive velocity feedback. Perturbations below the low-back natural frequency (~1 Hz) led to trunk-on-pelvis stabilization behavior, mainly attributed to increased intrinsic damping. This indicates that bandwidth effects should not be ignored and that experiments with high-bandwidth perturbations do not fully represent the intrinsic and reflexive behavior during most (low-bandwidth) daily life activities. The neck stabilized the head orientation effectively (head rotation amplitudes 2 % of trunk), but did not effectively stabilize the head in space (global head translations exceeded trunk translations by 20 %). Motor imagery relies strongly on motor representations. Currently, it is widely accepted that both the imagery and execution of actions share the same neural representations (Jeannerod, Neuroimage 14:S103-S109, ). Comparing mental with actual movement durations opens a window through which to examine motor representations and how they relate to cognitive motor processes. The present experiment examined mental durations reported by participants standing upright who imagined walking either with or without an additional load while actually carrying or not carrying that same load. Results showed a robust effect of longer durations when imagining the additional load during mental walking, whereas physical walking with an additional load did not extend movement durations accordingly. However, experiencing an actual load during imagery did not influence mental durations substantially. This dissociation of load-related effects can be interpreted as being due to an interaction of motor processes and their cognitive representation along with a reduction in neural activity in vestibular and somatosensory areas during imagery of locomotion. Studies on bimanual control inevitably deal with questions about interactions between the two effectors, mostly under conditions of sensory feedback of the same modality-usually vision-for both hands. This study used a novel paradigm in which one hand performed target-directed movements under visual control, while the other hand operated under predominantly kinesthetic control, without visual feedback. By introducing an abrupt visual feedback perturbation in the 'visible' hand, resulting in an update of its visuo-motor map, the robustness of the kinesthetic-motor map of the 'invisible' hand against interference from the visually controlled hand could be tested. Results show that the visuo-motor adaptation resulted in asymmetric directional interference: when the 'visible' right hand adapted to the perturbation, it interfered substantially more with the 'invisible' left hand (group 1) than when the left hand was under visual control and the right hand under kinesthetic control (group 2). The results support recent theories of functional lateralization postulating dominance of the right hand for trajectory control and demonstrate that on the level of arm kinematics, this interference crosses modality boundaries. Interestingly, while in most participants interference manifested itself in isodirectional deviations of the kinesthetically guided hand, the deviations in a small subsample of participants mirrored those of the visually guided hand. The literature reports that anticipatory postural adjustments (APAs) are programmed according to movement velocity. However, the linkage between APAs and velocity has been highlighted within single subjects who were asked to voluntarily change movement velocity; therefore, till now, it has been impossible to discern whether the key factor determining APA latency was the intended movement velocity or the actual one. Aim of this study was to distinguish between these two factors. We analyzed the APA chain that stabilizes the arm during a brisk index finger flexion in two groups of subjects: (1) 29 who composed our database from previous experiments and were asked to 'go-as-fast-as-possible' ( go- fast), but actually performed the movement with different speeds (238-1,180°/s), and (2) ten new subjects who performed the go- fast movement at more than 500°/s and were then asked to go- slow at about 50 % of their initial velocity, thus moving at 300-800°/s. No correlation between APA latency and actual movement speed was observed when all subjects had to go- fast ( p > 0.50), while delayed APAs were found in the ten new subjects when they had to go- slow ( p < 0.001). Moreover, in the speed range between 300 and 800°/s, the APA latency depended only on movement instruction: subjects going fast showed earlier APAs than those going slow ( p < 0.001). These data suggest a stronger role of the intended movement velocity versus the actual one in modifying the timing of postural muscles recruitment with respect to the prime mover. The present experiment examined the influence of final target position on grasp posture planning during a three-segment object manipulation task in which the required object orientation at the first target position was unconstrained. Participants grasped a cylindrical object from a home position, placed it at an intermediate position in a freely chosen orientation, and subsequently placed it at one of four final target positions. Considerable