The insula is important for the processing of pleasant aspects of touch whereas its role in the processing of emotionally neutral touch has been less explored. Here, we used a network approach to investigate the insular processing of pleasant stroking touch and emotionally neutral vibratory touch, analysing functional magnetic resonance imaging data from 23 healthy adult participants. Vibration and skin stroking activated areas in the posterior, middle and anterior insula. Psychophysiological interaction analyses suggested that skin stroking increased functional connectivity between the posterior and ventral anterior insula. Vibration instead increased functional connectivity between the posterior and dorsal anterior insula, and induced a stronger decrease of the default mode network activity compared to stroking. These results confirmed findings from previous studies showing that the posterior insula processes affective touch information. We suggest that this is accomplished by relaying tactile information from the posterior insula to ventral anterior insula, an area tightly connected to the emotional parts of the brain. However, our results also suggested that the insula processes tactile information with less emotional valence. A central hub in this processing seemed to be the right dorsal anterior insula. Highlights • Depressed mothers engage in less touching that is playful and interactive in nature. • Depressed mothers touch their infants less during face-to-face interactions. • Nondepressed mothers maintain high levels of touch following a perturbation period. • Depressed mothers decrease playful/stimulating touch after a perturbation period. • Implications for intervention programs of early touch stimulation for at-risk dyads., Touch is a critical channel of communication used by mothers to communicate and interact with their infants and to contribute to their infants’ socio-emotional development. The present study examined maternal touching in 41 mothers with and without depressive symptomatology. Mothers and their 4-month-old infants participated in the Still-Face (maternal emotional unavailability) and Separation (maternal physical unavailability) procedures. Maternal touching behaviours were video-recorded and coded using the Caregiver Infant Touch Scale (CITS). Results indicated that mothers with higher levels of depressive symptoms engaged in less touching following the perturbation period in the Still-Face procedure, whereas mothers with lower levels of depressive symptoms maintained stable levels of touching across both interaction periods. Mothers with higher levels of depressive symptoms displayed less playful/stimulating types of touching. Taken together, these results underscore the importance of touch and suggest key differences in touching behaviour between dyads with maternal depressive symptomatology and those without.

Increasing evidence shows that maternal touch may promote emotion regulation in infants, however less is known about how parental higher-order social cognition abilities are translated into tactile, affect-regulatory behaviours towards their infants. During 10 min book-reading, mother-infant sessions when infants were 12 months old (N = 45), we investigated maternal mind-mindedness (MM), the social cognitive ability to understand an infant’s mental state, by coding the contingency of maternal verbal statements towards the infants’ needs and desires. We also rated spontaneous tactile interactions in terms of their emotional contingency. We found that frequent non-attuned mind-related comments were associated with touch behaviours that were not contingent with the infant’s emotions; ultimately discouraging affective tactile responses from the infant. However, comments that were more appropriate to infant’s mental states did not necessarily predict more emotionally-contingent tactile behaviours. These findings suggest that when parental high-order social cognitive abilities are compromised, they are also likely to translate into inappropriate, tactile attempts to regulate infant’s emotions. The neural mechanisms facilitating the experience of vicarious social touch are largely unknown. The right inferior frontal gyrus (rIFG) has been suggested as part of a simulation observation-execution neural network that plays a key role in the perception of tactile stimuli. Considering that vicarious social touch involves vicarious sharing of emotions, we hypothesized that emotional empathy, i.e., the ability to feel what another individual is feeling, modulates the neural responses to vicarious touch. To examine the role of the rIFG in vicarious touch and its modulation by levels of emotional empathy, we used anodal transcranial direct current stimulation (tDCS) on forty participants who observed photos depicting social touch, nonsocial touch or no touch during tDCS or sham stimulation. The results show that while participants with high levels of emotional empathy exhibited no change in ratings of vicarious social touch, participants with low levels of emotional empathy rate human touch as more emotional following anodal stimulation of the rIFG than following sham stimulation. These findings indicate that emotional responses to vicarious social touch are associated with rIFG activity and are modulated by levels of emotional empathy. This result has major therapeutic potential for individuals with low empathic abilities, such as those with ASD. Touch provides more than sensory input for discrimination of what is on the skin. From early in development it has a rewarding and motivational value, which may reflect an evolutionary mechanism that promotes learning and affiliative bonding. In the present study we investigated whether affective touch helps infants tune to social signals, such as faces. Four-month-old infants were habituated to an individual face with averted gaze, which typically does not engage infants to the same extent as direct gaze does. As in a previous study, in the absence of touch, infants did not learn the identity of this face. Critically, 4-month-old infants did learn to discriminate this face when parents provided gentle stroking, but they did not when they experienced a non-social tactile stimulation. A preliminary follow-up eye-tracking study (Supplementary material) revealed no significant difference in the visual scanning of faces between touch and no-touch conditions, suggesting that affective touch may not affect the distribution of visual attention, but that it may promote more efficient learning of facial information. Highlights • The sense of touch enables parent-child communication from birth onward. • We theorized that parental touch would reduce children’s social vigilance. • Parental touch reduced children’s attention to social threat. • Parental touch increased trust, specifically among socially anxious children. • These effects occurred before (not after) the transition to adolescence., The sense of touch develops in utero and enables parent-child communication from the earliest moments of life. Research shows that parental touch (e.g., licking and grooming in rats, skin-to-skin care in humans) has organizing effects on the offspring’s stress system. Little is known, however, about the psychological effects of parental touch. Building on findings from ethology and psychology, we propose that parental touch—even as subtle as a touch on the shoulder—tells children that their environment is safe for exploration, thus reducing their social vigilance. We tested this hypothesis in late childhood (ages 8–10) and early adolescence (ages 11–14) in 138 parent-child dyads. Parents were randomly assigned to touch or not touch their child briefly and gently on the shoulder, right below the deltoid. Parental touch lowered children’s implicit attention to social threat. While parental touch lowered trust among socially non-anxious children, it raised trust among those who needed it the most: socially anxious children. The effects were observed only in late childhood, suggesting that parental touch loses its safety-signaling meaning upon the transition to adolescence. Our findings underscore the power of parental touch in childhood, especially for children who suffer from social anxiety.

