Abstract

International audience; Pollutants can have severe detrimental effects on insects, even at sublethal doses, damaging developmental and cognitive processes involved in crucial behaviours. Agrochemicals have been identified as important causes of pollinator declines, but the impacts of other anthropogenic compounds, such as metallic trace elements in soils and waters, have received considerably less attention. Here, we exposed colonies of the European honey bee Apis mellifera to chronic field-realistic concentrations of lead in food and demonstrated that consumption of this trace element impaired bee cognition and morphological development. Honey bees exposed to the highest of these low concentrations had reduced olfactory learning performances. These honey bees also developed smaller heads, which may have constrained their cognitive functions as we show a general relationship between head size and learning performance. Our results demonstrate that lead pollutants, even at trace levels, can have dramatic effects on honey bee cognitive abilities, potentially altering key colony functions and the pollination service.

Abstract

Petroleum can pollute pristine shorelines as a consequence of accidental spills or chronic leaks. In this study, the fate of petroleum hydrocarbons in soft pristine sediment of Caleta Valdés (Argentina) subject to ex situ simulated oil pollution was assessed. Sedimentary columns were exposed to medium and high concentrations of Escalante Crude Oil (ECO) and incubated in the laboratory during 30 days. Levels of aliphatic hydrocarbons at different depths of the sedimentary column were determined by gas chromatography. Oil penetration was limited to the first three centimetres in both treatments, and under this depth, hydrocarbons were clearly biogenic (terrestrial plants) as in the whole sedimentary column of the control assay. Bioturbation by macrobenthic infauna was strongly impacted by oil pollution which resulted in reduced sediment oxygenation and low burial of petroleum hydrocarbons. This may partly explain the limited hydrocarbon biodegradation observed, as indicated by the relatively high values of the ratios nC17/pristane, nC18/phytane, and total resolved aliphatic hydrocarbons/unresolved complex mixture. Correspondingly, at the end of the experiment the most probable number of hydrocarbon-degrading bacteria reached ~ 103 MPN g−1 dry weight. These values were lower than those found in chronically polluted coastal sediments, reflecting a low activity level of the oil-degrading community. The results highlight the low attenuation capacities of Caleta Valdés pristine sediments to recover its original characteristics in a short time period if an oil spill occurs. Abstract

International audience; Species Sensitivity Distributions (SSD) are widely used in environmental risk assessment to predict the con- centration of a contaminant that is hazardous for 5% of species (HC5). They are based on monospecific bioassays conducted in the laboratory and thus do not directly take into account ecological interactions. This point, among others, is accounted for in environmental risk assessment through an assessment factor (AF) that is applied to compensate for the lack of environmental representativity. In this study, we aimed to assess the effects of in- terspecific competition on the responses towards isoproturon of plant species representative of a vegetated filter strip community, and to assess its impact on the derived SSD and HC5 values. To do so, we realized bioassays confronting six herbaceous species to a gradient of isoproturon exposure in presence and absence of a compe- titor. Several modelling approaches were applied to see how they affected the results, using different critical effect concentrations and investigating different ways to handle multiple endpoints in SSD. At the species level, there was a strong trend toward organisms being more sensitive to isoproturon in presence of a competitor than in its absence. At the community level, this trend was also observed in the SSDs and HC5 values were always lower in presence of a competitor (1.12–11.13 times lower, depending on the modelling approach). Our dis- cussion questions the relevance of SSD and AF as currently applied in environmental risk assessment.

Abstract

International audience; Diesel exhaust particulate matter (PM) can have an impact on the environment due to its chemical constitution. A large number of substances such as organic compounds, sulfates, nitrogen derivatives and metals are adsorbed to the particles and desorption of these contaminants could promote genotoxic effects. The objective of this study was to assess the in vivo genotoxicity profile of diesel exhaust PM from heavy-duty engines. Extracts were obtained through leaching with pure water and chemical extraction using three organic solvents (dichloromethane, hexane, and acetone). The in vivo Vicia faba micronucleus test (ISO 29200 protocol) was used to assess the environmental impact of the samples collected from diesel exhaust PM. The solid diesel PM (soot) dissolved in water, and the different extracts, showed positive results for micronucleus formation. After the addition of EDTA, the aqueous extracts did not show a genotoxic effect. The absence of metals in the organic solvent extract indicated that organic compounds also had a genotoxic effect, which was not observed for a similar sample cleaned in a C18 column. Thus, considering the ecological importance of higher plants in relation to ecosystems (in contrast to Salmonella spp., which are commonly used in mutagenicity studies), the Vicia micronucleus test was demonstrated to be appropriate for complementing prokaryotic or in vitro tests on diesel exhaust particulate matter included in risk assessments.

