Drought stress is one of the most important environmental factors that limit plant growth and development, thus reducing yield. The objective of the present research was to correlate the genetic structure of different Fragaria genotypes, as assessed by Expressed Sequence Tag (EST) and Amplified Fragment Length Polymorphism (AFLP) markers, and plant responses to drought stress. Firstly, physiological parameters related to the plant response to drought stress such as leaf relative water content (RWC) and water losing rate (WLR) were measured. WLR and RWC were compared for 20 cultivars of the octaploid Fragaria × ananassa, two ecotypes of the diploid species F. vesca and one octaploid species F. chiloensis. These parameters could discriminate genotypes showing a contrasting response to water stress. Secondly, AFLP and ESTs were compared in terms of their information content and efficiency in the study of genetic diversity and relationships among these 23 Fragaria genotypes. To evaluate the genetic basis for the observed variation in the measured physiological parameter, the effect of specific AFLP/EST loci on WLR and RWC for the different Fragaria genotypes was quantified by Kruskal-Wallis analysis. By Mantel testing, the hierarchical clustering of the Fragaria genotypes based on associated EST or AFLP markers was compared to the observed eco-physiological relevant grouping. A better discriminating capacity for associated markers was noted, enabling a functional marker selection approach to screen the strawberry gene pool for drought tolerance. Sunflower is one of the major annual world crops grown for edible oil and its meal is a potential source of protein for human consumption. It contains tocopherol that decreases potential risk of chronic diseases in human. The objectives of the current research are to assess the genetic variability and to identify AFLP markers and candidate genes associated with seed-quality traits under well-irrigated and water-stressed conditions in gamma-induced mutants of sunflower. Two mutant lines, M8-826-2-1 and M8-39-2-1, with significant increased level of oleic acid were identified that can be used in breeding programs for quality increase high oxidative stability and heart-healthy properties. The significant increased level of tocopherol in mutant lines, M8-862-1N1 and M8-641-2-1, is justified by observed polymorphism for tocopherol pathway-related gene; MCT. The most important marker for total tocopherol content is E33M50_16 which explains 33.9% of phenotypic variance. One of the most important candidate genes involving fatty acid biosynthesis, FAD2 ( FAD2- 1), is linked to oleic and linoleic acids content and explained more than 53% of phenotypic variance. Common markers associated with different seed-quality traits in well-irrigated and water-stressed conditions could be used for marker-assisted selection (MAS) in both conditions. Solanum gourlayi and Solanum spegazzinii, wild potatoes endemic to Argentina, possess desirable traits for breeding. In periodical regenerations of accessions, variability was detected for morphology and breeding barriers. The persistence of these populations in nature was evaluated after more than 20 years. Both species were observed in all visited sites, along with other wild and cultivated potatoes. Chromosome numbers coincided with the originally reported, except for one population of Solanum gourlayi, with diploid and tetraploid cytotypes. The accompanying flora and environmental conditions revealed important alterations as the result of road construction, excessive stocking rates and overgrazing. Principal coordinate and cluster analyses and an AMOVA using AFLP data of three original accessions and the corresponding new accessions revealed high molecular variability and extensive overlapping. Plant grouping of accessions occurred at a distance of 0.58 for S. gourlayi, 0.62 for S. spegazzinii and 0.67 for both species. Fruit flavour of traditional tomato ecotypes differs from that of modern varieties, the latter being often blamed for lack of flavour. To respond to consumer complaints breeders should know the range of genetic variability available in tomato resources, including local ecotypes, for improving flavour. For this purpose, multitrait characterization, including biochemical, sensorial and molecular analyses, was performed on 12 Italian tomato ecotypes that belong to different types (San Marzano, Sorrento, and Vesuvio). The ecotypes showed a significant variation both in glucose and citric acid content. As for amino acid determination, lysine, aspartic acid and serine