Perfluorooctane sulfonate (PFOS) is associated with male reproductive disorder, but the related mechanisms are still unclear. In this study, we used in vivo and in vitro models to explore the role of Sertoli cell-derived exosomes (SC-Exo)/miR-9-3p/StAR signaling pathway on PFOS-induced suppression of testosterone biosynthesis. Forty male ICR mice were orally administrated PFOS (0.5-10 mg/kg/bw) for 4 weeks. Bodyweight, organ index, sperm count, reproductive hormones were evaluated. Primary Sertoli cells and Leydig cells were used to delineate the molecular mechanisms that mediate the effects of PFOS on testosterone biosynthesis. Our results demonstrated that PFOS dose-dependently induced a decrease in sperm count, low levels of testosterone, and damage in testicular interstitium morphology. In vitro models, PFOS significantly increased miR-9-3p levels in Sertoli cells and SC-Exo, accompanied by a decrease in testosterone secretion and StAR expression in Leydig cells when Leydig cells were exposed to SC-Exo. Meanwhile, inhibition of SC-Exo or miR-9-3p by their inhibitors significantly rescued PFOS-induced decreases in testosterone secretion and the mRNA and protein expression of the StAR gene in Leydig cells. In summary, the present study highlights the role of the SC-Exo/miR-9-3p/StAR signaling pathway in PFOS-induced suppression of testosterone biosynthesis, advancing our understanding of molecular mechanisms for PFOS-induced male reproductive disorders.

Colloids are ubiquitous in soils, and it has been reported that colloids can act as carriers to increase the mobility of poorly soluble contaminants in subsurface environments. Addition of sulfur (S) fertilizer greatly influences on heavy metal behavior in paddy soil, while the influence of S fertilizer on Cu migration and transformation in colloids of soil pore water has not yet been studied. The influence of S fertilizer (S0 and Na2SO4) applied in paddy soils on Cu migration and transformation in colloids of soil pore water from the rice rhizosphere region was explored in this study. The speciation of Cu in colloids of soil pore water from the rice rhizosphere region was explored by advanced synchrotron-based X-ray absorption near-edge spectroscopy (XANES) techniques. The morphology of colloids was characterized by field emission scanning electron microscopy coupled to energy dispersive X-ray spectroscopy (SEM-EDX). At a depth of 20 cm, the concentration of Cu in colloids of the rhizosphere soil pore water in the control was 2.4- and 6.5- fold higher than that in treatments of S0 and Na2SO4, respectively. The colloids in soil pore water were all positively charged, ranging from 2.4 to 7.8 mV, and the size of colloids was 440–740 nm. The proportion of Fe in colloids in the rhizosphere region decreased with S fertilizer application, while the proportions of C and O increased. Sulfur fertilizer application, increased the proportion of Cu-Cysteine, while the proportion of Cu2S decreased in soil colloids. In conclusion, application of sulfur fertilizer in paddy soil decreased the Cu concentration in soil pore water and colloids of the rhizosphere region, thereby decreasing the vertical migration of Cu in soil pore water.

Particulate organic matter (POM) acts as a metals sink in soil, but only a few studies focused on the interaction of POM and heavy metals in paddy soil. The aim of this study is to investigate the interaction between POM and Copper (Cu)/Zinc (Zn). Two levels of Cu (100, 400 mg kg−1) and Zn (250, 500 mg kg−1) were used in a soil culture experiment. Our results showed that POM was porous structure and varied in size. Hydroxyl and carboxyl involved in POM adsorption of Cu and Zn. Rhizosphere effects roughen the surface of POM and enhanced the capacity of POM on heavy metals absorption. Cu-humic (26.2–33.9%) and Cu-citrate (38.5–42.4%) were dominated in POM, and Cu-goethite (41.7–57.7%), Cu-sulphide (6.6–27.6%) was dominated in soil. Rhizosphere effects decreased the proportion of organic-bond Cu along with the increasing the proportion of Cu-sulphide in POM. Addition of Cu and Zn inhibited the degradation of POM but rhizosphere effects promoted. Carbon content was increased in POM by heavy metal and rhizosphere effects. Our findings indicated that POM tended to retain the heavy metals in soil and heavy metals inhibited the degradation of POM, however, rhizosphere effects decreased the stability of POM-metals interactions.