Highlights • The infant brain is sensitive to gentle skin stroking within the first weeks of age. • The postcentral gyrus and posterior insular cortex are responsive to stroking. • Social touch activates both somatosensory and socio-affective brain areas in infancy., Physical expressions of affection play a foundational role in early brain development, but the neural correlates of affective touch processing in infancy remain unclear. We examined brain responses to gentle skin stroking, a type of tactile stimulus associated with affectionate touch, in young infants. Thirteen term-born infants aged 11–36 days, recruited through the FinnBrain Birth Cohort Study, were included in the study. Soft brush strokes, which activate brain regions linked to somatosensory as well as socio-affective processing in children and adults, were applied to the skin of the right leg during functional magnetic resonance imaging. We examined infant brain responses in two regions-of-interest (ROIs) known to process gentle skin stroking – the postcentral gyrus and posterior insular cortex – and found significant responses in both ROIs. These results suggest that the neonate brain is responsive to gentle skin stroking within the first weeks of age, and that regions linked to primary somatosensory as well as socio-affective processing are activated. Our findings support the notion that social touch may play an important role in early life sensory processing. Future research will elucidate the significance of these findings for human brain development.

Touch, such as a caress, can be interpreted as very pleasant. The emotional valence assigned to touch is likely related to certain bottom-up factors, such as optimal activation of C-tactile (CT) afferents. It is however unclear if besides somatosensory input, contextual factors related to the own body also play a role in the perceived pleasantness of touch. To test this, we manipulated visual appearance of the participant’s arm (veridical vision, no vision, pixelated moving statistic projected onto the arm (i.e. crawling skin)). We used slow velocity stroking (CT optimal stroking) with a soft brush to induce pleasant touch, and fast velocity stroking as a control condition. After each visual condition we asked participants (N = 23) to rate the emotional valence of the stroking they felt. After slow velocity stroking ratings on perceived pleasantness (but not on perceived unpleasantness) were modulated by visual condition, with veridical vision of the arm resulting in higher pleasantness ratings than both no vision and pixelated vision. We conclude that contextual processes affect the perceived pleasantness of touch. These findings shed a new light on the underlying mechanisms of how humans experience pleasant touch and show that pleasant touch not solely dependents on bottom up information. Recently, many workflows and tools that aim to increase the reproducibility and replicability of research findings have been suggested. In this review, we discuss the opportunities that these efforts offer for the field of developmental cognitive neuroscience. We focus on issues broadly related to statistical power and to flexibility and transparency in data analyses. Critical considerations relating to statistical power include challenges in recruitment and testing of young populations, how to increase the value of studies with small samples, and the opportunities and challenges related to working with large-scale datasets. Developmental studies also involve challenges such as choices about age groupings, modelling across the lifespan, the analyses of longitudinal changes, and neuroimaging data that can be processed and analyzed in a multitude of ways. Flexibility in data acquisition, analyses and description may thereby greatly impact results. We discuss methods for improving transparency in developmental cognitive neuroscience, and how preregistration of studies can improve methodological rigor in the field. While outlining challenges and issues that may arise before, during, and after data collection, solutions and resources are highlighted aiding to overcome some of these. Since the number of useful tools and techniques is ever-growing, we highlight the fact that many practices can be implemented stepwise.

Highlights • We tested the impact of early life stress (ELS) on age-white matter (WM) relations. • Higher ELS youth showed greater age-WM associations in several tracts. • These tracts connected frontal regions with limbic, occipital, and parietal regions. • Older youth with higher ELS showed a negative uncinate-internalizing association. • More mature uncinate morphometry may be adaptive in ELS-exposed adolescents., Early life stress (ELS) is associated with increased risk for internalizing disorders and variations in gray matter development. It is unclear, however, whether ELS affects normative age-related changes in white matter (WM) morphology, and if such maturational differences are associated with risk for internalizing psychopathology. We conducted comprehensive interviews in a cross-sectional sample of young adolescents (N = 156; 89 F; Ages 9–14) to assess lifetime exposure to stress and objective cumulative ELS severity. We used diffusion-weighted imaging to measure WM fixel-based morphometry and tested the effects of age and ELS on WM fiber density and cross-section (FDC), and associations between WM FDC and internalizing problems. Age was positively associated with FDC in all WM tracts; greater ELS severity was related to stronger age-WM associations in several association tracts connecting the frontal lobes with limbic, parietal, and occipital regions, including bilateral superior and inferior longitudinal and uncinate fasciculi (UF). Among older adolescents with greater ELS severity, a higher UF FDC was associated with fewer internalizing problems. Greater ELS severity predicted more mature WM morphometry in tracts implicated in emotion regulation and cognitive processing. More phenotypically mature UF WM may be adaptive against internalizing psychopathology in adolescents exposed to ELS.

Neuronal oscillations emerge in early human development. These periodic oscillations are thought to rapidly change in infancy and stabilize during maturity. Given their numerous connections to physiological and cognitive processes, understanding the trajectory of oscillatory development is important for understanding healthy human brain development. This understanding is complicated by recent evidence that assessment of periodic neuronal oscillations is confounded by aperiodic neuronal activity, an inherent feature of electrophysiological recordings. Recent cross-sectional evidence shows that this aperiodic signal progressively shifts from childhood through early adulthood, and from early adulthood into later life. None of these studies, however, have been performed in infants, nor have they been examined longitudinally. Here, we analyzed longitudinal non-invasive EEG data from 22 typically developing infants, ranging between 38 and 203 days old. We show that the progressive flattening of the EEG power spectrum begins in very early development, continuing through the first months of life. These results highlight the importance of separating the periodic and aperiodic neuronal signals, because the aperiodic signal can bias measurement of neuronal oscillations. Given the infrequent, bursting nature of oscillations in infants, we recommend using quantitative time domain approaches that isolate bursts and uncover changes in waveform properties of oscillatory bursts., Highlights • We assess oscillatory and aperiodic activity in longitudinal infant EEG recordings. • Infant EEG activity is predominantly of aperiodic nature. • The aperiodic exponent shows a strong decrease in the first half year of life. • We confirm a developmental increase in alpha-frequency of infant oscillatory bursts.

