Management of tailings at metal mine smelter sites can reduce the potential hazards associated with exposure to toxic metal(loid)s and residual organic flotation reagents. In addition, microbes in the tailings harboring multi-resistance genes (e.g., tolerance to multiple antimicrobial agents) can cause high rates of morbidity and global economic problems. The potential co-selection mechanisms of antibiotic resistance genes (ARGs) and metal(loid) resistance genes (MRGs) during tailings sulfate-reducing bacteria (SRB) treatment have been poorly investigated. Samples were collected from a nonferrous metal mine tailing site treated with an established SRB protocol and were analyzed for selected geochemical properties and high throughput sequencing of 16S rRNA gene barcoding. Based on the shotgun metagenomic analysis, the bacterial domain was dominant in nonferrous metal(loid)-rich tailings treated with SRB for 12 months. KEGGs related to ARGs and MRGs were detected. Thiobacillus and Sphingomonas were the main genera carrying the bacA and mexEF resistance operons, along with Sulfuricella which were also found as the main genera carrying MRGs. The SRB treatment may mediate the distribution of numerous resistance genes. KOs based on the metagenomic database indicated that ARGs (mexNW, merD, sul, and bla) and MRGs (czcABCR and copRS genes) were found on the same contig. The SRB strains (Desulfosporosinus and Desulfotomaculum), and the acidophilic strain Acidiphilium significantly contributed to the distribution of sul genes. The functional metabolic pathways related to siderophores metabolism were largely from anaerobic genera of Streptomyces and Microbacterium. The presence of arsenate reductase, metal efflux pump, and Fe transport genes indicated that SRB treatment plays a key role in the metal(loid)s transformation. Overall, our findings show that bio-treatment is an effective tool for managing ARGs/MRGs and metals in tailings that contain numerous metal(loid) contaminants. [ABSTRACT FROM AUTHOR]

A comprehensive understanding of the mobility of both nitrogen (N) and phosphorus (P) and the inter-relationships between P, N, and iron (Fe) in sediments is important for controlling the "internal loadings" of nutrients in lakes. In this research, diffusive gradients in thin film (DGT) assemblies with binding layers (ZrO-AT, chelex, and ZrO) were designed for PO4-P, Fe, ammonium (NH4-N), and nitrate (NO3-N) at sediment/water interface (SWI) in Western Lake Taihu (China). The biogeochemical processes of N and P related to the physicochemical properties, the dynamic P transfer, the distribution characteristics of P microniches, and the estimation of the release risks in sediments in Western Lake Taihu were simultaneously revealed by the passive sampling technique-DGT with the high spatial resolutions (millimeter and sub-millimeter). Based on DGT concentration (CDGT) related to physicochemical properties in sediments, (1) P biogeochemical reactions included P release from Fe-bound P during Fe reduction, algae biomass decomposition, and phosphatase enzyme activity increased by NH4-N; (2) denitrification and dissimilatory nitrate reduction to ammonium (DNRA) led to exchangeable ammonium (NH4ex) enrichment and NH4-N release; anammox depleted NH4-N transfer; organic matter (OM) mineralization favored NH4-N release; and (3) aerobic nitrification led to NO3-N remobilization; denitrification and DNRA reduced NO3-N release. Redox status, OM, Fe, aluminum, or calcium influenced mobilization of nutrients. The numerical model of DGT-induced fluxes in sediments was used for dynamic P transfers with resupply types ("slow" ~ "fast") controlled by labile P pool, resupply constant, response time, and Dspt rate. The formation of P microniches in two dimensions was revealed. Sediment P release risk index (0.49 ~ 36.85 [lg (nmol cm−3 d−1)]) with "light" ~ "high" risks and diffusive fluxes across SWI (µg m−2 d−1) of 15.0 ~ 639 (PO4-P), − 1403 ~ 5010 (NH4-N), and − 1395 ~ 149 (NO3-N) were derived and lake management strategies were provided. In this work, a new resin gel incorporated with layered double hydroxide nanoparticles modified with diethylenetriaminepentaacetic acid is developed for application in diffusive gradients in thin-film devices (abbreviated as LDHs DGT) to monitor eight anions and cations (such as Fe, Mn, Co, Ni, Cu, Cd, Pb, and As) in natural waters and soils. The accumulated anions and cations were quantitatively recovered by one-step elution using 0.5 mol·L−1 HNO3 with an optimized elution time of 30 min. The