A conjugal block phenotype is described for the Tetrahymena pattern mutant, janA. janA exhibits a characteristic "janus" phenotype in which cells develop with a global mirror-image duplication of the ventral pattern of cortical organelles. janA cells are competent to form mating pairs, but later become irreversibly fused as heteropolar doublets. The few pairs that successfully dissociate fail to undergo postconjugal oral replacement and perish. The janA conjugal block is 100% penetrant, is under prezygotic macronuclear control, and is lethal. Here we characterize this conjugal block genetically and cytologically and demonstrate that it can be rescued by a transferable, wild-type product. New insights into late conjugal events, especially the replacement of the oral apparatus, are reported for wild-type cells as well.

In Paramecium, the morphogenesis of the cortex at cell division, which assures reconstruction of shape and surface pattern, has been shown to involve transcellular signals which spread across the cortex like a wave, originating principally from the oral apparatus. One of the events these signals control is the reorganization of the ciliary rootlets through a cycle of regression and regrowth. The ciliary rootlets are nucleated on the ciliary basal bodies and form a scaffold extending over the entire cell surface that is important in aligning the basal bodies and the unit territories organized around them in longitudinal rows. We present evidence that the mechanism underlying their reorganization is cell-cycle-dependent phosphorylation of the structural proteins which compose the ciliary rootlets. We have isolated the rootlets and prepared a polyclonal antibody against them. In situ immunofluorescence of dividing cells with the anti rootlet antibody, and with the monoclonal antibody MPM-2 specific for phosphoproteins shows that a wave of phosphorylation of the ciliary rootlets spreads across the cell at division and just precedes their regression. Two-dimensional Western blot analysis of cytoskeleton and isolated rootlets along with alkaline phosphatase treatment demonstrates that the rootlets are composed of phosphoproteins, while experiments with interphase and dividing cells provide direct evidence that hyperphosphorylation of these proteins at division brings about disassembly of the structure.

Ciliated protozoa possess cellular axes reflected in the arrangement of their ciliature. Upon transverse fission, daughter cells develop an identical ciliary pattern, ensuring perpetuation of the cellular phenotype. Experimentally manipulated cells can be induced to form atypical phenotypes, capable of intraclonal propagation and regeneration after encystment. One such phenotype in the ciliate Tetmemena pustulata (formerly Stylonychia pustulata) is the mirror-imaged doublet. These cells possess two distinct sets of ciliature, juxtaposed on the surfaces in mirror image symmetry, with a common anterior–posterior axis. We have examined whether individual ciliary components of Tetmemena mirror-image doublets are mirror imaged. Ultrastructural analysis indicates that despite global mirror imaging of the ciliature, detailed organization of the membranelles is reversed in the mirror-image oral apparatus (OA), such that the ciliary effective stroke propels food away from the OA. Assembly of compound ciliary structures of both OAs starts out identically, but as the structures associated with the mirror-image OA continue to form, the new set of membranelles undergoes a 180° planar rotation on the ventral surface relative to the same structures in the typical OA. The overall symmetry of the OA thus appears to be separable from the more localized assembly of individual basal bodies. True mirror imagery of the membranelles would require new enantiomorphic forms of the individual ciliary components, particularly the basal bodies, which is never observed. These observations suggest a mechanistic hypothesis with implications for the development of left–right asymmetry not only in ciliates, but perhaps also in development of left–right asymmetry in general.