Aluminum dialkyl phosphinates (ADPs) are a promising class of chemicals offering superior flame retardance. However, knowledge on their behavior in vivo is scarce. Hydrolysis has been suggested as one of the major routes of environmental degradation of ADPs. Herein, aluminum methylcyclohexyl phosphinic (AMHP), a kind of ADPs with industrial production in China, and its hydrolysate methyl cyclohexyl phosphinic acid (MHPA) were continuously exposed to Sprague Dawley (SD) rats for 28 days in this study. The same ratio of MHPA in organs to serum and the same daily excretion of MHPA were observed for AMHP exposure group and MHPA exposure group, suggesting the hydrolysis of AMHP in vivo. The hydrolysis of AMHP to MHPA was further confirmed by in vitro simulated human gastric intestinal juice. Therefore, both AMHP and MHPA distributed in liver, kidney and even brain in the form of MHPA. More than 80% of AMHP and MHPA could be excreted by feces and urine. Feces are the main route of excretion of AMHP and MHPA. The denseness of the inflammatory cell in the hepatic portal area and biochemical indexes showed the obvious dose-effect relationship. However, the toxicity of AMHP and MHPA was quite low even with exposure level up to 100 mg/kg dw/day. The low cumulative ability and mild toxicity indicated AMHP as a promising substitute for brominated flame retardant.

As the metabolites of tetrabromobisphenol A (TBBPA), tetrabromobisphenol A mono- and di-methyl ethers (TBBPA MME and TBBPA DME) have been detected in various environmental media. However, knowledge of the contribution of plants to their environmental fates, especially to the interactions between TBBPA DME and TBBPA, is quite limited. In this study, the metabolism and behaviors of TBBPA DME was studied with pumpkin plants through 15-day hydroponic exposure. The TBBPA were also studied separately using in-lab hydroponic exposure for comparison. The results showed that more TBBPA DME accumulated in pumpkin roots and translocated up to stems and leaves compared with TBBPA. Transformation of TBBPA DME occurred later and more slowly than that of TBBPA. Interconversion between TBBPA DME and TBBPA was verified in intact plants for the first time. Namely, TBBPA DME can be biotransformed to TBBPA MME (transformation ratio in mole mass, TRMM 0.50%) and to TBBPA (TRMM 0.53%) within pumpkin; and TBBPA can be biotransformed to TBBPA MME (TRMM 0.58%) and to TBBPA DME (TRMM 0.62%). In addition, two single benzene-ring metabolites, 2,6-dibromo-4-(2-(2-hydroxyl)-propyl)-anisole (DBHPA, TRMM 3.4%) with an O-methyl group and 2,6-dibromo-4-(2-(2-hydroxyl)-propyl)-phenetole (DBHPP, TRMM 0.57%) with an O-ethyl group, were identified as the transformation products in the TBBPA exposure experiments. The transformation and interconversion from TBBPA DME back to TBBPA is reported as a new pathway and potential source for TBBPA in the environment.

Hydroxylated (OH-) and methoxylated (MeO-) polybrominated diphenyl ethers (PBDEs) have been identified ubiquitous in wildlife and environment. However, understanding on their trophic accumulation and human exposure was hitherto limited. In this study, the occurrences and trophic behaviors were demonstrated for OH- and MeO-PBDEs using the biota samples collected from Dalian, a coastal city near Chinese Bohai Sea. ∑OH-PBDEs exhibited a wider concentration range (MDL (method detection limit)-25 ng/g dry weight (dw)) compared with ∑MeO-PBDEs (MDL-2 ng/g dw) and ∑PBDEs (MDL-2 ng/g dw). The congener profiles and distribution patterns revealed that majority of OH- and MeO-PBDEs in marine biota were naturally produced and largely attributed to preying on lower trophic level biota. Though tertiary consumers accumulated more MeO-PBDEs and PBDEs, these chemicals did not show statistically significant biomagnification in the selected food web. Conversely, trophic dilution was determined for ortho-substituted OH-tetraBDEs and OH-pentaBDEs, revealing that trophic dilution was prevalent for naturally produced OH-PBDEs. The dietary intake evaluation of OH-PBDEs (0.4 ng/kg/d) and MeO-PBDEs (0.8 ng/kg/d) via seafood consumption showed that coastal residents were in higher exposure risks to OH-PBDEs and MeO-PBDEs via the massive seafood consumption.

