Abstract

The increasing severity and prevalence of powdery mildew aided by extensive use of semi-dwarf cultivars and high levels of nitrogenous fertilizers are causing significant yield losses in wheat. Resistant cultivars are the most cost-effective and environmentally friendly approach to manage the disease. The objective of this study was to identify quantitative trait loci (QTL) for powdery mildew resistance in a doubled haploid (DH) population from a cross between leading Chinese cultivars, Yangmai 16 and Zhongmai 895. A high-density genetic map comprising of 14,480 non-redundant markers (equal to 148,179 SNPs) in 21 wheat chromosomes was constructed by genotyping the population with the Wheat 660 K SNP array. The DH population was phenotyped for powdery mildew resistance at the adult plant stage in multiple field trials, including four environments in the 2016–2017 cropping season and two environments in 2017–2018. Composite interval mapping detected six stable QTL explaining 3.8–23.6% of the phenotypic variance across environments. QPmyz.caas-5DS, QPmyz.caas-6BL and QPmyz.caas-7BS, are probably new QTL for powdery mildew resistance. One SNP marker closely linked to QPmyz.caas-6BL, the most stable QTL, was converted into a Kompetitive Allele-Specific PCR marker (K_AX-94973433) and validated on 103 commercial wheat cultivars. Significantly lower maximum disease severities of cultivars with the resistance-associated allele than those with the susceptibility-associated allele at QPmyz.caas-6BL in some environments indicated partial effectiveness of the marker. Abstract

Improvement of end-use quality in bread wheat depends on a thorough understanding of current wheat quality and the influences of genotype (G), environment (E), and genotype by environment interaction (G x E) on quality traits. Thirty-nine spring-sown spring wheat (SSSW) cultivars and advanced lines from China were grown in four agro-ecological zones comprising seven locations during the 1998 and 1999 cropping seasons. Data on 12 major bread-making quality traits were used to investigate the effect of G, E, and G x E on these traits. Wide range variability for protein quantity and quality, starch quality parameters and milling quality in Chinese SSSW was observed. Genotype and environment were found to significantly influence all quality parameters as major effects. Kernel hardness, flour yield, Zeleny sedimentation value and mixograph properties were mainly influenced by the genetic variance components, while thousand kernel weight, test weight, and falling number were mostly influenced by the environmental variance components. Genotype, environment, and their interaction had important effects on test weight, mixing development time and RVA parameters. Cultivars originating from Zone VI (northeast) generally expressed high kernel hardness, good starch quality, but poor milling and medium to weak mixograph performance; those from Zone VII (north) medium to good gluten and starch quality, but low milling quality; those from Zone VIII (central northwest) medium milling and starch quality, and medium to strong mixograph performance; those from Zone IX (western/southwestern Qinghai-Tibetan Plateau) medium milling quality, but poor gluten strength and starch parameters; and those from Zone X (northwest) high milling quality, strong mixograph properties, but low protein content. Samples from Harbin are characterized by good gluten and starch quality, but medium to poor milling quality; those from Hongxinglong by strong mixograph properties, medium to high milling quality, but medium to poor starch quality and medium to low protein content; those from Hohhot by good gluten but poor milling quality; those from Linhe by weak gluten quality, medium to poor milling quality; those from Lanzhou by poor bread-making and starch quality; those from Yongning by acceptable bread-making and starch quality and good milling quality; and those from Urumqi by good milling quality, medium gluten quality and good starch pasting parameters. Our findings suggest that Chinese SSSW quality could be greatly enhanced through genetic improvement for targeted well-characterized production environments.

Abstract

The endosperm storage proteins of 205 Chinese bread wheat cultivars and advanced lines were fractionated by SDS-PAGE to determine their high molecular weight (HMW) glutenin subunit composition. Seventeen alleles were identified: three at Glu-A1, eight at Glu-B1, and six at Glu-D1. The most common alleles were Null, 1, 7+8, 7+9, and 2+12. The results indicate that wheats from different regions differ in their frequencies of HMW glutenin subunits, however, none of the subunits could be related to specific environments. The glutenin quality scores of Chinese wheats ranged from 3 to 10, with an average of 6.7. Increasing quality scores have implications in improving steam-bread making quality for Chinese consumers. On the basis of HMW glutenin subunit composition, Chinese wheats are close to European wheats, especially Italian wheats because several Italian introductions are widely distributed in the pedigrees of Chinese wheat.

