Pulp and paper mill wastewater (PPMWW) contains high concentrations of recalcitrant compounds that cause toxicity to organisms. Advanced oxidation processes (AOPs) have the ability to degrade these compounds and reduce overall toxicity. Physicochemical characterization and Lactuca sativa toxicity test were conducted to compare the effectiveness of two post-treatments: UV/H 2 O 2 and photo-Fenton. A comparison of four phytotoxicity indexes was carried out. PPMWW from a Brazilian treatment plant was characterized by high values of phenols, color, integrated spectral area (ISA), and chemical oxygen demand (COD), and caused significant inhibition to seedling development. The use of both post-treatments allowed the removal of over 75% of phenols, color, ISA, and COD. Although UV/H 2 O 2 was more effective in removing phenols and ISA, photo-Fenton better reduced phytotoxicity. The most sensitive phytotoxicity indexes were RGIC 0.8 and GIC 80% , whereas SGC 0 , REC -0.25 and REC -0.50 better showed the effectiveness of the post-treatments. We suggest the combined use of two phytotoxicity indexes: one that evaluates the effects on seed germination and, another, on root elongation, e.g., SGC 0 and RGIC 0.8 . Additionally, we recommend the use of ISA for monitoring programs of wastewater treatments because it is a cost-effective approach that allows narrowing down the search and identification of compounds present in complex mixtures., (Copyright © 2020 Elsevier Inc. This study investigates whether pulse exposure to Chilean pulp and paper mill effluent solid phase extracted (SPE) extracts via intraperitoneal injection (IP), would result in changes in the activities of the respiratory metabolic enzymes citrate synthase (CS) and lactate dehydrogenase (LDH) in rainbow trout livers. It also investigated if an alteration in liver metabolic capacity influenced the liver detoxification processes and estrogenic effects previously reported. Besides, a comparison of those enzymatic activities with fish IP injected with SPE extracts of two model effluents coming from industries that process 100% different type of feedstock (softwood, SW and hardwood, HW) was also evaluated. An initial induction of the anaerobic metabolism (increase in LDH enzymatic activity) was detected in all Chilean pulp mill effluent extracts evaluated, contrary to the initial unaltered aerobic metabolism (CS enzymatic activity) observed. A compensatory relationship in energy metabolism (Pasteur effect) was observed when comparing both enzymatic activities of fish exposed to those effluent extracts. LDH and CS activities observed in fish injected with Chilean extracts seem to be related to the effects observed in fish injected with SW extracts. This study showed that intraperitoneal injection of pulp and paper mill effluent extracts affected the anaerobic and aerobic metabolic capacities in rainbow trout livers, but this metabolic alteration did not affect detoxification capability or estrogenic effect previously reported., (Copyright © 2010 Elsevier Inc. Two experiments using rainbow trout (Oncorhynchus mykiss) were conducted to examine the combined effects of energy intake as manipulated by ration and pulp and paper mill effluent exposure over either one, or two consecutive reproductive cycles. This study demonstrated that the level of energy intake affected the full range of measured parameters from energy allocation to somatic growth and the gonadal development, steroid production and hematology. Increasing ration level expectedly increased growth, condition, liver and gonad size. Female trout in the higher ration treatments produced more follicles and had larger eggs, investing the same relative proportion of total energy into ovarian development. Sex steroid levels and hematological parameters were also positively influenced by increasing ration level in males and females. By far, the most dramatic impact of reduced ration on reproduction was to substantially reduce the frequency of sexually maturing fish. The effects of effluent exposure were not as marked as those linked to ration level and typically did not manifest unless fish were exposed through two consecutive reproductive cycles. The physiological effects of pulp and paper effluent exposure observed in these experiments were not consistent between the two experiments conducted herein, nor were they consistent with previously observed impacts in similar experiments with this effluent. Effluent exposure over one reproductive cycle did not impact physiological parameters in trout. However, when effluent exposure was maintained over two reproductive cycles, a new pattern of effluent response emerged including increased condition factor in both sexes, a decrease in the potential ability of the blood of females to transport oxygen, and increased sex steroids and reproductive investment in males. Effluent was also observed to cause reduced growth in male trout over two years. The effects of ration on gonad and liver size were far more obvious and consistent when a longer exposure was employed, thus, it appears to take more than one full year for energy intake changes to be reflected in those particular physiological endpoints.