The prevalence of risky behavior such as substance use increases during adolescence; however, the neurobiological precursors to adolescent substance use remain unclear. Predictive modeling may complement previous work observing associations with known risk factors or substance use outcomes by developing generalizable models that predict early susceptibility. The aims of the current study were to identify and characterize behavioral and brain models of vulnerability to future substance use. Principal components analysis (PCA) of behavioral risk factors were used together with connectome-based predictive modeling (CPM) during rest and task-based functional imaging to generate predictive models in a large cohort of nine- and ten-year-olds enrolled in the Adolescent BrainCognitive Development (ABCD) study (NDA release 2.0.1). Dimensionality reduction (n = 9,437) of behavioral measures associated with substance use identified two latent dimensions that explained the largest amount of variance: risk-seeking (PC1; e.g., curiosity to try substances) and familial factors (PC2; e.g., family history of substance use disorder). Using cross-validated regularized regression in a subset of data (Year 1 Fast Track data; n1,500), functional connectivity during rest and task conditions (resting-state; monetary incentive delay task; stop signal task; emotional n-back task) significantly predicted individual differences in risk-seeking (PC1) in held-out participants (partial correlations between predicted and observed scores controlling for motion and number of frames [r

There has been a shift in the study of childhood adversity towards a focus on dimensions of adversity as opposed to a focus on cumulative risk or specific adversities. The Dimensional Model of Adversity and Psychopathology (DMAP) proposes deprivation and threat as core dimensions of childhood adversity. Previous work using DMAP has found links between deprivation and cognitive development and threat and emotional development in adolescence, but few studies have applied this framework to a poverty context, in which children are at heightened risk for adversity experiences, and none have examined outcomes in early childhood. We use data from the Family Life Project (n = 1,292) to examine deprivation and threat at child age 24 months as developmental mediators in the association between socioeconomic status (SES) measured at 15 months and executive functions (EF) measured at 48 months. In a multiple mediation model, lower SES was related to higher deprivation and threat. Deprivation was negatively associated with EF, and threat was not associated with EF. Deprivation fully mediated association between SES and EF. These results expand previous work using the DMAP and point to new directions in understanding children’s cognitive adaptations to adversity.

Characterizing typologies of childhood adversity may inform the development of risk profiles and corresponding interventions aimed at mitigating its lifelong consequences. A neurobiological grounding of these typologies requires systematic comparisons of neural structure and function among individuals with different exposure histories. Using seed-to-whole brain analyses, this study examined associations between childhood adversity and amygdala resting-state functional connectivity (rs-fc) in adolescents aged 11–19 years across three independent studies (N = 223; 127 adversity group) in both general and dimensional models of adversity (comparing abuse and neglect). In a general model, adversity was associated with altered amygdala rs-fc with clusters within the left anterior lateral prefrontal cortex. In a dimensional model, abuse was associated with altered amygdala rs-fc within the orbitofrontal cortex, dorsal precuneus, posterior cingulate cortex, and dorsal anterior cingulate cortex/anterior mid-cingulate cortex, as well as within the dorsal attention, visual, and somatomotor networks. Neglect was associated with altered amygdala rs-fc with the hippocampus, supplementary motor cortex, temporoparietal junction, and regions within the dorsal attention network. Both general and dimensional models revealed unique regions, potentially reflecting pathways by which distinct histories of adversity may influence adolescent behavior, cognition, and psychopathology.

Highlights • ERPs were used to compare multiplication fact processing in children and adults. • Different brain responses were elicited by children (N400) and adults (P300). • Results were directly compared to a language task (word-picture verification). • Access to arithmetic facts resembles language processing in children. • Adults interpret solutions as targets and categorize them efficiently., Multiplication tables are typically memorized verbally, with fluent retrieval leading to better performance in advanced math. Arithmetic development is characterized by strategy shifts from procedural operations to direct fact retrieval, which would not necessitate access to the facts’ conceptual meaning. This study tested this hypothesis using a combination of event related brain potentials (ERP) and behavioral measures with 3rd-5th grade children and young adults. Participants verified the solutions to simple multiplication problems (2 × 3 = 6 or = 7) and the semantic fit of word-picture pairs, separately. Children showed an N400 effect to multiplication solutions with larger (more negative) amplitude for incorrect than correct solutions, reflecting meaning-level processing. A similar ERP response was observed in the word-picture verification task, with larger negative amplitude for word-picture pairs that were semantically mismatched compared to matched. In contrast, adults showed a P300 response for correct solutions, suggesting that they treated these solutions as potential targets in over-rehearsed mathematical expressions. This P300 response was specific to math fact processing, as the word-picture verification task elicited a classic N400 in adults. These ERP findings reveal an overlooked developmental transition that occurs after fifth grade, and speak to theories of arithmetic that have been based primarily on adult data.

Phonological processing skills are known as the most robust cognitive predictor of reading ability. Therefore, the neural determinants of phonological processing have been extensively investigated by means of either neurofunctional or neurostructural techniques. However, to fully understand how the brain represents and processes phonological information, there is need for studies that combine both methods. The present study applies such a multimodal approach with the aim of investigating the pre-reading relation between neural measures of auditory temporal processing, white matter properties of the reading network and phonological processing skills. We administered auditory steady-state responses, diffusion-weighted MRI scans and phonological awareness tasks in 59 pre-readers. Our results demonstrate that a stronger rightward lateralization of syllable-rate (4 Hz) processing coheres with higher fractional anisotropy in the left fronto-temporoparietal arcuate fasciculus. Both neural features each in turn relate to better phonological processing skills. As such, the current study provides novel evidence for the existence of a pre-reading relation between functional measures of syllable-rate processing, structural organization of the arcuate fasciculus and cognitive precursors of reading development. Highlights • Previous studies have shown automatic vOT activation when processing spoken language. • vOT is organized from small onset to large rime grain size. • Higher fluency in 7−8 year olds only correlated with rhyme processing in anterior vOT. • Results suggest that these children have transitioned to larger grain size. • Exploratory analyses show reliance on phonology in STG for processing onsets., Previous studies have shown that reading skill in 3- to 6-year-old children is related to the automatic activation of the posterior left ventral occipitotemporal cortex (vOT) during spoken language processing, whereas 8- to 15-year-old children and adult readers activate the anterior vOT. However, it is unknown how children who are between these two age groups automatically activate orthographic representations in vOT for spoken language. In the current study, we recruited 153 7- to 8-year-old children to fill the age gap from previous studies. Using functional magnetic resonance imaging (fMRI), we measured children’s reading-related skills and brain activity during an auditory phonological task with both a small (i.e. onset) and a large (i.e. rhyme) grain size condition. We found that letter fluency, but not reading accuracy, was correlated with activation in the anterior vOT for the rhyme condition. There were no reading-related skill correlations for the posterior vOT or for activation during the onset condition in this age group. Our findings reveal that automatic activation in the anterior vOT during spoken language processing already occurs in higher skilled 7- to 8-year-old children. In addition, increases in naming automaticity is the primary determinant of the engagement of vOT during phonological awareness tasks.