In Tetrahymena, two unique cell-surface structures, the oral apparatus and the cytoproct, are formed at opposite ends of one ciliary row, the reference meridian, which is propagated longitudinally during clonal growth. A third set of unique structures, the contractile vacuole pore(s) (CVP), is located at a nearly constant proportion of the cell circumference to the cell's right of the reference meridian. Three allelic recessive temperature-sensitive mutations, collectively named hypoangular (hpo), alter both the geometry of propagation of the reference meridian and the location of the CVPs. In mutant cells, the reference meridian typically undergoes a steady rightward shift in successive cell generations ("cortical slippage"); concomitantly, CVP sets come to lie closer to the reference meridian. Although CVP location is still proportional to the cell circumference, the constant of proportionality (the "CVP angle") is reduced. Another effect is an alteration in the widths of morphogenetic domains within the cortex. As the temperature is raised (made more restrictive), these effects are accentuated and the CVP angle becomes reduced further. At the extreme, the CVP angle collapses to zero and less, i.e., there is a topological switch such that CVPs come to lie to the left of the reference meridian, and the direction of cortical slippage reverses from rightward to leftward. These observations are hard to reconcile with existing formal models of pattern specification in this system and suggest that the hpo locus might specify a key component of the intracellular positional system.

Homopolar doublets of Tetrahymena thermophila which have two normal oral systems directly opposite one another may undergo a global transformation of cell surface geometry to create transient imitations of mirror-image configurations brought about by mutations at janus gene loci. The process by which a typical doublet transforms into a janus-like organization involves loss of capacity to form oral structures at one of the two normal oral meridians, followed by interpolation of reversed oral structures at a new location to the cell's right of the disappearing normal oral meridian. At the same time, the contractile vacuole pore (CVP) set on the side of the cell that is undergoing the transformation shifts to the left. The combination of these events creates a symmetrical large-scale organization in which both of the CVP sets are situated on one side of the cell, between the normal and the partially reversed oral apparatus. This unilateral positioning of CVP sets is commonly manifested even when reversed oral structures are absent. These configurations probably represent intermediate stages in the transformation of balanced typical doublets into singlets. We propose that this pathway of regulation from the doublet to the singlet state, like the more common one that starts from unbalanced typical doublets (described in the preceding paper), involves reverse intercalation. The remarkable resemblance between the transient configuration described here and the stable configuration of janus mutant cells leads us to suggest that the phenotype of the mutant is also a consequence of reverse-intercalation, in that case provoked by a loss of capacity to maintain positional values rather than by a geometrical instability in the system of positional values.

By two-dimensional electrophoresis it is demonstrated that the macrostomal oral apparatus of Tetrahymena vorax contains about 55 unique polypeptides in addition to a set of about 145 polypeptides also found in the microstomal oral apparatus.

Differentiation of the oral apparatus in the heterotrich ciliate Stentor coeruleus is accompanied by striking changes in macronuclear morphology. The macronucleus normally exists as a chain of nodes, but during oral differentiation it coalesces into a rounded, irregular mass in the center of the cell, then elongates into a long rod, and finally renodulates. The technique of nuclear transplantation has been used to analyze each one of these macronuclear changes in terms of nucleocytoplasmic interactions. Macronuclei in various morphological states were transferred into atypical cytoplasmic environments. The experimental results lead to the conclusion that the three changes in macronuclear morphology which accompany oral differentiation are regulated by a corresponding sequence of cytoplasmic states, all of which are experimentally distinguishable from conditions prevailing in the cytoplasm of the non-differentiating cell. Various properties of these nucleocytoplasmic interactions have also been analyzed by nuclear transplantation. Among the findings are evidence that the nucleus cannot respond immediately to the cytoplasmic change initiating its coalescence and that it cannot complete coalescence or elongation in the absence of the cytoplasmic conditions which initiate these processes. The results are discussed with regard to their implication for the control of nuclear morphology and function in ciliates and metazoan cells.