Abstract

Three groups of characters selected using three methods were used to calculate genetic distance among two groups of winter wheat lines using Euclidean distance based on principal components. These wheat lines were then clustered by the unweighted pair-group arithmetic average method. The relationship between genetic distance and standardized potence was seemingly influenced by genetic materials. In order to obtain promising crosses, it is advisable to select parents from intercluster rather than intracluster materials. The results suggested that the repeatability method was better than the conventional and weighting methods for selecting characters for distance analysis.

Abstract

Arabinoxylans (AX) are major polymers of wheat grain cell walls and affect the end-use properties and nutritional quality. Total (TOT-AX), water-unextractable (WU-AX), and water-extractable (WE-AX) AX contents were determined in grain of 240 recombinant inbred wheat lines (RILs), derived from the cross PH82-2/Neixiang 188. A total of 195 SSR and STS markers were used to construct a genetic map and to detect quantitative trait loci (QTL). Wide ranges of variation in TOT-AX, WU-AX, and WE-AX contents were observed among the RILs, with line effects contributing the largest phenotypic variation. Broad sense heritabilities ( h) were 0.50, 0.38 and 0.71, respectively. Inclusive composite interval mapping (ICIM) identified four additive QTL for TOT-AX content on chromosomes 1B, 1D, 3B and 5B, explaining 3.7-14.6 % of the phenotypic variance (PV), among which two were involved in significant additive × season interactions, contributing 0.5-2.4 % of the PV. Two additive QTL for WU-AX content were identified on chromosomes 1B and 1D, accounting for 6.4 and 8.6 % of the PV, respectively; both were involved in significant additive × season interactions, contributing 0.6-1.4 % of the PV. Nine additive QTL for WE-AX content were identified on chromosomes 1A, 1B, 2B, 3B, 5A, 5B, 6B, 7A and 7B, explaining 3.8-15.2 % of the PV, and three of them were involved in significant additive × season interactions, contributing 0.5-2.1 % of the PV. Two pairs of epistatic QTL, with significant epistatic × season interactions, were identified for TOT-AX and WU-AX contents, contributing 16.6 and 18.6 % of the PV, respectively. Abstract

Fusarium head blight (FHB) poses a challenge for wheat breeders worldwide; there are limited sources of resistance and the genetic basis for resistance is not well understood. In the mid-1980s, a shuttle breeding and germplasm exchange program launched between CIMMYT-Mexico and China, enabled the incorporation of FHB resistance from Chinese bread wheat germplasm into CIMMYT wheat. Most of the Chinese wheat materials conserved in the CIMMYT germplasm bank had not been fully characterized for FHB reaction under Mexican environments, until 2009, when 491 Chinese bread wheat lines were evaluated in a FHB screening nursery in Mexico, and 304 (61.9 %) showed FHB indices below 10 %. Subsequent testing occurred in 2010 for plant height (PH), days to heading (DH), and leaf rust response. In 2012, 140 elite lines with good agronomic types were further evaluated for field FHB reaction and deoxynivalenol (DON) accumulation. Most of the tested lines showed good resistance: 116 (82.9 %) entries displayed FHB indices lower than 10 %, while 89 (63.6 %) had DON contents lower than 1.0 ppm. Significant negative correlations were observed between FHB traits (FHB index, DON content, and Fusarium damaged kernels) and PH, DH, and anther extrusion. A subset of 102 elite entries was selected for haplotyping using markers linked to 10 well known FHB quantitative trait loci (QTL). 57 % of the lines possessed the same 2DL QTL marker alleles as Wuhan 1 or CJ 9306, and 26.5 % had the same 3BS QTL allele as Sumai 3. The remaining known QTL were of low frequency. These materials, especially those with none of the above tested resistance QTL (26.5 %), could be used in breeding programs as new resistance sources possessing novel genes for FHB resistance and DON tolerance.