Word learning is a significant milestone in language acquisition. The second year of life marks a period of dramatic advances in infants’ expressive and receptive word-processing abilities. Studies show that in adulthood, language processing is left-hemisphere dominant. However, adults learning a second language activate right-hemisphere brain functions. In infancy, acquisition of a first language involves recruitment of bilateral brain networks, and strong left-hemisphere dominance emerges by the third year. In the current study we focus on 14-month-old infants in the earliest stages of word learning using infant magnetoencephalography (MEG) brain imagining to characterize neural activity in response to familiar and unfamiliar words. Specifically, we examine the relationship between right-hemisphere brain responses and prospective measures of vocabulary growth. As expected, MEG source modeling revealed a broadly distributed network in frontal, temporal and parietal cortex that distinguished word classes between 150–900 ms after word onset. Importantly, brain activity in the right frontal cortex in response to familiar words was highly correlated with vocabulary growth at 18, 21, 24, and 27 months. Specifically, higher activation to familiar words in the 150–300 ms interval was associated with faster vocabulary growth, reflecting processing efficiency, whereas higher activation to familiar words in the 600–900 ms interval was associated with slower vocabulary growth, reflecting cognitive effort. These findings inform research and theory on the involvement of right frontal cortex in specific cognitive processes and individual differences related to attention that may play an important role in the development of left-lateralized word processing.

Highlights • Children and adolescents performed the Monetary Incentive Delay Task. • We examined anticipatory and outcome ERPs of reward and punishment processing. • SPN amplitudes for anticipating loss or no gain decreased with increasing age. • Sensitivity to negative outcomes decreases from childhood to adolescence., Reward and punishment processing are subject to substantial developmental changes during youth. However, little is known about the neurophysiological correlates that are associated with these developmental changes, particularly with regard to both anticipatory and outcome processing stages. Thus, the aim of this study was to address this research gap in a sample of typically developing children and adolescents. Fifty-four children and adolescents (8–18 years) performed a Monetary Incentive Delay Task comprising a monetary reward and punishment condition. Using event-related brain potential recordings, the cue-P3 and the stimulus-preceding negativity (SPN) were analyzed during the anticipation phase, while the Reward Positivity and the feedback-P3 were analyzed during the outcome phase. When anticipating monetary loss or no gain, SPN amplitude in the right hemisphere decreased with age. Moreover, exploratory analyses revealed a decrease in feedback-P3 amplitudes in response to monetary loss with increasing age. No other group differences were observed. Age-related changes in the SPN and fP3 component suggest that sensitivity to negative outcomes decreases from childhood to late adolescence, supporting the notion that adolescence is associated with reduced harm-avoidance. Longitudinal research including young adults is needed to substantiate our findings and its clinical implications regarding disturbed developmental trajectories in psychiatric populations.

There is growing interest in the role of stress in the connectivity between the amygdala and medial prefrontal cortex (mPFC), a circuit that subserves automatic emotion regulation. However, the specific timing and mechanisms that underlie the association between stress and amygdala–mPFC connectivity are unclear. Many factors, including variations in fetal exposure to maternal stress, appear to affect early developing brain circuitry. However, few studies have examined the associations of stress and amygdala–mPFC connectivity in early life, when the brain is most plastic and sensitive to environmental influence. In this longitudinal pilot study, we characterized the association between prenatal stress and amygdala–mPFC connectivity in young infants (approximately age 5 weeks). A final sample of 33 women who provided data on preconception and prenatal stress during their pregnancy returned with their offspring for a magnetic resonance imaging scan session, which enabled us to characterize amygdala–mPFC structural and functional connectivity as a function of prenatal stress. Increased prenatal stress was associated with decreased functional connectivity and increased structural connectivity between the amygdala and mPFC. These results provide insight into the influence of prenatal maternal stress on the early development of this critical regulatory circuitry.

Early childhood is an important period of sensory, motor, cognitive and socio-emotional maturation, yet relatively little is known about the brain changes specific to this period. Voxel-based morphometry (VBM) is a technique to estimate regional brain volumes from magnetic resonance (MR) images. The default VBM processing pipeline can be customized to increase accuracy of segmentation and normalization, yet the impact of customizations on analyses in young children are not clear. Here, we assessed the impact of different preprocessing steps on T1-weighted MR images from typically developing children in two separate cohorts. Data were processed with the Computational Anatomy Toolbox (CAT12), using seven different VBM pipelines with distinct combinations of tissue probability maps (TPMs) and DARTEL templates created using the Template-O-Matic, and CerebroMatic. The first cohort comprised female children aged 3.9-7.9 years (N = 62) and the second included boys and girls aged 2.7-8 years (N = 74). We found that pipelines differed significantly in their tendency to classify voxels as grey or white matter and the conclusions about some age effects were pipeline-dependent. Our study helps to both understand age-associations in grey and white matter volume across early childhood and elucidate the impact of VBM customization on brain volumes in this age range.

Highlights • fNIRS and looking responses to emotional faces were measured in 5- and 7-month-olds. • Emotional faces had varying identities within happy, angry, and fearful blocks. • Temporo-parietal and frontal activations were observed, particularly to happy faces. • Infants looked longer to the mouth region of angry faces. • No difference in behavior or neural activity observed between 5- and 7-month-olds., The processing of facial emotion is an important social skill that develops throughout infancy and early childhood. Here we investigate the neural underpinnings of the ability to process facial emotion across changes in facial identity in cross-sectional groups of 5- and 7-month-old infants. We simultaneously measured neural metabolic, behavioral, and autonomic responses to happy, fearful, and angry faces of different female models using functional near-infrared spectroscopy (fNIRS), eye-tracking, and heart rate measures. We observed significant neural activation to these facial emotions in a distributed set of frontal and temporal brain regions, and longer looking to the mouth region of angry faces compared to happy and fearful faces. No differences in looking behavior or neural activations were observed between 5- and 7-month-olds, although several exploratory, age-independent associations between neural activations and looking behavior were noted. Overall, these findings suggest more developmental stability than previously thought in responses to emotional facial expressions of varying identities between 5- and 7-months of age.