performance of the LDHs DGT was independent of solution pH (5–8) and ionic strengths (5–100 mmol·L−1). The capacities of the LDHs DGT for Mn(II), Fe(II), Co(II), Ni(II), Cu(II), As(V), Cd(II), and Pb(II) individually are determined to be 202.9, 363.6, 246.9, 88.8, 99.5, 75.3, 159.8, and 671.7 μg·cm−2. During the field deployments in a nature river, LDHs DGT measured concentrations of cations and anions were almost like those measured by the traditional sampling method (except Fe(II), Cd(II), and Co(II)). Chemical pollutants present a substantial threat to the survival of the green turtle (Chelonia mydas). In this study, the concentrations of 12 trace elements (TEs) in seawater, sediments, and green turtle forage and eggshells from the Xisha Islands in the South China Sea, along with their patterns of transfer and accumulation, were identified. The results revealed that the median TE concentrations in seawater and sediments were lower than the first-grade limit values of the national standard in China, indicating a low ecological risk. The concentrations (μg·g−1) of TEs in forage ranged from 0.05–0.69, 3.43–14.4, 157–2391, 27.9–124, 2.05–9.39, 0.30–9.78, 2.01–80.50, 0.18–5.76, 0.06–0.98, 2.00–18.4, 0.02–0.24, and 0.01–0.09 for Cr, Mn, Sr, Fe, Ni, Cu, Zn, Se, Cd, As, Pb, and Hg, respectively. Seawater, sediments, turtle forage, and eggshells exhibited different TE profiles, which were driven by Hg, Sr, Cr, and Pb in seawater and sediments; Fe and Ni in sediments; Cd and As in forage; and Zn, Se, and Cu in eggshells. The contents of Cu, Zn, and Se increased slightly with trophic level, indicating that they were transferred through dietary pathways. Although Cd and As appeared to bioaccumulate in green turtle forage, it was not transferred to their eggshells, which may be related to the excretion and metabolism process in the mother's body. Progressive industrialization in recent decades has contributed to the increase of metal levels in the environment, which has a dangerous impact on human health, primarily pregnant women. In this study, we aimed to compare levels of various elements in maternal and umbilical cord (UC) plasma samples collected from 125 healthy pregnant women, conduct correlation analysis among paired plasma samples, and compare our data with other populations worldwide. The study design included the following elements: essential (Mn, Co, Cu, Zn, Se, Mo), non-essential (Be, Al, Ni, As, Rb, Sr, Cd, Sb, Pb, U), rare earth (La, Pr, Ce, Nd, Sm, Eu, Gd, Dy, Ho, Er), and noble metals (Ru, Rh, Re, Pt). Levels of 30 elements were higher in maternal plasma than in UC plasma samples. However, no disparities at the statistically significant level were found for Be, Zn, Rb, Cd, Ce, and Ho. Correlation analysis among paired plasma samples revealed only positive/synergistic correlations of different strengths between most elements. Compared to other countries across the globe, our participants had considerably lower plasma levels of Zn and higher levels of Co, Ni, and As. This study provides not only a new and deeper comprehension, but also the first insight into the levels, correlation, distribution, and potential transplacental transfer of 30 elements. In recent years, as the abuse of antibiotics, hormones, and herbicides has worsened in aquaculture industry, it is important to monitor the concentrations of those trace contaminants in aquaculture water more effortlessly. The aim of this study was to develop a reliable sampling method for chemical monitoring in aquaculture ponds based on the technique of diffusive gradients in thin films (DGT). A binding material (XDA-1 resin) with high adsorption capacity for antibiotics, hormones, and herbicides was selected. In laboratory, the diffusion coefficients of 11 antibiotics, 3 hormones, and 3 herbicides of the XDA-1-DGT devices were tested and ranged from 1.0×10−6 to 8.7×10−5 cm2/s. During the in situ application of XDA-1-DGT devices in different aquaculture ponds, concentrations of 11 antibiotics, 3 hormones, and 3 herbicides in aquaculture water (CSOLN) via grab sampling were also analyzed and found ranging from 0.03 to 6.3 ng/L, lower than the results based on DGT (CDGT) (1.2–1.3×102 ng/L). The values of CDGT/CSOLN were larger than one unit, suggesting that the rates of resupply of target chemicals (antibiotics, hormones, and herbicides) by the desorption and diffusion of pond water were higher than the rates of uptake by XDA-1-DGT, and pond water was proved a sufficient reservoir of antibiotics for DGT application. The value of Log (CDGT/CSOLN) of 