Experiments were conducted to examine the effects of a thermomechanical (TMP)/bleached kraft pulp and paper mill effluent (BKME), dehydroabietic acid (DHAA), hypoxia, and combinations of hypoxia and effluent on juvenile rainbow trout. In the first two experiments, trout were exposed for 4 weeks to 0%, 10%, 30%, and 70% TMP/BKME or 0, 35, 110, and 250 microgL(-1) DHAA, respectively. Endpoints of those dose-response studies included critical swimming speed, oxygen consumption, and hematology. Reduced swimming performance was found for fish exposed to 70% TMP/BKME. Moderate increases in mean cell hemoglobin concentration at 70% TMP/BKME and blood glucose at 30% and 70% TMP/BKME were also seen. The opposite trend for glucose was found for DHAA-exposed fish, where a slight decrease in glucose was seen at 110 and 250 microgL(-1) DHAA. The third experiment examined the effects of 15% v/v TMP/BKME exposure at 2.5 and 5.0 mgL(-1) dissolved oxygen (DO) for 4 weeks. This experiment found no effect of low DO on swimming ability. An interactive effect between DO and effluent exposure was seen only on hematocrit, where effluent caused an increase in hematocrit at 5 mgL(-1) and a decrease at 2.5 mgL(-1) DO. Effluent exposure in this experiment resulted in a greater number of smaller red blood cells. The current study demonstrated physiological effects in rainbow trout exposed to varying concentrations (15-70% v/v) of a TMP/BKME and no substantial effects of DHAA exposure. With the exception of the reduced swimming performance in fish exposed to TMP/BKME, the observed effects are considered relatively small in magnitude but are occurring at concentrations of effluent that occur in the receiving environment.

Aquatic habitat is affected by paper mill effluent discharge in many ways. The effect of paper mill effluent on the gill ATPases was studied in freshwater fish Channa punctata (Bloch) exposed to 1%(v/v) of effluent for 15, 30, and 60 days. There was a time-dependent significant (P<0.05-0.001) decrease in all the ATPase activities measured, viz., total, Na(+), K(+)--and ouabain-insensitive ATPase in gill. ATPases play an important role in maintenance of functional integrity of plasma membrane and in several intracellular functions and are considered to be a sensitive indicator of toxicity. In addition to this, branchial ATPases are intimately involved in osmoregulation, acid-base regulation, and respiration of fish. The inhibition of ATPases in gills by, e.g., paper mill effluent could cause disruption of these processes. It is suggested that measurement of ATPases could also be used as a surrogate biomarker of exposure to chemical pollutants.

In order to validate previous field observations by the authors on whitefish, Coregonus lavaretus L. s.l., a 30-day laboratory experiment with concentrations (0, 1.3, 2.3, 3.5, and 7 vol%) of bleached kraft pulp and paper mill effluent (BKME) simulating those occurring in a polluted lake was conducted. Chlorine dioxide had almost entirely replaced chlorine gas in the bleaching of pulp. As a consequence, the concentrations of adsorbable organic halogens and chlorinated phenolics (CPs) in BKME were significantly lowered compared to earlier studies. This reduction was also seen in the concentrations of CPs in the bile and CPs and extractable organic halogens in the intestinal lipids: the concentrations were low and did not depend on the dilution of BKME. In contrast, the resin acid content of bile decreased with decreasing BKME concentration. The growth of fish was speeded up in all BKME concentrations. However, at the highest BKME concentration (7 vol%) the increase was lowest. The induction of hepatic ethoxyresorufin O-deethylase (EROD) activity revealed strong dose-response relationship with BKME. At 3.5 vol% BKME (corresponding to a distance of 3.3 km from the mill sewer in the field) the EROD activity increased 12-fold. There was a tendency for lower activity of uridinediphosphate glucuronosyltransferase in the liver, but the decrease (34%; P < 0.05) was statistically significant only at 7 vol% BKME. The activity of liver glutathione S-transferase remained unchanged. All dilutions of BKME significantly depressed the concentrations of plasma immunoglobulin M (IgM). Erythrocytic concentrations of nucleotide triphosphates decreased and of sodium increased as the BKME concentration increased. Also some other blood parameters (hematocrit, hemoglobin, plasma glucose, and aspartate aminotransferase) were changed in all BKME exposures, although without obvious dependence on effluent concentration. In conclusion, there was a good agreement between field studies and laboratory experiments using BKME concentrations occurring in the field, confirming close or similar causes for responsive toxicity endpoints.