Psychosocial stress increases risky decision-making (DM). It is widely accepted that individual variation in neural phenotypes underlie variability in this behavioral tendency in adults, but is less examined in adolescents. Our goal was to test the hypothesis that the relation between neural phenotypes and stress-related risky DM is better characterized by individual variation than by age. Using diffusion tensor imaging (DTI) tractography to characterize the accumbofrontal tract, we determined if it uniquely moderated how stress affects risky DM, over and above age. A daily diary design monitored participants' daily stress for two weeks. Participants completed a DTI scan and performed a task in which decisions varied by expected value, once each on a day when they endorsed feeling higher (and lower) than usual levels of stress. Multilevel logistic regression analyses revealed that all participants were more likely to take risks as expected reward value increased; this behavior was greater under high versus low stress for individuals with low accumbofrontal tract integrity, whereas DM was less influenced by stress for individuals with high accumbofrontal tract integrity, regardless of age. Results suggest that individual differences in brain structure may be more germane to characterizing risky decisions in adolescents, rather than ontogeny.

Highlights • Test-retest reliability of structural MRI and DWI data was good. • Test-retest reliability of resting-state and task-related fMRI was moderate. • Overall, global brain measures are more reliable than local brain measures., The YOUth cohort study is a unique longitudinal study on brain development in the general population. As part of the YOUth study, 2000 children will be included at 8, 9 or 10 years of age and planned to return every three years during adolescence. Magnetic resonance imaging (MRI) brain scans are collected, including structural T1-weighted imaging, diffusion-weighted imaging (DWI), resting-state functional MRI and task-based functional MRI. Here, we provide a comprehensive report of the MR acquisition in YOUth Child & Adolescent including the test-retest reliability of brain measures derived from each type of scan. To measure test-retest reliability, 17 adults were scanned twice with a week between sessions using the full YOUth MRI protocol. Intraclass correlation coefficients were calculated to quantify reliability. Global brain measures derived from structural T1-weighted and DWI scans were reliable. Resting-state functional connectivity was moderately reliable, as well as functional brain measures for both the inhibition task (stop versus go) and the emotion task (face versus house). Our results complement previous studies by presenting reliability results of regional brain measures collected with different MRI modalities. YOUth facilitates data sharing and aims for reliable and high-quality data. Here we show that using the state-of-the art YOUth MRI protocol brain measures can be estimated reliably.

EEG is a widely used tool to study the infant brain and its relationship with behavior. As infants usually have small attention spans, move at free will, and do not respond to task instructions, attrition rates are usually high. Increasing our understanding of what influences data loss is therefore vital. The current paper examines external factors to data loss in a large-scale on-going longitudinal study (the YOUth project; 1279 five-month-olds, 1024 ten-months-olds, and 109 three-year-olds). Data loss is measured for both continuous EEG and ERP tasks as the percentage data loss after artifact removal. Our results point to a wide array of external factors that contribute to data loss, some related to the child (e.g., gender; age; head shape) and some related to experimental settings (e.g., choice of research assistant; time of day; season; and course of the experiment). Data loss was also more pronounced in the ERP experiment than in the EEG experiment. Finally, evidence was found for within-subject stability in data loss characteristics over multiple sessions. We end with recommendations to limit data loss in future studies.

Behaviors, traits and characteristics are transmitted from parents to offspring because of complex genetic and non-genetic processes. We review genetic and non-genetic mechanisms of intergenerational transmission of psychopathology and parenting and focus on recent methodological advances in disentangling genetic and non-genetic factors. In light of this review, we propose that future studies on intergenerational transmission should aim to disentangle genetic and non-genetic transmission, take a long-term longitudinal perspective, and focus on paternal and maternal intergenerational transmission. We present four large longitudinal cohort studies within the Consortium on Individual Development, which together address many of these methodological challenges. These four cohort studies aim to examine the extent to which genetic and non-genetic transmission from the parental generation shapes parenting behavior and psychopathology in the next generation, as well as the extent to which self-regulation and social competence mediate this transmission. Conjointly, these four cohorts provide a comprehensive approach to the study of intergenerational transmission.

This study is the first to systematically review and quantify the effects of physical activity on brain structure and neurophysiological functioning in children. Electronic data bases were searched for relevant studies. Studies that met the following criteria were included: (1) used an RCT or cross-over design, (2) examined the effects of physical activity on brain structure and/or neurophysiological functioning, (3) included children (5–12 years old) (4) included a control group (RCTs) or control condition (cross-over trials). A total of 26 and 20 studies were included in the systematic review and meta-analysis, respectively, representing and accompanying 973 and 782 unique children. Main analyses were separated for short-term and long-term physical activity and for effects on brain structure and neurophysiological functioning with a distinction between children from healthy and clinical populations. We found evidence for significant beneficial effects of long-term physical activity on neurophysiological functioning (d = 0.39, p < 0.001). In addition, short-term physical activity may induce changes in neurophysiological functioning (d = 0.32, p = 0.044), although this evidence showed limited robustness. No meta-analytic evidence was found for positive effects on brain structure. The results underline the importance of physical activity for brain development in children.

A key goal of human neurodevelopmental research is to map neural and behavioral trajectories across both health and disease. A growing number of developmental consortia have begun to address this gap by providing open access to cross-sectional and longitudinal 'big data' repositories. However, it remains challenging to develop models that enable prediction of both within-subject and between-subject neurodevelopmental variation. Here, we present a conceptual and analytical perspective of two essential ingredients for mapping neurodevelopmental trajectories: state and trait components of variance. We focus on mapping variation across a range of neural and behavioral measurements and consider concurrent alterations of state and trait variation across development. We present a quantitative framework for combining both state- and trait-specific sources of neurobehavioral variation across development. Specifically, we argue that non-linear mixed growth models that leverage state and trait components of variance and consider environmental factors are necessary to comprehensively map brain-behavior relationships. We discuss this framework in the context of mapping language neurodevelopmental changes in early childhood, with an emphasis on measures of functional connectivity and their reliability for establishing robust neurobehavioral relationships. The ultimate goal is to statistically unravel developmental trajectories of neurobehavioral relationships that involve a combination of individual differences and age-related changes.

