Endophytic filamentous brown algae are known to invade stipes and fronds of kelps with potentially negative effects for the hosts. They have simple filamentous thalli and are difficult to identify based on morphology. We investigated the molecular diversity of 56 endophytes isolated from seven different kelp species from Europe, Chile, Korea and New Zealand by sequencing two unlinked molecular markers (5'COI and ITS1). A majority of 49 of the isolated endophytes (88%) belonged to the genera Laminarionema and Laminariocolax. The endophyte Laminarionema elsbetiae was isolated from Saccharina latissima and S. japonica tissues in Europe and Korea, respectively, and showed highly similar sequences in both regions. Three different species of the genus Laminariocolax were identified, the most common of which was L. aecidioides, an endophyte with a worldwide distribution and a broad host range. The other two species, L. tomentosoides and a species described here as Laminariocolax atlanticus sp. nov., were associated with different kelp species in the northern hemisphere and the North Atlantic, respectively. Our results suggest that specific host-endophyte patterns could exist locally, as found in kelps in Brittany where all endophytes isolated from S. latissima were L. elsbetiae, all endophytes isolated from Laminaria digitata were Laminariocolax tomentosoides, and those isolated from Laminaria hyperborea were Laminariocolax atlanticus and L. aecidioides. The genetic diversity of the green algal genus Ulva sensu lato in the New Zealand region was surveyed, examining rbcL sequences of 581 samples from a wide geographical range. Twenty-four genetically distinct taxa were discovered in New Zealand waters, belonging to three genera-Ulva (19 species), Umbraulva (four species) and Gemina (one species). Of the 19 species of Ulva reported here, 13 could be identified to the species level based on morphological and genetic data. The remaining six species cannot currently be assigned to known species groups due to a lack of close homology with sequences in GenBank. These species may include undescribed endemic taxa, recognised taxa for which rbcL sequences are not yet available, or may represent cryptogenic species. The genus Umbraulva is recorded for the first time for the New Zealand region and for the Southern Hemisphere. Of the four species distinguished, one is considered to be introduced to the region and the other three are undescribed indigenous taxa. Subantarctic samples provide the first evidence of the genus Gemina since its description in 1952: sequence data confirmed that Gemina is distinct from Ulva and Umbraulva. A number of the species identified in this study can be distinguished through a combination of growth form, morphological, ecological and distributional characters. However there remain considerable problems in distinguishing a number of other species by morphological characters alone. Based on information such as distribution in New Zealand (percentage of samples occurring in highly modified environments and/or areas with frequent vessel traffic), as well as the genetic similarity of New Zealand samples to material from overseas, we have concluded that at least five species have been introduced to the New Zealand region: Ulva armoricana, U. californica, U. flexuosa, U. lactuca and Umbraulva olivascens. The phylogenetic affinities of the brown alga Herpodiscus durvillaeae, an obligate parasite of Durvillaea antarctica (Fucales, Phaeophyceae) endemic to New Zealand, were analysed using combined partial nuclear encoded ribosomal DNA (26S) and plastid encoded RuBisCO (rbcL) gene sequences. Results from phylogenetic analyses place this species within the order Sphacelariales. Molecular data were supported by two morphological features characteristic for the Sphacelariales sensu stricto: the presence of apical cells and the transitory blackening of the cell wall with sodium hypochlorite solution ('Eau de Javel'). However, the strongly heteromorphic life history distinguishes H. durvillaeae from all other members of the Sphacelariales of which the life cycle is known. This variability in life history provides a new systematic character for the order and should be taken into account in any comprehensive systematic revision of the Sphacelariales.