1.1 derived from the application in the aquaculture ponds of 14 regions in Eastern China can be a reference value for future chemical monitoring based on DGT technique. And the accuracy of this value was found hardly affected by the physical and chemical properties of chemicals with pKa ranging from −1 to 12, Log Kow ranging from 0 to 4.5, and Log solubility ranging from 0.0 to 4.0. The trace element of As and Sb have resulted in serious threats to ecological sustainability and human health in the Xijiang River basin, China. The geoaccumulation index (Igeo) results showed the sediment of Xijiang River was heavily contaminated by Sb as well as moderately contaminated by As. The effect of inorganic anions on the released As and Sb from the sediment increases in the following sequence: Cl− < SO42− < CO32− < PO43−. The presence of PO43− and CO32− had the greater effect over As mobility from the sediment compared to Sb. The effect of Ca(II), Al(III), and Mg(II) on the released amount of Sb from the sediment is negligible. Meanwhile, in the case of As, Ca(II) and Mg(II) had small impacts, but the released amount of As increased slightly with an increase of Al(III) concentration. The stability of As and Sb in the sediment was found to be the best at pH 5. The effect of organic compounds (≤ 0.05 mol/L) on the dissolved As percentages from the sediment increased in the following sequence: ethylenediaminetetraacetate (EDTA) < oxalate < citrate, and the effect on Sb release was oxalate < EDTA < citrate at concentrations (≤ 0.025 mol/L). Soil phosphorus fractions in wetland ecosystems have received increasing attention due to its high eutrophication risks. Soil samples were collected to 40 cm depth in three sampling seasons to investigate the seasonal dynamics of organic and inorganic phosphorus fractions, bioavailability, and relationship between those and soil properties in a seasonal-flooding wetland in the Yellow River Estuary. The results showed that inorganic phosphorus (IP) and organic phosphorus (OP) contents exhibited much higher levels in the top 10 cm soils, and declined along soil profiles in spring. IP kept constant along soil profiles in fall, while OP decreased in summer and fall. They were greatly affected by water content (WC), pH, Cl−/SO42−, soil organic matter (SOM), and electrical conductivity (EC). Middle labile organic phosphorus (MLOP) and non-labile organic phosphorus (NLOP) accounted for higher percentages of total OP in summer and fall respectively than labile organic phosphorus (LOP) in spring. MLOP and NLOP levels showed a decrease along soil profiles in spring and in spring/fall, respectively, while NLOP significantly increased with depth in summer. Ca-P was the dominant IP fraction in all soils in three sampling seasons, declined with depth in spring/fall and increased in summer. Comparatively, soluble/loosely-P(S/L-P) generally decreased with depth along soil profiles in three sampling seasons. And residual P (Res-P) kept little change with depth in spring. Fe/Al-P levels decreased firstly and then increased with depth in spring and summer. Available phosphorus and potential bioavailable phosphorus contents decreased with depth in spring and summer not in fall, and had a strong significant positive correlation with WC and SOM. Alkaline phosphatase not acid phosphatase was the key factor influencing soil MLOP levels. Generally, the fractions and bioavailability of phosphorus as well as phosphatase in this region were affected by soil depth, sampling seasons, and soil properties (e.g., WC, pH, Cl−/SO42−, SOM, and EC). Water-level regime alteration-associated redox fluctuation plays a primary role in governing exchange and transformation of nitrogen (N) in water-level fluctuation zones (WLFZs), while few understanding of how hydrological regimes under reservoir operation affected N cycling across the sediment-water interface (SWI), giving rise to uncertainties in reservoir N nutrient management. Batch microcosm simulation experiments with intact sediment cores from WLFZs of the Three Gorges Reservoir (TGR) were conducted for 24 days to identify holistic flooding-drying process mechanism on N-cycling patterns. Our results showed a distinct transition of N-cycling mode across the SWI, shifting from biological denitrogen loss dominated in initial period of flooding to enhance endogenous N retention. A dramatic source-sink switch of nitrous oxide (N2O) occurred in the first 1.5 days during the flooding period. However, combined accelerating migration of NH4+-N from sediment to