showed significant variation among the three types analyzed. Sensory analysis clearly evidenced differences among genotypes: the San Marzano ecotypes revealed sensory profiles that differ from the control processing variety in perception of sweetness and sourness, whereas the Sorrento and Vesuvio ecotype profiles were similar to the fresh market control. In particular, Vesuvio genotypes exhibited a good intensity of tomato aroma and sweetness and an intermediate level of acidity. Finally, molecular characterization performed through AFLP markers provided evidence for a very high level of polymorphism: an ecotype-specific AFLP pattern was identified for at least 11 genotypes, thus defining their molecular fingerprints. AFLP markers were used to assess the genetic diversity of 77 breeding lines from three of the world's major canola quality Brassica juncea breeding programs from Canada (Agriculture and Agri-Food Canada and Saskatchewan Wheat Pool) and Australia (Agriculture Victoria). The objectives of the paper were to assess the genetic diversity within and between these three breeding programs and to assess genetic diversity of the canola quality germplasm as compared to mustard quality B. juncea. Fifteen lines of mustard quality B. juncea from India, China, Russia and Australia were also included in the investigation. Ten EcoR1/Mse1 based primer pairs generated 751 scorable fragments with an average of 26 polymorphic bands per primer pair (35%). Similarity coefficients were calculated using the Simple Matching coefficient and a dendrogram was developed using the UPGMA procedure, resulting in germplasm being partitioned into five main groups. Line specific markers were discovered that have potential in enhancing the efficiency of individual breeding programs using breeding techniques like accelerated backcrossing. Nineteen of the major strawberry ( Fragaria × ananassa Duch.) cultivars grown in the UnitedStates and Canada were examined for AFLP markerpolymorphisms. For the AFLP reactions, the EcoRI-ACC primer was used in combination with four MseI primers ( MseI-CAC, MseI-CAG, MseI-CAT, or MseI-CTT). Each set ofprimers produced 46–66 scorable fragments ranging insize between 50 and 500 bp. The polymorphic fragmentsproduced from each set of primers were more thansufficient to distinguish among all the cultivars,demonstrating the usefulness of AFLP markers forcultivar identification. Similarity coefficients werecalculated based on data from 228 AFLP markers anddata from 15 previously characterized RAPD markers. The RAPD markers had been specifically selected forfingerprinting purposes because they succesfullydistinguish 41 strawberry cultivars, including the 19cultivars analyzed in this study. Separatedendrograms were constructed based on analysis of theAFLP and RAPD marker data using a neighbor-joiningalgorithm. The dendrograms were compared and found tobe very different. Correlations between similaritycoefficients calculated from AFLP marker data,similarity coefficients calculated from RAPD markerdata, and coefficients of coancestry calculated frompedigree information were evaluated. The genetic diversity of two winter oilseed rape (Brassica napus L.) collections was investigated using amplified fragment length polymorphism marker technology. The first collection of 78 genotypes was analyzed using 11 primer-enzyme combinations and PCR products were resolved by 13.35% denaturing polyacrylamide gels. The second collection was investigated using seven fluorescent-labeled AFLP primer combinations and PCR products were separated using capillary electrophoresis. A total of 262 polymorphic AFLP markers were obtained for the first collection and 423 polymorphic markers for the second. On the basis of all markers, principal component analysis was performed for both collections separately. The unweighted pair group method with arithmetic method based on the coefficient of dissimilarity separated the genotypes in collection 1 into two clusters and those in the second collection into three distinct clusters. Resynthesized lines formed a cluster that was clearly distinct from 42 winter oilseed rape parental lines in the cytoplasmic male sterility ogura system. Analysis of molecular variance showed that 79% and 81% of the detected variation was found to be within the groups (in the two collections, respectively), while the variation between groups contributed, respectively, to only 21% and 19% of the variance. Our results indicate that AFLP technology can be useful for the creation of a gene pool of parental components of winter oilseed rape hybrid cultivars.