inter-individual differences in initial grasp selection were observed which also led to differences in final grasp postures. Whereas some participants strongly adjusted their initial grasp postures to the final target orientation, and thus showed a preference for end-state comfort, other participants showed virtually no adjustment in initial grasp postures, hence satisfying initial-state comfort. Interestingly, as intermediate grasp postures were similar regardless of initial grasp adjustment, intermediate-state comfort was prioritized by all participants. These results provide further evidence for the interaction of multiple action selection constraints in grasp posture planning during multi-segment object manipulation tasks. Whereas some constraints may take strict precedence in a given task, other constraints may be more flexible and weighted differently among participants. We explored stability of multi-finger cyclical accurate force production action by analysis of responses to small perturbations applied to one of the fingers and inter-cycle analysis of variance. Healthy subjects performed two versions of the cyclical task, with and without an explicit target. The 'inverse piano' apparatus was used to lift/lower a finger by 1 cm over 0.5 s; the subjects were always instructed to perform the task as accurate as they could at all times. Deviations in the spaces of finger forces and modes (hypothetical commands to individual fingers) were quantified in directions that did not change total force (motor equivalent) and in directions that changed the total force (non-motor equivalent). Motor equivalent deviations started immediately with the perturbation and increased progressively with time. After a sequence of lifting-lowering perturbations leading to the initial conditions, motor equivalent deviations were dominating. These phenomena were less pronounced for analysis performed with respect to the total moment of force with respect to an axis parallel to the forearm/hand. Analysis of inter-cycle variance showed consistently higher variance in a subspace that did not change the total force as compared to the variance that affected total force. We interpret the results as reflections of task-specific stability of the redundant multi-finger system. Large motor equivalent deviations suggest that reactions of the neuromotor system to a perturbation involve large changes in neural commands that do not affect salient performance variables, even during actions with the purpose to correct those salient variables. Early frontal somatosensory evoked potentials (SEPs) (i.e., N30) are known to be modulated by movement. Furthermore, individuals with prefrontal lesions have enhanced early frontal SEPs. However, it is currently unclear through what mechanism the prefrontal cortex may modulate early frontal SEPs. The current study investigated whether prefrontal modulatory effects on frontal SEPs may depend on the relevancy of somatosensory input for movement (i.e., interaction with motor areas). Two experiments were conducted to determine whether selective spatial attention alone ( Experiment 1- Attend and Mentally Count) or when using attended somatosensory input in the preparation of finger sequences with the limb contralateral to somatosensory stimulation ( Experiment 2- Attend for Movement Preparation) could modulate SEPs. In Experiment 1, SEPs elicited by median nerve (MN) stimulation at both wrists were measured in trials when individuals attended and mentally counted vibrotactile (VibT) input at either index finger. In Experiment 2, SEPs elicited by MN stimulation at the left wrist were measured in trials when individuals used attended VibT input at the left index finger to prepare finger sequences that were contralateral to MN stimulation. In both experiments, control conditions were performed where participants received passive VibT and MN stimulation. Results from Experiment 1 confirmed that selective spatial attention alone does not modulate frontal N30 peak amplitudes. However, Experiment 2 revealed that frontal N30 peak amplitudes were decreased (i.e., gated) when individuals used attended VibT input at the left index finger to prepare contralateral finger sequences. These results support a role of sensory gating of early frontal SEPs during finger sequence preparation of the limb contralateral to MN stimulation that may result from increased activity in prefrontal, motor preparatory areas, and basal ganglia. A decision to select an action from alternatives is often guided by rules that flexibly map sensory inputs to motor outputs when certain conditions are satisfied. However, the neural mechanisms underlying rule-based decision making remain poorly understood. Two complementary types of neurons in the supplementary eye field (SEF) of macaques have been identified that modulate activity differentially to interpret rules in an ocular go-nogo task, which stipulates that the animal either visually pursue a moving object if it intersects a visible zone ('go'), or maintain fixation if it does not ('nogo'). These neurons discriminate between go and nogo rule-states by increasing activity to signal their