Although the natural occurrence of arsenic-containing lipids (arsenolipids) in marine organisms is now well established, the possible role of these unusual compounds in organisms and in the cycling of arsenic in marine systems remains largely unexplored. We report the finding of arsenolipids in 61 plankton samples collected from surface marine waters of high- and low-nutrient content along a transect spanning the Gulf Stream in the North Atlantic Ocean. Using high-performance liquid chromatography (HPLC) coupled to both elemental and molecular mass spectrometry, we show that all 61 plankton samples contained six identifiable arsenolipids, namely, three arsenosugar phospholipids (AsPL958, 10-13%; AsPL978, 13-25%; and AsPL1006, 7-10% of total arsenolipids), two arsenic-containing hydrocarbons (AsHC332, 4-10% and AsHC360, 1-2%), and a methoxy-sugar arsenolipid that contained phytol (AsSugPhytol, 1-3%). The relative amounts of the six arsenolipids showed clear dependence on the nutrient status of the ambient water with plankton collected from high-nutrient waters having less of the arsenosugar phospholipids and more of the three non-P containing arsenolipids compared to low-nutrient waters. By combining these first field data of arsenolipids in plankton with reported global phytoplankton productivity, we estimate that the oceans' phytoplankton transform per year 50 000-100 000 tons of arsenic into arsenolipids.

Arsenic occurs in marine waters, typically at concentrations of 1-2 μg As kg -1 , primarily as the inorganic species arsenate. Marine animals, however, contain extremely high levels of arsenic (typically 2000-20 000 μg As kg -1 wet mass), most of which is present as arsenobetaine, an organic form of arsenic that has never been found in seawater. We report a method based on ion-exchange preconcentration and HPLC/mass spectrometry to measure arsenobetaine in seawater, and apply the method to samples of seawater collected at various depths from seven sites in the North Atlantic. Arsenobetaine was detected in most samples at levels ranging from 0.5 to 10 ng As kg -1 , and was found at depths down to 4900 m. Furthermore, we report the presence of 15 additional organoarsenicals in seawater, 14 of which had never been detected in marine waters. The arsenobetaine depth profile was related, albeit weakly, to that of chlorophyll; this relationship probably reflects arsenobetaine's release to water from marine animals associated with the euphotic zone rather than its direct biosynthesis by primary producers. Future application of the new method for seawater analysis will shed new light on the biogeochemical cycle of marine arsenic.

Arsenic is an ubiquitous toxic element in the environment, and organisms have evolved different arsenic detoxification strategies. Studies on arsenic biotransformation mechanisms have mainly focused on arsenate (As(V)) reduction, arsenite (As(III)) oxidation, and arsenic methylation; little is known, however, about the pathway for the biosynthesis of arsenosugars, which are significant arsenic transformation products. Here, the involvement of As(III) S-Adenosylmethionine methyltransferase (ArsM) in arsenosugar synthesis is demonstrated for the first time. Synechocystis sp. PCC 6803 incubated with As(III) or monomethylarsonic acid (MMA(V)) produced dimethylarsinic acid (DMA(V)) and arsenosugars, as determined by high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC/ICPMS). Arsenosugars were also detected in the cells when they were exposed to DMA(V). A mutant strain Synechocystis ΔarsM was constructed by disrupting arsM in Synechocystis sp. PCC 6803. Methylation of arsenic species was not observed in the mutant strain after exposure to arsenite or MMA(V); when Synechocystis ΔarsM was incubated with DMA(V), arsenosugars were detected in the cells. These results suggest that ArsM is a required enzyme for the methylation of inorganic arsenicals, but not required for the synthesis of arsenosugars from DMA, and that DMA is the precursor of arsenosugar biosynthesis. The findings will stimulate more studies on the biosynthesis of complex organoarsenicals, and lead to a better understanding of the bioavailability and function of the organoarsenicals in biological systems.

