Per- and polyfluoroalkyl substances (PFAS) are a class of persistent organic pollutants used in industrial applications due to their physicochemical properties that results in their ubiquitous presence across environmental matrices. To date, legacy PFAS have been well-studied, however, the concentration of alternative PFAS may exceed the concentration of legacy pollutants and more information is needed regarding the sub-lethal toxicity at the molecular-level of aquatic model organisms, such as Daphnia magna. Perfluorobutanoic acid (PFBA), perfluorohexanoic acid (PFHxA), perfluorohexanesulfonic acid (PFHxS), and perfluorononanoic acid (PFNA), are four widely detected PFAS alternatives of varying chain length and polar functionality that are quantified in aquatic environments. This study examines the metabolic perturbations of PFAS with varying chemistries to D. magna using targeted mass spectrometry -based metabolomics. Daphnia were acutely exposed to sub-lethal concentrations of PFBA, PFHxA, PFHxS, and PFNA before the polar metabolite profile was extracted from single organisms. Multivariate analysis demonstrated significant separation between the sub-lethal concentrations of PFHxA, PFHxS, and PFNA relative to the controls; in sum, longer chain lengths demonstrated greater overall perturbations to the extracted metabolic profiles. Univariate statistics revealed significant perturbations in the concentrations of several amino acids, nucleotides/nucleosides, and neurotransmitters with exposure to PFAS. These metabolic perturbations are consistent with disruptions in energy metabolism (pantothenate and Coenzyme A metabolism, histidine metabolism) and protein synthesis (aminoacyl-tRNA biosynthesis and amino acid metabolism) which were identified through biochemical pathway analysis. These results provide evidence that while PFAS chemistry (chain length and polar functional group) invokes unique metabolic responses there is also an underlying toxic mode of action that is common with select PFAS exposure. Overall, this work highlights the capabilities of environmental metabolomics to elucidate the molecular-level perturbations of pollutants within the same chemical class to model aquatic organisms which can be used to prioritize risk assessment of substituted PFAS alternatives. Atrazine toxicity to earthworms is still not fully understood, particularly at sub-lethal concentrations. Because of the ubiquity of atrazine in the environment, it is imperative to understand the impacts of atrazine presence on soil-dwelling organisms. To examine this in detail, we used 1H nuclear magnetic resonance (NMR)-based metabolomics to elucidate earthworm (Eisenia fetida) responses after a 48 h of atrazine exposure in contact tests. Earthworms were exposed to four sub-lethal concentrations of 362.4 ng/cm2, 181.2 ng/cm2, 90.6 ng/cm2 and 45.3 ng/cm2, which correspond to 1/8th, 1/16th, 1/32nd and 1/64th of the LC50 value respectively. After exposure, polar metabolites were isolated from earthworm tissues and analyzed using 1H NMR spectroscopy. Sub-lethal atrazine exposure induced a non-monotonic response with respect to exposure concentration and caused an overall suppression in earthworm metabolism. Maltose, fumarate, malate, threonine/lactate, adenosine-5'-triphosphate (ATP), betaine, scyllo-inositol, glutamate, arginine and glutamine were the metabolites identified as most sensitive to atrazine exposure. These observed fluctuations in the metabolic profile suggest that atrazine reduced ATP synthesis and negatively impacted the health of earthworms with acute sub-lethal exposure. Our study also demonstrates the utility of NMR-based metabolomics for the basic assessment of sub-lethal toxicity which can then be used for more targeted approaches with other molecular techniques. Anthropogenic activity is increasing the presence of contaminants that enter waterways through wastewater effluent and urban and/or agricultural runoff, generally in complex mixtures. Depending on the mode of action of the individual contaminant within the mixture, toxicity can occur due to contaminants having similar or dissimilar modes of action. However, it is unknown how the metabolome responds to sublethal contaminant mixtures in the keystone genus Daphnia. In the present study we examined