preferred (agonist) rule-state and decreasing activity to signal their non-preferred (antagonist) rule-state. In the current study, we found that SEF neurons decrease activity in anticipation of the antagonist rule-state, and do so more rapidly when the rule-state is easier to predict. This rapid decrease in activity could underlie a process of elimination in which trajectories that do not invoke the preferred rule-state receive no further computational resources. Furthermore, discrimination between difficult and easy trials in the antagonist rule-state occurs prior to when discrimination within the agonist rule-state occurs. A winner-take-all like model that incorporates a pair of mutually inhibited integrators to accumulate evidence in favor of either the decision to pursue or the decision to continue fixation accounts for the observed neural phenomena. Hemispheric brain injury resulting from a stroke is often accompanied by muscle weakness in contralateral limbs. In neurologically intact subjects, appropriate motoneuronal recruitment and rate modulation are utilized to optimize muscle force production. In the present study, we sought to determine whether weakness in an affected hand muscle in stroke survivors is partially attributable to alterations in the control of muscle activation. Specifically, our goal was to characterize whether the surface EMG amplitude was systematically larger as a function of (low) force in paretic hand muscles as compared to contralateral muscles in the same subject. We tested a multifunctional muscle, the first dorsal interosseous (FDI), in multiple directions about the second metacarpophalangeal joint in ten hemiparetic and six neurologically intact subjects. In six of the ten stroke subjects, the EMG-force slope was significantly greater on the affected side as compared to the contralateral side, as well as compared to neurologically intact subjects. An unexpected set of results was a nonlinear relation between recorded EMG and generated force commonly observed in the paretic FDI, even at very low-force levels. We discuss possible experimental as well as physiological factors that may contribute to an increased EMG-force slope, concluding that changes in motor unit (MU) control are the most likely reasons for the observed changes. When a cognitive and a motor task like walking or keeping one's balance are performed concurrently, performance usually deteriorates. Older adults have often been shown to prioritize their motor performance in such dual-task situations, possibly to protect themselves from falls. The current study investigates whether these prioritization behaviors can still be observed when several challenges are combined. Younger (20-30 years old) and older adults (60-70 years old; n = 24 in each group) were asked to walk through virtual environments with and without a cognitive load (3-back task). Walking difficulty was increased by walking on an elevated surface or on a narrow as opposed to a broad track, or both. Walking instructions emphasized speed and accuracy (avoiding missteps). No instruction was given concerning which performance dimension should be prioritized during dual-task trials. Participants decreased their 3-back performance while walking. Younger adults maintained their walking speed on elevated surfaces and were able to keep the number of missteps low, even when walking on a narrow track while performing the cognitive task. Older adults increased their walking speed on elevated relative to even surfaces and committed more missteps under cognitive load. The objective of this study was to assess changes in monosynaptic motoneuron responses to stimulation of Ia afferents after locomotor training in individuals with chronic spinal cord injury (SCI). We hypothesized that locomotor training modifies the amplitude of the soleus monosynaptic motoneuron responses in a body position-dependent manner. Fifteen individuals with chronic clinical motor complete or incomplete SCI received an average of 45 locomotor training sessions. The soleus H-reflex and M-wave recruitment curves were assembled using data collected in both the right and left legs, with subjects seated and standing, before and after training. The soleus H-reflexes and M-waves, measured as peak-to-peak amplitudes, were normalized to the maximal M-wave ( M). Stimulation intensities were normalized to 50 % M stimulus intensity. A sigmoid function was also fitted to the normalized soleus H-reflexes on the ascending limb of the recruitment curve. After training, soleus H-reflex excitability was increased in both legs in AIS C subjects, and remained unchanged in AIS A-B and AIS D subjects during standing. When subjects were seated, soleus H-reflex excitability was decreased after training in many AIS C and D subjects. Changes in reflex excitability coincided with changes in stimulation intensities at H-threshold, 50 % maximal H-reflex, and at maximal H-reflex, while an interaction between leg side and AIS scale for the H-reflex slope was also found. Adaptations of the intrinsic properties of soleus motoneurons and Ia afferents, the excitability profile of the soleus motoneuron pool, oligosynaptic inputs, and