Recent industrial developments have resulted in an increase in the use of so-called technology-critical elements (TCEs), for which the potential impacts on aquatic biota remain to be evaluated. In the present study, quantitative ion character-activity relationships (QICARs) have been developed to relate intrinsic metal properties to their toxicity toward freshwater aquatic organisms. In total, 23 metal properties were tested as predictors of acute median effect concentration (EC50) values for 12 data-rich metals, for algae, daphnids, and fish (with and without species distinction). Simple and multiple linear regressions were developed using the toxicological data expressed as a function of the total dissolved metal concentrations. The best regressions were then tested by comparing the predicted EC50 values for the TCEs (germanium, indium, gold, and rhenium) and platinum group elements (iridium, platinum, palladium, rhodium, and ruthenium) with the few measured values that are available. The 8 "best" QICAR models (adjusted r 2  > 0.6) used the covalent index as the predictor. For a given metal ion, this composite parameter is a measure of the importance of covalent interactions relative to ionic interactions. Toxicity was reasonably well predicted for most of the TCEs, with values falling within the 95% prediction intervals for the regressions of the measured versus predicted EC50 values. Exceptions included Au(I) (all test organisms), Au(III) (algae and fish), Pt(II) (algae, daphnids), Ru(III) (daphnids), and Rh(III) (daphnids, fish). We conclude that QICARs show potential as a screening tool to review toxicity data and flag "outliers," which might need further scrutiny, and as an interpolating or extrapolating tool to predict TCE toxicity. The environmental risk assessment of rare earth elements (REEs) requires data on their potential toxicity. In the present study, the toxicity of lanthanum (La) and cerium (Ce) was studied in relation to metal speciation in solution. For both La and Ce, the use of organic ligands demonstrated that the calculated free ion concentration was a good indicator of toxicity. Whether in the absence or presence of organic ligands, when based on free ion concentrations, the obtained half-maximal effective concentrations were similar. When all generated data were pooled, Ce and La showed identical toxicity thresholds after 120 h of exposure with free ion concentration-based median effective concentration values (95% confidence intervals) of 0.48 (0.38-0.60) µM and 0.47 (0.36-0.61) µM for La 3+ and Ce 3+ , respectively. The inhibition of algal growth was also correlated with the intracellular lanthanide concentrations, regardless of the ligand used. Finally, increasing the ambient calcium concentration protected the test algae by reducing the amount of lanthanide internalized into the cells. These results suggest that, at constant pH (5.5), REE accumulation and toxicity are linked to the free ion concentration and ambient calcium concentration, as predicted by the biotic ligand model. Biomolecules involved in handling cytosolic metals in the liver of the yellow perch (Perca flavescens) were characterized in juvenile fish collected from 4 lakes constituting metal contamination gradients. Using size-exclusion liquid chromatography coupled to an inductively coupled mass spectrometer, we determined metal distributions among ligands of different molecular weights in the cytosol, before and after a heat denaturation step designed to isolate metallothionein-like peptides and proteins. Silver, Cd, and Cu found in the heat-stable protein supernatants were indeed largely present as metallothionein-like peptide complexes; but Co, Ni, and Tl, also present in the heat-stable protein supernatants, did not coelute with metallothionein-like peptides and proteins. This difference in metal partitioning is consistent with the known preference of "soft" metals such as Ag, Cd, and Cu(I) for thiolated ligands and the contrasting tendency of Co and Ni to bind to ligands with oxygen and nitrogen as donor atoms. Metal handling in the whole cytosol also reflected these differences in metal-binding behavior. For Cd and Cu, the importance of the molecular weight pool that includes metallothionein-like peptides and proteins increased relative to the other pools as the total cytosolic metal concentration ([M] cytosol ) increased, consistent with a concentration-dependent detoxification response. In contrast, for Ni and Tl the increase in [M] cytosol was accompanied by a marked increase in the high-molecular weight (670-33 kDa) pool, suggesting that hepatic Ni and Tl are not effectively detoxified. Predicting metal availability and toxicity for chronic (several hours or days) metal exposure scenarios, even for unicellular algae, is a major challenge to existing toxicity models. This is because several factors affecting metal uptake and toxicity, such as the release of metal-binding exudates, changes in the kinetics of metal uptake and toxicity over time, and algal physiological acclimation to internalized metals, are still poorly understood. The present study assessed the influence of these factors on Cd uptake and toxicity in laboratory batch cultures of the freshwater alga Pseudokirchneriella