D. magna metabolic responses to acute sublethal exposure of propranolol, carbamazepine, and perfluorooctanesulfonic acid (PFOS) as well as in binary (propranolol-carbamazepine, propranolol-PFOS, carbamazepine-PFOS) and tertiary mixtures (carbamazepine-propranolol-PFOS), all at 10% of the median lethal concentration of the population (LC50). The metabolome was measured using 1 H nuclear magnetic resonance (NMR) and characterized using principal component analysis, regression analysis, and fold changes in metabolite relative to the unexposed (control) group. The averaged principal component analysis scores plots revealed that carbamazepine-PFOS and carbamazepine-propranolol-PFOS exposures were significantly different from the control treatment. After normalizing the toxicity of each contaminant, we found that some metabolites responded monotonically, whereas others displayed a nonmonotonic response with increasing toxicity units. The single contaminant exposures and 2 binary mixtures (propranolol-carbamazepine, and propranolol-PFOS) resulted in minimal changes in the identified metabolites, whereas the carbamazepine-PFOS and carbamazepine-propranolol-PFOS displayed increases in several amino acid metabolites and decreases in glucose. Overall, our results highlight the sensitivity of the metabolome to distinguish the composition of the contaminant mixtures, with some mixtures displaying heightened responses versus others. Nuclear magnetic resonance (NMR) spectroscopy is an essential tool for studying environmental samples but is often hindered by low sensitivity, especially for the direct detection of nuclei such as13C. In very heterogeneous samples with NMR nuclei at low abundance, such as soils, sediments, and air particulates, it can take days to acquire a conventional13C spectrum. The present study describes a prescreening method that permits the rapid prediction of experimental run time in natural samples. The approach focuses the NMR chemical shift dispersion into a single spike, and, even in samples with extremely low carbon content, the spike can be observed in two to three minutes, or less. The intensity of the spike is directly proportional to the total concentration of nuclei of interest in the sample. Consequently, the spike intensity can be used as a powerful prescreening method that answers two key questions: (1) Will this sample produce a conventional NMR spectrum? (2) How much instrument time is required to record a spectrum with a specific signal-to-noise (S/N) ratio? The approach identifies samples to avoid (or pretreat) and permits additional NMR experiments to be performed on samples producing high-quality NMR data. The use of consumer products and pharmaceuticals that act as contaminants entering waterways through runoff and wastewater effluents alters aquatic ecosystem health. Traditional toxicological endpoints may underestimate the toxicity of contaminants, as lethal concentrations are often orders of magnitude higher than those found within freshwater ecosystems. While newer techniques examine the metabolic responses of sublethal contaminant exposure, there has been no direct comparison with ontogeny in Daphnia. It was hypothesized that Daphnia magna would have distinct metabolic changes after 3 different sublethal contaminant exposures, because of differences in the toxic mode of action and ontogeny. To test this hypothesis, the proton nuclear magnetic resonance metabolomic profiles were measured in D. magna aged day 0 and 18 after exposure to 28% of the lethal concentration of 50% of organisms tested (LC50) of atrazine, propranolol, and perfluorooctanesulfonic acid (PFOS) for 48 h. Principal component analysis revealed significant separation of contaminants from the control daphnids in both neonates and adults exposed to propranolol and PFOS. In contrast, atrazine exposure caused separation from the controls in only the adult D. magna. Minimal ontogenetic changes in the targeted metabolites were seen after exposure to propranolol. For both atrazine and PFOS exposures ontogeny exhibited unique changes in the targeted metabolites. These results indicate that, depending on the contaminant studied, neonates and adults respond uniquely to sublethal contaminant exposure. Environ Toxicol Chem 2017;36:938-946. © 2016 SETAC.