Highlights • Infants were imaged at 1 month of age and attention was assessed at 6 months of age. • Infants with higher FA in several white matter tracts showed increased orienting. • Results indicate early neurodevelopment of pathways important in attention., Early infancy is characterized by rapid brain development that occurs alongside, and in response to, the development of cognitive and behavioral functions, including attention. Infants’ ability to orient and sustain attention to stimuli develops in concert with refinement of the orienting network in frontoparietal regions of the brain. Infants (n = 97) underwent magnetic resonance imaging at one-month of age and data were fit to a diffusion tensor imaging model to calculate fractional anisotropy (FA) and radial diffusivity (RD), as well as to a neurite orientation dispersion and density imaging model to calculate intracellular volume fraction (νic). Infant attention was assessed at six months of age using a dynamic puppet task (Cuevas and Bell, 2014). Infants with higher FA in the corpus callosum and anterior cingulum showed increased orienting behaviors. Our findings indicate that increased microstructure of the white matter tracts in the orienting network may play a role in the early neurodevelopment of attentional orienting behaviors.

In this study, we aimed to develop a behavioral task that measures pro-social decision making in rats. A fully automated, operant pro-social two-choice task is introduced that quantifies pro-social preferences for a mutual food reward in a set-up with tightly controlled task contingencies. Pairs of same-sex adult Wistar rats were placed in an operant chamber divided into two compartments (one rat per compartment), separated by a transparent barrier with holes that allowed the rats to see, hear, smell, but not touch each other. Test rats could earn a sucrose pellet either for themselves (own reward) or for themselves and the partner (both reward) by means of lever pressing. On average, male rats showed a 60 % preference for the lever that yielded a food reward for both themselves and their partner. In contrast, females did not show lever preference, regardless of the estrous cycle phase. Next, the impact of juvenile environmental factors on male rat social decision making was studied. Males were group-housed from postnatal day 26 onwards in complex housing Marlau (TM) cages that provided social and physical enrichment and stimulation in the form of novelty. Complex housed males did not show a preference for the pro-social lever.

Maternal odor is known to play an important role in mother-infant-interaction in many altricial species such as rodents. However, we only know very little about its role in early human development. The present study therefore investigated the impact of maternal odor on infant brain responses to emotional expression. We recorded the electroencephalographic (EEG) signal of seven-month-old infants watching happy and fearful faces. Infants in two control groups exposed to no specific odor (control 1) or the odor of a different infant’s mother (control 2) showed the expected EEG fear response. Crucially, this response was markedly absent in the experimental group exposed to their mother’s odor. Thus, infants respond differently to fear signals in the presence of maternal odor. Our data therefore suggest that maternal odor can be a strong modulator of social perception in human infants.

Research has yet to investigate trajectories of sensitivity to threat across childhood and adolescence. Further, neural associations of these trajectories remain unknown. The current 3-year study used a latent class growth curve analysis to investigate whether there were distinct trajectories of sensitivity to threat among children and adolescents over time (N = 363; age range at Time 1 = 8–14). We also examined whether alpha asymmetry (a neural index of motivational tendencies) was associated with the different trajectories. Results revealed three distinct trajectory groups (1) high-stable sensitivity to threat, (2) moderate-increasing sensitivity to threat and (3) low-stable sensitivity to threat. The high-stable sensitivity to threat group had greater right frontal asymmetry activation (i.e., greater neural avoidance motivation) than the other two groups. Additionally, females, those with higher parental education, and individuals with more advanced pubertal development (but not age) had greater odds of being part of the high-stable sensitivity to threat group compared to the other groups. Of interest, puberty rather than age may be an important indicator of heightened sensitivity to threat.

Anxiety is common in adolescence and has been linked to a plethora of negative outcomes across development. While previous studies of anxiety have focused on threat sensitivity, less work has considered the concurrent development of threat- and reward-related neural circuitry and how these circuits interact and compete during puberty to influence typical adolescent behaviors such as increased risk taking and exploration. The current review integrates relevant findings from clinical and developmental neuroimaging studies to paint a multidimensional picture of adolescent-onset anxiety against the backdrop of typical adolescent development. Ultimately, this paper argues that longitudinal neuroimaging studies tracking approach and avoidance motivations across development are needed to fully understand the mechanisms underlying the development of anxiety in adolescence and to identify and provide effective interventions for at-risk youth.

Highlights • Fibre properties derived from fixel-based analysis are sensitive to development. • Age-related patterns are consistent with previous diffusion tensor studies. • Pubertal stage correlates with fibre density and morphology in the right SLF. • Pubertal stage correlates with fibre morphology in the right ILF. • Pubertal progression interacts with age for longitudinal fibre density changes., The pubertal period involves dynamic white matter development. This period also corresponds with rapid gains in higher cognitive functions including attention, as well as increased risk of developing mental health difficulties. This longitudinal study comprised children aged 9–13 years (n = 130). Diffusion magnetic resonance imaging (dMRI) data were acquired (b = 2800s/mm2, 60 directions) at two time-points. We derived measures of fibre density and morphology using the fixel-based analysis framework and performed a tract-based mixed-effects modelling analysis to understand patterns of white matter development with respect to age, sex, pubertal stage, and the change in pubertal stage. We observed significant increases in apparent fibre density across a large number of white matter pathways, including major association and commissural pathways. We observed a linear relationship between pubertal stage and fibre density and morphology in the right superior longitudinal fasciculus, and fibre morphology in the right inferior longitudinal fasciculus. Finally, we report a significant interaction between the change in pubertal stage and age in the development of fibre density, for left-lateralised association tracts. Overall, white matter development across ages 9–13 years involves the expansion of major white matter fibre pathways, with key association pathways linked with pubertal stage.