overlying water, and subsequently enhanced transformation of NH4+-N to NO3−-N formed from flooding to drying rotation, thereby increasing N loading to overlying water. The reason for this investigation could be attributed to intensive N loss through coupled nitrification and denitrification in oxic-anoxic microenvironments after flooding. With oxygen replenishment from atmosphere during drying phase, persistent ammonification of organic N in sediments provided sufficient source of NH4+-N for the formation of NO3−-N fraction in a more oxic overlying water. Therefore, water-level regime alteration by reservoir operation was capable of weakening N removal from water body and lengthening internal N turnover time across redox-variable SWI. These findings elucidate new understanding of holistic hydrological regime mechanisms on N cycling across SWI and provide insight to biogenic N nutrient management for improving the green credentials of hydroelectric reservoir. A passive sampling method based on diffusive gradients in thin films (DGT) using ceria oxide (CeO2) binding gel was developed for in situ measurement of dissolved reactive phosphorus (DRP). CeO2-based DGT showed excellent uptake performance for DRP, and the uptake mass was consistent with the predication by DGT equation. pH (4.2~9.4) and ionic strength (0.01~500 mM) had no effects on the uptake of DRP. Filed deployment of CeO2-DGT in reservoir water and seawater showed that the measureable concentrations of DRP were comparable to those obtained by grab sampling. CeO2-DGT was deployed in sewage sludge, and results showed the ratios (RS) between the concentration (CDGT) by CeO2-DGT and the concentration (CS) obtained by a traditional centrifugation method ranged from 0.23 to 0.58. This result indicated that sludge solid phase was a potential pool of DRP in sludge solution, and the DRP released from sludge solid phase could compensate partly the consumption of DRP at the interface of DGT device during the deployment. The ratios RS had positive correlation with the content of Fe (r = 0.847, p < 0.01) but were reversed with the level of Ca (r = − 0.879, p < 0.01) in sewage sludge. Water samples and sediments from Liangzi Lake were taken and used to study the vertical distribution characteristics of nitrogen (N) and phosphorus (P) in both the overlying and pore water. Fluxes of ammonia (NH4+-N) and phosphate (PO43−-P) were calculated using a one-dimensional transport-reaction model based on Fick's First Law. The results showed that the mean NH4+-N and NO3−-N concentrations in the overlying water of Liangzi Lake were 2.59 and 0.46 mg L−1, respectively. The mean PO43−-P concentrations were lower than the detection limit. Both N and P displayed peaks at the sediment-water interface. For example, the mean concentration of ammonia in pore water in the surficial layer (0–5 cm) was 4.29 ± 2.74 mg·L−1, which was twice than that of the overlying water. Two PO43−-P vertical profile regimes were identified; one had a gradually increasing trend, while the other first increased and then decreased. The mean orthophosphate concentration in the pore water of the surface layer (0–5 cm) was 0.01 ± 0.01 mg·L−1. The spatial distribution of ammonia flux values was highly heterogeneous. Using these data, the annual load contribution of autochthonous ammonia was calculated to be 481 t a−1. Representative sampling of suspended particulate matter is fundamental for assessing river sediment quality, including the distribution and physicochemical characterisation of particles at different hydrological events. This study compares time-integrated samplers and discrete sampling methods, focusing on (a) the representativeness of the different methods and (b) the comparability between the various sampling techniques. The study investigates whether different sampling devices used under the same conditions can reproduce the annual mean. Two time-integrated sampling techniques (Binnensammler floating collector (BS); self-constructed Phillips sampler (PS)) and two discrete sampling systems (continuous-flow centrifuge (CFC); hydrocyclone (HC)) were compared. The monitoring program (August 2013 to August 2014) was conducted using a 4-week sampling frequency at two different monitoring stations in the Rhine river basin. The analysis of physicochemical parameters included total organic carbon (TOC), particulate phosphorus (PP), grain size distribution (GSD), metals, and organic pollutants (PCBs, HCB, PAH). A nested analysis of variance (ANOVA) was used to evaluate the dependence of