Nuclear magnetic resonance (NMR) microimaging is a noninvasive and nondestructive technique that has great potential for the study of soil processes. Hydrogen-1 NMR microimaging techniques were used to examine the distribution of water in four different soil cores. Fluorine-19 NMR microimaging is also used to study the transport of three model contaminants (hexafluorobenzene, sodium fluoride, and trifluralin) in soil columns. The 1H water distribution studies demonstrate that NMR microimaging can provide unique detail regarding the nature and location of water in soils. Image distortion (magnetic susceptibility) was observed for soil samples low in water (20–28% by weight) and that contained an iron content of 0.73 to 0.99%. Highly resolved images were obtained for the organic-rich soil (Croatan sample) and also facilitated the analysis of bound and unbound soil water through varying spin echo times. The contaminant studies with 19F NMR demonstrated that preferential flow processes can be observed in soil cores in as little as 16 h. Studies with hexafluorobenzene produced the highest quality images whereas the definition decreased over time with both trifluralin and sodium fluoride as the compounds penetrated the soil. Nonetheless, both 1H and 19F NMR microimaging techniques demonstrate great promise for studying soil processes.

The earthworm Eisenia fetida is frequently used in ecotoxicological studies; however, it has not yet been investigated using proton nuclear magnetic resonance ((1)H NMR) metabolic profiling methods. The present study investigates the impact of depuration time, sample homogenization, and different extraction solvents on the quality and reproducibility of the (1)H NMR spectra of E. fetida with the goal of determining whether this species is suitable for future metabonomic studies. A depuration time of 96 h, followed by intact lyophilization before homogenization and extraction into a deuterium oxide (D(2)O)-based phosphate buffer, was found to produce extracts with excellent (1)H NMR reproducibility. The D(2)O buffer extracted the largest quantity of the widest variety of earthworm metabolites, which is consistent with the results from other studies using different earthworm species. Nuclear magnetic resonance assignments of the major metabolites in the D(2)O-based buffer also were performed and found to be similar to those for other earthworm species, such as Eisenia veneta, but also to have characteristic attributes in E. fetida. The major metabolites identified include amino acids (alanine, arginine, glutamic acid, glutamine, glycine, leucine, lysine, phenylalanine, serine, tyrosine, and valine), two sugars (glucose and maltose), the sugar alcohol mannitol, and the polyalcohol inositol. Two other earthworm species (Lumbricus rubellus and Lumbricus terrestris) also were examined using protocols developed for E. fetida, and of the three species, the (1)H NMR spectra of E. fetida had the least variation, indicating this species is well-suited for future metabolomic-based ecotoxicity studies.

We examined the molecular-level interactions of aromatic compounds with a humic acid that was extensively characterized with one- and two-dimensional nuclear magnetic resonance (NMR) spectroscopy. Interactions of target compounds were evaluated by monitoring their NMR relaxation properties. Solid-state NMR revealed that the humic sample contains paraffinic carbon (31%), substituted aliphatic carbon (30%), and aromatic carbon (28%). The liquid-state experiments further identified amino acids, a range of carbohydrates, methoxylated aromatics (likely derived from lignin), and a series of aliphatic chains. The 13C spin-lattice relaxation time (T1) of site-specifically labeled naphthalene, 1-naphthol, and quinoline was found to decrease with increasing additions of humic acid. The 1H T1 values also were measured and demonstrated that the association with the humic acid was not specific because all the protons acquired the T1 value of the humic acid at the same rate. The lack of a chemical shift change and an increase in signal line broadening indicates that the interaction between these compounds and humic acid is noncovalent. These interactions were detected at low humic concentrations (5 mg C/L) and suggest that low concentrations of humic material, which are prevalent in both aquatic and terrestrial systems, will significantly affect the fate and transport of contaminants in the environment.

The sorptive behavior of 17 beta-estradiol (estradiol), estrone, and 17alpha-ethinylestradiol (EE2) from aqueous solutions to four soil samples, two clay minerals, and sand was examined. The measured sorption isotherms were found to be nonlinear and soil isotherm data fit the Freundlich model. Alternatively, both the Langmuir and Freundlich models were used for the mineral samples. The sorption affinity of steroid estrogens was found to be greater for montmorillonite than kaolinite and the sand. The soil Freundlich coefficients (K(F)) for estradiol, estrone, and EE2 were observed to increase with organic carbon (OC) content, and resulting Freundlich coefficients that were normalized to the OC content (K(F)OC) were observed to be within the same range for estradiol and estrone but not for EE2. Sorption of steroid estrogens in soil appears to be governed by OC and expanding clay mineral content; thus, estimating sorption coefficients from physicochemical properties may underestimate sorption in soils or sediments that are rich in OC and smectitic clay minerals. Analysis of soils by solid-state (13)C nuclear magnetic resonance did not reveal any trends between sorption capacity and organic matter structure. Competitive sorption experiments revealed that the degree of competition varied with the OC and mineral content, further suggesting that specific soil properties are important for understanding sorption of estrogens in terrestrial environments.

Many sorption studies aim to elucidate organic matter structure and contaminant sorption relationships. Through this pursuit, a great deal of insight has been gained about contaminant interactions with humic fractions, namely the fulvic and humic acid isolates. Comparatively, less is known about the structure and environmental reactivity of the humin fraction; however, researchers have reported that the humin fraction consistently produces higher sorption coefficients than the corresponding source material and other humic fractions. In this paper, we report on a study that uses solid-state 13C nuclear magnetic resonance (NMR) to characterize six humin samples extracted from soil. In addition, 1-naphthol sorption was measured for each whole-soil and humin sample. With the exception of the peat sample, the humin samples yielded significantly higher organic carbon-normalized sorption coefficients (K(oc)) compared with the whole-soil samples. The solid-state 13C NMR analysis reveals the presence of amorphous, polymethylene-rich domains in all of the humin samples. Other researchers have indicated that these domains exhibit a high affinity for hydrophobic organic contaminants. Consequently, we hypothesize that the concentration of amorphous, polymethylene-rich domains in soil humin is responsible for the high sorption coefficients reported here and by other researchers.