Since the first neurodevelopmental models that sought to explain the influx of risky behaviors during adolescence were proposed, there have been a number of revisions, variations and criticisms. Despite providing a strong multi-disciplinary heuristic to explain the development of risk behavior, extant models have not yet reliably isolated neural systems that underlie risk behaviors in adolescence. To address this gap, we screened 2017 adolescents from an ongoing longitudinal study that assessed 15-health risk behaviors, targeting 104 adolescents (Age Range: 17-to-21.4), characterized as high-or-average/low risk-taking. Participants completed the Monetary Incentive Delay (MID) fMRI task, examining reward anticipation to “big win” versus “neutral”. We examined neural response variation associated with both baseline and longitudinal (multi-wave) risk classifications. Analyses included examination of a priori regions of interest (ROIs); and exploratory non-parametric, whole-brain analyses. Hypothesis-driven ROI analysis revealed no significant differences between high- and average/low-risk profiles using either baseline or multi-wave classification. Results of whole-brain analyses differed according to whether risk assessment was based on baseline or multi-wave data. Despite significant mean-level task activation, these results do not generalize prior neural substrates implicated in reward anticipation and adolescent risk-taking. Further, these data indicate that whole-brain differences may depend on how risk-behavior profiles are defined.

Highlights • We report longitudinal ERP data of 80 infants in a face-discrimination task. • P1, N290, Nc are all sensitive to faces in five-month-olds. • P1, N290, Nc show equal face-categorization in infants tested longitudinally. • N290 shows less variation in face-categorization trajectories than P1 or Nc. • Visual ERPs increase in amplitude over infancy, but this is not face-specific., From infancy onwards, EEG is widely used to measure face-categorization, i.e. differential brain activity to faces versus non-face stimuli. Four ERP components likely signal infants’ face-sensitivity but reflect different underlying mechanisms: the P1, N290, P400, Nc. We test whether these components reveal similar developmental patterns from early to late infancy, using a longitudinal dataset of 80 infants tested at 5 and 10 months. The P1, N290, and the Nc show face-categorization already in 5-months-olds, a pattern which did not change over time. Development is visible as increased amplitudes in all components, but similar for face and non-face stimuli. By using Markov models, we illustrate that there are differences in the distribution of individual trajectories of face-categorization components from 5 to 10 months. Whereas individual trajectories appear more varied for the Nc and the P1, the N290 reveals a more consistent pattern: a larger proportion of 5-month-olds shows the dominant group response; a larger proportion of 10-month-olds remains in this group, and larger proportions of the alternative trajectories from 5- to 10-month-olds move towards the dominant group. This is vital information when one wants to examine individual differences in infant ERPs related to face-categorization.

Anxiety is the most common form of psychopathology, and it is often characterized by chronic impairment across the lifespan. Researchers have identified core neural markers that confer risk for anxious outcomes. An increased error-related negativity (ERN) in anxious individuals has been shown to prospectively predict onset of anxiety disorders across development. Hence, it is critical to examine environmental factors that may shape the ERN. In the current study, we use a large sample of 170 female adolescents aged 10-17 to investigate whether the ERN mediates the relationship between parenting style and anxiety diagnostic status. This study replicates previous findings, and it extends previous work by suggesting that this relationship is more robust in young children as compared to adolescents. Interventions targeting the ERN via parenting may be most effective during childhood.

Highlights • Low peer preference was linked to higher brain activation during early social exclusion experiences. • Activation was found in an area implicated in emotion regulation and attentional control. • Results suggest that low peer preferred boys become sensitive to social exclusion. • Low peer preference seems to become embodied in children’s neurobiology., Peer preference among classmates is a highly influential factor in children’s social development and not being preferred by peers has long-term consequences for children’s developmental outcomes. However, little is known about how a history of low peer preference during primary school is associated with neural responses to a new social exclusion experience in childhood. In this functional magnetic resonance imaging (fMRI) study, we examined self-reported social distress and neural responses to social exclusion using the Cyberball paradigm in primary school boys (Mage = 10.40 years) with a history of low (n = 27) versus high peer preference (n = 28). Boys were selected from a longitudinal classroom-based study in which children’s peer social preferences were assessed in three consecutive years prior to this study. Neuroimaging results showed that low peer preferred boys exhibited increased activation in the lateral prefrontal cortex during early social exclusion relative to later social exclusion experiences as compared to high peer preferred boys. Increased neural activity was not accompanied by higher self-reported levels of social distress during social exclusion in low versus high peer preferred children. Findings of this study may provide insight into the neural processes associated with real-life peer experiences in children attending primary school.

Highlights • We examined efficiency of and adaptiveness in proactive control use as a function of contextual cue reliability in 6- and 9-year-olds and adults. • All age groups showed greater behavioural, ERP and pupil dilation markers of proactive task selection when cues were reliable. • Early developing use of contextual cue reliability ensures some degree of adaptiveness in proactive control engagement from 6 years onwards. • Only adults additionally engaged in motor preparation with reliable precues, suggesting refinement in proactive control efficiency with age., Age-related progress in cognitive control reflects more frequent engagement of proactive control during childhood. As proactive preparation for an upcoming task is adaptive only when the task can be reliably predicted, progress in proactive control engagement may rely on more efficient use of contextual cue reliability. Developmental progress may also reflect increasing efficiency in how proactive control is engaged, making this control mode more advantageous with age. To address these possibilities, 6-year-olds, 9-year-olds, and adults completed three versions of a cued task-switching paradigm in which contextual cue reliability was manipulated. When contextual cues were reliable (but not unreliable or uninformative), all age groups showed greater pupil dilation and a more pronounced (pre)cue-locked posterior positivity associated with faster response times, suggesting adaptive engagement of proactive task selection. However, adults additionally showed a larger contingent negative variation (CNV) predicting a further reduction in response times with reliable cues, suggesting motor preparation in adults but not children. Thus, early developing use of contextual cue reliability promotes adaptiveness in proactive control engagement from early childhood; yet, less efficient motor preparation in children makes this control mode overall less advantageous in childhood than adulthood.