Abstract

International audience; Freshwater mussels such as the invasive Dreissena polymorpha and the indigenous Unio tumidus nourish by high filtration rates and may accumulate cyanobacteria and their toxins during cyanobacterial blooms. Physiological adaptations to cyanotoxins enable organisms to endure cyanobacteria lblooms but may differ between species. Biotransformation and excretion capacities for cyanobacteria and anthropogenic pollutants have been demonstrated for Dreissena polymorpha but less for unionid species. This study compares the activities of biotransformation (glutathioneS-transferase, GST) and antioxidant enzymes (superoxide dismutase, SOD and catalase, CAT) in Dreissena polymorpha to Unio tumidus in response to cyanotoxin exposure (10 mg L 1 and 50 mg L 1 microcystin-LR, respectively, total microcystin from a cyanobacterial crude extract) for 24h and 7 d exposure duration. Enzyme activities in Dreissena polymorpha were measured in the whole mussel tissue, digestive gland and in gills and in Unio tumidus in the digestive gland, gills, mantle, foot as well as in the remaining tissue. The sGST was elevated for the entire exposure period in the whole mussel tissue of Dreissena polymorpha but despite higher basal activities in digestive gland and gills of Unio tumidus, it was rather inhibited or unaltered in most of their tissues. Elevated SOD activity indicated oxidative stress response in Dreissena polymorpha, but not in Unio tumidus. The CAT activity was barely affected in both species, rather inhibited in Unio tumidus, despite again higher basal activities in digestive gland and remaining tissue. Compared to the indigenous Unio tumidus, the investigated biotransformation and oxidative stress combating enzymes respond stronger in the invasive Dreissena polymorpha.

Abstract

International audience; Formation of organometallic complexes in soil solution strongly influence metals phytoavailability. However, only few studies deal with the influence of metalspeciation both on plant uptake and genotoxicity. In the present study, Viciafaba seedlings were exposed for 6 h in controlled hydroponic conditions to 5 μM of lead nitrate alone and chelated to varying degrees by different organic ligands. Ethylenediaminetetraacetic acid and citric acid were, respectively, chosen as models of humic substances and low weight organic acids present in natural soil solutions. Visual Minteq software was used to estimate free lead cations concentration and ultimately to design the experimental layout. For all experimental conditions, both micronucleus test and measure of leaduptake by plants were finally performed. Chelation of Pb by EDTA, a strong chelator, dose-dependently increased the uptake in V. faba roots while its genotoxicity was significantly reduced, suggesting a protective role of EDTA. A weak correlation was observed between total lead concentration absorbed by roots and genotoxicity (r2=0.65). In contrast, a strong relationship (r2=0.93) exists between Pb2+ concentration in exposure media and genotoxicity in the experiment performed with EDTA. Citric acid induced labile organometallic complexes did not demonstrate any significant changes in leadgenotoxicity or uptake. These results demonstrate that metalspeciation knowledge could improve the interpretation of V. fabagenotoxicity test performed to test soil quality.