Although epoxiconazole is the worldwidely used fungicide, limited information is known about its toxic effects and bioaccumulation behavior in freshwater ecosystems. In this study, zebrafish were exposed to epoxiconazole at concentrations of 100 and 1000 μg L−1 for 21 d. 1H NMR-based metabolomics analysis showed that low- and high-dose epoxiconazole exposure resulted in two similar but not identical patterns for the change of endogenous metabolites related to energy, lipid and amino acid metabolism. The expression of genes associated with mitochondrial respiratory chain, ATP synthesis and fatty acid β-oxidation were further measured to explore the reason for the disturbed energy metabolism, finding epoxiconazole had an inhibition effect on the genes expression of the above ways. Significant enantioselectivity was observed with (+)-epoxiconazole enrichment in the bioaccumulation process. These results will be of great importance in understanding the toxic effects induced by epoxiconazole and provide important basis for its comprehensive environmental assessment.

Short chain chlorinated paraffins (SCCPs) are ubiquitously present as persistent organic pollutants in the environment. However, little information on the interaction of SCCPs with plants is currently available. In this work, young pumpkin plants (Cucurbita maxima × C. Moschata) were hydroponically exposed to the congener of chlorinated decane, 1,2,5,5,6,9,10-heptachlorodecane (1,2,5,5,6,9,10-HepCD), to investigate the uptake, translocation and transformation of chlorinated decanes in the intact plants. It was found that parent HepCD was taken up by the pumpkin roots, translocated from root to shoots, and phytovolatilized from pumpkin plants to air via the plant transpiration flux. Our data suggested that dechlorination of 1,2,5,5,6,9,10-HepCD to lower chlorinated decanes and rearrangement of chlorine atoms in the molecule were all mediated by the whole pumpkin seedlings. Chlorinated decanes were found in the shoots and roots of blank controls, indicating that chlorinated decanes in the air could be absorbed by leaves and translocated from shoots to roots. Lower chlorinated congeners (C10H17Cl5) tended to detain in air compared to higher chlorinated congeners (C10H16Cl6 and other C10H15Cl7). Potential transformation pathway and behavior of 1,2,5,5,6,9,10-HepCD in pumpkin were proposed based on these experiments.

Lead contamination in soil has become a worldwide threat on food security and human health. To assess the Pb bioavailability and evaluate the safe use of low Pb polluted soil for food production, the speciation of Pb in 19 types of paddy soil were investigated by chemical extraction and X-ray absorption near-edge structure (XANES), and the uptake and accumulation characteristics of Pb in different soil-rice systems were investigated. Moreover, an empirical model was established to predict the content of Pb in rice grain, and field validation was conduct to evaluate model performance. Results showed that the proportion of available Pb in different soil satisfied normal distribution N (0.47, 0.23). Pb(CH3COO)2, GSH-Pb, PbO, PbHPO4 and Pb3(PO4)2 performed well in characterizing the speciation of Pb in different rhizosphere soils, and PbHPO4 accounted for more than 70%. The exceedance of Pb in grain in CK, 0.5X and 1X treatment were 10.5%, 36.1% and 42.1%, respectively, and the accumulation of Pb in grain was significantly related with Pb content in root. Carbonate and organic bound Pb in rhizosphere soil were two major Pb species that influenced the accumulation of Pb in rice. Moreover, content of total Pb, clay and SOM performed well in predicting the Pb content in grain, both for pot and field samples. Arsenic (As) has been recognized as one of the most toxic metalloids present in the surface soil contaminating food chain and posing threat to human life. Sulfur (S) fertilizer is often supplied in paddy soil for rice growth, but its impact on As mobility and related bacteria remains poorly understood. In this study, a pot experiment was set up with two different types of sulfur treatments (element sulfur and Na2SO4) to evaluate the effect of sulfur fertilizers on As speciation in porewater, As fractions in soil, As accumulation in rice plants. Besides, rhizosphere bacterial composition and functional genes that might influence As mobility were also studied. The results revealed that the addition of 150 mg/kg Na2SO4 decreased As(III) and As(V) concentrations in soil porewater at maturation stage by 77% and 64%, respectively. With the same sulfur content, Na2SO4 was more effective than element sulfur. The addition of sulfur fertilizers promoted rice growth and reduced As accumulation in shoots, further reduced As translocation from root to above-ground parts by 39–59%. The addition of sulfur fertilizers had little effect on genes involved in As metabolism. However, the relative abundance of Fe(III) and sulfate reduction related genera increased with the addition of 150 mg/kg Na2SO4, consistent with the increase of Fe(III) reducing bacteria Geobacteraceae and sulfate reducing gene dsrA. The phenomenon likely influenced the decrease of As concentrations in soil porewater and rice uptake. The outcomes indicate that promoting Fe- and S- reducing bacteria in the rhizosphere by sulfur fertilizers may be one way to reduce As risk in the soil-rice system.