suspended matter quality parameters on the sampling method, the sampling device, and the sampling station. According to ANOVA, physical and chemical parameters as well as persistent organic pollutants are homogenous for the time-integrated samplers PS and BS. Comparing PS and the reference technique CFC, only the mean annual concentration of TOC is significantly higher for CFC (5.91%; PS, 4.53%) due to degradation processes. With the exception of TOC, Ni, and GSD, data of BS was comparable with that of CFC. Comparing CFC and HC, there are significant differences for GSD, Zn, and Ni. An analysis of the time series of GSD < 63 μm and sum of PAHs confirms the findings of the nested ANOVA, showing that HC time series display significant differences compared with the other samplers. In the present study, a comparative analysis was performed on the extraction of nickel ions (Ni2+) from agricultural wastewater using nanosilica (NS) synthesized from barley (NS-B) and wheat (NS-W) grass waste with a yield of 92.4%. The experimental procedure was conducted on barley and wheat waste to obtain an 85% pure NS that served as the adsorbent for nickel extraction in wastewater. The NS was characterized and studied using X-ray fluorescence (XRF), which demonstrated that NS synthesized from barley contained 94.2% SiO2, while NS synthesized from wheat contained 93.0% SiO2. Scanning electron microscopy (SEM) and atomic force microscopy (AFM) were used to determine the surface morphology of the nanoparticles. The energy-dispersive X-ray (EDX) analysis and Fourier transform infrared (FTIR) analysis were used to determine the elements and functional groups of the synthesized particles, respectively. Lastly, particle size and surface area analyses were performed using the Brunauer–Emmett–Teller (BET) method, which determined that the nanoparticles were 70 and 102 nm for NS-B and NS-W, respectively. The adsorption of nickel ions from agricultural wastewater was studied at various concentrations (10–200 mg/L). The kinetic models indicate that sorption equilibrium time was 65 min and that the reaction followed the pseudo-first-order kinetics model with a regression coefficient (R2) of 0.9289. Corresponding studies indicated that the Freundlich isotherms best describe the sorption reaction with an R2 value of 0.9958, which indicates the multilayer adsorption of nickel on the adsorbent. The ambiguous mechanism that selenite seems to be absorbed by roots via phosphorus (P) and silicon (Si) transporters signifies P and Si may affect selenite uptake. However, the role of P and Si in phloem-mediated selenium (Se) transport within plant tissue is unknown. Therefore, in this work, tomato (Solanum lycopersicum L.) seedlings were exposed to selenite under different hydroponic conditions firstly. And then, split-root experiments were conducted. Results showed that Se uptake decreased as external pH increased. At pH 8, more selenite in the form of SeO32− was assimilated under P-deficient conditions than under P-normal conditions. Silicate inhibited Se uptake only at pH 3 (27.5% H2SeO3 +72.5% HSeO3−). The results of split-root experiments showed that Se concentrations in seedlings increased under heterogeneously high P or Si. Selenium transport from shoots to roots immersed in solution without selenite was also enhanced. This study illustrated that the affinity of tomato roots to assimilate selenite species followed the order of H2SeO3 >HSeO3− >SeO32−. H2SeO3 was absorbed into roots via Si transporters, whereas HSeO3− and a portion of SeO32− were absorbed via low- and high-affinity P transporters, respectively. Wetlands act as kidneys of land and facilitate remediation of metals and other harmful pollutants through uptake by aquatic macrophytes. The aim of the present study was to investigate metal concentrations in sediments and plants, sources of metal origin, and contamination level in Uchalli Wetland Complex. Sediment samples were collected from 15 randomly selected sites. Metal concentrations (Cd, Pb, Ni, Cu, Zn, Cr, As, Mn) in sediments and macrophytes were determined during summer and winter seasons using the inductively coupled plasma technique. Metal concentrations in sediments during summer and winter seasons were in the order as follows: As > Mn > Zn > Cr > Ni > Cd > Pb > Cu and As > Mn > Zn > Cr > Ni > Pb > Cd >Cu respectively. All analyzed metals were within European Union (EU) limits. In macrophytes, these metals were in the order as follows: Mn > As > Ni > Zn > Cr > Cd > Cu > Pb and As > Mn > Zn > Ni > Cr > Cd > Pb during