Abstract

The aim of the present study was the assessment of the sub-chronic effects of silver (AgNPs) and gold nanoparticles (AuNPs) of 40 nm primary size either stabilised with citrate (CIT) or coated with polyethylene glycol (PEG) on the freshwater invertebrate Gammarus fossarum. Silver nitrate (AgNO3) was used as a positive control in order to study the contribution of silver ions potentially released from AgNPs on the observed effects. A multibiomarker approach was used to assess the long-term effects of AgNPs and AuNPs 40 nm on molecular, cellular, physiological and behavioural responses of G. fossarum. Specimen of G. fossarum were exposed for 15 days to 0.5 and 5 µg L−1 of CIT and PEG AgNPs and AuNPs 40 nm in the presence of food. A significant uptake of both Ag and Au was observed in exposed animals but was under the toxic threshold leading to mortality of G. fossarum. Silver nanoparticles (CIT-AgNPs and PEG-AgNPs 40 nm) led to an up-regulation of Na+K+ATPase gene expression. An up-regulation of Catalse and Chitinase gene expressions due to exposure to PEG-AgNPs 40 nm was also observed. Gold nanoparticles (CIT and PEG-AuNPs 40 nm) led to an increase of CuZnSOD gene expression. Furthermore, both AgNPs and AuNPs led to a more developed digestive lysosomal system indicating a general stress response in G. fossarum. Both AgNPs and AuNPs 40 nm significantly affected locomotor activity of G. fossarum while no effects were observed on haemolymphatic ions and ventilation.

Abstract

International audience; The persistence of soil contamination after cessation of oil activities remains a major environmental issue in tropical regions. The assessment of the contamination is particularly difficult on vegetated sites, but promising advances in reflectance spectroscopy have recently emerged for this purpose. This study aimed to exploit vegetation reflectance for estimating low concentrations of Total Petroleum Hydrocarbons (TPH) in soils. A greenhouse experiment was carried out for 42 days on Cenchrus alopecuroides (L.) under realistic tropical conditions. The species was grown on oil-contaminated mud pit soils from industrial sites, with various concentrations of TPH. After 42 days, a significant decrease in plant growth and leaf chlorophyll and carotenoid contents was observed for plants exposed to 5–19 g kg−1 TPH in comparison to the controls (p 0.7). The same indices also succeeded in estimating the concentrations of TPH using random forest regression, at leaf and plant scales (RMSE = 1.46 and 1.63 g kg−1 and RPD = 5.09 and 4.44, respectively). Four out of the 33 indices contributed the most to the models (>75%). This study opens up encouraging perspectives for monitoring the cessation of oil activities in tropical regions. Further researches will focus on the application of our approach at larger scale, on airborne and satellite imagery.

Abstract

International audience; The objective of this study was to investigate chemical, biological and eco-toxicological parameters of a compost produced through the co-composting of dewatered primary sludge (DPS) and date palm waste to evaluate in which extent it can exploited as a bio-fertilizer. DPS and date palm waste were co-composted in aerobic conditions for 210 days. Physico-chemical parameters were evaluated during composting (total organic carbon, total nitrogen, pH, available forms of phosphorus). Furthermore, heavy metals (Cd, Cu, Cr, Pb, Ni, Zn) and antibiotics (fluoroquinolones, macrolides and tetracyclines) content were analyzed in the DPS. To evaluate the genotoxicity of substrates, Vicia faba micronucleus test was carried out. Single and combined toxicities of a mixture of antibiotics (ciprofloxacin, enroflxacin, nalidixic acid, roxithromycin and sulfapyridin) and chromium (Cr2 (SO4)3 and K2Cr2O7) were examined. Although the final compost product showed a significant decrease of the genotoxicity, almost 50% of the micronucleus frequency still remained, which could be explained by the persistence of several recalcitrant compounds such as chromium and some antibiotics. Overall, the presence of antibiotics and chromium showed that some specific combination of contaminants represent an ecological risk for soil health and ecosystems even at environmentally negligible concentrations.

Abstract

International audience; The impact of anaerobic digestion on the bioavailability of copper and zinc from pig slurry was assessed. Both chemical and biological approaches were used independently on raw slurry (RS) and anaerobically digested pig slurry (DS). This work, using ultracentrifugation pellets from the same pig slurry before and after an anaerobic treatment, confirmed that Cu and Zn behave differently in terms of bioavailability, and contrasting results were obtained by chemical and biological assessments. A chemical approach combined a preliminary study of the pH effect on particulate/dissolved metal partitioning, sequential extraction, and biochemical fractionation. This approach tended to show a lower mobility of metals from digested slurry (DS). A biological approach was carried out with Zea mays and Vicia faba to study Cu and Zn uptake in soil amended with RS or DS. This assay could not differentiate the two slurries.