This paper presents further evidence that the cortex controls macronuclear replication and basal body production during the cell cycle of Stentor . At the onset of cell division, basal body production occurs on the ventral side of the cell to form an oral primordium; this structure develops slowly into the oral apparatus destined for the posterior daughter cell. Meanwhile, a series of morphological changes in the macronucleus (coalescence, elongation, nodulation) doubles the number of nodes in preparation for division. When a cell undergoing oral development is grafted to a morphostatic cell of equal size, oral development is usually induced in the morphostatic component and the two members of the graft complex eventually become synchronized with respect to macronuclear morphology. However, primordium induction and nuclear synchronization usually do not occur when the 2 members of the graft complex are separated by cortical discontinuities which heal gradually, even though the graft components demonstrably share a common endoplasm throughout the experiment. These results suggest that the cell surface may control the replication of organelles such as the macronucleus and basal bodies which are normally kept “in step” with the cell cycle in such a way that they are not lost or reproduced too frequently.

Much of the cell surface on the ciliate Tetrahymena thermophila is covered by a polarized lattice of cytoskeletal structures that are associated with basal bodies of the ciliary rows. Unique structural landmarks, including an oral apparatus and contractile vacuole pores, develop before cell division in localized domains located, respectively, posterior and anterior to the transverse fission zone. All of these structures can be visualized by specific monoclonal antibodies. A single-locus recessive mutation, disorganized-A (disA), primarily affects the striated rootlets of the ciliary-row basal bodies and brings about a severe disorganization in the positioning and orientation of these basal bodies and associated cytoskeletal elements. Nonetheless, the new oral apparatus, contractile vacuole pores, and other unique structures appeared at or near their normal sites along the anteroposterior axis of disA cells, indicating that the positioning of these localized structures is not dependent on the integrity of the ciliary rows. Abnormalities were present in the details of construction of some of the localized structures and in aspects of cell shape that may be influenced by these details. In the main, however, analysis of disA mutant cells indicates that intracellular domains near the cell poles develop independently of the vectorial polarity of the ciliary rows.

This study shows that development of the new soma during sexual reproduction in ciliates can be conceptualized on the same basis as embryogenesis in multicellular organisms. In conjugating Paramecium, development of a new oral apparatus takes place during fertilization and the first three divisions of the zygotic nucleus and completes well before the postsexual cell undergoes the first cell fission. The control of oral development is analyzed by microsurgical removal of the zygotic nucleus or the postzygotic nuclei from conjugants. The enucleated exconjugants can pass through an early hurdle in oral development (the initiation of oral membranelle assembly) and subsequently develop an oral apparatus. Such oral apparatuses nevertheless exhibit structural and functional abnormalities including fragmentation and misalignment of oral membranelles, absence of the postoral microtubular bundle, reduction in the length of buccal cavity, and impaired phagocytosis. Other stomatogenic aspects, such as the arrangement of basal bodies in the oral membranelles, remain unaffected. The two groups of exconjugants, one derived from cells enucleated at the zygotic stage, and the other at the postzygotic stage, exhibit the same types of oral abnormality. We conclude that (i) the zygotic nucleus is not essential for the initiation of oral membranelle assembly. The existence of zygotic signals for subsequent oral development is not ruled out, but these are insufficient. (ii) Postzygotic nuclei, as well as maternal nuclei (the old somatic nucleus and meiotic derivatives of the germ nucleus), control oral development. This reveals a parallelism between postsexual development in ciliates and the early embryology of multicellular organisms, in their reliance on information provided by maternal, as well as early postzygotic nuclei. (iii) The activity of the old somatic nucleus alone is not sufficient for the later stages of oral development. Probably, some stomatogenic functions of the old somatic nucleus normally utilized for the later stages of oral development in binary fission are inactivated during sexual reproduction. Alternatively, the old somatic nucleus may rely on some critical conditions prescribed by the postzygotic nuclei in order to act.

The patterns of increase in numbers of argentophilic elements (kinetosomes) are studied during the cell cycle of Tetrahymena pyriformis, syngen 1. The patterns suggest that new ciliary units are added uniformly to all ciliary rows during the early part of the cell cycle. After the start of the formation of the new oral apparatus, the rate of increase of the row to the right of the oral apparatus is increased and the rate of growth of the row to the left is decreased. The increase in total ciliary units, both somatic and oral, may well be constant for the entire cell cycle.