summer and winter seasons respectively. Contamination degree (Cd) (1.023–5.309) for these lakes showed low contamination during both seasons; mCd values (below 1.5) showed very little contamination degree, while the pollution load index (0.012 to 0.0386) indicated no metal pollution in these lakes. PCA applied on sediment showed that Pb, Zn, Cr, Cu, and Cd had anthropogenic sources of origin. As and Mn were due to natural processes while Ni could be resultant of both anthropogenic and natural sources. PCA on macrophytes showed that Ni, Pb, Cr, Zn, Cu; Cd, As; Mn had anthropogenic, natural, and anthropogenic + natural sources of origin. Dissolved sulfide, iron (Fe), and phosphorus (P) in a mudflat (Jiaozhou Bay, China) impacted by shellfish aquaculture were measured in situ by the diffusive gradients in thin films (DGT) technique. A combination of porewater and solid-phase chemistry was used to characterize the interplays of Fe and S, and their control on P mobilization. Below the subsurface layer, two times higher fluxes (FDGT) of dissolved Fe2+ from porewater to the DGT device than those of dissolved sulfide indicate that dissimilatory iron reduction (DIR) dominates over sulfate reduction (SR). Spatial coupling of dissolved Fe2+ and P points to P release driven mainly by reductive dissolution of Fe. Much higher FDGT values of dissolved Fe2+ relative to dissolved P imply that oxidative regeneration of Fe oxides at the sediment-water interfaces (SWIs) of the transitional mudflat serves as an effective "iron curtain" of upward diffusing P. In the mudflat sediments of DIR prevalence, the accumulation of total reduced inorganic sulfur (TRIS) is dampened, which can largely ascribed to enhanced oxidative loss of sulfide and/or limited availability of degradable organic carbon in the dynamic regimes. Low dissolved sulfide concentrations in the sediments leave the majority of reactive Fe unsulfidized and thus abundantly available to buffer newly produced sulfide. [ABSTRACT FROM AUTHOR]

This study deals with further and systematic laboratory evaluation of the already known ammonium 12-molybdophosphate (AMP)-diffusive gradient in thin film (DGT) method, which is used for measuring total Cs concentration in environmental waters. This study confirms that the AMP-binding gel is not stable for pH > 6. In order to reveal a potential impact of AMP degradation on DGT application, time-series experiments were performed by deploying AMP-DGT samplers in Cs-doped moderately basic soft and hard water up to total AMP-binding gel degradation (60 and 175 h of deployment time, respectively). Linear accumulation of Cs by AMP-DGT samplers was observed up to 48 and 58 h in hard and soft waters, respectively. For this deployment time range, AMP-DGT measured over 77 ± 10 and 94 ± 16% of total Cs concentration in hard and soft water, respectively. The difference in DGT response was attributed to Ca2+ and Mg2+ competition reducing the uptake of AMP-DGT samplers in hard water. Shrinkage of agarose-polyacrylamide diffusive gel was experimentally observed only in hard water due to more intensive AMP-binding gel degradation in hard water. Even if the AMP-DGT response was not impacted in this study, it is recommended to use agarose hydrogel as standard diffusive gel. This study aimed to investigate the Sb and As co-accumulating processes of Pteris vittata under soil culture condition, including the transformation of Sb and As, and the difference in co-accumulating ability among different plant species/populations. Two populations of P. vittata and one population of As-tolerant species Holcus lanatus L. were grown on soil co-contaminated by Sb and As. Sb and As speciation in plants was assessed by X-ray absorption near-edge structure (XANES) spectroscopy. P. vittata displayed strong As- but limited Sb-accumulating ability, with the highest shoot concentrations of As and Sb reaching 455 and 26 mg kg−1, respectively. After 28 days culture, the concentrations of Sb and As in the soil solution were reduced by up to 22% and 36% in the P. vittata treatments, respectively. Holcus lanatus showed limited uptake for both metalloids. In P. vittata, the reduction of arsenate to arsenite occurred (with As in shoots all existing as arsenite), but limited reduction of antimonate to antimonite (with more than 90% of Sb in shoots existing as antimonate) was observed. In terms of the differences in metalloid uptake between the two P. vittata populations, the population from the habitat with higher soil As concentration showed 35% higher As uptake than