Abstract

The aim of this study was to identify a metallothionein (MT) coding sequence from the freshwater bivalve Corbicula fluminea and to measure the seasonal transcriptional pattern of MT in parallel with several detoxification genes: superoxide dismutase (SOD), catalase (CAT), glutathione S-transferases (GST) and glutathione peroxidases (GPx), in the digestive gland and the gills of this bivalve during a 1-year period. We identified a C. fluminea MT complete cDNA sequence using RT-PCR and RACE-PCR. The amino acid sequence deduced from the coding sequence encodes for a protein of 73 amino acids containing 21 cysteine residues. This protein exhibits high identities and similarities with the MT sequences of numerous bivalves. MT, SOD, CAT, pi-GST and Se-GPx expression patterns did not exhibit major seasonal variations. A slight increase of MT was observed in July. Therefore, the mRNA expression of these five genes could be used as biomarkers for monitoring studies.

Abstract

International audience; The toxicity of cadmium for the Collembola Folsomia candida was studied by determining the effects of increasing Cd concentrations on growth, survival and reproduction in three cultivated and forested soils with different pH (4.5-8.2) and organic matter content (1.6-16.5%). The Cd concentration in soil CaCl2 exchangeable fraction, in soil solution and in Collembola body was determined. At similar total soil concentrations, the Cd concentration in soil solutions strongly decreased with increasing pH. Reproduction was the most sensitive parameter. Low organic matter content was a limiting factor for reproduction. Effect of Cd on reproduction was better described by soil or body concentrations than by soil solution concentration. Values of EC50-Repro expressed on the basis of nominal soil concentration were 182, 111 and 107 μg g−1, respectively, for a carbonated cultivated soil (AU), an acid forested soil with high organic matter (EPC) and a circumneutral cultivated soil with low organic content (SV). Sensitivity to Cd was enhanced for low OM content and acidic pH. The effect of Cd on reproduction is not directly related to Cd concentration in soil solution for carbonated soil: a very low value is found for EC50-Repro (0.17) based on soil solution for the soil with the highest pH (AU; pH=8.2). Chronic toxicity cannot be predicted on the basis of soluble fractions. Critical concentrations were 8×10−5, 1.1, 0.3 μg mL−1, respectively, for AU, EPC and SV soils.

Abstract

International audience; This study concerns a comparative analysis of the acridineorange and Giemsastaining procedures for the fish erythrocyte micronucleusassay. The goal was to optimize the assay in the context of field watermonitoring. Fish (Carassius carassius) were exposed to a reference genotoxic agent, cyclophosphamide monohydrate 5 mg l−1 for 2, 4, and 6 days before testing. Slides from each individual were scored using the two procedures. The results show that the assay was more sensitive when acridineorange was used. When slides were Giemsa stained, the presence of ambiguous artefacts, leading to false positives and increasing random variance, reduced the contrast between exposed and control samples. AcridineOrangestaining was then applied in the context of waterqualitymonitoring. Fish were exposed for 4 days to water sampled in two hydrological contexts: basal flow and spring flood. The results show that exposure to spring flood water in an agricultural stream can induce mutagenicity.

Abstract

Contaminant loadings to the Venice Lagoon peaked from 1950s–1980s and although they have since declined, contaminant concentrations remain elevated in sediment and seafood. In order to identify the relative importance of contaminant sources, inter-media exchange and removal pathways, a modified 10-segment fugacity/aquivalence-based model was developed for octachlorodibenzodioxin/furan (OCDD/F), PCB-180, Pb and Cu in the Venice Lagoon. Results showed that in-place pollution nearby the industrial area, current industrial discharges, and tributary loadings were the main sources of contaminants to the lagoon, with negligible contributions from the atmosphere. The fate of these contaminants was governed by sediment-water exchange with simultaneous advective transport by water circulation. Contaminants circulated amongst the northern and central basins with a small fraction reaching the far southern basin and the Chioggia inlet. As a consequence, we estimated limited contaminant transfer to the Adriatic Sea, trapping the majority of contaminants in the sediment in this “average” circulation scenario which does not account for periodic flooding events.