Since 1988, the State of Maine has monitored dioxin levels in the tissues of predatory and bottom feeding species of fish taken from the state's major water bodies, including five rivers which receive the effluent of bleached kraft paper mills. Data are also available on dioxin concentrations in the effluent of these mills. Monitoring results were analyzed using simple models to yield field bioaccumulation factors (BAFs) for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in Maine rivers. Three BAFs were computed for each fish species at each site to account for declining TCDD levels in mill effluents over time. Mean BAFs (across rivers) ranged from 11,500 to 24,600 for smallmouth bass fillets, from 17,900 to 28,300 for brown trout fillets, from 3000 to 7500 for white perch fillets, and from 78,500 to 106,000 for white sucker whole bodies. Field BAFs obtained in this analysis are similar to factors obtained in recent laboratory studies of TCDD accumulation in fish. Our results suggest that EPA's currently recommended BCF value of 5000 is probably too low. Instead, a value between 15,000 and 25,000 is considered a reasonable estimate of the BAF to use for regulatory purposes in the state of Maine.

Toxic effects of unbleached (sulfate or sulfite) and bleached (sulfate) paper mill effluents were studied in a primary culture of rainbow trout liver cells. The effluents and control water from a clean area were extracted with diethyl ether and added to the cultures dissolved in dimethyl sulfoxide. Plasma membrane integrity was studied by measuring lactate dehydrogenase (LDH) leakage. The cellular content of glutathione (GSH) was used as an indicator of oxidative stress and the formation of reactive intermediates. Dose-response studies indicated that unbleached effluents contained more potent toxic substances than bleached effluents. Both unbleached and bleached effluents contained organic diethyl ether-extractable substances which increased cytochrome P450-dependent 7-ethoxyresorufin-O-deethylase (EROD) activities. The inducing effects were seen at concentrations substantially lower than those decreasing GSH content and increasing LDH leakage. Possible EROD inducing substances in bleached effluents are chlorinated organic compounds. Inducing compounds in unbleached effluents are yet to be identified. Furthermore, at higher concentrations the effluents contained substances that inhibited the cytochrome P450 system. The results show that the trout primary hepatocyte cultures afford a convenient in vitro method for screening cytochrome P450 inducing components extracted from industrial effluents to investigate mechanisms by which wastewaters cause injury in cells.

In order to study immunomodulation, controlled laboratory experiments were carried out with roach (Rutilus rutilus) exposed to bleached kraft mill effluent (BKME) or spent bleach liquor (SBL) from two pulp and paper mills using elementary chlorine and chlorine dioxide for bleaching. The total number of immunoglobulin-secreting cells (ISC) and the number of specific antibody-secreting cells (ASC) were determined by ELISPOT assay. Exposure to BKME resulted in decreased numbers of ISC in the spleen. To study the capability of response against foreign antigens the fish were immunized with bovine gamma-globulin. Exposure to BKME or SBL reduced antigen-specific ASC response in fish. Moreover, the suppression of ASC response did not require exposure prior to immunization. Depuration of fish in clean water reveals that the immunosuppression caused by BKME is reversible. The ASC response in the BKME-pretreated fish returned to the same or even a higher level compared to untreated immunized fish and, in addition, the number of ISC increased greatly. The results of this study verify an earlier finding on poor antibody response due to exposure in a lake contaminated with BKME. Exposure to SBL alone suppresses antibody-mediated immunity, suggesting that compounds formed in bleaching are at least partly responsible for immunotoxic effects.

Roach (Rutilus rutilus), from a unpolluted lake, were caged in a lake receiving treated waste waters from a pulp and paper mill 16 km upstream, and simultaneously a group of fish were held in cages in the unpolluted reference lake. The level of serum immunoglobulin of the fish kept in cages in these lakes was quantified. Serum immunoglobulin levels decreased when the fish were transferred to the contaminated lake and remained significantly reduced for the 8-week period fish were monitored. In another experiment fish were immunized with bovine gamma-globulin (BGG) after 5 weeks caging in the lakes. The kinetics of specific antibody synthesis were determined by assaying the number of antibody secreting cells in the spleen using enzyme-linked immunospot assay and by measuring the concentration of specific antibody in the serum. The peak response of the effluent exposed fish against BGG was lower than that for fish kept in the unpolluted reference lake, and the antibody titers, due to immunization, increased more slowly in the exposed fish. These results demonstrate that subchronic exposure of fish in a lake receiving treated waste waters from a bleached kraft mill affects the antibody-mediated immunity resulting in, besides lower levels of Ig, weak responsiveness against antigen.