The biogeochemical cycling of sulfur in soil is closely associated with the mobility and bioavailability of heavy metals; however the influence of sulfur on the behavior of metal-based nanoparticles has not yet been studied. The influence of S fertilizer (S0 and Na2SO4) applied in paddy soils on CuO NPs behavior in soil pore water was explored in the present study. Synchrotron-based techniques were applied to investigate the migration and speciation transformation of CuO NPs in soil pore water colloids. The application of sulfur fertilizer increased the zeta potential of soil colloids from the rice rhizosphere region and reduced the size of the colloids. Sulfur fertilization decreased the concentration of Cu in soil pore water in the rice rhizosphere region. S0 fertilizer reduced the Cu concentration in soil colloids (by 55.8%-73.5%), while Na2SO4 increased the Cu concentration in soil colloids (by 173.8%-265.1%). Sulfur fertilization changed the spatial distribution of Fe3+ and Cu2+ in colloids, making these ions more likely to be aggregated on the edges of soil colloids. Speciation transformation of CuO NPs happened during the process of migration. The main Cu speciation in the soil colloids were CuO NPs, Cu-Cysteine, Cu2S and Cu-Citrate. Sulfur fertilization increased the proportion of Cu2S (by 40.5%) in soil pore water colloids from the rice rhizosphere region, while the proportion of CuO NPs was reduced (by 18.4%). Sulfur fertilization changed the morphology and elementary composition of colloids in soil pore water, thus influencing the migration of CuO NPs in the soil column through soil colloids.

Currently, copper oxide nanoparticles (CuO NPs) have been widely used in industry, manufacturing and agriculture. The transport and retention of CuO NPs are vital to understanding the fate as well as the life cycle of CuO NPs in the environment. This study systematically investigates the transport and retention of CuO NPs in saturated porous media, and the experimental results were explained by the CFT and DLVO theory. The van der Waals force between CuO NPs and collector was repulsive, resulting in the unfavorable deposition condition. Column experiments were conducted with saturated quartz sand under environmentally relevant pH (6, 8, 9), ionic strength (IS, 1, 10, 50 mM), and humic acid (HA, 0.1–10 mg-C/mL). Experimental results show that the breakthrough curves (BCTs) were affected by different pH and IS. Under pH 6 and 9, the mobility of CuO NPs was enhanced by high IS while the mobility was inhibited by high IS under pH 8. The mobility of CuO NPs was enhanced by humic acid and the effect was best at 0.5 mg-C/mL HA. The experimental results were successfully explained by CFT and DLVO theory, the main mechanisms were aggregation of CuO NPs, interaction energy and collision between CuO NPs and collector. In general, these findings can improve our understanding of the transport and retention of CuO NPs in subsurface environments, and suggest pH, IS, HA may be key factors governing mobility and stability of CuO NPs in natural environment.

A set of toxic metals, i.e. As, Hg, Pb, Cd, Cu, Zn, Ni and Cr, in urban and suburban SDSs were investigated comparatively in the biggest metropolitan area of China, Shanghai. Results showed that all of the metals except As were accumulated greatly, much higher than background values. Geo-accumulation index indicated that metal contamination in urban SDSs was generally heavier than that in suburban SDSs. Potential ecological risk index demonstrated that overall risks caused by metals were considerable. Cd contributed 52% to the overall risk. Multivariate statistical analysis revealed that in urban SDSs, Zn, Ni, Cd, Pb, Cu and Cr were related to traffic and industry; coal combustion led to elevated levels of Hg; soil parent materials controlled As contents. In suburban SDSs, Pb, Cu, As and Cd largely originated from traffic pollution; Zn, Ni and Cr were associated with industrial contaminants; Hg was mainly from domestic solid waste.

A detailed investigation was conducted to understand the contamination characteristics of a selected set of potentially toxic metals in Shanghai. The amount of Pb, Zn, Cu, Cr, Cd and Ni were determined from 273 soil/dust samples collected within urban area. The results indicated that concentration of all metals except Ni in soils was significant, and metal pollution was even severer in roadside dust. A series of metal spatial distribution maps were created through geostatistical analysis, and the pollution hotspots tended to associate with city core area, major road junctions, and the regions close to industrial zones. In attempt of identifying the source of metals through geostatistical and multivariate statistical analyses, it was concluded as follows: Pb, Zn and Cu mainly originated from traffic contaminants; soil Ni was associated with natural concentration; Cd largely came from point-sourced industrial pollution; and Cr, Ni in dust were mainly related to atmospheric deposition.