Childhood adversity is heterogeneous with potentially distinct dimensions of violence exposure and social deprivation. These dimensions may differentially shape emotion-based neural circuitry, such as amygdala–prefrontal cortex (PFC) white matter connectivity. Amygdala–orbitofrontal cortex (OFC) white matter connectivity has been linked to regulation of the amygdala’s response to emotional stimuli. Using a preregistered analysis plan, we prospectively examined the effects of childhood exposure to two dimensions of adversity, violence exposure and social deprivation, on the adolescent amygdala–PFC white matter connectivity. We also reproduced the negative correlation between amygdala–PFC white matter connectivity and amygdala activation to threat faces. 183 15–17-year-olds were recruited from the Fragile Families and Child Wellbeing Study — a longitudinal, birth cohort, sample of predominantly low-income youth. Probabilistic tractography revealed that childhood violence exposure and social deprivation interacted to predict the probability of adolescent right hemisphere amygdala–OFC white matter connectivity. High violence exposure with high social deprivation related to less amygdala–OFC white matter connectivity. Violence exposure was not associated with white matter connectivity when social deprivation was at mean or low levels (i.e., relatively socially supportive contexts). Therefore, social deprivation may exacerbate the effects of childhood violence exposure on the development of white matter connections involved in emotion processing and regulation. Conversely, social support may buffer against them.

Children that have experienced psychosocial neglect display impairments in self-monitoring and controlling their behavior (cognitive control) and are at broad, transdiagnostic risk for psychopathology. However, the neural underpinnings of such effects remain unclear. Event-related mediofrontal theta oscillations reflect a neural process supporting cognitive control that may relate to transdiagnostic psychopathology risk. Recent work demonstrates reduced mediofrontal theta in rodent models of neglect; however, similar findings have not been reported in humans. Here, 136 children reared in Romanian institutions were randomly assigned to either a high-quality foster care intervention and placed with families or remained in institutions; 72 never-institutionalized children served as a comparison group. The intervention ended at 54 months; event-related mediofrontal theta and psychopathology were assessed at 12- and 16-year follow-up assessments. Institutional rearing (neglect) predicted reduced mediofrontal theta by age 16, which was linked to heightened transdiagnostic risk for psychopathology (P factor); no specific associations with internalizing/externalizing factors were present once transdiagnostic risk was accounted for. Earlier placement into foster care yielded greater mediofrontal activity by age 16. Moreover, foster care placement was associated with the developmental trajectory of mediofrontal theta across the adolescent period (ages 12-16), which was, in turn, associated with greater reductions in transdiagnostic risk across this same period. These data reflect the first experimental evidence that the development of mediofrontal theta is impacted by removal from situations of neglect in humans, and further characterizes the importance of studying developmental change in mediofrontal theta during the adolescent period.

Highlights • Reduced positive valence systems (PVS) function increases risk for psychopathology. • PVS function is shaped by genetics/heritability, temperament, parenting, and stress. • Individual factors have relatively modest and inconsistent effects. • There may be distinct developmental trajectories leading to reduced PVS function., Reduced activation of positive valence systems (PVS), including blunted neural and physiological responses to pleasant stimuli and rewards, has been shown to prospectively predict the development of psychopathology. Yet, little is known about how reduced PVS activation emerges across development or what implications it has for prevention. We review genetic, temperament, parenting, and naturalistic and laboratory stress research on neural measures of PVS and outline developmentally-informed models of trajectories of PVS activation. PVS function is partly heritable and appears to reflect individual differences in early-emerging temperament traits. Although lab-induced stressors blunt PVS activation, effects of parenting and naturalistic stress on PVS are mixed and depend on the type of stressor, developmental timing, and interactions amongst risk factors. We propose that there may be multiple, dynamic developmental trajectories to reduced PVS activation in which combinations of genes, temperament, and exposure to severe, prolonged, or uncontrollable stress may exert direct and interactive effects on PVS function. Critically, these risk factors may alter PVS developmental trajectories and/or PVS sensitivity to proximal stressors. Distinct factors may converge such that PVS activation proceeds along a typical, accelerated, chronically low, or stress-reactive trajectory. Finally, we present directions for future research with translational implications.

Highlights • A novel, cutting-edge measure for studying active learning in infancy is introduced: theta oscillations. • Theta rhythms are linked to expectation of information, memory, and reward processing, making it an ideal candidate for studying active learning. • Existing evidence and potential applications of theta oscillations in research on infant active learning are discussed., Active learning is a critical component of human development, however, the mechanisms supporting it are not fully understood. Given that early learning experiences may affect both infants' immediate learning success, as well as their motivation to learn, it is particularly important to investigate the mechanisms of active learning in this period, when the foundations of learning habits and curiosity are built. Traditional behavioural approaches of studying infant learning face challenges that emerging tools from neuroscience may help relieve. We introduce one such tool, EEG theta oscillations, and propose this neural marker has great potential for offering novel insights into active learning. Theta activity, recorded prior to or during learning, has been shown to be predictive of learning success. We argue that this involvement in memory formation, combined with theta activity’s tight association with reward processing, makes theta oscillations a uniquely suited tool for the investigation of motivational mechanisms underlying active learning. We outline research questions as well as methodological approaches pertinent to infant learning and suggest how and why theta oscillations may offer complementary insights. As such, we aim to bridge the gap between cognitive and neural approaches, and advance our knowledge of active learning in development more broadly.

Highlights • 18-month-old infants differentiate between legal and illegal phonotactics. • Infants show adult-like processing mechanisms in the electrophysiological signal. • Vascular responses do not differentiate between legal and illegal phonotactics. • Methodological and brain maturational aspects explain vascular responses., The present study investigated neural correlates of implicit phonotactic processing in 18-month-old children that just reached an important step in language development: the vocabulary spurt. Pseudowords, either phonotactically legal or illegal with respect to their native language, were acoustically presented to monolingually German raised infants. Neural activity was simultaneously assessed by means of electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS). The former method excellently tracks fast processing mechanisms, whereas the latter reveals brain areas recruited. Results of the present study indicate that 18-month-olds recognize the linguistic properties of their native language based on phonotactics. This manifested in an increased N400 for legal compared to illegal pseudowords in the EEG conforming to adult-like mechanisms. Unfortunately, fNIRS findings did not support this discrimination ability. Possible methodological and brain maturational reasons might explain this null finding. This study provides evidence for the advantage of a multi-methodological approach in order to get a clear picture on neural language development.