the population from the habitat with lower As concentration. This populational difference may partly result from varying As transformation efficiencies. However, no significant difference was observed in Sb accumulation between the two populations. The soils around the world's largest antimony mine have been contaminated by high concentrations of Sb and As, which might influence microbial diversity in the surrounding soils. The ecological effects of bioavailable Sb and As on the composition and diversity of microbial community in soils remain unknown. In this study, the relative abundance, taxonomic diversity and composition of bacterial community in soils from a typical Sb mine area, and the relationship between the bacterial community and bioavailable concentrations as well as environmental factors have been investigated comprehensively using high-throughput sequencing (HTS) and diffusive gradients in thin films (DGT). The results indicated that Proteobacteria, Acidobacteria, Chloroflexi, Bacteroidetes, Actinobacteria, Gemmatimonadetes, and Cyanobacteria were the dominant bacterial populations at phylum level in all soil samples, accounting for more than 80% of the bacteria sequenced. The abundance and diversity of bacterial community vary along a metal contamination gradient. Redundancy discriminate analysis (RDA) revealed that 74.74% of bacterial community variation in the contaminated soils was explained by six environmental factors (pH, Sb, As, potential ecological risk index (RI), TC, TN), among which pH, Sb, and As were dominant factors influencing the composition and diversity of bacteria. The in situ stabilization has been widely used to remediate metal-contaminated soil. However, the long-term retaining performance of heavy metals and the associated risk after in situ stabilization remains unclear and has evoked amounting concerns. Here, Pb- or As-contaminated soil was stabilized by a commercial amendment. The availability of Pb and As after in situ stabilization were estimated by ten different in vitro chemical extractions and DGT technique. After amendment application, a significant decline in extractable Pb or As was observed in treatments of Milli-Q water, 0.01 M CaCl, 0.1 M NaNO, 0.05 M (NH)SO, and 0.43 M HOAc. Potential available metal(loid)s determined by DGT also showed remarkable reduction. Meanwhile, the results of in vivo uptake assays demonstrated that Pb concentrations in shoots of ryegrass Lolium perenne L. declined to 12% of the control samples, comparable to the extraction ratio of 0.1 M NaNO (15.8%) and 0.05 M (NH)SO (17.3%). For As-contaminated soil, 0.43 M HOAC provided a better estimation of relative phytoavailability (64.6 vs. 65.4% in ryegrass) compared to other extraction methods. We propose that 0.1 M NaNO or 0.05 M (NH)SO for Pb and 0.43 M HOAc for As may serve as surrogate measures to estimate the lability of metal(loid)s after soil remediation of the tested contaminated soils. The diffusive gradients in thin films (DGT) technique was applied to obtain high spatial resolution of dissolved reactive phosphorus (DRP) concentrations in overlying water and sediment porewater in Lake Taihu. A strong positive correlation between total phosphorus (TP) in sediments and DRP concentrations using DGT was found in both of the sampled lake regions. For stations ZSW and DPG, which have high TP background values in sediments, DRP concentrations in overlying water and porewater were much higher than those at stations MLW, DPG, and HX. Also, a high potential for P release at the sediment-water interface was revealed at stations ZSW and DPG due to a significant concentration gradient between overlying water and porewater. In sediment cores from stations ZSW and DPG, DRP porewater concentration profiles using DGT with three diffusive gel thicknesses displayed a similar trend, increasing down to −2 cm or −4 cm, then decreasing down to −8 cm, and then becoming relatively stable in deeper layers. High-resolution profiles of DRP were related to dissolved oxygen concentrations and organic matter concentrations in different sediment layers at stations ZSW and DPG. For most sediment with low TP concentrations at stations MLW, DPG, and HX, DRP concentrations could not be determined at and near the sediment-water interface, then increased gradually down to −8 cm or −10 cm, and remained stable. At stations MLW, DPG, and HX, most DRP (DRP concentrations measured by DGT) values were 10-30 % of the DRP concentrations in porewater, indicating a partial resupply of the sediments to porewater.