subcapitata. To do so, changes in the free Cd(2+) concentrations caused by the release of metal-binding algal exudates were monitored, (109)Cd accumulation in algal cells was measured, and Cd-induced inhibition of algal growth as a function of exposure time (from 12 h to 96 h) was followed. Results indicate that metal-binding exudates may decrease the proportion of the free Cd(2+) ion in solution up to 2-fold, a decrease that affects Cd uptake and toxicity. Pseudokirchneriella subcapitata has the capacity to decrease net Cd uptake rate on short time scales (<24 h), but this reduction in the Cd uptake rate disappeared after 24 h, and Cd toxicity occurred at relatively high Cd concentrations in solution. The present review describes and discusses key interactions between mercury (Hg) and phytoplankton to highlight the role of phytoplankton in the biogeochemical cycle of Hg and to understand direct or indirect Hg effects on phytoplankton. Phytoplankton are exposed to various Hg species in surface waters. Through Hg uptake, phytoplankton affect the concentration, speciation, and fate of Hg in aquatic systems. The mechanisms by which phytoplankton take up Hg are still not well known, but several studies have suggested that both facilitated transport and passive diffusion could be involved. Once internalized, Hg will impact several physiological processes, including photosynthesis. To counteract these negative effects, phytoplankton have developed several detoxification strategies, such as the reduction of Hg to elemental Hg or its sequestration by intracellular ligands. Based on the toxicological studies performed so far in the laboratory, Hg is unlikely to be toxic to phytoplankton when they are exposed to environmentally relevant Hg concentrations. However, this statement should be taken with caution because questions remain as to which Hg species control Hg bioavailability and about Hg uptake mechanisms. Finally, phytoplankton are primary producers, and accumulated Hg will be transferred to higher consumers. Within the biotic ligand model (BLM) construct, major cations are considered to be simple competitors for metal binding to uptake sites and may offer some protection against metal-induced toxicity, but the influence of essential trace elements and cell preconditioning to different micronutrient concentrations on metal uptake and toxicity is considered negligible. To test these underlying assumptions, we monitored Cd uptake and toxicity in a green alga (Chlamydomonas reinhardtii) after long-term exposures (60 h) to a range of environmentally realistic free Zn(2+) , Co(2+) , Fe(3+) , Mn(2+) , Ca(2+) , and Cu(2+) concentrations buffered with nitrilotriacetic acid. A 200-fold increase in free [Mn(2+) ] as well as a 100-fold increase in free [Fe(3+) ] did not affect Cd uptake or toxicity, whereas a 50-fold increase in free [Ca(2+) ] effectively offered some protection, as predicted by the BLM. However, a 10-fold increase in free [Cu(2+) ] significantly enhanced Cd toxicity by a factor of approximately 2, whereas a 100-fold increase in free [Zn(2+) ] and [Co(2+) ] from 10(-11) to 10(-9) M significantly decreased Cd uptake and toxicity by more than twofold. These effects did not change with prior algal acclimation to different essential micronutrient concentrations. Enzymes representing a variety of metabolic pathways were examined in yellow perch (Perca flavescens) collected from a metal-contaminated region (Rouyn-Noranda, Québec, Canada) to determine which were most closely related to fish condition factor, pyloric caeca weight, and visceral lipid accumulation, as well to seek a better understanding of the influence of metal contamination on the physiology and biometrics of perch. Compared to laboratory fish, wild perch were under important energy restrictions. The condition factor of wild fish was correlated with indicators of aerobic metabolism (citrate synthase, cytochrome C oxidase), protein anabolism (nucleoside diphosphokinase), and indicators of lipid accumulation (glucose-6-phosphate dehydrogenase, visceral lipid index). Pyloric caeca weights were well correlated with indicators of protein anabolism, but only when both seasons were examined together, possibly indicating a lag in the response of enzymes to changes in diet. The addition of contaminant stress to existing energy restrictions led to changes in the relationships between enzymes and biometrics, reducing the predictive power of the models for perch in contaminated lakes. Larvae of the insect Chaoborus are used to monitor cadmium (Cd) in lakes. We set out to determine if this animal possesses a metallothionein-like protein to which its Cd could be bound and if the concentrations of such a protein are correlated with those of Cd in the insect and in lakewater. To achieve our goals, we collected water and larvae of several Chaoborus species from 10 lakes situated along an environmental Cd gradient. We found that all of the Chaoborus species possess a metallothionein-like protein and that concentrations of the protein and of Cd differed among species. Concentrations of the metallothionein-like protein were directly related to those in Chaoborus and in lakewater. These direct relationships support the use of Chaoborus larvae as a Cd biomonitor.