Hepatic monooxygenase (MO) and conjugation enzyme activities, metabolites of chlorinated phenolics in the bile, and blood ionoregulatory parameters were studied in juvenile whitefish (Coregonus muksun Pallas and C. muksun X Coregonus peled Gmelin hybrid) held in cages downstream from a mill producing chlorine-bleached kraft pulp and printing paper. MO activities, measured as benzo[a]pyrene hydroxylase, 7-ethoxycoumarin O-deethylase, and 7-ethoxyresorufin O-deethylase, were significantly induced in whitefish caged about 5 km from the effluent outlet. The highest mean increase detected was 17 times the control value. In the nearest caging station (3 km) the induction was lower, indicating inhibition or toxicity caused by the effluent. The levels of bile metabolites of chlorinated phenolics showed highest concentrations at the nearest station and decreased levels at more distant locations over the whole water area studied (15 km). Bile metabolites in whitefish exposed in control areas confirmed low-level background pollution of the lake system due to chlorinated phenolics. Observations on blood ionic concentrations suggest that whitefish were able to regulate their hydromineral balance despite the environmental pollution affecting physiology of biotransformation.

Examining the concentration and types of airborne bacteria in waste paper and cardboard sorting plants (WPCSP) is an urgent matter to inform policy makers about the health impacts on exposed workers. Herein, we collected 20 samples at 9 points of a WPCSP every 6 winter days, and found that the most abundant airborne bacteria were positively and negatively correlated to relative humidity and temperature, respectively. The most abundant airborne bacteria (in units of CFU m−3) were: Staphylococcus sp. (72.4) > Micrococcus sp. (52.2) > Bacillus sp. (30.3) > Enterococcus sp. (24.0) > Serratia marcescens (20.1) > E. coli (19.1) > Pseudomonas sp. (16.0) > Nocardia sp. (1.9). The lifetime average daily dose (LADD) for the inhalation and dermal routes for the intake of airborne bacteria ranged from 3.7 × 10−3 ≤ LADDInhalation ≤ 2.07 × 101 CFU (kg d)−1 and 4.75 × 10−6 ≤ LADDDermal ≤ 1.64 × 10−5 CFU (kg d)−1, respectively. Based on a sensitivity analysis (SA), the concentration of airborne bacteria (C) and the exposure duration (ED) had the most effect on the LADDInhalation and LADDDermal for all sampling locations. Although the Hazard Quotient of airborne bacteria was HQ 2), indicating worker’s exposure to an infected environment. Therefore, in the absence of sufficient natural ventilation the indoor ambient conditions of the WPCSP studied should be controlled by supplying mechanical ventilation.

The toxicity of pulp and paper mill effluents was estimated by exposing rainbow trout to the effluents in situ during four seasons. The experiments were carried out using one control group and three test groups at different distances from the discharge. After a 10-day exposure mainly hematological parameters were determined from the caged fish. The caging site affected most of the 15 parameters analyzed (blood Hct, MCHC, erythrocyte, and leucocyte counts; plasma gluc, prot, urea, Ca2+, Mg2+). Some changes were detected even at the farthest caging site, 8 km from the discharge. The time of year strongly influenced the results. Seasonal variation exceeded the variation caused by caging site. In addition, the season influenced the responses of rainbow trout to the effluent in many parameters. None of the responses detected were observed in all the experiments. The only significant changes in the median values of the parameters from the caging experiments were a decrease in the plasma Mg2+ and an increase in the plasma K+ concentration.

A number of high molecular weight (HMW) lignin derivatives possessing varied chemical properties were screened for their biological effects in order to obtain more information on the possible structural features of HMW lignin-related effects. The studied compounds were both commercial and in-house extracted lignin derivatives. Bioassays used include reverse electron transport (RET), Vibrio fischeri, Daphnia magna, and juvenile rainbow trout (Oncorhynchus mykiss) hepatocytes. The studied lignin derivatives inhibited the in vitro systems and luminescence of V. fischeri bacteria to some extent-daphnids were not affected. It seems that, at least in the RET assay, certain pH-dependent functional groups in lignin may be of importance regarding the biological effects., (Copyright © 2010. Published by Elsevier Inc.)