Arsenic occurs naturally in many types of seafood as water- and fat-soluble organoarsenic compounds. Although water-soluble compounds have been well characterized, the fat-soluble compounds, so-called arsenolipids, have until recently remained unknown. We report that sashimi-grade tuna fish, with a total arsenic content of 5.9 microg of As/g dry mass, contains approximately equal quantities of water- and fat-soluble arsenic. The water-soluble arsenic comprised predominantly arsenobetaine (>95%) with a trace of dimethylarsinate. Two fat-soluble compounds, which together accounted for about 40% of the lipid-arsenic, were isolated and characterized. The first was identified as 1-dimethylarsinoylpentadecane [(CH(3))(2)As(O)(CH(2))(14)CH(3)] by comparison of HPLC/mass spectrometric data and accurate mass data with those of an authenticated synthesized standard. The second arsenolipid was postulated as 1-dimethylarsinoyl all-cis-4,7,10,13,16,19-docosahexane from mass spectrometric data and analogy with non-arsenic-containing lipids found in fish. The remaining fat-soluble arsenic consisted of less polar arsenolipids of currently unknown structure. This is the first identification of arsenolipids in commonly consumed seafood.

Information on the effects of long-term organoarsenical consumption by mammals is limited despite the fact that foodstuffs, especially seafood, often contain organoarsenicals at very high concentrations. Here we evaluate the intake, uptake, and excretion (urine and feces) of arsenic by sheep that live on North Ronaldsay in the Orkney Islands and naturally consume large amounts of arsenosugars through their major food source-seaweed. The sheep eat a broad variety of seaweed species, and arsenic concentrations were determined in all the species observed eaten by the sheep (5.7-74.0 mg kg(-1) dry mass). Because of preference and availability, they feed mostly on the seaweed species found to contain the highest arsenic concentrations: Laminaria digitata and Laminaria hyperborea (74 +/- 4 mg kg(-1) dry mass). To quantify the arsenic intake by the sheep, a feeding experiment reflecting natural conditions as close as possible was set up. In the feeding trial, the average daily intake of arsenic by 12 ewes was 35 +/- 6 mg (97% of water-extractable arsenic was present as arsenosugars) gained from feeding on the two brown algae. To test the possible influence of microflora on the metabolism of arsenosugars, six of the sheep were adapted to feeding on grass for 5 months before the start of the trial (control sheep), and the remaining six sheep were kept on their normal seaweed diet (wild sheep). No significant difference in seaweed/arsenic intake and arsenic excretion was found between the two groups of sheep. The arsenic excreted in the feces represents 13 +/- 10% (n = 12) of the total consumed, and on the assumption of that, the average urinary excretion is estimated to 86%.The main arsenic metabolite excreted in urine was dimethylarsinic acid (DMA(V)) (60 +/- 22%) and minor amounts of dimethylarsinoylethanol (DMAE), methylarsonic acid (MA(V)),tetramethylarsonium ion (TMA+), and arsenate (As(V)) together with seven unknown arsenic compounds were also excreted. The urinary arsenic excretion pattern showed a lag period (>4 h) before significant quantities appeared in the urine, an excretion rate that peaked between 4 and 28 h after seaweed intake and a relatively slow half-life (17 h) after end of intake.

Arsenic compounds in aqueous extracts of the marine polychaetes Nereis diversicolor and Nereis virens were determined by HPLC-ICP-MS. Both polychaetes contained most of their water-soluble arsenic as arsenobetaine (approximately 60%), and the tetramethylarsonium ion was also a significant arsenic constituent (approximately 20% in N. diversicolor and approximately 30% in N. virens). Trimethylarsoniopropionate and arsenosugars were present in the Nereis species as minor constituents, and traces of arsenocholine, trimethylarsine oxide, and arsenate were also detected. When N. diversicolor and N. virens were exposed in laboratory experiments to different concentrations of arsenate in seawater (10, 50, 100, 500, and 1000 microg of As L(-1)), both species accumulated arsenic in a dose-dependent manner. Atthe highest exposure level, they achieved mean arsenic concentrations (dry mass) of approximately 70 mg of As kg(-1) (N. virens) and 90 mg of As kg(-1) (N. diversicolor), which represented about a 5-fold increase when compared with the control animals. The arsenic taken up by the polychaetes was readily methylated, and the major metabolite was the tetramethylarsonium ion (up to approximately 85% of the accumulated arsenic). Two other products of arsenic methylation, methylarsonate and trimethylarsine oxide, were also produced to a small extent; however, dimethylarsinate, another likely intermediate in the formation of the tetramethylarsonium ion, could not be detected in these experiments. The remaining accumulated arsenic was present as unchanged arsenate. This is the first report of significant biomethylation of arsenic to the tetramethyl stage and provides a ready explanation for the widespread occurrence of tetramethylarsonium ion in marine animals.