corticospinal inputs may all contribute to these changes. The findings of this study demonstrate that locomotor training impacts the amplitude of the monosynaptic motoneuron responses based on the demands of the motor task in people with chronic SCI. We investigated the effect of repetitive transcranial magnetic stimulation (rTMS) applied either during one session of stimulation, or by ten sessions of low-frequency stimulation over the left parietal cortex, on hemispatial neglect in stroke patients. We enrolled 34 subjects that had experienced a stroke. All subjects received 1,200 real rTMS over the left parietal cortex at an intensity of 90 % of motor thresholds with 1 Hz. Subjects were divided into two groups. One group of subjects ( n = 19) received real rTMS over the left parietal cortex in a single session of stimulation, and the other group ( n = 15), underwent a total of ten sessions of daily stimulations for 2 weeks. Letter cancelation test, line bisection test, and Ota's task were administered to compare the effects of different rTMS protocols, before and after rTMS. The results showed no difference in baseline value between the single session group and the ten sessions group. Total ten sessions of low-frequency rTMS over the left parietal cortex, compared with the single session of rTMS, significantly improved hemispatial neglect in letter cancelation, line bisection, and Ota's task ( P < 0.01). Finally, a total of ten sessions of low-frequency rTMS can be used in treatment by rTMS for patients suffering from hemispatial neglect after stroke. [ABSTRACT FROM AUTHOR]

It has been reported that the 90° arm abduction force counteracting external adduction loads appeared to be smaller under teeth clenching condition than under non-clenching condition. To elucidate the physiological mechanism underlying the possible inhibitory effect of teeth clenching on the arm abduction, we have attempted to quantify the difference in the force induced against the fast and slow ramp load between the arm abductions under teeth non-clenching and clenching conditions. When the load of adduction moment was linearly increased, the abductor force increased to a maximal isometric contraction force (MICF) and further increased to a maximal eccentric contraction force (MECF) with forced adduction. The MICF measured under teeth clenching was significantly lower than that under non-clenching, despite no significant difference in the MECF between the two conditions. The reduction in MICF caused by teeth clenching was enhanced by increasing the velocity of the load. These results suggest that clenching inhibits abduction force only during isometric contraction phase. The invariability of MECF would indicate the lack of involvement of fatigue in such inhibitory effects of clenching. To discover the source of the inhibition, we have examined the effects of teeth clenching on the stretch reflex in the deltoid muscle. The stretch reflex of deltoid muscles was inhibited during clenching, contrary to what was expected from the Jendrassik maneuver. Taken together, our results suggest that the teeth clenching reduced the MICF by depressing the recruitment of deltoid motoneurones presumably via the presynaptic inhibition of spindle afferent inputs onto those motoneurones. Facial nerve injury is a common clinical trauma involving long-term functional deficits with facial asymmetry leading to associated psychological issues and social hardship. We have recently shown that repair by hypoglossal-facial or facial-facial nerve surgical end-to-end anastomosis and suture [hypoglossal-facial anastomosis (HFA) or facial-facial anastomosis (FFA)] results in collateral axonal branching, polyinnervation of neuromuscular junctions (NMJs) and poor function. We have also shown that another HFA repair procedure using an isogenic Y-tube (HFA + Y-tube) and involving a 10-mm gap reduces collateral axonal branching, but fails to reduce polyinnervation. Furthermore, we have previously demonstrated that manual stimulation (MS) of facial muscles after FFA or HFA reduces polyinnervation of NMJs and improves functional recovery. Here, we examined whether HFA + Y-tube and MS of the vibrissal muscles reduce polyinnervation and restore function. Isogenic Y-tubes were created using abdominal aortas. The proximal hypoglossal nerve was inserted into the long arm and sutured to its wall. The distal zygomatic and buccal facial nerve branches were inserted into the two short arms and likewise sutured to their walls. Manual stimulation involved gentle stroking of the vibrissal muscles by hand mimicking normal whisker movement. We evaluated vibrissal motor performance using video-based motion analysis, degree of collateral axonal branching using double retrograde labeling and the quality of NMJ reinnervation in target musculature using immunohistochemistry. MS after HFA + Y-tube reduced neither collateral branching, nor NMJ polyinnervation. Accordingly, it did not improve recovery of function. We conclude that application of MS after hypoglossal-facial nerve repair using an isogenic Y-tube is contraindicated: it does not lead to functional recovery but, rather, worsens it.