Epidermal papillomatosis in fish has been proposed as an indicator of environmental stress but experimental evidence of connection between contaminants and papillomatosis in fish is scarce. We studied changes in the intensity of epidermal papillomatosis and the expression of heat shock protein 70 (HSP70) in roach, Rutilus rutilus, exposed to treated pulp mill and municipal effluents. In male roach, the increase in papillomatosis intensity was higher in fish exposed to 15% than in fish exposed to 1.5% concentration of municipal effluent. No differences were observed in papillomatosis development in females, or in HSP70 expression. In all the experiments conducted, the increasing effect of effluents seemed to be more pronounced in male fish suggesting that sex-related factors affected the intensity of papillomatosis after exposure to effluents. The present results indicate that environmentally relevant concentrations of municipal effluents may be contributing to the development of papillomatosis in fish.

Anadramous Atlantic salmon (Salmo salar L.) returning to Exploits River, Newfoundland, Canada, to spawn encounter low concentrations of thermomechanical pulp (TMP) effluent as they migrate upstream past an integrated newsprint mill. Various physiological responses of adult Atlantic salmon from the Exploits River were studied under laboratory conditions. The effects of a 6-h exposure to 0%, 6%, 12%, or 25% (v/v) TMP effluent or an increasing concentration gradient of effluent (0-25%) on cardiac output (Q ), critical swimming performance (U(crit)), hematocrit, and blood glucose, cortisol, lactate, and osmolality were examined. Relative to other treatment groups, Q during routine and low-level activity was 7-10% higher in fish exposed to at least 12% effluent. The 25% exposure group had a distinctly lower U(crit) and scope for increase in Q than the other treatment groups. These findings suggest that effluent exposure elevates physiological maintenance and repair costs, resulting in a reallocation of energetic resources.

Potential masculinization, detoxification, and oxidative stress responses were assessed in domesticated female guppies (Poecilia reticulata) exposed for 42 days to diluted effluent from a modern Swedish kraft pulp mill or a model androgen. Methyltestosterone induced male-like coloration and transformation of the anal fin into a gonopodium-like structure. The effluent did not induce any apparent changes of the anal fin morphology; however, the exposed guppies became more colored than control fish, which could be an androgenic response. A better understanding of the physiological mechanisms involved in these responses would be required for a full evaluation. Both primary effluent and effluent which had undergone activated sludge treatment caused a moderate but significant induction of hepatic ethoxyresorufin-O-deethylase (EROD) activity. However, the general toxicity of both effluents was low, as mortality was negligible even at 25% dilutions. There was a continuous production of offspring in all groups (47-62% female fry), except by methyltestosterone-treated females, which did not reproduce. There were no indications that either effluent caused oxidative stress since hepatic glutathione reductase, glutathione S-transferase and DT-diaphorase activities remained unchanged compared with controls.

Juvenile female rainbow trout was exposed for 4.5 months (June to October) to two dilutions of untreated and activated sludge treated whole mill effluent from a pulp mill producing bleached ECF pulp. Two controls were used, on fed ad libitum and a second receiving 0.5% feed of the body weight. All effluent exposed groups were fed ad libitum. Mean weight of the fish was measured monthly. At the end of the experiment a number of physiological and biochemical parameters were analyzed in order to establish the physiological status of the exposed fish in comparison with unexposed fish that obtained ad libitum or restricted amount of feed. The fish exposed to treated effluent grew significantly more than ad libitum control fish until August, whereupon growth retarded in fish exposed to the lower effluent dilution (400 v/v). The growth of fish exposed to untreated effluent did not deviate significantly from the control fed ad libitum. The results from the hematological analysis clearly showed that fish fed restricted amount of feed deviated significantly in most parameters compared with the control fed ad libitum. Fish exposed to treated effluent showed a response pattern similar to that of the control fed restricted amount of feed, whereas the fish exposed to untreated effluent showed a response pattern that did not deviate from that of the ad libitum control. The metabolic parameters suggested that fish exposed to treated effluent had a higher metabolic demand than ad libitum control and that the energy allocation at the end of the experiment was directed to processes other than growth. The responses on hematology were mainly a consequence of the increased energy demand and were not primary effects.
